Simultaneous enrichment and speciation of lead and mercury by magnetic solid-phase extraction coupled to HPLC-ICP-MS based on magnetic hydrazine-linked covalent organic frameworks

BackgroundTrace levels of organic and inorganic lead and mercury species in the environment, including divalent lead (Pb2+), trimethyllead (TML), divalent mercury (Hg2+), monomethylmercury (MeHg), and ethylmercury (EtHg), are highly toxic to humans and ecology. It is of great importance for speciation of lead and mercury to evaluate the toxicity of lead and mercury and their biogeochemistry in the environment. However, simultaneous multi-elemental enrichment and speciation at trace level remains a challenge. There are few reports of simultaneous magnetic solid-phase extraction (MSPE) of organic and inorganic lead and mercury species at trace level in the real water. ResultsIn this work, a novel core-shell magnetic hydrazine-linked covalent organic frameworks (Fe3O4@COF-TCH) was prepared for the first time by grafting hydrazine-linked COFs on the Fe3O4 nanoparticles. Fe3O4@COF-TCH with abundant thione and imino groups has strong adsorption for lead and mercury species. Based on it, a simple and practical magnetic solid-phase extraction high-performance liquid chromatography-inductively coupled plasma mass spectrometry (MSPE-HPLC-ICP-MS) method was developed for extraction and determination of trace lead and mercury species, including Hg2+, MeHg, EtHg, Pb2+ and TML. The limits of detection (3δ) of the developed method were 0.08, 0.81, 0.90, 0.56 and 0.88 ng L−1 with the enrichment factors (EFs) of 384, 376, 379, 389 and 360-fold for Pb2+, TML, Hg2+, MeHg and EtHg, respectively. The high accuracy and reproducibility have been proved by the spiked recoveries (94.4–103 %) in real samples. SignificanceThe proposed method with simple operation and high sensitivity has been successfully applied to simultaneous speciation of lead and mercury at trace levels in the water samples with complicated matrices, including underground water, surface water, sea water. Meanwhile, it has the advantages of cost-saving, labor-saving and time-saving and is suitable for the investigation and risk assessment in water. The development of MSPE-HPLC-ICP-MS method provides ideas and guidance for the simultaneous multi-elemental enrichment and speciation.

Facile synthesis of Fe3O4@CS@ZIF-67 for the magnetic solid phase extraction-high performance liquid chromatographic determination of benzoylurea insecticides in tea infusions

In this paper, a novel magnetic metal–organic framework composite with chitosan as linker (Fe3O4@CS@ZIF-67) was synthesized by a facile method at normal pressure and normal temperature. After characterization, Fe3O4@CS@ZIF-67 was used as adsorbent for the magnetic solid phase extraction of benzoylurea insecticides (BUs) in tea infusions prior to high performance liquid chromatographic analysis. The effects of the amount of adsorbent, ionic strength, pH, adsorption time, type of eluent and elution time were investigated. The adsorption could be completed in 1.5 min with only 15 mg adsorbent for the extraction of 10 mL sample solution. Good linearity was obtained for the analysis of BUs with R2 between 0.9988 and 0.9999. The limits of detection were in the range of 0.72–1.48 μg·L−1. Relative standard deviations (RSDs) for intra-day precision and inter-day precision were in the range of 2.62–4.32 % and 3.60–6.29 %, respectively. RSDs for batch-to-batch precision of Fe3O4@CS@ZIF-67 were between 0.41 % and 7.52 %. The spiked recoveries ranged from 75.7 % to 112.5 %. The proposed method has been successfully applied to the determination of BUs in tea infusion samples.

Online solid phase extraction high-performance liquid chromatography – Isotope dilution – Tandem mass spectrometry quantification of organophosphate pesticides, synthetic pyrethroids, and selected herbicide metabolites in human urine

Analytical methods to quantify pesticide biomarkers in human population studies are critical for exposure assessment given the widespread use of pesticides for pest and weed control and their potential for affecting human health. We developed a method to quantify, in 0.2 mL of urine, concentrations of 10 pesticide biomarkers: four organophosphate insecticide metabolites (3,5,6-trichloro-2-pyridinol (TCPy), 2-isopropyl-6-methyl-4-pyrimidinol, para-nitrophenol, malathion dicarboxylic acid); five synthetic pyrethroid insecticide metabolites (4-fluoro-3-phenoxybenzoic acid, 3-phenoxybenzoic acid, cis and trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane carboxylic acid (DCCA), cis-3-(2,2-dibromovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid); and the herbicide 2,4-dichlorophenoxyacetic acid.he method is based on enzymatic hydrolysis of conjugated urinary metabolites, extraction and pre-concentration of the deconjugated metabolites using automated online solid-phase extraction, and separation and quantification using liquid chromatography-isotope dilution tandem mass spectrometry.Depending on the analyte, method detection limits were 0.1–0.6 ng/mL; mean accuracy, calculated as spike recoveries, was 91–102%, and total precision, given as percent variation coefficient, was 5.9–11.5%. Percent differences associated with three freeze-thaw cycles, 24-h benchtop storage, and short-term processed sample stability were <14%.Method suitability was assessed by recurring successful participation in external quality assessment schemes and by analyzing samples from subjects with suspected exposure to pesticides (n = 40) or who self-reported consuming an organic diet (n = 50). Interquartile ranges were considerably lower for people consuming an organic diet than for those potentially exposed for cis-DCCA (0.37 ng/mL vs 0.75 ng/mL), trans-DCCA (0.88 ng/mL vs 1.78 ng/mL) and TCPy (1.81 ng/mL vs 2.48 ng/mL). This method requires one-fifth of the sample used in our previous method and is suitable for assessing background exposures to select pesticides in large human populations and for studies with limited sample volumes.

Nanoconfinement of functionalized ionic liquid for enhanced adsorption and rapid sensitive detection of phenylurea herbicides in food and environmental samples

The novel ILs@CNTs was synthesized by encapsulating task-specific ionic liquids (ILs) within carbon nanotubes (CNTs) derived from ZIF-67. These hybrid materials served as multifunctional adsorbents enabling simultaneous sorptive removal, sensitive detection, molecular sieve selection, and magnetic separation. In contrast to pristine CNTs, ILs@CNTs demonstrate significantly enhanced adsorption of phenylurea herbicides (PUHs). The complex interactions between ILs@CNTs and PUHs were comprehensively analyzed using a combination of experimental results and theoretical calculations. Furthermore, a magnetic solid phase extraction-high performance liquid chromatography (MSPE-HPLC) method was developed for the determination of multiple trace PUHs in real samples. The method exhibited lower detection limits (0.02–0.03 μg L−1) and higher enrichment factors (131 < EFs < 185). Interestingly, a portable lab-in-a-syringe device was developed to facilitate rapid on-site extraction and enrichment of PUHs. Additionally, the developed methods successfully applied in river water, tea drinks, and cucumber samples, highlighting its substantial potential for rapid PUH detection.

Occurrence and Risk of Organophosphate Flame Retardants in Multiple Urban Water of Beijing, China

Investigating the organophosphate flame retardants (OPFRs) concentration and attenuation in multiple water of Beijing would provide valuable insight into OPFR management and reduction; thus, 6 OPFRs were determined by solid phase extraction-high performance liquid chromatography-tandem mass spectrometry to explore the pollution characteristics, and ecological risk of OPFRs. The results showed that 5 OPFRs were detected with a concentration ranging from ND to 394.77 ng/L except for tris(2,3-dibromopropyl) phosphate (TDBP). The chloroalkyl phosphate was the main compound and tris(chloropropyl) phosphate (TCPP) was the most abundant OPFRs with the highest concentration. The concentration of aryl-OPFRs in surface water and groundwater was much lower than that of chlorinated ones, and the concentration of chlorinated OPFRs in surface water was higher than that in groundwater. The treatment effect of chlorinated OPFRs including TCPP, TCEP, and TDCPP in STPs was negative. Ecological risk assessment showed that low risk of OPFRs for the organisms (algae, crustacean, and fish).

Construction of hierarchically porous metal-organic framework HP-UiO-66-30% for sensitive determination of benzoylurea insecticides

Due to widespread application of benzoylurea insecticides (BUs) and its persistence in environment, the effective capture of benzoylurea insecticides residues in environment is an important issue of environmental safety monitoring. To obtain excellent adsorption performance, creating defective structure in metal-organic frameworks (MOFs) can be employed as the method for adjusting its properties. Zirconium(Ⅳ)-based MOF termed as UiO-66-30% was constructed with 2-aminoterephthalic acid (NH2-BDC) and terephthalic acid (H2BDC) as building blocks. After calcination and removal of thermal-sensitive ligand (NH2-BDC), hierarchically porous UiO-66-30% (HP-UiO-66-30%) with multistage pore structure and good stability was obtained. The unique structure of HP-UiO-66-30% endowed it to achieve instantaneous equilibrium (within 2 min) when it was used as a dispersed solid phase extraction (d-SPE) adsorbent to extract BUs from environmental samples, greatly reducing the operation time. A wide linear range (0.05–200 ng mL−1), good linearity (R2 ≥ 0.9980), low detection limits (0.01–0.03 ng mL−1) and quantification limits (0.05–0.1 ng mL−1) were obtained for BUs. In addition, the HP-UiO-66-30% material possessed the good reusability and the adsorption capacity did not change significantly over 16 adsorption-desorption cycles. Finally, the established dispersed solid phase extraction-high performance liquid chromatography-diode array detector (d-SPE-HPLC-DAD) method was successfully applied to determination of BUs residues in environmental soil samples. The results demonstrated that HP-UiO-66-30% was an excellent sorbent for extraction BUs from environmental samples.

Simultaneous determination of 22 antibiotics in environmental water samples by solid phase extraction-high performance liquid chromatography-tandem mass spectrometry

The widespread and frequent use of antibiotics to treat diseases or encourage animal growth has resulted in their persistence and accumulation in water, soil, and sediments. As a typical emerging pollutant in the environment, antibiotics have become an important research focus in recent years. Antibiotics are commonly found at trace levels in water environments. Unfortunately, the determination of various types of antibiotics, all of which exhibit different physicochemical properties, remains a challenging endeavor. Thus, developing pretreatment and analytical techniques to achieve the rapid, sensitive, and accurate analysis of these emerging contaminants in various water samples is an essential undertaking.In this paper, a solid phase extraction-high performance liquid chromatography-tandem mass spectrometry (SPE-HPLC-MS/MS) method for the simultaneous determination of 22 antibiotics including 4 penicillins, 12 quinolones and 6 macrolides in environmental water samples was developed. Based on the characteristics of the screened antibiotics and sample matrix, the pretreatment method was optimized, focusing on the SPE column, pH of the water sample, and amount of ethylene diamine tetra-acetic acid disodium (Na2EDTA) added to the water sample. Prior to extraction, a 200 mL water sample was added with 0.5 g of Na2EDTA and pH-adjusted to 3 using sulfuric acid or sodium hydroxide solution. Water sample enrichment and purification were achieved using an HLB column. HPLC separation was carried out on a C18 column (100 mm×2.1 mm, 3.5 μm) via gradient elution with a mobile phase composed of acetonitrile and 0.15% (v/v) formic acid aqueous solution. Qualitative and quantitative analyses were performed on a triple quadrupole mass spectrometer in multiple reaction monitoring mode using an electrospray ionization source. The results showed correlation coefficients greater than 0.995, indicating good linear relationships. The method detection limits (MDLs) and limits of quantification (LOQs) were in the ranges of 2.3-10.7 ng/L and 9.2-42.8 ng/L, respectively. The recoveries of target compounds in surface water at three spiked levels ranged from 61.2% to 157%, with relative standard deviations (RSDs) of 1.0%-21.9%. The recoveries of target compounds in wastewater at three spiked levels were 50.1%-129%, with RSDs of 1.2%-16.9%. The method was successfully applied to the simultaneous determination of antibiotics in reservoir water, surface water, sewage treatment plant outfall, and livestock wastewater. Most of the antibiotics were detected in watershed and livestock wastewater. Lincomycin was detected in 10 surface water samples, with a detection frequency of 90%, and ofloxacin showed the highest contents (127 ng/L) in livestock wastewater. Therefore, the present method exhibits excellent performance in terms of MDLs and recoveries compared with previously reported methods. The developed method presents the advantages of small water sample volumes, wide applicability, and fast analysis times; thus, it can be considered a rapid, efficient, and sensitive analytical method with excellent potential for monitoring emergency environmental pollution. The method could also provide a reliable reference for formulating antibiotic residue standards. The results provide strong support for and an improved understanding of the environmental occurrence, treatment, and control of emerging pollutants.

Determination of 15 carbonyl compounds in soil using improved solid phase extraction-high performance liquid chromatography

An improved solid phase extraction (SPE)-high performance liquid chromatography method was established to determine 15 carbonyl compounds, namely, formaldehyde (FOR), acetaldehyde (ACETA), acrolein (ACR), acetone (ACETO), propionaldehyde (PRO), crotonaldehyde (CRO), butyraldehyde (BUT), benzaldehyde (BEN), isovaleraldehyde (ISO), n-valeraldehyde (VAL), o-methylbenzaldehyde (o-TOL), m-methylbenzaldehyde (m-TOL), p-methylbenzaldehyde (p-TOL), n-hexanal (HEX), and 2,5-dimethylbenzaldehyde (DIM), in soil. The soil was ultrasonically extracted with acetonitrile, and the extracted samples were derivatized with 2,4-dinitrophenylhydrazine (2,4-DNPH) to generate stable hydrazone compounds. The derivatized solutions were cleaned using an SPE cartridge (Welchrom® BRP) packed with N-vinylpyrrolidone/divinylbenzene copolymer. Separation was performed on an Ultimate® XB-C18 column (250 mm×4.6 mm, 5 μm), isocratic elution was performed with acetonitrile-water (65∶35, v/v) as the mobile phase, and detection was performed at a wavelength of 360 nm. The 15 carbonyl compounds in the soil were then quantified using an external standard method. The proposed method improves the sample processing method described in the environmental standard HJ 997-2018: Soil and sediment-Determination of carbonyl compounds-High performance liquid chromatography. A series of experiments revealed the following optimal conditions for soil extraction: acetonitrile as the extraction solvent, extraction temperature of 30 ℃, and extraction time of 10 min. The results showed that the purification effect of the BRP cartridge was significantly better than that of the conventional silica-based C18 cartridge. The 15 carbonyl compounds showed good linearities, and all correlation coefficients were above 0.996. The recoveries ranged from 84.6% to 115.9%, the relative standard deviations (RSDs) ranged from 0.2% to 5.1%, and the detection limits were 0.02-0.06 mg/L. The method is simple, sensitive, and suitable for the accurate quantitative analysis of the 15 carbonyl compounds in soil specified in HJ 997-2018. Thus, the improved method provides reliable technical support for studying the residual status and environmental behavior of carbonyl compounds in soil.

Genome mining and chemical characterization of a new cyclic lipopeptide associated with MDN-0066 from Pseudomonas moraviensis HN2 cultured in a valine-rich medium.

A new cyclic lipopeptide (CLP) MDN-0066-β (1) and MDN-0066 (2) were isolated and characterized from the bacterial cultures of P. moraviensis HN2 in this study. The CLPs were purified by solid-phase extraction (SPE) and reversed-phase high performance liquid chromatography (RP-HPLC). Moreover, chemical structures of two CLPs were characterized by genome mining and analysis, nuclear magnetic resonance (NMR), high-resolution mass spectrometry (HR-MS), Marfey's method and (C-H)α NMR fingerprint matching approach. MDN-0066 (2) has an amino acid sequence of L-Leu1, D-Glu2, D-allo-Thr3, D-Leu4, D-Leu5, D-Ser6, L-Leu7, L-Ile8 linked to a saturated C10 β-hydroxyl fatty acid moiety (R-configuration for 3-OH). The new CLP MDN-0066-β (1) differs MDN-0066 (2) in the 8th position of L-valine in its peptide moiety, this variation in structure could be attributed to the supplement of L-valine in the cultural medium during liquid fermentation. Further antimicrobial tests showed that the two CLPs display moderate antagonistic activity against Staphylococcus aureus and Escherichia coli.

Preparation of magnetic molybdenum disulfide for the extraction of neonicotinoid insecticides from environmental water samples

In this study, magnetic molybdenum disulfide hybrid composites (Fe3O4/MoS2) were successfully synthesized by in situ fabrication method and firstly applied as magnetic adsorbent for extracting neonicotinoid insecticides from environmental water samples. The Fe3O4/MoS2 composites were characterized in detail by scanning electron microscope, fourier transform infrared, X-ray diffraction and vibrating sample magnetometer, and possessed the faster adsorption capacity towards neonicotinoid insecticides (only 10 min). The parameters (solution pH, amounts of adsorbents, adsorption time, elution conditions and sample volume) effecting the performance of magnetic solid-phase extraction were carefully investigated. Under the optimal conditions, the magnetic solid-phase extraction-high performance liquid chromatography method based on Fe3O4/MoS2 composites for enriching and detecting neonicotinoid insecticides in environmental water samples was established. The good linearity (0.01–10 μg/mL) with correlation coefficients between 0.9949 and 0.9999 was obtained. The limits of detection and the limits of quantifications were 0.48–0.67 ng/mL and 1.44–2.02 ng/mL, respectively. With spiked at three different concentration levels (50, 100,500 ng/mL), the recovery was 66.0–91.3 % with relative standard deviations from 1.4 % to 8.3 %. These data demonstrated that the developed method was highly sensitive, repeatable and easily operated. Also, this study provided new adsorbents for other trace pollutants in the environment.

Analysis of parabens in environmental water samples by covalent organic framework-based magnetic solid-phase extraction-high performance liquid chromatography

Parabens are a class of antimicrobial preservatives that are widely used in cosmetics, pharmaceuticals, and food products because of their ease of production, antimicrobial effect, and low price. The widespread use of these parabens, poses potential risks to human health. Therefore, it is necessary to establish a simple and rapid method for the detection of parabens. The large number of substrate interferences in complex samples is an important factor affecting the sensitivity of analytical methods. Magnetic solid-phase extraction (MSPE) has received much attention because of its advantages of easy operation, short extraction time, small sample amount, low cost, and environmental friendliness. Covalent organic frameworks (COFs) with high crystallinity, high specific surface area, adjustable pore size, regular porosity, as well as high chemical and thermal stability are now widely used in separation and analysis. Therefore, a sample pretreatment method combining MSPE and COF for the analysis of parabens in complex matrices is very promising. A magnetic covalent organic framework, Fe3O4@TbBd, was successfully synthesized by the Schiff base reaction of 1,3,5-triformylbenzene (Tb) and benzidine (Bd) at room temperature using Fe3O4 nanoparticles as magnetic cores. Characterization by scanning electron microscopy (SEM), thermogravimetric analysis (TGA), X-ray diffraction (XRD), vibrating sample magnetometer (VSM) measurements, etc. revealed that the magnetic COF has high magnetic responsiveness, as well as good thermal and chemical stability, which make it an ideal adsorbent for the MSPE of parabens. Some factors related to the extraction efficiency, including the amount of adsorbent, extraction time, pH, desorption solvent, desorption time, and number of desorption were systematically investigated. A method involving MSPE and high performance liquid chromatography-ultraviolet detection (HPLC-UV) based on the Fe3O4@TbBd was developed for the determination of four parabens (ethylparaben, propylparaben, butylparaben, and benzylparaben) in environmental water samples. Under the optimal extraction conditions, the method showed good linearities. The limits of detection and limits of quantification were 0.2-0.4 μg/L and 0.7-1.4 μg/L for the four analytes, respectively. The recoveries at three spiked levels were in the range of 86.1%-110.8% with intra-day and inter-day RSDs of less than 5.5% and 4.9%, respectively. The method was successfully applied to the determination of parabens in East Lake water, Yangtze water, and domestic wastewater. Ethyl paraben and propyl paraben were detected in domestic wastewater at the levels of 1.8 μg/L and 0.4 μg/L, respectively. The recoveries of the parabens at different spiked levels ranged from 80.7% to 117.5%, with RSDs of 0.2%-8.8%. The method has good potential for the determination of parabens in environmental water samples because of its operational simplicity, short extraction time, high sensitivity, and environmental friendliness.

Simultaneous determination of four antibiotics in human urine and hair samples employing a simple and efficient dispersive micro solid-phase extraction-high performance liquid chromatography.

Antibiotics containing azithromycin, amoxicillin, doxycycline, tetracycline, and azithromycin are widely utilized in the treatment of bacterial infections, and their determination in biological samples is critical to evaluate their side effects. A dispersive micro solid-phase extraction (DMSPE) as an efficient sample preparation was developed for the simultaneous extraction of the antibiotics in biological samples. For this purpose, chitosan@Fe3O4 nanoparticles were prepared as a green and magnetic sorbent based on a chemical coating of Fe3O4 nanoparticles with chitosan. The sorbent was compared with several sorbents such as ZnO NPs, CuO NPs, and Fe3O4 NPs to extract the antibiotics. In addition, effective factors for antibiotic extraction were evaluated and optimized using a central composite design and one factor at a time. The method has wide linearity in the concentration ranges of 2.1-800.0, 3.5-1000.0, 2.0-800.0, and 2.7-1000.0μgL-1 with an R-squared higher than 0.9958 for the determination of azithromycin, amoxicillin, doxycycline, and tetracycline, respectively. The limit of detection and limit of quantitation was lower than 1.1 and 3.5μgL-1, respectively. The relative standard deviations (RSDs) were also lower than 4.90% for the determination of the analytes at a concentration of 20.0µgL-1. High recoveries in the percentage range of 94.70-106.75%, with RSDs between 4.64 and 5.83%, were obtained to analyze spiked urine and hair samples. The DMSPE-HPLC-DAD procedure displayed various advantages for determining antibiotics, including simple operation, high recoveries, low sample solution and desorption solvent consumption, high preconcentration factors, proper determination time, and simple and inexpensive method of sorbent preparation.

Simultaneous Determination of Protoberberine Alkaloids Derived from Coptidis Rhizoma and Phellodendri Cortex in Pharmaceuticals by On-line Solid-Phase Extraction (SPE)—High Performance Liquid Chromatography with Ultraviolet Detection (HPLC-UV)

An automated on-line solid-phase extraction (SPE) high-performance liquid chromatography (HPLC) method was developed for the determination of six protoberberine alkaloids in compound Chinese medicines containing Coptidis Rhizoma and Phellodendri Cortex. The sample solution extracted by acidic-methanol was loaded into the constructed system directly to perform enrichment and the purification of the analytes was performed automatically. A cation-exchanged sorbent based on turbulent chromatography was employed for on-line SPE to remove macro and neutral matrix interferences from samples. The target alkaloids were retained on the SPE column and eluted onto the trapping C18 column by a mobile phase of methanol/500 mM ammonium acetate (85:15) at a flow rate of 0.3 mL/min. Ion pair reverse phase high-performance liquid chromatography was employed to complete the determination of six analytes. The results showed the linear correlation coefficients for all analytes were above 0.999. The peak area of relative standard deviation in reproducibility measurements were from 0.55% to 2.39%. Recovery measurements spiked at two levels were from 92.71% to 104.28%. The reported method was suitable for the determination of six active alkaloids in multicomponent Chinese medicines containing Coptidis rhizoma and Phellodendri cortex.

Simultaneous determination of raloxifene and tamoxifen using dispersive micro solid-phase extraction and HPLC method in real water, human hair and serum samples

ABSTRACT A sensitive determination procedure was presented for two anticancer drugs (tamoxifen and raloxifene) in real water and biological samples based on magnetic dispersive micro solid-phase extraction–high-performance liquid chromatography with an ultraviolet detector. Fe3O4 NPs@SiO2@TiO2 nanocomposite was synthesised as a magnetic sorbent by the sol-gel technique. The effective parameters in the procedure were optimised with an experimental design, including screening and optimisation steps. Under the optimum determination conditions, the procedure displayed wide linear ranges of 5.3–735.0 and 7.0–806.0 ng mL−1 with R-squared of 0.9916 and 0.9908, limit of detections of 1.6 and 2.1 ng mL−1 for the raloxifene and tamoxifen determination, respectively. The intra-day relative standard deviations were 3.76 and 3.59% for three times the raloxifene and tamoxifen measurements, respectively. The preconcentration factors for the raloxifene and tamoxifen determination with a concentration of 20 ng mL−1 were 506.1 and 546.7, respectively. The method was applied to analyse well water, tap water, and hair samples and determine raloxifene and tamoxifen simultaneously with recoveries between 88.5–96.4 and relative standard deviations lower than 8.4%. The method’s advantages for raloxifene and tamoxifen measurement included easy operation, low sorbent and desorption solvent consumption, simple sorbent preparation and separation, and suitable analysis time.

Determination of trans-4-hydroxy-2-alkenals in thermally treated soybean oil by SPE-HPLC

Trans-4-hydroxy-2-hexenal (4-HHE) and trans-4-hydroxy-2-nonenal (4-HNE) are secondary lipid peroxidation products in edible oils, which are cytotoxic and genotoxic. They could covalently bind with protein, phospholipids and DNA, further disrupting the normal function of liver, lung and brain. Derivation process was generally conducted during pretreatment before detection and quantification of 4-HHE and 4-HNE. However, the derivation procedures were time consuming and chemical degradation may occur during the process. Hence, this paper aims to establish a simple solid phase extraction-high performance liquid chromatography (SPE-HPLC) method to determine the 4-HHE and 4-HNE contents in thermally treated soybean oil. C18 solid phase extraction was applied in the pretreatment process. Firstly, the reliability of the method was evaluated. Good linearity was observed in the range of 0.1–0.5 μg/mL and 0.5–10 μg/mL for 4-HHE and 4-HNE. The limit of detection (LOD) of 4-HHE and 4-HNE were 0.0486 and 0.0129 μg/mL, respectively. And the limit of quantitation (LOQ) of 4-HHE and 4-HNE were 0.1458 and 0.0431 μg/mL, respectively. Recovery rate were in the range of 89.11%–91.58% and 71.83%–79.40% for 4-HHE and 4-HNE, respectively. The method achieved the extraction, purification and detection of 4-HHE and 4-HNE simultaneously and had the advantages of simple operation, effectiveness, high precision, good repeatability. Then, the method was applied to monitor the concentrations of 4-HHE and 4-HNE in soybean oil heated at 180 °C for 40 h. The contents of 4-HHE and 4-HNE were 0–0.32 μg/g and 0–6.97 μg/g, respectively, which provided guidance for evaluating health risks of thermally treated soybean oil during heating.

Effect of Bisphenol A-Mediated RBP-4 on Pregnancy Outcomes in Nonobese Pregnant Female with Subclinical Hypothyroidism.

Hypothyroidism is a systemic hypometabolic syndrome caused by the thyroxine resistance or a reduction in its extent. It is an endocrinopathy secondary to gestational diabetes and occurs usually without significant symptoms. This study explored the effect of bisphenol A (BPA)-mediated retinol-binding protein 4 (RBP-4) on pregnancy outcomes in a nonobese pregnant female with subclinical hypothyroidism. Three hundred nonobese pregnant females who had that established pregnancy files and had regular obstetric examinations from January 2021 to March 2022 were enrolled and classified with 100 cases in each group as early pregnancy (6–12 weeks of gestation), second-trimester (13–24 weeks of gestation), and third-trimester groups (25–36 weeks of gestation). Thirty pregnant women with subclinical hypothyroidism were selected as subjects, and another thirty pregnant women with normal thyroid function were selected as the normal control group. Thyroid function (thyroid-stimulating hormone (TSH), free triiodothyronine (FT3), free T4 (FT4), and thyroid peroxidase antibody (TPO-Ab)) was measured by immunoelectrochemiluminescence. The level of BPA in urine was determined by solid-phase extraction high-performance liquid chromatography-tandem mass spectrometry. Serum RBP4 levels were determined by enzyme-linked immunosorbent assay. The level of TSH in the third-trimester group was higher than that in the first- and second-trimester groups, while the levels of FT3, FT4, and TPO-Ab were lower than those in the other two groups (P < 0.05). TSH in the second-trimester group was higher than that in the first-trimester group, while FT3, FT4, and TPO-Ab levels were lower than those in the first-trimester group (P < 0.05). The levels of BPA and RBP4 in gestational diabetes mellitus and hypertension were higher than those in the nongestational period, and the levels of BPA and RBP4 in gestational intrahepatic cholestasis and anemia were higher than those in the nongestational period, and the levels of BPA and RBP4 in preterm delivery were higher than those in nongestational period (P < 0.05). Also, the level of urinary BPA in the hypothyroidism group was higher than that in the normal control group (P < 0.05) and the level of serum RBP4 in the hypothyroidism group was higher than that in the normal control group (P < 0.05). According to multivariate logistic regression analysis, age ≥30 years and the ascending BPA and RBP4 were risk factors for subclinical hypothyroidism during pregnancy in the nonobese female. BPA and RBP-4 are closely related to the pregnancy outcome of nonobese subclinical hypothyroidism in the pregnant female. The degree of BPA and RBP-4 in adverse pregnancy outcomes is increased, which is the risk factor for nonobese subclinical hypothyroidism.

Distribution and Ecological Risk Assessment of Pharmaceuticals and Personal Care Products in Sediments of North Canal, China

The pollution of water bodies by pharmaceuticals and personal care products (PPCPs) has attracted widespread concern due to their widespread use and pseudo-persistence, but their effects on sediments are less known. In this study, solid-phase extraction-high performance liquid chromatography–tandem mass spectrometry (SPE-LC/MSMS) was used to investigate the occurrence and ecological risks of five typical pharmaceuticals and personal care products (PPCPs) in thirteen key reservoirs, sluices, dams, and estuaries in the Haihe River Basin. At the same time, the PPCP exchanges of surface water, groundwater, and sediments in three typical sections were studied. Finally, the PPCP’s environmental risk is evaluated through the environmental risk quotient. The results showed that the five PPCPs were tri-methoprazine (TMP), sinolamine (SMX), ibuprofen (IBU), triclosan (TCS), and caffeine (CAF). The average concentration of these PPCPs ranged from 0 to 481.19 μg/kg, with relatively high concentrations of TCS and CAF. The relationship between PPCPs in the surface sediments was analyzed to reveal correlations between SMX and TMP, CAF and IBU, CAF and TCS. The risk quotients (RQ) method was used to evaluate the ecological risk of the five detected PPCPs. The major contributors of potential environmental risks were IBU, TCS and CAF, among which all the potential environmental risks at the TCS samples were high risk. This study supplemented the research on the ecological risk of PPCPs in sediments of important reaches of the North Canal to reveal the importance of PPCP control in the North Canal and provided a scientific basis for pollution control and risk prevention of PPCPs.

Exposure assessment of polycyclic aromatic hydrocarbons in refined coal tar sealant applications.

Refined coal tar sealant (RCTS) emulsions are used to seal the surface of asphalt pavement. Nine of the 22 polycyclic aromatic hydrocarbons (PAHs) evaluated in this study are classified as known, probable, or possible human carcinogens. Exposure assessment research for RCTS workers has not been published previously. The overall objective of this study was to develop a representative occupational exposure assessment of PAH exposure for RCTS workers based on worksite surveys. The specific aims were to: 1) quantify full-shift airborne occupational exposures to PAHs among RCTS workers; 2) quantify workers' dermal exposures to PAHs; 3) quantify biomarkers of PAH exposure in workers' urine; 4) identify specific job titles associated with RCTS exposure; and 5) apply these results to a biological exposure index to assess risk of potential genotoxicity from occupational exposures. A total of twenty-one RCTS workers were recruited from three companies. Personal and area air samples were collected using a modification of NIOSH Method 5515. Dermal exposure was assessed by hand and neck wipes before and after shifts. Twenty-two PAHs were quantified via gas chromatography-mass spectrometry selected ion monitoring. Internal dose was estimated by quantifying select PAH metabolites in pre- and post-shift urine samples using on-line solid phase extraction-high performance liquid chromatography-tandem mass spectrometry. PAH levels in the worker breathing zones were highest for naphthalene, acenaphthene, and phenanthrene, with geometric means of 52.1, 11.4, and 9.8μg/m3, respectively. Hand wipe levels of phenanthrene, fluoranthene and pyrene were the highest among the 22 PAHs with geometric means of 7.9, 7.7, and 5.5μg/cm2, respectively. Urinary PAH biomarkers for naphthalene, fluorene, phenanthrene, and pyrene were detected in all workers and were higher for post-shift samples than those collected pre-shift. Urinary concentrations of the metabolite 1-hydroxypyrene were greater than the American Conference of Governmental Industrial Hygienists (ACGIH) Biological Exposure Index (BEI) for this metabolite in 89 percent of post-shift samples collected on the final day of the work week or field survey. Statistically significances were found between concentrations of fluorene, naphthalene, and phenanthrene in the breathing zone of workers and their corresponding urinary PAH biomarkers. Workers were placed in two work place exposure groups: applicators and non-applicators. Applicators had higher total PAH concentrations in personal breathing zone (PBZ) air samples than non-applicators and were more likely to have post-shift hand wipe concentrations significantly higher than pre-shift concentrations. Concentrations of post-shift urinary biomarkers were higher, albeit not significantly, for applicators than non-applicators. The exposure results from RCTS worker samples cannot be explained by proximal factors such as nearby restaurants or construction. Air and skin concentration levels were substantially higher for RCTS workers than previously published levels among asphalt workers for all PAHs. PAH profiles on skin wipes were more consistent with RCTS sealant product than air samples. Last day post-shift urinary concentrations of 1-hydroxypyrene greatly exceeded the ACGIH BEI benchmark of 2.5μg/L in 25 of 26 samples, which suggests occupational exposure and risk of genotoxicity. When pyrene and benzo[a]pyrene were both detected, concentration ratios from personal exposure samples were used to calculate the adjusted BEI. Concentrations of 1-hydroxypyrene exceeded the adjusted BEIs for air, hand wipes, and neck wipes in most cases. These results indicate the need to increase safety controls and exposure mitigation for RCTS workers.

Determination and level investigation of 45 hormones in edible bird's nest by solid phase extraction-high performance liquid chromatography-tandem mass spectrometry

建立了固相萃取-高效液相色谱-串联质谱(SPE-HPLC-MS/MS)同时测定食用燕窝中皮质激素、雌激素、雄激素及孕激素等5类45种激素的多残留分析方法。采用乙腈-乙酸乙酯(1∶1, v/v)超声辅助提取、亲水亲脂平衡固相萃取柱净化,甲醇洗脱。分别在正、负电喷雾电离源、多反应监测模式下检测45种激素。正离子模式下的流动相为乙腈-0.1%甲酸水溶液,负离子模式下的流动相为乙腈-水,色谱柱为Phenomenex Kinetex C18柱(100 mm×2.1 mm, 2.6 μm)。在优化条件下,45种激素在0.20~20.0 μg/L范围内线性关系良好,相关系数(R2)≥0.9990,方法的检出限(LOD)为0.04~0.70 μg/kg,定量限(LOQ)为0.16~2.00 μg/kg。按三水平(2.0、4.0、20.0 μg/kg)进行加标回收试验,氟米龙、布地奈德、醛固酮、醋酸氟轻松、炔雌醇的回收率为40.2%~63.6%,可对这5种激素进行定性分析,其余40种激素的平均加标回收率为72.2%~112.3%,相对标准偏差(RSD)为2.5%~11.6%,该方法可对40种激素进行准确定性定量,精密度好,灵敏度高,简便、快速。从2017~2021年,通过研究建立的方法对来自马来西亚、印度尼西亚、泰国和越南等国家的1021个燕窝样品进行监测,仅勃地酮、雄烯二酮、孕酮有检出(大于检出限),其他激素均小于检出限。孕酮检出率为100%,勃地酮、雄烯二酮检出率分别为79%和89%, 3种激素含量范围分别为0.097~3.58、0~0.096和0~1.77 μg/kg。与同为动物源性食品的鸡蛋、纯牛奶、乳制品相比,所有测定的鸡蛋样品中均检出雄烯二酮,含量比其他3类产品略高;勃地酮在4类产品中的含量差别不大,均为微量;孕酮含量在鸡蛋中最高,其次是纯牛奶,燕窝中含量最低。研究结果表明,食用燕窝带入的激素种类少,含量低,对健康影响小。

Hierarchical porous phenolic polymer for efficient adsorption of triazine herbicides: Novel preparation strategies and potential applications

Efficient removal of pollutants based on porous adsorbents is a research hotspot now. Triazine herbicides (TRZHs), a class of broad-spectrum, low-cost pesticides, have become an environmental concern owing to the widespread use and abuse these chemicals. In this study, an ionic liquid (IL)-functionalized porous m-aminophenol formaldehyde polymer (IL-PMAPFP) used to adsorb four TRZHs (simazine, cyanazine, atrazine, and terbuthylazine) in environmental water was synthesized. The role of the ethylene oxide/propylene oxide block copolymer (F127) and IL in pore formation was confirmed by microscopic morphology and specific surface area analysis of IL-PMAPFP. The specific surface area of IL-PMAPFP increased 13-fold after adding F127 and the IL. The adsorption of TRZHs on IL-PMAPFP conformed to the pseudo-second-order kinetic and Freundlich isotherm models. The adsorption process is spontaneous, endothermic, and has a strong anti-interference ability. The adsorption equilibrium of IL-PMAPFP could be reached within 1 h, and the adsorption capacities of the four TRZHs are 4.26, 4.82, 5.21, and 8.53 mg/g, respectively, which were higher than many reported adsorbents. Theoretically, the ability of the adsorbent to treat TRZH-contaminated water is 450–900 L/g. IL-PMAPFP is a powerful adsorbent owing to its large surface area, high adsorption amount, fast mass transfer, and good regeneration capability (>eight cycles). IL-PMAPFP combined with dispersive solid phase extraction/miniaturized pipette tip solid phase extraction-high performance liquid chromatography is expected to accurately quantify TRZHs. In addition, the synergistic pore formation strategy of F127 and IL present a new method for preparing of porous polymer. This study provides a new perspective for developing efficient purification techniques to monitor and remove pollutants.
