Pseudomonas plecoglossicida (NR_114226) as a novel biocontrol agent against Fusarium crown rot of wheat

AbstractFusarium crown rot (FCR) poses a significant threat to wheat crops, causing substantial grain loss and mycotoxin contamination. While chemical fungicides have been effective in controlling this disease, the need for environmentally friendly alternatives is paramount due to concerns about human health and fungicide resistance. This study delves into the efficacy of Pseudomonas plecoglossicida (NR_114226) as a plant growth‐promoting and biocontrol agent against Fusarium graminearum (ON685926) and Fusarium pseudograminearum (ON687723). Greenhouse experiments demonstrated a significant reduction in disease severity by 66.06% through seed treatment with this rhizobacterial strain in soil already infested with fungal pathogens. Moreover, significant growth enhancement occurred in F. pseudograminearum‐infected seedlings treated with P. plecoglossicida (F4 + P57), increasing plant height from 13.8 to 33.1 cm and root length from 9.56 to 19.62 cm, with notable improvements in fresh and dry weights. The study further validated these findings through enzymatic assays and analysis of synthesized secondary metabolites, including chitinase, protease, hydrocyanic acid, indole acetic acid, phosphate solubilization and siderophore production. These results strongly suggest that P. plecoglossicida has the potential to serve as an effective biocontrol agent against soilborne cereal pathogens.

Repeated exposure of wheat to the fungal root pathogen Bipolaris sorokiniana modulates rhizosphere microbiome assembly and disease suppressiveness

BackgroundDisease suppressiveness of soils to fungal root pathogens is typically induced in the field by repeated infections of the host plant and concomitant changes in the taxonomic composition and functional traits of the rhizosphere microbiome. Here, we studied this remarkable phenomenon for Bipolaris sorokiniana in two wheat cultivars differing in resistance to this fungal root pathogen.ResultsThe results showed that repeated exposure of the susceptible wheat cultivar to the pathogen led to a significant reduction in disease severity after five successive growth cycles. Surprisingly, the resistant wheat cultivar, initially included as a control, showed the opposite pattern with an increase in disease severity after repeated pathogen exposure. Amplicon analyses revealed that the bacterial families Chitinophagaceae, Anaerolineaceae and Nitrosomonadaceae were associated with disease suppressiveness in the susceptible wheat cultivar; disease suppressiveness in the resistant wheat cultivar was also associated with Chitinophagaceae and a higher abundance of Comamonadaceae. Metagenome analysis led to the selection of 604 Biosynthetic Gene Clusters (BGCs), out of a total of 2,571 identified by AntiSMASH analysis, that were overrepresented when the soil entered the disease suppressive state. These BGCs are involved in the biosynthesis of terpenes, non-ribosomal peptides, polyketides, aryl polyenes and post-translationally modified peptides.ConclusionCombining taxonomic and functional profiling we identified key changes in the rhizosphere microbiome during disease suppression. This illustrates how the host plant relies on the rhizosphere microbiome as the first line of defense to fight soil-borne pathogens. Microbial taxa and functions identified here can be used in novel strategies to control soil-borne fungal pathogens.

Field evaluation of fungicides for the management of Neofabraea leaf lesion of olive in California.

Field experiments were conducted during the fall-winter seasons of 2017-18 and 2018-19 to evaluate the efficacy of various fungicides to control Neofabraea leaf lesion of olive. Field trials were conducted in the highly susceptible cultivar Arbosana in a commercial, super-high-density orchard in San Joaquin County, California. Up to eight fungicidal products were applied using an air blast backpack sprayer and their efficacy was compared with different application strategies. Results showed that most products were effective in reducing infection by the pathogens and limiting disease severity. Overall, best disease control was achieved by thiophanate-methyl, cyprodinil, difenoconazole + cyprodinil, and chlorothalonil, providing up to 75% reduction in disease severity. Copper hydroxide did not control the disease. In 2018-19, the fungicides difenoconazole + cyprodinil and ziram were evaluated in additional field trials using different application strategies (single, dual, and combined applications) suitable for pathogen resistance management. Results showed that both products provided significant reduction in disease severity (~50%) although no differences in efficacy were found between the two products nor between the different application strategies. Both products performed equally using one or two applications at two-week interval following harvest.

Acceleration of histopathological and biochemical defense patterns in mung bean with biopriming of Amycolatopsis sp. SND-1 against Cercospora leaf spot disease

ObjectivesFailure of chemical control methods in conferring protection against Cercospora leaf spot (CLS) disease in mung bean crop led to the investigation of biocontrol agents for effective management of the CLS. The current study was focused on increased defense response (histochemical and biochemical) in mung bean plants by alginate pellet formulation of endophytic Amycolatopsis sp. SND-1 (SND-1) against CLS disease. MethodsThe sodium alginate bioformulation of SND-1 was prepared by standard method using CaCl2 and evaluated for viability and contamination. Greenhouse study of prepared SND-1 formulation was performed on mung bean plants against CLS infection. Further, the acceleration of histochemical depositions (lignin, H2O2 and phenol) by differential staining methods and biochemical defense enzymes like PAL, GLU, and POX, phenolic, flavonoid and chlorophyll contents and cell death analysis were evaluated through microscopic and spectroscopic studies. ResultsThe viability assay of the prepared formulation exhibited a negligible reduction in SND-1 spores (99.5% to 86.55%) throughout the six months of storage period. In-vivo (Greenhouse) study exhibited increased growth parameters in alginate pellet formulation of SND-1challenged with Cercospora pathogen (SND-1+Pathogen) treated mung bean plants in comparison with Control (SDW) and only pathogen infested plants. Consequently, the plants with SND-1+Pathogen treatment showed significant reduction in disease severity up to 56%. Furthermore, the microscopic evaluation of histochemical defense markers depicted increased deposition of lignin (82.43±0.31%), H2O2 (47.97±0.66%), and phenol (76.74±0.79%) and cell death analysis through microscopic observation exhibited lesser dead cells of 31% in SND-1+Pathogen treatment in comparison with other treatments. Defense patterns at biochemical level hinted the elevation in activities of defense enzymes like PAL, GLU and POX at 12 and 24 h post pathogen inoculation (hpi) in SND-1+Pathogen treatment. The formulation treatment with pathogen (SND-1+Pathogen) also substantiated that enhanced total phenolic, flavonoid, and chlorophyll contents at different time intervals. ConclusionsOverall the present investigation showed that Amycolatopsis sp. SND-1 formulation confers the protection in mung bean upon Cercospora infection and it might be an effective eco-friendly approach towards sustainable agriculture.

Biocontrol of Rhizoctonia Solani in Cucumber Plants Using Endobacteria: Effects On Disease Severity and Chlorophyll Content

This study aimed to investigate the potential of endobacteria isolated from different parts of cucumber plants (roots, stems, and leaves) to inhibit the growth of the pathogenic fungus Rhizoctonia solani. Molecular identification confirmed the presence of R. solani in cucumber plants displaying symptoms of wilting and yellowing. Laboratory experiments revealed variations in citrate consumption and gram stain results among different endobacterial isolates. In field experiments, treatments involving Bacillus subtilis with Tricozone and Pseudomonas fluorescens with Tricozone demonstrated significant reductions in disease severity and increased chlorophyll content in cucumber plants compared to the pathogen control. Additionally, Acinetobacter baumannii with Hymazole at the first concentration showed superior effects on leaf area. These findings highlight the potential of endobacteria as biocontrol agents against R. solani, with implications for improving cucumber plant health and productivity.

Comparative Efficacy of Systemic and Combination Fungicides for the Control of Alternaria Leaf Spot of Cabbage

Alternaria leaf spot of cabbage, caused by the Alternaria brassicicola, affects leaves of cabbages and often results in head rots causing severe decline in yield. In this work, the effects of systemic and combination fungicides on A. brassicicola mycelia growth in vitro and disease severity in field trials were investigated. The results of in vitro evaluation revealed that both fungicides significantly inhibited (p < 0.05) the growth of A. brassicicola under in vitro conditions. However, metalaxyl-M 6% was less effective with 100 μg/mL having only 30 ± 3.5% inhibition. On the other hand, 100 μg/mL of mancozeb 63% + carbendazim 12% had 94 ± 3.5% growth inhibition of A. brassicicola, respectively, under the same conditions. Dose-response analysis of the efficacy of the two fungicides showed that the LC50 of metalaxyl-M 6% and mancozeb 63% + carbendazim 12% were 125.52 ppm and 57.22 ppm, respectively, indicating the superiority of combination fungicide over systemic fungicide alone. Field studies showed that while manure type significantly impacted on biomass production (p < 0.001), it did not significantly affect disease severity. On the other hand, the frequency of fungicide application impacted on disease severity, with biweekly application leading to a significant reduction in disease severity after 10 weeks.

Role of myeloid-derived suppressor cells in prediction of the effectiveness of biologics in children with psoriasis

Psoriasis is a chronic inflammatory skin disease characterized by increased proliferation of epidermal cells, impaired keratinization, and an inflammatory reaction in the dermis due to activation of T-lymphocytes and synthesis of proinflammatory cytokines. Pathophysiology of psoriasis is associated not only with activation of proinflammatory reactions, but also with decreased anti-inflammatory functions of immunosuppressive cells. It is known that Treg, Breg and MDSCs do not perform their classical homeostatic functions in psoriasis. In recent years, there have been more and more cases of developing resistance to ongoing therapy with genetically engineered biological drugs (GEBD) in childhood, requiring replacement or discontinuation of the drug. The aim of our work was to estimate the content of MDSCs subpopulations and their functional activity in the peripheral blood of children with psoriasis at different effectiveness of biotherapeutic drugs. We examined 110 children with psoriasis vulgaris before the appointment of biologics, at 16 and 52 weeks of therapy with adalimumab, etanercept and ustikinumab, aged 6 to 18 years. Сomparison group consisted of 34 healthy children. The effectiveness of therapy was assessed by the achievement of PASI 75 by one year of therapy. Contents of myeloid-derived suppressor cells (MDSCs) and their subpopulations, and the activity of arginase-1 were assessed by multicolor flow cytometry. An increased content of MDSCs was found in children with psoriasis against the comparison group (p = 0.000). Analysis of the effectiveness of biologics in children with psoriasis, according to PASI, showed a significant reduction in disease severity in the group of patients with good effect, both at week 16 of therapy (p = 0.000) and by one year (p = 0.017). In the group of patients with good effect of biological therapy, percentage of total MDSCs population was higher, both before start of treatment and by 52nd week of therapy (p 0.01). Children with psoriasis showed increased immunosuppressive function of MDSCs by arginase-1 activity versus the comparison group (p = 0.000). The arginase-1 activity in patients with psoriasis at the stage of disease regression (PASI 10) was significantly increased relative to children in progressive stage of psoriasis (PASI10; p = 0.001). Thus, the content of MDSCs and their suppressive activity in children with psoriasis is an informative efficiency predictor of the biological drugs. Fading of biotherapy effect after the induction course is accompanied by decreased number of MDSCs and their functional activity.

Plant Growth Promoting and Colonization of Endophytic Streptomyces albus CINv1 against Strawberry Anthracnose

Strawberry anthracnose is a serious disease, and fungicides are currently widely used by farmers. Thus, biological control is a good alternative. This study aims to identify the species of endophytic Streptomyces CINv1 that was previously isolated from Cinnamomum verum J. Presl. and to evaluate its properties as a biocontrol agent, plant growth promoter, and plant colonizing endophyte. This strain was identified by analysis of its 16S rRNA gene sequences, and the result shows 100% similarity to Streptomyces albus CINv1. The CINv1 strain displayed high resistance (81.83%) against Colletotrichum sp. isolate CA0110, as tested by the dual culture technique. Additionally, inhibited pathogen growth on IMA-2 agar was observed under a compound microscope. The results demonstrated swelling, bulbousness, and cytoplasmic aggregation of abnormal hyphal, which were confirmed by SEM as well. Furthermore, the functional media used to evaluate plant growth-promoting properties, including nitrogen fixation, phosphate solubilization, and siderophore production, yielded positive results. Analyses of plant hormones by HPLC found their ability to produce indole-3-acetic acid (IAA). Thus, a biological control trial in greenhouse conditions was conducted by spraying a spore suspension of the strain onto strawberry seedlings once a week, which showed a significant reduction in disease severity. After the seventh spraying, the assessment of the number of leaves and canopy height of the seedling showed significant promotion. In addition, the CINv1 strain established a mutualistic interaction with the plant cells through colonization inter-and intracellularly in strawberry roots, leaves, and petioles. Moreover, using LC-MS/MS to analyze the secondary metabolites of this strain, various groups of compounds were found that could potentially benefit pharmaceutical and agricultural uses.

Undaria pinnatifida (Harvey) Suringar aqueous extract activates potato defense responses against Phytophthora infestans

Undaria pinnatifida is an invasive macroalga on the Argentinean coast and its use as a biostimulant was rarely investigated. The present work aimed to evaluate the effect of U. pinnatifida aqueous extract (AE) as an inducer of potato defense responses. Our results showed that AE protected potato plants against Phytophthora infestans infection. Application of AE by foliar spray (10% dilution), resulted in a significant reduction in disease severity. However, when antimicrobial activity was tested in vitro, a higher concentration of AE was needed to reduce P. infestans growth, showing hyphae morphological alterations and a reduction in the number of sporangia. These results indicate that AE at 10% induced defense responses in treated plants. The analysis of PR1 and IPII genes (Salicylic Acid and Jasmonic acid pathways markers, respectively), showed that the SA pathway would participate in the AE-induced resistance. Additionally, the accumulation of β-1,3-glucanase, guaiacol peroxidase activity, and changes in free polyamines were analyzed. An accumulation or enhanced activity was stimulated by AE. Moreover, the effect of AE treatment might be positive for the plant performance, since all the dilutions of AE tested showed an increment of chlorophyll content. These preliminary results strongly encourage further studies of the potential use of AE as an alternative tool for potato disease management using locally available seaweed in environmentally friendly conditions.

Breast Implant Illness: A Cohort Study.

Background Breast implant illness (BII) is a clinical disease defined by a constellation of symptoms that patients experience as a result of their breast implants. This retrospective, cohort study evaluated the benefit of breast implant explantation with total capsulectomy on patients' symptoms. Methodology This is a single-center, single-arm,cohort study utilizing retrospectively collected data. All participants included in this study voluntarily presented to the department of plastic and reconstructive surgery and requested breast implant removal. A total of 229 patients were enrolled in the study over a three-year period from 2018 to 2021.The primary endpoints of the study were to objectively grade the improvement of symptomatology following surgical intervention. The secondary endpoints were to identify co-factors such as age, comorbidities, implant characteristics, the timing of symptoms, and other data that were potentially influenced by or influencers of the breast implant illness. Results The study achieved a total of 549-point decrease in symptom frequencies following surgery. Furthermore, with an average preoperative symptom score of 3.5 (scored 1-5) and a postoperative average of 1.9, the study demonstrated a score reduction of 1.6 across all symptoms. Furthermore, the study was able to eliminate on average 2.8 symptoms of breast implant illness from every patient following explantation. Conclusion Breast implant illness is a true clinical entity that affects an extensive population of patients who have undergone breast augmentation. This study has not only highlighted the extensive morbidity of breast implant illnessbut has also demonstrated that there is an opportunity to standardize treatment for this disease. These outcomes have proven that a significant reduction in disease severity can be achieved with breast implant explantation and total capsulectomy.

Nanocapsules of ZnO Nanorods and Geraniol as a Novel Mean for the Effective Control of Botrytis cinerea in Tomato and Cucumber Plants.

Inorganic-based nanoparticle formulations of bioactive compounds are a promising nanoscale application that allow agrochemicals to be entrapped and/or encapsulated, enabling gradual and targeted delivery of their active ingredients. In this context, hydrophobic ZnO@OAm nanorods (NRs) were firstly synthesized and characterized via physicochemical techniques and then encapsulated within the biodegradable and biocompatible sodium dodecyl sulfate (SDS), either separately (ZnO NCs) or in combination with geraniol in the effective ratios of 1:1 (ZnOGer1 NCs), 1:2 (ZnOGer2 NCs), and 1:3 (ZnOGer2 NCs), respectively. The mean hydrodynamic size, polydispersity index (PDI), and ζ-potential of the nanocapsules were determined at different pH values. The efficiency of encapsulation (EE, %) and loading capacity (LC, %) of NCs were also determined. Pharmacokinetics of ZnOGer1 NCs and ZnOGer2 NCs showed a sustainable release profile of geraniol over 96 h and a higher stability at 25 ± 0.5 °C rather than at 35 ± 0.5 °C. ZnOGer1 NCs, ZnOGer2 NCs and ZnO NCs were evaluated in vitro against B. cinerea, and EC50 values were calculated at 176 μg/mL, 150 μg/mL, and > 500 μg/mL, respectively. Subsequently, ZnOGer1 NCs and ZnOGer2 NCs were tested by foliar application on B. cinerea-inoculated tomato and cucumber plants, showing a significant reduction of disease severity. The foliar application of both NCs resulted in more effective inhibition of the pathogen in the infected cucumber plants as compared to the treatment with the chemical fungicide Luna Sensation SC. In contrast, tomato plants treated with ZnOGer2 NCs demonstrated a better inhibition of the disease as compared to the treatment with ZnOGer1 NCs and Luna. None of the treatments caused phytotoxic effects. These results support the potential for the use of the specific NCs as plant protection agents against B. cinerea in agriculture as an effective alternative to synthetic fungicides.

Impact of non-fumigant nematicides on reproduction and pathogenicity of Meloidogyne enterolobii and disease severity in tobacco.

Meloidogyne enterolobii is a highly aggressive quarantine pathogen which threatens the multibillion-dollar tobacco industry and is not manageable with the currently available management methods in tobacco. There is currently no known host plant resistance in tobacco and previous studies have shown that the lower level of the currently recommended rate of non-fumigant nematicides does not provide satisfactory management of M. enterolobii. The current study was conducted with the hypothesis that M. enterolobii can be better managed using a single soil application of the maximum allowed rate of non-fumigant nematicides. Treatments involved three non-fumigant chemical nematicides (oxamyl, fluopyram, and fluensulfone), a biological nematicide derived from Burkholderia, and a non-treated control. Fluensulfone significantly suppressed the nematode reproduction relative to the control, the suppression being 71% for eggs and 86% for the second stage juveniles (J2). Fluopyram also suppressed nematode reproduction, although this was statistically insignificant, with the suppression being 26% and 37% for eggs and J2, respectively. Oxamyl significantly suppressed J2 (80%), but not eggs (50%) in relation to the control. The most significant reduction of disease severity was achieved by the application of fluensulfone (64%), followed by oxamyl (54%) and fluopyram (48%). Except for fluensulfone, which significantly reduced the root biomass, none of the nematicides significantly impacted root and shoot biomass. The biological nematicide did not significantly affect nematode reproduction, pathogenicity, or disease severity. The results from the current study suggest that while the non-fumigant nematicides provided a good level of the nematode suppression, more research is needed to improve the efficacy of non-fumigant nematicides through employing better application methods or finding better chemistries.

Global longitudinal strain as a potential marker for earlier treatment referral for severe aortic stenosis

Abstract Background Aortic stenosis (AS) is the most prevalent valvular disease in the developed countries. The disease progression is long and It is of greatest importance to choose the optimal intervention timing and to secure best long term prognosis. Purpose To establish left ventricular strain as independent predictor for left ventricular function improvement after AVR which could be used for treatment decision making. Methods In this prospective, crossectional study 76 patients were included that fulfilled the criteria for severe aortic stenosis according to the guidelines from professional associations. In all patients before the AVR detailed clinical, functional, angiographic examination was performed as well as transthoracic echocardiography using Philips Epique 7. With 2D TTE the left ventricular systolic and diastolic function was assessed and with speckle tracking echocardiography global longitudinal strain (GLS %) was evaluated. Same TTE evaluation was performed 4 months after AVR. Measurements were analyzed using QLAB 7.1. Statistical analysis was performed using SPSS 25.0. Results Surgical treatment of the aortic valve significantly reduced aortic stenosis severity in all patients. Age didn't have any impact on change of left ventricular function after AVR. In both genders there was significant reduction in disease severity as well as improvement in systolic and diastolic function of the left ventricle (Figure 1) with greater improvement of GLS values in the female gender. Analysis of GLS changes after AVR in patients divided by EF showed significant improvement of GLS in patients no matter the value of preoperative EF (Figure 2). For EF improvement after AVR independent predictors were: left ventricular systolic dimension and GLS. Analysis of our cohort before AVR showed significant correlation of impaired GLS with male gender, obesity, more severe AS, larger LVD dimension and wall thickness, as well as LV systolic and diastolic volumes, higher LVMI, lower EF and lower stroke index, reduced MAPSE and s'TDI, higher wall motion score index, higher Zva and lower left atrial EF. After AVR there was significant improvement of GLS in all patient, however patients with reduced preoperative EF had significantly worse GLS after AVR. In the patients with preserved EF ≥50%, GLS before AVR was independent predictor for postoperative improvement of EF and number of segment with LS less than 13% was independent predictor for positive remodeling only in patients with low flow. Conclusions Impairment of LV GLS is of particular significance in the wide variety of patients with preserved EF that vary in ratio between flow and gradient were worse values of GLS imply worse prognosis. The results from our study support promotion of LV GLS as a marker for decision on AVR timing in respect of its prognostic implications. It is our expectation that GLS evaluation would find its place in future guidelines for diagnosis and treatment of patients with AS. Funding Acknowledgement Type of funding sources: None.

Trichoderma harzianum sensu lato TSM39: A wheat microbiome fungus that mitigates spot blotch disease of wheat (Triticum turgidum L. subsp. durum) caused by Bipolaris sorokiniana

Bipolaris sorokiniana - the causal agent of wheat spot blotch - is an emerging phytopathogenic fungus in the Yaqui Valley, Sonora, Mexico. At present, its control relies on the use of harmful chemicals, partly due to the lack of effective alternatives. Thus, this work aimed to identify and characterize indigenous wheat-associated fungal strains with biocontrol capacity against B. sorokiniana that can be integrated into control strategies for spot blotch management. Out of 59 fungal strains evaluated, we identified 5 strains with antagonistic activity against B. sorokiniana by dual confrontation assay, and these were taxonomically assigned as Trichoderma harzianum sensu lato (strains TSM39 and TSO39), Aspergillus terreus (strains TRQ89 and TRQ93), and Aspergillus nidulans (strain TRQ83) based on phylogenetic analysis using the ITS region of the rRNA cistron. Trichoderma harzianum TSM39 showed interesting biological control traits (i.e., strong antagonistic activity, high growth rate, non-cytotoxicity) and its application to wheat plants - under highly conducive conditions for spot blotch outbreak- resulted in a significant reduction of disease severity. In vitro experiments revealed that the chitinolytic system of strain TSM39 responds to B. sorokiniana lysates (signals), which could have potential biotechnological applications to improve its biocontrol activity. Additionally, a solid-state fermentation system was successfully established for spore production of strain TSM39. Taken together, Trichoderma harzianum sensu lato TSM39 is a biocontrol agent against B. sorokiniana with the potential to be integrated into spot blotch management strategies and contribute to the sustainability of the region.

Combination of Sodium Bicarbonate (SBC) with Bacterial Antagonists for the Control of Brown Rot Disease of Fruit.

Simultaneous treatment with antagonistic bacteria Bacillus amylolquefaciens (SF14), Alcaligenes faecalis (ACBC1), and the food additive sodium bicarbonate (SBC) to control post-harvest brown rot disease caused by Monilinia fructigena, and their effect on the post-harvest quality of nectarines were evaluated. Four concentrations of SBC (0.5, 2, 3.5, and 5%) were tested. Results showed that bacterial antagonists displayed remarkable compatibility with different concentrations of SBC and that their viability was not affected. The results obtained in vitro and in vivo bioassays showed a strong inhibitory effect of all treatments. The combination of each bacterial antagonist with SBC revealed a significant improvement in their biocontrol efficacies. The inhibition rates of mycelial growth ranged from 60.97 to 100%. These results also indicated that bacterial antagonists (SF14 or ACBC1) used at 1 × 108 CFU/ mL in combination with 2, 3.5, or 5% SBC significantly improved the control of M. fructigina by inhibiting the germination of spores. Interestingly, disease incidence and lesion diameter in fruits treated with SF14, ACBC1 alone, or in combination with SBC were significantly lower than those in the untreated fruits. In vivo results showed a significant reduction in disease severity ranging from 9.27 to 64.83% compared to the untreated control, while maintaining the appearance, firmness, total soluble solids (TSS), and titratable acidity (TA) of fruits. These results suggested that the improved disease control by the two antagonistic bacteria was more likely due to the additional inhibitory effects of SBC on the mycelial growth and spore germination of the pathogenic fungus. Overall, the combination of both bacteria with SBC provided better control of brown rot disease. Copper oxide nanostructures as a potential method for control of zucchini yellow mosaic virus in squash.

Zucchini yellow mosaic virus (ZYMV) infects cucurbits and has been identified as a major limiting factor in their production. The purpose of this study was to create copper oxide nanostructures (CONS) to control ZYMV in squash plants. Protection of squash against ZYMV was assessed in terms of virus severity, ZYMV concentration, transcription of pathogenesis-related genes and growth enhancement of treated squash. The findings revealed that squash plants treated with CONS had a significant reduction in disease severity when compared with untreated plants. In squash plants treated with CONS, defense genes associated with the salicylic acid signaling pathway were strongly expressed compared with untreated plants. The structural characteristics of CONS, such as their small size and appropriate shape, added to their excellent anti-ZYMV efficacy. CONS-treated squash plants show significantly improved growth traits compared with untreated plants. Based on the results of this study, CONS may be a new strategy for the control of ZYMV in squash. This represents an unconventional solution to control this virus, particularly as no chemical pesticides can control viral diseases. © 2022 Society of Chemical Industry.

A Novel In Planta Enrichment Method Employing Fusarium graminearum-Infected Wheat Spikes to Select for Competitive Biocontrol Bacteria.

This work introduces an alternative workflow for the discovery of novel bacterial biocontrol agents in wheat against Fusarium head blight (FHB). Unlike the mass testing of isolate collections, we started from a diverse inoculum by extracting microbiomes from ears of field-grown plants at grain filling stage. Four distinct microbial communities were generated which were exposed to 3 14-day culture-independent experimental enrichments on detached wheat spikes infected with F. graminearum PH1. We found that one bacterial community reduced infection symptoms after 3 cycles, which was chosen to subsequently isolate bacteria through limiting dilution. All 94 isolates were tested in an in vitro and in planta assay, and a selection of 14 isolates was further tested on detached ears. The results seem to indicate that our enrichment approach resulted in bacteria with different modes-of-action in regard to FHB control. Erwinia persicina isolate C3 showed a significant reduction in disease severity (Fv/Fm), and Erwinia persicina C3 and Pseudomonas sp. B3 showed a significant reduction in fungal biomass (cGFP). However, the mycotoxin analysis of both these treatments showed no reduction in DON levels. Nevertheless, Pantoea ananatis H3 and H11 and Erwinia persicina H2 were able to reduce DON concentrations by more than 50%, although these effects were not statistically significant. Lastly, Erwinia persicina H2 also showed a significantly greater glucosylation of DON to the less phytotoxic DON-3G. The bacterial genera isolated through the enrichment cycles have been reported to dominate microbial communities that develop in open habitats, showing strong indications that the isolated bacteria can reduce the infection pressure of F. graminearum on the spike phyllosphere.

How effective is cyanoacrylate closure in small saphenous vein insufficiency? A single center experience.

The present study was aimed to evaluate the efficacy and early outcomes of n-butyl cyanoacrylate (NBCA) ablation in small saphenous vein (SSV) insufficiency. In this single-center, retrospective, single-arm study, NBCA ablation was performed in 80 patients with SSV insufficiency between September 2018 and May 2020. Primary outcomes (anatomic success rate and occlusion rate) and secondary outcomes (VCSS and AVVQ scores) of the patients were analyzed. No technical failure and device-related complications were encountered. Anatomic success rate was 100% after procedures. Each treated SSV was occluded on colored Doppler ultrasonography immediately after the procedure, and all veins remained occluded at 2weeks after the procedure. Partial recanalization was observed in five patients at 12-month follow-up. Kaplan-Meier analysis revealed an occlusion rates at 6months and 12months follow-up were 97.5% and 93.75%, respectively. The mean pretreatment VCSS (4.72 ± 2.04) decreased to 1.61 ± 0.93, 0.87 ± 0.58, and 0.73 ± 0.52 at 2weeks, 6months, and 12months after treatment, respectively (p < .001). The mean pretreatment AVVQ score (11.92 ± 2.23) decreased to 8.2 ± 1.89, 4.2 ± 1.16, and 3.32 ± 1.19 at 2weeks, 6months, and 12months after treatment, respectively (p < .001). The Clinical, Etiologic, Anatomic, and Pathophysiology clinical classification at 12months demonstrated a significant reduction in disease severity compared with preoperative values (p < .001). There was no mortality and major adverse events including anaphylaxis and pulmonary thromboembolism (PTE) related to procedure after follow-ups. Moreover, there were no symptoms or signs of any sural nerve injury and no cases of skin necrosis, infection, or hyperpigmentation. In addition, no hematoma, deep venous thrombosis, and hypersensitivity reactions were observed. Phlebitis-like abnormal reaction was observed in three patients (3.8%). In conclusion, in patients with SSV insufficiency, NBCA ablation with VenaBlock® system appears to be an effective and reliable treatment method. At the 12-month follow-up, the NBCA of SSVs showed a low recanalization rate and had a satisfactory improvement on the VCSS and AVVQ scores.

Diabetes and coronavirus (SARS-CoV-2): Molecular mechanism of Metformin intervention and the scientific basis of drug repurposing.

Coronavirus Disease 2019 (COVID-19), caused by a new strain of coronavirus called Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), was declared a pandemic by WHO on March 11, 2020. Soon after its emergence in late December 2019, it was noticed that diabetic individuals were at an increased risk of COVID-19–associated complications, ICU admissions, and mortality. Maintaining proper blood glucose levels using insulin and/or other oral antidiabetic drugs (such as Metformin) reduced the detrimental effects of COVID-19. Interestingly, in diabetic COVID-19 patients, while insulin administration was associated with adverse outcomes, Metformin treatment was correlated with a significant reduction in disease severity and mortality rates among affected individuals. Metformin was extensively studied for its antioxidant, anti-inflammatory, immunomodulatory, and antiviral capabilities that would explain its ability to confer cardiopulmonary and vascular protection in COVID-19. Here, we describe the various possible molecular mechanisms that contribute to Metformin therapy’s beneficial effects and lay out the scientific basis of repurposing Metformin for use in COVID-19 patients.
