The usefulness of ecotoxicological tools to improve the assessment of water bodies in a climate change reality

This study aimed to analyse the added value of using ecotoxicological tools to complement and improve the assessment of natural water bodies status, in situations of climate change, with a higher frequency of extreme events as floods or droughts.Four water bodies of streams in the Guadiana Basin (Álamos, Amieira, Lucefécit, Zebro) were studied in 2017 and 2018 and classified based on the Water Framework Directive (WFD) parameters: Biological Quality Element – Phytobenthos (diatoms), General chemical and physicochemical elements, Specific pollutants, and Priority Substances. Complementarily, bioassays (including lethal and sublethal parameters) were carried out with organisms of different trophic levels: (i) the bacteria Aliivibrio fischeri; (ii) the microalgae Pseudokirchneriella subcapitata; (iii) the crustaceans Daphnia magna, Thamnocephalus platyurus and Heterocypris incongruens. A classification system with 5 scores was developed, permitting to classify water bodies from non-toxic (EC50 > 100 %; growth and feeding rate > 80 %; blue) to highly toxic (EC50 < 10 %; growth and feeding rate < 10 %; red). The comparison between the classification based on the WFD parameters and on ecotoxicological endpoints showed similar results for 71 % of the samples, and significant positive Pearson correlations were detected between the diatom-based Specific Polluosensitivity Index (SPI) and EC50V.fisheri, the algae growth rate and Shannon diversity index. These results indicate that when the biological quality elements cannot be used (namely under drought or flooding conditions) the application of ecotoxicological bioassays may be a good alternative. Further, when ecotoxicological parameters were included, an increase of worse classifications (Bad and Poor) was observed, revealing an improvement in the sensitivity of the classification, mainly in presence of specific and priority substances. So, the ecotoxicological analysis appears to provide useful information regarding the potential presence of both known and unknown contaminants at concentrations that cause biological effects (even within the WFD limits), in agreement with several authors that have already suggested its use in biomonitoring.

River Diatoms Reflect Better Past than Current Environmental Conditions

The time required for diatom assemblages to integrate the abiotic variables of the aquatic environment has been subject of discussion for many years, with contrasting opinions as shown by field observations and experiments. Numerous diatom-based water quality indices are being used in the bioassessment of freshwaters; one of the most widely used (Specific Polluosensitivity Index or SPI) is compared in this study with water quality status as measured by the Water Quality Index (WQI) obtained from limnological parameters measured at 37 fluvial stations belonging to the Duero Basin (NW Spain). The correlation existing between both metrics was determined using different time lags between water and diatom sample collection. Results show that the correlation coefficient increases as the time gap decreases, but is maximal when comparing WQI scores measured one month before diatom sampling. We thus confirm the initial hypothesis that diatom indices actually indicate past (1 month ago) and not current water conditions in rivers. The practical consequences of this finding are briefly discussed.

The response of benthic diatom community to anthropogenic eutrophication of a river basin under agricultural influence in NE China

Anthropogenic eutrophication has universally threatened river ecological health and has been a key issue in river conservation. Agricultural sewage, which leads to increased nutrient levels and results in the loss of ecological function and biodiversity, results from the growth of agricultural activity and human populations. Consequently, attention has increasingly been focused on developing a relevant tool to describe the relationship between the biotic communities and anthropogenic eutrophication. The benthic diatom community has been widely used as an effective tool for indicating the ecological status of river systems especially in the context of recent international Water Framework Directive policies (WFD). In this study, a statistical method consisting of trophic status index (TSI), Specific Polluosensitivity Index (IPS), Descy Index (DES), redundancy analysis (RDA), and the geographic information system (GIS) technique was conducted to explore the response of benthic diatom community to anthropogenic eutrophication. Initially, the TSI, IPS and DES were determined to differentiate the trophic gradient along a gradient in agricultural intensity. Subsequently, RDA was used to identify spatial distribution patterns of environmental parameters and benthic diatom communities. Finally, the RDA scores as calculated and spatially interpolated were applied to GIS technology to reveal the response of benthic diatom communities to anthropogenic impacts along the trophic gradient. Our results revealed that the studied basin mostly exceeded the eutrophic level. Two different patterns of diatom communities response to ecological gradient were identified based on RDA, one representing the agriculture eutrophication discharges in electrical conductivity (EC) and the other representing organic pollution as chemical oxygen demand (COD). The developed method in this study highlight that the EC was more effective than the nutrient indices in determining diatom distribution in a eutrophic agriculturally-influenced river system.

How good is good ecological status? A test across river typologies, diatom indices and biological elements

Good ecological status is the environmental objective of EU water legislation to be achieved by all European water bodies. However, good ecological status varies depending on national criteria, typology approach, and classification systems used. Since nutrient enrichment is the main cause of river degradation, it is important to establish nutrient criteria that consistently support good ecological status across these influential factors. This study analyzes good ecological status, depending on the typology, classification system and biological element used, and it discusses potential implications of the results for river management. We used a database of 425 sites from northern Spain, corresponding to 11 river types of the Spanish typology derived from physiographic data, or to the four river types resulting from NORTIdiat predictive model, derived from regional diatom reference assemblages. PERMANOVA analysis found significant differences among diatom assemblages across the four river types derived from the NORTIdiat system. Among the classification systems currently in use, or of potential use in the area, the upper P-PO43− threshold, established as the P95 of the class distribution for good ecological status, both NORTIdiat (50.7 μg l−1) and the Multimetric Diatom index (MDIAT; 26.4 μg l−1) were close to proposed thresholds for good status at the EU level. However, this value was much higher for the Specific Polluosensitivity Index (IPS; 118.1 μg l−1). Nutrient thresholds for good status also varied among bioindicators, since the predictive invertebrate-based model NORTI classified 67% of samples with high P-PO43− content in good ecological status, whereas the NORTIdiat classified only 33% of them in good status. Results suggest that current nutrient criteria used to establish good ecological status should be revised, accounting for biological specificity and response of biological elements, to provide a more ecologically coherent approach to preserving or restoring good ecological status.

Periphyton colonization and changes in the diatom assemblages of an artificial urban pond

Artificial ecosystems are sometimes the only opportunity for citizens to experience nature. Maintaining their quality is essential for continuing to enjoy the ecosystem services. We studied an urban pond in the Northwest Iberian Peninsula (Aveiro, Portugal) as a model for testing the suitability of diatom assemblages and indices of water quality in these particular ecosystems. Weekly sampling of three replicas was performed over 28 days on stones and ceramic tiles in a shaded site (Site-SH) and in a sunnier one (Site-SU). The best model of the distance-based analysis included temperature and NH4+as the variables that best explained the diatom assemblages (R 2= 0.43). Chlorophyll-α taken as a proxy for algae biomass, and ash-free dry mass increased until the third week, when a turbid runoff entered the system. After this disturbance, chlorophyll-α increased only at Site-SH (from 14.4 mg m –2 to 62.4 mg m–2); ash-free dry mass increased at Site-SU (from 7.2 mg m–2 to 42.3 mg m–2). The Specific Polluosensitivity Index of the diatoms showed low values below 16 that further decreased with the incubation time. Shannon diversity did not vary between sites, while ANOSIM (analysis of similarity) enabled the identification of differences in the species composition among sites and over time. Cyclotella meneghiniana and Sellaphora nigri were abundant at both sites, but other species showed a prevalence to one of the sites: Nitzschia amphibia and Gomphonema parvulum dominated at site-SH, whereas Amphora pediculus and Grunowia solgensis were abundant at site-SU. This study provides evidence that the use of benthic diatoms is a useful tool for evaluating the ecological potential of artificial ecosystems. Moreover, such systems in the close vicinity of urban areas need better management and monitoring to reduce the risk of deterioration.

Understanding divergences between ecological status classification systems based on diatoms

A large number of diatom-based classification systems have been developed worldwide in recent years. These new systems, together with the oldest, emerged on the need to assess the water quality of rivers, but knowledge on possible divergences resulting from their simultaneous application within a territory is limited. This study aimed to compare the ecological status classification provided by conceptually different methodological approaches, of use or potential use within the same region. 402 monitoring samples were collected from Atlantic siliceous streams (NW-Iberian Peninsula) and temporary Mediterranean streams (Balearic Islands, Spain). Two multimetric indices specifically developed for these areas (MDIAT and DIATMIB, respectively) were calculated, as well as the Specific Polluosensitivity Index (IPS). Multimetric indices were more sensitive methods at diagnosing degradation than IPS since they took directly account of abundance (i.e. chlorophyll a in DIATMIB) or indirectly by its proved inverse relationship with Chl a (MDIAT), together with their use of the regional reference diatom community. Alteration gradients were identified in both studied regions based on the distribution of diatoms, with the first axis of distance-based redundancy analyses (dbRDA) being related to nutrient enrichment and organic loads. Threshold Indicator Taxa ANalysis (TITAN) performed on diatoms sampled along environmental (dbRDA axis 1 and phosphate) and biological gradients (as Ecological Quality Ratio scores of classifications), pointed to lower than current Good/Moderate boundaries for phosphate maximum values (e.g. 22.5 and 71.6 μg L−1 for Galicia and the Balearic Islands, respectively) as well as for higher Good/Moderate boundaries for the MDIAT and IPS classifications. A ‘transition group’ of species was classified as sensitive or as tolerant depending on the regional nutrients range. Findings of the present study highlight the need to perform auto-ecological studies to increase the knowledge on regional diatoms and their optimal survival ranges across regions prior to adopt other non-regional diatom indices.

Responses of the Benthic Diatom Community to Nutrients and the Identification of Nutrient Thresholds in Three Aquatic Ecoregions of the Huntai River, Northeast China

Benthic diatom communities and nutrient gradients were investigated from 287 sampling sites in three aquatic ecoregions (AE) of the Huntai River to characterize the spatial distribution of nutrients and the benthic diatom communities. Locally weighted scatterplot smoothing (LOWESS) was used to analyze the thresholds for nitrogen and phosphorus. The results showed that:① The concentration of ammonia nitrogen and total phosphorus significantly differs in the three AEs, and shows a tendency of AEⅠ < AE Ⅱ < AE Ⅲ. ② To reveal the structure of benthic diatom communities, various benthic diatom indexes, including the sportive diatom percentage, sensitive diatom percentage, stipitate diatom percentage, Pielou evenness index, specific polluosensitivity index (IPS), biological diatom index (IBD), and generic diatom index (IDG) were analyzed. The sportive diatom percentage varied significantly in AEI, AEⅡ, and AE Ⅲ, with the highest percentage observed in AEⅢ and the lowest in AEⅠ. However, the other six indexes exhibited an opposite trend. All revealed that AE Ⅲ has been seriously damaged, while AEⅠ is less disturbed than AEⅡ and AE Ⅲ. ③ LOWESS fitting curves show thresholds for ammonia nitrogen (NH4+ -N) in the three aquatic ecoregions as 0.13, 0.30, and 1.98 mg·L-1, respectively and for total phosphorous (TP) were 0.04, 0.06, and 0.20 mg·L-1. All results were tested by independent-sample T tests. This study will provide assistance for effectively protecting the benthic diatom community in different aquatic ecoregions and also provide a theoretical basis for water management.

Assessing ecological status with diatoms DNA metabarcoding: Scaling-up on a WFD monitoring network (Mayotte island, France)

Diatoms are excellent ecological indicators of water quality because they are broadly distributed, they show high species diversity and they respond rapidly to human pressures. In Europe, the Water Framework Directive (WFD) gives the legal basis for the use of this indicator for water quality assessment and its management. Several quality indices, like the Specific Polluosensitivity Index (SPI), were developed to assess the ecological quality status of rivers based on diatom communities. It is based on morphological identifications and count of diatom species present in natural biofilms using a microscope. This methodology requires high taxonomic skills and several hours of analysis per sample as 400 individuals must be identified to species level. Since several years, a molecular approach based on DNA metabarcoding combined to High-Throughput Sequencing (HTS) is developed to characterize species assemblages in environmental samples which is potentially faster and cheaper. The ability of this approach to provide reliable diatom inventories has been demonstrated and its application to water quality assessment is currently being improved. Despite optimization of the DNA metabarcoding process with diatoms, few studies had yet extended it at the scale of a freshwater monitoring network and evaluated the reliability of its quality assessment compared to the classical morphological approach.In the present study we applied DNA metabarcoding to the river monitoring network of the tropical Island Mayotte. This island is a French département since 2011 and the WFD has to be applied. This offered the opportunity to scale up the comparison of molecular and morphological approaches and their ability to produce comparable community inventories and water quality assessments. Benthic diatoms were sampled following WFD standards in 45 river sites in 2014 and 2015 (80 samples). All samples were submitted in parallel to the molecular and the morphological approaches. DNA metabarcoding was carried out using Genelute DNA extraction method, rbcL DNA barcode and PGM sequencing, while microscopic counts were carried out for the classical methodology. Diatom community structures in terms of molecular (OTUs) and of morphological (species) were significantly correlated. However, only 13% of the species was shared by both approaches, with qualitative and quantitative variation due to i) the incompleteness of the reference library (82% of morphological species are not represented in the database), ii) limits in taxonomic knowledge and iii) biases in the estimation of relative abundances linked to diatom cell biovolume. However, ecological quality status assessed with the molecular and morphological SPI values were congruent, and little affected by sequencing depth. DNA metabarcoding of diatom communities allowed a reliable estimation of the quality status for most of the rivers at the scale of the full biomonitoring network of Mayotte Island.

Impact of Drought on Diatom Communities and the Consequences for the use of Diatom Index Values in the River Maureillas (Pyrénées‐Orientales, France)

AbstractIn the context of climate change, with an increase in the incidence of seasonally dry streambeds, we examined the impact of drought, and its duration, on the structure of diatom communities and the consequences of this on diatom index values (Biological Diatom Index; Specific Polluosensitivity Index) in the Mediterranean river Maureillas. The effects of several durations of droughts or low flow events (1, 2 and 4 weeks) on diatom‐dominated biofilms were studied by removing pebbles from the river and comparing the growing communities after rewetting with those of control biofilms (pebbles left in the streambed) and with those of newly formed biofilms (cleaned‐up pebbles). Sampling was performed six times during the 28‐day rewetting phase. We determined the specific composition of diatom communities, the mortality rate after rewetting, and the Biological Diatom Index and Specific Polluosensitivity Index scores for each sample, distinguishing between potentially living diatoms (full frustules with chloroplasts) and dead diatoms (empty frustules). Our results showed a notable and durable impact of even short drought on diatom communities. The different trajectories of biofilm communities observed between the types of pebbles showed the importance of the biofilm history for the community structure. This should be taken into account to improve the sampling practices used for monitoring. We also observed a stable mortality rate whatever the drought duration was. Diatom index values were stable despite the observed impact of treatment on diatom communities. These results emphasize the reliability of the diatom index to evaluate biological quality according to European requirements. In addition, the distinction of empty and full frustules did not induce significant differences, but it may reduce biases in interpretation. Copyright © 2014 John Wiley &amp; Sons, Ltd.

Effect of riparian vegetation on diatom assemblages in headwater streams under different land uses

Differences in the structure of diatom assemblages in headwaters with contrasting shading conditions and different land use in the buffer zone and upper catchment were studied in order to evaluate the influence of the lack of riparian vegetation on the biofilm. The objective was to ascertain whether a riparian buffer can mitigate the negative influence of human induced disturbance and pollution on diatom assemblages in headwaters. Four streams were selected in order to maximize the differences in the land cover and minimize other environmental gradients. Multivariate statistics, different comparative and permutation tests and correlations were applied to compare the diatom assemblages, the Specific Polluosensitivity Index (IPS) and the diatom ecological guilds (low profile, high profile and motile) among the sites studied and to evaluate their responses to disturbances. The analysis showed that low profile diatoms typically dominated in forested headwaters with limited resources, whilst assemblages at impacted sites showed a wider range of growth forms. In unimpacted streams, the diatom assemblages were influenced by temperature, pH, conductivity and calcium, as usually reported for oligotrophic streams with high natural disturbance due to fast current and shading. In both shaded and unshaded impacted streams, the importance of nutrients and land use disturbance, especially urbanization, prevailed. This trend was also reflected by the IPS index that showed consistently lower values at impacted sites, correlating most significantly with nutrients. The diatom species composition as well as diatom guilds at impacted sites were similar, regardless of the presence or absence of riparian vegetation, and were significantly influenced by seasonal changes.Our results indicate that diatoms react sensitively to alterations of the water environment in headwaters, induced by anthropogenic activities, and these impacts are not buffered by an intact riparian zone. Diatoms closely reflected land use practices in the upper catchment regardless of the buffer zone status.

Application of diatom biotic indices in the Guadalquivir River Basin, a Mediterranean basin. Which one is the most appropriated?

The diatom community was studied in 110 sites within the Guadalquivir River catchment area, South Spain, in order to test the applicability of diatom biotic indices developed in other European regions to this site and to provide a useful tool for monitoring water quality in the river basin. We identified 399 taxa and calculated five diatomic indices (Specific Polluosensitivity Index (IPS), Biological Diatom Index, Trophic Diatom Index, Index of the European Economic Community, and Diatom-based Eutrophication Pollution Index (EPI-D)). Since the indices analyzed were highly correlated, their results could be compared. The indices that gave the best results were the EPI-D followed by the IPS, the latter being the most widely used index in Iberian catchments. Nevertheless, the EPI-D presented certain advantages: (1) this index correlated the best with the water chemistry in the catchment area; (2) EPI-D is not sensitive to the presence of taxa belonging to the Achnanthidium minutissimum complex frequently present in the Guadalquivir basin. Nevertheless, EPI-D retains its effectiveness and thus constitutes an easier index for application from a taxonomical standpoint. We estimated the general water quality of the entire basin on the basis of EPI-D. According to these results, 55% of the sites had either high or good water quality. The species that better characterized each water quality category in the study area were: A. minutissimum (high and good), Amphora pediculus (moderate), Nitzschia frustulum (poor), and Nitzschia capitellata (bad).

Use of biological indicators to assess water quality of the Ul River (Portugal)

Diatoms and macroinvertebrates have been extensively used as water quality indicators in Europe for the last two decades. In Portugal, the use of biological indicators to assess water quality in rivers has increased greatly. The aim of this work was to assess the water quality and ecological status of the Ul River in order to evaluate its ability for the establishment of a fluvial beach, using periphytic diatoms and macroinvertebrates as indicators. Four sites were selected along the Ul River. At each site, biological, physical, and chemical parameters were investigated. Epilithic diatoms and macroinvertebrates were sampled. The Specific Polluosensitivity Index and the Biological Diatom Index were applied to diatom data, while for macroinvertebrates, the Iberian Biological Monitoring Working Party (IBMWP) was used. According to the results obtained, it was possible to conclude that up to now, this river does not possess the ideal conditions for the establishment of a fluvial beach. We concluded that epilithic diatoms and macroinvertebrates provided consistent information on water quality assessment and can be used as biological indicators of the water quality in Ul River.

A comparison between biotic indices and predictive models in stream water quality assessment based on benthic diatom communities

Diatoms are widely used in stream bioassessment due to their broad distribution, extraordinary variability and the ability to integrate changes in water quality. The indices Specific Polluosensitivity Index (SPI), standardized Biological Diatom Index (BDI), European Economic Community Index (CEC) and Generic Diatom Index (GDI), originally developed in France, are often applied in Portugal to evaluate stream ecological quality based on diatom communities. Alternatively, predictive models resulting from the comparison between the communities of the study site and those of a set of reference sites representing undisturbed or the best available conditions of a given region have been proposed as valuable methods for evaluating the ecological status of streams. In the present study, we applied the four above-mentioned widely used diatom-based indices (SPI, BDI, CEC and GDI) and a predictive model (MoDi) to 54 sites located in central Portugal to assess the sensitivity of the five methods to a range of anthropogenic disturbances cumulatively affecting streams and represented by 27 variables (e.g., organic enrichment, changes in morphology of the channels, integrity of the riparian corridor, land use in the catchment). The results were analyzed comparatively through Spearman correlations, Boxplots and Stepwise Discriminant Analysis. This study confirmed the sensitivity of diatoms to organic and nutrient contamination (showed by the MoDi, BDI, CEC and SPI) and revealed the importance of suspended solids (through the MoDi, GDI, and SPI). The relevance of modifications in land use to diatoms was shown by all methods applied, except for the GDI. The MoDI also revealed the importance of changes in the structure and morphology of the reach and the channel, like the construction of artificial walls or embankments and connectivity; the BDI also related its assessments with the riparian zone integrity; and the SPI was not useful in detecting morphological pressures. The GDI produced the most divergent assessments and was less effective in revealing the anthropogenic disturbances. The use of the predictive model (MoDi) is therefore a good method for the assessment of streams in central Portugal because it expresses a great diversity of quantitative and qualitative changes in freshwater systems reflected in the structure (species richness and abundance) of diatom communities.

Diatom Communities and Water Quality Assessment in Mountain Rivers of the Upper Segre Basin (La Cerdanya, Oriental Pyrenees)

Epilithic diatoms of mountain rivers from the upper Segre catchment (Oriental Pyrenees) were studied in 1998, during three different seasons: March, July and September. Four rivers, the river Segre and its three most important tributaries, Duran, Molina and Querol, were sampled in upstream and downstream stretches. The diatom communities were comparable in all upstream stretches of these mountain rivers draining siliceous substrates. Dominant taxa were Achnanthidium subatomus, Diatoma mesodon, Encyonema cf. minutum, E. silesiacum, Fragilaria arcus, F. capucina, Gomphonema calcifugum, G. pumilum, Meridion circulare and Nitzschia pura. Changes in water quality in the downstream stretches lead to the appearance of pollution tolerant taxa, such as Eolimna minima, Gomphoneis minuta, Navicula gregaria, and Nitzschia inconspicua. As a result, the values obtained with the diatom water quality indices (IPS Specific Polluosensitivity Index, CEE and IBD Biological Diatom Index) decreased. The diatom community composition and the derived water quality values did not change in the upstream stretches over the year. In contrast, significant changes were observed in the downstream stretches with best water␣quality in July, during high flows due to melting snow, and worst values in September, during low␣discharge. The diatom indices, especially the IPS, showed a good performance in these mountain rivers.

Utilisation des indices diatomiques pour la mesure de la qualité des eaux du bassin Artois-Picardie : bilan et perspectives

Because of the special nature of the streams in its basin, the Water Agency of Artois-Picard ie adopted in 1988 the diatoms and diatom indices for the assessment of water quality. Six indices have been tested. The results show that the IPS or Specific Polluosensitivity Index (Coste in CEMAGREF 1982) is the only on provide a realistic assessment of water quality. This index integrates three variables : organic pollution expressed as DB0 5, DCO or ammonium, salinity expressed as conductivity and chlorides, and eutrophication which, when actual, is expressed as chlorophyll levels and, when potential, is expressed as phosphate levels. These three variables are the principal factors responsible for variations in water quality within the basin. In addition, the physicochemical conditions were markedly stable throughout the year and therefore the diatom indices calculated in September are representative of summer water quality. Different simula­ tions have been made, using the program OMNIDIA, in order to determine a practical index that can be used routinely. The results show that the best approach is to work at the generic level with occasional determinations to species.
