Colorimetric Determination of Atropine, Homatropine, Scopolamine, and Their Derivatives by the Ferric Hydroxamate Method

The purpose of this study was to develop a procedure for the quantitative analysis of the salts of the solanaceous alkaloids in the presence of their hydrolysis products. The salts taken were atropine sulfate, homatropine hydrobromide, homatropine methyl-bromide, scopolamine hydrobromide, and scopolamine methyl-bromide. The ferric hydroxamate procedure was used. The conditions necessary for a reproducible reaction were established; a procedure of sufficient sensitivity was developed which will allow unit dose analysis; and an analysis was developed which will permit the salts of the solanaceous alkaloids to be quantitatively analyzed in the presence of their degradation products.

Gas Chromatography of Barbiturates, Phenolic Alkaloids, and Xanthine Bases: Flash-Heater Methylation by Means of Trimethylanilinium Hydroxide

Injection of methanol solutions of trimethylanilinium salts of barbiturates, phenolic alkaloids, and dimethylxanthines produced thermal decomposition in the injection port to give methyl derivatives suitable for quantitative gas chromatography. Trimethylanilinium hydroxide appeared to be superior to tetramethylammonium hydroxide for instantaneous methylation of these compounds. Quantitative methylation was achieved under a variety of experimental conditions. The change in retention time resulting from methylation may be used for identification purposes. The 1,3-dimethyl structure of methylated barbiturates was confirmed by NMR spectroscopy.

A coulometric microdetermination of salts of certain organic bases and alkaloids is described, based on precipitation as the tetraphenylborates and the coulometric determination of the tetraphenylborate content of the precipitate with electrogenerated silver ion. Eleven compounds were determined with satisfactory accuracy.

Gas Chromatography of Alkaloids, Alkaloidal Salts, and Derivatives

A number of alkaloids and alkaloidal derivatives have been gas chromatographed on columns of silicone rubber SE-30. The method has also been applied to alkaloidal extracts of crude drugs. Phenolic alkaloids are readily gas chromatographed as the trimethylsilyl ethers. Alkaloidal salts have been gas chromatographed directly. The salt dissociates in the flash heater and the alkaloid is eluted as the base. Under certain conditions, many alkaloids decompose when subjected to gas chromatography. These decompositions often appear to be catalyzed by the glass wool used on top of the column packing.

A method is described for the assay of alkaloids or alkaloidal salts either in the pharmaceutical preparation or plant extract which is based on separating the alkaloids by ion exchange resin and determination of them in the nonaqueous solution, either by visual indicator method or potentiometry.

The behaviour of acetone solutions of free acids or their alkaloid salts on aluminium oxide columns can be explained by assuming that: (1) different ions occupy the surface of the exchanger to a different extent, (2) polybasic acids are bound to aluminium oxide with only one valency. Acid aluminium oxide columns that have been prepared with sulphuric acid can exchange alkali cations in acetone solution for hydroxonium ions.

The hemolytic method has been employed to determine the isotonic coefficient, i , for various alkaloidal salts. It was thought that these data would be of value in the preparation of solutions isotonic with blood, since methods based on physicochemical data are not always reliable. Direct determinations were made of the degree of hemolysis occurring in solutions of various alkaloidal salts as compared with the degree of hemolysis in solutions of sodium chloride. Experiments were conducted using rabbit and human erythrocytes.

Behavior of Some Inorganic Anions as Bases in Glacial Acetic Acid

Data are presented to show that all common inorganic anions, except bisulfate, act as bases in glacial acetic acid. Supporting evidence includes results obtained by titrating a number of alkaloidal salts and many inorganic salts. Theoretical basis for this behavior is discussed.

The idea is generally accepted by most pharmacologists that the physiological activity and toxicity of powerful alkaloids are practically the same as that of their salts, provided allowance is made for the slight increase in molecular weight of the salts due to the acid radicles, and for the difference in solubility between the alkaloids and their salts.
