Physical and chemical difference of seed coat between hard and soft seeds of Licorice (Glycyrrhiza Uralensis Fisch)

In this paper, the physical and chemical differences of seed coat between hard and soft seeds from the same Licorice lot were identified. Electron microscopy-based examination revealed that the cracks on hard seeds coat were much less obvious (shallower and narrower) than those of soft seeds, and that hilar fissure from hard seeds was considerably narrower. Endosperm of hard seeds was markedly thicker than that of soft seeds. Finally, X-ray fluorescence spectrometry revealed significant enrichment for 12 out of the 14 tested mineral elements in hard seed coat. It could be concluded that these above features of hard seeds would increase their resistance to physical impact and protect against cracks in seed coat surface, thus contributing to the impermeability of hard seeds.

Cistus incanus from Strandja Mountain as a Source of Bioactive Antioxidants.

The purpose of the present study is to survey the extraction conditions and explore the antioxidant potential of the wild herb Cistus incanus, which is non-traditional in Bulgarian ethnomedicine and widespread in the Strandja Mountain. The influence of the extraction time (0–500 min) and solvent composition (0–50% ethanol in water) on the polyphenols, flavonoid yields and on the antioxidant capacity of the extracts of leaves, stalks (wood parts) and bud mixture were studied. The antioxidant capacity (AOC) was evaluated by use of scavenging assays of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals. Total polyphenol and flavonoid contents were quantified using UV–vis (ultraviolet-visible) spectrophotometry. The optimal yield of the desired components was obtained with 30% ethanol in water solvent at the 390th min of extraction time. In addition, the influence of seasonality (winter and summer Cistus incanus), and of the different aerial parts—hard-coated seeds, buds, and a mixture of leaves and stalks of the wild plant—on the presence of polyphenols, flavonoids, and AOC were investigated. The present work revealed that the high values of polyphenols, flavonoids and the high AOC occurred not only in the summer leaves, but were also found in the winter leaves, hard-coated seeds, buds, and stalks. Based on the obtained results, the Cistus incanus from Strandja Mountain could be an excellent new source of natural antioxidants in food and for the pharmaceutical industries.

Enhancement of Seed Dehiscence by Seed Treatment with Talaromyces flavus GG01 and GG04 in Ginseng (Panax ginseng).

Seed dehiscence of ginseng (Panax ginseng C. A. Mayer) is affected by moisture, temperature, storage conditions and microbes. Several microbes were isolated from completely dehisced seed coat of ginseng cultivars, Chunpoong and Younpoong at Gumsan, Korea. We investigated the potential of five Talaromyces flavus isolates from the dehiscence of ginseng seed in four traditional stratification facilities. The isolates showed antagonistic activities against fungal plant pathogens, such as Cylindrocarpon destructans, Fusarium oxysporum, Rhizoctonia solani, Sclerotinia nivalis, Botrytis cinerea, and Phytophthora capsici. The dehiscence ratios of ginseng seed increased more than 33% by treatment of T. flavus GG01, GG02, GG04, GG12, and GG23 in comparison to control (28%). Among the treatments, the reformulating treatment of T. flavus isolates GG01 and GG04 showed the highest of stratification ratio of ginseng seed. After 16 weeks, the reformulating treatment of T. flavus isolates GG01 and GG04 significantly enhanced dehiscence of ginseng seed by about 81% compared to the untreated control. The candidate’s treatment of T. flavus GG01 and GG04 showed the highest decreasing rate of 93% in seed coat hardness for 112 days in dehiscence period. The results suggested that the pre-inoculation of T. flavus GG01 and GG04 found to be very effective applications in improving dehiscence and germination of ginseng seed.

Seed heat tolerance and germination of six legume species native to a fire-prone longleaf pine forest

Recognition of spatial heterogeneity of fire at fine scales is emerging, particularly in ecosystems characterized by frequent, low-intensity fire regimes. Differences in heat flux associated with variation in fuel and moisture conditions create microsites that affect survivorship and establishment of species. We studied the mechanisms by which fire affects seed germination using exposure of seeds to fire surrogates (moist and dry heat). Tolerance (survival) and germination responses of six perennial, herbaceous legume species common to the fire-prone longleaf pine–wiregrass ecosystem of the southeastern USA were examined the following heat treatments. Moist heat was more effective in stimulating germination than dry heat flux for most species examined. We also compared intrinsic seed properties (relative seed coat hardness, percent moisture, and seed mass) among species relative to their heat tolerance and heat-stimulated germination responses. Seed coat hardness was closely associated with the probability of dry and moist heat-stimulated germination. Variation among species in optimal germination conditions and degree of heat tolerance likely reflects selection for specific microsites among a potentially diverse suite of conditions associated with a low-intensity fire regime. Fire-stimulated germination, coupled with characteristics of seed dormancy and longevity in the soil, likely fosters favorable recruitment opportunities in restoration situations aimed at reintroducing a frequently prescribed burn regime to a relict longleaf pine site. In a restoration context in which externally available seed pool inputs are limited, this regenerative mechanism may provide a significant source of recruitment for vegetative recovery in a post-fire landscape.

Germination sensitivity to water stress of eight Cistaceae species from the Western Mediterranean

AbstractWater availability regulates timing of germination, especially in those environments where it is a limiting factor. However, the water requirements for germination of most wild Mediterranean species are unknown. In this work we analysed the germination response to water stress of eight Cistaceae species with hard-coated seeds, which are typical for the Mediterranean shrublands. Seeds were exposed to a heat shock in order to overcome physical dormancy and then incubated under five water potentials (0, −0.2, −0.4, −0.6 and −0.8 MPa). Ungerminated seeds following these treatments were transferred to water in order to evaluate the recovery of germination. Additionally, at the end of the experiment, viability of still ungerminated seeds was examined. We analysed final germination percentage, time to reach 50% of the final germination (t50), recovery of germination and seed viability in relation to water stress treatments. Furthermore, hydrotime analysis was performed; sensitivity to water stress, as characterized by Ψb(50), was checked in relation to seed size of species. Overall, water stress delayed and decreased germination but species showed different germination sensitivities, which were not related to seed size. Recovery of germination was high after disappearance of water stress but seed viability decreased in some species. Consequently, post-fire germination of Cistaceae must be strongly regulated by water availability and their seeds can recover germination capacity when sufficient water is available after long periods of incomplete hydration. However, some seeds die during the wait.

Strong potential for endozoochory by waterfowl in a rare, ephemeral wetland plant species, Astragalus contortuplicatus (Fabaceae)

It was proposed previously that passive dispersal by migratory aquatic birds explain the widespread distribution of many wetland organisms. Several experimental studies have shown that many widespread wetland plant species can be readily dispersed within the guts of Anatidae. However, it is unclear whether plants with a more restricted distribution are able to disperse via waterbirds. This paper addresses the dispersal ability and germination ecology of the little-known Hungarian milkvetch <em>Astragalus contortuplicatus</em>, which occurs on banks of continental rivers and has a limited and unpredictable distribution. To test whether limited capacity for endozoochory by waterfowl could explain the sporadic appearance of this species, we force-fed ten captive mallards (<em>Anas platyrhynchos</em>) with 100 milkvetch seeds each. Droppings were collected for up to 45 h after feeding. Intact and viable seeds were found in the droppings of each mallard, and altogether 24.7% of seeds fed were recovered intact. The proportion of retrieved seeds that germinated (27.0%) was significantly higher than that of untreated control seeds (0.5%), but significantly lower than that of mechanically scarified seeds (96.0%). Retrieved seeds that germinated developed into healthy mature plants. Given the average flight velocity of mallards, seeds of <em>A. contortuplicatus</em> may travel up to 1600 km inside the digestive tract of migrating individuals. Our results suggest that avian vectors may be more important for the dispersal of rare higher plants (especially those with a hard seed-coat) than hitherto considered. Moreover, they suggest that rarity does not necessarily indicate limited dispersal ability, and may instead be explained by specific habitat requirements.

Trait‐mediated seed predation, dispersal and survival among frugivore‐dispersed plants in a fragmented subtropical forest, Southwest China

By tracking the fate of individual seeds from 6 frugivore-dispersed plants with contrasting seed traits in a fragmented subtropical forest in Southwest China, we explored how rodent seed predation and hoarding were influenced by seed traits such as seed size, seed coat hardness and seed profitability. Post-dispersal seed fates varied significantly among the 6 seed species and 3 patterns were witnessed: large-seeded species with a hard seed coat (i.e. Choerospoadias axillaries and Diospyros kaki var. silvestris) had more seeds removed, cached and then surviving at caches, and they also had fewer seeds predated but a higher proportion of seeds surviving at the source; medium-sized species with higher profitability and thinner seed coat (i.e. Phoebe zhennan and Padus braohypoda) were first harvested and had the lowest probability of seeds surviving either at the source or at caches due to higher predation before or after removal; and small-seeded species with lower profitability (i.e. Elaeocarpus japonicas and Cornus controversa) had the highest probability of seeds surviving at the source but the lowest probability of seeds surviving at caches due to lower predation at the source and lower hoarding at caches. Our study indicates that patterns of seed predation, dispersal and survival among frugivore-dispersed plants are highly determined by seed traits such as seed size, seed defense and seed profitability due to selective predation and hoarding by seed-eating rodents. Therefore, trait-mediated seed predation, dispersal and survival via seed-eating rodents can largely affect population and community dynamics of frugivore-dispersed plants in fragmented forests.

Post‐dispersal seed predation and its relations with seed traits: a thirty‐species‐comparative study

AbstractPost‐dispersal seed predation is a key process determining the variability in seed survival in forests, where most seeds are handled by rodents. Seed predation is thought to affect seedling regeneration, colonization ability and spatial distribution of plants. Basic seed traits are the essential factors affecting rodent foraging preferences and thus seed survival and seedling recruitment. Many studies have discussed several seed traits and their effects upon seed predation by rodents. However, the results of those previous studies are usually equivocal, likely because few seed traits and/or plant species tend to be incorporated into these studies. In order to elucidate the relationships between seed predation and seed traits, we surveyed the predation of 48 600 seeds in a natural pine forest, belonging to 30 species, for three consecutive years. The results demonstrated that: (i) seed size and seed coat hardness did not significantly affect seed predation; (ii) total phenolics had a negative effect upon seed predation; (iii) positive effects of nitrogen content upon seed predation were found. From our study, it seems that the better strategy to prevent heavy predation is for plants to produce seeds with higher total phenolics content rather than physical defenses (i.e. hard seed coat) or larger seeds. Additionally, rodent foraging preference may depend more on Nitrogen content than other nutrient content of seeds.

Seed biology and pre-sowing treatment of Peltophorum pterocarpum (DC.) Backer ex K. Heyne

Peltophorum pterocarpum a multipurpose tree species produces hardcoated and light brown colour seeds. Seed has physical dormancy due to hardiness of testa. Suitable pretreatment to break the dormancy was investigated. Germination was enhanced from 19.50 % (untreated) to 92.30 % (mechanical scarification). Although mechanical scarification treatment gives early, uniform and highest germination but it is not practicable in large-scale purposes. Therefore, hot water soaking for 24 hours or sulphuric acid for 30 minutes soaking is recommended.

Seed-coat anatomy and proanthocyanidins contribute to the dormancy of Rubus seed

Rubus seed has a deep double dormancy that restricts germination due to seed coat structure and chemical composition. Improved germination of diverse Rubus species required for breeding improved blackberry and raspberry cultivars is partly dependent on the seed coat structure. This study evaluated the seed coat structure of three species with thin ( R. hoffmeisterianus Kunth & C. D. Bouché), medium ( R. occidentalis L.) and thick ( R. caesius L.) seed coats. The three species exhibited distinctive seed-coat cell composition. The very thin testa (0.086 mm) of R. hoffmeisterianus had little exotesta (surface) reticulation; with the meso- and endotesta composed of sclereids of homogenous shape and size. R. occidentalis had a thick testa (0.175 mm) and a highly reticulate exotesta; the meso- and endotesta were composed of several diverse types of sclereids. R. caesius had the thickest seed coat (0.185 mm) but only moderate exotesta reticulation; the meso- and endotesta were composed of large, irregular, loosely arranged sclereids. R. occidentalis, a medium size seed, was the most heavily lignified with seed-coat thickness similar to R. caesius, the largest seed. Proanthocyanidins (PAs) from dry seed of six Rubus species were extracted and quantified by high performance liquid chromatography. R. hoffmeisterianus, a thin only slightly hard seed, had half the PA (0.45 μg/seed) of R. occidentalis with a thick, extremely-hard seed coat and diverse sclereids (1.07 μg/seed). PA content and sclereid composition both appear contribute to seed coat hardness and resulting seed dormancy. The effectiveness of sulfuric acid for Rubus seed scarification is likely due to degradation of PAs in the testa.

Seed Removal and Predation as Factors Affecting Seed Availability of Tree Species in Degraded Habitats and Restoration Plantings in Rainforest Areas of Queensland, Australia

The post-dispersal removal or predation of seeds of native tree species was investigated in Queensland, Australia, at degraded habitats and rainforest restoration sites where direct seeding might be used to facilitate tree regeneration (old fields or open habitats, lantana thicket, rainforest edge, and 5- and 10-year-old restoration plantings). Seed removal/predation was assessed in relation to tree seed weight and canopy density of the habitats during the wet season period. Results indicated that seed removal/predation imposes limitations on seed availability, particularly for small seeded species. In most situations, larger seeds were less removed/predated, most likely due to the more limited range of large seed consumers. The use of large, hard-coated seeds may potentially reduce seed loss in open situations (from both seed removal and desiccation), unless large seed consumers frequent the site. Canopy cover exerted an influence on seed removal/predation, though trends varied in relation to site and the time of season. Broadcast sowing of seed under planted tree canopies at the more advanced stages of closure may in some areas result in higher seed removal/predation. Likewise, seeding in areas dominated by woody weeds may result in high seed losses to consumers such as rodents. Results suggested that undertaking direct seeding to coincide with the maximal period of fruit production may in some situations be beneficial to minimize seed loss. Overall, site context, canopy cover, and species selection appear to be important considerations when aiming to reduce loss of seeds to animal seed consumers in restoration work.

Effects of soil temperature regimes after fire on seed dormancy and germination in six Australian Fabaceae species

In addition to direct fire cues such as heat, smoke and charred wood, the passage of fire leads indirectly to changes in environmental conditions which may be able to break physical dormancy in hard-coated seeds. After a fire, the open canopy and the burnt material lying on the surface alter the thermal properties of the soil, resulting in elevated soil temperatures for long periods of time. We simulated daily temperature regimes experienced at different depths of soil profile after a summer fire. Our aim was to determine whether these temperature regimes and the duration of exposure (5, 15 and 30 days) play an important role breaking physical seed dormancy in six legumes from south-eastern Australia. Our results showed that simulated temperature regimes break seed dormancy. This effect is specially pronounced at temperatures that are expected to occur near the soil surface (0–2 cm depth). The duration of exposure interacts with temperature to break dormancy, with the highest germination rates reached after the longest duration and highest temperatures. However, the germination response varied among species. Therefore, this indirect post-fire cue could play a role in the regeneration of plant communities, and could stimulate seedling emergence independent of direct fire cues as well as in interaction with direct cues.

Using endocarp-remains of seeds of wild apricot Prunus armeniaca to identify rodent seed predators

Abstract Some rodent-dispersed seeds have a hard seed-coat (e.g.woody endocarp). Specific scrapes or dental marks on the hard seed-coat left by rodents when they eat these seeds can be used to identify seed predators. In this study we measured the morphological traits of endocarp-remains of seeds of wild apricot Prunus armeniaca used by Chinese white-bellied rats Niviventor confucianus and Korean field mice Apodemus peninsulae. We established their Fisher’s linear discriminant functions to separate endocarp-remains between the two predators. A total of 90.0% of the endocarp-remains left by Korean field mice and 88.0% of those left by Chinese white-bellied rats were correctly classified. The overall percentage of correct classification was 89.0%. One hundred and sixty endocarp-remains of unknown what species predated them were classified using the functions. The method may allow more reliable quantitative studies of the effects of Chinese white-bellied rats and Korean field mice on seed consumption and dispersal of wild apricot and this study might be used for reference in other studies of seed predators identification on hard seeds.

Seed germination and in vitro propagation of Piliostigma thonningii – an important medicinal plant

Piliostigma thonningii is a multipurpose tree of high priority for conservation in Nigeria. Almost all its parts are used in traditional medicine and its seeds are a good source of antioxidant micronutrients, rich in crude protein and carbohydrate. Its seeds are however dormant and the plant is uncultivated. Seed germination and in vitro regeneration experiments were carried out on P. thonningii. Seed dormancy was successfully broken by physical and chemical scarification using concentrated sulphuric acid for 15 min with 91.7 ± 4.01 and 95.0 ± 2.24% germination, respectively. The seed dormancy was due mainly to the hard seed-coat. Callus production was observed in both cotyledon and hypocotyl explants cultured in auxin-supplemented Murashige and Skoog (MS) medium. Explant orientation on the medium and the position from which the explant is taken affected callus production. Complete plantlet was regenerated from the hypocotyl-derived callus in medium containing auxin only.

Dietary plant use by middle Holocene foragers in the Bonneville Basin, western North America

Middle Holocene foragers in the Bonneville basin, western North America, developed a subsistence strategy that differed in emphasis and orientation compared with their early Holocene predecessors. This paper examines one aspect of this changing middle Holocene subsistence orientation, the increasing dietary use of plants such as small hard-coated seeds (including iodinebush, pepperweed, goosefoot, and others), bulrush rhizomes, cactus pads and fruits, and to a lesser degree a few upland plant foods (pine nuts and various fruits). Middle Holocene archaeobotanical assemblages are reviewed for four sites: Danger Cave, Bonneville Estates Rockshelter, Camels Back Cave, and Hogup Cave. The middle Holocene shift toward extensive use of small seeds and plant foods from varied upland and desert environments reflects intensified use and processing of low-ranked plant resources, focusing on resource reliability, in the context of widespread regional aridification.

Fire drives phylogenetic clustering in Mediterranean Basin woody plant communities

Summary 1 Many Mediterranean plant species persist after fire because their seeds are protected from the heat of the fire (e.g. hard-coated seeds, serotinous cones), thus permitting rapid post-fire recruitment. For simplicity, this trait will hereafter be called P and its two possible phenotypes P+ (seeder) and P– (non-seeder). 2 Because P+ appears in a narrow taxonomic spectrum and confers persistence under high fire frequencies, we test the extent to which communities with different fire histories have different phenotypic and phylogenetic structures. Specifically, we compare coastal vegetation growing in a warm and dry Mediterranean climate subject to high fire frequency (HiFi vegetation) with montane vegetation subject to a subhumid climate where fires are rare (LowFi) under the hypothesis that P+ species will be over-represented in HiFi communities, thus producing phenotypic and phylogenetic clustering. 3 Trait conservatism on P is evaluated by testing the presence of a phylogenetic signal, phenotypic clustering is tested by correlating co-occurrence and phenotypic distance matrices, and the phylogenetic structure is evaluated by testing whether the phylogenetic distances between species in each community are different from those expected by chance. 4 The results suggest that: (a) P is a strongly conserved trait; (b) co-occurring species have similar P phenotypes (phenotypic clustering); and (c) the phylogenetic structure in HiFi vegetation is significantly clustered while LowFi vegetation tends to be overdispersed. 5 Synthesis: Fire is a strong driving force in assembling HiFi communities while other forces, such as competitive interactions, are the main assembly mechanisms in LowFi communities. This result supports the role of recurrent disturbances as filters driving phylogenetic community structure.

Seed morphology and ultrastructure in Citrus garrawayi (Rutaceae) in relation to germinability

Combinational traits of seed size and seed-coat hardness in Citrus garrawayi (F.M.Bailey) (syn. of Microcitrus garrowayi) were investigated as markers for estimation of seed morphological and physiological maturity. Seed size (length) and coat hardness correlated well with changes in seed coat and embryo morphological development, dry-weight accumulation, decreases in moisture content and a significant increase in germinability. Seed moisture content decreased from 82 ± 1% in immature seeds to 40 ± 1% at seed maturation. The outer integument of immature seeds consisted of thin-walled epidermal fibres from which outgrowths of emerging protrusions were observed. In comparison, mature seed coats were characterised by the thickening of the cell walls of the epidermal fibres from which arose numerous protrusions covered by an extensive mucilage layer. Immature seeds, with incomplete embryo and seed-coat histodiffereniation, had a low mean germination percentage of 4 ± 4%. Premature seeds, with a differentiated embryonic axis, were capable of much higher levels of germination (51 ± 10%) before the attainment of mass maturity. Mature seeds, with the most well differentiated embryonic axis and maximum mean dry weight, had the significantly highest level of germination (88 ± 3%).

Effects of logging, liana tangles and pasture on seed fate of dry forest tree species in Central Brazil

We assessed seed fate of six tree species in a seasonally deciduous forest of Central Brazil. Seed germination, predation, removal and death caused by pathogens or dessication were followed in an undisturbed forest site, a logged forest site, and an active pasture. In both forest types we sowed seeds under patches of liana tangles (Low Forest habitat) and under patches of mature forest (High Forest habitat). Seeds were monitored weekly for up to 8 months. Overall, small-seeded species ( Astronium fraxinifolium and Tabebuia impetiginosa) had high germination, although germination was lower in the pasture due to insect predation. The hard-coated seeds of Erythrina sp. had higher germination rates in the pasture. The large-seeded Cavanillesia arborea and Swartzia multijuga had high seed removal (ca. 90%) in the pasture and under High Forest, but only 23–34% under Low Forest in the logged site. Lower removal of large seeds in Low Forest suggests that this habitat conceals the seeds and constrains the movement of large mammals. Both species were favored in Low Forest, suggesting that caution should be used when managing lianas. When protected from predation, seeds with a soft coat and high water content, such as the seeds of Eugenia dysenterica and S. multijuga, died from desiccation in the pasture. Restoration efforts must take into consideration high mortality caused by desiccation or insect predation in abandoned pastures of dry forests.

Effects of burial by sand and inundation by fresh- and seawater on seed germination of five tropical beach species

Plants growing on the beach are exposed to substrate mobility, freshwater inundation, and immersion in seawater, which greatly affect germination and species spatial distribution. In greenhouse experiments, we tested whether the most abundant species on the beach at La Mancha (on the Gulf of Mexico) were tolerant to burial and flooding. We buried seeds of Canavalia maritima, Ipomoea pes-caprae, Chamaecrista chamaecristoides, Palafoxia lindenii, and Ambrosia artemisiifolia at different depths. We used fresh- and seawater flooding with two frequencies: permanent and intermittent. The hard-coated seeds of Canavalia, Ipomoea, and Chamaecrista were left scarified and not scarified to test if dormancy was broken by the flooding regimes. Germination decreased as burial depth increased. We calculated a tolerance index for the burial responses, which indicated that Chamaecrista was the most tolerant species, followed by Canavalia and Palafoxia. Flooding inhibited germination, but more so permanent seawater flooding; it was especially lethal for species closest to the ocean such as Canavalia and Ipomoea. After transfer to freshwater, seeds of Chamaecrista, Palafoxia, and Ambrosia germinated. There was no evidence that hard-coat dormancy was broken by flooding. Tolerance to burial seems to be related to the spatial distribution of species but not to germination responses to flooding.Key words: germination, sand burial, freshwater, seawater, flooding, Gulf of Mexico.

Influence of a low-intensity fire on a Pinus halepensis Mill. forest seed bank and its consequences on the early stages of plant succession

The effect of an experimental low-intensity fire on the soil seed bank and its consequences on the early stages of post-disturbance succession were analyzed. The hypothesis that low-intensity fire may produce a population decline of hard-seeded species by the elimination of established plants and the lack of a seed germination promotion effect was investigated. Consecutive pre- and post-burning soil samplings revealed extremely low impact of fire on seed banks. In contrast, some specific seed banks increased with fire, probably due to a seed rain from fire-opening of capsule fruits. Fire neither promoted germination of hard-coated seeds nor affected their viability. Both the length of the seed-unproductive juvenile phase and post-fire habitat conditions were revealed as critical in determining the recovery of hard-seeded species after a low-intensity fire.
