AbstractBackgroundBrucellosis is a serious zoonotic disease affecting human and all domestic animals. It is considered to be one of the great public health problems all over the world. Little attention has been paid to bovine brucellosis in small holder dairy farm and in animal owners and farm workers in central highland of Ethiopia. This research was conducted with the aim of determining seropositivity and identifying the potential risk factors of brucellosis in dairy cows with recent cases of abortion in the central highlands of Ethiopia.MethodsThis was a cross‐sectional study with purposive sampling carried out on dairy cows in the farms and kebeles from December 2019 to May 2020. The serum samples were initially screened for brucellosis using the Rose Bengal plate test (RBPT) followed by the complement fixation test (CFT) for confirmation. The risk factors for bovine brucellosis were assessed using univariable Firth bias reduced logistic regression.ResultsA total of 352 samples were tested. Serological positivity for bovine brucellosis was detected in 1.2% (95% CI: 0.47–2.97) by RBPT, and bovine brucellosis confirmed by CFT was 0.6% (95% CI: 0.16–2.09). The risk factors for confirmed bovine brucellosis were the late stage of abortion (OR = 1.046, p = < 0.0041), retained fetal membranes (OR = 32.74, p = 0.006), and market‐based stock replacement (OR = 1.0638, p = 0.0008), which are significantly associated at 95% confidence interval.ConclusionThe proportion of brucellosis among aborted cows seems to be low in the study area. However, given the lack of control strategies in the area, there is a potential risk of transmission in dairy cattle and the human population in the study areas.

Detection of Antibodies Against Brucellosis and Associated Risk Factors in Cross Breed Dairy Cattle in Smallholder Farmers, Southern Ethiopia.

A cross-sectional study was conducted in selected districts of Gamo zone, southern Ethiopia over a period of November 2019 and September 2020 to estimate seroprevalence and associated risk factors and assess knowledge and practices of smallholder farmers about bovine brucellosis. Two districts and four kebeles from each district were purposively selected, and individual animals were sampled using a simple random sampling technique. A total of 384 sera samples were collected, and concurrently, 236 dairy cattle owners were interviewed. The samples were tested for antibodies against Brucella using both Rose Bengal Plate test and Complement Fixation test following OIE standard protocol. Risk factors associated with bovine brucellosis were analyzed using univariate and multivariate logistic regressions. The survey result has shown that 95% and 97% of the small holder farmers did not know the cause and symptoms of bovine brucellosis, respectively, and the majority have engaged in risky practices. Eight (2.08%) of the collected serum samples were positive with screening test (RBT) and only six (1.5%) were positive with confirmatory test (CFT). Multivariable logistic regression analysis showed a statistically significant association between herd sizes and the disease. The seropositivity of the disease is higher in small-sized herds followed by medium-sized herds. The seroprevalence of bovine brucellosis was found to be at a low percentage with confirmatory tests even if there was a presence of associated risk factors for the disease in the study area. Again, the results suggest that smallholder farmers have poor knowledge and risky practices, which expose them to the disease. Awareness creation about the disease is of paramount importance even if the prevalence was low in this serological study. The implementation of a test and slaughter program before the disease becomes widespread, along with the testing of new stock before introduction to the farms is recommended.

Brucellosis in Borena Cattle: - Seroprevalence and Awareness of the Pastoral Community in Yabello Ethiopia

The study was cross-sectional and the objectives of the study were: estimation of prevalence of bovine brucellosis, assessment of risk factors and assessment of knowledge of pastoralist about the disease and its risk factors in Borana cattle at Dida Tuyura ranch and its surrounding. The study animals were selected by multi-stage sampling. Blood was collected from selected animals and serum was extracted. The Sera samples were screened using the rose Bengal plate (RBPT) test and those which tested positive were further tested using Complement fixation test (CFT) for confirmation. Sixteen (16) cattle out of 661 (2.4%; 95 5 CI: 1.39, 3.9) tested using RBPT were found to be positive. However, only 5 animals were found positive with CFT in animal sampled from Dida Tuyura Ranch yielding a prevalence of 1.47% (95% CI: 0.48, 3.41). from Six animals which gave positive reaction to RBPT from pastoralists’ herd in the vicinity of the ranch only two gave positive reaction to CFT yielding a prevalence of 0.62% (95% CI: 0.162, 4.73). Taken together the seroprevalence of bovine brucellosis as revealed by CFT 1.1% (95% CI: 0.43, 2.17). Univariable logistic regression analysis showed that previous history of abortion and retained fetal membranes were significantly associated with seropositivity to brucellosis (P<0.05) whereas sex, age, parity, body condition and PAs were not associated with infection with Brucella (P>0.05). In the multivariable analysis, only abortion (OR=13.46, p<0.05)) remained to be independently associated with brucellosis seropositivity whereas other not. The results of questionnaire survey revealed that the majority of the pastoralists or cattle attendants do not have sufficient knowledge about brucellosis and are at risk of acquiring the infection. Therefore, educating the pastoralists about the disease through extension service on the handling of aborted fetuses and assistance of delivery is important.

Seroprevalence of Bovine Brucellosis in Selected Sites of Central Highland of Ethiopia.

Brucellosis is a contagious, economically significant bacterial disease that affects animals worldwide and is one of the most neglected zoonotic diseases in the world. The disease poses a barrier to the trade of animals and animal products, represents a public health hazard, and is an impediment to free animal movement. A cross-sectional study was carried out from December 2019 to May 2020 in order to determine seroprevalence and identify potential risk factors for brucellosis in dairy cows in the Central Highlands of Ethiopia with recent cases of abortion. Purposive sampling was carried out on the farms and kebeles in question to screen for recent cases of abortion in dairy cows. For the purpose of performing serological testing, 352 blood samples from dairy cattle were obtained. The Rose Bengal Plate test was used to initially screen the serum samples, and the Complement Fixation test was utilized as a confirmatory test. Using combined RBPT and CFT tests, the overall seroprevalence of bovine brucellosis was 0.6% (95% CI: 0.16-2.09). Retained fetal membrane (OR = 32.74, p = 0.006), market-based stock replacement (OR = 16.55, p = 0.002), breeding method (OR = 7.58, p = 0.027), and late stage of abortion (OR = 14.74, p = 0.0002) are all significantly associated risk factors. The present seroprevalence study revealed that brucellosis is prevalent at a lower rate among dairy cattle in the study areas. However, there is a possible risk of brucellosis transmission in dairy cattle and the exposed human population in research locations because no control measures were put in place there. Implementing a test and slaughter method with compensation for farmers is advised due to the low prevalence of bovine brucellosis in government-owned and small-holder farms.

BackgroundBrucellosis is a contagious bacterial disease, which affects domestic animals, humans, and wildlife in Ethiopia and other countries.MethodsA cross-sectional study design using a simple random sampling technique was conducted in Diga, Guto, Gida, and Sibu Sire districts of East Wollega Zone, Western Oromia, from November 2019 to July 2020 to determine the prevalence of bovine brucellosis and identify associated risk factors. A lottery method was used during serum collection and risk factors identified during interview.ResultsA total of 1152 serum samples were collected from cattle of 6 months age and above. Serum samples were tested by enzyme linked immunosorbent assay (I-ELISA). An overall seroprevalence of brucellosis was 1.82% (21/1152). Statically significant variation (P<0.05) in seroprevalence of bovine brucellosis was recorded among potential risk factors such as breed [P≤0.05; OR:8.905; CI:1.568–50.573], parity [P≤0.05; OR:0.017; CI:0.042–5.195], retained fetal membrane [P≤0.001; OR:0.018; CI:0.00–2.169], and abortion history [P≤0.001; OR:0.030; CI:0.004–0.212]. Conversely, seroprevalence variations in sex, district, villages, body condition score, and age were statistically insignificant (P>0.05).ConclusionThe current study revealed bovine brucellosis is posing a threat to livestock with no strict control and prevention scheme in place. Therefore, public awareness creation about the zoonotic importance of the disease is crucial and the Government should establish legislation, support, control, and prevention of the disease recommended.

Seroprevalence and risk factors for bovine brucellosis in the state of Paraná, Brazil: an analysis after 18years of ongoing control measures.

Seroprevalence and risk factors of bovine brucellosis (Brucella abortus) in herds and cattle were estimated by a cross-sectional study in the state of Paraná, Brazil. The state was divided into seven regions and a random, two-stage sampling was performed on properties and cattle from each region between 2018 and 2019. Serum samples were collected from 11,592 cows over 24months from 1,757 properties and a questionnaire was applied to identify potential risk factors. As recommended by the National Program for the Control and Eradication of Animal Brucellosis and Tuberculosis (PNCEBT), serological testing for the detection of anti-Brucella antibodies included the buffered plate agglutination test (screening test) and the fluorescence polarization assay (confirmatory test). The seroprevalence of bovine brucellosis on properties and in cattle was 4.87% (95% confidence interval [CI]: 3.98-5.93%) and 2.24% (95% CI: 1.47-3.41%), respectively. Multiple logistic regression analysis identified larger herd size and failure to test for brucellosis as risk factors for the presence of anti-B. abortus antibodies. These results demonstrate no change in the prevalence when comparing initial studies conducted in 2002. Given our findings, it is recommended that policies for brucellosis control include a widespread vaccination program for higher prevalence areas and eradication approach to lower prevalence areas. All steps related to correct immunization of the herds should be verified and improved by training and education. Health education action must be carried out informing farmers about the risks of introducing animals not tested for brucellosis into their herds and the benefits of testing their herds regularly.

Brucellosis is one of the major zoonotic diseases yet prevalent in Ethiopia. It is contagious and has harmful effects on free animal movement and export. A cross-sectional study was conducted from November 2016 to April 2017 in the Loko and Uke peasant association of Guto Gida District. The goal of this study is to determine the seroprevalence and associated risk factors of bovine brucellosis. The questionnaire survey was conducted on 200 respondents to collect the livestock owner's perception of this disease. Eighty (80) blood samples were collected from grazing cattle above six months of age. Serum was tested by complement fixation test (CFT) for Brucella antibody detection. Pearson chi-square is used to assess the relation of independent variables such as sex, site, and age with seroprevalence in a 95% confidence interval if P value is less than 0.05, recoded as significant. Seroprevalence of bovine brucellosis in the present study is 22.5%. Bovine brucellosis prevalence recorded in Uke (28.6%) was higher than that in Loko (21.2%). Again bovine brucellosis is higher in males (30.4%) than females (19.2%). There is prevalence variation among site, sex, and age which is statistically insignificant (P value>0.05). Survey findings revealed that 52% (104/200) of respondents did not know the causes of abortion, while 48% (96/200) of them confirmed abortion in their herd and 64% (124/200) of respondents removed retained fetal membrane by themselves. Brucellosis is a contagious reproductive disease of cattle with zoonotic implications and needs to design and implement control measures aiming at preventing further spread of the disease.

Seroprevalence of Bovine Brucellosis in Selected Districts of Zambia.

Brucellosis is an infectious zoonosis that has huge economic and public health implications globally. The disease is prevalent in humans, livestock and wildlife in Sub-Saharan Africa. A cross-sectional study was conducted between 6 May 2017 and 31 July 2020 during which 1712 sera from 175 cattle herds in five districts from Southern, Western and Eastern Provinces of Zambia were collected and screened against brucellosis. The Rose Bengal Test (RBT) and competitive Enzyme-linked Immuno Assay (c-ELISA) were used in serial testing for the detection of antibodies against Brucella species. A total of 127 animals from 37 herds tested positive, giving overall individual animal and herd-level seroprevalences of 7.53% (95% CI: 6.28–8.78%) and 21.14% (95% CI: 15.0–27.2%), respectively. Namwala district had the highest herd seroprevalence (33.9%, 95% CI: 21.6–46.1%), while Lundazi did not record any seropositivity. Comparably, Southern Province had the highest individual animal (8.97%, 95% CI: 7–11%) and herd-level (28.5%, 95% CI: 20.3–36.7%) seroprevalences, although this was not statistically significant. Within Southern Province, higher seropositivity was observed in Namwala district (OR: 8.55; CI: 2.66–27.44), among female animals (OR: 2.48; CI: 1.38–4.46) and in those aged 11 years and above (OR: 2.67; CI: 1.34–5.34) as well as in gravid cows (OR: 4.34; CI: 2.08–8.92). Seropositivity was also observed among some animals with hygromas (OR: 6.5; CI: 0.45–94.08) and those with a history of abortion (OR: 1.13; CI: 0.18–7.28) although the findings were not statistically significant. Brucella seroprevalence among traditional cattle in Zambia remains high. Control programs against bovine brucellosis must be introduced to reduce its impact on human health and animal production.

Seroprevalence of bovine brucellosis and analysis of risk factors in cattle and livestock Handler's in Gilgit – Pakistan, 2019

Background: Brucellosis is one of the silent killers in livestock population with zoonotic effect in all over the world. The objective of this study was to determine the seroprevalence of bovine brucellosis and identify risk factors associated with transmission of Brucella abortus. Methods & Materials: A cross sectional study was conducted in district Gilgit during October 2018 to May 2019. Total 150 serum samples were collected from different breeds of Cattles. Twenty (20) sera were also collected from livestock handlers. Rose Bengal plate test (RBPT) was performed for the initial screening, while the suspected and positive samples were confirmed through Tube agglutination test (TAT) and indirect-ELISA. A predesigned questionnaire was used to collect information on risk factors; animal breed, age, sex, breeding method, abortion history and retained placenta. Information was also collected on human related risk factors including occupation, consuming raw milk and animal handling. Data analysis was done using SPSS version 20. Results: All serum samples (n = 170) were initially screened with RBPT and 16 were found seropositive to brucellosis. All human samples (n = 20) were found negative. The overall seroprevalence of bovine brucellosis was 10.7% while the seroprevalence in Jersy breed was 28% and the local cattle breed were found negative. Out of the total 16 RBPT seropositive sera, 11 were showing seropositive titer in TAT. Six sera were showing acute infection having the titer 1:160 with higher sensitivity 92.4% while only four (7.02%) with positive antibody titer were confirmed in indirect ELISA. Risk factors age (p-value 0.004), breed (p-value 0.002), retained placenta (p-value 0.002) and history of abortion (p-value 0.002) was associated significantly with the disease. Conclusion: The prevalence of brucellosis in Jersy cattle was much higher than the local cattle breed. Age, retained placenta and abortion history were positively correlated. Safe management practices might be helpful to limit the transmissibility.

Sero-prevalence and associated risk factors of bovine brucellosis in Sendafa, Oromia Special Zone surrounding Addis Ababa, Ethiopia.

Bovine brucellosis is an infectious bacterial disease caused by members of genus Brucella, affecting both animals and humans, and resulting in a serious economic loss in animal production sector and deterioration of public health. A cross-sectional study was conducted from November 2014 to April 2015 to determine the seroprevalence and associated risk factors of bovine brucellosis in Sendafa, Oromia Special Zone, Ethiopia. A total of 503 blood samples were collected using a simple random sampling technique from dairy cattle of above 6 months of age with no history of previous vaccination against brucellosis. All sera samples were subjected to both Rose Bengal Plate Test for screening and Complement Fixation Test for confirmation. Accordingly, the overall seroprevalence of bovine brucellosis in the study area was 0.40%. The result showed that the seroprevalence of bovine brucellosis in the study area was not statistically significant with all proposed risk factors. No reactors were observed in male animals. The seroprevalence was observed in animals without previous history of abortion. Moreover, information was gathered on individual animal and farm-level risk factors and other farm characteristics using a questionnaire. Awareness among society was poor, so the positive animals can be a potential hazard to animals and humans in the study area. Therefore, public education should be done to improve the awareness of the community on bovine brucellosis and its public health impact with due consideration on the safe consumption of food of animal origin.

Sero-prevalence of bovine brucellosis in the Bojanala Region, North West Province, South Africa 2009–2013

Bovine brucellosis affects food safety, food security and human health in rural communities in the North West Province, South Africa. The World Organisation for Animal Health suggests routine sero-surveillance and vaccination of cattle for control and to prevent zoonotic transmission. Although sero-surveillance and subsidised vaccination have been in place for decades, data from Bojanala have not previously been analysed. The aim of this study was to retrospectively analyse historical data on routine sero-surveillance of bovine brucellosis and state subsidised vaccination, in communal, commercial and dairy cattle in the study area. This was a descriptive, cross-sectional retrospective analysis of records from all adult cows bled by the state veterinary services during routine sero-surveillance for bovine brucellosis, in the Bojanala Region, North West Province, between 2009 and 2013. Fewer communal (N = 11 815) and dairy (N = 6696), than commercial beef (N = 28 251) cows, were tested. Overall herd prevalence (33.33%), differed significantly from individual prevalence (3.18%) in all groups. Communal herds had both the highest herd prevalence (38.8%) and the highest individual prevalence (5.2%). Both herd and individual sero-prevalence were lowest in dairy cattle, possibly because registered dairy herds are routinely tested. Over the 5-year study period, only 24 086 (7.15%) of the 342 500 cows eligible for free vaccination, were vaccinated. The annual number of cattle tested was highly variable. Dairy cattle that were regularly tested had a significantly lower herd and individual prevalence. Herd prevalence would be useful for spatial mapping, whilst individual prevalence could better reflect the risk of zoonotic transmission.

Sero-prevalence of brucellosis (Brucella abortus) in bovines from Caquetá state, Colombia

Brucellosis is a disease with implications in the public health due both to its zoonotic character as its easy transmission trough of dairy products contaminated with the microorganism Brucella sp. The aim of this article was determinate the prevalence of bovine brucellosis in nine municipalities that carrying the 75% of bovine population in the Caquetá state, Colombia. Blood samples were collected in 100 herds dedicates to dual-purpose cattle, obtained blood serum from 1000 animals, of which 882 corresponding to cows. Rose Bengal Plate Test was performed and the confirmations of positive cases were made by competitive Elisa. The sero-prevalence of bovine brucellosis in Caquetá state was of 3.23% (95%, CI: 0-6.53%), in males 2.6% and females 3.29% respectively. At the municipal level, the sero-prevalence varied of 0 to 6.06%. The prevalence of brucellosis at herd level was of 22% (95%, CI: 12.24-31.76%). In conclusion, the sero-prevalence is low in bovines but moderate at the level of herds, therefore, it is necessary to increase the performance of the official control entities at the level of herds to reduce the high prevalence of the disease for guaranteed the safety of milk and thus the health of consumers.

A cross-sectional study was carried out between November 2015 and January 2016 to determine the seroprevalence of Brucella antibodies in cattle raised under communal, fenced farms and tethering systems and the associated factors. Seven hundred twenty-eight bovine sera were collected and tested with rose Bengal test as a screening test and the indirect enzyme-linked immunosorbent assay as a confirmatory test. Animal- and herd-level data were collected and binary logistic regression was used to assess the potential risk factors. True animal- and herd-level prevalence was highest in the fenced farms (4.5% (95%CI, 2.3-6.9) and 19.5% (95%CI, 8.2-32.7) respectively). The risks for natural brucellosis infection were sharing water with wild animals (OR = 0.21, 95%CI, 0.104-0.83), herd size (medium: OR = 0.089, 95%CI 0.017-0.449; large: OR = 0.024, 95%CI 0.003-0.203), fenced farms (OR = 3.7, 95% CI, 1.7-7.9), sex (OR = 0.03, 95%CI, 0.01-0.079), and lactation (OR = 0.013, 95%CI, 0.004-0.049). Changes in rangeland tenure and the shift towards intensive cattle production have influenced brucellosis epidemiology. Future studies should aim at identifying the infecting Brucellae and examining the role of wildlife in brucellosis epidemiology.

Sero-prevalence of Bovine Brucellosis and associated risk factors in mbeya region, Southern highlands of Tanzania

A cross-sectional study was conducted to establish the seroprevalence of brucellosis and associated risk factors in indigenous and exotic breeds of cattle from 178 farms in Mbeya region. A total of 1211 cattle (929exotic cattle from 108 commercial farms and 282 indigenous cattle from 70 traditional farms) were tested for Brucella antibodies using the Rose Bengal Plate Test (RBPT) and competitive Enzyme Linked Immunosorbent Assay (c-ELISA) as screening and confirmatory tests, respectively. The overall animal-level seroprevalence was 9.3%; 11.3% (95% CI: 9.4–13.5) in indigenous cattle and 2.8% (95% CI:1.4–5.6) in exotic cattle. Further, the overall herd level seroprevalence was 32.0%; 50.5% (95% CI: 40.9–59.9) in indigenous cattle and 4.2% (95% CI: 1.3–12.4) in exotic cattle. Infections were higher in cattle aged 6–10 years old, (39.8%; 95% CI: 31.2–49.1) followed by those aged 1–5 years (5.8%; 95% CI: 4.8–6.6) and 11–15years old (2.7%; 95% CI: 0.8–8). When compared to cattle sampled from herds size of 1–50, those sampled from the herd sizes of 51–100 and 101–150 had higher odds of brucellosis seropositivity [(OR=3.6, CI: 1.76–7.16, p<0.001) and (OR=3.0, CI: 1.09–8.04, p=0.033). The odds of seropositivity in animals which calved on pasture was 3.0 (CI: 1.1–7.8, p=0.028) compared to those that calved at home. Brucella seroprevalence was also observed to vary according to districts, with Mbarari district recording the highest (45.4%). It is evident from the study that Brucellosis is present in Mbarari, Mbeya and Momba districts of Mbeya Region. The findings of this study provide some baseline data that could contribute to the design and implementation of brucellosis control measures in the study areas.

Seroprevalence and risk factors associated with bovine brucellosis in the Potohar Plateau, Pakistan

BackgroundThe seroprevalence and risk factors of bovine brucellosis were studied at animal and herd level using a combination of culture, serological and molecular methods. The study was conducted in 253 randomly selected cattle herds of the Potohar plateau, Pakistan from which a total of 2709 serum (1462 cattle and 1247 buffaloes) and 2330 milk (1168 cattle and 1162 buffaloes) samples were collected. Data on risk factors associated with seroprevalence of brucellosis were collected through interviews using questionnaires. Univariable and multivariable random effects logistic regression models were used for identifying important risk factors at animal and herd levels.ResultsOne hundred and seventy (6.3%) samples and 47 (18.6%) herds were seropositive for brucellosis by Rose Bengal Plate test. Variations in seroprevalence were observed across the different sampling sites. At animal level, sex, species and stock replacement were found to be potential risk factors for brucellosis. At herd level, herd size (≥9 animals) and insemination method used were important risk factors. The presence of Brucella DNA was confirmed with a real-time polymerase chain reaction assay (qRT-PCR) in 52.4% out of 170 serological positive samples. In total, 156 (6.7%) milk samples were positive by milk ring test. B. abortus biovar 1 was cultured from 5 positive milk samples.ConclusionThis study shows that the seroprevalence of bovine brucellosis is high in some regions in Pakistan. Prevalence was associated with herd size, abortion history, insemination methods used, age, sex and stock replacement methods. The infected animal may act as source of infection for other animals and for humans. The development of control strategies for bovine brucellosis through implementation of continuous surveillance and education programs in Pakistan is warranted.

A total of 240 blood samples were collected from semi-intensively and extensively managed cattle. The Rose Bengal Plate Test (RBPT) was used as a screening test. Those serum samples reacting positively to (RBPT) detected 9 of 240 (3.75%) of the samples as brucellosis positive. The positive sera when further retested using CFT 5 out of 9 RBPT positive sera were confirmed to be positive. The prevalence of brucellosis based on CFT in and around kombolcha was 2.08%, and all positive sera were from old aged female cattle. An attempt was also made to investigate the prevalence rate of abortion and fetal membrane retention in both extensive and semi-intensively management systems. A higher prevalence rate of abortion was recorded in extensively managed cows (10.8) than semi-intensively managed cows (2.08%). The difference in prevalence rate was statistically significant (P<0.05). A relatively higher prevalence rate of retained fetal membrane was found in extensive managed cattle (13.8%) than semi-intensively managed cattle (4.1%). The difference in prevalence rate was statistically significant (p<0.05). In conclusion, the prevalence rate of brucellosis is low in and around kombolcha. However, this low infection of bovine brucellosis may be spreaded in the study area and may cause economic loss and human infection unless control strategy should be conducted.
