In vitro antiradical properties and total phenolic contents in methanol extract/fractions from bark of Schleichera oleosa (Lour.) Oken

This study was aimed at evaluating the free radical scavenging activity of methanol extract and fractions from the bark of Schleichera oleosa (Lour.) Oken by employing various well-established in vitro systems such as 2,2′-diphenyl-1-picrylhydrazyl (DPPH), deoxyribose degradation (non-site specific and site specific), reducing power, chelating power, and plasmid nicking assays. Total Phenol Content of the extracts was determined by the assay based on Folin-Ciocalteu’s method. In all the assays, it was observed that the residue fraction, left after the precipitation, was more effective in scavenging the free radicals than the aqueous extract and precipitates. The higher activity of residue fraction may be attributed to the greater amount of phenolic content present in it (942.4 mg/g GAE) as compared to precipitates and aqueous extract. The extract and fractions were found to possess potent antiradical properties, which may be due to either direct scavenging of free radicals or through metal chelation.

Free Radical Scavenging Activity of Methanol Extracts of Chungkukjang

To further the goal of isolating Bacillus sp. from commercial chungkukjang (CKJ) for a development of a probiotic dietary adjunct using soymilk or milk, antioxidant activity of CKJ purchased from the Sunchang Traditional Village in Chunbook province was examined. Six CKJ samples were evaluated and 3 were selected based on the results of the physicochemical analysis and sensory evaluation for further antioxidant study. IC 50 for DPPH scavenging activity of methanol extracts of CKJ ranged from 238.1 to 345.7 ㎍/mL. CKJ exhibited over 80% scavenging of ?OH and ONOO- at concentrations of 100 ㎍/mL and 250 ㎍/mL, respectively. O₂? and NO scavenging activities of three CKJ were increased in a dose dependent manner with the concentration tested from 100 to 1000 ㎍/mL. In this study, the methanol extract of CKJ exhibited a great reduction capability and powerful free radical scavenging activity, especially against OH? and ONOO-, which are the most toxic radicals responsible for oxidative damage in the body. However, radical scavenging effects of CKJ on DPPH, O₂?, and nitrite radical were rather moderate. In conclusion, CKJ may reduce the oxidative stress in the body by scavenging the free radicals.

Antioxidant and free radical scavenging activity of methanol extract of chungkukjang

An assessment of chungkukjang (traditional fermented soyfood) fermented at 42 °C for 72 h with six sample groups of Korean black soybean ( Glycine max) was carried out. Total phenol and isoflavone contents of large black soybean (LBS) group extracts were higher than that of small black soybean (SBS) group extracts. Genistein contents were higher than daidzein in all groups. In total anthocyanin pigment, LBS groups were similar to SBS groups. LBS group extracts exhibited good inhibition rate of about 70–90% in antioxidant and some scavenging activities of free radicals as well as hydrogen peroxide. Bacillus megaterium SMY-212 was a suitable fermenting strain to promote the antioxidant and free-radical scavenging activities in cooked black soybean.

Antioxidant and Free Radical Scavenging Activities of Ervatamia coronaria Stapf. leaves

The present study was carried out to evaluate the antioxidant and free radical scavenging activity of methanolic extract of Ervatamia coronaria leaves (Apocynaceae) in various systems. DPPH radical, superoxide anion radical, nitric oxide radical and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the extract. The antioxidant activity of methanolic extract increased in a dose dependent manner. About 50, 100, 250 and 500 µg of methanol extract of Ervatamia coronaria (MEEC) showed 61.33, 66.21, 72.04 and 76.83% inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MEEC on reducing power increases in a dose dependent manner. In DPPH radical scavenging assay the IC50 value of the extract was found to be 167.09 µg/ml. MEEC was found to inhibit the nitric oxide radicals generated from sodium nitroprusside. The IC50 value was found to be 83.375 µg/ml, whereas the IC50 value of curcumin was 20.4 µg/ml. Moreover, the MEEC was found to scavenge the superoxide generated by PMS/NADH-NBTsyst em. MEEC was also found to inhibit the hydroxyl radical generated by Fenton's reaction, where the IC50 value of MEEC was found to be more than 1000 µg/ml and for catechin the IC50 value was found to be 5 µg/ml, which indicates the prooxidant activity of MEEC. The amounts of total phenolic compounds were also determined in this study. The results obtained in the present study indicate that the MEEC can be a potential source of natural antioxidant.

Biological activities of the essential oils and methanol extract of Origanum vulgare ssp. vulgare in the Eastern Anatolia region of Turkey

Abstract The present study was conducted to evaluate the antimicrobial activities, antioxidant and properties of essential oils and methanol extracts of Origanum vulgare ssp. vulgare plants. The chemical composition of a hydrodistilled essential oil of O. vulgare ssp. vulgare was analyzed by a GC/MS system. A total 62 constituents were identified. Caryophyllene and spathulenol were found to be the main constituents, followed by germacrene-D and α-terpineol. Antioxidant activity was measured employing two methods namely, scavenging of free radical DPPH and the inhibition of linoleic acid oxidation by methanol extracts and the essential oil of O. vulgare ssp. vulgare . Antioxidant studies suggested that methanol extract behaved as a strong free radical scavenger providing IC 50 at only 9.9 μg/ml, whereas the oil showed weaker activity with IC 50 at 8.9 mg/ml. Total phenolic constituents based on gallic acid equivalents revealed the presence of total soluble phenolics in the extract as 220 μg/mg dry extract (22%, w/w) and, most probably, they are responsible for the radical scavenging activity of methanol extracts. Methanol extract was not effectively able to inhibit linoleic acid oxidation and only 32% inhibition was achieved at 2 mg/ml concentration, far below that of the positive control (butylated hyroxytoluene, BHT) at the same concentration. However, 2.2 mg/ml essential oil solutions provided 50% inhibition in the linoleic acid oxidation test system. The antimicrobial test results showed that the essential oil of O. vulgare ssp. vulgare had great potential of antimicrobial activity against all 10 bacteria, and 15 fungi and yeast species tested. In contrast, the methanol extract from aerial parts of O. vulgare plant showed no antimicrobial activity. The result may suggest that the essential oil O. vulgare ssp. vulgare possesses compounds with antimicrobial properties as well as antioxidant activity, and therefore can be used as a natural preservative ingredient in food and/or pharmaceutical industry.
