Guardians of the vault: unmasking online threats and fortifying e-banking security, a systematic review

Purpose The purpose of this paper is to investigate and analyze the adoption of digital technologies in the banking industry and its impact on the rise of digital fraudulent activities, specifically focusing on online banking frauds. This paper aims to provide insights into the current technologies implemented by banks to secure their online banking systems and explores the methods used by cybercriminals to exploit security vulnerabilities in these systems. Design/methodology/approach In order to understand how digital technologies in banking can be secured against online fraud, this research conducted a systematic literature review (SLR) on digital banking, online banking fraud, and security measurements. The review encompasses a variety of sources from online databases such as Emerald Insight, Google Scholar, IEEE, JSTOR, Springer and Science Direct. Findings The key finding of the paper is that the adoption of digital technologies in the banking industry has led to a significant increase in digital fraudulent activities, particularly in the form of online banking frauds. This paper emphasizes that these frauds have become a global concern and have evolved into an industry where cybercriminals use sophisticated tools such as phishing attacks, denial-of-service attacks, Trojan horses, malware infections, identity theft and computer viruses. Research limitations/implications This study relies solely on a literature review without incorporating primary data or case studies; therefore, it might miss out on the firsthand experiences and perspectives of banks and cybersecurity professionals. Practical implications This study emphasizes the need for banks to adopt advanced security measures to safeguard their online banking systems. Social implications This study underscores the importance of ongoing training and awareness programs for both bank employees and customers. Originality/value This study specifically addresses the adoption of digital technologies in the banking industry and its correlation with the increase in digital fraudulent activities. This focus on the intersection of technology and fraud in the banking sector is a distinctive aspect. This study conducts a SLR to examine the current technologies implemented by banks to safeguard their online banking systems. This comprehensive approach provides insights into the diverse security measures used by banks to protect against various types of cyber threats.

Landslide hazard cascades can trigger earthquakes.

While earthquakes are well-known to trigger surface hazards and initiate hazard cascades, whether surface hazards can instead trigger earthquakes remains underexplored. In 2018, two landslides on the Tibetan plateau created landslide-dammed lakes which subsequently breached and caused catastrophic outburst floods. Here we build an earthquake catalog using machine-learning and cross-correlation-based methods which shows there was a statistically significant increase in earthquake activity (local magnitude ≤ 2.6) as the landslide-dammed lake approached peak water level which returned to the background level after dam breach. We further find that ~90% of the seismicity occurred where Coulomb stress increased due to the combined effect of direct loading and pore pressure diffusion. The close spatial and temporal correlation between the calculated Coulomb stress increase and earthquake activity suggests that the earthquakes were triggered by these landslide hazard cascades. Finally, our Coulomb stress modeling considering the properties of landslide-dammed lakes and reservoir-induced earthquakes globally suggests that earthquake triggering by landslide-dammed lakes and similar structures may be a ubiquitous phenomenon. Therefore, we propose that earthquake-surface hazard interaction can include bidirectional triggering which should be properly accounted for during geological hazard assessment and management in mountainous regions.

Effects of dietary nano-selenium concentration on growth, proximate chemical composition and antioxidant enzymes, and physiological responses to stressors by juvenile Catla catla

This study evaluated effects of nano‑selenium (nano-Se) on growth, feed utilization and antioxidant status of Catla catla fed isonitrogenous (26.25% protein) and isolipidic (13.12% lipid) diets formulated to contain nano-Se at 0, 0.75, 1.5 and 2.25 mg/kg (analysed Se 0.32, 1.08, 1.81 and 2.61 mg/kg). There were three tanks (89 × 58 × 61 cm) for each of the four feed treatments, and each tank of 25 fish (initial weight of 6.41 ± 0.02 g) was fed to satiation twice each day for 90 days. After 90 days, growth performance and survival rate were noted and then 9 fish from each replicate were harvested and analysed for body proximate composition, serum biochemistry and antioxidant enzyme activities. The remaining 16 fish from each replicate were subjected to temperature and hypoxia stress (8 fish against each stressor). Orthogonal polynomial contrast analysis revealed dietary nano-Se supplementation significantly improved the growth performance and feed utilization in linear, quadratic trends. Fish fed diet containing 1.08 mg/kg nano-Se showed highest weight gain (28.00 ± 0.84 g) and specific growth rate (1.87 ± 0.01%/day) and lowest FCR (1.07 ± 0.03). However, no significant effect was observed on the feed intake or survival rates of the fish. Furthermore, whole body moisture, crude protein (CP) and ash contents showed significant linear and quadratic trends in response to dietary nano-Se. Before and after stress challenges, significant increase in liver superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase activities (GPx) and decrease in thiobarbituric acid reactive substances (TBARS) were observed in fish fed 1.08 mg/kg nano-Se diets compared with the control group. Before stress, nano-Se supplementation had no effect on the levels of serum glucose (GLU), serum cortisol (CORT) or the survival rate (SR), however, significant elevations in these parameters were observed after stress challenges. Fish fed 1.08 mg/kg nano-Se diet showed significantly lower values of GLU, CORT and SR as compared to the control group after each stress challenge. In conclusion, nano-Se supplementation improved the growth performance, health status and response to stressor in Catla catla. The optimum nano-Se concentration in the diets of C. catla was estimated to be 1.54, 1.60, 1.58 and 1.64 mg/kg on the basis of weight gain percent, FCR, TBARS and GLU, respectively.

The effect of the probiotic drug "Ecoflor" on the proteolytic activity of the sterlet intestine

The effect of the probiotic preparation "Ecoflor" on proteolytic activity of the intestinal mucosa and chyme of sterlet was studied. This probiotic is a consortium of strains of lacto- and bifidobacteria: B. bifidum, B. longum and L. acidophilus, L. casei, L. plantarum, immobilized on carbon-mineral sorbent SUMS-1. For the experiment, fish were divided into 4 groups: control and three experimental groups. Experimental groups of fish were given feed with probiotic in the amount of 2, 4, 6 g/kg of feed. Sampling was carried out before the beginning of the experiment, on 7, 14, and 21 days. Material for the study was taken from 5 individuals from each group. To obtain enzymatically active preparations, fish intestines were freed from fat and dried on ice bath glass. After incision, the chyme was collected with a spatula and scraper, then the mucous membrane was removed. The total proteinase activity was calculated by the ratio of the difference between the spectrophotometer readings of the sample with substrate and the blank sample to the product of the intestinal weight (g) by the incubation time (min) EA = ΔE425/(g×min). Determinations were performed in three repetitions. Data analysis showed the stimulating effect of the preparation on proteolytic activity. The maximum increase in enzymatic activity in the experimental groups compared to the control group was recorded on the 7th day of the experiment. A more significant increase in proteolytic activity of the intestinal mucosa compared to chyme was found. At the beginning of feeding, the stimulating effect of the preparation was found in all experimental groups compared to the control. In further observation periods the effectiveness of the probiotic significantly decreased.

IL-17RA-Mediated Epithelial Cell Activity Prevents Severe Inflammatory Response to Helicobacter pylori Infection.

Helicobacter pylori is a Gram-negative pathogen that colonizes the stomach, induces inflammation, and drives pathological changes in the stomach tissue, including gastric cancer. As the principal cytokine produced by Th17 cells, IL-17 mediates protective immunity against pathogens by inducing the activation and mobilization of neutrophils. Whereas IL-17A is largely produced by lymphocytes, the IL-17 receptor is expressed in epithelial cells, fibroblasts, and hematopoietic cells. Loss of the IL-17RA in mice results in impaired antimicrobial responses to extracellular bacteria. In the context of H. pylori infection, this is compounded by extensive inflammation in Il17ra-/- mice. In this study, Foxa3creIl17rafl/fl (Il17raΔGI-Epi) and Il17rafl/fl (control) mice were used to test the hypothesis that IL-17RA signaling, specifically in epithelial cells, protects against severe inflammation after H. pylori infection. The data indicate that Il17raΔGI-Epi mice develop increased inflammation compared with controls. Despite reduced Pigr expression, levels of IgA increased in the gastric wash, suggesting significant increase in Ag-specific activation of the T follicular helper/B cell axis. Gene expression analysis of stomach tissues indicate that both acute and chronic responses are significantly increased in Il17raΔGI-Epi mice compared with controls. These data suggest that a deficiency of IL-17RA in epithelial cells is sufficient to drive chronic inflammation and hyperactivation of the Th17/T follicular helper/B cell axis but is not required for recruitment of polymorphonuclear neutrophils. Furthermore, the data suggest that fibroblasts can produce chemokines in response to IL-17 and may contribute to H. pylori-induced inflammation through this pathway.

Effect of light and noise pollution on oxidative stress and proximate composition in Dreissena polymorpha

In this study, it was aimed to determine the effect of sound and light pollution on Zebra mussel (Dreissena polymorpha) by biomarker and nutritional composition. For this purpose, an experimental design was carried out in laboratory conditions where the model was exposed to live sound, light, and combined sound + light parameters for 30 min every hour over 96 h. Changes in the biomarkers superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities, as well as glutathione (GSH) and thiobarbituric acid (TBARS) levels, were assessed in the model organism exposed to sound, light, and sound + light pollution. The changes in protein and fat amounts were determined. When the changes in biomarkers were examined statistically compared to the control, there was no significant decrease in SOD activity (p < 0.05), a significant increase in CAT activity in sound and sound+light, but a significant decrease in light (p < 0.05), no significant difference in GPx activity (p > 0.05). It was determined that there was a significant increase in GSH and TBARS levels in all three parameters. Significant changes were also observed in the proximate composition. As a result, exposure to sound, light and sound+light pollution caused an increase in oxidative damage and decreases in protein and lipid composition were detected.

Anti-inflammatory and immunoregulatory effects of colistin sulphate on human PBMCs.

In previous studies, CST has been identified as having an immunostimulatory effect on Caenorhabditis elegans and macrophage of rats. Here, we further investigated its immunomodulatory effects on human peripheral blood mononuclear cells (PBMCs). LPS-stimulated PBMCs inflammatory model was established. Flow cytometry was applied to measure phagocytosis of PBMCs. Cytokine mRNA and protein expression levels of LPS-stimulated PBMCs with or without CST were measured by qRT-PCR and ELISA. The transcriptomic profile of CST-treated PBMCs was investigated by RNA-sequencing. Gene Ontology (GO) and Kyoto Encylopedia of Genes and Genomes (KEGG) were applied to find potential signalling pathways. PBMCs showed a significant increase in phagocytic activity at 6 h after being incubated with CST at the concentration of 10 μg/mL. In the presence of LPS, CST maintained and promoted the expression of TNF-α and chemokine CCL24. The content of pro-inflammatory cytokines, such as IL-1β, IL-6 and IFN-γ, which were released from LPS-stimulated PBMCs, was reduced by CST at 6 h. Anti-inflammatory cytokines, such as IL-4, IL-13 and TGF-β1, were significantly increased by CST at 24 h. A total of 277 differentially expressed immune-related genes (DEIRGs) were detected and cytokine-cytokine receptor interaction was highly enriched. CST presented obvious anti-inflammatory and immunoregulatory effects in LPS-induced PBMCs inflammatory model not only by improving the ability of PBMCs to clear pathogens but also by decreasing pro-inflammatory cytokines and increasing anti-inflammatory cytokines. And the mechanism may be related to cytokine-cytokine receptor interaction.

Effect of medium supplementations and extraction conditions on cellulase production through solid state fermentation of oil palm empty fruit bunches

In this study, cellulase enzyme was produced through solid state fermentation (SSF), employing oil palm empty fruit bunches (EFB) as the primary substrate. Two key aspects were explored to enhance crude enzyme yield, which are medium supplementation effects during SSF and cellulase recovery. During medium supplementation, glucose and/or Tween 80 were added alongside EFB substrate and other nutrients. Enzyme yield was determined using a filter paper assay and expressed as enzyme activity. The initial addition of glucose during fermentation led to increased crude enzyme activity, as measured by the filter paper assay. The peak crude enzyme activity was observed with the addition of 3 mg of glucose, with higher amounts showing no further increase in activity. Conversely, the addition of Tween 80 did not yield any significant increase in crude enzyme activity across all concentrations tested. The extraction conditions were varied to study cellulase recovery, specifically by adjusting the solid‐to‐solvent ratio and the number of extraction stages. Higher enzyme activity was achieved with lower solid‐to‐liquid ratios, as the increased solvent volume facilitated greater enzyme extraction. However, increasing the number of extraction steps did not significantly affect the resulting cellulase activity. Overall, this research underscores the need for further process optimization for cellulase production via SSF, utilizing the widely available EFB in Indonesia.

Valorization of sugarcane bagasse for high-yield production of laccase through Aspergillus terreus for effective azo dye decolourization

Synthetic dyes such as azo dyes are significant pollutants in the wastewater released from various textile industries. The low biodegradability and production from synthetic sources with high shelf life make azo dyes a challenging material for degradation. This study used chemically mutated Aspergillus terrus in the laccase production under solid-state fermentation using sugarcane bagasse. Initially, the wild-type strain produced a laccase activity of 4.12 U/mL. Later, the alkaline pretreatment of sugarcane bagasse showed a significant increase in laccase activity by 38.9%. Further, random mutagenesis treatment with 100 mM EMS generated a hyper laccase-producing strain with a 2.3-fold increment in laccase activity compared to the wild-type strain. The enzyme displayed optimal activity at pH 6.5 and 35 °C. The metal ions such as Fe3+ (29.4 U/mL), Fe2+ (20.8 U/mL) and Cu2+ (18.05 U/mL) showed positive effects on laccase activity. The crude laccase was used to bioremediate Congo red, a prominent azo dye used in textile and pharmaceutical industries. The preliminary studies with a crude enzyme displayed 68.86% dye decolourization after 24 h of incubation. Additionally, with Taguchi orthogonal array optimization experiments, the maximal dye decolorization of 78.24% was achieved by maintaining crude enzyme concentration (20 U), dye concentration (25 mg/L) and pH 4.5.

The protein methyltransferase TrSAM inhibits cellulase gene expression by interacting with the negative regulator ACE1 in Trichoderma reesei

Protein methylation is a commonly posttranslational modification of transcriptional regulators to fine-tune protein function, however, whether this regulation strategy participates in the regulation of lignocellulase synthesis and secretion in Trichoderma reesei remains unexplored. Here, a putative protein methyltransferase (TrSAM) is screened from a T. reesei mutant with the ability to express heterologous β-glucosidase efficiently even under glucose repression. The deletion of its encoding gene trsam causes a significant increase of cellulase activities in all tested T. reesei strains, including transformants of expressing heterologous genes using cbh1 promotor. Further investigation confirms that TrSAM interacts with the cellulase negative regulator ACE1 via its amino acid residue Arg383, which causes a decrease in the ACE1-DNA binding affinity. The enzyme activity of a T. reesei strain harboring ACE1R383Q increases by 85.8%, whereas that of the strains with trsam or ace1 deletion increases by more than 100%. By contrast, the strain with ACE1R383K shows no difference to the parent strain. Taken together, our results demonstrate that TrSAM plays an important role in regulating the expression of cellulase and heterologous proteins initiated by cbh1 promotor through interacting with ACE1R383. Elimination and mutation of TrSAM and its downstream ACE1 alleviate the carbon catabolite repression (CCR) in expressing cellulase and heterologous protein in varying degrees. This provides a new solution for the exquisite modification of T. reesei chassis.

Functional identification of CCR1 gene in apple (Malus halliana) demonstrates that it enhances saline-alkali stress tolerance

BackgroundLignin is a complex aromatic polymer that plays an important biological role in maintaining plant structure and defending plants against biotic and abiotic stresses. Cinnamoyl-CoA reductase (CCR) is a key enzyme involved in the lignin synthesis-specific pathway and regulates lignin biosynthesis and accumulation.MethodsBased on transcriptome data, MhCCR1, which was significantly induced by saline-alkali stress, was cloned from Malus halliana. The physicochemical properties, evolutionary relationships and cis-acting elements were analyzed. We obtained transgenic materials by heterologous transformation of MhCCR1 into Arabidopsis thaliana and tobacco, as well as homologous transformation into apple callus, and characterized their saline-alkali stress resistance by a series of physiological and biochemical experiments. And yeast two-hybridization technique was applied to screen and validate the interacting proteins.ResultsWe found that overexpression of MhCCR1 enhanced the tolerance of A. thaliana, tobacco and apple calli under saline-alkali stress, and caused a variety of physiological and biochemical changes. As compared to the wild type, the transgenic plants showed better growth, higher lignin, chlorophyll and proline contents, lower conductivity and MDA content, and significant increase in antioxidant enzyme activities (SOD, POD, CAT) in the transgenic lines under stress condition. In addition, expression of saline-alkali stress-related genes in overexpressed A. thaliana were also higher than in WT, including the antioxidant genes, the Na+ transporter genes, and the H+-ATPase genes, while expression of the K+ transporter genes displayed opposite changes. Meanwhile, the expression levels of genes related to lignin synthesis, AtPAL1, AtCOMT, AtC4H, At4CL1, and AtCCOAOMT, were also significantly up-regulated. At last, the Y2H experiment confirmed the interaction between MhCCR1 and MhMYB4, MhMYB1R1, MhHXK, and MhbZIP23 proteins.ConclusionsThese results suggest that MhCCR1 may play a positive regulatory role in saline-alkali tolerance of transgenic lines by regulating the lignin content, osmoregulatory substances, chlorophyll content, antioxidant enzyme activities, and genes related to saline-alkali stress, thus providing excellent resistance genes for the stress-responsive regulatory network of apples, and providing a theoretical basis for the cultivation of saline and alkali-resistant apple varieties.Graphical

Abstract 5473: Combined inhibition of MDM2 and PARP lead to a synergistic anti-tumoral response in p53 wild-type rhabdomyosarcoma

Abstract Targeting MDM2-p53 interaction to enhance p53 activity represents an attractive antitumoral strategy for p53 wild-type tumors. However, early-phase clinical trials in hematological and solid cancers have shown limited efficacy of MDM2 inhibitors highlighting the necessity for exploring more effective combinations with additional agents. In this study, we provide results from a 22-drug combination screening demonstrating the therapeutic potential of combining Siremadlin (MDM2 inhibitor) and Olaparib (PARP inhibitor) for treating p53 wild-type rhabdomyosarcoma (RMS). Cell survival, cell death and apoptosis analysis revealed synergistic effects when combining Siremadlin and Olaparib in vitro. Combination of both drugs resulted in a significant increase of p53 activity as evidenced by p53 accumulation and K382 acetylation along with an increased expression of p53 targets. Finally, combination of both drugs resulted in significant reduction of tumor growth and increased overall survival outperforming individual treatments and control groups. In summary, our study demonstrates, for the first time, the synergistic effect of the combination between Siremadlin and Olaparib in the inhibition of p53 wild-type RMS tumor growth in vitro and in vivo. These findings advocate for the exploration of this drug combination in clinical trials and underscore the need for further investigation in tumors sharing similar molecular features. Effects of a phytogenic diet on growth, haemato-immunological parameters, expression of immune- and stress-related genes, and resistance of Oncorhynchus mykiss to Lactococcus garvieae infection

Phytogenic additives have been implicated in aquafeed owing to their proven growth-stimulating, antimicrobial, antioxidant, and immune-enhancing efficacies. Herein, a short-term feeding experiment (21 days) was designed to explore the effects of a phytogenic additive, PASSIF MOOD® (PM, a mixture of Valeriana officinalis and Passiflora incarnata extracts), on growth, haemato-biochemical variables, immunity, and mRNA expression responses of rainbow trout fingerlings. After feeding, fish were experimentally infected with Lactococcus garvieae and monitored for an additional 20 days. Six groups were fed a commercial diet supplied with different PM levels as 0, 470, 940, 1410, 1880, and 2350 mg/kg and labeled as PM0 (control), PM1, PM2, PM3, PM4, and PM5 groups, respectively. Results revealed significant improvements in the growth of PM-supplied groups compared to the control. Dietary PM significantly increased RBC count, hemoglobin, and hematocrit values compared to those fed on a control diet. Stress indicators such as blood glucose and cortisol levels were significantly decreased in all PM groups than in controls. Moreover, there were significant increases in myeloperoxidase, respiratory burst, and lysozyme activities in PM groups than the control. The highest expression levels of the lysozyme II gene were found in the PM3, PM4, and PM5 groups. Moreover, the mRNA transcripts of il-10 and IgM heavy chain genes were significantly increased in all PM groups compared to the controls. Alternatively, the expression of tnf-α and hsp70 genes were significantly decreased in all PM groups compared to the controls. After challenge, the survival rates in PM0, PM1, PM2, PM3, PM4, and PM5 groups were 37.34%, 84.0%, 85.34%, 68.0%, 64.0%, and 36.0%, respectively, suggesting enhancement of the fish resistance in PM1, PM2, PM3, and PM4 groups compared to control. The results suggest the growth-promoting, antistress, and immunomodulatory roles of the tested phytogenic additive.

In Vitro Investigation of the Cytotoxic and Antiproliferative Effects of Haberlea rhodopensis Total Extract: A Comparative Study

Haberlea rhodopensis Friv., known also as Rhodope silivryak and the Orpheus flower, is a Balkan endemic “resurrecting” plant belonging to the Gesneriaceae family. In folk medicine, the leaves of Haberlea rhodopensis Friv. were widely used to treat wounds and some infectious diseases of stock such as foot-and-mouth disease and hoof rot, while the herb of Haberlea rhodopensis Friv. is still used to cleanse the stomach, liver, kidneys, and blood vessels. Because of the content of myconoside, during the last decade, Haberlea rhodopensis Friv. extracts have been recognized as valuable cosmetic ingredients. In the present study, we aim to (i) evaluate the cytotoxic and antiproliferative activity of two herb extracts of Haberlea rhodopensis Friv. that are commercially used for the preparation of cosmetic ingredients on different cancer cells, with one normal cell line used as a reference, and (ii) compare the investigated effects with those observed for the reference anticancer, non-selective compound doxorubicin. Herein, we observed a decrease in the inhibitory activity of both extracts compared to those of doxorubicin against all tested cell lines. However, the myconoside-enriched Haberlea rhodopensis Friv. plant Extract 2 (designated also as M2) showed increased inhibitory activity (cytotoxicity and antiproliferative effects) compared to the Haberlea rhodopensis Friv. plant Extract 1 (designated also as E1). Moreover, the Haberlea rhodopensis Friv. plant Extract 2 showed a significant increase in cytotoxicity (at 24 h) and antiproliferative activity (at 48 and 72 h post-treatment) at its highest-tested concentration of 100 µg/mL compared to Haberlea rhodopensis Friv. plant Extract 1.

Turning fishery waste into aquafeed additives: Enhancing shrimp enzymes immobilization in alginate-based particles using electrohydrodynamic atomization

Alternative protein ingredients are gradually replacing fish meal in aquafeeds. However, farmed animals often struggle to digest the provided feed, which leads to inefficient nutrient assimilation. This results in a high feed conversion ratio and in water quality deterioration. Fish waste-derived proteases show great potential as bioactive ingredients to enhance protein digestibility. To achieve this, a suitable carrier is essential to include exogenous proteases in the fish feed formulation. Therefore, this research aimed to immobilize a shrimp enzyme extract into different alginate-based microcapsules through electrodynamic atomization and compare their structural and functional properties. Morphology, physicochemical and thermal properties, and in vitro protein release were evaluated for the following microcapsules made from various materials: alginate (A), alginate-chitosan (AC), alginate-bentonite (AB), alginate-bentonite previously subjected to a sonication treatment (ABs), and a combination of all materials (ABsC or ABC). The average sizes ranged from 360 to 790 μm (wet) and 260–516 μm (freeze-dried), confirming the expected differences, with larger sizes for those including chitosan and bentonite. ABsC proved to be the most suitable system for immobilizing exogenous proteases, as these particles exhibited high encapsulation efficiency in both wet and freeze-dried versions and displayed controlled protein release under typical conditions of the proximal intestine. This was further confirmed through an in vivo trial. Two bioassays were conducted with tilapia (Oreochromis niloticus) (n = 42, 2.6 ± 0.58 g) to evaluate the distribution of ABsC-rodamine labeled particles in digestive organs and digestive enzyme activity. ABsC remained in the intestine for over 7 h, and a significant increase in total proteolytic activity was observed 60 min after feeding. These findings suggest that ABsC microcapsules could serve as effective carriers for exogenous proteases in feeds for aquaculture.

Adaptive changes in immunological, hematological and metabolic profiles of pregnant cows

Pregnancy triggers a series of complex interactions within the body, involving both the immune system and metabolic processes. In the case of pregnant cows, these mechanisms are essential for supporting the developing fetus and maintaining the overall health of the mother. The aim of the work was to study the hematological, biochemical, and immunological parameters of cows at different periods of pregnancy. Research was conducted on cows (Bos Taurus) of the Ukrainian Black-and-White dairy breed. Groups were formed based on the principle of analogs. The research was carried out using the methodology of group-period experiments. The control group consisted of non-pregnant cows. The first observation period was conducted on pregnant cows with confirmed pregnancy (30 days). The second observation period was carried out at 90 days of pregnancy, the third at 180 days of gestation, and the fourth at 270 days. Laboratory studies included immunological research conducted in the specialized laboratory of immunology for reproduction animals. The first notable observation was the gradual increase in the population of CD3+ lymphocytes from the beginning of gestation, which remained stable until the 90th day of pregnancy and then gradually decreased. The population of B cell immune cells CD22+ had its peculiarities: dynamic and stable growth from the start of pregnancy until the middle of the second trimester (180 days), followed by a gradual decrease until the onset of labor. The redistribution of T and B lymphocytes occurred with dynamic fluctuations in the number of undifferentiated 0-lymphocytes. The processes of transformation and adaptation of homeostasis affected both enzyme activity and indicators of mineral metabolism. Thus, from the beginning of pregnancy (from the 30th day of gestation), the experimental cows showed a significant increase in alkaline phosphatase activity (P &lt; 0.05), alanine aminotransferase (P &lt; 0.05), and aspartate aminotransferase activity (P &lt; 0.05). During the pregnancy of cows, body changes occur. From the early period of gestation throughout the entire period of embryogenesis, changes in the system of hematopoiesis, alterations in the activity of enzymes, exchange of Ca and P, accompanied by changes in immunological reactions take place. These processes are adaptive and ensure the maintenance of the homeostasis system. The perspective of further research will aim at studying changes in immunological and biochemical parameters during physiological pregnancy and when it is complicated.

Copper-substituted magnetite as a Fenton-like catalyst boosted with electromagnetic heating

Copper-substituted magnetite samples Fe3-xCuxO4 (x = 0.0; 0.02; 0.05; 0.1) were synthesized using the co-precipitation method. XRD data revealed that the synthesized samples retain the cubic spinel structure of magnetite. The obtained materials were tested as heterogeneous Fenton catalysts to destroy organic pollutants and inactivate bacteria. The model organic pollutants were oxytetracycline, Congo Red dye, and Methylene Blue dye having neutral, anionic, and cationic nature, respectively. These organic pollutants differ in their adsorption on magnetite surface: oxytetracycline < Congo Red < Methylene Blue. The same order stays for their decomposition rates. The copper-substituted magnetite catalysts also ensure effective inactivation of E.coli bacteria. A contact time of 60 min is sufficient to reduce the number of bacteria by 6-log. In all tests, the catalytic activity of magnetite increases with the increase in Cu content. The maximum catalytic activity was recorded for the Fe2.9Cu0.1O4 sample. Since the copper-substituted magnetite samples are ferromagnetic, they absorb a high-frequency electromagnetic field. Electromagnetic heating results in a significant increase in catalytic activity: the rate constants of the studied catalytic reactions are increased by 1.5–5 times. Thus, copper-doped magnetite is a hetero-Fenton catalyst capable of increasing activity by electromagnetic heating. The adjustable catalytic activity may be useful in water disinfection, where bacterial load varies greatly.

Advances in Understanding the Antioxidant and Antigenic Properties of Egg-Derived Peptides.

Pepsin, trypsin and proteinase K were used in the present study to hydrolyse the proteins from whole eggs, yolks or whites, and the resulting hydrolysates were characterised in terms of antioxidant and IgE-binding properties, using a combination of in vitro and in silico methods. Based on the degree of hydrolysis (DH) results, the egg yolk proteins are better substrates for all the tested enzymes (DH of 6.2-20.1%) compared to those from egg whites (DH of 2.0-4.4%). The SDS-PAGE analysis indicated that pepsin and proteinase K were more efficient compared to trypsin in breaking the intramolecular peptide bonds of the high molecular weight egg proteins. For all the tested substrates, enzyme-assisted hydrolysis resulted in a significant increase in antioxidant activity, suggesting that many bioactive peptides are encrypted in inactive forms in the parent proteins. The hydrolysates obtained with proteinase K exhibited the highest DPPH radical scavenging activity (124-311 µM Trolox/g protein) and the lowest residual IgE-binding capacity. The bioinformatics tools revealed that proteinase K is able to break the integrity of the main linear IgE-binding epitopes from ovalbumin and ovomucoid. It can be concluded that proteinase K is a promising tool for modulating the intrinsic properties of egg proteins.

Tinnitus-related increases in single-unit activity in awake rat auditory cortex correlate with tinnitus behavior

Tinnitus is known to affect 10–15 % of the population, severely impacting 1–2 % of those afflicted. Canonically, tinnitus is generally a consequence of peripheral auditory damage resulting in maladaptive plastic changes in excitatory/inhibitory homeostasis at multiple levels of the central auditory pathway as well as changes in diverse nonauditory structures. Animal studies of primary auditory cortex (A1) generally find tinnitus-related changes in excitability across A1 layers and differences between inhibitory neuronal subtypes. Changes due to sound-exposure include changes in spontaneous activity, cross-columnar synchrony, bursting and tonotopic organization. Few studies in A1 directly correlate tinnitus-related changes in neural activity to an individual animal's behavioral evidence of tinnitus. The present study used an established condition-suppression sound-exposure model of chronic tinnitus and recorded spontaneous and driven single-unit responses from A1 layers 5 and 6 of awake Long-Evans rats. A1 units recorded from animals with behavioral evidence of tinnitus showed significant increases in spontaneous and sound-evoked activity which directly correlated to the animal's tinnitus score. Significant increases in the number of bursting units, the number of bursts/minute and burst duration were seen for A1 units recorded from animals with behavioral evidence of tinnitus. The present A1 findings support prior unit recording studies in auditory thalamus and recent in vitro findings in this same animal model. The present findings are consistent with sensory cortical studies showing tinnitus- and neuropathic pain-related down-regulation of inhibition and increased excitation based on plastic neurotransmitter and potassium channel changes. Reducing A1 deep-layer tinnitus-related hyperactivity is a potential target for tinnitus pharmacotherapy.

Speciation, bioaccumulation, and toxicity of the newly deposited atmospheric heavy metals in soil-earthworm (Eisenia fetida) system near a large copper smelter

The speciation, bioaccumulation, and toxicity of the newly deposited atmospheric heavy metals in the soil-earthworm (Eisenia fetida) system were investigated by a fully factorial atmospheric exposure experiment using soils exposed to 0.8-year and 1.8-year atmospheric depositions. The results shown that the newly deposited metals (Cu, Cd, and Pb) primarily accumulated in the topsoil (0–6 cm) and were present as the highly bioavailable speciation. They can migrate further to increase the concentrations of Cu, Cd, and Pb in soil solution of the deeper layer (at 10 cm) by 12 %–436 %. Earthworms tended to preferentially accumulate the newly deposited metals, which contributed 10 %–61 % of Cu, Cd, and Pb in earthworms. Further, for the unpolluted and moderately polluted soils, the newly deposited metals induced the significant oxidative stress in earthworms, resulting in significant increases in antioxidant enzyme activities (SOD, CAT, and GSH-Px). No significant differences were observed in the levels of heavy metals in soil solutions, bioaccumulation, and enzyme activities in earthworms exposed to 0.8-year and 1.8-year depositions, indicating the bioavailability of atmospheric metals deposited into soils was rapidly decreased with time. This study highlights the high bioaccumulation and toxicity of heavy metals to earthworm from the new atmospheric deposition during the earthworm growing period.
