BT is a primary source of at least 21 allergenic proteins, although most studies have been performed with recombinant proteins. Particularly recombinant Blo t 2 is not regarded so far a major allergen. The aim of this work was the purification and assessment of allergenic activity of native Blo t2 in allergic Cuban patients. Methods: Protein extract was obtained by sonication of the culture medium dissolved in PBS. Clarification was achieved by 60% ammonium sulfate precipitation of high MW proteins. Supernatant of the salting-out step was subjected to Sephadex 75 gel filtration chromatography. The resulting protein peak was identified as Blo t2 by SDS-PAGE and Mass Spectrometry. For allergenic characterization, patients reactive to BT extract were screened by skin prick test (SPT) from a cohort of patients with a history of asthma. Serum anti-Blo t2 IgE in BT reactive patients was assessed by Western Blotting. Results: A Blo t 2 protein was obtained with high purity and a yield of 0.9 g per 20g of culture medium. This protein migrated as two bands between 14 and 16 kDa in SDS-PAGE. Sera of 80% of patients reactive to BT allergen extract by SPT showed IgE binging to this band by Western Blotting. Conclusions: These results indicate that Blo t2 is a major allergen from Blomia tropicalis house dust mite, with a high allergenicity in Cuban asthmatic patients. The characterization of this protein will allow the development of diagnostic methods and vaccine candidates for allergen specific immunotherapy.
