Antimicrobial and antioxidant effects of an edible coating of Lepidium sativum seed mucilage and Satureja hortensis L. essential oil in Uncooked lamb meat

The current research aimed to fabricate an active edible coating, Lepidium sativum seeds mucilage (LSSM) loaded with Satureja hortensis essential oil (SHEO) (0, 0.5, 1, and 1.5 % (w/w)) for further extension of the shelf life of refrigerated lamb meat during storage for 18 days at (4 °C). The control and the coated lamb meat samples were continually analyzed for microbiological (total count, psychrotrophic count, yeast and mold), chemical (pH, moisture content, PV, and TBA), and sensory characteristics. Consistent with the microbiological analysis, the LSSM incorporated with 1, and 1.5% SHEO caused a significant shelf-life extension of the lamb meat by 15 and 18 days, respectively. These results suggested that (LSSM+1.5%S) sufficiently hindered the overall bacterial growth in the lamb meat sample since the shelf life in this treatment was 12 days more in comparison to the control. Hence, LSSM containing SHEO is very useful as an active packaging in order to preserve the quality and to prolong the lamb meat shelf life at 4 °C.

Design and characterization of chitosan-based films incorporated with summer savory (Satureja hortensis L.) essential oil for active packaging

In this study, biodegradable films were fabricated by using cross-linked chitosan nanoparticles containing different concentrations (0, 1.0, 1.2, 1.4, and 1.5 %, v/v) of Satureja hortensis essential oil (SHEO) and their physicochemical, mechanical, antimicrobial, morphological, structural and antioxidant properties were analyzed. SHEO incorporation improved antibacterial, antioxidant and thermal properties of the films. Four studied bacteria were efficiently inhibited by films loaded with SHEO. According to the FE-SEM analysis, control film had nano-sized pores while micron sized particles were present in SHEO incorporated films. Increase in color difference was well correlated with SHEO concentration. The addition of SHEO decreased elongation at break (EB) and caused an irregular fluctuation in the tensile strength (TS) values. Increase in essential oil concentration resulted in lower water solubility. The FTIR spectra of films showed evidence of interactions and molecular arrangements when SHEO was added to the polymer matrix. Overall, the findings demonstrated that SHEO incorporated chitosan-based films were successfully prepared by cross linking and film properties were remarkably affected from SHEO concentration.

Protective effects of summer savory (Satureja hortensis) oil on growth, biochemical, and immune system performance of common carp exposed to pretilachlor herbicide

There are many reports on the deleterious effects of herbicides on aquatic organisms which lead to tremendous biological, environmental and economical damage. In this regard, in the present study, the protective effect of summer savory (Satureja hortensis) essential oil (SEO) against pretilachlor, one of the most used herbicides was investigated in common carp (Cyprinus carpio). The fish assigned to six treatment groups (T1: control treatment; T2: 25% LC50 pretilachlor herbicide; T3: 50% LC50 pretilachlor herbicide; T4: 1% SEO; T5: 25% LC50 pretilachlor herbicide + 1% SEO; and T6: 25% LC50 pretilachlor herbicide + 1% SEO) for 21 days. The results showed that the SEO-containing treatments significantly increased the survival rate (SR) (P < 0.05). The highest final weight (FW), specific growth rate (SGR), and feed conversion ratio (FCR) were observed in the T4 treatment (P < 0.05). There was a significant increase in glucose (GLU) level in pretilachlor treatments and a significant decrease in SEO-containing treatments compared to the control (P < 0.05). The significantly highest total protein (TP) content was observed in T4 treatment containing SEO. Cholesterol (CHOL) and triglyceride (TRIG) levels decreased in SEO-containing treatments with the lowest level in T4 treatment (P < 0.05). Alternative complement pathway activity (ACH50), activity levels of superoxide dismutase (SOD), and glutathione peroxidase (GPX) showed an increasing trend in SEO-containing treatments with the highest level in T4 treatment (P < 0.05). The activity of liver enzymes showed a significantly lowest level in T4 treatment. To conclude, our findings revealed that the use of SEO in fish exposed to pretilachlor herbicide could improve growth, strengthen the immune system and exert a protective effect on common carp.

Green Synthesis of Magnetic Nanoparticles Using Satureja hortensis Essential Oil toward Superior Antibacterial/Fungal and Anticancer Performance.

The biological synthesis of nanoparticles, due to their environmental and biomedical properties, has been of particular interest to scientists and physicians. Here, iron nanoparticles (FeNPs) were synthesized using Satureja hortensis essential oil. Then, the chemical, functional, and morphological properties of these nanoparticles were characterized by typical experiments such as Uv-Vis, FTIR, XRD, FE-SEM, PSA, zeta potential, EDX, and EDX mapping. The results indicated Fe nanoparticles' formation with a cubic morphological structure and a particle size in the range of 9.3-27 nm. The antimicrobial effects of these nanoparticles were further evaluated using disc diffusion, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and minimum fungal concentration (MFC) against two gram-positive bacterial strains (Staphylococcus aureus and Corynebacterium glutamicum), two gram-negative bacterial strains (Pseudomonas aeruginosa and Escherichia coli), and one fungus species Candida albicans. The results showed that green-synthesized Fe nanoparticles possessed higher antimicrobial properties than Satureja hortensis essential oil against selected pathogenic microorganisms, especially Gram-negative bacteria. Finally, the anticancer effect of these Fe nanoparticles was investigated on human cancer cells, K-562, and MCF-7, by the MTT assay. The results showed the anticancer effect of these nanoparticles against selected cell lines.

Shelf‐life extension of pomegranate arils using chitosan nanoparticles loaded with Satureja hortensis essential oil

This study was conducted in two parts to improve the antimicrobial activity and stability of Satureja hortensis essential oil (SEO) and its impacts on the quality of pomegranate arils. In the first part, SEO was encapsulated by an ionic gelation technique into 142.2-267.7nm chitosan nanoparticles (CSNPs). In the second part of the experiment, the CSNPs and CSNPs-SEO were applied to improve storability of pomegranate arils. Arils were dipped in water (control), CSNPs and CSNPs-SEO for 5min. After superficial water removal, arils were packed into polystyrene boxes and stored at 5°C for 18 days. Based on spectrophotometry analysis, the encapsulation efficiency (EE) of SEO-loaded CSNPs (CSNPs-SEO) decreased from 26.57% to 7.41% and their loading capacity (LC) increased from 4.72% to 6.17%, respectively, upon increasing the initial SEO content from 0.125 to 0.5g g-1 of chitosan. Phytochemicals and water content were maintained, and microbial counts were reduced in the coated arils during storage. Total phenol and antioxidant activity decreased during storage. At the end of storage, the highest total phenol content (2980.0mg gallic acid equivalents L-1 ) was found in arils treated with CSNPs-SEO, whereas ascorbic acid content was maximal (6.32 mg L-1 ) in arils treated with CSNPs. The encapsulation of savory essential oil in chitosan nanoparticles did not have undesirable effects in pomegranate arils. Pre-storage treatment of pomegranate arils with CSNPs-SEO could be considered a beneficial treatment to better maintain the biochemical and sensorial quality during storage. © 2020 Society of Chemical Industry.

The Lethal Effect of a Nano Emulsion of Satureja hortensis Essential Oil on Protoscoleces and Germinal Layer of Hydatid Cysts

New scolicidal agents and novel therapeutic drugs are essential for better management of the zoonotic infection, hydatid disease. This study evaluated the effect of a nanoemulsion (NE) of Satureja hortensis essential oil (SHEO) on protoscoleces and germinal layer of hydatid cysts. This study was conducted from July to October 2016 in Shiraz University, Shiraz, Iran. Gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) were performed to identify the main components of SHEO. To determine the scolicidal power of the NE of SHEO, live protoscoleces of hydatid cysts were exposed to two concentrations (1 and, 2 mg/mL) of the NE and incubated at 37 °C for 10 and 20 min. To evaluate the anti-hydatid effect of the NE of SHEO, the collected hydatid cysts from the abdominal cavities of the experimentally infected mice were immersed in the NE (0.5 mg/ml) and incubated at 22 °C for 24 h. Carvacrol and γ-terpinene were the major components of the SHEO. NE of SHEO at the concentrations of 1 and 2 mg/mL showed 100% scolicidal power after 20 and 10 min respectively. Exposure of the hydatid cysts to the NE of SHEO resulted in crumpling of their germinal layer and detachment of this layer from the laminated layer. NE of SHEO showed a strong scolicidal activity as well as a profound lethal effect on the germinal layer of hydatid cysts. Accordingly, this product may be used as a natural scolicidal agent in hydatid cyst surgery. Furthermore, it may be used as a therapeutic tool for treatment of hydatid disease.

Exploring Ecological Alternatives for Crop Protection Using Coriandrum sativum Essential Oil.

Essential oils (EOs) are a natural source of active compounds with antifungal, antimycotoxigenic, and herbicidal potential, and have been successfully used in organic agriculture, instead of chemical compounds obtained by synthesis, due to their high bioactivity and the absence of toxicity. The aim of this study was to highlight the importance of Coriandrum sativum essential oil (CEO) as a potential source of bioactive constituents and its applications as an antifungal and bioherbicidal agent. The CEO was obtained by steam distillation of coriander seeds and GC-MS technique was used to determine the chemical composition. Furthermore, in vitro tests were used to determine the antifungal potential of CEO on Fusarium graminearum mycelia growth through poisoned food technique, resulting in the minimum fungistatic (MCFs) and fungicidal concentrations (MCFg). The antifungal and antimycotoxigenic effect of CEO was studied on artificially contaminated wheat seeds with F. graminearum spores. Additionally, the herbicidal potential of CEO was studied by fumigating monocotyledonous and dicotyledonous weed seeds, which are problematic in agricultural field crops in Romania. The in vitro studies showed the antifungal potential of CEO, with a minimum concentration for a fungistatic effect of 0.4% and the minimum fungicidal concentration of 0.6%, respectively. An increase in the antifungal effects was observed in the in vivo experiment with F. graminearum, where a mixture of CEO with Satureja hortensis essential oil (SEO) was used. This increase is attributed to the synergistic effect of both EOs. Moreover, the synthesis of deoxynivalenol (DON)-type mycotoxins was found to be less inhibited. Hence, CEO has shown an herbicidal potential on weed seeds by affecting inhibition of germination.

Encapsulation of Satureja hortensis L. (Lamiaceae) in chitosan/TPP nanoparticles with enhanced acaricide activity against Tetranychus urticae Koch (Acari: Tetranychidae)

The objective of this study was the fabrication of encapsulated Satureja hortensis essential oil (S.EO) in chitosan/tripolyphosphate nanoparticles (CS/TPP-NPs) via ionic gelation technique and investigation of its acaricidal effect. A high encapsulation efficiency of 96.17% was obtained, which shows successful encapsulation of EOs in CS-TPP nanoparticles. Transmission electron microscopy (TEM) analysis proved the formation of spherical S. hortensis EO-loaded chitosan nanoparticles (S.EO@NPs). Fourier-transform infrared spectroscopy (FTIR) analysis demonstrated the presence of encapsulated S.EO in CS/TPP nanoparticles. The average size of nanoparticles was found to be 192.1 ± 11 nm using dynamic light scattering (DLS) technique. Moreover, durability and fumigant acaricide activity of S.EO@NPs against Tetranychus urticae Koch were investigated. The obtained results demonstrated that there were considerable differences between pure S. hortensis EO and S.EO@NPs in their ovicidal, adulticidal, and persistence activities against T. urticae. The LC50 values of pure EO and as-prepared S.EO@NPs against adult mite were 4.95, 46.98 µL/L after 24 h exposure and 2.02, 31.30 µL/L after 72 h exposure, respectively. Fumigation exposure for 24 and 72 h showed that the sensitivity of adults T. urticae were more than the eggs of T. urticae. The LC50 values of fumigant toxicity of pure S.EO and S.EO@NPs against eggs of T. urticae were measured after 24 and 72 h. The experimental results for 24 h treatment showed 6.71 and 211.66 µL/L air LC50 values for pure S.EO and S.EO@NPs, respectively. In the case of 72 h exposure, The LC50 values of pure S.EO and S.EO@NPs were 4.15 and 107.38 µL/L air, respectively. A sustained release of S.EO from S.EO@NPs was observed during 25 days of the study, indicating the persistence acaricide activity for a long time. The as-prepared S.EO@NPs and pure S.EO illustrated 67% and 2% mortality at 18th day exposure, respectively. The notable increasing of the residual fumigant toxicity may be related to the slow and sustainable release of the active ingredient of EO. Based on this study, the S.EO@NPs showed significantly residual adulticidal activity against adults of T. urticae. S.EO@NPs would be recommended as an alternative for pure EOs and other common acaricides.

Antimicrobial efficacy of some plant extracts on bacterial ring rot pathogen, clavibacter michiganensis ssp. sepedonicus

Abstract Control of plant bacterial diseases remains difficult due to the limited availability of efficient plant protection products with reduced negative effects either in the environment or with human and animal health. In order to reduce the usage of chemical pesticides alternative strategies for controlling plant pathogens and improve plant disease resistance are promoted. The aim of the study was to investigate the antibacterial activity of some natural compounds (plant extracts of Tamarix ramosissima, Rosmarinus officinalis, Chelidonium majus, Silybum marianum, Satureja hortensis essential oil and propolis) against bacterial ring rot pathogen, Clavibacter michiganensis ssp. sepedonicus (Cms). An agar diffusion method was used for the screening of the inhibitory effect of natural compounds on bacterial strains’ growth. Minimum inhibition concentrations (MICs) were determined by a twofold serial dilution method. The anti-pathogenic activity was investigated by the study of anti-biofilm activity of natural substances. The analyzed natural substances showed a good microbicidal activity and anti-biofilm activity. The results obtained from this study may contribute to the development of new bio-control agents as alternative strategies for prevention and control of ring rot pathogen.

This experiment was carried out to study the effects of dietary supplementation of Satureja hortensis essential oil on growth, carcass parts, pH and luminal microbe in broiler chicken. A total of 400 one week old ROS 308 male chicks were used in a completely randomize design model with 5 treatments and 4 replicates, each composing of 20 chicks. Duration of experiment was 35 days until the age of 42 days. Treatments were a basal diet supplemented with 50, 100 and 150 mg/kg of S. hortensis essential oil with a negative (no supplementation) and a positive control (containing 300 g/1000 kg) of Orego-Stim (a commercial compound). Feed intake and body weight gain were measured weekly and feed conversion ratio was calculated accordingly. Chicks were reared on litter and were fed ad libitum with antibiotic and coccidiostat free diet. At 35th day, litter samples was collected and sent to laboratory for determining its moisture and bacterial total count. At the 42nd day, two chicks per cage were slaughtered for determining carcass parts ratio, ileum digesta pH and bacterial count by dilution method. Results showed that S. hortensis essential oil and Orego-Stim compound had no effects on broiler growth. Liver relative weight increased by 50 and 150 ppm S. hortensis essential oil as compared to the control group. No change of pH was observed due to treatments. Chicks that received 150 ppm S. hortensis essential oil showed higher litter bacterial count as compared to the control and other treatments. Antibacterial Activity of Essential Oil of Sature jahortensis Against Multi-Drug Resistant Bacteria

Results: The antibiotic resistance profile of the E. coli isolates were as follows: ceftazidime (50%) cefixime (41.6%), tetracycline (75%), erythromycin (58.3%). However K. pneumoniae isolates showed resistance to ceftazidime (33.3%), cefixime (58.3%), erythromycin (75%) and S. aureus isolates were resistant to cefixime (33.3%), trimethoprim-sulfamethoxazole (41.66%), penicillin (50%), oxacillin (83.3%), ceftazidime (66.6%) and vancomycin (8.3%). The essential oil of this plant had inhibitory effect against most isolates. More than 1/3 of the E. coli isolates showed the lowest MIC (10 ppm) whereas more than 1/3 of the K. pneumoniae isolates showed the highest (250 ppm) MIC values. In contrast, equal number of S. aureus isolates showed the low MIC values (10 and 50 ppm), while the heighest MIC (250 ppm) was seen in 1/3 of isolates and moderate MIC (100 ppm) was seen in one isolate only. Conclusions: The Saturejahortensis essential oil has a potent antimicrobial activity against multi-drug resistant bacteria. The present study confirms the usefullness of this essential oil as antibacterial agent but further research is required to evaluate the practical value of this plant before proving its therapeutic applications.

Incorporation of essential oil in alginate microparticles by multiple emulsion/ionic gelation process

In this study, an o/w/o multiple emulsion/ionic gelation method was developed for production of alginate microparticles loaded with Satureja hortensis essential oil (SEO). It was found that the essential oil concentration has significant influence on encapsulation efficiency (EE), loading capacity (LC) and size of microparticles. The values of EE, LC and particle mean diameter were about 52–66%, 20–26%, and 47–117μm, respectively, when the initial SEO content was 1–3% (v/v) .The essential oil-loaded microparticles were porous, as displayed by scanning electron micrograph. The presence of SEO in alginate microparticles was confirmed by Fourier transform-infrared (FT-IR) spectroscopy and differential scanning calorimetry (DSC) analyses. SEO-loaded microparticles showed good antioxidant (with DPPH radical scavenging activity of 40.7–73.5%) and antibacterial properties; this effect was greatly improved when the concentration of SEO was 3% (v/v). S. aureus was found to be the most sensitive bacterium to SEO and showed a highest inhibition zone of 304.37mm2 in the microparticles incorporated with 3% (v/v) SEO. In vitro release studies showed an initial burst release and followed by a slow release. In addition, the release of SEO from the microparticles followed Fickian diffusion with acceptable release.
