Chromosomal single-strand break repair and neurological disease: Implications on transcription and emerging genomic tools

Cells are constantly exposed to various sources of DNA damage that pose a threat to their genomic integrity. One of the most common types of DNA breaks are single-strand breaks (SSBs). Mutations in the repair proteins that are important for repairing SSBs have been reported in several neurological disorders. While several tools have been utilised to investigate SSBs in cells, it was only through recent advances in genomics that we are now beginning to understand the architecture of the non-random distribution of SSBs and their impact on key cellular processes such as transcription and epigenetic remodelling. Here, we discuss our current understanding of the genome-wide distribution of SSBs, their link to neurological disorders and summarise recent technologies to investigate SSBs at the genomic level.

Electromagnetic fields act via activation of voltage‐gated calcium channels to produce beneficial or adverse effects

The direct targets of extremely low and microwave frequency range electromagnetic fields (EMFs) in producing non-thermal effects have not been clearly established. However, studies in the literature, reviewed here, provide substantial support for such direct targets. Twenty-three studies have shown that voltage-gated calcium channels (VGCCs) produce these and other EMF effects, such that the L-type or other VGCC blockers block or greatly lower diverse EMF effects. Furthermore, the voltage-gated properties of these channels may provide biophysically plausible mechanisms for EMF biological effects. Downstream responses of such EMF exposures may be mediated through Ca2+/calmodulin stimulation of nitric oxide synthesis. Potentially, physiological/therapeutic responses may be largely as a result of nitric oxide-cGMP-protein kinase G pathway stimulation. A well-studied example of such an apparent therapeutic response, EMF stimulation of bone growth, appears to work along this pathway. However, pathophysiological responses to EMFs may be as a result of nitric oxide-peroxynitrite-oxidative stress pathway of action. A single such well-documented example, EMF induction of DNA single-strand breaks in cells, as measured by alkaline comet assays, is reviewed here. Such single-strand breaks are known to be produced through the action of this pathway. Data on the mechanism of EMF induction of such breaks are limited; what data are available support this proposed mechanism. Other Ca2+-mediated regulatory changes, independent of nitric oxide, may also have roles. This article reviews, then, a substantially supported set of targets, VGCCs, whose stimulation produces non-thermal EMF responses by humans/higher animals with downstream effects involving Ca2+/calmodulin-dependent nitric oxide increases, which may explain therapeutic and pathophysiological effects.

Repair of DNA Strand Breaks in a Minichromosome In Vivo: Kinetics, Modeling, and Effects of Inhibitors

To obtain an overall picture of the repair of DNA single and double strand breaks in a defined region of chromatin in vivo, we studied their repair in a ∼170 kb circular minichromosome whose length and topology are analogous to those of the closed loops in genomic chromatin. The rate of repair of single strand breaks in cells irradiated with γ photons was quantitated by determining the sensitivity of the minichromosome DNA to nuclease S1, and that of double strand breaks by assaying the reformation of supercoiled DNA using pulsed field electrophoresis. Reformation of supercoiled DNA, which requires that all single strand breaks have been repaired, was not slowed detectably by the inhibitors of poly(ADP-ribose) polymerase-1 NU1025 or 1,5-IQD. Repair of double strand breaks was slowed by 20–30% when homologous recombination was supressed by KU55933, caffeine, or siRNA-mediated depletion of Rad51 but was completely arrested by the inhibitors of nonhomologous end-joining wortmannin or NU7441, responses interpreted as reflecting competition between these repair pathways similar to that seen in genomic DNA. The reformation of supercoiled DNA was unaffected when topoisomerases I or II, whose participation in repair of strand breaks has been controversial, were inhibited by the catalytic inhibitors ICRF-193 or F11782. Modeling of the kinetics of repair provided rate constants and showed that repair of single strand breaks in minichromosome DNA proceeded independently of repair of double strand breaks. The simplicity of quantitating strand breaks in this minichromosome provides a usefull system for testing the efficiency of new inhibitors of their repair, and since the sequence and structural features of its DNA and its transcription pattern have been studied extensively it offers a good model for examining other aspects of DNA breakage and repair.

Abstract 5497: An inhibitor of phosphatase PP2A enhances in vivo anticancer activity of a phospho (ADP-ribosyl) polymerase inhibitor (PARPi) combined with temozolomide (TMZ)

Abstract We previously showed that a novel inhibitor of PP2A enhances the in vivo activity of TMZ against human glioblastoma xenografts (Lu et al, PNAS 2009;104:11697-11702). Several mechanisms underlie this effect of PP2A inhibition and include an increased rate of cancer cell entry into DNA synthesis, inhibition of cell cycle arrest at G1 and G2/M, and a marked reduction in p53. Because inhibitors of PARP1 have been shown to enhance the activity of cytotoxic cancer drugs by inhibiting repair of single-strand breaks in cells with impaired mechanisms for double-stranded DNA repair (BrCa mutants, triple negative breast cancer cells), we reasoned that blocking other types of DNA-damage defense mechanisms by PP2A inhibition might further enhance the activity of a PARPi combined with TMZ in cancers, not necessarily already deficient in DNA repair. We treated mice bearing xenografts of human melanoma, line A2058, with single agent LB-100, a novel water soluble PP2A inhibitor (Lixte Biotechnology Holdings, Inc., East Setauket, NY; Lu et al, J Neurosurg 2010; 113: 225-233) at 1.5mg/kg by continuous intraperitoneal (i.p.) infusion for 21 days and TMZ at 80mg/kg i.p. days 3,6,9,12,15,18. LB-100 and TMZ were given alone and in combination with each other and in 2-drug combinations with the PARPi, ABT-888 (Sigma), at 6.25 mg/kg i.p. on days 3,6,9,12,15,18, and all three drugs were given together at the same doses and schedules as when used alone or in 2-drug combinations. (ABT-888 was inactive as a single agent and was not used alone). The combination of LB-100 + TMZ was more inhibitory to the xenografts at 21 days than ABT-888 + TMZ, but the 3-drug regimen was most effective and no more toxic than any of the 2-drug regimens. Histologic analysis of xenografts 24 hours after treatment of animals with each of the single agents, each 2-drug regimen, and the 3-drug regimen showed increased apoptosis and necrosis by the 3-drug combination compared to the two-drug regimens, which in turn were more effective that the single agents. Similar results were found in vitro by flow cytometric analysis of A2058 cells 24 hours after exposure to the single agent and 2- and 3-drug regimens. Inclusion of a PP2A inhibitor may increase and extend the effectiveness of cytotoxic regimens containing a PARPi to cancers without acquired mutations in DNA repair. Influence of a static magnetic field (250 mT) on the antioxidant response and DNA integrity in THP1 cells

The aim of this study was to investigate the effect of static magnetic field (SMF) exposure in antioxidant enzyme activity, the labile zinc fraction and DNA damage in THP1 cells (monocyte line). Cell culture flasks were exposed to SMF (250 mT) during 1 h (group 1), 2 h (group 2) and 3 h (group 3). Our results showed that cell viability was slightly lower in SMF-exposed groups compared to a sham exposed group. However, SMF exposure failed to alter malondialdehyde (MDA) concentration (+6%, p > 0.05) and glutathione peroxidase (GPx) (−5%, p > 0.05), catalase (CAT) (−6%, p > 0.05) and superoxide dismutase (SOD) activities (+38%, p > 0.05) in group 3 compared to the sham exposed group. DNA analysis by single cell gel electrophoresis (comet assay) revealed that SMF exposure did not exert any DNA damage in groups 1 and 2. However, it induced a low level of DNA single strand breaks in cells of group 3. To further explore the oxidative DNA damage, cellular DNA for group 3 was isolated, hydrolyzed and analysed by HPLC-EC. The level of 8-oxodGuo in this group remained unchanged compared to the sham exposed group (+6.5%, p > 0.05). Cells stained with zinc-specific fluorescent probes zinpyr-1 showed a decrease of labile zinc fraction in all groups exposed to SMF.Our data showed that SMF exposure (250 mT, during 3 h) did not cause oxidative stress and DNA damage in THP1 cells. However, SMF could alter the intracellular labile zinc fraction.

Causes of DNA single-strand breaks during reduction of chromate by glutathione in vitro and in cells

Carcinogenic chromates induce DNA single-strand breaks (SSB) that are detectable by conventional alkali-based assays. However, the extent of direct breakage has been uncertain because excision repair and hydrolysis of Cr–DNA adducts at alkaline pH also generate SSB. We examined mechanisms of SSB production during chromate reduction by glutathione (GSH) and assessed the significance of these lesions in cells using genetic approaches. Cr(VI) reduction was biphasic and the formation of SSB occurred exclusively during the slow reaction phase. Catalase or iron chelators completely blocked DNA breakage, as did the use of GSH purified by a modified Chelex procedure. Thus, the direct intermediates of GSH–chromate reactions were unable to cause SSB unless activated by H 2O 2. SSB repair-deficient XRCC1 −/− and proficient XRCC1 + EM9 cells had identical survival at doses causing up to 60% clonogenic death and accumulation of 1 mM Cr(VI). However, XRCC1 −/− cells displayed higher lethality in the more toxic range and the depletion of GSH made them hypersensitive even to moderate doses. Elevation of cellular catalase or GSH levels eliminated survival differences between XRCC1 −/− and XRCC1 + cells. In summary, formation of toxic SSB in cells occurs at relatively high chromate doses, requires H 2O 2, and is suppressed by high GSH concentrations.

Effects of motexafin gadolinium on DNA damage and X-ray-induced DNA damage repair, as assessed by the Comet assay

To investigate the effects of motexafin gadolinium (MGd) on the levels of reactive oxygen species (ROS), glutathione (GSH), and DNA damage in EMT6 mouse mammary carcinoma cells. The ability of MGd to alter radiosensitivity and to inhibit DNA damage repair after X-ray irradiation was also evaluated. Reactive oxygen species and GSH levels were assessed by 2,7-dichlorofluorescein fluorescence flow cytometry and the Tietze method, respectively. Cellular radiosensitivity was assessed by clonogenic assays. Deoxyribonucleic acid damage and DNA damage repair were assessed in plateau-phase EMT6 cells by the Comet assay and clonogenic assays. Cells treated with 100 mumol/L MGd plus equimolar ascorbic acid (AA) had significantly increased levels of ROS and a 58.9% +/- 3.4% decrease in GSH levels, relative to controls. Motexafin gadolinium plus AA treatment increased the hypoxic, but not the aerobic, radiosensitivity of EMT6 cells. There were increased levels of single-strand breaks in cells treated with 100 mumol/L MGd plus equimolar AA, as evidenced by changes in the alkaline tail moment (MGd + AA, 6 h: 14.7 +/- 1.8; control: 2.8 +/- 0.9). The level of single-strand breaks was dependent on the length of treatment. Motexafin gadolinium plus AA did not increase double-strand breaks. The repair of single-strand breaks at 2 h, but not at 4 h and 6 h, after irradiation was altered significantly in cells treated with MGd plus AA (MGd + AA, 2 h: 15.8 +/- 3.4; control: 5.8 +/- 0.6). Motexafin gadolinium did not alter the repair of double-strand breaks at any time after irradiation with 10 Gy. Motexafin gadolinium plus AA generated ROS, which in turn altered GSH homeostasis and induced DNA strand breaks. The MGd plus AA-mediated alteration of GSH levels increased the hypoxic, but not aerobic, radiosensitivity of EMT6 cells. Motexafin gadolinium altered the kinetics of single-strand break repair soon after irradiation but did not inhibit potentially lethal damage repair in EMT6 cells.

The Involvement of Topoisomerase I in the Induction of DNA-Protein Crosslinks and DNA Single-Strand Breaks in Cells of Ultraviolet-Irradiated Human and Frog Cell Lines

Exposure of GM 4390 human skin fibroblasts and ICR 2A frog cells to 10 kJ m(-2) of ultraviolet B (UVB) radiation resulted in the formation of DNA-protein crosslinks (DPCs) and DNA single-strand breaks (SSBs). However, upon incubation, there were rapid increases in the yields of both DPCs and SSBs. An enhancement in these DNA alterations was detected within 12 min after irradiation and their levels continued to rise by 5-8-fold within 15 h after exposure to UV radiation. Using an antibody-based assay that measures covalent complex formation between topoisomerase (topo) I and genomic DNA, it was found that topo I is one of the proteins involved in these DPCs induced by UV radiation. The levels and rate of increase of topo I-DNA covalent complexes were similar to the UV-radiation-dependent formation of DPCs and SSBs. A UV-radiation-sensitive mutant frog cell line, DRP 153, was also examined and was found to be deficient in this induction of DPCs and SSBs by UV radiation. When these cells were transfected with the human SUVCC3 gene, the resulting transformant displayed kinetics for the induction of DPCs and SSBs similar to the human and parental frog cells. However, human topo I was not defected in the transformed frog cells, indicating that SUVCC3 does not encode topo I. It is likely that SUVCC3 encodes an associated enzymatic activity which permits normal stimulation of topo I-DNA covalent complexes in UV-irradiated cells.

Involvement of DNA polymerase δ and/or ϵ in joining UV-induced DNA single strand breaks in human fibroblasts (comparison of effects of butylphenyldeoxyguanosine with aphidicolin)

DNA polymerases involved in ultraviolet (UV)-induced DNA repair were studied in human fibroblasts using the inhibitors of DNA polymerases, aphidicolin which inhibits DNA polymerases α, δ and ϵ, and butylphenyldeoxyguanosine (BuPGdR) which inhibits DNA polymerase α strongly and weakly inhibits δ and ϵ. Both inhibitors inhibited replicative DNA synthesis in a dose dependent manner as measured by thymidine incorporation. However, BuPGdR did not accumulate single strand breaks in cells irradiated with 5 J/m 2 UV-light even at the highest dosage tested, indicating that BuPGdR does not inhibit DNA repair. On the other hand, aphidicolin accumulated single strand breaks in UV-light irradiated cells. These results suggest that DNA polymerase δ and/or ϵ are mainly involved in UV-induced DNA repair.

Antagonistic effect of aclarubicin on camptothecin induced cytotoxicity: Role of topoisomerase I

The cellular target of camptothecin and several of its derivatives has been identified as topoisomerase I. Central to the cytotoxic action of camptothecin is the drug's ability to stimulate formation of topoisomerase I mediated DNA cleavages. Here we demonstrate that the intercalating antitumor agent aclarubicin inhibits camptothecin induced DNA single strand breaks in cells as measured by alkaline elution. When purified topoisomerase I was reacted with DNA, aclarubicin inhibited the formation of enzyme mediated DNA breaks induced by camptothecin. High aclarubicin concentrations (10 and 100 μM) caused a slight stimulation of topoisomerase I mediated DNA cleavage at a few distinct DNA sites. The cytotoxicity associated with camptothecin treatment measured in clonogenic assays was antagonized by preincubation with aclarubicin. This inhibitory effect of aclarubicin upon camptothecin action holds implications for the scheduling of aclarubicin in combination therapy with anticancer agents directed against topoisomerase I. Aclarubicin also inhibits the effect of topoisomerase II directed agents [such as etoposide (VP16), amsacrine (mAMSA), etc.] suggesting that aclarubicin acts against the two topoisomerases.

Radiation-Induced DNA Single-Strand Breaks in Freshly Isolated Human Leukocytes

Single-strand breaks are a major form of DNA damage caused by ionizing radiation, and measurement of strand breaks has long been used as an index of overall cellular DNA damage. Most assays for DNA single-strand breaks in cells rely on measuring fractionated DNA samples following alkali denaturation. Quantification is usually achieved by prelabeling cells with radioactive DNA precursors; however, this is not possible in the situation of nondividing cells or freshly isolated tissue. It has previously been demonstrated that the alkali unwinding assay of DNA strand breaks can be quantified by blotting the recovered DNA on nylon membranes and hybridizing with radiolabeled sequence-specific probes. We report here improvements to the technique, which include hot alkali denaturation of DNA samples prior to blotting and the use of carrier DNA that is non-complementary to the radiolabeled probe. Our method allows both single- and double-stranded DNA to be quantified with the same efficiency, thereby improving the sensitivity and reproducibility of the assay, and allows calibration for determination of absolute levels of DNA strand breaks in cells. We also used this method to assay radiation-induced DNA strand breaks in freshly isolated human leukocytes and found them to have a strand break induction rate of 1815 strand breaks/cell/Gy.

Breakage and binding of DNA by reaction products of hypochlorous acid with aniline, 1-naphthylamine, or 1-naphthol

Hypochlorous acid (HOCl) is a chemically reactive oxidant and a potent microbicidal agent that is synthesized in phagosomes of inflammatory neutrophils and released into extracellular spaces. Besides reducing pathogenicity by reacting with phagocytized infectious agents, HOCl may damage tissues and yield toxic products upon reaction with various other molecules, including xenobiotics. As model xenobiotics, the substituted aryl compounds aniline, 1-naphthylamine, and 1-naphthol (1-NOH) were investigated herein for their potential to react with HOCl and be transformed into genotoxic products. The compounds were first exposed to HOCl (25–150 μ m) in phosphate buffer and afterward used to treat human fibroblasts or purified DNA. DNA single-strand breaks in cells and the binding of HOCl-reacted 1-[ 14C]NOH to purified DNA were assessed by DNA alkaline elution and scintillation spectrometry, respectively. It was found that neither HOCl nor compounds alone could break cellular DNA. But HOCl-reacted compounds produced up to 400 rat equivalents of DNA breaks. HOCl reaction products of aniline and the model bicyclic aryl compounds differed in their DNA-breaking characteristics. HOCl-reacted 1-[ 14C]NOH was stable and bound to DNA at up to 124 pmol/mg DNA. Sodium thiosulfate, glutathione, and taurine inhibited the transformation reactions; but only the former two blocked binding of HOCl-reacted 1-NOH to DNA. Ultraviolet spectra showed that HOCl reacted rapidly (<1 min) and equally well with 1-NOH at pH 7.2 or at an intraphagosomal pH of 5.0. Reaction concentrations of HOCl in this study were 2- to 11-fold lower than levels generated in vitro by stimulated neutrophils. These results show that certain aryl compounds can react readily with approximated physiological levels of HOCl ( −OCl) to form relatively long-lived products that bind DNA and age genotoxic to human cells.

DNA BREAKS CAUSED BY MONOCHROMATIC 365 nm ULTRAVIOLET‐A RADIATION OR HYDROGEN PEROXIDE and THEIR REPAIR IN HUMAN EPITHELIOID and XERODERMA PIGMENTOSUM CELLS

The induction and repair of DNA single-strand breaks (SSB) assayed by alkaline filter elution was compared in human epithelioid P3 and xeroderma pigmentosum (XP) cells exposed to monochromatic 365-nm UV-A radiation and H2O2. Initial yields of SSB were measured with the cells held at 0.5 degrees C during exposure. The yield from exposure to 365-nm radiation was slightly greater in XP than in P3 cells, whereas H2O2 produced more than three times as many SSB in P3 compared with XP cells. o-Phenanthroline (50 mM) markedly inhibited the yields of SSB induced in XP cells by H2O2, but had no effect on those produced by 365-nm UV-A. These results are consistent with the fact that P3 cells, unlike XP cells, have undetectable levels of catalase. The measured production of trace amounts of H2O2 by the actual 365-nm UV-A exposures was not sufficient to account for the numbers of breaks that were observed. Single-strand breaks produced by both agents were completely repaired after 50 min in P3 cells, as were H2O2-induced SSB in XP cells. However, 25% of the 365-nm UV-A-induced SSB in XP cells remained refractory to repair after 60 min. The results show that SSB produced by these two agents are different and that 365 nm radiation produces most SSB in cells by mechanisms other than by production of H2O2.

On the Measurement of DNA Double-Strand Breaks by Neutral Elution

Neutral elution is a sensitive and convenient method for measuring double-strand breaks in cellular DNA, but results obtained with this method are controversial, particularly as regards form of dose-response relationship. We pointed out in a recent publication (1) that explanation for different results obtained by neutral elution compared with neutral sedimentation remains obscure. The possibility exists that types of radiation damage other than double-strand breaks might affect neutral elution behavior. In a recent Letter to Editor, Hutchinson (2) stated that single-strand breaks might alter neutral elution because of configurational changes in DNA and/or increased susceptibility to shear but the necessary experiments to determine effect of single-strand breaks on neutral elution have not been performed. These necessary experiments must test whether single-strand breaks can modulate elution behavior of DNA containing a significant number of double-strand breaks, and we have performed such experiments. We induced double-strand breaks in DNA of human P3 epithelioid cells using 125I decay exactly as described previously (3), then, after allowing for accumulations of specific numbers of decays, we induced various numbers of single-strand breaks in cells by using treatments known to produce many single-strand breaks and few or no double-strand breaks: H202 (4) and bromodeoxyuridine photolysis (5), as well as y radiation. In no case (three measurements for each single-strand breakinducing treatment at different levels of induced singleand double-strand breaks) did extra single-strand breaks have any measurable effect on elution profiles (manuscript in preparation). Thus we have experimental evidence that neutral elution is not affected by concomitant singlestrand breaks, and this explanation for discrepancy between neutral sedimentation and neutral elution appears to be invalid. With respect to effect of protein crosslinked to DNA, more rigorous DNA lysis and cleaning procedures were described by Okayasu and Iliakis (6), who obtained double-strand break dose responses without a shoulder and, in some cases, straight elution profiles following these treatments. We have repeated those experiments as closely as possible using P3 cells, but saw neither exponential profiles nor dose responses without shoulders that Okasayu and Iliakis obtained. Therefore, DNA-to-protein crosslinking does not appear to be a determinant of neutral elution behavior in all types of cells. Thus question of different form of dose response measured by elution and sedimentation is still not resolved. Hutchinson's critique of neutral elution did not mention several positive aspects of assay or generally negative aspects of neutral sedimentation covered very thoroughly by van der Schans (7). The validity of elution assay is supported by fact that singleto doublestrand break ratios caused by different agents (which may differ by more than two orders of magnitude) detected by neutral elution correspond well with results obtained by other methods. Neutral sedimentation is beset with problems of irreproducibility caused in part by susceptibility of DNA to shear (7), as well as insensitivity. (Equivalent sensitivity to that of neutral elution has been achieved (8) but only by using extraordinarily laborious sample-handling procedures and 4-day sedimentation times.) Although neutral sedimentation, unlike neutral elution, is based upon mathematical theory, extent to which sedimentation theory is both untested and problematic in range of molecular weights relevant to low-dose studies in mammalian cells (9) should not be ignored. Despite unresolved probms, neutral elution remains only practical means of determining double-strand breaks at relatively low radiation doses.

Techniques to measure DNA single-strand breaks in cells: a review.

Alkaline sucrose sedimentation was for a number of years the standard procedure for the measurement of single-strand breaks. Some years ago a number of new techniques with improved sensitivity were introduced. The following techniques are presented and discussed: alkaline unwinding, alkaline filter elution, nucleoid sedimentation, viscoelastometry, microelectrophoresis of single cells, DNA precipitation, pulse field gel electrophoresis, fluctuation spectroscopy and nick translation.

Radiation-induced DNA single-strand breaks in the intestinal mucosal cells of mice treated with the radioprotectors WR-2721 or 16-16 dimethyl prostaglandin E2.

Both S-2-(3-aminopropylamino) ethylphosphorothioic acid (WR-2721) and 16-16 dimethyl prostaglandin E2 (dm PGE2) protected the intestinal clonogenic cells to some degree from the effects of 137Cs gamma-irradiation. The D0 was increased from 1.1 +/- 0.12 Gy in controls to 1.55 +/- 0.48 Gy in 16-16 dm PGE2 treated and 2.12 +/- 0.20 Gy in WR-2721 treated mice. Both agents also increased the shoulder of the clonogenic-cell survival curve. Studies were done to measure the effects of these two different radioprotectors on radiation-induction of DNA single-strand breaks in cells comprising the murine intestinal mucosa. The number of DNA single-strand breaks increased with increasing doses of gamma-rays in animals killed immediately following exposure. WR-2721 reduced the number of initial radiation-induced DNA single-strand breaks when given one-half hour before exposure; the time of maximum protection. In contrast, 16-16 dm PGE2 given 1 hour before irradiation (the time required to afford maximum protection from radiation cytotoxicity) did not reduce the number of initial DNA breaks. Both agents impeded the rate of rejoining of DNA breaks with increasing time after irradiation. However, the relationship between these effects on the rate of strand rejoining and cell survival is unknown. These results suggest that either both agents are similarly distributed within the cells but the mechanisms of radioprotection are different, or the mechanisms by which these agents protect are similar, but the two agents affect different subcellular targets, the protection of which contributes to increased cell survival.

The Oxygen Enhancement Ratio for Single- and Double-Strand Breaks Induced by Tritium Incorporated in DNA of Cultured Human T1 Cells. Impact of the Transmutation Effect

The effect of oxygen, expressed as the oxygen enhancement ratio (OER), on the number of single-strand breaks (SSB) and double-strand breaks (DSB) induced in DNA by the radioactive decay of tritium was measured in human T1 cells whose DNA had been labeled with tritium at carbon atom number 6 of thymidine. Decays were accumulated in vivo under aerobic conditions at 0-1 degrees C and at -196 degrees C and in a nitrogen atmosphere at 0-1 degrees C. The number of SSB and DSB produced was analyzed by sucrose gradient centrifugation. For each tritium decay there were 0.25 DSB in cells exposed to air at 0-1 degrees C and 0.07 in cells kept under nitrogen, indicating an OER of 3.6, a value expected for such low-LET radiation. However, for each tritium decay there were 1.25 SSB in cells exposed to air at 0-1 degrees C and 0.76 in cells kept under nitrogen indicating an OER of only 1.7. The corresponding values for 60Co gamma radiation, expressed as SSB per 100 eV absorbed energy, were 4.5 and 1.0, giving an OER of 4.5. The low OER value found for SSB induced by tritium decay can be explained if 31% of the total SSB produced in air result from transmutation by a mechanism which does not produce DSB and is unaffected by oxygen.

Induction of Single-Strand DNA Breaks in Human Cells by H 2 O 2 Formed in Near-uv (Black Light)-Irradiated Medium

When Dulbecco's modified Eagle's medium (depleted of phenol red) was irradiated for up to 3 h by 4 to 5 W/m/sup 2/ black light, hydrogen peroxide (H/sub 2/O/sub 2/) was produced. Generation of H/sub 2/O/sub 2/ resulted from riboflavin-sensitized photooxidation of tryptophan and tyrosine. Reagent H/sub 2/O/sub 2/, or hydrogen peroxide generated in black light-exposed aqueous solutions containing riboflavin and tryptophan, induced 2 x 10/sup 4/ single-strand breaks per 10/sup 16/ daltons of DNA in intact, physiologically viable human D98/AH/sub 2/ cells. Concomitant with the single-strand breaks in the cells was loss of cellular reproductive viability. Two classes of photoproducts were identified: H/sub 2/O/sub 2/ and non-H/sub 2/O/sub 2/. The H/sub 2/O/sub 2/ component of the photoproducts was responsible for all the single-strand break induction but for only partial loss of reproductive viability. The non-H/sub 2/O/sub 2/ photoproducts, accountable for the remainder of cell lethality, caused no single-strand breaks.
