Digital Image Analysis as a Technique for Alewife Fecundity Estimation in a New Hampshire River

AbstractFecundity estimates are important in fisheries management to determine the reproductive potential and spawning stock biomass of a population. Traditional methods for determining these estimates, however, have been time consuming and tedious, often requiring long‐term sample storage in toxic preservatives. Recently, studies incorporated the use of scanners and image analysis software to produce equally reliable fish fecundity estimates. This study was undertaken to demonstrate the application of this technique with Alewife Alosa pseudoharengus, “a species of concern.” Ovaries were collected from fish ranging in size from 25 to 34 cm throughout their annual spawning migration (Newmarket, New Hampshire) and used to compare fecundity estimated gravimetrically following long‐term storage and by use of a flatbed scanner. Gonadosomatic indices were calculated throughout the collection period but were not found to differ over time. Comparisons within ovaries demonstrated that oocyte size and number did not differ among anterior, center, and posterior sections. Fecundity estimates did not differ between the two methods and increased with fish age and size ranging from 121,000 and 130,200 oocytes/female (25 cm) to 332,500 and 364,900 oocytes/females (34 cm), respectively. These results demonstrate that fecundity estimates using digital image analysis software were equally reliable as those produced by traditional methods but were generated more quickly and with less reliance on toxic preservatives.

Measuring Stolons and Rhizomes of Turfgrasses Using a Digital Image Analysis System.

Length and diameter of stolons or rhizomes are usually measured using simple rulers and calipers. This procedure is slow and laborious, so it is often used on a limited number of stolons or rhizomes. For this reason, these traits are limited in their use for morphological characterization of plants. The use of digital image analysis software technology may overcome measurement errors due to human mistakes, which tend to increase as the number and size of samples also increase. The protocol can be used for any kind of crop but is particularly suitable for forage or grasses, where plants are small and numerous. Turf samples consist of aboveground biomass and an upper soil layer to the depth of maximum rhizome development, depending on the species of interest. In studies, samples are washed from the soil, and stolons/rhizomes are cleaned by hand before analysis by digital image analysis software. The samples are further dried in a laboratory heating oven to measure dry weight; therefore, for each sample, the resultant data are total length, total dry weight, and average diameter. Scanned images can be corrected before analysis by excluding visible extraneous parts, such as remaining roots or leaves not removed with the cleaning process. Indeed, these fragments normally have much smaller diameters than stolons or rhizomes, so they can be easily excluded from analysis by fixing the minimum diameter below which objects are not considered. Stolon or rhizome density per unit area can then be calculated based on sample size. The advantage of this method is quick and efficient measurement of the length and average diameter of large sample numbers of stolons or rhizomes.

Shaping Ability of ProTaper Universal, ProTaper NEXT and WaveOne Primary in Severely Curved Resin Blocks

Complex anatomy of roots requires endodontic rotary instruments that respect and follow that anatomy. The purpose of this study was to compare the shaping ability of three engine-driven nickel-titanium instruments in severely curved simulated canals. Forty-eight severely curved simulated canals in resin blocks were prepared to an apical size of 25 using the following systems (n = 16 per group): ProTaper Universal (PTU), ProTaper NEXT (PTN), and WaveOne Primary (WO) (all Dentsply Maillefer, Ballaigues, Switzerland). Composite images were made from the superimposition of pre- and post-instrumentation images. The amount of resin removed by each system was measured by using a digital template and image analysis software. Shaping ability of each system was compared using different parameters: total resin removal, centering ability, canal transportation in the apical, middle and coronal third of canal. Preparation times were also recorded. The data were statistically analyzed by using analysis of variance, paired t-test, and Tukey's post hoc test. Canals prepared with PTN were better centered in the apical part than those prepared with WO and PTU (p < 0.05). WO removed significantly more resin at the outer aspect of the maximum point of curvature. WO and PTU caused similar canal transportations at 7 points out of 11 measuring points. At measuring points 3, 4, and 5 WO caused more canal straightening than PTU. Instrumentation with PTU required more time than with the two other instruments (p <0.05). No preparation error or instrument separation occurred. In the apical part of the canals PTN obtained the best results with regard to canal transportation. All of the rotary instruments were safe in preparing severely curved canals and PTN showed better preparation of apical part of the canal.

Clinimetric analysis of recently applied quantitative tools in evaluation of vitiligo treatment.

Vitiligo affects about 1% of the world's population, however, there is currently no universally used standardized measure to assess its response to treatment. To find the most effective technique for the quantitative assessment of therapeutic results in vitiligo patients. The study was performed in three stages: (1) Conducting an adapted Delphi survey to check current dermatologists' attitudes regarding the topic of study. (2) Conducting a pilot study that involves testing the selected digital image analysis software in the laboratory to validate future tasks. (3) The chief clinimetric study that implicates selecting actual vitiligo lesion models and evaluating them. Regarding the surface area measuring techniques, the most accurate results were gained through the digital image analysis for surface area, followed by point-counting technique. The digital image analysis for color measurement was accurate and reliable in getting a percentage representation of color improvement within the vitiligo lesions, in response to therapy. Many dermatologists lack understanding of basic concepts about imaging techniques. The study does not include a traditional assessment method such as vitiligo area scoring index. Our designated digital image analysis technique was able to efficiently assess the changes that occur both on surface area and the color of vitiligo lesions in response to therapy.

Digital Microscopy, Image Analysis, and Virtual Slide Repository.

Advancements in technology and digitization have ushered in novel ways of enhancing tissue-based research via digital microscopy and image analysis. Whole slide imaging scanners enable digitization of histology slides to be stored in virtual slide repositories and to be viewed via computers instead of microscopes. Easier and faster sharing of histologic images for teaching and consultation, improved storage and preservation of quality of stained slides, and annotation of features of interest in the digital slides are just a few of the advantages of this technology. Combined with the development of software for digital image analysis, digital slides further pave the way for the development of tools that extract quantitative data from tissue-based studies. This review introduces digital microscopy and pathology, and addresses technical and scientific considerations in slide scanning, quantitative image analysis, and slide repositories. It also highlights the current state of the technology and factors that need to be taken into account to insure optimal utility, including preanalytical considerations and the importance of involving a pathologist in all major steps along the digital microscopy and pathology workflow.

Sunscreen application to the face persists beyond 2 hours in indoor workers: an open-label trial

Background: The American Academy of Dermatology recommends reapplication of sunscreen every two hours for adequate sun protection when outdoor. However, the frequency of reapplication needed to achieve adequate protection in indoor workers remains unknown.Objective: To investigate the persistence of sunscreen applied once in the morning on the face of indoor workers throughout a normal 8-hour workday.Methods: This open-label trial included 20 healthy volunteers who work indoors. Volunteers applied 1 g of sunscreen (2 mg/cm2) mixed with 2% invisible blue fluorescent agent on the face in the morning. Photographs were taken by VISIA-CR booth in UV mode at 8 am and then every 2 hours thereafter until 4 pm with limited outdoor activity less than 1 hour. Six areas of the face were analyzed using digital image analysis software. The primary outcome was the total amount of sunscreen diminution during the 8-hour study period.Results: The amount of sunscreen decreased with mean peak reduction of 16.3% at 2 hours, and minimal reduction thereafter. Total sunscreen reduction was 28.2% at the end of the 8-hour day.Limitations: Small study population.Conclusion: For indoor workers who applied adequate amount of sunscreen once in the morning, reapplication of sunscreen may be unnecessary.

Morphometric properties of the facial nerve in fetal temporal bones

ObjectivesThe main aim of the study was to examine the development and course of the facial nerve within fetal temporal bones from an anatomical and neuro-otological perspective. MethodsThe study was conducted on 32 temporal bones from obtained fetuses (7 females, 9 male), on a mean gestational age of 26.75 ± 4.36 (range, 20–34) weeks from the collection of the Anatomy Department of Medicine Faculty. All the measurements were collected with a digital image analysis software. ResultsNeither male/female nor right/left significant differences were observed in relation with the algebraic data of the segment lengths and angles of the facial nerve (p > 0.05). Linear functions for meatal, labyrinthine, tympanic, and mastoid segment lengths of the facial nerve were calculated as: y = −1.206 + 0.200 × Age (weeks), y = −1.868 + 0.153 × Age (weeks), y = −2.327 + 0.325 × Age (weeks), and y = −1.507 + 0.246 × Age (weeks), respectively. In addition, linear functions for first and second genu angles were calculated as: y = 105.475–0.117 × Age (weeks) and y = 140.446–0.042 × Age (weeks), respectively. ConclusionThe regression equations and the scatter plot with increment curve, representing the growth dynamics of the facial nerve can be used for estimating its lengths and for understanding its development. The data suggest that there is a dramatic change transition from fetal life to the gathered data of adulthood in the length of meatal and mastoid segments as well as in the second genu angle; in addition, there is a partial change in the length of labyrinthine and tympanic segments as well as in the first genu angle.

Effects of different resin sealing therapies on nanoleakage within artificial non-cavitated enamel lesions.

The aim of this study was to evaluate nanoleakage within the different lesion-sealing therapies applied to artificial non-cavitated enamel lesions. Thirty-two human anterior teeth were used. Artificial subsurface enamel lesions were produced on the labial surfaces of teeth. The specimens were then randomly divided into three groups (n=10): Group I- Clinpro Sealant application; Group II- ExciTE F adhesive resin application; and Group III- ICON resin infiltrant application. Each group was further divided into two subgroups: control and thermocycler. Nanoleakage was calculated by the digital image analysis software. In the control and thermocycled groups, there was no statistically significant difference between the Groups I, II, and III (p>0.05). The only significant leakage scores were obtained between the Group III control and thermocycler groups (p=0.027). ICON infiltrant can be used as an alternative to dental adhesives and fissure sealants in the sealing of initial non-cavitated enamel lesions. But the resin may become more affected by the water sorption than other resin materials over time. More studies are needed to evaluate long-term durability of resin infiltrants.

Quantitative and Neurovascular Anatomy of the Growing Gracilis Muscle in the Human Fetuses.

This study is intended to obtain the algebraic growth dynamics of the gracilis in fetuses and determine the variations of neurovascular pedicle(s) of the gracilis, to aid infant surgeries. Forty fetuses (19 males and 21 females) were included in the study. Gestational mean age of the fetuses was 22.40 ± 2.67 (range, 18-28) weeks. Numerical values were obtained using a digital caliper and a digital image analysis software. Linear functions for the surface area, width, anterior, and posterior margin lengths of the gracilis were calculated, respectively, as: y = -289.307 + 20.501 × age (weeks), y = -7.113 + 0.622 × age (weeks), y = -24.421 + 3.434 × age (weeks), and y = -24.397 + 3.314 × age (weeks). In addition, length and width of the gracilis tendon were calculated as y = -6.676 + 0.934 × age (weeks) and y = -0.732 + 0.074 × age (weeks), respectively. Parameters of the gracilis had no statistically significant difference regarding side and sex (P > 0.05). In all the specimens, the gracilis was innervated by the anterior branch of the obturator nerve. Blood supply of the gracilis was identified to be derived from 1 single artery in 38 sides of total 80 (47.5%), from 2 arteries in 36 (45%) and from 3 arteries in 6 (7.5%). In 74 sides (92.5%), the nerve was superficial to the main artery, whereas in 6 sides (7.5%), it was deeper. The data of the present study could be beneficial for surgeons in infant surgeries to treat conditions such as obstetrical brachial plexus paralysis, facial palsy, or anal incontinence. Linear functions can be utilized to better evaluate the growth course of the gracilis in fetuses and to predict the dimensions thereof. Additionally, comprehending the structure and recognizing the variations of the gracilis nerves and arteries can help to protect the neurovascular pedicle(s) of the gracilis during the operations.

In vivo regeneration functionalities of experimental organo-biomaterials containing water-soluble nacre extract

BackgroundNovel multifunctional biomaterials were recently designed to allow for an optimized tissue regeneration process. PurposeTo comprehensively assess (photographic, radiographic and histological) the in vivo functionality of demineralized bovine bone matrix (DBM) associated with an experimental marine organic extract (MOE) from nacre in a sheep ectopic grafting model. Materials and methodsSynthesis of MOE was based on mixing powdered nacre (0.05 g, particles average size <0.1 mm) with acetic acid (5 mL, pH 7) under constant stirring for 72 hours (25 °C). Polyethylene tubes (3/animal, n = 4, diameter: 5.0 mm × length: 10.0 mm) from the control (empty) or experimental groups (DBM or DBM + MOE) were then intramuscularly implanted into the lumbar regions of sheep (n = 8, 2-years old, ≈45 kg). Animals were euthanized at 3 and 6 months to allow for the collection of tissue samples. Tissue samples were fixed in formalin 10% (buffered, 7 days) in preparation for photographic, radiographic and histological assessments. Acquired images were then analyzed using digital image analysis software to quantify the amount of neoformed tissues, whereas radiographic and histological analyses were performed to determine radiopacity and classification of tissues deposited inside of the tubes. ResultsPhotographic and radiographic analyses have shown that both pure (unaltered) and MOE-modified DBM were capable of depositing neoformed tissues (at 3 and 6 months), where higher levels of deposition and radiopacity were observed on groups treated with experimental materials. Histological results, however, demonstrated that tissues formed from both unaltered and MOE-modified DBM were only fibrous connective in origin. ConclusionsAs an ectopic grafting in sheep, the experimental organo-biomaterial association applied did not reveal any osteoinductive property but led to a fibrous tissue repair only.

Determination of flakiness index (FI) and shape index (SI) using open source digital image analysis software

The aim of the article was to verify the possibility of using open source digital image analysis software to determine the particle shape. Devices using laser diffraction or dynamic image analysis could be used to determine size and shape of particles. However, the laboratory devices for digital image analysis are relatively expensive. For this reason, it was decided to use open source digital image analysis software Fiji Is just ImageJ and mobile phone with a 12 Mpx camera. Coarse aggregates of basalt and granite were used in the study. Flakiness index (FI) and shape index (SI) were determined using two methods: in accordance with the European Standard and with the use of digital image analysis (reference method). Based on the obtained results it was found that there is an appropriate relationship between the European Standard method and the reference method.

Quantitative assessment of the growth dynamics of the teres major in human fetuses.

The main objective of the study was to examine the use of teres major (TM) as a flap in the pediatric surgeries from an anatomical perspective by: (1) revealing the growth dynamics of the developing TM; (2) assessing the possible interconnecting structures between TM and latissimus dorsi (LD); (3) exposing the innervation patterns of TM in human fetuses. Study was conducted on 50 fetuses (26 females and 24 males), on a mean gestational age of 22.86 ± 3.21 (range 18-30) weeks. All the measurements were collected with a digital caliper and a digital image analysis software. Additionally, structural relations between TM and LD were examined in detail and further classified. No significant difference of side and sex was detected on TM measurements (p > 0.05). Linear functions for the surface area, width, length of the superior and inferior margins of TM were detected as follows: y = - 257.142 + 18.334 × age (weeks), y = - 5.497 + 0.545 × age (weeks), y = - 1.621 + 1.068 × age (weeks), and y = - 2.147 + 1.284 × age (weeks), respectively. As classified in five types, a number of 33 muscular or tendinous connections between TM and LD were detected. Musculo-tendinous slips from TM to triceps brachii (TB) were evident in four sides. Innervation of TM was observed to be providing by the lower subscapular nerve in all the cases. First, linear functions, representing the developing fetal morphometry obtained by our study can be adapted for estimating the morphometric of this muscle in early childhood. Besides, acknowledging the diverse appointments of TM with the surrounding muscles such as LD and TB may facilitate the prevention of neurovascular structures and the application of surgical procedures during tendon transfers. In brief, our findings are highly potent to bring the attention of pediatric surgeons.

Definition and quantification of drapeability through the measurement of constituent effects

Although considerable work has been carried out in the field of drapeability research, both the definition of what drapeability comprises as well as the tools to measure and quantify it must be further developed. This paper presents a definition of drapeability that is meant to broaden its scope to include not only the important effects of shearing and wrinkling, but all constituent effects such as gap formation, undulations, loops (tow buckling) and fabric anisotropy. This definition is completed by an experimental test device to measure and quantify all of these effects in a single, standardized setup (available as the Textechno Drapetest).The Drapetest utilizes optic measurement technology coupled with digital image analysis software to detect and measure fibre positions and orientation, and fabric topology. For each of the defined drapeability effects characteristic values were generated from this optical data that can describe the intensity, distribution and extent of the effect in question. The device and methodology was empirically validated using a set of 72 specially manufactured glass fibre NCF. The interdependencies between textile design, process parameters and drapeability effects as well as a semi-analytical equation for them are shown. A method to use these semi-analytical equations in the composite design process is introduced [1].

Validation of the systematic scoring of immunohistochemically stained tumour tissue microarrays using QuPath digital image analysis.

Output from biomarker studies involving immunohistochemistry applied to tissue microarrays (TMA) is limited by the lack of an efficient and reproducible scoring methodology. In this study, we examine the functionality and reproducibility of biomarker scoring using the new, open-source, digital image analysis software, QuPath. Three different reviewers, with varying experience of digital pathology and image analysis, applied an agreed QuPath scoring methodology to CD3 and p53 immunohistochemically stained TMAs from a colon cancer cohort (n = 661). Manual assessment was conducted by one reviewer for CD3. Survival analyses were conducted and intra- and interobserver reproducibility assessed. Median raw scores differed significantly between reviewers, but this had little impact on subsequent analyses. Lower CD3 scores were detected in cases who died from colorectal cancer compared to control cases, and this finding was significant for all three reviewers (P-value range = 0.002-0.02). Higher median p53 scores were generated among cases who died from colorectal cancer compared with controls (P-value range = 0.04-0.12). The ability to dichomotise cases into high versus low expression of CD3 and p53 showed excellent agreement between all three reviewers (kappa score range = 0.82-0.93). All three reviewers produced dichotomised expression scores that resulted in very similar hazard ratios for colorectal cancer-specific survival for each biomarker. Results from manual and QuPath methods of CD3 scoring were comparable, but QuPath scoring revealed stronger prognostic stratification. Scoring of immunohistochemically stained tumour TMAs using QuPath is functional and reproducible, even among users of limited experience of digital pathology images, and more accurate than manual scoring.

Effects of Injection Molding Screw Tips on Polymer Mixing

In all fields of industry it is important to produce parts with good quality. Injection molded parts usually have to meet strict requirements technically and aesthetically. The aim of the measurements presented in our paper is to investigate the aesthetic appearance, such as surface color homogeneity, of injection molded parts. It depends on several factors, the raw material, the colorants, the injection molding machine and the processing parameters. In this project we investigated the effects of the injection molding machine on surface color homogeneity. We focused on injection molding screw tips and investigated five screw tips with different geometries. We produced flat specimens colored with a masterbatch and investigated color homogeneity. To evaluate the color homogeneity of the specimens, we used digital image analysis software developed by us. After that we measured the plastication rate and the melt temperature of the polymer melt because mixing depends on these factors. Our results showed that the screw tips (dynamic mixers) can improve surface color homogeneity but they cause an increase in melt temperature and a decrease in the plastication rate.

Morphometric properties of the levator scapulae, rhomboid major, and rhomboid minor in human fetuses

The main objective of this study is to showcase the growth dynamics of levator scapulae, rhomboid major, and rhomboid minor algebraically, to help plan surgeries on newborns and young infants. Twenty-five formalin-fixed fetuses (11 male-14 female) with a mean gestational age of 21.80 ± 2.61 (range 18-27) weeks present in the inventory of Mersin University Faculty of Medicine Anatomy Department were dissected. Surface area of levator scapulae, rhomboid major, and rhomboid minor was calculated using digital image analysis software; width and length parameters were measured using digital calipers. Neither sex nor side significant differences were observed in relation with the numerical data of levator scapulae, rhomboid major, and rhomboid minor (p > 0.05). Algebraic parameters such as surface area, width and length were detected to exhibit a linear growth from 18th to 27th week. Linear functions for levator scapulae, rhomboid major and rhomboid minor surface areas were obtained as y = - 136.871 + 10.598 × age (weeks), y = - 480.567 + 33.147 × age (weeks) and y = - 128.090 + 8.843 × age (weeks), respectively. The results and data of this study can potentially be beneficial in planning of surgeries for many infancy diseases such as trapezius paralysis, obstetrical brachial plexus palsy, Sprengel's disease, or winged scapula. Linear functions calculated in this study are expected to provide a better understanding of the growth dynamics of levator scapulae, rhomboid major, and rhomboid minor in fetal term and more precise estimation of muscle sizes.

Abstract P2-03-07: Digital image analysis of Ki67 in hot spots is superior to alternative proliferation associated markers in breast cancer

Abstract INTRODUCTION Proliferative activity is one of the most important prognostic parameters in cancer. During the pathological examination of breast tumors, it is routinely evaluated by a count of the number of mitoses. Adding immunohistochemical stains of the nuclear protein Ki67 provides extra prognostic and predictive information. However, the currently used methods for both of these evaluations battle imperfections, primarily in reproducibility. In this study, we make an equally broad and detailed evaluation of mitoses, Ki67 and the more recently described Phosphohistone H3 (PHH3) in primary breast cancer using digital image analysis (DIA). Furthermore, we aim to investigate the prognostic and predictive value of proliferation-associated biomarkers in breast cancer stromal cells in relation to patient outcome. MATERIALS AND METHODS Two cohorts of primary breast cancer specimens (total n=297) with clinicopathological data including &amp;gt;10 years survival data, were sectioned and stained for Ki67, PHH3 and pancytokeratin (CKMNF116) and all glass slides were digitally scanned at x20. The DIA software used was the Visiopharm Integrator System (VIS) by Visiopharm A/S, Hoersholm, Denmark. VIS operates by a 'digital fusion' method that automatically excludes non-epithelial tissue restricting the analysis of the biomarkers (Ki67 and PHH3) to CKMNF116 positive cells. Both manual and DIA scores were compared for sensitivity and specificity for the gene expression based Luminal B versus A subtype, for high versus low transcriptomic grade as well as for their prognostic value in terms of Cox regression hazard ratios and breast cancer specific and overall survival. Further, we investigated whether the expression of Ki67 in the tumors' hot spots, invasive edges or as an average across all regions should be assessed for maximum power in relation to these outcomes. In addition, by inverting the DIA algorithm run by the VIS on the same cohorts, the expression of Ki67 and PHH3 was evaluated in the tumor stromal compartment. RESULTS Regardless of tumor region, DIA of Ki67 outperformed the other markers in sensitivity and specificity for gene expression subtypes and transcriptomic grade. In contrast to mitotic counts, tumors with high expression of Ki67 as defined by DIA, had significantly increased hazard ratio for all-cause mortality within 10 years from diagnosis. DIA of Ki67 was superior to manual Ki67 and PHH3 evaluations as well as to mitotic counts in terms of separation of patients with poor versus relatively good survival. Finally, we replaced the manual mitotic counts with DIA of Ki67 in hot spots as the marker for proliferation when determining histological grade. This increased the differences in estimated mean overall survival between the highest and lowest grades and added significantly more prognostic information to the classic Nottingham histological grade. CONCLUSIONS We conclude that digital image analysis of Ki67 in hot spots should be suggested as the marker of choice for proliferative activity in breast cancer. Digital Radiography in Total Hip Arthroplasty: Technique and Radiographic Results.

Obtaining the ideal acetabular cup position in total hip arthroplasty remains a challenge. Advancements in digital radiography and image analysis software allow the assessment of the cup position during the surgical procedure. This study describes a validated technique for evaluating cup position during total hip arthroplasty using digital radiography. Three hundred and sixty-nine consecutive patients undergoing total hip arthroplasty were prospectively enrolled. Preoperative supine anteroposterior pelvic radiographs were made. Intraoperative anteroposterior pelvic radiographs were made with the patient in the lateral decubitus position. Radiographic beam angle adjustments and operative table adjustments were made to approximate rotation and tilt of the preoperative radiograph. The target for cup position was 30° to 50° abduction and 15° to 35° anteversion. Intraoperative radiographic measurements were calculated and final cup position was determined after strict impingement and range-of-motion testing. Postoperative anteroposterior pelvic radiographs were made. Two independent observers remeasured all abduction and anteversion angles. Of the cups, 97.8% were placed within 30° to 50° of abduction, with a mean angle (and standard deviation) of 39.5° ± 4.6°. The 2.2% of cups placed outside the target zone were placed so purposefully on the basis of intraoperative range-of-motion testing and patient factors, and 97.6% of cups were placed between 15° and 35° of anteversion, with a mean angle of 26.6° ± 4.7°. Twenty-eight percent of cups were repositioned on the basis of intraoperative measurements. Subluxation during range-of-motion testing occurred in 3% of hips despite acceptable measurements, necessitating cup repositioning. There was 1 early anterior dislocation. Placing the acetabular component within a target range is a critical component to minimizing dislocation and polyethylene wear in total hip arthroplasty. Using digital radiography, we positioned the acetabular component in our desired target zone in 97.8% of cases and outside the target zone, purposefully, in 2.2% of cases. When used in conjunction with strict impingement testing, digital radiography allows for predictable cup placement in total hip arthroplasty.

Semi-automated digital measurement as the method of choice for beta cell mass analysis.

Pancreas injury by partial duct ligation (PDL) activates beta cell differentiation and proliferation in adult mouse pancreas but remains controversial regarding the anticipated increase in beta cell volume. Several reports unable to show beta cell volume augmentation in PDL pancreas used automated digital image analysis software. We hypothesized that fully automatic beta cell morphometry without manual micrograph artifact remediation introduces bias and therefore might be responsible for reported discrepancies and controversy. However, our present results prove that standard digital image processing with automatic thresholding is sufficiently robust albeit less sensitive and less adequate to demonstrate a significant increase in beta cell volume in PDL versus Sham-operated pancreas. We therefore conclude that other confounding factors such as quality of surgery, selection of samples based on relative abundance of the transcription factor Neurogenin 3 (Ngn3) and tissue processing give rise to inter-laboratory inconsistencies in beta cell volume quantification in PDL pancreas.

Reduced angiogenic factor expression in intrauterine fetal growth restriction using semiquantitative immunohistochemistry and digital image analysis.

To localize, quantify and compare angiogenic factors, vascular endothelial growth factor (VEGF), placental growth factor (PlGF), as well as their receptors fms-like tyrosine kinase receptor (Flt-1) and kinase insert domain receptor (KDR) in the placentas of normal pregnancy and complications of preeclampsia (PE), intrauterine fetal growth restriction (IUGR) and PE + IUGR. In a prospective cross-sectional case-control study, 30 pregnant women between 24-40 weeks of gestation, were recruited into four clinical groups. Representative placental samples were stained for VEGF, PlGF, Flt-1 and KDR. Analysis was performed using semiquantitative methods and digital image analysis. The overall VEGF and Flt-1 were strongly expressed and did not show any conclusive difference in the expression between study groups. PlGF and KDR were significantly reduced in expression in the placentas from pregnancies complicated by IUGR compared with normal and preeclamptic pregnancies. The lack of PlGF and KDR may be a cause for the development of IUGR and may explain the loss of vasculature and villous architecture in IUGR. Automated digital image analysis software is a viable alternative method to the manual reading of placental immunohistochemical staining.
