The Promising Effects of Erdosteine and Vitamin B in the Liver Ischemia/Reperfusion Model in Anesthetized Rats.

Background and Objectives: Erdosteine (Erd) is an antioxidant and anti-inflammatory drug. Vitamin B has been reported to exert anti-inflammatory and antioxidant effects. In this study, we investigated the effect of erdosteine and vitamin B complex on a liver ischemia/reperfusion (I/R) model. Materials and Methods: Thirty-two Wistar Albino male rats weighing 350-400 g were used. The animals were randomly selected and divided into four groups. The groups are as follows: first group (Sham), second group (I/R), third group (I/R + vit B), and fourth group (I/R + vit B + Erd). Rats were subjected to 45 min of hepatic ischemia, followed by a 45 min reperfusion period in the I/R and Vitamin B + Erd groups. An amount of 150 mg/kg/day of erdosteine was given orally for 2 days, and 0.05 mL/kg of i.p. vitamin B complex was given 30 min before the reperfusion. Serum biochemical parameters were measured. Serum Total Antioxidant Status (TAS) and Total Oxidant Status (TOS) were measured, and the Oxidative Stress Index (OSI) was calculated. Hepatic tissue samples were taken for the evaluation of histopathological features. Results: In terms of all histopathological parameters, there were significant differences in the I/R + vit B group and I/R + vit B + Erd group compared with the I/R group (p < 0.01). In terms of aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), TNF-alpha, and IL-6 levels, there were significant differences between the I/R group and treatment groups (p < 0.01). The lowest TOS and OSI levels were obtained in the treatment groups, and these groups had statistically significantly higher TAS levels compared with the sham and I/R groups (p < 0.01). Conclusions: As a preliminary experimental study, our study suggests that these agents may have potential diagnostic and therapeutic implications for both ischemic conditions and liver-related diseases. These results suggest that the combination of vit B + Erd may be used to protect against the devastating effects of I/R injury. Our study needs to be confirmed by clinical studies with large participation.

Higher oxidative stress and inflammation in obese compared to lean patients with type 2 diabetes mellitus

AimTo compare mRNA [messenger RNA] expression of RELA, NFκB1, TNF-α, IL-6 and MCP-1 in whole blood & serum Total Antioxidant status [TAS] in newly diagnosed lean and obese patients with T2DM. MethodsNewly diagnosed treatment naïve patients of T2DM were enrolled in this study. The patients were divided into two groups of 30 patients each, lean (BMI< 18.5 kg/m2) and obese (BMI >25 kg/m2) groups. mRNA expression of RELA, NFκB1, TNF-α, IL-6 and MCP-1 was measured by real time PCR. Serum TAS was measured using a commercially available kit. ResultsThere was a 2.7-fold increase in mRNA expression of RELA in obese group compared to the lean group. There was a 1.3-fold increase in mRNA expression of NFκB1, a 3.24-fold increase in mRNA expression of TNF-α, a 4.7-fold increase in mRNA expression of IL 6 and a 3.8-fold increase in mRNA expression of MCP-1 in obese group compared to the lean group. Mean fasting serum insulin levels were 16.07 ± 8.39 μIU/mL in the lean group and 27.11 ± 4.91μIU/mL in the obese group (p = 0.001). Mean TAS level was 5.39 ± 2.28 μM Trolox Equivalents in the obese group and 3.85 ± 3.33 μM Trolox Equivalents in the lean group (p = 0.001). ConclusionInflammation and OS is higher in obese patients of T2DM compared to lean patients of T2DM. This could be the result of excess adipokines production or resistance to the anti-inflammatory effects of insulin with multiple explanations. Our study suggests a difference in the pathogenic mechanism in lean patients when compared with obese T2DM patients.

Serum testosterone levels and oxidative stress in type 1 diabetes, type 2 diabetes, and obesity.

Obesity, type 1 diabetes mellitus (T1DM), and type 2 diabetes mellitus (T2DM) are metabolic diseases that continue to be a global problem. Testosterone levels in men are affected by several factors, including obesity and DM. Although the relationship between diabetes and testosterone is not fully understood, oxidative stress is thought to play a major role. The aim of this study was to compare serum testosterone levels and oxidative stress markers [total antioxidant status (TAS), total oxidant capacity (TOS), oxidative stress index (OSI), and ischaemic modified albumin (IMA)] among the control group and experimentally induced obese, T1DM, and T2DM rats. The study included 28 male Sprague-Dawley rats divided into 4 groups: the obesity group were fed a high-fat diet (HFD), the T2DM group received a HFD plus a single dose of streptozocin (STZ), the T1DM group received only STZ, and there was a control group. Serum testosterone, TAS, TOS, OSI, and IMA were analysed. Serum testosterone levels were lower in the T1DM and T2DM groups compared to the control and obesity groups. The TOS levels were highest in the T2DM group, followed by the T1DM group, the obesity group, and finally the control group. No significant difference was found between the obesity group and the control group in terms of TOS levels. Regarding TAS levels, the order observed was control group > obesity group > T2DM > T1DM. Testosterone was positively correlated with TAS and negatively correlated with TOS and OSI. Increased oxidative stress in diabetes may be an important factor that decreases serum testosterone levels.

Prolidase could be considered a sign of inflammation associated with cigarette smoking.

Smoking causes inflammation, thickening, and narrowing of the airways. This inflammatory process is a reaction to free radicals and oxidants. Smoking affects collagen metabolism and tissue remodeling. Prolidase enzyme hydrolyzes iminodipeptides with hydroxyproline and C terminal proline. It plays a crucial role in the metabolism of collagen and the remodeling of the matrix. The present study aims to reveal the association of prolidase with inflammation caused by smoking and to compare serum prolidase levels with oxidative-antioxidative status in healthy individuals. A total of 76 participants (38 smokers and 38 nonsmokers) were involved in the present study. Serum cotinine levels were measured to show the exposure to nicotine in tobacco smoke by using the competitive inhibition enzyme immunoassay method. Serum prolidase, total oxidant status (TOS), and total antioxidant status (TAS) were determined by the enzyme-linked immunosorbent (ELISA) method, respectively. The correlation between smoking, serum prolidase levels, TOS, and TAS was investigated. TAS and serum prolidase levels of smokers were considerably lower than those in non-smokers (p < 0.001, p = 0.012 respectively). However, no differences were observed in TOS between the two groups. There was no statistically significant correlation between serum prolidase levels, TAS, and TOS. Moreover, no relationship was observed between respiratory function parameters and serum prolidase levels. To the best of our knowledge, the present study is the first study to demonstrate the role of prolidase in smoking-related inflammation. The results achieved in the present study suggest that smoking creates an imbalance in the oxidant-antioxidant activity. Smoking decreases prolidase levels, leading to decreased collagen turnover. Chronic pulmonary disease might be related to this decrease in collagen turnover.

Oxidative stress markers in breast cancer

Purpose: Oxidative stress is thought to play a role in the etiology of breast cancer. The aim of this study was to compare serum Total Antioxidant Status (TAS), Total Oxidant Status (TOS), Oxidative Stress Index (OSI) and Ischemia Modified Albumin (IMA) levels in breast cancer patients and healthy women and to determine whether there is a relationship between oxidative stress markers and breast cancer.&#x0D; Materials and methods: Newly diagnosed 106 breast cancer patients at the Bozyaka Training and Research Hospital General Surgery Clinic and 30 healthy women were included in the study. Serum levels of IMA, TAS, TOS were analyzed by spectrophotometric methods. &#x0D; Results: IMA, TOS and OSI values were found significantly higher in the breast cancer group. Although the TAS values in patients with breast cancer were lower than the control group, the difference was not statistically significant. &#x0D; Conclusions: In our study, it was seen that oxidative stress levels increased in breast cancer patients due to decreased TAS, significantly increased IMA, TOS and OSI results. Therefore, we think that increased oxidative stress may be related to breast cancer and that IMA, TOS and OSI can be used as markers of oxidative stress to differentiate breast cancer patients from healthy women.&#x0D; Keywords: breast cancer, cancer, biomarkers, ischemia-modified albumin, oxidative stress

Effects of Quince Gel and Hesperidin Mixture on Experimental Endometriosis.

Endometriosis (EM) is the presence of endometrial tissue outside the uterus. This study aimed to examine the effects of quince gel and hesperidin treatment on uterine tissue in an experimental endometriosis model. Thirty-two rats were categorized into four groups as sham, EM, EM+quince gel (QG), and EM+QG+Hesperidin (HES). The endometriosis (EM) model was induced with surgical intervention. Estradiol benzoate (EB) was used to induce endometrial hyperplasia. In the EM group, EB was given to rats for 7 days. The EM+QG group received 2 cc QG for 21 days. HES treatment was given for 21 days after EM induction. At the end of the experiment, blood was taken from the animals and the serum total antioxidant status (TAS) and total oxidant status (TOS) values were studied. Uterine tissues were dissected and processed for histological paraffin embedding. Tissues were fixed in 4% glutaraldehyde solution and processed for ultrastructural analysis. After EM, QG and HES treatment significantly increased the TAS and decreased the TOS value. EM caused epithelial and glandular degeneration, thinning of the basal membranes, and vascular dilatation with increased fibrosis and edema. QG+HES restored the pathology and showed protective effects in uterine tissues. Caspase-3 expression was increased in the epithelium, glands, and muscle layers of the EM group. In EM+QG+HES, hesperidin protected cell survival and decreased Caspase-3 expression in uterine tissues. TNF-α expression was intense in inflammatory cells and the muscle layer in the EM group. HES reduced inflammation by decreasing the TNF-α expression. MAPK expression was increased after EM induction in epithelial, glandular, and inflammatory cells in the EM group. After HES treatment, MAPK expression was mainly negative in cells of uterine tissue in the EM+QG+HES group. Ultrastructurally, in the EM group, organelles were disrupted and dilated and degenerated after EM induction. QG and HES treatment improved cellular organelles. Local vaginal applications can be an alternative treatment method in the endometriosis model via QG+HES treatment promoting cell proliferation and angiogenesis and preventing cell death.

Relationships between Serum Biomarkers of Oxidative Stress and Tobacco Smoke Exposure in Patients with Mental Disorders.

The role of cigarette smoking as an aggravating factor of systemic oxidative stress in patients with mental disorders has not been extensively investigated, although significantly higher rates of smoking are recorded in these subjects in comparison with the general population. In the present study, we tested the hypothesis that smoking might be an exacerbator of systemic oxidative stress, being directly correlated with the degree of exposure to tobacco smoke. We analyzed, in 76 adult subjects from a public health care unit, the relationships between serum cotinine levels as a marker of tobacco smoke exposure, and three biomarkers of oxidative stress: the serum glutathione (GSH), the advanced oxidation protein products (AOPPs), and the total serum antioxidant status (FRAP). The results indicate that the degree of tobacco smoke exposure was inversely associated with GSH levels in both passive and active smokers, suggesting that smoke particulate components' toxicity is associated with a systemic GSH depletion. Paradoxically, the lowest AOPP levels which were positively associated with GSH, were recorded in active smoking patients whereas in passive smokers individual values of AOPPs decreased along with the increase in GSH levels. Our data suggest that an enhanced inhalation of particulate constituents of cigarette smoke could induce critical changes in systemic redox homeostasis and GSH can no longer exert its antioxidant role.

Effects of chronic core training on serum and erythrocyte oxidative stress parameters in amputee football players.

Objective: The positive impact of aerobic exercise on blood oxidative stress parameters is well documented. However, the effect of core exercises on these parameters in amputee football players (AF) remains unclear. Therefore, this study aims to investigate the impact of core exercises on blood oxidative stress parameters in this population. Methods: Experimental method was adopted in the study. Eleven elite AF players participated in the study. The participants were divided randomly into two groups a core exercise group (CEG) and a control group (CG). Blood measurements were taken before and after the 8-week core exercise program. Blood measurements included erythrocyte Total Oxidant Status (eTOS), erythrocyte Total Antioxidant Status (eTAS), erythrocyte oxidative stress index (eOSI), serum nitric oxide (sNO), serum Total Oxidant Status (sTOS), serum Total Antioxidant Status (sTAS), serum oxidative stress index (sOSI), serum total thiol (sTT), serum native thiol (sNT), and serum disulfide (sDS) parameters were studied. Results: According to the results of the study, a significant difference was found between the 0th and eighth week pre-aerobic training load (ATL) sTOS (p = .028) values of CEG values. A significant difference was found in sTOS (p = .028) and sOSI (p = .028) values after the 0th and eighth-week pre-ATL. A significant difference was found in the sTOS (p = .043) and sOSI values (p = .043) of CG at week 0th and eighth-week pre-ATL. Conclusion: Overall, the results suggest that core exercises had a positive effect on blood oxidative stress parameters in AF players by reducing blood total oxidant levels.

ВЫРАЖЕННОСТЬ ОКСИДАТИВНОГО СТРЕССА У БОЛЬНЫХ С ИНФАРКТОМ МИОКАРДА НА ФОНЕ ХОБЛ

Introduction. Oxidative stress is a pathogenetic link in the development of many diseases, including pathology of the respiratory system and cardiovascular events. Comorbidity is a topical problem of modern medicine. One of the frequent combinations is myocardial infarction against the background of chronic obstructive pulmonary disease. The peculiarities of changes in protein and lipid peroxidation, as well as the state of antioxidant protection in comorbid patients are of interest. Aim. To study protein and lipid peroxidation processes in patients with myocardial infarction against the background of chronic obstructive pulmonary disease. Material and Methods: 195 patients with myocardial infarction against the background of chronic obstructive pulmonary disease were examined. Comparison groups were represented by 130 patients with myocardial infarction without chronic obstructive pulmonary disease and 104 patients with chronic obstructive pulmonary disease. The control group included 110 somatically healthy individuals. The level of malondialdehyde was assessed as a marker of lipid peroxidation, the level of deep protein oxidation products was assessed as a marker of free radical oxidation of proteins. Antioxidant protection was assessed using plasma level of total superoxide dismutase. To evaluate non-enzymatic components of antioxidant protection, total serum antioxidant status was determined by spectroscopy. The data were statistically processed using the SPSS 26.0 software package. Results and discussion. In patients with chronic obstructive pulmonary disease the levels of protein and lipid oxidation markers were higher than in myocardial infarction. The highest levels were noted in comorbid patients. Antioxidant levels were decreased in all groups compared to controls. Patients with monosomatic myocardial infarction had lower levels of antioxidants compared with patients with chronic obstructive pulmonary disease. The lowest values were observed in the group of comorbid patients. Conclusion. Chronic obstructive pulmonary disease is accompanied by a more pronounced activation of lipid and protein peroxidation processes, but accompanied by activation of adaptive mechanisms, which is not observed in acute coronary pathology. The greatest severity of oxidative stress is characteristic of comorbid patients.

Comparison of oxidative stress markers in the saliva, gingival crevicular fluid, and serum samples of pregnant women with gestational diabetes and healthy pregnant women.

To compare oxidative stress (OS) markers in the saliva, gingival crevicular fluid (GCF), and serum samples of pregnant women with gestational diabetes (GDM) and healthy pregnant women and to investigate the association between periodontal health/diseases and OS and GDM. Eighty women with GDM and 80 healthy pregnant women were included in the study. Medical and clinical anamnesis was obtained from all the pregnant women included in the study, and their plaque index (PI), gingival index (GI), bleeding on probing (BoP), probing pocket depth (PPD), and clinical attachment level (CAL) measurements were performed. GCF, saliva, and serum samples were collected for the measurements of the local and systemic total antioxidant status (TAS) and total oxidant status (TOS). Clinical periodontal parameters were found to be significantly higher in the GDM group compared to the control group. The serum and saliva TAS, TOS, and TAS/TOS values were significantly lower in the GDM group than in the control group. In the analysis of the GCF samples, the mean TAS and TAS/TOS values were significantly lower and the TOS value was significantly higher in the GDM group than in the control group. The multivariate reduced model indicated that gravidity, salivary TAS/TOS, and GCF TAS were significant independent variables in the development of GDM (p < .05). Our results indicated that the OS of serum, saliva, and GCF samples increased in patients with GDM compared to healthy pregnant women. The role of local OS parameters in GDM may be associated with elevated clinical periodontal parameters.

The usability of MCP-1, fetuin-A, TAS, and TOS levels in the diagnosis of acute myocardial infarction.

Our aims were to determine whether the levels of plasma monocyte chemotactic protein-1 (MCP-1), fetuin-A, serum total antioxidant status (TAS), and serum total oxidant status (TOS) are cardiac biomarkers and to clarify their relationship with each other in acute myocardial infarction (AMI). The study included 90 participants: 60 patients with AMI [30 with and 30 without ST-segment elevation myocardial infarction (STEMI)] and 30 cardiac patients without AMI. The diagnostic values of serum Hs-cTnT, MCP-1, fetuin-A, TAS, and TOS levels in predicting AMI were evaluated statistically. Median levels of MCP-1 [120.10 ng/L (interquartile range: 76.94-230.54 ng/L)] and TOS [2.89 U/MI (IQR: 2.31-3.94 U/Ml)] were statistically higher, and median levels of fetuin-A [433.52 mg/L (IQR: 387.89-584.49 mg/L)] and TAS (3.10 ± 0.86 U/mL) were lower in patients with AMI than in controls. The parameter with the area under the curve (0.815), sensitivity (73.3%), and specificity (66.7%) closest to those of Hs-cTnT was fetuin-A, followed by MCP-1, TOS, and TAS, respectively. A one-unit increase in MCP-1 levels increased the probability of AMI by 1.023 times (p = 0.002). A one-unit increase in fetuin-A levels decreased the probability of AMI by 0.995 times (p = 0.003). A one-unit increase in serum TOS levels was 1.29 times more characteristic of STEMI than of NSTEMI (p = 0.044). MCP-1, oxidative stress parameters, and fetuin-A might support Hs-cTnT levels in the early diagnosis of AMI. Fetuin-A and MCP-1 levels may be independent risk factors for AMI, whereas TOS could be used to distinguish STEMI from NSTEMI.

Determinants Related to Oxidative Stress Parameters in Pediatric Patients with Type 1 Diabetes Mellitus

Adequate glycemic management is one of the main goals in treating type 1 diabetes mellitus (T1DM) and preventing the early onset of diabetic complications. Improperly controlled diabetes mellitus (DM) will result in oxidative stress (OS) and lead to further related health issues. Therefore, the aim of this study was to evaluate the body’s ability to defend against OS depending on the duration of T1DM, metabolic management, antioxidant intake and modern glycemic monitoring systems (GMS). The study included 103 adolescents with T1DM aged 10–17 years. The control group consisted of 65 healthy peers. The patients’ blood was assayed for antioxidant enzymes, minerals and toxic elements. In addition, their dietary intake of antioxidant components was assessed. The T1DM group had higher total oxidant status, oxidative stress index and Cu/Zn ratio values, higher concentrations of malondialdehyde and lower total antioxidant status (TAS) and chromium, zinc, superoxide dismutase and catalase levels than their healthy peers. The comparison between GMS types revealed favorable changes in OS parameters for the flash and continuous systems. Furthermore, an effect of vitamin A and C dietary intake on serum TAS concentrations was detected. More than 82% of the patients with high TAS fulfilled the estimated average requirement norm for vitamin A, and more than 60% fulfilled the vitamin C requirement. In youths with T1DM, it is advisable to observe the antioxidant activity of the body to prevent the accelerated development of diabetic complications.

Lipid Peroxidation and Serum Total Antioxidant Status in Patients with Recurrent Aphthous Ulceration Treated by Herbal Medicine

Background: Recurrent aphthous is a common but poorly understood oral mucosal disorder. Impaired oxidant/antioxidant balance is associated with the tissue damage in recurrent aphthous. Aim: To evaluate lipid peroxidation and serum total antioxidant status (TAS) in aphthous patients treated locally by herbal medicine.Patients and methods: Patients were diagnosed and treated for recurrent ulceration in Outpatient Clinic, Mosul, Iraq, from September 2011 to February 2012. A total of 42 patients with recurrent aphthous ulceration (31 males and 11 females) and 25 healthy subjects as controls (19 males and 6 females) were included in the study. The patients were treated with aphthous cream that contained clove, thyme, mint, and liquerice, applied six times daily until cure or for two weeks. The patients were advised to use the cream for two weeks even the aphthous is healed. Blood samples were taken (5 mL) from patients before and after two weeks of treatment and other blood samples from the control group and analyzed for serum malodialdehyde (MDA) and total antioxidant status (TAS) by colorimetric methods. Results: In aphthous patients, serum MDA was significantly higher (p ≤ 0.001) than that in controls (1.82 ± 0.19 vs 1.06 ± 0.12 μmol/L), while serum TAS was significantly lower (p ≤ 0.001) than in the control group (1.16 ± 0.11 vs 1.70 ± 0.16 mmol/L). After treatment, serum MDA was decreased significantly (p ≤ 0.001) compared with the value before treatment (1.82 ± 0.19 vs 1.74 ± 0.17 μmol/L); however, serum TAS was increased significantly (p ≤ 0.05) after treatment compared with the value before treatment (1.16 ± 0.11 vs 1.2 ± 0.08 mmol/L). Both serum MDA and TAS in patients after treatment did not return to the control levels. The aphthous healed in most of the treated patient after 5-7 days, only two patients cured after 10 days of treatment. Conclusion: Aphthous ulceration in patients raises lipid peroxidation and reduces antioxidants. Aphthous treated with cream contained clove, thyme, ment and liquerice, healed within a week and the cured patients were associated with reduced lipid peroxidation and raised antioxidants status.

Maternal supplementation of L-carnosine improves reflexive motor behaviors in mice offspring

This study aimed to investigate the effect of maternal supplementation of L-carnosine on improved reflexive motor behaviors in mice offspring. Forty pregnant female NMRI mice were allocated into four groups. In the control group, mice received water, while in groups 2–4, female mice received supplementation of the L-carnosine (0.001, 0.01, or 0.1 mg/kg) at gestation days (G.D.) 5, 8, 11, 14, and 17. Newborn male pups were selected, and reflexive motor behaviors were analyzed on days 5, 7, 10, and 10‐15, respectively. Serum malondialdehyde(MDA), superoxide dismutase(SOD), glutathione peroxidase(GPx) and total antioxidant status(TAS) of was determined in offspring’s. According to findings, prenatal supplementation of the L-carnosine significantly increased ambulation score, surface righting, hind-limb suspension score, grip strength, front-limb suspension time, and negative geotaxis in mice offspring (P < 0.05). Hind-limb foot angle decreased in mice offspring by maternal supplementation of the L-carnosine (P < 0.05). Prenatal supplementation of the L-carnosine significantly decreased the MDA and increased the SOD, GPx, and TAS levels in offspring (P < 0.05). These results suggested maternal supplementation of the L-carnosine improved reflexive motor behaviors and antioxidant status in mice offspring.

NF-ĸB expression in endometriosis induced rat uterine tissue

Objective: Our aim in this study is to examine the effects of endometriosis on uterine tissue.&#x0D; Materials and methods: Two groups were formed, with eight rats in each group. Sham group was subjected only saline solution for 7 days. The endometriosis (EM) model was induced with estradiol benzoate (EB). In the EM group, EB was given to rats for 7 days. At the end of the experiment, blood was taken from the animals and serum total antioxidant status (TAS) and total oxidant status (TOS) values were studied. Uterine tissues were fixed in formalin and embedded in paraffin blocks. NF-ĸB 3 immune staining were performed on uterine sections. The results were examined under the microscope.&#x0D; Results: Compared to sham group, TAS values were significantly decreased and TOS values were significantly increased in serum of rats belonging to EM group. In the analysis of NF-ĸB expression in uterine tissue of sham group, the expression was slight and found only in connective tissue cells and inflammatory cells. NF-ĸB expression was mainly negative. After EM induction, NF-ĸB expression was increased in degenerated epithelial cells and gland cells, inflammatory cells around the glands and blood vessel endothelial cells and lamina propria.&#x0D; Conclusion: We think that NF-κB signal may be a determinant in the treatment of endometriosis.&#x0D; Keywords: endometriosis, immunohistochemistry, NF-κB, biochemistry, rat

Determination of Serum Trace Elements and Oxidative Stress in Bitches with Transmissible Venereal Tumor

Background: This study was conducted to compare the changes in oxidative stress levels with some trace elements in blood serum in healthy bitches and bitches with transmissible venereal tumor (TVT). Methods: A total of 40 bitches were used in the study. According to the genital organ examinations, healthy bitches constituted the control group (n=20) and those with venereal tumors constituted the TVT group (n=20). Total oxidant status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) were determined spectrophotometrically. In addition, some serum trace element levels such as magnesium (Mg), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn) and selenium (Se) were measured using an ICP-MS analysis device. Result: The serum Se, Mg and Zn levels were lower in the TVT group compared to the control group (p less than 0.01) and serum Fe and Cu levels were higher in the TVT group than in the control group (p less than 0.01). Serum TOS and OSI levels were higher (p less than 0.001) and serum TAS levels were lower (p less than 0.001) in the TVT group compared to the control group. The study results showed that oxidative stress is high in bitches with TVT, and in parallel with this, changes occur in the amount of certain trace elements that are associated with oxidative stress.

Serum total oxidant and anti-oxidant status in children with cancer.

Oxidative stress has a potential role in carcinogenesis. Anti-oxidant enzymes have a neutralizing effect on both cancer initiation and progression. We aimed to assess the oxidant and anti-oxidant levels of pediatric cancer patients and to compare the levels in healthy controls. The study involved 105 pediatric cancer patients (40 undergoing chemotherapy, 65 survivors) and 40 healthy children. The serum total oxidant status (TOS) and total anti-oxidant status (TAS) were measured. The oxidative stress index was significantly lower in pediatric cancer patients compared to the levels in the controls (0.20 ± 0.07 vs. 0.26 ± 0.10; P = 0.001). The mean serum TAS level was significantly higher in patient groups compared to the level in the control (1.87 ± 0.48 vs. 1.63 ± 0.32 mmol/L, P = 0.001). The TAS level of children with cancer in survivors was also found to be significantly higher compared to the levels in the control group (1.85 ± 0.45 vs. 1.63 ± 0.32 mmol/L, P = 0.005). Radiotherapy, surgery, relapsed disease, presence of metastases, and receiving enteral nutritional support caused no change in the TAS/TOS level. It has been revealed for the first time that the serum total anti-oxidant level was high in children undergoing chemotherapy and the survivor group as well. Moreover, the oxidative stress index was low in children with cancer. Longitudinal prospective studies are needed to reveal the alterations in oxidant status among children with cancer.

BENİGN, PRE-MALİGN VE MALİGN ENDOMETRİYAL LEZYONLARDA PROLİDAZ AKTİVİTESİ VE OKSİDATİF STRES

OBJECTIVE: The present study aims to investigate the prolidase activity, total oxidant status (TOS) and total anti-oxidant status (TAS) in women who have been diagnosed with benign, pre-malignant and malignant endometrial pathologies. MATERIAL AND METHODS: Ninety women who underwent endometrial biopsy due to abnormal uterine bleeding were divided into three groups according to their histopathological findings: Benign endometrial pathology (n=65), endometrial hyperplasia (n=12) and endometrial cancer (n=13). These groups were compared with respect to oxidative stress markers and prolidase activity in serum and endometrial tissue. RESULTS: When compared to the benign endometrial pathology group, the endometrium cancer group had significantly higher age, shorter height and higher incidences of menopause and positive family history for gynecological malignancy (p=0.001, p=0.023, p=0.001 and p=0.001). When compared to the benign endometrial pathology group, tissue prolidase activity was significantly higher in the endometrium hyperplasia and endometrium cancer groups (p=0.001 for both). However, tissue prolidase activity was statistically similar in the endometrial hyperplasia and endometrial cancer groups (p=0.166). All study groups had statistically similar serum prolidase activity, serum and tissue TOS, serum and tissue TAS, tissue malondialdehyde and glutathione values. CONCLUSIONS: Prolidase activity in endometrial tissue has enhanced in pre-malignant and malignant endometrial lesions when compared to benign endometrial lesions. The assessment of prolidase activity in endometrial tissue might help to distinguish pre-malignant and malignant lesions in case histopathological characteristics are insufficient for the differentiation of endometrial lesions.

Oxidative stress and antioxidant status in threatened preterm labor.

The purpose of this study was the evaluation of total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI) and superoxide dismutase (SOD) levels in women with threatened preterm labor (TPL) and also to compare the levels of these oxidative stress biomarkers of TPL pregnancies that had preterm and term deliveries. This case-control study was conducted on 46 patients diagnosed with TPL and 47 healthy pregnant women matched for gestational age. Patients with threatened preterm labor were divided into two groups: true preterm birth (TPB) group (n = 16) and false preterm birth (FPB) group (n = 30) groups. Maternal serum SOD, TOS and TAS levels were measured by a spectrophotometric method using a commertial kit. OSI level for each patient was calculated by using the formula: (TOS (μmol·H2O2·equiv/L) × 100)/(TAS (μmol·Trolox·equiv/L)). The mean TAS levels of the TPB and FPB groups were significantly lower than those of the control group (0.96 ± 0.3 vs 1.36 ± 0.34, p1 < 0.001; 0.97 ± 0.22 vs 1.36 ± 0.34, p2 < 0.001, respectively). The mean SOD, TOS and OSI levels of the TPB and FPB groups were significantly higher than those of the control group (p < 0.001). There was no significant difference between the TPB and FPB groups for any oxidative stress biomarkers. The maternal serum oxidative stress biomarkers are increased in pregnancies with TPL. However, these are not effective in predicting preterm birth in pregnancies with TPL.

Evaluation of Oxidative Stress Levels and Dynamic Thiol-disulfide Balance in Patients with Retinopathy of Prematurity

Purpose The aim of this study is to evaluate both dynamic thiol-disulfide homeostasis and oxidative stress (OS) levels in patients with retinopathy of prematurity (ROP). Methods A total of 129 infants of <34 weeks gestational age were enrolled in the present study. The thiol-disulfide homeostasis was determined by using the new, cost-effective and fully automated colorimetric method. Total antioxidant status (TAS), Total oxidant status (TOS) and Oxidative stress index (OSI) levels were evaluated. Results We found serum TAS levels were lower while serum TOS and OSI levels were significantly higher in patients with ROP compare to the without ROP group (p < .05). However, native, total and disulfide values were not statistically significant between the groups (p > .05). In addition, we also evaluated the native, total and disulfide levels in patients with ROP according to grades and no statistically significant results were found (p > .05). Low birth weight (p = .001), gestational age (p = .001) and 5-min Apgar score were significantly lower in the ROP group. Conclusion This study revealed that dynamic thiol-disulfide homeostasis was changed in patients with ROP. Increased TOS and decreased TAS levels may be associated with functional reduction of the antioxidant system due to increased OS. This indicate that ROP patients are highly sensitive to OS. The dynamic thiol-disulfide homeostasis may conduce to the pathophysiological mechanism and disease follow-up in patients with ROP. The results of this study show that ROP patients are highly sensitive to oxidative stress.
