A M�ssbauer spectroscopy study of cellular acquisition of iron from pyoverdine byPseudomonas aeruginosa

Mössbauer spectroscopy was used to investigate the cellular acquisition of iron by Pseudomonas aeruginosa which had been incubated with ferripyoverdine for 20, 40, 60, 120 or 360 min. Studies revealed that no ferripyoverdine accumulated in the cells at any of these times and that the amounts and kinds of iron complexes produced by cellular metabolism vary with time. At 20 and 40 min a ferric species, with isomer shift delta = 0.38-0.42 mm/s and quadrupole splitting delta EQ = 0.94-0.92 mm/s, was the major iron metabolite comprising approximately 80% of the iron. At later times at least three other ferric species appeared with delta = 0.54----0.72, delta EQ = 0.84----1.07 mm/s. Ferrous species, delta = 1.43----1.77 mm/s and delta EQ = 2.69----1.82 mm/s, were also seen at times as early as 20 min and comprised as much as 17% of the total iron at 20 and 40 min. The parameters of all these species identify them as being six-coordinated high-spin complexes. In addition a low-spin species, delta = 0.19 mm/s delta EQ = 0.67----0.91 mm/s, never before reported in cells, appeared at 60, 120, and 360 min as one of the major iron metabolites (50% or more). All isomer shifts are measured with respect to natural iron.

A six-coordinate high spin protoporphyrin IX iron(III) complex as a model for ferric haemproteins: Mössbauer spectrum of bisaquo(protoporphyrinato IX)iron(III) encapsulated in aqueous detergent micelles

The Mössbauer spectrum is reported for a six- coordinate high spin iron(III) protoporphyrin complex, [(PPIX)Fe(H 2O) 2] +, which is monodispersed in an aqueous frozen solution of sodium dodecyl sulphate micelles. The isomer shift of 0.46 mm −1 and the quadrupole splitting of 1.19 mm 1̄ are similar to those of aquo met myoglobin. These results are compared with the literature results on other high spin six-coordinated models and with those of some proteins. This is the first report of a Mössbauer study on a haem encapsulated in a detergent micelle.

Studies of the reactions of ferric iron with glutathione and some related thiols. Part V. Solid complexes containing Fe II and glutathione or Fe III with oxidized glutathione

A number of polymeric iron(III) oxidised glutathione materials have been prepared. Mössbauer spectroscopic, magnetic susceptibility data and electronic reflectance spectra studies on these iron(III) materials are discussed. Two stoichiometries 1:1 and 1:2 have been established for iron(II)-glutathione materials. Spectroscopic studies show that the 1:1 materials contain high spin iron(II) in distorted five and six coordinated environments, whereas the 1:2 materials contain only distorted six-coordinated high spin iron(II). The nature of the coordinating ligands are discussed.

The equilibrium spatial configuration of ligands around a central ion in six-coordinated high-spin (ionic) complexes with electron configurations dn (n=1,2,...10) in the approximation of the crystal field theory has been determined theoretically. Formulas have been derived for all cases whereby it is possible to determine the deviations of the ligand positions from a regular octahedron (internal asymmetry), and formulas for additional splitting of electronic terms due to internal asymmetry. The results of applications of the theory so investigations of the physicochemical properties of complexes will be given in future communications.
