Exploring the neural underpinnings of chord prediction uncertainty: an electroencephalography (EEG) study

Predictive processing in the brain, involving interaction between interoceptive (bodily signal) and exteroceptive (sensory) processing, is essential for understanding music as it encompasses musical temporality dynamics and affective responses. This study explores the relationship between neural correlates and subjective certainty of chord prediction, focusing on the alignment between predicted and actual chord progressions in both musically appropriate chord sequences and random chord sequences. Participants were asked to predict the final chord in sequences while their brain activity was measured using electroencephalography (EEG). We found that the stimulus preceding negativity (SPN), an EEG component associated with predictive processing of sensory stimuli, was larger for non-harmonic chord sequences than for harmonic chord progressions. Additionally, the heartbeat evoked potential (HEP), an EEG component related to interoceptive processing, was larger for random chord sequences and correlated with prediction certainty ratings. HEP also correlated with the N5 component, found while listening to the final chord. Our findings suggest that HEP more directly reflects the subjective prediction certainty than SPN. These findings offer new insights into the neural mechanisms underlying music perception and prediction, emphasizing the importance of considering auditory prediction certainty when examining the neural basis of music cognition.

Neural Correlates of Anhedonia in Major Depressive Disorder: Insights from Concurrent Analysis of Feedback-Related Negativity and Stimulus-Preceding Negativity.

Anhedonia, a core symptom of major depressive disorder (MDD), is explored in this study, focusing on the neural underpinnings through the examination of two event-related potential (ERP) components: feedback-related negativity (FRN) and stimulus-preceding negativity (SPN). This cross-sectional study was conducted in China from March 2022 to March 2023. It involved 35 MDD patients and 31 healthy controls (HC) participating in a modified 2-door task with simultaneous EEG recordings. Depression severity and anhedonia were assessed using the Hamilton Depression Scale (HAMD) and the Temporal Experience of Pleasure Scale (TEPS-CV), respectively. FRN and SPN metrics, along with correlations with each other and clinical assessments, were examined. In comparison to the HC group, the MDD group exhibited significantly lower scores in TEPS-CV (t = 2.854, p = 0.006) and its subscales (t = -3.596, p = 0.001 and t = 2.434, p = 0.018, respectively), along with consistently reduced amplitudes of FRN (F 1.64= 4.726, p = 0.033) and SPN (F 1.64= 4.195, p = 0.045) across all conditions. Limited correlations were observed between ERP metrics and clinical indicators, except for positive correlations between FRN amplitudes (loss minus win) and HAMD scores (r = 0.392, p = 0.020), and SPN amplitudes after losing (SPN-L) and TEPS-CV consumption subscale scores (r = 0.357, p = 0.035). Notably, while the HC group displayed no significant FRN-SPN correlations, the MDD group exhibited positive FRN-SPN correlations under distinct conditions (r = 0.376, p = 0.026 and r = 0.355, p = 0.037, respectively). Our data reveal subjective and objective anhedonia in both consumption and anticipation, suggesting a shared impairment in reward feedback processing and anticipatory neural mechanisms in individuals with MDD. These findings deepen our understanding of anhedonia's neural foundations and may guide targeted interventions for this core symptom.

Acute stress selectively blunts reward anticipation but not consumption: An ERP study

Stress-induced dysfunction of reward processing is documented to be a critical factor associated with mental illness. Although many studies have attempted to clarify the relationship between stress and reward, few studies have investigated the effect of acute stress on the temporal dynamics of reward processing. The present study applied event-related potentials (ERP) to examine how acute stress differently influences reward anticipation and consumption. In this study, seventy-eight undergraduates completed a two-door reward task following a Trier Social Stress Task (TSST) or a placebo task. The TSST group showed higher cortisol levels, perceived stress, anxiety, and negative affect than the control group. For the control group, a higher magnitude of reward elicited a reduced cue-N2 but increased stimulus-preceding negativity (SPN), suggesting that controls were sensitive to reward magnitude. In contrast, these effects were absent in the stress group, suggesting that acute stress reduces sensitivity to reward magnitude during the anticipatory phase. However, the reward positivity (RewP) and P3 of both groups showed similar patterns, which suggests that acute stress has no impact on reward responsiveness during the consummatory phase. These findings suggest that acute stress selectively blunts sensitivity to reward magnitude during the anticipatory rather than the consummatory phase.

The impact of threat of shock-induced anxiety on alerting, orienting, and executive function in women: an ERP study.

The present study used a combination of the Threat-of-Shock paradigm and the Attention Network Test (ANT) to investigate how induced anxiety affects alerting, orienting, and executive control and whether individual differences in threat sensitivity moderate these effects. Forty-two female subjects completed the ANT task in alternation under shock-threat and no-shock ("safe") conditions while event-related potentials (ERPs) were recorded. The results showed that anxiety induced by the threat of shock had a significant impact on alerting and executive control functions at the neural level. Specifically, alerting-related N1 and stimulus-preceding negativity (SPN) differences between double cue and no cue conditions were greater in the threat versus safe state, suggesting that the induced anxiety promoted the early perception of cues and preparation for the target. Moreover, executive control-related P3 and sustained potential (SP) differences between incongruent and congruent trials were greater in the threat versus safe state, indicating that the induced anxiety might improve the attentional allocation efficiency and stimulate subjects to recruit more cognitive resources to resolve conflicts. However, orienting-related ERPs were not affected by the threat of shock, but the threat of shock promoted the processing efficiency of spatial-cue at the behavioral level. Analysis of individual differences revealed that trait anxiety moderated the attentional allocation efficiency when performing executive control related tasks in the threat versus safe state. Our findings demonstrate the adaptive significance of the threat of shock-induced anxiety in that being in an anxious state can enhance individuals' alerting, orienting, and executive functions.

Neural Differences in Relation to Risk Preferences during Reward Processing: An Event-Related Potential Study.

Inter-individual variability in risk preferences can be reflected in reward processing differences, making people risk-seekers or risk-averse. However, the neural correlates of reward processing in individuals with risk preferences remain unknown. Consequently, this event-related potential (ERP) study examined and compared electrophysiological correlates associated with different stages of reward processing in risk-seeking and risk-averse groups. Individuals scoring in the bottom and top 20% on the Balloon Analogue Risk Task (BART) were deemed risk-averse and risk-seeking, respectively. Participants engaged in a gambling task while their electroencephalogram (EEG) was recorded. Risk-seekers tended to choose high-risk options significantly more frequently than low-risk options, whereas risk-averse individuals chose low-risk options significantly more frequently than high-risk ones. All participants selected the low-risk alternative more slowly than the high-risk option. During the anticipation stage, the low-risk option elicited a relatively attenuated stimulus-preceding negativity (SPN) response from risk-seekers compared to risk-averse participants. During the outcome stage, feedback-related negativity (FRN) increased in risk-seekers responding to greater losses but not in risk-averse participants. These results indicate that ERP components can detect differences in reward processing during risky situations. In addition, these results suggest that motivation and cognitive control, along with their associated neural processes, may play a central role in differences in reward-based behavior between the two groups.

Electrophysiological markers for anticipatory processing of nocebo-augmented pain.

Nocebo effects on pain are widely thought to be driven by negative expectations. This suggests that anticipatory processing, or some other form of top-down cognitive activity prior to the experience of pain, takes place to form sensory-augmenting expectations. However, little is known about the neural markers of anticipatory processing for nocebo effects. In this event-related potential study on healthy participants (n = 42), we tested whether anticipatory processing for classically conditioned nocebo-augmented pain differed from pain without nocebo augmentation using stimulus preceding negativity (SPN), and Granger Causality (GC). SPN is a slow-wave ERP component thought to measure top-down processing, and GC is a multivariate time series analysis used to measure functional connectivity between brain regions. Fear of pain was assessed with the Fear of Pain Questionnaire-III and tested for correlation with SPN and GC metrics. We found evidence that both anticipatory processing measured with SPN and functional connectivity from frontal to temporoparietal brain regions measured with GC were increased for nocebo pain stimuli relative to control pain stimuli. Other GC node pairs did not yield significant effects, and a lag in the timing of nocebo pain stimuli limited interpretation of the results. No correlations with trait fear of pain measured after the conditioning procedure were detected, indicating that while differences in neural activity could be detected between the anticipation of nocebo and control pain trials, they likely were not related to fear. These results highlight the role that top-down processes play in augmenting sensory perception based on negative expectations before sensation occurs.

Intolerance of uncertainty and neural measures of anticipation and reactivity for affective stimuli.

Intolerance of uncertainty (IU) is a transdiagnostic construct referring to the aversive interpretation of contexts characterized by uncertainty. Indeed, there is a growing body of research examining individual differences in IU and how these are associated with emotional anticipation and reactivity during periods of certainty and uncertainty, however, how these associations are reflected via neurophysiological indices remain understudied and poorly understood. The present study examined the relationship between self-reported IU and neurophysiological measures of emotional anticipation and reactivity, namely stimulus preceding negativity (SPN) and late positive potential (LPP), and self-report measures of emotional experiences. These measures were captured during an S1-S2 picture viewing tasks in which participants were presented with cues (S1) that either indicated the affective valence of upcoming picture (S2) or provided no information about the valence. Findings here provide evidence for significant associations between SPN amplitude and IU scores during uncertain and certain-positive cueing conditions, and significant associations between LPP amplitude and IU scores during both certain- and uncertain-negative picture viewing conditions that appear driven by prospective IU sub-scores. These positive associations between IU and SPN amplitude are suggestive of heightened emotional anticipation following S1 cues, while positive associations between IU and LPP are suggestive of heightened emotional reactivity following S2 images. These findings are discussed in detail relative to existing IU literature, and potential implications of these findings.

The influence of self-focused attention on emotional picture processing: An ERP study.

Research has documented neurophysiological indicators of anticipation (Stimulus Preceding Negativity [SPN]) and perception (Late Positive Potential [LPP]) of threat, yet little is known as to how self-focused attention manipulations influence emotion processing within the context of cued picture viewing. With self-referent attention moderating attention to external stimuli, it is necessary to document how self-focused attention impacts attention and the ability to emotionally process external threat. The goal of the present study was to evaluate the impact of self-focused attention on the anticipation and perceptual processing of unpleasant pictures within a cued-picture viewing paradigm among 33 participants. Overall, the results suggest that the self-focused attention manipulations disrupted anticipation but not processing of pictures, as indexed by the SPN and LPP respectively. Self-focused attention appears to disrupt the preparatory attention for upcoming unpleasant stimuli, potentially through loading cognitive resources or activation of associative defensive responding. Collectively, these findings demonstrate the impact of self-focused attention within the context of emotional picture processing and suggest further areas of investigation.

Transdiagnostic Fear and Anxiety: Prospective Prediction Using the No-Threat, Predictable Threat, and Unpredictable Threat Task

BackgroundFear and anxiety are distinct dimensions of psychopathology that may be characterized by differences in dimensional threat reactivity. Heightened response to predictable threat is hypothesized to underlie fear symptomatology, whereas increased response to unpredictable threat may underlie anxiety. Despite widespread acceptance of this model, these purported associations have rarely been tested, and the prognostic value of predictable and unpredictable threat responding is unclear. Here we examined multilevel indicators of predictable and unpredictable threat response as cross-sectional correlates and prospective predictors of transdiagnostic fear and anxiety. MethodsFifty-two individuals with varying levels of internalizing psychopathology (31 female) performed the no-threat, predictable threat, and unpredictable threat task. Transdiagnostic fear and anxiety were assessed at baseline (time 1) and approximately 1.5 years later (time 2). We used event-related potential, the stimulus-preceding negativity, as a measure of threat anticipation and startle eyeblink as a measure of defensive reactivity during the no-threat, predictable threat, and unpredictable threat task. These probes were assessed as cross-sectional correlates and prospective predictors of fear and anxiety. ResultsParticipants with larger time 1 stimulus-preceding negativities to predictable threat were characterized by greater time 1 fear. Larger time 1 stimulus-preceding negativities to unpredictable threat were associated with greater increases in time 2 anxiety. Heightened time 1 startle to predictable threat predicted larger increases in time 2 fear. ConclusionsResults validate predictable and unpredictable threat responding as dimensional correlates of transdiagnostic fear versus anxiety and suggest that psychophysiological measures of predictable and unpredictable threat response hold promise as prospective predictors of trajectories of fear and anxiety.

Fear memory in humans is consolidated over time independently of sleep

Fear memories can be altered after acquisition by processes, such as fear memory consolidation or fear extinction, even without further exposure to the fear-eliciting stimuli, but factors contributing to these processes are not well understood. Sleep is known to consolidate, strengthen, and change newly acquired declarative and procedural memories. However, evidence on the role of time and sleep in the consolidation of fear memories is inconclusive. We used highly sensitive electrophysiological measures to examine the development of fear-conditioned responses over time and sleep in humans. We assessed event-related brain potentials (ERP) in 18 healthy, young individuals during fear conditioning before and after a 2-hour afternoon nap or a corresponding wake interval in a counterbalanced within-subject design. The procedure involved pairing a neutral tone (CS+) with a highly unpleasant sound. As a control, another neutral tone (CS−) was paired with a neutral sound. Fear responses were examined before the interval during a habituation phase and an acquisition phase as well as after the interval during an extinction phase and a reacquisition phase. Differential fear conditioning during acquisition was evidenced by a more negative slow ERP component (stimulus-preceding negativity) developing before the unconditioned stimulus (loud noise). This differential fear response was even stronger after the interval during reacquisition compared with initial acquisition, but this effect was similarly pronounced after sleep and wakefulness. These findings suggest that fear memories are consolidated over time, with this effect being independent of intervening sleep.

The contralateral effects of anticipated stimuli on brain activity measured by ERP and fMRI.

The present study examined the effects of unilateral stimulus presentation on the right hemisphere preponderance of the stimulus-preceding negativity (SPN) in the event-related potential (ERP) experiment, and aimed to elucidate whether unilateral stimulus presentation affected activations in the bilateral anterior insula in the functional magnetic resonance imaging (fMRI) experiment. Separate fMRI and ERP experiments were conducted using visual and auditory stimuli by manipulating the position of stimulus presentation (left side or right side) with the time estimation task. The ERP experiment revealed a significant right hemisphere preponderance during left stimulation and no laterality during the right stimulation. The fMRI experiment revealed that the left anterior insula was activated only in the right stimulation of auditory and visual stimuli whereas the right anterior insula was activated by both left and right stimulations. The visual condition retained a contralateral dominance, but the auditory condition showed a right hemisphere dominance in a localized area. The results of this study indicate that the SPN reflects perceptual anticipation, and also that the anterior insula is involved in its occurrence.

Dissociated modulations of intranasal vasopressin on prosocial learning between reward-seeking and punishment-avoidance.

As an integral ingredient of human sociality, prosocial behavior requires learning what acts can benefit or harm others. However, it remains unknown how individuals adjust prosocial learning to avoid punishment or to pursue reward. Given that arginine vasopressin (AVP) is a neuropeptide that has been involved in modulating various social behaviors in mammals, it could be a crucial neurochemical facilitator that supports prosocial learning. In 50 placebo controls and 54 participants with AVP administration, we examined the modulation of AVP on the prosocial learning characterized by reward and punishment framework, as well as its underlying neurocomputational mechanisms combining computational modeling, event-related potentials and oscillations. We found a self-bias that individuals learn to avoid punishment asymmetrically more severely than reward-seeking. Importantly, AVP increased behavioral performances and learning rates when making decisions to avoid losses for others and to obtain gains for self. These behavioral effects were underpinned by larger responses of stimulus-preceding negativity (SPN) to anticipation, as well as higher punishment-related feedback-related negativity (FRN) for prosocial learning and reward-related P300 for proself benefits, while FRN and P300 neural processes were integrated into theta (4-7 Hz) oscillation at the outcome evaluation stage. These results suggest that AVP context-dependently up-regulates altruism for concerning others' losses and reward-seeking for self-oriented benefits. Our findings provide insight into the selectively modulatory roles of AVP in prosocial behaviors depending on learning contexts between proself reward-seeking and prosocial punishment-avoidance.

When the Counterpart Chooses the Opposite: The First Mover's Anticipation and Evaluation of the Final Feedback in Gambles.

This research examines the effect of response (in)consistency on the first mover’s anticipation and evaluation of the performance feedback in gambles. In a two-player gambling task, the participant played as the first mover while the confederate served as the second mover, who made their gambles in sequence. A more pronounced feedback-related negativity (FRN) was observed when the first mover noticed that the second mover chose a different option from him/her. An enlarged stimulus-preceding negativity (SPN) was observed when the first mover was anticipating the final feedback in this condition. Interestingly, consistent responses gave rise to a more pronounced FRN difference wave (d-FRN) during the feedback stage. Taken together, these results suggest that response discrepancy would modulate the first mover’s anticipation and evaluation of the final feedback in gambles.

Reduced neural activity when anticipating social versus nonsocial rewards in schizophrenia: Preliminary evidence from an ERP study

Diminished social motivation is a core feature of schizophrenia that might reflect disturbances in social reward processing. It is not known whether these disturbances reflect anticipatory (“wanting”) and/or consummatory (“liking”) pleasure deficits. The primary aim of this study was to examine social versus nonsocial reward processing during these temporally distinct substages using event-related potential (ERP) components. Twenty-three schizophrenia participants and 20 healthy participants completed an incentive delay task with social (i.e., smiling expressions) and nonsocial (i.e., money) rewards. We measured two anticipatory ERPs (i.e., “wanting”) (target anticipation: Contingent Negative Variation [CNV]; feedback anticipation: Stimulus Preceding Negativity [SPN]) and one consummatory ERP (i.e., “liking”) (feedback receipt: P300). As a secondary aim, we examined correlations between the ERPs and interview-rated motivational negative symptoms and social functioning. Schizophrenia participants showed overall less target anticipation (blunted CNV) across all trials (social and nonsocial) than healthy participants. Importantly, schizophrenia participants exhibited less anticipation of social rewards relative to nonsocial rewards (SPN), whereas healthy participants showed similar anticipation for both reward types. Both groups showed similar responses to social and nonsocial reward receipt (P300). Furthermore, social reward anticipation during the incentive delay task was associated with more social approach behaviors in the real-world. Together, these findings provide preliminary evidence for intact social reward “liking” and impaired “wanting” in schizophrenia.

Possible Event-Related Potential Correlates of Voluntary Attention and Reflexive Attention in the Emei Music Frog.

Simple SummaryWe investigated auditory event-related potentials (ERP) related to auditory attention in music frogs. Our objective was to explore whether ERP components related to voluntary attention and reflexive attention exist in frogs. We found that the amplitudes of stimulus preceding negativity (SPN, related to voluntary attention and under up-down control) evoked by silence replacement in the telencephalon were the largest when the sequence of acoustic stimuli could be predicted, while the N1 amplitudes (related to reflexive attention and under bottom-up control) evoked in the mesencephalon were the largest when the sequence of acoustic stimuli could not be predicted. This suggests that human-like ERP components related to voluntary attention and reflexive attention exist in the lower vertebrates also.Attention, referring to selective processing of task-related information, is central to cognition. It has been proposed that voluntary attention (driven by current goals or tasks and under top-down control) and reflexive attention (driven by stimulus salience and under bottom-up control) struggle to control the focus of attention with interaction in a push–pull fashion for everyday perception in higher vertebrates. However, how auditory attention engages in auditory perception in lower vertebrates remains unclear. In this study, each component of auditory event-related potentials (ERP) related to attention was measured for the telencephalon, diencephalon and mesencephalon in the Emei music frog (Nidirana daunchina), during the broadcasting of acoustic stimuli invoking voluntary attention (using binary playback paradigm with silence replacement) and reflexive attention (using equiprobably random playback paradigm), respectively. Results showed that (1) when the sequence of acoustic stimuli could be predicted, the amplitudes of stimulus preceding negativity (SPN) evoked by silence replacement in the forebrain were significantly greater than that in the mesencephalon, suggesting voluntary attention may engage in auditory perception in this species because of the correlation between the SPN component and top-down control such as expectation and/or prediction; (2) alternately, when the sequence of acoustic stimuli could not be predicted, the N1 amplitudes evoked in the mesencephalon were significantly greater than those in other brain areas, implying that reflexive attention may be involved in auditory signal processing because the N1 components relate to selective attention; and (3) both SPN and N1 components could be evoked by the predicted stimuli, suggesting auditory perception of the music frogs might invoke the two kind of attention resources simultaneously. The present results show that human-like ERP components related to voluntary attention and reflexive attention exist in the lower vertebrates also.

An event-related potential investigation of distanced self-talk: Replication and comparison to detached reappraisal

Emotion regulation is critical for managing stress, but many regulatory strategies consume high levels of cognitive resources to implement, which are depleted under stress. This raises a conundrum: the tools we have to feel better may be ineffective when they are most needed. Recent event-related potential (ERP) research indicates that distanced self-talk (i.e., reflecting on one's experiences using non-first-person singular pronouns and one's name) reduces negative emotional reactivity without overtaxing cognitive resources. Here, we report the first direct replication of this work and extend it by examining how distanced self-talk compares to detached reappraisal, one of the most frequently studied regulatory techniques. Sixty-seven participants were randomly assigned to an emotion regulation picture task and instructed to reflect on the feelings they experienced in response to viewing negative emotional images using distanced self-talk or detached reappraisal while ERPs were measured. Directly replicating past findings, distanced self-talk led to a reduction in an affective arousal ERP, the late positive potential (LPP), without increasing stimulus preceding negativity (SPN), an ERP that reflects anticipatory and preparatory processing. These results further bolster support for distanced self-talk as a relatively effortless emotion regulation strategy. On the other hand, detached reappraisal was neither associated with the modulation of the LPP nor the SPN. Due to the failed replication of the reappraisal effect, a direct comparison between emotion regulation strategies was not conducted. Methodological limitations that may have contributed to the reappraisal failure and future directions for comparisons between emotion regulation strategies are discussed.

P320. Waiting for the Payoff: Electrical Anticipation Response in Individuals With Depression/Anxiety Disorders

Reward processing dysfunction is considered a core characteristic of depression and anxiety disorder (DEP/ANX). However, research in DEP/ANX has focused mainly on reward receipt as opposed to anticipation. The stimulus preceding negativity (SPN) is a slow event-related potential recorded during electroencephalography (EEG), thought to reflect anticipatory brain activity. We aimed to examine the effect of MDD on SPN amplitude and its relationship with future treatment completion (>7 sessions of behavioral activation or exposure therapy).

Neural dynamics of effort-modulated reward processing.

Previous studies have established that effort can either decrease reward value as a cost or increase reward value as an augmenter, which is referred to as the effort paradox. The present event-related potential study investigated the neural dynamics underlying the modulation of reward processing by effort expenditure. Thirty-two participants completed a modified monetary incentive delay task in a high-effort context and a low-effort context to earn monetary rewards while their EEG was recorded during effort anticipation, effort execution, and effort completion, respectively. Results revealed the multiphase nature of the effort-reward relationship, which unfolds over time. During effort anticipation, effort discounts reward value as reflected by a reduced contingent negative variation for high- versus low-effort trials. During effort exertion, the target-P3 reward effect was accrued by effort expenditure. Finally, effort also provides its own value following effort completion, as revealed by an enhanced stimulus-preceding negativity for high-effort relative to low-effort trials. These findings provide new insights into the relationship between effort expenditure and reward processing and are helpful to mitigate the effort paradox.

Modulating social feedback processing by deep TMS targeting the medial prefrontal cortex: Behavioral and electrophysiological manifestations

Neuroimaging studies have suggested that the medial prefrontal cortex (mPFC) is a key brain region for social feedback processing, but previous findings are largely based on correlational approaches. In this study, we use the deep transcranial magnetic stimulation (dTMS) to manipulate mPFC activity, then investigate participants’ behavioral performance and event-related potentials (ERPs) during the Social Judgment Paradigm. A between-subject design was applied, such that both the active dTMS group and the sham group consisted of 30 participants. We found that the sham group was more likely to predict that they would be socially accepted (rather than rejected) by peers, but the same was not true in the active group. Additionally, this study is the first one to observe ERP signal changes in response to dTMS manipulation. ERP results show that both the expectation stage and the experience stage of social feedback processing were modulated by dTMS: (1) at the expectation stage, the P1 component was smaller in the active group than the sham group, while the stimulus-preceding negativity showed a stronger differentiating effect between positive and negative prediction in the sham group than the active group; (2) at the experience stage, the sensitivity of the late positive potential to the valence and predictability of social feedback was stronger in the sham group than the active group. These results improve our understanding about the relationship between the mPFC and social feedback processing.

Investigating Changes in Reward-Related Neural Correlates After PEERS Intervention in Adolescents With ASD: Preliminary Evidence of a "Precision Medicine" Approach.

Background: The Social Motivation Hypothesis proposes that individuals with autism spectrum disorder (ASD) experience social interactions as less rewarding than their neurotypical (TD) peers, which may lead to reduced social initiation. Existing studies of the brain's reward system in individuals with ASD report varied findings for anticipation of and response to social rewards. Given discrepant findings, the anticipation of and response to social rewards should be further evaluated, particularly in the context of intervention outcome. We hypothesized that individual characteristics may help predict neural changes from pre- to post-intervention.Methods: Thirteen adolescents with ASD received the Program for the Education and Enrichment of Relational Skills (PEERS) intervention for 16 weeks; reward-related EEG was collected before and after intervention. Fourteen TD adolescents were tested at two timepoints but did not receive intervention. Event-related potentials were calculated to measure anticipation of (stimulus-preceding negativity; SPN) and response to (reward-related positivity; RewP) social and non-social rewards. Additionally, measures of social responsiveness, social skills, and intervention-engagement were collected. Group differences were analyzed as well as individual differences using prediction models.Result: Parent-reported social responsiveness and social skills improved in adolescents with ASD after participation in PEERS. ASD adolescents displayed marginally decreased anticipation of social rewards at post-intervention compared to pre-intervention. Regression models demonstrated that older adolescents and those with lower parent-reported social motivation prior to participation in PEERS displayed marginally increased social reward anticipation (more robust SPN) from pre- to post-intervention. Participants who displayed more parent-reported social motivation before intervention and were more actively engaged in the PEERS intervention evidenced increased social reward processing (more robust RewP) from pre- to post-intervention.Conclusion: Findings suggest that there may be differences in saliency between wanting/anticipating social rewards vs. liking/responding to social rewards in individuals with ASD. Our findings support the hypothesis that identification of individual differences may predict which adolescents are poised to benefit the most from particular interventions. As such, reported findings set the stage for the advancement of “precision medicine.” This investigation is a critical step forward in our ability to understand and predict individual response to interventions in individuals with ASD.
