Novel Pickering bigels stabilized by whey protein microgels: Interfacial properties, oral sensation and gastrointestinal digestive profiles

In the ongoing quest to formulate sensory-rich, low-fat products that maintain structural integrity, this work investigated the potential of bigels, especially those created using innovative Pickering techniques. By harnessing the unique properties of whey protein isolate (WPI) and whey protein microgel (WPM) as interfacial stabilizers, WPM-based Pickering bigels exhibited a remarkable particle localization at the interface due to specific intermolecular interactions. The rise in protein concentration not only intensified particle coverage and interface stabilization but also amplified attributes like storage modulus, yield stress, and adhesiveness, owing to enhanced intermolecular forces and a compact gel matrix. Impressively, WPM-based Pickering bigels outshone in practical applications, showcasing exceptional oil retention during freeze–thaw cycles and extended flavor release-a promising indication for frozen food product applications. Furthermore, these bigels underwent a sensory evolution from a lubricious texture at lower concentrations to a stable plateau at higher ones, offering an enriched consumer experience. In a comparative digestibility assessment, WPM-based Pickering bigels demonstrated superior prowess in decelerating the release of free fatty acids, indicating slowed lipid digestion. This study demonstrates the potential to fine-tune oral sensations and digestive profiles in bigels by modulating Pickering particle concentrations.

Multiscale Simulations of Self-Assembling Peptides: Surface and Core Hydrophobicity Determine Fibril Stability and Amyloid Aggregation.

Assemblies of peptides and proteins through specific intermolecular interactions set the basis for macroscopic materials found in nature. Peptides provide easily tunable hydrogen-bonding interactions, which can lead to the formation of ordered structures such as highly stable β-sheets that can form amyloid-like supramolecular peptide nanofibrils (PNFs). PNFs are of special interest, as they could be considered as mimics of various fibrillar structures found in nature. In their ability to serve as supramolecular scaffolds, they could mimic certain features of the extracellular matrix to provide stability, interact with pathogens such as virions, and transduce signals between the outside and inside of cells. Many PNFs have been reported that reveal rich bioactivities. PNFs supporting neuronal cell growth or lentiviral gene transduction have been studied systematically, and their material properties were correlated to bioactivities. However, the impact of the structure of PNFs, their dynamics, and stabilities on their unique functions is still elusive. Herein, we provide a microscopic view of the self-assembled PNFs to unravel how the amino acid sequence of self-assembling peptides affects their secondary structure and dynamic properties of the peptides within supramolecular fibrils. Based on sequence truncation, amino acid substitution, and sequence reordering, we demonstrate that peptide-peptide aggregation propensity is critical to form bioactive β-sheet-rich structures. In contrast to previous studies, a very high peptide aggregation propensity reduces bioactivity due to intermolecular misalignment and instabilities that emerge when fibrils are in close proximity to other fibrils in solution. Our multiscale simulation approach correlates changes in biological activity back to single amino acid modifications. Understanding these relationships could lead to future material discoveries where the molecular sequence predictably determines the macroscopic properties and biological activity. In addition, our studies may provide new insights into naturally occurring amyloid fibrils in neurodegenerative diseases.

Liquid–liquid phase separation of the prion protein is regulated by the octarepeat domain independently of histidines and copper

Liquid-liquid phase separation (LLPS) of the mammalian prion protein is mainly driven by its intrinsically disordered N-terminal domain (N-PrP). However, the specific intermolecular interactions that promote LLPS remain largely unknown. Here, we used extensive mutagenesis and comparative analyses of evolutionarily distant PrP species to gain insight into the relationship between protein sequence and phase behavior. LLPS of mouse PrP is dependent on two polybasic motifs in N-PrP that are conserved in all tetrapods. A unique feature of mammalian N-PrP is the octarepeat domain with four histidines that mediate binding to copper ions. We now show that the octarepeat is critical for promoting LLPS and preventing formation of PrP aggregates. Amphibian N-PrP, which contains the polybasic motifs but lacks a repeat domain and histidines, does not undergo LLPS and forms non-dynamic protein assemblies indicative of aggregates. Insertion of the mouse octarepeat domain restored LLPS of amphibian N-PrP, supporting its essential role in regulating phase transition of PrP. This activity of the octarepeat domain was neither dependent on the four highly conserved histidines nor on copper binding. Instead, the regularly spaced tryptophan residues were critical for regulating LLPS, presumably via cation-π interactions with the polybasic motifs. Our study reveals a novel role for the tryptophan residues in the octarepeat in controlling phase transition of PrP and indicates that the ability of mammalian PrP to undergo LLPS has evolved with the octarepeat in the intrinsically disordered domain, but independently of the histidines.

Ru(II)‐Nitrophenylhydrazine/Chlorophenylhydrazine Complexes: Nanoarchitectonics, Biological Evaluation and In silico Study

AbstractRu(II)‐arene compounds are being investigated as anticancer agents due to the biocompatibility of ruthenium and their structural diversity. Two newly synthesized Ru(II) complexes, [RuCl(η6‐p‐cymene)(3‐DNPH)] (chlorido(η6‐p‐cymene)(3‐nitrophenylhydrazine‐k2N,N′)ruthenium(II)) (1) and [RuCl(η6‐p‐cymene)(3‐CNPH)] (chlorido(3‐chlorophenylhydrazine‐k2N,N′)(η6‐p‐cymene)ruthenium(II)) (2), are experimentally (IR, NMR) and theoretically (B3LYP/6‐31+G(d,p)(H,C,N,Cl)/LanL2DZ(Ru)) characterized. Experimental and theoretical values of 1H and 13C chemical shifts and position of the most intense vibrational bands showed high correlation coefficients and low mean absolute errors, proving the predicted structure and applicability of the selected level of theory. Cell viability studies performed on MDA‐MB‐468, BT‐474, and PC3 cells using MTT and CV assay indicated the activity of the second complex similar to the activity of cisplatin towards BT‐474 breast cancer cells. The spectrofluorimetric measurements of Bovine Serum Albumin showed the binding process‘s spontaneity of complexes and protein, with a binding energy of around −30 kJ mol−1. Detailed molecular docking analysis allowed the elucidation of the binding mechanism through specific intermolecular interactions. Both compounds showed a higher affinity towards BSA than naproxen and cisplatin. Molecular docking simulations proved the spontaneity of the complexes binding to DNA. Based on these promising results, further biological examinations of these compounds are advised.

Design and synthesis of nucleotidyl lipids and their application in the targeted delivery of siG12D for pancreatic cancer therapy

Nucleic acid drugs are attracting significant attention as prospective therapeutics. However, their efficacy is hindered by challenges in penetrating cell membranes and reaching target tissues, limiting their applications. Nucleotidyl lipids, with their specific intermolecular interactions such as H-bonding and π-π stacking, offer a promising solution as gene delivery vehicles. In this study, a novel series of nucleotide-based amphiphiles were synthesized. These lipid molecules possess the ability to self-assemble into spherical vesicles of appropriate size and zeta potential in aqueous solution. Furthermore, their complexes with oligonucleotides demonstrated favorable biocompatibility and exhibited antiproliferative effects against a broad range of cancer cells. Additionally, when combined with the cationic lipid CLD, these complexes displayed promising in vitro performance and in vivo efficacy. By incorporating DSPE-PEGylated cRGD into the formulation, targeted accumulation of siG12D in pancreatic cancer cells increased from approximately 6% to 18%, leading to effective treatment outcomes (intravenous administration, 1 mg/kg). This finding holds significant importance for the liposomal delivery of nucleic acid drugs to extrahepatic tissues.

Refractive index and related properties for the mixture (1, 4-dioxane + water) at temperature T= (295.15, 298.15, 301.15, 304.15, 307.15, 310.15 and 313.15) K

ABSTRACT The refractive index n D for the mixture (1,4-dioxane (1) + water (2)) was measured over the whole composition range at different temperatures and atmospheric pressure. From measured values of the refractive indices, the molar refraction R m , reduced molar free volumes V m R m , molecular radii r and internal pressure, P int were calculated. On the other hand, some excess parameters were also deduced. The obtained results have been discussed and interpreted in terms of the type and nature of specific intermolecular interactions existed between the components. Furthermore, we have used nine mixing rules for refractive index proposed by different authors to calculate the refractive indices of the present mixture. A comparative study was carried out between the measured and calculated indices. The Redlich–Kister polynomial equation has been used to fit the presented excess parameters to derive the adjustable fitting coefficients and estimate the standard error values.

Atomistic, macromolecular model of the Populus secondary cell wall informed by solid-state NMR.

Plant secondary cell walls (SCWs) are composed of a heterogeneous interplay of three major biopolymers: cellulose, hemicelluloses, and lignin. Details regarding specific intermolecular interactions and higher-order architecture of the SCW superstructure remain ambiguous. Here, we use solid-state nuclear magnetic resonance (ssNMR) measurements to infer refined details about the structural configuration, intermolecular interactions, and relative proximity of all three major biopolymers within air-dried Populus wood. To enhance the utility of these findings and enable evaluation of hypotheses in a physics-based environment in silico, the NMR observables are articulated into an atomistic, macromolecular model for biopolymer assemblies within the plant SCW. Through molecular dynamics simulation, we quantitatively evaluate several variations of atomistic models to determine structural details that are corroborated by ssNMR measurements.

Investigations on the thermo physical properties of the binary mixtures of 2-methyl-pentanediol-2,4 with n-alkanols at various temperatures (hexylene glycol with 1-octanol, 1-nonanol, 1-decanol)

ABSTRACT Densities (ρ), speeds of sound (u) and viscosities (η) for three binary mixtures of hexylene glycol (2-methyl-2,4-pentanediol or MPD) with 1-octanol (D1), 1-nonanol (D2) and 1-decanol (D3) are calculated at temperatures ranging from 303.15 to 313.15 K and atmospheric pressure over the entire composition range. Experimental measurements are used to calculate the excess molar volume, excess isentropic compressibility, deviation in viscosity and excess Gibbs-free energy of activation of viscous flow. Partial molar properties and infinite dilution molar partial properties are also calculated for each binary system. The results obtained can be discussed in terms of weaker interactions and dispersion forces with the absence of specific intermolecular interactions in binary mixtures. Finally, the Prigogine-Flory-Patterson (PFP) theory is applied to identify the most predominant molecular interaction. Jouyban-Acree model results are discussed in terms of mean relative deviation (MRDs) and individual relative deviation (IRD) between calculated and experimental densities, speeds of sound and viscosities as an accuracy criterion.

Exploring the mechanism of asymmetric growth of organic polar crystals: Inherent properties, specific intermolecular interactions

Organic polar crystals (OPCs) are the foundation for photonic and non-linear optical applications in advanced materials. High-quality crystals are a key in achieving efficient performance for OPCs. This work explores the properties and mechanisms of OPC growth along the polar axis. Melt microdrop growth experiments confirm that asymmetric growth is an inherent property of OPCs and relates to the intrinsic crystal structure. Notably, the solvent also plays a non-negligible role in OPCs growth. Solvent-crystal face interactions are strongly influenced by the crystal face structure. A rough surface has a slower growth rate, and a specific recognition of the solvent molecules. Asymmetric growth correlates with the modified attachment energy model and reveals the role of the solvent in inhibiting of crystal growth. Finally, the growth pattern in different solvents is explained by the interfacial adsorption model. This work will further enrich the asymmetric growth mechanism of OPCs and provide guidance for obtaining high-quality OPCs in solution.

Experimental analysis of volumetric, acoustic, viscometric and optical properties of water with 1-amino-2-propanol, 1,2-diaminopropane and 1,3-diaminopropane at T=298.15 -318.15 K: Molecular Modelling by Graph, PFP and IR spectroscopy investigations

BackgroundLiquid mixtures are used as a solvent in most of the natural systems and chemical processes. Therefore, the physicochemical properties of liquid mixtures at different compositions and at different temperatures have a significant role in the optimization of chemical processes. The present report involves the analysis of the thermophysical properties of water (1) and amines (2). MethodsDensity, ρ, dynamic viscosity, η, refractive index, nD, and speed of sound, u, data were measured for binary mixtures of water (1) and amines (1A2P (1-amino-2-propanol), 1,2-DAP (1,2-diaminopropane) and 1,3-DAP (1,3-diaminopropane) (2) at five different temperatures (T = 298.15 to 318.15 K) and at 0.1 MPa pressure. Based on experimental data, the excess molar volume, VmE, deviation in dynamic viscosity, Δη, excess Gibbs free energy of activation, G*E, deviation in refractive index, ΔnD, excess ultrasonic speed, uE, excess isentropic compressibility, KsE, deviation in free volume, ΔVf, excess available volume, VaE, and excess intermolecular free length, LfE, were calculated and these evaluated properties were fitted to Redlich-Kister polynomial (R.K.). The VmE data was analyzed by Prigogine-Flory-Patterson (PFP) theory. The dynamic viscosity, η, values were studied by different correlations. The VmE and Δη data were also analyzed by Graph theoretical approach (GTA). Raman and FT-IR spectroscopic studies give important information about the intermolecular interactions present between unlike molecules water (1) and amines (2) which is also recognized by GTA. Significant findingsThe value of VmE and KsE values were found negative over the whole composition range, and it follow the sequence as: 1,2-DAP > 1,3-DAP > 1A2P with water at equimolar composition. GTA and PFP well supported the VmE data. GTA studies help in investigating the specific intermolecular interactions between unlike components. Further, Graph theory studies predicted OH—–NH interaction is dominant interaction among all interactions which were also supported by Raman and FTIR spectroscopic studies.

Atomistic-Level Effects of Noncovalent Interactions and Crystalline Packing for Organic Material Structural Integrity upon Exposure to Gamma Radiation.

Developing an atomistic understanding of ionizing radiation induced changes to organic materials is necessary for intentional design of greener and more sustainable materials for radiation shielding and detection. Cocrystals are promising for these purposes, but a detailed understanding of how the specific intermolecular interactions within the lattice upon exposure to radiation affect the structural stability of the organic crystalline material is unknown. This study evaluates atomistic-level effects of γ radiation on both single- and multicomponent organic crystalline materials and how specific noncovalent interactions and packing within the crystalline lattice enhance structural stability. Dose studies were performed on all crystalline systems and evaluated via experimental and computational methods. Changes in crystallinity were evaluated by p-XRD and free radical formation was analyzed via EPR spectroscopy. Type of intermolecular interactions and packing within the crystal lattice was delineated and related to the specific free radical species formed and the structural integrity of each material. Periodic DFT and HOMO-LUMO surface mapping calculations provided atomistic-level identifications of the most probable sites for the radicals formed upon exposure to γ radiation and relate intermolecular interactions and molecular packing within the crystalline lattice to experimental results.

SASH1: A Novel Eph Receptor Partner and Insights into SAM-SAM Interactions

The Eph (erythropoietin-producing human hepatocellular) receptor family, the largest subclass of receptor tyrosine kinases (RTKs), plays essential roles in embryonic development and neurogenesis. The intracellular Sterile Alpha Motif (SAM) domain presents a critical structural feature that distinguishes Eph receptors from other RTKs and participates in recruiting and binding downstream molecules. This study identified SASH1 (SAM and SH3 domain containing 1) as a novel Eph receptor-binding partner through SAM-SAM domain interactions. Our comprehensive biochemical analyses revealed that SASH1 selectively interacts with Eph receptors via its SAM1 domain, displaying the highest affinity for EphA8. The high-resolution crystal structure of the EphA8-SASH1 complex provided insights into the specific intermolecular interactions between these proteins. Cellular assays confirmed that EphA8 and SASH1 co-localize and co-precipitate in mammalian cells, with cancer mutations (EphA8 R942H or G978D) impairing this interaction. We demonstrated that SAM-SAM interaction is critical for SASH1-mediated regulation of EphA8 kinase activity, shedding new light on the Eph signaling pathway and expanding our understanding of the molecular basis of the tumor suppressor gene SASH1.

Competition between β-Sheet and Coacervate Domains Yields Diverse Morphologies in Mixtures of Oppositely Charged Homochiral Polypeptides.

Biomolecular assembly processes involving competition between specific intermolecular interactions and thermodynamic phase instability have been implicated in a number of pathological states and technological applications of biomaterials. As a model for such processes, aqueous mixtures of oppositely charged homochiral polypeptides such as poly-l-lysine and poly-l-glutamic acid have been reported to form either β-sheet-rich solid-like precipitates or liquid-like coacervate droplets depending on competing hydrogen bonding interactions. Herein, we report studies of polypeptide mixtures that reveal unexpectedly diverse morphologies ranging from partially coalescing and aggregated droplets to bulk precipitates, as well as a previously unreported re-entrant liquid-liquid phase separation at high polypeptide concentration and ionic strength. Combining our experimental results with all-atom molecular dynamics simulations of folded polypeptide complexes reveals a concentration dependence of β-sheet-rich secondary structure, whose relative composition correlates with the observed macroscale morphologies of the mixtures. These results elucidate a crucial balance of interactions that are important for controlling morphology during coacervation in these and potentially similar biologically relevant systems.

Highly Luminescent Lanthanide Metal‐Organic Frameworks with Tunable Color for Nanomolar Detection of Iron(III), Ofloxacin and Gossypol and Anti‐counterfeiting Applications

AbstractSolvothermal reaction of 5,5′‐(pyridine‐2,6‐diylbis(oxy))diisophthalic acid (H4L) with europium(III) or terbium(III) nitrates in acetonitrile‐water (1 : 1) at 120 °C gave rise to isostructural 2D coordination polymers, [Ln(HL)(H2O)3]∞ (NIIC‐1‐Eu and NIIC‐1‐Tb), the layers of which are composed by eight‐coordinated lanthanide(III) ions interconnected by triply deprotonated ligands HL3−. The layers are packed in the crystal without any specific intermolecular interactions between them, allowing the facile preparation of stable water suspensions, in which NIIC‐1‐Tb exhibited top‐performing sensing properties through luminescence quenching effect with exceptionally low detection limits towards Fe3+ (LOD 8.62 nM), ofloxacin (OFX) antibiotic (LOD 3.91 nM) and cotton phytotoxicant gossypol (LOD 2.27 nM). In addition to low detection limit and high selectivity, NIIC‐1‐Tb features fast sensing response (within 60–90 seconds), making it superior to other MOF‐based sensors for metal cations and organic toxicants. The photoluminescence quantum yield of NIIC‐1‐Tb was 93 %, one of the highest among lanthanide MOFs. Mixed‐metal coordination polymers NIIC‐1‐EuxTb1−x demonstrated efficient photoluminescence, the color of which could be modulated by the excitation wavelength and time delay for emission monitoring (within 1 millisecond). Furthermore, an original 2D QR‐coding scheme was designed for anti‐counterfeiting labeling of goods based on unique and tunable emission spectra of NIIC‐1‐Ln coordination polymers.

A novel interface between the N-terminal and coiled-coil domain of STAT1 functions in an auto-inhibitory manner

BackgroundSTAT1 is an intracellular signaling molecule that is crucially involved in the regulation of the innate immune system by activation of defense mechanisms against microbial pathogens. Phosphorylation-dependent activation of the STAT1 transcription factor is associated with a conversion from an antiparallel to parallel dimer configuration, which after nuclear import binds to DNA. However, not much is known about the specific intermolecular interactions that stabilize unphosphorylated, antiparallel STAT1 complexes prior to activation.ResultsIn this study, we identified a previously unknown interdimeric interaction site, which is involved in the termination of STAT1 signaling. Introduction of the glutamic acid-to-alanine point mutation E169A in the coiled-coil domain (CCD) by site-directed mutagenesis led to increased tyrosine phosphorylation as well as accelerated and prolonged nuclear accumulation in transiently transfected cells. In addition, DNA-binding affinity and transcriptional activity were strongly enhanced in the substitution mutant compared to the wild-type (WT) protein. Effects of Dispersed Carbon Nanotubes and Emerging Supramolecular Structures on Phase Transitions in Liquid Crystals: Physico-Chemical Aspects

The current state of the study of different liquid crystalline (LC) systems doped with carbon nanotubes (CNTs) is discussed. An attempt is endeavored to outline the state-of-the-art technology that has emerged after two past decades. Systematization and analysis are presented for the integration of single- and multi-walled carbon nanotubes in thermotropic (nematic, smectic, cholesteric, ferroelectric, etc.) and lyotropic LCs. Special attention is paid to the effects of alignment and supramolecular organization resulting from orientational coupling between CNTs and the LC matrix. The effects of the specific inter-molecular and inter-particle interactions and intriguing microstructural, electromagnetic, percolation, optical, and electro-optical properties are also discussed.

Isoxazolyl-Derived 1,4-Dihydroazolo[5,1-c][1,2,4]Triazines: Synthesis and Photochemical Properties

New fluorescent dyes containing an assembled 1,4-dihydroazolo[5,1-c][1,2,4]triazine (DAT) core and an isoxazole ring were synthesized through a reaction between diazopyrazole or diazoimidazoles and isoxazolyl-derived enamines in mild conditions. The photophysical characteristics (maxima absorption and emission, Stokes shifts, fluorescent quantum yields, and fluorescence lifetimes) of the new fluorophores were obtained. The prepared DATs demonstrated emission maxima ranging within 433-487 nm, quantum yields within 6.1-33.3%, and a large Stokes shift. The photophysical characteristics of representative DAT examples were studied in ten different solvents. Specific (hydrogen bonds) and non-specific (dipole-dipole) intermolecular and intramolecular interactions were analyzed using XRD data and spectral experiments. Solvatochromism was analyzed using Lippert-Mataga and Dimroth-Reichardt plots, revealing the relationship between the DAT structure and the nature of solute-solvent interactions. The significant advantages of DATs are the fluorescence of their powders (QY up to 98.7%). DAT-NMe210 expressed bright aggregation-induced emission (AIE) behavior in DMSO and THF as the water content increased. The numerous possible variations of the structures of the heterocycles included in the DATs, as well as substituents, create excellent prospects for adjusting their photophysical and physicochemical properties.

Cage-Like Sodalite-Type Porous Organic Salts Enabling Luminescent Molecule's Incorporation and Room-temperature Phosphorescence Induction in Air.

Expression of room-temperature phosphorescence (RTP) in organic materials requires complicated molecular design and specific intermolecular interactions, and therefore types of RTP materials are restricted. This work presents cage-like sodalite-type porous organic salts (s-POSs) as host materials for luminescent molecules to induce RTP, using tetrasulfonic acid with an adamantane core and triphenylmethylamines that are modified with substituents in the para-positions of benzene rings (TPMA-X). By adding a representative luminescent molecule (pyrene) to a reaction solution during construction of s-POSs, the molecule is incorporated in a facile manner. s-POSs with a heavy halogen atom (X: Iodine) on the pore surface give heavy atom effects, suppression of thermal vibration, and protection from oxygen, for the incorporated molecule, which induce its RTP even in air. This strategy can be applied to various luminescent molecules, which may lead to the achievement of RTP of various colors.

Short aramid fiber reinforced thermotropic liquid crystalline polyester composites: Fabrication, thermal, and mechanical properties

We conducted a study to analyze the impact of short aramid fibers (AFs) on the melt-rheological behavior, thermal transition, thermal stability, and mechanical durability of thermotropic liquid crystal polyesters (TLCPs). By using different AF loading contents ranging from 3–15 wt%, we produced TLCP matrix composites through masterbatch-based melt-compounding and injection-molding. The SEM images and FT-IR spectra demonstrate that the AFs are dispersed in the TLCP matrix with a microfibrillar structure through good interfacial adhesion caused by specific intermolecular interactions between the TLCP and AFs. As a result, the complex viscosity, shear storage/loss moduli, and thermal transition (melt-crystallization, glass transition, and melting) temperatures of the composites increase with increasing AF filler content. However, the melt-crystallization and melting enthalpies increase only at low AF loading contents of 3–5 wt%. At high AF contents of 7–15wt%, the enthalpies decrease owing to the partial aggregation of AF fillers. The thermogravimetric analysis proves that the thermal stability of TLCP/AF composites improves when the AF filler is introduced. The dynamic mechanical analysis using the stepped isothermal method shows that the addition of 5 wt% AF to the TLCP leads to an approximately 150% improvement in elastic moduli and long-term mechanical durability at elevated temperatures.
