Spatial variations impact the soil fungal communities of Larix gmelinii forests in Northeast China

Soil fungi play a critical role in the biogeochemical cycles of forest ecosystems. Larix gmelinii is a strong and important timber tree species, which forms close associations with a wide range of soil fungi. However, the temporal-spatial disparity effects on the assembly of soil fungal communities in L. gmelinii forests are poorly understood. To address these questions, a total of 120 samples, including 60 bulk soil and 60 root samples, were collected from Aershan and Genhe in July (summer) and October (autumn)2021. We obtained 7,788 operational taxonomic units (OTUs) after merging, filtering, and rarefying using high-throughput sequencing. The dominant phyla are Basidiomycota, Ascomycota, Mortierellomycota, and Mucoromycota. There were 13 dominant families, among which the families with average relative abundance more than 5% included Thelephoraceae, Mortierellaceae, Archaeorhizomycoaceae, and Inocybaceae. In the functional guilds, symbiotrophic fungi had a relative advantage in the identified functions, and the relative abundances of pathotrophic and saprotrophic fungi varied significantly between sites. There were 12 families differentially expressed across compartments, 10 families differentially expressed between seasons, and 69 families were differentially expressed between sites. The variation in alpha diversity in the bulk soil was greater than that in the rhizosphere soil. Among the three parts (compartment, season, and site), the site had a crucial effect on the beta diversity of the fungal community. Deterministic processes dominated fungal community assembly in Genhe, whereas stochastic processes dominated in Aershan. Soil physicochemical properties and climatic factors significantly affected fungal community structure, among which soil total nitrogen and pH had the greatest effect. This study highlights that spatial variations play a vital role in the structure and assembly of soil fungal communities in L. gmelinii forests, which is of great significance for us in maintaining the health of the forests.

Effects of Maize/Peanut Intercropping and Nitrogen Fertilizer Application on Soil Fungal Community Structure

Maize/peanut intercropping may improve soil health through reducing nitrogen (N) fertilization. However, the effects of maize/peanut intercropping combined with reduced N fertilization on the soil fungal community structure have not been well reported. Using a long-term localized micro-zone experiment, we investigated the combined effects of intercropping and N fertilizer application on soil fungal community diversity and composition. Three cropping patterns (maize/peanut intercropping, maize monoculture, and peanut monoculture) and three N application levels (0 kg·hm−2, 150 kg·hm−2, and 300 kg·hm−2) were assessed. The results showed that the total numbers of fungal species and unique species (operational taxonomic units, OTUs) in both maize and peanut soils tended to first increase and then decrease with increasing N application. Compared with monoculture, the numbers of total OTUs and unique OTUs in intercropped maize soil decreased by 4.14% and 12.79%, respectively, but the total numbers of OTUs and unique OTUs in peanut soil increased by 1.08% and 3.78%, respectively. With increasing N application, the soil fungal Ace and Chao indices of maize soil first increased and then decreased, while the fungal Shannon, Ace, and Chao indices of peanut soil decreased. Compared with the monoculture system, intercropping significantly reduced the maize soil fungal Ace and Chao indices but increased the peanut soil fungal Shannon, Ace, and Chao indices. Nitrogen application and intercropping significantly altered the fungal community structure of maize soil, while N application had no significant effect on the fungal community structure of peanut soil, though intercropping significantly changed the fungal community structure of peanut soil. At the phylum level, Ascomycota, Basidiomycota, Mortierellomycota, unclassified_k_Fungi, and Chytridiomycota were the dominant taxa. Redundancy analysis (RDA) showed that soil nitrate (NO3−) content was the main environmental factor shaping the soil fungal community. In conclusion, excessive N fertilization (300 kg·hm−2) can reduce soil fungal community diversity; maize/peanut intercropping reversed the negative effect of N application on fungal community of peanut soil, but not that of maize soil. Soil NO3− content is the primary environmental driver of soil fungal communities.

Nitrogen addition changed soil fungal community structure and increased the biomass of functional fungi in Korean pine plantations in temperate northeast China

Nitrogen (N) deposition is a global environmental issue that can have significant impacts on the community structure and function in ecosystems. Fungi play a key role in soil biogeochemical cycles and their community structures are tightly linked to the health and productivity of forest ecosystems. Based on high-throughput sequencing and ergosterol extraction, we examined the changes in community structure, composition, and biomass of soil ectomycorrhizal (ECM) and saprophytic (SAP) fungi in 0–10 cm soil layer after 8 years of continuous N addition and their driving factors in a temperate Korean pine plantation in northeast China. Our results showed that N addition increased fungal community richness, with the highest richness and Chao1 index under the low N treatment (LN: 20 kg N ha−1 yr−1). Based on the FUN Guild database, we found that the relative abundance of ECM and SAP fungi increased first and then decreased with increasing N deposition concentration. The molecular ecological network analysis showed that the interaction between ECM and SAP fungi was enhanced by N addition, and the interaction was mainly positive in the ECM fungal network. N addition increased fungal biomass, and the total fungal biomass (TFB) was the highest under the MN treatment (6.05 ± 0.3 mg g−1).Overall, we concluded that N addition changed soil biochemical parameters, increased fungal activity, and enhanced functional fungal interactions in the Korean pine plantation over an 8-year simulated N addition. We need to consider the effects of complex soil conditions on soil fungi and emphasize the importance of regulating soil fungal community structure and biomass for managing forest ecosystems. These findings could deepen our understanding of the effects of increased N deposition on soil fungi in temperate forests in northern China, which can provide the theoretical basis for reducing the effects of increased N deposition on forest soil.

Differential response patterns of soil fungal and bacterial communities to typical vegetation types in the Yellow River floodplain ecosystem

Soil microorganisms are one of the primary driving factors of biogeochemical cycles in floodplain ecosystems. Vegetation type has an impact on the activities of soil microorganisms. However, it is currently unclear whether the composition and structure of soil bacterial and fungal communities in floodplain ecosystems have a consistent response pattern to vegetation communities. In this research, Illumina Mi-Seq sequencing technology was employed to study the soil fungal and bacterial communities covered by three typical vegetation communities in the Yellow River floodplain ecosystem. Our results showed that the underground biomass, total carbon (TC) and nitrogen (TN) were significantly different between the three vegetation communities. In the surface soil of summer, the Shannon index of fungi community was the highest in Tamarix chinensis (T. chinensis) and the lowest in Phragmites australis (P.australis). In autumn, the Shannon and Simpson indices of bacterial communities in surface soil were the highest in the P.australis and the lowest in the Sonchus arvensis (S.arvensis), but there was no significant difference in the β-diversity of bacterial community among the three vegetation communities. In autumn, the Chao1 index of fungal community in subsoil was the highest in P.australis and the lowest in S.arvensis, while there was no significant difference in the α-diversity of bacterial community among the three vegetation communities. In addition, there were considerable disparities in the fungal and bacteria community structure in various vegetation soils. Redundancy analysis (RDA analysis) showed that TN, TC, AP, pH, NO3 -, NH4 + and moisture content were key factors driving bacterial community structure, while TP, NH4 +, pH and moisture content were essential factors driving fungal community composition. These outcomes help to improve our understanding of the ecological model of soil fungal and bacterial community in floodplain ecosystems and give assistance in improving the role of an ecological barrier and floodplain service possibilities.

Soil Environments Regulate Dominant Soil Fungal Communities along an Elevational Gradient in Subtropical Forests

Soil fungal communities play a critical role in the promotion of nutrient cycling and the stabilization of ecosystem functions in subtropical forests. Yet, clarifying the relationships between soil fungal diversity and microclimate variability along an elevational gradient, as well as understanding the driving mechanisms of their variations in subtropical forests, remains insufficiently understood. In this study, we recorded the composition and soil fungal diversity along an elevational gradient in Daiyun Mountain of China, aiming to elucidate the primary factors influencing the structure of the dominant soil fungal along an elevational gradient in subtropical forests. The results showed that (1) the dominant phylum of soil fungi at different elevations were Basidiomycota, Ascomycota (relative abundance > 10%) and Zygomycota (relative abundance > 1%). The Simpson index of soil fungi showed a clear upward trend along the elevational gradient, while no significant difference was observed in the other indices, and both overall reached their maximum value at the elevation of 1200 m. (2) The mean annual soil temperature and moisture, soil pH and available phosphorus were the main factors driving the dominant soil fungal along the elevational gradient. (3) Co–occurrence network analyses revealed a distinct modular structure of dominant soil fungal communities at different elevations, with Ascomycetes identified as the key taxa in fungi network relationships. Our research holds ecological significance in understanding the pivotal role of soil environmental factors in shaping the complex composition and interactions within soil fungal communities.

Effect of Biogas Slurry on the Soil Properties and Microbial Composition in an Annual Ryegrass-Silage Maize Rotation System over a Five-Year Period.

Soil health is seriously threatened by the overuse of chemical fertilizers in agricultural management. Biogas slurry is often seen as an organic fertilizer resource that is rich in nutrients, and its use has the goal of lowering the amount of chemical fertilizers used while preserving crop yields and soil health. However, the application of continuous biogas slurry has not yet been studied for its long-term impact on soil nutrients and microbial communities in a rotation system of annual ryegrass-silage maize (Zea mays). This study aimed to investigate the impacts on the chemical properties and microbial community of farmland soils to which chemical fertilizer (NPK) (225 kg ha-1), biogas slurry (150 t ha-1), and a combination (49.5 t ha-1 biogas slurry + 150 kg ha-1 chemical fertilizer) were applied for five years. The results indicated that compared to the control group, the long-term application of biogas slurry significantly increased the SOC, TN, AP, and AK values by 45.93%, 39.52%, 174.73%, and 161.54%, respectively; it neutralized acidic soil and increased the soil pH. TN, SOC, pH, and AP are all important environmental factors that influence the structural composition of the soil's bacterial and fungal communities. Chemical fertilizer application significantly increased the diversity of the bacterial community. Variation was observed in the composition of soil bacterial and fungal communities among the different treatments. The structure and diversity of soil microbes are affected by different methods of fertilization; the application of biogas slurry not only increases the contents of soil nutrients but also regulates the soil's bacterial and fungal community structures. Therefore, biogas slurry can serve as a sustainable management measure and offers an alternative to the application of chemical fertilizers for sustainable intensification.

Nutrients addition decreases soil fungal diversity and alters fungal guilds and co-occurrence networks in a semi-arid grassland in northern China

Anthropogenic eutrophication is known to impair the diversity and stability of aboveground community, but its effects on the diversity, composition and stability of belowground ecosystems are not yet fully understood. In this study, we conducted a 9-year nitrogen (N) and phosphorus (P) addition experiment in a semi-arid grassland of Northern China to elucidate the impacts of nutrients addition on soil fungal diversity, functional guilds, and co-occurrence networks. The results showed that N addition significantly decreased soil fungal diversity and altered fungal community composition, whereas P addition had no impact on them. The relative abundance of arbuscular mycorrhizal fungi and leaf_saprotroph were reduced by N and P addition, but P addition enhanced the abundance of saprotrophic fungi. Co-occurrence network analysis revealed that N addition destabilized fungal network complexity and stability, while P addition slightly increased the network complexity. Additionally, the network analysis of N × P interaction revealed that P addition mitigated negative effects of N addition on network complexity and stability. Structural equation modeling (SEM) results suggested that nutrients addition directly or indirectly influenced the fungal community structure through the loss of plant richness and the increase of perennial grass biomass. These findings indicate that in comparison to P addition, N addition exhibits a pronounced negative effect on soil fungal communities. Our findings also suggest that changes in plant functional groups under nutrients deposition are pivotal in shaping soil fungal community structure in semi-arid grassland and highlight the need for a better understanding of the belowground ecosystem dynamics.

Effects of Transformation of Inefficient Camellia oleifera Plantation on Soil Quality and Fungal Communities

Camellia oleifera, a key economic forestry species in southern China, struggles with low productivity due to suboptimal planting management. Recently, transforming old or unadopted varieties of C. oleifera plantations has been recognized as a means to enhance economic benefits and production. However, the impact of these transformations on soil properties and fungal communities has received little attention. In this study, we targeted pre-renewal old C. oleifera and post-renewal young C. oleifera, Pinus massoniana, and Cunninghamia lanceolata. Through field sampling and soil physicochemical property analysis, we developed a soil quality evaluation system that effectively analyzes fungal community structures and identifies key arbuscular mycorrhizal fungi (AMF) species for soil health. We found that the soil quality evaluation system for this region comprises pH, TK, AK, NO3−, PO4− BG, ACP, F.simpson, AMF.shannon, and AMF.ace, which collectively indicated significant improvements in soil quality following transformation. Notably, the nutritional characteristics of the dominant fungal communities underwent marked changes, with an increase in pathogenic fungi in young C. oleifera and an expansion of ectomycorrhizal fungi in P. massoniana forests. The AMF communities in all four types of forest exhibited aggregation, and Scutellospora and Diversispora emerged as key species in the AMF community of C. oleifera. Additionally, Mortierella and Trichoderma were found to enhance plant resistance to pathogenic fungi. This study demonstrates that forestland transformation positively impacts soil quality and fungal community structure in C. oleifera, which provides valuable insights for future soil management in the region, both in terms of soil quality evaluation and fungal conservation.

Differences in soil fungal communities under salinity gradients in arid and semiarid regions

Salinization is considered a significant threat to soil ecosystem and plant growth in arid and semiarid regions. Soil microorganisms are supposed to be essential in relieving soil salinization and enhancing plant salt tolerance. However, although the responses of soil fungal diversity and community structure to salt stress have been investigated, major knowledge gaps remain in the interactions and community assembly of abundant and rare fungal taxa, and fungal functional characteristics under different salt intensities. Here, next-generation sequencing technology were used to investigate the response of all, abundant and rare fungal taxa to salt stress in arid and semiarid regions. The results demonstrated that high salt stress reduced the complexity and stability of fungal networks and increased fungal collaborations. Rare taxa held a key position in the co-occurrence network and exhibited greater proficiency in collaborating with intermediate and rare taxa, thereby augmenting the resistance of the community and maintaining its stability. Notably, with increasing salt stress, the importance of rare taxa gradually enhanced, while the importance of abundant taxa decreased. Moreover, fungal communities were manipulated by stochastic processes under salt stress, while were also influenced by deterministic processes (heterogeneous selection), especially for rare taxa. Furthermore, low salinity conditions promoted the colonization of saprophytic and symbiotic fungi and inhibited the growth of pathogenic fungi. Functional groups associated with fungal parasite-soil saprotroph-undefined saprotroph function were extremely sensitive to soil salinity and exerted a pivotal influence in sustaining community stability. Our findings may broaden our understanding of how abundant and rare fungi react to salt stress, making it easier to forecast the possible loss of biodiversity due to soil salinization, and offering microbial references for the development of saline-alkaline soil.

Enhancing boreal forest resilience: A four-year impact of biochar on soil quality and fungal communities

Boreal forests commonly suffer from nutrient deficiency due to restricted biological activity and decomposition. Biochar has been used as a promising strategy to improve soil quality, yet its impacts on forest soil microbes, particularly in cold environment, remains poorly understood. In this study, we investigated the effects of biochar, produced at different pyrolysis temperatures (500 °C and 650 °C) and applied at different amounts (0.5 kg·m−2 and 1.0 kg·m−2), on soil property, soil enzyme activity, and fungal community dynamics in a boreal forest over a span of two to four years. Our results showed that, four-year post-application of biochar produced at 650 °C and applied at 1.0 kg·m−2, significantly increased the relative abundance of Mortierellomycota and enhanced fungal species richness, α-diversity and evenness compared to the control (CK) (P < 0.05). Notably, the abundance of Phialocephala fortinii increased with the application of biochar produced at 500 °C and applied at 0.5 kg·m−2, exhibiting a positively correlation with the carbon cycling-related enzyme β-cellobiosidase. Functionally, distinct fungal gene structures were formed between different biochar pyrolysis temperatures, and between application amounts in four-year post-biochar application (P < 0.05). Additionally, correlation analyses revealed the significance of the duration post-biochar application on the soil properties, soil extracellular enzymes, soil fungal dominant phyla, fungal community and gene structures (P < 0.01). The interaction between biochar pyrolysis temperature and application amount significantly influenced fungal α-diversity (P < 0.01). Overall, these findings provide theoretical insights and practical application for biochar as soil amendment in boreal forest ecosystems.

Responses of soil bacterial and fungal community structure and functions to different plant species in a mixed forest plantation in a semi-arid region of China

Soil microbial communities play important roles in biogeochemical cycles and ecosystem functioning. However, how tree and grass species affect soil microbial structure and functions in semi-arid regions is poorly understood. In this study, we used sequencing of representative genes and bioinformatic analysis to compare the soil bacterial and fungal communities and their functional groups among legume and non-legume broad-leaved plantations, conifer plantations, artificial grassland, and adjacent farmland in a semi-arid area of Northern China. We found that soil bacterial diversity and abundance were significantly increased by legume plantations, especially Robinia pseudoacacia, compared to farmland. Soil bacterial communities were dominated by K-strategists in plantation soils and by r-strategists in farmland and grassland soils. Soil fungal communities showed contrasting patterns between Ascomycetes and Basidiomycota across treatments, and the highest abundance of nutrient indicator taxa Mortierellomycota was observed in plantation soils. Functional analysis revealed that afforestation altered the capacity of soil bacteria for organic compound mineralization and C and N cycling, and that different plant species enriched different types of ectomycorrhizal and saprophytic fungi. Moreover, we found that plantations and grasslands formed more connected but less complex and stable bacterial co-occurrence networks than farmland, and that conifer plantations increased the closeness of soil fungal networks compared to broad-leaved plantations. Our results suggest that tree and grass species play a key role in shaping the soil microbial community structure and functional groups, and provide valuable information for choosing suitable tree and grass species for ecosystem restoration and management in semi-arid regions.

Contrasting fungal functional groups influence nutrient cycling across four Japanese cool-temperate forest soils

Understanding soil dynamics and nutrient cycling is crucial for the sustainable management of Japanese forests covering 70 % of the national land area. These forests are dominated by tree species with contrasting traits, influencing soil dynamics differently. We investigated how changes in soil characteristics across different forest stands shift in composition and functioning of fungal communities. Four different forest stands dominated by two different mycorrhizal types were selected: Fagus crenata and Larix kaempferi, representing ectomycorrhizal (ECM) types, and Cryptomeria japonica and Robinia pseudoacacia, representing arbuscular mycorrhizal (AM) types. In total, 62 composite topsoil samples from two depths were analyzed for their physicochemical properties and fungal communities were profiled by DNA sequencing. Ectomycorrhizal fungi dominated soils of Fagus crenata and Larix kaempferi forests, while fungal saprotrophs were more abundant in Cryptomeria japonica and Robinia pseudoacacia forests. Forest stand type rather than soil depth determined the composition and structure of soil fungal communities. Soil pH was positively correlated with abundances of saprotrophic fungi (P < 0.05) and negatively with ECM fungi. Soil C:N ratio was positively correlated, and nitrate was negatively correlated with relative abundances of root-associated fungi, primarily ECM fungi. No links between C nor N stocks with fungal guilds were found across the dataset. Observed links between soil C:N ratio and relative abundances of root-associated fungi and saprotrophs stress the importance of these guilds for influencing nutrient cycling economy across contrasting forest types. The lack of correlation between fungal communities and soil C and N stocks suggests distinct mechanisms driving stocks in these soils.

An Earthworm Peptide Alters Soil Nematode, Microbial, and Nutrient Dynamics: A Novel Mechanism of Soil Food Web Feedbacks

Earthworms are soil macrofauna that control soil ecosystems by strongly influencing soil nematodes, microorganisms, and nutrient cycling, as well as soil environmental factors. We have discovered an earthworm cyclic peptide that disrupts nematode DNA, affecting its lifespan, reproduction, and feeding preferences. To investigate the effects of this peptide on soil, it was added to soil, and changes in soil nematode, bacterial and fungal communities, soil nutrient contents, and basal respiration were measured on days 5 and 21. The results showed that the peptide reduced soil basal respiration on day 5 and soil NO3-N on day 21, decreased soil fungivores nematodes on day 5 and soil nematode abundance on day 21, and increased soil fungal community richness and diversity. It also altered the soil bacterial community structure between day 5 and the soil fungal community structure on days 5 and 21. The peptide regulates the soil environment by influencing the structure of soil bacterial and fungal communities through the soil nematode community, as demonstrated by partial least squares path modelling (PLS-PM) analyses. Earthworm cyclic peptides mediates tri-trophic interactions between earthworms, nematodes, microbes, and environmental factors, providing new insights into soil biota interactions and feedback in dynamic soil food webs.

PFOA and PFOS induces mineralization of soil organic carbon by accelerating the consumption of dissolved organic carbon

The wide spread of Perfluoroalkyl substances (e.g., PFOA and PFOS) in soil can alter native soil geochemical properties and microbial communities through various approaches. Nonetheless, it is unclear that how PFOA and PFOS in soil affect the mineralization of soil organic carbon (SOC). A laboratory-scale culture experiment (180 d) was performed to explore the contribution of PFOA and PFOS to SOC dynamics and soil geochemical processes. Results showed that positive priming effect (PE) occurred in the short term (i.e., 30 days after PFOA and PFOS addition) and subsequently shifted to a slightly negative PE (i.e., 90 days) and remained in a significant negative PE thereafter (i.e., > 180 days). The PEs caused by PFOA and PFOS both accelerated the consumption of dissolved organic carbon (DOC), resulting in a significant increase in SOC mineralization (10.45%–127.36%) under the short-term incubation (30 d). As for long-term exposure (> 90 d), the organic carbon mineralization rate was significantly reduced (58.30%–65.24%) due to the excessive DOC consumption in the initial stage. High throughput analysis indicated that both PFOA and PFOS changed soil bacterial and fungal community structures, altered the relevant metabolic pathways and resulted in the enrichment of specific taxa.Graphical

Response of Soil Fungal Community to Biochar Application Under Different Irrigation Water Salinity

Saline water irrigation can alleviate the shortage of freshwater resources in the northwest arid zone, but long-term saline water irrigation can damage the soil fungal community structure. To alleviate the harm caused by salinity, biochar is used as a soil amendment to improve the soil fungal community structure. To investigate the intrinsic link between biochar application and the structural diversity of fungal communities in saline soils, two irrigation water salinity levels were set:0.35 dS·m-1 (fresh water) and 8.04 dS·m-1 (saline water). At each irrigation water salinity, two levels of biochar application were set:0 t·hm-2 (no application) and 3.7 t·hm-2 (application). High-throughput sequencing results showed that compared to that under fresh water irrigation, saline water irrigation increased fungal community species diversity and decreased fungal community species richness; biochar application under saline water irrigation reduced soil fungal community species diversity and species richness. The dominant fungal phyla in the soils of each treatment were Ascomycota, Mortierellomycota, Basidiomycota, Chytridiomycota, Glomeromycota, Rozellomycota, and Cysticercales, and the dominant genera were Gibberella, Chaetomium, Sarocladium, Stachybotrys, and Fusarium. Compared to that under freshwater irrigation, saline water irrigation significantly increased the relative abundance of Basidiomycota and Chytridiomycota and significantly decreased the relative abundance of Ascomycota and Rozellomycota. The application of biochar under saline irrigation significantly increased the relative abundance of Ascomycota and Sarocladium but significantly decreased the relative abundance of Basidiomycota, Chaetomium, and Fusarium. LEfSe analysis showed that under the condition of no biochar application, saline irrigation reduced the number of potential biomarkers of fungal communities, whereas the application of biochar under the condition of saline irrigation increased the number of potential biomarkers of fungal communities. These results indicated that the application of biochar can improve the saline soil environment and fungal community structure and provide a theoretical basis for reasonable brackish water irrigation and soil fertilization in arid areas.

Response Characteristics of Soil Fungal Community Structure to Long-term Continuous Cropping of Pepper

This study aimed to clarify the effect of long-term continuous cropping of pepper on soil fungal community structure, reveal the mechanism of continuous cropping obstacles, and provide a theoretical basis for the ecological safety and sustainable development of pepper industry. We took the pepper continuous cropping soil in the vegetable greenhouse planting base of Tongren City as the research object. The diversity and community structure of fungi in farmland soil were analyzed using Illumina MiSeq high-throughput sequencing, the responses of soil physio-chemical properties and fungal community characteristics to long-term continuous pepper cropping were discussed, and the relationships between the characteristics of fungal community structure and environmental factors were determined using CCA and correlation network analysis. The results showed that with the extension of pepper continuous cropping years, the soil pH value and organic matter (OM) content decreased, total phosphorus (TP) and available phosphorus (AP) contents increased, hydrolyzed nitrogen (AN) and available potassium (AK) contents decreased first and then increased, and total nitrogen (TN) and total potassium (TK) contents did not change significantly. Long-term continuous cropping decreased the Chao1 index and observed species index and decreased the Shannon index and Simpson index. The change in continuous cropping years had a significant effect on the relative abundance of soil fungal dominant flora. At the phylum level, the relative abundance of Mortierellomycota decreased with the extension of pepper continuous cropping years, the relative abundance of Ascomycota increased first and then decreased, and the relative abundance of Basidiomycota decreased first and then increased. At the genus level, with the increasing of pepper continuous cropping years, the relative abundance of Fusarium increased, and the relative abundance of Mortierella and Penicillium decreased. In addition, long-term continuous cropping simplified the soil fungal symbiosis network. CCA analysis indicated that pH, OM, TN, AN, AP, and AK were the driving factors of soil fungal community structure, and correlation network analysis showed that pH, OM, TN, TP, TK, AN, AP, and AK were the driving factors of soil fungal community structure, including Fusarium, Lophotrichus, Penicillium, Mortierella, Botryotrichum, Staphylotrichum, Plectosphaerella, and Acremonium. In conclusion, continuous cropping changed the soil physical and chemical properties, affected the diversity and community structure of the soil fungal community, changed the interaction between microorganisms, and destroyed the microecological balance of the soil, which might explain obstacles associated with continuous cropped pepper.

Response of soil microbial properties in the life cycle of potatoes to organic substitution regimes in North China

Organic substitution regimes (OSR) can improve soil microbial properties in crop production. However, the dynamic responses of soil microbial flora and its driving factors in crop growth periods under OSR are still unclear. Here, a 2-year fertilizer field trial was conducted to explore the response of soil chemistry and microbial properties to OSR in the life cycle of potatoes (Solanum tuberosum L.) in North China. The fertilization treatment included only mineral fertilizer (ConN), substitution of 30% mineral N with organic N (M30), and substitution of 60% mineral N with organic N (M60); soil samples were collected and analyzed in the early flowering period (EP), flowering period (MP), and maturity period (LP) of potatoes. As the potato growth period progressed, M30 and M60 increased soil organic carbon (SOC) compared with ConN. Soil total nitrogen (pH) increased (decreased) under M60, while soil available potassium was similar across three treatments. Compared with ConN, soil bacterial Chao, Shannon, and PD indexes significantly increased under M30 and M60 in MP and LP, and the fungal Shannon index increased in LP. Soil bacterial and fungal community structures under M30 and M60 were only significantly separated from ConN in MP. The bacterial communities of dominant phyla or genera responded more sensitively to the fertilization regimes than the flora of fungi, and the changes in the flora mainly occur in EP and MP. Redundancy analysis revealed that SOC and available phosphorus were highly correlated with soil microbial community compositions. Compared with ConN, soil microbial network complexity and fungal network robustness were higher under M30 and M60, while bacterial network robustness showed the opposite trend. Partial least squares pathway modeling showed that the conversion in the fertilization system or growth period had a direct positive effect on the C/N-cycling function of soil bacteria, but not on the C-degradation function of fungi. Our research has important theoretical implications for the development of microbial fertilizers and fertilization management in different potato growth periods.

Forest management and former land use have no effect on soil fungal diversity in uneven-aged mountain high forests

Key messageMetabarcoding analysis of soil fungal communities in French mountain forests revealed that harvesting intensity, time since last harvest and former land use had no effect on fungal community composition compared to key abiotic factors. Low-intensity management in these uneven-aged mountain forests therefore has limited effects on soil fungal community composition which is mainly driven by elevation and edaphic properties.ContextPast and current human activities are known to affect forest biodiversity. However, the effects of former land use and forest management have been studied much more extensively on higher plants than on fungi.AimsOur objectives were to assess the effects of harvesting intensity, duration since last harvest and former land use on soil fungal communities in uneven-aged mountain high forests.MethodsOn the basis of historical land-use maps drawn between 1862 and 1864 and on historical forest management archives, we selected 62 sites in the French Alps with contrasting land-use histories (ancient forests, which were already forested on historical maps vs recent forests, which have recovered following abandonment of pastures) and different durations since last harvest (from 1 to over 50 years). We carried out soil sampling and assessed fungal diversity by metabarcoding analysis. We analysed soil fungal molecular operational taxonomic units (MOTU) diversity as a whole and for the main lifestyle groups (such as wood saprotrophic or ectomycorrhizal fungi) using multiple linear regressions on Shannon’s diversity index and fungal taxonomic composition using canonical correlation analysis.ResultsWe found no significant effect of harvesting intensity, time since last harvest or land-use history on total fungal MOTU diversity, fungal lifestyle diversity or taxonomic composition. In contrast, we observed significant effects of elevation, pH, organic carbon and available phosphorus content on the taxonomic and functional composition of soil fungal communities.ConclusionsThe structure of soil fungal communities (i.e. diversity and species composition) was mainly determined by elevation and edaphic factors, indicating a high-context dependency, as previously found in similar studies. Our study in mountain forests shows that recent forests established on former pastures had no legacy effect on soil conditions and fungal communities, in contrast to previous results in lowland areas, where recent forests were mainly established on former cropland. Uneven-aged forest management had no effect on fungal diversity, in contrast to previous results observed in even-aged high forests.

Effect of fire and post-fire management on soil microbial communities in a lower subtropical forest ecosystem after a mountain fire

Wildfires and post-fire management exert profound effects on soil properties and microbial communities in forest ecosystems. Understanding microbial community recovery from fire and what the best post-fire management should be is very important but needs to be sufficiently studied. In light of these gaps in our understanding, this study aimed to assess the short-term effects of wildfire and post-fire management on both bacteria and fungi community composition, diversity, structure, and co-occurrence networks, and to identify the principal determinants of soil processes influencing the restoration of these communities. Specifically, we investigated soil bacterial and fungal community composition, diversity, structure, and co-occurrence networks in lower subtropical forests during a short-term (<3 years) post-fire recovery period at four main sites in Guangdong Province, southern China. Our results revealed significant effects of wildfires on fungal community composition, diversity, and co-occurrence patterns. Network analysis indicated reduced bacterial network complexity and connectivity post-fire, while the same features were enhanced in fungal networks. However, post-fire management effects on microbial communities were negligible. Bacterial diversity correlated positively with soil microbial biomass nitrogen, soil organic carbon, and soil total nitrogen. Conversely, based on the best random forest model, fungal community dynamics were negatively linked to nitrate-nitrogen and soil water content. Spearman's correlation analysis suggested positive associations between bacterial networks and soil factors, whereas fungal networks exhibited predominantly negative associations. This study elucidates the pivotal role of post-fire management in shaping ecological outcomes. Additionally, it accentuates the discernible distinctions between bacterial and fungal responses to fire throughout a short-term recovery period. These findings contribute novel insights that bear significance in evaluating the efficacy of environmental management strategies.
