We report a rapid magnetic nanoparticle-mediated protocol to extract and detect DNA of E. coli O157:H7. Bare iron oxide nanoparticles (Fe3O4 NPs) were used as solid phase support to extract high purity DNA from concentrated and spiked water samples. Electrochemical detection of the DNA is achieved by employing polyaniline-functionalized magnetic nanoparticles (PANI-MNPs) which serve as DNA pre-concentrator and transducer. The DNA-based nanobiosensor shows good selectivity and sensitivity (detection limit of 0.10 ng/µL DNA) and offers faster and comparable results relative to conventional methods. This DNA extraction and detection method is expected to be applicable in food, water, and environmental samples.
