Phytochemical Screening, Antioxidant, Cytotoxic, Analgesic, Antidiarrheal Activity and GC-MS Analysis of the Leaves of Stachyphrynium Placentarium

The goal of the present study is to analyze the phytochemicals, antioxidants, cytotoxic, analgesic and anti-diarrheal properties of the leaves of Stachyphrynium placentarium methanol extract as well as its dichloromethane soluble fraction using GC-MS technique. The leaves of the S. placentarium were extracted with methanol and designated as SPM. Phytochemicals investigation exhibited the presence of several important secondary metabolites. Antioxidants such as total phenolic content, flavonoids content, total antioxidant, reducing power capacity and DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging assay SPM revealed non-significant antioxidant properties. The cytotoxicity study of the extract showed significant cytotoxicity (LC50 35.1093 µg/mL) compared to standard vincristine sulfate (LC50 0.482 µg/mL). Analgesic study of S. placentarium by hot plate model indicates both the doses exhibited significant central analgesic activity compared with the control. The maximum analgesic activity was observed at 90 min. SPM (500 mg/kg) is 60.21 %. SPM250 also showed a significant percentage of elongation or latency from 30 to 150 min 57.00, 59.64, 59.88, 58.07 and 53.71 %, respectively 30 min time interval. In the acetic acid writhing test, S. placentarium significantly outperformed the control and standard (SPM250 - 52.27 %, SPM500 - 34.09 and STD - 27.27 %), showing a significant increase in peripheral analgesic activity (p < 0.001). Antidiarrheal activity of SPM showed that SPM250 and SPM500 substantially decreased the feces in castor oil-induced diarrhea (p < 0.05). According to the study, the incidence and severity of diarrhea are significantly reduced by SPM500. Four chemicals are found in the dichloromethane-soluble portion of the methanol extract of S. placentarium leaves, according to GC-MS studies such as Octadecane, 6-methyl- (1.326 %), Dodecane (1.02 %), 1-Iodo-2-methylundecane (0.47 %), tetradecanoic acid 12-methyl, methyl ester (72.64 %). Tetradecanoic acid exhibits anti-inflammatory, anticancer, 5-reductase inhibitory, antioxidant, hemolytic and antifibrinolytic properties. HIGHLIGHTS The phytochemicals, antioxidants, cytotoxic, analgesic and anti-diarrheal properties of Stachyphrynium placentarium methanol extract (SPM) leaves. The dichloromethane soluble fraction is examined using Gas chromatography–mass spectrometry (GC-MS) and identified Octadecane, 6-methyl-, Dodecane, 1-Iodo-2-methylundecane, tetradecanoic acid 12-methyl, methyl ester. The brine shrimp lethality test exhibited that SPM has significant cytotoxicity (LC50 35.1093 µg/mL) compared to vincristine sulphate (LC50 0.482µg/mL), the extract showed significant cytotoxicity (LC50 35.1093 µg/mL). S. placentarium at both dosages (250 and 500 mg) provided significant central analgesia compared to the control in a hot plate model. In the acetic acid writhing test, S. placentarium showed significantly higher peripheral analgesic action (SPM250 - 52.27, SPM500 - 34.09 and STD - 27.27 %) compared to control and standard. In castor oil-induced diarrhoea, SPM250 and SPM500 (250 and 500 mg) showed significantly reduced faeces production (p < 0.05). GRAPHICAL ABSTRACT

Aqueous Seed Extract of Nigella Sativa Ameliorated Metronidazole-Induced Testicular Damage Via Up-Regulations of the Antioxidant System

Background: Infertility is a condition characterized by the failure to achieve a pregnancy after 12 months or more of regular, unprotected sexual intercourse. Objectives: To determine the effect of aqueous seed extract of Nigella sativa on testicular antioxidant levels in metronidazole-induced infertility in male Wistar rats. Methods: A total of thirty-five (35) male Wistar rats (aged 7-10 weeks old) were randomly distributed into five different groups (n = 7 each); Group A (Control group), Group B (Metronidazole only), Group C (Metronidazole + aqueous seed extract of Nigella sativa), Group D (Metronidazole Recovery), and Group E (Metronidazole + aqueous seed extract of Nigella sativa + Recovery). Experimental animals received 500 mg/kg of metronidazole and 300 mg/kg of aqueous seed extract of Nigella sativa orally. Results: Metronidazole treatment caused a significant (p<0.05) reduction in the level of testicular glutathione (Control group - 3.50±0.50; MTZ only - 2.00±0.30; MTZ+NS - 2.50±0.40; MTZ+ NS+ Recovery - 2.80±0.40). Similar reductions occurred in the activities of testicular superoxide dismutase (Control - 1.58±0.10; MTZ only - 0.23±0.06; MTZ+NS - 0.68±0.10; MTZ+ NS+ Recovery - 0.68±0.10) and catalase (Control - 2.70±0.35; MTZ only - 0.70±0.25; MTZ+NS - 1.23±0.20; MTZ+ NS+ Recovery: 1.25±0.30) which was followed by histo-architectural alterations in the testes of rats. Thereafter, aqueous seed extract of Nigella sativa treatment improved all the initial alterations caused by metronidazole treatment. Two - weeks recovery period was accompanied by a significant (p = 0.017) increase in antioxidant enzyme activities of group E when compared with group B and group D. Conclusions: The aqueous seed extract of Nigella sativa exerts its ameliorative effect by improving antioxidant enzyme activities in the testes which then protect the testes against oxidative damage, enhancing sperm quality, preventing apoptosis, supporting testicular function, and aiding tissue repair.

Site-directed mutagenesis of nattokinase: Unveiling structure-function relationship for enhanced functionality

Site-directed mutagenesis was employed to investigate the structure-function relationship of nattokinase (NK) and its effect on the enzymatic activity, thermostability, pH tolerance, and fibrinolytic properties of NK. Specific mutations (T270S, V271I, E262D, and A259T) were introduced within the nk gene, targeting regions predicted to be involved in substrate binding. The NK(E262D) mutant exhibited a significant increase in enzymatic activity (2-fold) and catalytic efficiency (2.2-fold) as assessed by N-Succinyl-Ala-Ala-Pro-Phe p-nitroanilide (Suc-AAPF-pNA) hydrolysis, compared to the wild type. In silico analysis supported these findings, demonstrating lower binding energy for the NK(E262D) mutant, suggesting stronger fibrin affinity. Thermostability assays revealed that NK(E262D) and NK(A259T) displayed exceptional stability, retaining enzyme activity at 60 °C. All mutants exhibited a broader pH tolerance range (pH 5.0–10.0) compared to the wild-type NK. The fibrinolytic activity assay revealed that the E262D mutant possessed the highest fibrinolytic activity (2414 U/mg), surpassing the wild-type. This study reported an NK variant with improved enzymatic activity, thermostability, and fibrinolytic properties.

Multiple dimensions of syntactic structure are resolved earliest in posterior temporal cortex

How we combine minimal linguistic units into larger structures remains an unresolved topic in neuroscience. Language processing involves the abstract construction of ‘vertical’ and ‘horizontal’ information simultaneously (e.g., phrase structure, morphological agreement), but previous paradigms have been constrained in isolating only one type of composition and have utilized poor spatiotemporal resolution. Using intracranial recordings, we report multiple experiments designed to separate phrase structure from morphosyntactic agreement. Epilepsy patients (n = 10) were presented with auditory two-word phrases grouped into pseudoword-verb (‘trab run’) and pronoun-verb either with or without Person agreement (‘they run’ vs. ‘they runs’). Phrase composition and Person violations both resulted in significant increases in broadband high gamma activity approximately 300 ms after verb onset in posterior middle temporal gyrus (pMTG) and posterior superior temporal sulcus (pSTS), followed by inferior frontal cortex (IFC) at 500 ms. While sites sensitive to only morphosyntactic violations were distributed, those sensitive to both composition types were generally confined to pSTS/pMTG and IFC. These results indicate that posterior temporal cortex shows the earliest sensitivity for hierarchical linguistic structure across multiple dimensions, providing neural resources for distinct windows of composition. This region is comprised of sparsely interwoven heterogeneous constituents that afford cortical search spaces for dissociable syntactic relations.

Exploring rice tolerance to salinity and drought stresses through Piriformospora indica inoculation: understanding physiological and metabolic adaptations.

Drought and salinity are significant challenges to global food security. This study investigated the interactive impacts of Piriformospora indica inoculation with salinity and drought stresses on rice. Two greenhouse experiments were conducted. Thefirst experiment evaluated two P. indica inoculation levels and three salinity levels (0-, 50-, and 100-mM sodium chloride), while the subsequent experiment assessed two inoculation levels under three drought intensities (25%, 50%, and 100% of available water content). P. indica spores were inoculated following optimized seed disinfection and germination processes. The shoot and root biomass under salinity stress were consistently higher in inoculated plants compared to controls. Sodium concentrations in shoots and roots exhibited an overall upward trend, with the trend being less pronounced in inoculated plants due to increased potassium uptake. Under salinity stress, nitrogen, magnesium, and calcium concentrations significantly increased in inoculated plants. With increasing salinity, there was a significant increase in catalase enzyme activity and soluble carbohydrate concentrations across all treatments, with a greater increase in inoculated plants. Plants under drought stress experienced reduced root and shoot biomass, but inoculated plants maintained higher biomass. Increasing drought stress led to decreased nitrogen, magnesium, and calcium concentrations in all treatments, with the reduction being less severe in inoculated plants. Catalase enzyme activity and carbohydrate increased with rising drought stress, with the increase being more pronounced in inoculated plants compared to non-inoculated ones. By promoting plant growth, nutrient uptake, and stress tolerance, P. indica inoculation has a significant potential to enhance crop productivity in extreme climate conditions.

Biological transformation of herbal extracts through Lactic Acid Bacteria fermentation: A study on Milk thistle and liquorice root extract

Herbal extracts have recently gained attention in pharmaceuticals, supplements, and cosmetics. This study explored the impact of Lactic Acid Bacteria (LAB) fermentation on the functional properties of milk thistle and licorice root extracts to develop health-enhancing beverages. Biological transformation using microorganisms or enzymes was employed to improve the stability and efficacy of these extracts. After 36 h of fermentation, bacterial growth in both extracts surged approximately two-fold. The licorice root extract showed a particularly significant increase in total phenolic content and antioxidant activity, attributed to bacterial enzymes breaking down complex, high-molecular-weight phenolic compounds into simpler, more potent free phenols. Despite a reduction in total flavonoid content across all samples, this did not indicate a decline in biological activity. High-Performance Liquid Chromatography (HPLC) analysis revealed a notable increase in silymarin concentration in the milk thistle extract, rising from 0.59 mg mL−1 to 2.5 mg mL−1 after fermentation with Lactiplantibacillus plantarum. Glycyrrhizic acid in the licorice root extract slightly decreased following fermentation with L. plantarum, while no significant changes were observed in extracts fermented with L. acidophilus. Cell viability studies during cold storage showed that the bacterial count in the licorice root extract remained within acceptable limits (106 CFU mL−1) for four weeks, ensuring ongoing health benefits. However, in the milk thistle-fermented extract, no viable cells were detected by the third week. These findings demonstrate the potential of LAB-fermented extracts for developing functional beverages with enhanced health benefits and therapeutic properties.

Possible protective effect of remifentanil against testicular ischemia-reperfusion injury

Abstract Objectives This study aims to evaluate the protective efficacy of remifentanil against testicular ischemia-reperfusion injury. Methods The study included 24 male rats. The rats were randomized into three groups: Group 1 was the control group. Group 2 was subjected to a testicular torsion/detorsion model. Group 3 underwent similar procedures and additionally received remifentanil (0.6 μg/kg/min) intravenously for the first 20 min of reperfusion. Blood samples were taken for biochemical analyses, and orchiectomy was performed for histopathologic examination. Results Biochemical analysis of blood samples showed a significant increase in antioxidant enzyme activity, including superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in Group 3 compared to Group 2 (p:0.004 and p:0.002, respectively). There was a dramatic decrease in the levels of proinflammatory cytokines, including interleukin-1 beta (IL-1 Beta), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-alpha) in Group 3 compared to Group 2 (p:0.001, p:0.046, and p:0.004, respectively). Similarly, malondialdehyde (MDA) levels decreased in Group 3 compared to Group 2 (p:0.004). Histopathologic examination of Group 3 rats showed positive changes in inflammation, hemorrhage, edema, and congestion levels compared to Group 2 (p<0.001). Similarly, there was a positive effect on the Johnsen and Cosentino score in Group 3 compared to Group 2 (p:0.001 and p<0.001, respectively). Conclusions In our study, it has been documented that remifentanil protects against testicular ischemia-reperfusion injury.

Smear Layer Removing and Pulp Dissolving Ability of Sodium Hypochlorite Mixed with Two Etidronate-Containing Irrigants in Continuous Chelation Technique

The study aimed to assess the effect of two etidronate-containing irrigants and EDTA on the ability of sodium hypochlorite (NaOCl) to remove the smear layer and dissolve organic tissues. This study evaluated the following solutions: distilled water, 3% NaOCl, 3% NaOCl + EDTA, and 3% NaOCl mixed with etidronate powder from two manufacturers [Dual Rinse, (DR); IsraDent, (ID)] to obtain 9%, 15%, and 18% solutions. To assess the proteolytic activity, bovine muscle tissue samples (56.1 ± 5.4 mg; n = 10 per group) were immersed in the tested solutions for 15 min. Absolute weight changes and percentages of weight changes (wt%) were calculated. To assess smear layer removal, the root canals of human wisdom teeth were instrumented, irrigated with the tested solutions (n = 10 per group), and evaluated using SEM. Statistical analysis employed an ANOVA with a post-hoc Tukey test and paired t-test, the Kruskal–Wallis test with a pairwise Wilcoxon rank sum test, and the Friedman test with a pairwise Wilcoxon signed-rank test. The mean weight loss in the NaOCl group comprised 17.3 mg (32 wt%). Sequential use of NaOCl and EDTA resulted in a significant increase in proteolytic activity of the former (57 wt%), while mixing these solutions led to a reduction of proteolytic activity (17 wt%). All NaOCl/DR groups exhibited a significantly greater dissolution activity than that of NaOCl alone, with the mean weight loss ranging from 23.3 mg (40 wt%) to 24 mg (41 wt%). ID9% and ID15% significantly decreased the proteolytic activity of NaOCl. In most groups, the apical thirds of the root canals demonstrated a significantly heavier smear layer compared to the middle and coronal thirds. The most effective smear layer removal was observed after irrigation with NaOCl combined with EDTA or DR (all concentrations); however, in the NaOCl + EDTA and DR18% groups, it was accompanied by moderate to severe erosion. Irrigation with ID did not result in smear layer removal or dentin erosion. In conclusion, the smear layer removal and pulp dissolving effects of continuous chelation using etidronate were manufacturer/composition-dependent. DR9% combined with NaOCl demonstrated the most promising results.

IN VITRO CHARACTERIZATION AND ANALYSIS OF RICE VARIETIES AGAINST VARIOUS LEVELS OF SALT STRESS

Salinity is a significant environmental factor that greatly impacts plant yield, particularly in arid and semi-arid regions. The study examined the response of two rice varieties (Swat-1 and Pakhal) to salinity stress on the callus. Various salt concentrations were utilized. Among the two varieties, Swat-1 displayed the highest rate of callogenesis in the initial results. The Calli were exposed to varying concentrations of salt (50mM, 100mM, and 150mM), with each concentration tested in three replicates. Under the influence of stress, Swat-1 experienced a decrease in its Relative growth rate (RGR), whereas Pakhal remained steady at 50mM and 100mM. It was only at 150mM that Pakhal also exhibited a decline in RGR. Regarding water content, the stress caused a decrease in the content. At the highest stress level (150mM), the lowest water content was observed. The impact on soluble sugars was also noteworthy. As stress levels rose, concentrations of total sugars also increased. Similar findings were noted for proline, with stress leading to an increase in its concentration. Out of the two varieties, Pakhal appeared to have the highest proline accumulation. Similarly, there was a noticeable upward trend observed in catalase activity. There was a significant increase in catalase activity in both varieties after being exposed to stress for 30 and 60 days. The findings indicate that both varieties effectively developed strategies to combat salinity stress. They exhibited high concentrations of total soluble sugars and proline, as well as demonstrated significant catalase activity.

Lactobacilli Cell-Free Supernatants Modulate Inflammation and Oxidative Stress in Human Microglia via NRF2-SOD1 Signaling

Microglia are macrophage cells residing in the brain, where they exert a key role in neuronal protection. Through the gut–brain axis, metabolites produced by gut commensal microbes can influence brain functions, including microglial activity. The nuclear factor erythroid 2-related factor 2 (NRF2) is a key regulator of the oxidative stress response in microglia, controlling the expression of cytoprotective genes. Lactobacilli-derived cell-free supernatants (CFSs) are postbiotics that have shown antioxidant and immunomodulatory effects in several in vitro and in vivo studies. This study aimed to explore the effects of lactobacilli CFSs on modulating microglial responses against oxidative stress and inflammation. HMC3 microglia were exposed to lipopolysaccaride (LPS), as an inflammatory trigger, before and after administration of CFSs from three human gut probiotic species. The NRF2 nuclear protein activation and the expression of NRF2-controlled antioxidant genes were investigated by immunoassay and quantitative RT-PCR, respectively. Furthermore, the level of pro- and anti-inflammatory cytokines was evaluated by immunoassay. All CFSs induced a significant increase of NRF2 nuclear activity in basal conditions and upon inflammation. The transcription of antioxidant genes, namely heme oxygenase 1, superoxide dismutase (SOD), glutathione-S transferase, glutathione peroxidase, and catalase also increased, especially after inflammatory stimulus. Besides, higher SOD1 activity was detected relative to inflamed microglia. In addition, CFSs pre-treatment of microglia attenuated pro-inflammatory TNF-α levels while increasing anti-inflammatory IL-10 levels. These findings confirmed that gut microorganisms’ metabolites can play a relevant role in adjuvating the microglia cellular response against neuroinflammation and oxidative stress, which are known to cause neurodegenerative diseases.Graphical Gut-brain crosstalk: molecular point of view. Metabolites contained in the supernatant derived from Lactobacilli can cross the gut barrier and reach the central nervous system, where they are taken up by microglial cells. They induce the activation of the NRF2 pathway and the production of inflammatory mediators. This interaction attenuates two important events: oxidation (with high levels of NRF2) and inflammation (with high levels of IL-10 and low levels of TNF-α).

Lamellipodia-Mediated Osteoblast Haptotaxis Guided by Fibronectin Ligand Concentrations on a Multiplex Chip.

Skull morphogenesis is a complex, dynamic process involving two different germ layers and progressing to the coordinated, directional growth of individual bones. The mechanisms underlying directional growth toward the apex are not completely understood. Here, a microfluidic chip-based approach is utilized to test whether calvarial osteoblast progenitors undergo haptotaxis on a gradient of Fibronectin1 (FN1) via lamellipodia. Mimicking the embryonic cranial mesenchyme's FN1 pattern, FN1 gradients is established in the chip using computer modeling and fluorescent labeling. Primary mouse calvarial osteoblast progenitors are plated in the chip along an array of segmented gradients of adsorbed FN1. The study performs single-cell tracking and measures protrusive activity. Haptotaxis is observed at an intermediate FN1 concentration, with an average directional migration index (yFMI) of 0.07, showing a significant increase compared to the control average yFMI of -0.01. A significant increase in protrusive activity is observed during haptotaxis. Haptotaxis is an Arp2/3-dependent, lamellipodia-mediated process. Calvarial osteoblast progenitors treated with the Arp2/3 (Actin Related Protein 2/3 complex) inhibitor CK666 show significantly diminished haptotaxis, with an average yFMI of 0.01. Together, these results demonstrate haptotaxis on an FN1 gradient as a new mechanism in the apical expansion of calvarial osteoblast progenitors during development and shed light on the etiology of calvarial defects.

Region of interest determination algorithm of lensless calcium imaging datasets.

Advances in fluorescence imaging technology have been crucial to the progress of neuroscience. Whether it was specific expression of indicator proteins, detection of neurotransmitters, or miniaturization of fluorescence microscopes, fluorescence imaging has improved upon electrophysiology, the gold standard for monitoring brain activity, and enabled novel methods to sense activity in the brain. Hence, we developed a lightweight and compact implantable CMOS-based lensless Ca2+ imaging device for freely moving transgenic G-CaMP mouse experiments. However, without a lens system, determination of regions of interest (ROI) has proven challenging. Localization of fluorescence activity and separation of signal from noise are difficult. In this study, we report an ROI selection method using a series of adaptive binarizations with a gaussian method and morphological image processing. The parameters for each operation such as the kernel size, sigma and footprint size were optimized. We then validated the utility of the algorithm with simulated data and freely moving nociception experiments using the lensless devices. The device was implanted in the dorsal raphe nucleus to observe pain-related brain activity following a formalin test to stimulate pain. We observed significant increases in fluorescence activity after formalin injection compared to the control group when using the ROI determination algorithm.

The anti-angiogenic, anti-inflammatory and anticoagulant potential of a polysaccharide extracted from the brown alga Cystoseira humilis

The present study aimed to evaluate the properties of a sulfated polysaccharide (PSCH) extracted from the brown alga Cystoseira humilis. We analyzed the chemical composition, anti-angiogenesis activities, anticoagulant, and anti-inflammatory effects of the PSCH, using various techniques encompassing UV–visible spectroscopy, fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (X-RD), scanning electron microscopy (SEM), steric exclusion chromatography (SEC) and high-performance liquid chromatography coupled to refractive index detector (HPLC-RID). The study found that PSCH was a sulfated heteropolysaccharide consisting of glucuronic acid, arabinose, galacturonic acid, xylose and fructose, and has a semi-crystalline nature with a molecular weight of about 379.44 kDa. The PSCH contains 41.77 % carbohydrates, 12.33 % proteins, and 4.59 % sulfates. The PSCH fraction has anti-angiogenic potential highlighted through assays on the chorioallantoic membrane, where vascular branching was inhibited by about 41.65 % and 61.38 % at concentrations of 50 and 100 μg/egg, respectively, compared to the untreated group. PSCH displayed an anti-edematous effect in vivo by reducing malondialdehyde and advanced oxidation protein products in paw tissue after five hours of carrageenan injection. When rats were co-treated with PSCH, there was evidence of a significant increase in antioxidant enzyme activity (superoxide dismutase, glutathione peroxidase, and glutathione) in edematous paw tissues compared to untreated rats. Furthermore, PSCH significantly influenced hematological parameters and the rate of anti-inflammatory serum protein fractions compared to the inflamed group. Interestingly, PSCH may exhibit anticoagulant properties by extending activated partial thrombosis time, thrombin time and prothrombin time. The present study has shown that Cystoseira humilis polysaccharides have antioxidant, anti-angiogenic, anti-inflammatory, and anti-coagulant properties, making them suitable for therapeutic purposes.

Bioactivated Glucoraphanin Modulates Genes Involved in Necroptosis on Motor-Neuron-like Nsc-34: A Transcriptomic Study.

Research on bioactive compounds has grown recently due to their health benefits and limited adverse effects, particularly in reducing the risk of chronic diseases, including neurodegenerative conditions. According to these observations, this study investigates the activity of sulforaphane (RS-GRA) on an in vitro model of differentiated NSC-34 cells. We performed a transcriptomic analysis at various time points (24 h, 48 h, and 72 h) and RS-GRA concentrations (1 µM, 5 µM, and 10 µM) to identify molecular pathways influenced by this compound and the effects of dosage and prolonged exposure. We found 39 differentially expressed genes consistently up- or downregulated across all conditions. Notably, Nfe2l2, Slc1a5, Slc7a11, Slc6a9, Slc6a5, Sod1, and Sod2 genes were consistently upregulated, while Ripk1, Glul, Ripk3, and Mlkl genes were downregulated. Pathway perturbation analysis showed that the overall dysregulation of these genes results in a significant increase in redox pathway activity (adjusted p-value 1.11 × 10-3) and a significant inhibition of the necroptosis pathway (adjusted p-value 4.64 × 10-3). These findings suggest RS-GRA's potential as an adjuvant in neurodegenerative disease treatment, as both increased redox activity and necroptosis inhibition may be beneficial in this context. Furthermore, our data suggest two possible administration strategies, namely an acute approach with higher dosages and a chronic approach with lower dosages.

The elicitation effects of diode and He-Ne laser irradiations on the alleviation of nutrient-deficiency induced damage in anthocyanin-producing red-fleshed apple cell suspension

Purpose We explored the elicitation role of the laser irradiations on the alleviation of nutrient-deficiency induced damage in anthocyanin-producing red-fleshed apple cell suspension in continuous production of anthocyanin. Methods Anthocyanin-producing red-fleshed apple cells were irradiated by 4 intensity levels of red He-Ne (RHNL) and blue diode (BDL) lasers for 20 min. Results Nutrient deficiency indicated negative effect on total soluble proteins (TSP), superoxidase dismutase (SOD) activity, and total phenolics content (TPC) while it displayed a positive effect on malondialdehyde (MDA), total flavonoids content (TFC), O2 -, H2O2 - , and lipoxygenase (LOX) and polyphenol oxidase (PPO) activities in light controls, illustrating oxidative stress. The laser irradiations on suspension cells indicated variable effects on measured parameters and were time of growth-, levels of intensity-, and laser type-dependent. Likewise, the elicitation effects of lasers relied on a critical threshold among ROS generation and antioxidative system which determines the fate of cells against oxidative stress. The same trend was displayed by RHNL at 6.46 mWcm−2 intensity and BDL at 13.73 mWcm−2. These intensities resulted in a significant increase in SOD, APX, POD, and CAT activities and TSP, TPC, TFC, proline, and glycine betaine accumulation, while induced decrease in LOX, and PPO activities and MDA, and ROS generation, alleviating cellular injury from prolonged nutrient deficiency by diminishing lipid peroxidation and oxidative damages of cell membrane. Conclusion Results suggested that lasers application on mitigating nutrient deficiency stress relied on establishing a suitable balance between ROS generation and antioxidative system, which enables the nutrient-starved anthocyanin-producing cells to continuously produce anthocyanin.

Construction of more oxygen vacancies of BaCo0.4Fe0.4Zr0.2O3-δ for high-performance proton-conducting solid oxide fuel cell cathodes

The cathodic catalytic activity for the oxygen reduction reaction (ORR) plays a critical role in determining the performance of proton-conducting solid oxide fuel cells (P-SOFCs). BaCo0.4Fe0.4Zr0.2O3-δ (BCFZ) has emerged as a promising cathode for P-SOFCs due to its triple-phase conductivity. Nevertheless, its suboptimal proton conductivity and hydration ability at intermediate temperatures prevent it from achieving the anticipated ORR catalytic activity. To address these limitations, this study explores the enhancement of electrocatalytic activity in BCFZ through alkali metal doping and A-site defect construction. The effects of such modifications on oxygen surface exchange kinetics and ORR catalytic activity are systematically investigated. The findings reveal that BCFZ exhibits relatively low parameters in terms of electronic conductivity, oxygen ion conductivity, oxygen vacancy concentration, kchem, and Dchem. Conversely, these parameters are markedly improved in A-site deficient BCFZ (D-BCFZ) and Na/K-doped BCFZ. Consequently, the enhancement of these properties yields a significant increase in ORR catalytic activity, with D-BCFZ demonstrating the best electrochemical performance. Specifically, P-SOFC with D-BCFZ cathode achieves an Rp value of just 0.073 Ω cm2 and a Pmax value of 0.928 W cm-2 at 650 °C. This study provides theoretical insight into the mechanisms by how alkali metal doping and defect construction enhance the ORR catalytic activity of BCFZ. These findings offer valuable guidance for the development and optimization of high-performance P-SOFC cathodes.

EFFECT OF THREE QUINOA VARIETIES AND EDIBLE FLOWERS ON ANTIOXIDANT PROPERTIES OF ALE BEER

Fermentation experiments were conducted with a top-fermenting brewing yeast strain and wort enriched with three varieties of quinoa (with white, red and black grain color), as well as edible flowers (marigold and dandelion) to study their influence on the antioxidant properties of ale beer. Plant additives led to a slight increase in pH and a lower alcohol content of the beer. By itself, quinoa did not improve the functional properties of the beverage, but the combination of black quinoa and edible flowers resulted in an increased content of polyphenols in beer; marigold showed a 48.7 % increase in this parameter compared to the control, and dandelion - 51.2 %. The content of flavonoids in beer was not affected by the addition of plant additives to the wort. A slight increase in anthocyanins (0.7 - 4.4 %) was reported for black quinoa beer as well as black quinoa with flowers. A significant increase in antioxidant activity was reported for the variants with flowers (54 % for marigold and 56.6 % for dandelion). The increased antioxidant capacity combined with the reduced alcohol content of the beers with black quinoa and flowers suggests that a beverage with improved functional properties was obtained. Black quinoa, combined with edible flowers, was more favorable for increasing the antioxidant capacity of beer than other pseudo-cereals (amaranth and chia).

New Utilization of Fraxinus mandshurica Leaves: As a Safe and Promising Natural Antioxidant.

In this paper, an in-depth study on Fraxinus mandshurica (FM) was conducted, focusing on the chemical constituents, in vitro and in vivo antioxidant activities of flavonoids, acute oral toxicity testing, network pharmacology, and molecular docking in the leaves of FM. The in vitro antioxidant results revealed that the total flavonoid extract (TFE), kaempferol, quercetin, and rutin exhibited similar antioxidant activities, with TFE demonstrating significantly better scavenging ability against hydroxyl radical compared to the other flavonoids. Moreover, in vivo antioxidant findings indicated that TFE led to a significant increase in glutathione peroxidase and superoxide dismutase activities along with a decrease in malondialdehyde levels in the liver tissues of mice in an ethanol-induced oxidative stress model, outperforming quercetin. The acute oral toxicity test established 5000 mg/kg of bw as the LD50 for TFE in rats. Through network pharmacological analysis, it was observed that all seven flavonoids in FM exhibited spontaneous binding to their respective key targets, reinforcing their potential antioxidant properties. Consequently, based on the experimental outcomes, TFE appears to be a safe and promising antioxidant source, indicating its potential as a new natural antioxidant resource.

The role of peripheral venous distension reflex in regulating hemodynamics: mini review

Significant volume is pooled in veins in humans and the amount is dramatically altered by various physiological stresses and diseases. Several animal and human studies demonstrated that limb venous distension evoked significant increases in blood pressure and sympathetic nerve activity (venous distension reflex, VDR). VDR has attracted much attention because of its potential to explain the still unknown mechanism of autonomic dysfunction in several diseases, which would lead to a new treatment approach. This mini review discusses accumulated evidence of VDR at this point and what should be investigated in the future to apply the current understanding of VDR in clinical practice.

Impact of Hadang and Bentengan Game on Students’ Physical Fitness Levels

This research aims to determine the effectiveness of blocking and fortification games on the physical fitness level of elementary school students. The research method used was experimental using a two-group pretest-posttest design. The sampling technique used purposive sampling and the sample consisted of 54 students. Data collection techniques are 1) documents taken from the results of learning notes during the treatment delivery process, and 2) physical fitness tests is to know physical fitness level. The data analysis technique uses the t-test, but the data analysed must have a normal distribution so that the level of normality and homogeneity of the data is first tested. The t-test results for the Hadang and Bentengan games showed statistically significant increases in physical fitness (e.g., t = 29.105 for Hadang, t = 21.236 for Bentengan, both p 0.05)., so it can be concluded that there was an increase in physical fitness through the Bentengan game. Students' physical fitness levels experienced a significant increase in sprints, pull-ups, sit-ups, vertical jumps and short-distance running activities. The results of this research also have implications for the development of various types of traditional games for other physical fitness learning activities or other health, sports and physical fitness learning activities. Teachers can design types of games as teaching techniques and can be adjusted to the age needs of students such as for elementary or junior high school levels.
