Effects of Pb2+ on the transient outward potassium current in acutely dissociated rat hippocampal neurons.

Modulation of the voltage-dependent transient outward potassium current (IA) by Pb2+ was studied in acutely dissociated rat hippocampal pyramidal cells from the CA1 region at postnatal ages 7-14 days using the conventional whole-cell patch-clamp technique. In the presence of different concentrations of external Pb2+, the initial delay and activation time of IA were concentration-dependently lengthened. In particular, the initial delay was even longer in 1 mM Pb2+, showing no signs of saturation. Pb2+ also slowed the inactivation of IA, for decay time constants in the presence of Pb2+ were increased under the same experimental protocols. The activation curves, which were reasonably fitted by a single Boltzmann function, illustrated that Pb2+ increased the voltage threshold of IA and shifted the normalized activation current-voltage curves to more depolarizing voltage commands. Moreover, Pb2+ significantly affected the steady-state inactivation of IA. The application of Pb 2+ made the curves of the steady-state inactivation of IA shift to more depolarizing voltages with little change in the slopes factors. In brief, the results demonstrated that Pb2+ is a dose- and voltage-dependent, reversible blocker of IA currents of hippocampal CA1 neurons. The observations were fitted by the revised "Kuo and Chen type model", which postulates a Pb2+-selective site near the pore of the IA channel and that modulation of the IA channel by Pb2+ is the result of the competitive influences of Pb2+ on opening and inactivating different pathways.

Protein kinase-dependent phosphorylation and cannabinoid receptor modulation of potassium A current (IA) in cultured rat hippocampal neurons.

The potent cannabinoid receptor agonist WIN 55,212-2 produces positive shifts in steady-state inactivation of the potassium A current (IA) in rat hippocampal neurons via an adenosine 3',5'-cyclic monophosphate (cAMP)-, protein kinase A (PKA)-dependent process. This effect is probably mediated by phosphorylation or dephosphorylation of the IA channel protein. The role of protein phosphorylation in this cascade was tested by testing cannabinoid actions in cultured hippocampal neurons (pyramidal cells) that were exposed also to either the catalytic subunit of PKA (PKAc), a PKA-specific phosphorylation inhibitor (IP-20, Walsh peptide), or a potent protein phosphatase inhibitor (okadaic acid). Cannabinoids such as WIN 55,212-2 produce a positive (rightwards) shift in the steady-state inactivation of IA, thus providing increased current at a given membrane voltage. Cells dialyzed with PKAc showed a negative shift in IA inactivation, opposite to that produced by cannabinoids, and similar to that produced by increased levels of cAMP. In addition, PKAc completely blocked the positive shift produced by WIN 55,212-2. In contrast, dialysis of cells with IP-20 produced a positive shift in steady state inactivation of IA, similar to that produced by WIN, but the effects were not additive with cannabinoid receptor activation. The phosphatase inhibitor, okadaic acid produced a small negative shift in IA steady-state inactivation when administered alone, and blocked the positive shift produced by WIN 55,212-2. Okadaic acid also enhanced the negative shift in IA inactivation when co-administered with forskolin. The effects of okadaic acid and WIN 55,212-2 were not additive, suggesting a common pathway. These results demonstrate that IA is altered by direct manipulations of the phosphorylation status of the channel protein, and that cannabinoid effects on IA are probably mediated by dephosphorylation of the IA channel.

Transient outward current in adult rat supraoptic neurones with slice patch-clamp technique: inhibition by angiotensin II.

1. Outward potassium currents were recorded from microscopically identified supraoptic neurones of adult Wistar male rats using the whole-cell patch-clamp technique in thin-slice preparations. The basic characteristics of transient outward current (IA or A-current) and the effects of angiotensin II (AII) on the currents were studied. 2. IA was isolated by subtracting outward currents elicited by stepping from two different holding potentials to a test potential or by applying 4-aminopyridine (4-AP) at 5 mM. The isolated IA had a threshold for activation between -55 and -65 mV and was characterized by fast activation and inactivation. Values of the time to peak and the inactivation time constants for current decay at different test potentials were voltage dependent. 3. Normalized currents for activation and steady-state inactivation of IA were fitted to the Boltzmann function. The mid-points and the slope factors were, respectively, -35.0 and -14.3 +/- 0.40 mV (n = 5) for the activation curve, and -72.0 and 7.0 +/- 0.68 mV (n = 5) for the inactivation curve. 4. The time course of recovery from inactivation was best fitted to a single exponential function with the time constant of 37.8 +/- 6.6 ms (n = 6). 5. The effects of AII on IA and delayed rectifier current (IK) were investigated. According to their responses to AII, cells were classified into two groups, sensitive and low-sensitive. Bolus injection of AII (10 microM, 100 microliters) decreased the IA amplitude by 25.1 +/- 2.4% in seven (53.8%) of the thirteen neurones tested (sensitive group), whereas the other six neurones (low-sensitive group) changed by only 2.2 +/- 0.8%. Perfusion of AII (0.1 microM) decreased the IA amplitude by 21.3 +/- 3.1% in six (54.5%) of eleven neurones tested (sensitive group), whereas the other five neurones (low-sensitive group) changed only by 1.7 +/- 0.8%. Bolus injection of AII (10 microM, 100 microliters) decreased the IK amplitude 9.6 +/-1.6% mV in five (45.5%) of the eleven neurones tested (sensitive group), whereas the other six neurones (low-sensitive group) changed only by 0.46 +/- 0.27%. In the sensitive groups, the reduction of IA by AII was significantly larger than that of IK (P < 0.05). 6. Application of saralasin at 1 microM, an AII antagonist, blocked the effects of AII on IA. 7. These results suggest that the excitatory action of AII on supraoptic neurosecretory cells is mediated at least in part through suppression of IA.

Early regulation of membrane excitability by ras oncogene proteins

Two electrode voltage clamp conditions were used to study the early effects on ionic membrane channels of the intracellularly injected proto-oncogenic form of c-Ha-ras (c-ras) and its oncogenic counterpart v-Ha-ras (v-ras). These experiments were conducted on isolated somata of identified fully differentiated neurons of the sea snail Hermissenda. 20 min after c-ras, and 10 min after v-ras intracellular injections into type B medial photoreceptors of Hermissenda, the peak amplitude of two outward potassium currents (IA and IC), across the isolated Type B soma membrane begin to decrease. These two currents have been previously isolated by differences in activation and inactivation kinetics and their response to pharmacological blockers. c- or v-ras injections did not have any effect on a voltage-dependent inward calcium current. Reduction of IA preceded that of IC. Current reductions due to c-ras, but not to v-ras injection reversed spontaneously after 40 min. The voltage dependence of the steady state inactivation of IA shifted toward more negative potentials with ras injections. Ras-mediated cell transformations therefore, could involve, perhaps as initial events, prolonged modification of membrane currents.

Effects of Dopamine on Voltage-Dependent Potassium Currents in Identified Rat Lactotroph Cells

The effects of dopamine (DA) on voltage-dependent potassium currents were investigated in rat lactotrophs maintained in primary culture. Lactotroph cells were identified using the reverse hemolytic plaque assay. Membrane currents and potentials of lactotroph cells were recorded using the patch-clamp recording technique in the 'whole-cell' configuration. In the presence of cobalt (2 mM), two types of voltage-dependent K+ currents were recorded, a voltage-activated delayed K+ current (IK) and a voltage-activated transient K+ current (IA). The current IK was activated at membrane potentials varying from -20 to +40 mV and did not inactivate during prolonged voltage steps (up to 25 s); it was blocked by tetraethylammonium (10 mM). The current IA was activated at membrane potentials higher than -45 mV and showed a voltage-dependent inactivation between -110 and -40 mV; it was slightly inhibited by 4-aminopyridine (5 mM). Under current-clamp conditions, the majority of the cells (60%) showed spontaneous Ca2(+)-dependent action potentials (APs) while silent cells (40%) were excitable by depolarizing current pulses. Bath application of 10 nM DA evoked a hyperpolarizing response, blocked spontaneous APs and decrease the amplitude of evoked APs. Only the hyperpolarizing response faded during the course of the whole cell recording experiments. Under voltage-clamp conditions, DA induced a reversible increase in both voltage-dependent outward K+ currents, without modifying their thresholds. Steady-state inactivation of IA was not affected by DA. These DA-induced responses were dose-dependent and they involved D2 receptor activation. They were mimicked by the specific D2 receptor agonist bromocriptine (10 nM) and blocked by the specific D2 receptor antagonist sulpiride (100 nM), the D1 antagonist SCH 23390 being ineffective. The ability of DA to increase voltage-dependent K+ currents cannot be observed without GTP in the recording pipette. It was pertussis-toxin-sensitive but was affected neither by bath application of 1 mM forskolin nor by the presence of 500 microM cyclic AMP with 500 microM 3-isobutyl-1-methylxanthine in the pipette solutions. We conclude that in lactotroph cells DA specifically increases two voltage-dependent K+ currents via a pertussis-toxin-sensitive guanine nucleotide regulatory protein and appears to be independent of intracellular cyclic AMP. This effect leads to a decrease in the excitability of the cell, explaining in part the inhibitory effect of DA on prolactin release.
