Epistatic selection on a selfish Segregation Distorter supergene - drive, recombination, and genetic load.

Meiotic drive supergenes are complexes of alleles at linked loci that together subvert Mendelian segregation resulting in preferential transmission. In males, the most common mechanism of drive involves the disruption of sperm bearing one of a pair of alternative alleles. While at least two loci are important for male drive-the driver and the target-linked modifiers can enhance drive, creating selection pressure to suppress recombination. In this work, we investigate the evolution and genomic consequences of an autosomal, multilocus, male meiotic drive system, Segregation Distorter (SD) in the fruit fly, Drosophila melanogaster. In African populations, the predominant SD chromosome variant, SD-Mal, is characterized by two overlapping, paracentric inversions on chromosome arm 2R and nearly perfect (~100%) transmission. We study the SD-Mal system in detail, exploring its components, chromosomal structure, and evolutionary history. Our findings reveal a recent chromosome-scale selective sweep mediated by strong epistatic selection for haplotypes carrying Sd, the main driving allele, and one or more factors within the double inversion. While most SD-Mal chromosomes are homozygous lethal, SD-Mal haplotypes can recombine with other, complementing haplotypes via crossing over, and with wildtype chromosomes via gene conversion. SD-Mal chromosomes have nevertheless accumulated lethal mutations, excess non-synonymous mutations, and excess transposable element insertions. Therefore, SD-Mal haplotypes evolve as a small, semi-isolated subpopulation with a history of strong selection. These results may explain the evolutionary turnover of SD haplotypes in different populations around the world and have implications for supergene evolution broadly.

Distinct spermiogenic phenotypes underlie sperm elimination in the Segregation Distorter meiotic drive system.

Segregation Distorter (SD) is a male meiotic drive system in Drosophila melanogaster. Males heterozygous for a selfish SD chromosome rarely transmit the homologous SD+ chromosome. It is well established that distortion results from an interaction between Sd, the primary distorting locus on the SD chromosome and its target, a satellite DNA called Rsp, on the SD+ chromosome. However, the molecular and cellular mechanisms leading to post-meiotic SD+ sperm elimination remain unclear. Here we show that SD/SD+ males of different genotypes but with similarly strong degrees of distortion have distinct spermiogenic phenotypes. In some genotypes, SD+ spermatids fail to fully incorporate protamines after the removal of histones, and degenerate during the individualization stage of spermiogenesis. In contrast, in other SD/SD+ genotypes, protamine incorporation appears less disturbed, yet spermatid nuclei are abnormally compacted, and mature sperm nuclei are eventually released in the seminal vesicle. Our analyses of different SD+ chromosomes suggest that the severity of the spermiogenic defects associates with the copy number of the Rsp satellite. We propose that when Rsp copy number is very high (> 2000), spermatid nuclear compaction defects reach a threshold that triggers a checkpoint controlling sperm chromatin quality to eliminate abnormal spermatids during individualization.

Origin, evolution, and population genetics of the selfish Segregation Distorter gene duplication in European and African populations of Drosophila melanogaster.

Meiotic drive elements are a special class of evolutionarily "selfish genes" that subvert Mendelian segregation to gain preferential transmission at the expense of homologous loci. Many drive elements appear to be maintained in populations as stable polymorphisms, their equilibrium frequencies determined by the balance between drive (increasing frequency) and selection (decreasing frequency). Here we show that a classic, seemingly balanced, drive system is instead characterized by frequent evolutionary turnover giving rise to dynamic, rather than stable, equilibrium frequencies. The autosomal Segregation Distorter (SD) system of the fruit fly Drosophila melanogaster is a selfish coadapted meiotic drive gene complex in which the major driver corresponds to a partial duplication of the gene Ran-GTPase activating protein (RanGAP). SD chromosomes segregate at similar, low frequencies of 1-5% in natural populations worldwide, consistent with a balanced polymorphism. Surprisingly, our population genetic analyses reveal evidence for parallel, independent selective sweeps of different SD chromosomes in populations on different continents. These findings suggest that, rather than persisting at a single stable equilibrium, SD chromosomes turn over frequently within populations.

Large-scale selective sweep among Segregation Distorter chromosomes in African populations of Drosophila melanogaster.

Segregation Distorter (SD) is a selfish, coadapted gene complex on chromosome 2 of Drosophila melanogaster that strongly distorts Mendelian transmission; heterozygous SD/SD + males sire almost exclusively SD-bearing progeny. Fifty years of genetic, molecular, and theory work have made SD one of the best-characterized meiotic drive systems, but surprisingly the details of its evolutionary origins and population dynamics remain unclear. Earlier analyses suggested that the SD system arose recently in the Mediterranean basin and then spread to a low, stable equilibrium frequency (1–5%) in most natural populations worldwide. In this report, we show, first, that SD chromosomes occur in populations in sub-Saharan Africa, the ancestral range of D. melanogaster, at a similarly low frequency (∼2%), providing evidence for the robustness of its equilibrium frequency but raising doubts about the Mediterranean-origins hypothesis. Second, our genetic analyses reveal two kinds of SD chromosomes in Africa: inversion-free SD chromosomes with little or no transmission advantage; and an African-endemic inversion-bearing SD chromosome, SD-Mal, with a perfect transmission advantage. Third, our population genetic analyses show that SD-Mal chromosomes swept across the African continent very recently, causing linkage disequilibrium and an absence of variability over 39% of the length of the second chromosome. Thus, despite a seemingly stable equilibrium frequency, SD chromosomes continue to evolve, to compete with one another, or evade suppressors in the genome.

Defects in nuclear transport enhance Segregation Distortion

The Segregation Distorter (SD) system in Drosophila melanogaster causes the transmission of the SD chromosome at the expense of the SD+ chromosome. This occurs through a defect in sperm-specific chromatin condensation of the SD+-bearing spermatids of the SD/SD+ male. The Sd gene encodes a truncated form of RanGAP that is missing a nuclear export signal and is therefore trapped in the nucleus; normally RanGAP is found at the periphery of the nuclear membrane and is required for normal Ran-mediated nuclear transport. The presence of active RanGAP in the nucleus interferes with nuclear export and causes distortion. We show that mutations that affect nuclear import and export can enhance distortion in an SD background, thus verifying that the defect in nuclear transport is responsible for the unequal transmission of chromosomes. In addition, we identify several genes involved in chromatin condensation which also cause distortion in an SD background, opening the way to the dissection of the mechanism of segregation distortion.

Closing the (Ran)GAP on segregation distortion in Drosophila.

Segregation Distorter (SD) is a meiotic drive system in Drosophila that causes preferential transmission of the SD chromosome from SD/SD(+) males owing to induced dysfunction of SD(+) spermatids. Since its discovery in 1956, SD and its mode of action have baffled biologists. Recently, substantial progress has been made in elucidating this puzzle. Sd, the primary gene responsible for distortion encodes a mutant RanGAP, a key protein in the Ran signaling pathway required for nuclear transport and other nuclear functions. The mutant protein is enzymatically active but mislocalized to nuclei, which apparently disrupts Ran signaling by reducing intranuclear Ran-GTP levels. Some evidence suggests that a defect in nuclear transport may be the main cause of sperm dysfunction. Although important questions remain, the basic mechanism of distortion is now understood sufficiently well that specific hypotheses can be formulated and tested. This previously mysterious genetic system may now offer unique insights into novel aspects of regulation by Ran.

Segregation distortion induced by wild-type RanGAP in Drosophila.

Segregation Distorter (SD) is a meiotic drive system in Drosophila that causes preferential transmission of the SD chromosome from SD/SD(+) males owing to the induced dysfunction of SD(+) spermatids. The key distorter locus, Sd, is a dominant neomorphic allele encoding a truncated, but enzymatically active, RanGAP (RanGTPase-activating protein) whose nuclear mislocalization underlies distortion by disrupting the Ran signaling pathway. Here, we show that even wild-type RanGAP can cause segregation distortion when it is overexpressed in the male germ line or when the gene dosage of a particular modifier locus is increased. Both manipulations result in substantial nuclear accumulation of RanGAP. Distortion can be suppressed by overexpression of Ran or Ran guanine nucleotide exchange factor (RanGEF) in the male germ line, indicating that the primary consequence of nuclear mislocalization of RanGAP is reduction of intranuclear RanGTP levels. These results prove that segregation distortion does not depend on any unique properties of the mutant RanGAP encoded by Sd and provide a unifying explanation for the occurrence of distortion in a variety of experimental situations.

Defective nuclear transport: a prime suspect in segregation distortion

Segregation Distorter (SD) is an example of a meiotic drive system in Drosophila melanogaster, so designated because the SD locus is transmitted to virtually 100% of the progeny from a heterozygous male. Distortion occurs owing to abortive development of spermatids carrying the wild-type allele (SD+). A second locus, called Responder (Rsp), comprising heterochromatic repeat elements, is the target of Sd. A sensitive Rsp (Rsps) allele generally present on SD+ chromosomes renders the chromosome susceptible to distortion. SD chromosomes carry an insensitive Rsp (Rspi) allele and are resistant, therefore, to Sd-induced distortion.

Nuclear Mislocalization of Enzymatically Active RanGAP Causes Segregation Distortion in Drosophila

Segregation Distorter ( SD) is a meiotic drive system in Drosophila that causes preferential transmission of the SD chromosome from SD/SD + males owing to dysfunction of SD + spermatids. The Sd locus, which is essential for distortion, encodes a truncated RanGAP (Ran GTPase activating protein), a key nuclear transport factor. Here, we show that Sd-RanGAP retains normal enzyme activity but is mislocalized to nuclei. Distortion is abolished when enzymatic activity or nuclear localization of Sd-RanGAP is perturbed. Overexpression of Ran or RanGEF (Ran GTPase exchange factor) in the male germline fully suppresses distortion. We conclude that mislocalization of Sd-RanGAP causes distortion by reducing nuclear RanGTP, thereby disrupting the Ran signaling pathway. Nuclear transport of a GFP reporter in salivary glands is impaired by SD, suggesting that a defect in nuclear transport may underlie sperm dysfunction.

The Segregation Distorter (SD) complex and the accumulation of deleterious genes in laboratory strains of Drosophila melanogaster

Segregation Distorter (SD) associated with the second chromosome of D. melanogaster is found in nature at equilibrium frequencies lower than 5%. We report extremely high frequencies of SD (30-50%) in two selected strains, established in 1976, and show it to be responsible for the accumulation of deleterious genes in chromosome II. Samples of chromosomes extracted over a 4-year period were characterized with respect to distortion, sensitivity, lethality, sterility, and inversions. SD chromosomes were inversion-free as they have been shown to be in the Mediterranean area. The cosmopolitan inversion In(2L)t was found associated with SD (+) chromosomes. Lines polymorphic for SD have accumulated linked lethal and female-sterile genes approaching a near balanced system. It is proposed that deleterious genes linked in coupling to SD were accumulated by the balancing effect of distortion, while drift and restricted recombination account for the accumulation of deleterious genes linked in repulsion by a mechanism similar to Muller's ratchet. Our results should not be viewed as a particular case as SD chromosomes associated with detrimental genes and inversions are present in almost all populations around the world. The system could evolve in the way we describe whenever equilibrium conditions are broken down in small populations and lead to an increase in SD frequency.

Segregation Distortion in Drosophila melanogaster: Genetic and Molecular Analyses

The Segregation Distorter (SD) complex in the centromeric region of chromosome 2 in Drosophila melanogaster is responsible for a naturally occurring and strong system of male meiotic drive. Earlier recombinational dissection and deletional analysis showed that the SD complex resolves into three major loci: the euchromatic Sd, or Segregation distorter gene at the base of the left arm of the chromosome (2L), and two heterochromatic genes: E(SD), the Enhancer of SD, and Rsp, the Responder, in the center of the left arm and the right arm (2R), respectively. Rsp exists in the major allelic forms, RsPs, for sensitive, and Rspi, for insensitive. Males that are heterozygous for an SD chromosome (SdE(SD)Rspi) and a sensitive SD+ homologue (Sd+E(SD)+Rsps) transmit predominantly or even exclusively the SD-bearing chromosome to the progeny. The distortion of the segregation ratio is traceable to failure in chromatin condensation and maturation of those spermatids that receive the Rsps homologue during meiosis. Characterization of the properties of null alleles of Sd, E(SD), and Rsp that were generated by deletion determined the precise cytological locations of the components and established the functional relationship of each to its wild-type counterpart, suggesting a model whereby a deleterious action of Sd, along with E(SD), on the Rsps target sets in motion the events culminating in sperm dysfunction. Further genetic analysis of E(SD), a gene required for full expression of drive, showed that E(SD) in two doses can cause significant distortion even in the absence of Sd. This distortion is suppressible by a suppressor of SD action. Thus, E(SD) is more than a simple modifier of Sd; rather, it is an effector locus that, like Sd, can act at the Rsps target. Cloning and molecular analysis of the Sd locus reveal that the alteration uniquely associated with Sd is a 5-kb tandem duplication within the polytene band 37D5, which is where Sd is known to map. The Sd-associated duplication appears to be part of a large gene about 100 kb in size. A 4.2-kb SD-specific transcript has been identified, and analyses of the cDNAs indicate a complex transcription pattern. Since Responder plays such a key role in segregation distortion, recent progress in analyzing this locus has been exciting. Studies of Rsp at the genetic, cytogenetic, and molecular levels have been mutually reinforcing and consistent in demonstrating that Rsp is an extended locus that is (1) subdivisible and (2) associated with a 120-bp repeated sequence of DNA that is rich in adenine-thymine pairs and whose copy number is correlated with the degree of sensitivity. In an exceptional situation, there are certain SdRspi/Sd+ Rsps males cartying particular modifiers in which the Rspi chromosome can he transmitted at frequencies of less than 0.50 relative to the Rsps chromosome, suggesting that the genetics of segregation ratios may involve even further complexities, which need to be unraveled.

Segregation ratios withinSegregation Distorterlines ofDrosophila melanogasterconform to a beta-binomial distribution

Segregation Distorter (SD) chromosomes are preferentially recovered from SD/SD+ males due to the dysfunction of sperm bearing the SD+ chromosome. The proportion of offspring bearing the SD chromosome is given the symbol k. The nature of the frequency distribution of k was examined by comparing observed k distributions produced by six different SD chromosomes, each with a different mean, with k distributions predicted by two different statistical models. The first model was one where the k of all males with a given SD chromosome were considered to be equal prior to the determination of those gametes which produce viable zygotes. In this model the only source of variation of k would be binomial sampling. The results rigorously demonstrated for the first time that the observed k distributions did not fit the prediction that the only source of variation was binomial sampling. The next model tested was that the prior distribution of segregation ratios conformed to a beta distribution, such that the distribution of k would be a beta-binomial distribution. The predicted distributions of this model did not differ significantly from the observed distributions of k in five of the six cases examined. The sixth case probably failed to fit a beta-binomial distribution due to a major segregating modifier. The demonstration that the prior distribution of segregation ratios of SD lines can generally be approximated with a beta distribution is crucial for the biometrical analysis of segregation distortion.

Linkage and segregation distortion in Drosophila melanogaster

Segregation distortion is caused by a group of genetic elements in and near the centric heterochromatin of chromosome 2 of Drosophila melanogaster. These elements promote their preferential recovery in heterozygous males by rendering sperm bearing the homologous chromosome dysfunctional. Previous work has shown that numerous Y-autosome translocations are associated with the suppression of the segregation distorter phenotype. The present study examined the effects of translocations between the major autosomes upon the expression of segregation distortion. Autosomal translocations involving either the segregation distorter chromosome or its sensitive homologue had no significant effect upon the expression of segregation distortion. These results argue that linkage arrangement per se may not have a major effect on segregation distortion. The suppression of SD by specific Y-autosomal translocations may be due to the disruption of elements on the Y chromosome that are important for the expression of SD.

ADDITIVE EFFECTS OF MULTIPLE SEGREGATION DISTORTER (SD) CHROMOSOMES ON SPERM DYSFUNCTION IN DROSOPHILA MELANOGASTER

A portion of the Segregation distorter (SD) chromosome, including both the Sd and E(SD) loci, has been moved by insertional translocation from SD Roma into Y(L ). This Dp(2;Y)SD chromosome shows a negligible reduction in its ability to cause dysfunction of Rsp( s)-bearing sperm when compared to the parent SD chromosome, suggesting that SD can still act effectively, even when removed from its normal second chromosome milieu, and that its activity level does not depend on pairing with a normal autosomal homologue. Male genotypes have been constructed using this Dp(2;Y)SD along with a standard SD chromosome (either SD Roma or R( SD-36)-1(bw)) and a third chromosome suppressor of SD (TM6) in all possible three-way combinations. The observed level of SD-mediated dysfunction in each case is most compatible with a model that assumes that all SD elements act additively (in terms of M, the probit transformation of the probability of sperm dysfunction), rather than multiplicatively. The additive action of SD elements contrasts with the independent response to SD activity exhibited by multiple Rsp(s) copies.

Measuring segregation distortion in Drosophila melanogaster

Segregation distorter (SD) chromosomes are preferentially transmitted to offspring of a male heterozygous for SD and SD+ chromosomes, as a result of the dysfunction of SD+-bearing sperm. Segregation distortion is measured by the segregation ratio k, the proportion of progeny bearing the SD chromosome. However, k is an ordinal measure and is therefore of only limited usefulness. A transformation has been developed (the k-probit transformation) that has been used to provide an interval method of measuring segregation distortion. This method has not been adequately tested as previous comparisons of observed k distributions with those predicted by the k-probit transformation have not taken the effects of binomial sampling into consideration. We have compared k distributions produced by three different SD chromosomes with the k distributions predicted by the k-probit transformation, utilizing a computer simulation model to account for binomial sampling. The results observed with two of the SD chromosomes do not agree with the predictions of the transformation. Thus we do not support the use of the k-probit transformation to provide a valid interval measure for segregation distortion.Key words: segregation distortion, Drosophila.

On the components of segregation distortion in Drosophila melanogaster. III. Nature of enhancer of SD.

Analysis of X-ray-induced deletions in the Segregation Distorter (SD) chromosome, SD-5, revealed that this chromosome had a gene proximal to lt in the centric heterochromatin of 2L that strongly enhanced the meiotic drive caused by the SD chromosome. This Enhancer of Segregation Distortion [E(SD)] locus had not been characterized in earlier studies of SD chromosomes because it cannot be readily separated by recombination from the Responder (Rsp) locus in the proximal heterochromatin of 2R.--To determine whether E(SD) is a general component of all SD chromosomes and to examine further its effects on distortion, we produced deletions of E(SD) in three additional SD chromosomes. Analysis of these deletions leads to the following conclusions: (1) along with Sd and Rsp, E(SD) is common to all SD chromosomes; (2) the E(SD) allele on each SD chromosome enhances distortion by the same amount, which indicates that allelic variation at the E(SD) locus is not responsible for the different drive strengths seen among SD chromosomes; (3) E(SD) causes very little or no distortion by itself in the absence of Sd; (4) E(SD), like Sd, acts in a dosage-dependent manner; (5) E(SD) exerts its effect in cis or trans to Sd; and (6) if E(SD)+ exists, its function is not related to SD.

SUPPRESSOR SYSTEMS OF SEGREGATION DISTORTER (SD) CHROMOSOMES IN NATURAL POPULATIONS OF DROSOPHILA MELANOGASTER

Several natural populations of D. melanogaster were investigated for the presence (or absence) of the Segregation Distorter ( SD) chromosomes and their suppressor systems. The SD chromosomes were found, at frequencies of a few percent, in two independent samples taken in different years from a Raleigh, North Carolina, population, whereas no SD chromosomes were found in samples collected from several populations in Texas. The populations in these localities were found to contain suppressor X chromosomes in high frequencies (75% or higher). They also contained relatively low frequencies of partial suppressor or insensitive second chromosomes of varying degrees, but completely insensitive second chromosomes were practically absent in all populations examined. The frequencies of suppressor X chromosomes, as well as those of the partially insensitive or suppressor second chromosomes, were the same among the populations investigated. This suggests the possibility that the development of a suppressor system of SD in a population could be independent of the presence of an SD chromosome. Segregation distortion appeared to be occurring in natural genetic backgrounds, but the degree of distortion varied among males of different genotypes. There were many instances in which the SD chromosomes showed transmission frequencies from their heterozygous male parents that were smaller than 0.6 and, in several cases, even smaller than 0.5. The presence of a recessive suppressor, or suppressors, of SD in natural populations was suggested.

A THEORETICAL ANALYSIS OF THE EFFECTS OF SEX CHROMOSOME ANEUPLOIDY ON X AND Y CHROMOSOME MEIOTIC DRIVE.

Extra sex chromosomes are normally detrimental to the individual carrying them. In XY (or WZ) sex determining systems, an extra X chromosome in the homogametic sex generates enough X-autosome imbalance to usually cause inviability, or at least sterility. On the other hand, extra Y chromosomes are sometimes tolerated, although in mammals and other higher organisms where the Y is actively involved in sexual development, XXY individuals are generally sterile. In Drosophila and perhaps other invertebrates where the Y is largely genetically inert, a single extra Y may be permitted in both sexes, while two extra copies is only fertile in XXYY females (Cooper, 1956), if at all. Even if XXY and XYY genotypes were perfectly viable and fertile, abnormal meiotic segregation and the consequent production of unbalanced gametes would lead to enough zygotic lethality to reduce their overall fitness (see Fig. 1). Thus we expect sex chromosome aneuploids in nature to be rare, primarily arising from recurrent spontaneous nondisjunction events. However, the literature records many cases of unusual accessory or sex chromosome constitutions found in both plants and animals (particularly insects, see White, 1973) which must have faced at least a transient period of aneuploidy during the process of their establishment. The genetic forces which might be responsible for this form of karyotype evolution are at present largely in the realm of speculation (White, 1978). I recently (Lyttle, 1981a) reported an experimental population of Drosophila melanogaster in which such aneuploids were present at high frequency. This population had been segregating for a type of Y chromosome meiotic drive (pseudo-Y or pY drive, Lyttle, 1977), which operates by coupling the Y chromosome to the autosomal drive element Segregation distorter (SD) by reciprocal translocation. Normally, pY drive males produce only male offspring, as potential daughters are removed both by the SD mediated dysfunction of X;SD+ gametes and the concomitant lethality of the hypoploid zygotes arising from fertilization events involving X;SD sperm. Since the pY drive was expected to push the male frequency to near 100% of the population, the observed high frequency of aneuploids was interpreted as a strategy for reducing this deviation of the population sex ratio from the optimal 1:1 value. This would occur because XYY males and XXY females, owing to their production of gametes with unusual sex chromosome combinations, have proportionately more daughters than do their XY and XX counterparts under the conditions of pY drive (Lyttle, 1981a). That is, since the y2 portion of the translocation carries the large missing piece of the SD chromosome, its added presence in XY;SD sperm can rescue usually hypoploid zygotes to give autosomally euploid XXY females. To investigate this apparent selection for aneuploidy, I constructed a simple discrete generation model which included effects of chromosomal segregation, fitness, and meiotic drive, the latter defined as the excess recovery in the gametes of one member of a pair of heterozygous alleles or heteromorphic chromosomes (cf. Gametic frequency of second chromosomes of the T-007 type in a natural population of Drosophila melanogaster in Texas.

The T-007 second chromosome, which was isolated from a natural population of Drosophila melanogaster in south Texas in 1970, is known to show, when made heterozygous in males with a standard cn bw second chromosome, a transmission frequency (k) of 0.35-much lower than the theoretically expected 0.5. Natural populations of this species in Texas contain second chromosomes that, against the standard cn bw genetic background, are associated with distorted transmission frequencies comparable to that of the T-007 chromosome. In order to explain how such chromosomes can persist in natural populations in nontrivial frequencies, it has been postulated that, although such chromosomes show reduced k values when tested under the genetic background of a laboratory stock such as cn bw, they may show, on the average, k values larger than 0.5 under natural genetic backgrounds. If this were true, the frequency of chromosomes of the T-007 type (T chromosomes) should be higher in male than in female gametes under natural genetic backgrounds. The present study was conducted to examine this possibility. The results clearly showed that the frequency of such chromosomes was much higher among male than among female gametes, and that the transmission frequency of this type of chromosome was higher than 0.5 under natural genetic backgrounds. These results suggest that T chromosomes behave like Segregation Distorter (SD) chromosomes in natural populations of this species in Texas. A possible relationship between T-007 and SD chromosomes is suggested.

Genetic dissection of segregation distortion. III. Unequal recovery of reciprocal recombinants.

The genetic structure of a segregation distorter chromosome (a derivative of SD-36) has been analyzed in a system in which recombination in the second chromosome is blocked by inversions except for the critical region around the centromeric heterochromatin. The results confirm the map order and characteristics of four loci known to be involved in segregation distortion, namely Sd, E(SD), Rspins, M(SD). However, SD-36 carries a fifth major locus involved in distortion. This locus is near pr in 2L and has the effect of enhancing the degree of distortion. In addition, reciprocal recombinant products from SD-36 are recovered unequally. All recombinants carrying the pr region from SD-36 seem also to carry Sd, although Sd has previously been mapped 1.6 units to the left of pr. Both the enhancement of distortion and the unequal recovery of reciprocal products can be explained if it is assumed that the new locus near pr in SD-36 is actually a duplication of Sd.
