Growth performance and ruminal fermentation characteristics of Thai-native beef cattle fed cassava pulp fermented with Lactobacillus casei TH14 and additives.

Raw cassava pulp is a low-nutritional agro-industry residue with a high moist content that is susceptible to rot. Biodegradation of cassava pulp by adding Lactobacillus spp. plus stimulants could boost bio-acidity to preserve cassava pulp as ruminant feed. This investigation assessed the effect of ensiled cassava pulp without or with Lactobacillus casei TH14 (LTH14), urea, and molasses on the intake, digestibility, rumen fermentation, and growth performance of native Thai beef cattle. Eight Thai-native beef bulls (170.3 ± 20.2kg of initial body weight) were randomly assigned to two treatment groups in a completely randomized design: cassava pulp ensiled without additives (CPFNO; n = 4) and cassava pulp ensiled with the combination of LTH14 (2.5g per ton of fresh material), urea (4% DM), and molasses (4% DM) (CPFWA; n = 4). The intake of the concentrate diet and ensiled cassava pulp did not differ (p > 0.05) between the CPFNO and CPFWA groups, including final body weight, average daily gain (ADG), and feed conversion ratio (p > 0.05). However, CPFWA affected higher rice straw intake compared to the CPFNO group (p < 0.05). The intake and digestibility of crude protein (CP) were greater for the CPFWA group at 43.86% and 30.87%, respectively, compared to the CPFNO group (p < 0.05). Ruminal ammonia-nitrogen (NH3-N)and blood urea-nitrogen (BUN) were increased when animals were fed with CPFWA (p < 0.05). The average total volatile fatty acids (VFA) concentration of CPFWA was greater than that of CPFNO by 27.49% (p < 0.05). In conclusion, CPFWA supplementation can enhance rice straw intake, CP intake and digestion, total ruminal VFA concentration, and BUN levels, without adversely affecting growth performance.

476 Induced hindgut acidosis in sheep affected gut permeability and ruminal fermentation

Abstract Leaky gut may be caused by acidotic conditions in the hindgut and lead to systemic inflammation and negative effects on the gastrointestinal tract. The objective was to determine the effects of induced hindgut acidosis in sheep on cecal pH, ruminal fermentation, and gut permeability. Ruminally and cecally cannulated ewes [n = 11; body weight (BW) = 49 ± 4 kg] were assigned to one of two treatments: control (CON; n = 5) or induced hindgut acidosis (HGA; n = 6). To induce hindgut acidosis, 3 g wheat starch/kg BW per 24 h was continuously infused via the cecal cannula for 4 d. Control ewes received a constant infusion of deionized water. The diet contained 40% grass hay pellets, 35% whole shelled corn, 15% dried distillers grains, and 10% alfalfa cubes. Chromium EDTA was dosed once daily via the cecal cannula as a marker of gut permeability. Rumen, cecal, and fecal samples were collected to determine pH and volatile fatty acids (VFA). Rumen fluid was collected on d 4 for an ex vivo fermentation. Flasks were incubated for 24 h to determine pH, VFA, ammonia and in vitro dry matter digestibility. Tissue samples from the ileum, cecum, and colon were mounted in Ussing chambers to measure transepithelial electrical resistance (TEER). There was a treatment × time effect (P = 0.05) for cecal pH with HGA ewes having lesser cecal pH after d 1. By d 4, cecal pH had declined to 5.07 for HGA ewes compared with CON ewes which remained above 6.40 throughout the experiment. A treatment × time interaction was also observed (P &amp;lt; 0.01) for fecal pH and followed the same trend as cecal pH. Rumen pH was not affected (P = 0.87) by the interaction of treatment and time but was affected (P &amp;lt; 0.01) by treatment as ewes on the HGA treatment had a lesser rumen pH than CON ewes. Control ewes had lesser ruminal VFA and ammonia concentrations than HGA ewes (P &amp;lt; 0.01). Urinary Cr recovery was not affected by the interaction of treatment and time, or treatment (P ≥ 0.13). In vitro dry matter disappearance was not affected by cecal infusion treatment (P = 0.60). There was no effect (P = 0.78) of treatment on ex vivo pH. There were no effects (P ≥ 0.11) of treatment, time, or their interaction on ex vivo ammonia or total VFA concentration. In cecal tissue, TEER tended (P = 0.09) to be greater in CON ewes than HGA ewes. In contrast, TEER was not different (P ≥ 0.83) in ileal or colonic tissues between treatments. Induced hindgut acidosis in sheep altered rumen fermentation and tended to increase ex vivo measures of cecal permeability, but did not affect ex vivo rumen fermentation.

Mealworm larvae and black soldier fly larvae as novel protein supplements for cattle consuming low-quality forage.

The global population is projected to increase, indicating that there will be greater demand for animal protein to meet the associated food needs. This demand will place additional pressure on livestock systems to increase output while also minimizing natural resource inputs. Insect protein has emerged as a potential alternative to conventional protein feeds, such as soybean meal. Mealworm larvae (MWL; Tenebrio molitor) have been studied in poultry and swine as an alternative protein source; however, there is no research currently evaluating MWL for cattle. Black soldier fly larvae (BSFL; Hermetia illucens) have also received attention for their potential use in livestock feed due to their scalability and nutritional value, but research in cattle is limited. The objective of this study was to evaluate the effects of whole-dried MWL and defatted BSFL as protein supplements for cattle consuming forage. Five ruminally cannulated steers were utilized in a 5 × 5 Latin square experiment to determine how MWL and BSFL supplementations affect forage utilization. Steers consuming ad libitum low-quality forage (76.5% neutral detergent fiber [NDF], 4.2% crude protein) were provided one of the five treatments each period: 1) control with no supplement (CON), 2) soybean meal (CONV), 3) BSFL, 4) MWL, or 5) 50/50 by-weight blend of BSFL and MWL (MIX). All treatments were provided at 100 mg N/kg BW and periods included 8 d for treatment adaptation, 5 d for intake and digestion, and 1 d for ruminal fermentation measures. Protein supplementation stimulated forage organic matter intake (FOMI; P ≤ 0.01) relative to CON (3.28 kg/d). There was a significant difference in FOMI (P ≤ 0.01) between BSFL (4.30 kg/d) and CONV (4.71 kg/d), but not between CONV and MWL (P = 0.06, 4.43 kg/d). Total digestible OM intake (TDOMI) was also stimulated by the provision of protein (P ≤ 0.01), from 1.94 kg/d for CON to an average of 3.24 kg/d across protein supplements. Organic matter digestibility (OMD) and NDF digestibility (NDFD) were not affected by treatment (P ≥ 0.37), for an average OMD of 66.5% and NDFD of 62.7%. There was also no treatment effect on ruminal volatile fatty acid (P = 0.96) or ammonia-N (P = 0.22) concentrations. These data indicate that MWL may stimulate forage utilization by beef cattle to a greater extent than BSFL, but both are viable protein supplements.

PSVI-4 Effects of ruminal acidosis on rumen fermentation of steers adapted to a high concentrate diet

Abstract The objective was to characterize the effects of ruminal acidosis on rumen fermentation of steers adapted to a high concentrate diet. Cattle entering the feedlot undergo a transition period where the amount of concentrates in the diet is gradually increased to mitigate the risks of ruminal acidosis. Despite a dietary adaptation, rumen acidosis still affects feedlot cattle later in the finishing phase. Ruminally cannulated steers [n = 11; body weight (BW) = 795 kg ± 54) were blocked into 2 groups based on initial BW. Before the start of the study, cattle had been consuming a finishing diet (60% dry rolled corn, 15% modified distiller’s grains, 15% corn silage, and 10% ground corn based supplement) for more than 150 d. Steers were randomly assigned to 1 of 2 treatments: control (CON) or acidosis (ACD). For the experiment, steers were fasted for 24 h and at the conclusion received the same finishing diet. Steers on ACD steers were also ruminally dosed 0.05% BW of wheat starch on h 0 and h 12 to induce acidosis. Rumen fluid was collected every 4 h for 48 h to analyze ruminal pH, ammonia, volatile fatty acids (VFA), and lactate. Fecal grab samples were collected every 8 h for 48 h to determine fecal pH. Data were analyzed with the MIXED procedure of SAS 9.4 with the effects of treatment, time, and their interaction. A treatment × hour (P &amp;lt; 0.01) effect was observed for ruminal pH with the ACD steers having a lower pH than CON from h 12 to 32. Fecal pH was not affected by treatment, time, or their interaction (P ≥ 0.12). Steers fed ACD had greater total VFA concentration than CON (P &amp;lt; 0.01). A treatment × hour interaction (P &amp;lt; 0.01) was observed for acetate molar proportion with CON having greater percentage acetate at h 24 through 32. Additionally, there was a treatment × hour interaction (P &amp;lt; 0.01) for propionate molar proportion with ACD steers having greater propionate at h 28 and 32, and then at h 40 and 44. Butyrate was not affected (P ≥ 0.58) by treatment × hour or treatment. Acetate to propionate ratio (A:P) was affected by treatment × hour (P &amp;lt; 0.01) with CON steers having greater A:P after h 28. Rumen ammonia and lactate concentrations did not differ (P ≥ 0.25) by treatment or the interaction with h. In steers adapted to a high concentrate diet, acidosis only altered ruminal fermentation in regards to ruminal pH and VFA concentrations.

Effects of low-quality forage and starter protein content in starter diet of young calves on growth performance, rumen fermentation, and urinary purine derivatives.

Feeding low-quality forage (LQF) has been evaluated in mature ruminants and results show that it has been improved nitrogen utilization efficiency. The present study evaluated the interaction effect of feeding wheat straw as LQF (0 and 7.5%, DM basis) and starter protein level (20 vs. 24%, DM basis) on growth performance, ruminal fermentation, and microbial protein synthesis in Holstein dairy calves raised under moderate heat stress condition. Forty-eight 3-day old dairy calves (averaging 40.6 kg) were assigned in four experimental treatments as follow; 1) no LQF with 20% CP (NLQF-20CP), 2) no LQF with 24% CP (NLQF-24CP), 3) 7.5% LQF with 20% CP (LQF-20CP) and 4) 7.5% LQF and 24% CP (LQF-24CP). The calves were weaned on d 53 of age but the experiment extended until d 73 of age. Feeding LQF increased starter intake, average daily gain (tendency), ruminal acetate concentration, and improved fecal score of calves. The average daily gains before and after weaning were positively influenced with greater starter protein content. Hence, weaning and final BWs were improved when calves received greater CP content. In addition, greater starter CP content increased total ruminal volatile fatty acid concentration. With respect to the interaction effect between LQF feeding and starter protein content, the lower nitrogen excretion through urine was obtained for LQF-20CP diet among experimental treatments. The results of the current study showed that feeding LQF improved ruminal fermentation pattern and improved growth performance through increased starter intake. In addition, greater starter protein content is advisable during pre-weaning period for calves raised under mild heat stress condition. In conclusion, based on the results found in the current study, it can be suggested that feeding LQF for calves under heat stress condition can improve nitrogen utilization when dietary protein content is low. This can be opportunity to formulate starter diets with greater nitrogen utilization efficiency which is critical for accelerated growth programs at early stages of growth for young calves while calves raised under hot season condition.

Dietary supplementation with live or autolyzed yeast: Effects on performance, nutrient digestibility, and ruminal fermentation in dairy cows

This study was conducted to evaluate the effects of live or autolyzed yeast supplementation on dairy cow performance and ruminal fermentation. Two experiments were conducted to evaluate performance, feed sorting, total-tract apparent digestibility of nutrients, purine derivatives excretion, N utilization, ruminal fermentation, and abundance of specific bacterial groups in the rumen. In Exp. 1, 39 Holstein cows (171 ± 40 d in milk and 32.6 ± 5.4 kg/d milk yield) were blocked according to parity, days in milk, and milk yield and randomly assigned to the following treatments: Control (CON); autolyzed yeast (AY) fed at 0.625 g/kg DM (Levabon, dsm-firmenich); or live yeast (LY) fed at 0.125 g/kg DM (Vistacell, AB Vista). Cows were submitted to a 2-wk adaptation period followed by a 9-wk trial. In Exp. 2, 8 ruminal cannulated Holstein cows (28.4 ± 4.0 kg/d milk yield and 216 ± 30 d in milk), of which 4 multiparous and 4 primiparous, were blocked according to parity and enrolled into a 4 × 4 Latin square experiment with 21 d-periods (the last 7 d for sampling). Cows within block were randomly assigned to treatments sequences: control (CON2); live yeast (LY) using the same product and dietary concentration as described in Exp. 1; or autolyzed yeast fed at 0.625 g/kg DM (AY) or 0.834 g/kg DM (AY2). In Exp. 1 and 2, nutrient intake and total-tract apparent digestibility were not affected by treatments. Sorting for long feed particles (>19 mm) tended to be greater in cows fed yeast supplements than CON in Exp.1. Efficiency of N conversion into milk N was increased when feeding yeast supplements in Exp. 1. 3.5% Fat-corrected milk yield tended to be greater in cows fed yeast supplements than CON. Feed efficiency was increased when yeast supplements were fed to cows in relation to CON in Exp. 1. In Exp. 2, yield of fat-corrected milk and fat were greater in cows fed yeast supplements compared with CON2. Uric acid concentration and output in urine were increased when feeding yeast supplements when compared with CON2. Neither ruminal pH nor total volatile fatty acids were influenced by treatments. The current study did not reveal treatment differences in ruminal abundance of Anaerovibrio lipolytica, genus Butyrivibrio, Fibrobacter succinogenes, Butyrivibrio proteoclasticus, or Streptococcus bovis. Yeast supplementation can increase feed efficiency without affecting nutrient intake and digestibility, ruminal volatile fatty acid concentration, or ruminal abundance of specific bacterial groups. Supplementing live or autolyzed yeast, regardless of the dose, resulted in similar performance.

Almond hulls and shells as an alternative fiber source in limit-fed growing beef cattle diets.

Almond hulls and shells are a by-product of almond production that can be incorporated as a feed ingredient in beef cattle diets. Three experiments were conducted to determine the effects of hammermill screen size on almond hull and shell bulk density and inclusion of ground or non-ground almond hulls and shells in limit-fed growing diets on growth performance, diet digestibility, and ruminal fermentation characteristics of beef cattle. In experiment 1, almond hulls and shells were ground with a laboratory-scale hammermill using no screen, a 11.1-mm screen, a 19.1-mm screen, or a 25.4-mm screen. Each screen-size treatment was ground at three separate time points (n= 3 replications/treatment). Grinding almond hulls and shells with no screen increased bulk density by 111% and minimized proportions of fine particles; therefore, almond hulls and shells ground using no screen were included as a treatment in the following experiments. In experiment 2, 364 steers (initial body weight [BW]: 257± 20.7kg) were blocked by truckload (n = 4), stratified by BW, and assigned to pen within block. Pens were randomly assigned to 1 of 4 experimental diets (n= 10 pens/treatment). The control diet (CON) contained (DM basis) 39.5% dry-rolled corn, 7.5% supplement, 40% wet-corn gluten feed, and 13% prairie hay. Non-ground (13AH) or ground (13GAH) almond hulls and shells replaced prairie hay and were fed at 13% of diet DM or non-ground almond hulls and shells were fed at 26% of diet DM and replaced 13% prairie hay and 13% dry-rolled corn (26AH). Diets were limit-fed at 2.2% of BW daily (DM basis) for 56 d. Overall average daily gains (ADG) were greater (P ≤ 0.05) for CON, 13AH, and 13GAH compared with 26AH. In addition, ADG from days 14 to 56 were greater (P= 0.03) for 13GAH and tended to be greater (P = 0.09) for 13AH compared with CON. Experiment 3 was a 4 × 4 replicated Latin square in which 8 ruminally cannulated heifers (initial BW = 378 ± 44.0kg) were fed diets from experiment 2. Apparent dry matter digestibility did not differ (P = 0.21) among treatments. Total ruminal volatile fatty acid concentrations were greater (P ≤ 0.03) for 13GAH and 13AH compared with 26AH and tended (P = 0.06) to be greater for 13GAH compared with CON. Overall, almond hulls and shells can be utilized as an alternative to prairie hay in limit-fed growing diets without negatively influencing rates of gain or diet digestibility.

Ruminal volatile fatty acid concentration and methane gas production in sheep (Ovis aries) fed two types of basal diets with of without Gliricidia sepium legume forage supplementation

The aim of this study was to clarify whether the type of forage diets, Napier grass (Pennisetum purpureum) vs. urea-treated rice straw, and supplementation with Gliricidia sepium affect rumen VFA production and methane emission as well as intake and digestibility in ruminants. The experiment was set up in a randomized complete block design (RCBD) composed of four treatments namely: Napier grass (T1), Napier + legume forage (T2), urea-treated rice straw (T3), and urea-treated rice straw + legume forage (T4). Each treatment was divided into four blocks (n=16) based on body weight and sex combination. The daily dry matter intake tended to increase from treatments supplemented with G. sepium (T2 and T4) compared to treatment-fed pure basal diet alone (T1 and T3). Intake relative to metabolic weight (BW 0.75) is high in T2 and T4. The molar concentration of volatile fatty acid was higher in T1, which was found comparable with T2 and T3, while T4 has the lowest. The molar proportion of acetate tended to increase in T1 and T3, while the propionate concentration was significantly high in T2 and T4 leading to lesser CH4/CO2 production respectively. These results confirmed the potential of G. sepium as a methane-mitigating supplement to basal diets such as Napier and urea-treated rice straw as feed to ruminants.

Influence of the concentration and nature of total dissolved solids in brackish groundwater on water intake, nutrient utilization, energy metabolism, ruminal fermentation, and blood constituents in different breeds of mature goats and sheep

Brackish water is being progressively utilized in livestock farming across the globe where freshwater is scarce, potentially leading to detrimental consequences for animal health and performance. This study was conducted to determine effects of different brackish groundwater types on water and feed intake, nutrient utilization, ruminal fluid characteristics, and blood constituent concentrations in different breeds of goats and sheep in a 6 (animal types; AT) × 6 (water treatments; WT) Latin square design. Six AT (Boer, Spanish, and Tennessee Stiff-Leg does and Dorper, Katahdin, and St. Croix ewes) consumed drinking water differing in total dissolved solids (TDS) and mineral composition. Six WT were fresh water (FRW; 287 mg TDS/kg), a natural brackish water (BR) alone (100-BR; 5734 mg TDS/kg), BR with concentrations of all minerals increased by about 100 % (200-BR; 10,603 mg TDS/kg), FRW added with NaCl having similar TDS level (6654 mg/kg) as 100-BR (100-SL), FRW added with NaCl having similar TDS (12,197 mg/kg) as 200-BR (200-SL), and water with similar TDS (10,643 mg/kg) level as 200-BR by adding NaCl to 100-BR (200-BR/SL). Water treatment only affected (P < 0.05) water intake, total volatile fatty acids in ruminal fluid, blood phosphorus, blood aldosterone, and plasma osmolality. Water intake was greater (P < 0.05), but total ruminal volatile fatty acid concentrations were lower (P < 0.05) for 200-SL than other WT. Plasma osmolality and aldosterone concentrations were lower (P < 0.05) for 200-SL and 200-BR/SL than FRW. There were no significant interactions between WT and AT for any variable. Water and feed intake, digestibilities, ruminal ammonia concentration, and methane emission were similar among AT (P > 0.05). There were differences (P < 0.05) among AT for many ruminal fermentation and blood characteristics. Although there were some differences among WT, no AT seemed adversely affected by water with a TDS level near or slightly above 10,000 mg/kg regardless of mineral sources.

Isoacids supplementation improves growth performance and feed fiber digestibility associated with ruminal bacterial community in yaks.

This study was conducted to assess the effect of mixed isoacid (MI) supplementation on fermentation characteristics, nutrient apparent digestibility, growth performance, and rumen bacterial community in yaks. A 72-h in vitro fermentation experiment was performed on an ANKOM RF gas production system. MI was added to five treatments at doses of 0, 0.1%, 0.2%, 0.3%, 0.4%, and 0.5% on the dry matter (DM) basis of substrates using a total of 26 bottles (4 bottles per treatment and 2 bottles as the blank). Cumulative gas production was measured at 4, 8, 16, 24, 36, 48, and 72 h. Fermentation characteristics including pH, the concentration of volatile fatty acids (VFAs), ammonia nitrogen (NH3-N), microbial proteins (MCP), and the disappearance rate of dry matter (DMD), neutral detergent fiber (NDFD), and acid detergent fiber (ADFD) were measured after a 72-h in vitro fermentation to determine an optimal MI dose. Fourteen Maiwa male yaks (180-220 kg, 3-4 years old of age) were randomly assigned to the control group (without MI, n = 7) and the supplemented MI group (n = 7, supplemented with 0.3% MI on DM basis) for the 85-d animal experiment. Growth performance, nutrient apparent digestibility, rumen fermentation parameters, and rumen bacterial diversity were measured. Supplementation with 0.3% MI achieved the greatest propionate and butyrate content, NDFD and ADFD compared with other groups (P < 0.05). Therefore, 0.3% was used for the animal experiment. Supplementation with 0.3% MI significantly increased the apparent digestibility of NDF and ADF (P < 0.05), and the average daily weight gain of yaks (P < 0.05) without affecting the ruminal concentration of NH3-N, MCP, and VFAs. 0.3% MI induced rumen bacteria to form significantly different communities when compared to the control group (P < 0.05). g__norank_f__Bacteroidales_BS11_gut_group, g__norank_f__Muribaculaceae, g__Veillonellaceae_UCG-001, g__Ruminococcus_gauvreauii_group, g__norank_f__norank_o__RF39 and g__Flexilinea were identified as the biomarker taxa in responding to supplementation with 0.3% MI. Meanwhile, the abundance of g__Flexilinea and g__norank_f__norank_o__RF39 were significantly positively correlated with the NDF digestibility (P < 0.05). In conclusion, supplementation with 0.3% MI improved the in vitro rumen fermentation characteristics, feed fiber digestibility, and growth performance in yaks, which was associated with changes of the abundance of g__Flexilinea and g__norank_f__norank_o__RF39.

Inflammatory genes expression and calf responses to different dietary grains (steam flaked corn vs steam-rolled barley) and protein sources (pelleted soybean meal vs pelleted canola meal)

This study aimed to investigate the effects of grains, and protein sources in the starter diet on daily gain, pro- and anti-inflammatory genes expression, ruminal volatile fatty acid (VFA) concentration, and blood metabolites in the dairy calves. Forty Holstein calves were randomly assigned to treatments in a 2 × 2 factorial arrangement with the factors of grain source (steam-flaked corn, CG vs. steam-rolled barley, BG) and pelleted protein source (soybean meal, SBM vs. canola meal, CM). The daily gain of calves who fed with SBM was higher than those fed with CM. Total VFA concentration in the rumen of calves fed with CG was greater than BG at day 35 (p < 0.05), and calves receiving SBM had higher total VFA concentration than calves fed with CM. The relative expression of tumor necrosis factor-α and interleukin-1β genes were significantly decreased in the calves fed with BG and CM compared to calves fed with CG and SBM. Calves receiving the starter diet based on BG had the highest (p < 0.05) gene expression of interferon-γ. Feeding calves with SBM is recommended because it resulted in a greater daily gain than CM. Complete replacement of SBM with CM, and CG with BG is not recommended in dairy calves.

Effects of hempseed cake on ruminal fermentation parameters, nutrient digestibility, nutrient flow, and nitrogen balance in finishing steers.

Five ruminally and duodenally cannulated red angus steers (n = 5; initial body weight [BW] = 542 kg, SD = 40 kg) were used in a three-period Youden square design consisting of three 21-d periods, three treatments, and five steers (one or two steers per treatment within each period) to evaluate the effect of feeding hempseed cake on ruminal fermentation parameters, organic matter (OM) intake, total tract nutrient digestion, and nitrogen (N) balance in steers fed finishing diets. The control (CON) diet contained 75% dry-rolled corn, 20% corn silage, and 5% supplement (DM basis). The dried corn distillers grains plus solubles (DDGS) and hempseed cake (HEMP) diets contained 55% dry-rolled corn, 20% corn silage, 20% dried corn distillers grains plus solubles or hempseed cake, and 5% supplement (DM basis). Total ruminal volatile fatty acid concentration was greater (P < 0.01) in steers fed the HEMP diet than in steers fed the DDGS or CON diets. Ruminal fluid pH was not influenced (P = 0.93) by treatment. Organic matter intake tended (P = 0.07) to be greater and OM total tract digestibility was less (P = 0.03) in steers fed the HEMP diet compared with steers fed the DDGS or CON diets. Ruminal true and total tract apparent N digestibility was greater (P < 0.01) in steers fed the HEMP diet than steers fed the DDGS or CON diets. Duodenal flow of essential, nonessential, and total amino acids was not influenced (P ≥ 0.37) by dietary treatment, but the lack of response was likely because ruminally degradable protein (RDP) supply exceeded the RDP requirement. Steers fed the HEMP diet had greater (P < 0.01) N retention (g/d) than steers fed the DDGS diet, which was greater (P < 0.01) than steers fed the CON diet, suggesting that feeding hempseed cake improved utilization of N. Although inclusion of hempseed cake decreased total tract OM digestibility compared with dried corn distillers grains or corn, improvements in N utilization suggest that hempseed cake could be a useful alternative feed ingredient for finishing cattle diets.

Effects of starch sources varying in particle sizes on ruminal fermentation, nutrient flow, starch digestibility, and lactation performance of dairy cows.

Cereal grains are the predominant starch source (SS) for dairy cows; however, starch digestibility varies greatly depending on source, grain processing, and potentially interactions between these factors. The objective was to study the effects of the interactions between SS, and particle sizes (PS) on ruminal fermentation, nutrient flow, starch digestibility, and lactation performance of dairy cows. Four ruminally cannulated multiparous Holstein cows were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments. Two SS (corn or sorghum) used in this study were either finely or coarsely ground (using a 1- or 4-mm screen sieve). Digesta flow was quantified using the reticular sampling technique, applying the triple-marker method. Data were analyzed using the GLIMMIX procedure of SAS version 9.3 (SAS Institute Inc., Cary, NC, USA). For ruminal pH, data were analysed with time as repeated measure. There were no interactions between SS and PS on production or intake, flow, and digestibility of nutrients. Dry matter intake was greater for the corn diet compared to the sorghum diet (25.15 vs. 21.98 kg/d), which consequently affected nutrient intake, however, PS did not affect intake. Milk yield was not affected by SS; however, it was greater for cows fed fine grains than cows fed coarser grains (25.32 vs. 23.16 kg/d). Milk fat and milk protein were not affected by SS or PS. Interactions (SS × PS) were observed for ruminal pH, reticular pH, and volatile fatty acids (VFA) concentrations but not for ruminal NH3-N concentration. Ruminal and reticular pH were greater for sorghum when coarsely ground and the total VFA concentration was decreased, compared to coarse corn and fine sorghum; however, coarsely grinding corn did not affect ruminal or reticular pH nor VFA concentration. Acetate concentration was lower for corn when finely ground; however, finely grinding sorghum did not affect acetate. Decreasing PS increased ruminal digestibility of starch (87.18% vs. 83.43%), reduced the flow of starch to the reticulum (0.79 vs. 0.96 kg/d) but decreased neutral detergent fiber digestibility in the rumen (30.23% vs. 34.88%). Although SS were differently affected by processing, the effects of PS on production, intake, flow, and digestibility of nutrients were observed regardless of the SS. Furthermore, the effects of decreasing PS on pH and VFA concentrations were more pronounced in sorghum compared to corn.

Effect of substituting steam-flaked corn for course ground corn on in vitro digestibility, average daily gain, serum metabolites and ruminal volatile fatty acids, and bacteria diversity in growing yaks

Yaks are well adapted to the Qinqhai-Tibetan Plateau and, traditionally, only grazed natural pasture all year without supplements. Today, however, yaks are also raised in feedlots; consequently, more information is needed in choosing optimal feeding regimes. This study examined the effects of substituting steam-flaked corn (SFC) for coarse ground corn (CGC) on average daily gain (ADG), in vitro true digestibility (IVTD), concentrations of ruminal volatile fatty acids (VFAs) and serum metabolites, and rumen bacterial diversity of growing yaks. Twenty-one, 2-year-old male yaks (138 ± 37.3 kg) were divided randomly into 3 groups (n = 7 per group) and received a total mixed ration (TMR) for 143 days. The TMR contained 446 g corn per kg dry matter (DM), but the ratio of CGC: SFC differed among the three groups as: 1) 100:0 (SFC0), 2) 50:50 (SFC50), and 3) 0:100 (SFC100). To maintain conditions similar to the feedlots, the yaks were fed as a group and feed intake was recorded daily. The yaks were weighed on 6 occasions during the study and ADG was calculated for each yak from the regression equation of body weight (BW) on days of treatment. On the morning of d 144 before feed was offered, jugular vein blood was collected to measure the concentrations of serum metabolites, and rumen contents were collected to determine ruminal VFAs, fermentation parameters and bacterial diversity. The daily dry matter intake (DMI) averaged between 6.08 and 6.15 kg per yak for the three dietary treatments and the IVTD increased with an increase in SFC (P < 0.05). The ADG was higher (P < 0.05) by 13.2% (812 vs 717 g/d) in the SFC100 group than in the SFC0 group while the ratio of the average daily DMI per yak to the average ADG per yak was lowest numerically for the SFC100 group (7.51), followed by the SFC50 group (8.46) and then the SFC0 group (8.66). The serum concentrations of alkaline phosphatase (ALP), serum urea-N, and triglycerides (TG) decreased (P < 0.05) but of aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and total cholesterol (TC) increased (P < 0.05) with an increasing proportion of dietary SFC. The greater serum concentration of LDH and lesser concentration of serum urea-N suggests that the SFC50 and SFC100 groups were proliferating cells at a faster rate and depositing more protein in muscle tissue than the SFC0 group, as the ADG increased with an increase in the level of dietary SFC. The ruminal concentrations of VFAs increased and of NH3-N decreased with an increase in the proportion of SFC. The rumen bacterial diversity did not differ among dietary groups; Bacteroidetes and Firmicutes were the dominant phyla and Prevotella and Succiniclasticum were the dominant genera in all dietary treatments. These results demonstrated that increasing the proportion of SFC could improve IVTD, ruminal concentration of VFAs, ADG and the group DMI to ADG ratio.

Effects of rumen-protected methionine supplementation on production performance, apparent digestibility, blood parameters, and ruminal fermentation of lactating Holstein dairy cows.

This study aimed to evaluate the effects of reducing dietary CP and supplementing rumen protected-methionine (RPM) on production performance, blood parameters, digestibility of nutrients or ruminal fermentation in lactating Holstein dairy cows. A total of 96 lactating cows were randomly assigned to 1 of 2 treatments: a diet containing 17.3% CP without RPM (control group; CON; n = 49) or a diet containing 16.4% CP and supplemented with 15.0 g/d of RPM (treatment group; RPM; n = 47). No effect was observed in the RPM group on milk yield, milk composition and digestibility of nutrients. The results of blood parameters showed that cows in the RPM group exhibited lower blood urea nitrogen concentration than in CON group. Rumen microbial crude protein (MCP) was higher in the RPM group compared to the CON group. Ruminal volatile fatty acid (VFA) concentrations were not different between treatments except for butyrate and isovalerate, which were higher in the RPM group than the CON group 2 h after feeding. In conclusion, reducing dietary CP with RPM supplementation did not limit milk yield, milk composition or digestibility of nutrients, but could improve nitrogen utilization, synthesis of MCP and partially increase VFA production 2 h after feeding cows.

Using probiotics to improve the utilization of chopped dried date palm leaves as a feed in diets of growing Farafra lambs.

The study determined the ability of three probiotics to improve the nutritional value of date palm leaves in diets of growing lambs. Twenty male Farafra lambs (26 ± 0.33 kg) were randomly allocated to one of four treatments (n = 6) and fed: a control or basal diet (C; 70% concentrate + 30% date palm leaves without additives) and supplemented with Bacillofort containing 2 × 1011 CFU of Bacillus subtilis/g (BAC treatment), Lacotpro containing 1 × 1012 CFU of Lactobacillus acidophilus/g (LAC treatment) or ZAD containing 6 × 108 CFU of R. albus/g (ZAD treatment) at 4 g of all additives for 150 days. As a result of this study, LAC improved (P < 0.05) growth performance and feed efficiency compared to control. Additives increased (P = 0.001) concentrations of albumin, triiodothyronine, and thyroxine, hemoglobin concentration and red blood cells and decreased (P = 0.001) globulin and urea-N. Additives increased hot carcass (P = 0.040) while BAC increased Longissimus dorsi, meat and fat without affecting water holding capacity compared to other treatments. In the metabolism experiment, BAC increased the digestibility of crude protein, while BAC and ZAD increased the digestibility of dry matter, organic matter, and neutral detergent fiber. Additives did not affect nitrogen (N) intake and urinary N; however, decreased fecal N and increased N balance compared to the control. BAC and ZAD increased ruminal volatile fatty acids concentration compared to the control. Based on our results, Lacotpro could be used to improve growth performance and feed efficiency, while Bacillofort could be used to improve meat quality of in lambs.

Effect of 2-hydroxy-4-(methylthio)butanoate (HMTBa) on milk fat, rumen environment and biohydrogenation, and rumen protozoa in lactating cows fed diets with increased risk for milk fat depression.

Biohydrogenation-induced milk fat depression (MFD) is a reduction in milk fat synthesis caused by bioactive fatty acids (FA) produced during altered ruminal microbial metabolism of unsaturated FA. The methionine analog 2-hydroxy-4-(methylthio)butanoate (HMTBa) has been shown to reduce the shift to the alternate biohydrogenation pathway and maintain higher milk fat yield in high-producing cows fed diets lower in fiber and higher in unsaturated FA. The objective of this experiment was to verify the effect of HMTBa on biohydrogenation-induced MFD and investigate associated changes in rumen environment and fermentation. Twenty-two rumen cannulated high-producing Holstein cows [168 ± 66 d in milk; 42 ± 7 kg of milk/d (mean ± standard deviation)] were used in a randomized design performed in 2 blocks (1 = 14 cows, 2 = 8 cows). Treatments were control (corn carrier) and HMTBa (0.1% of diet dry matter). The experiment included a 7-d covariate period followed by 3 phases that fed diets with increasing risk of MFD. The diet during the covariate and low-risk phase (7 d) was 32% neutral detergent fiber with no additional oil. The diet during the moderate-risk phase (17 d) was 29% neutral detergent fiber with 0.75% soybean oil. Soybean oil was increased to 1.5% for the last 4 d. The statistical model included the random effect of block and time course data were analyzed with repeated measures including the random effect of cow and tested the interaction of treatment and time. There was no effect of block or interaction of block and treatment or time. There was no overall effect of treatment or treatment by time interaction for dry matter intake, milk yield, and milk protein concentration and yield. Overall, HMTBa increased milk fat percent (3.2 vs. 3.6%) and yield (1,342 vs. 1,543 g/d) and there was no interaction of treatment and dietary phase. Additionally, HMTBa decreased the concentration of trans-10 18:1 in milk fat and rumen digesta. Average total ruminal concentration of volatile FA across the day and total-tract dry matter and fiber digestibility were not affected by HMTBa, but HMTBa increased average rumen butyrate and decreased propionate concentration and increased total protozoa abundance. Additionally, HMTBa increased the fractional rate of α-linoleic acid clearance from the rumen following a bolus predominantly driven by a difference in the first 30 min. Plasma insulin was decreased by HMTBa. In conclusion, HMTBa prevented the increase in trans FA in milk fat associated with MFD through a mechanism that is independent of total volatile FA concentration, but involves modification of rumen biohydrogenation. Decreased propionate and increased butyrate and ruminal protozoa may also have functional roles in the mechanism.

Different feeding strategies can affect growth performance and rumen functions in Gangba sheep as revealed by integrated transcriptome and microbiome analyses.

Due to the harsh environment in the Tibetan Plateau, traditional grazing greatly limits the growth potential of local animals and causes severe ecosystem degradation. This is an urgent issue to be solved, which requires alternative strategies for grazing animals in the Tibetan alpine pastoral livestock systems. This study aimed to investigate the effects of different feeding strategies on growth performance and ruminal microbiota-host interactions in the local breed of sheep (Gangba sheep). Thirty 9-month old Gangba sheep (n = 10 per group) were assigned to natural grazing (G), semi-grazing with supplementation (T), and barn feeding (F) groups (supplementation of concentrate and oat hay) based on body weight. At the end of the experiment (75 d), all sheep were weighed, rumen fluid was obtained from six sheep per group, and ruminal epithelium was obtained from 3 sheep per group. The results showed that: (1) Compared with the G and T groups, the F group significantly increased dry matter intake, average daily gain, and feed conversion ratio of animals. Additionally, Gangba sheep in the F group had higher concentrations of ruminal short-chain volatile fatty acids (VFAs), especially propionate and butyrate (P <0.05) than sheep in the G and T groups. (2) The principal coordinates analysis indicated a significant difference in bacterial composition among different feed strategies. More specifically, the relative abundance of propionate (unidentified F082 and Succiniclasticum) and butyrate-producing (Eubacterium_coprostanoligenes_group) genera were also observed to be increased in the F group, in which unidentified F082 was identified as a differential biomarker among the three groups according to linear discriminant analysis effect size analysis. (3) The dynamics of the rumen epithelial transcriptome revealed that ECM-receptor interactions, focal adhesion, and PI3K-Akt signaling pathways, which are critical in mediating many aspects of cellular functions such as cell proliferation and motility, were upregulated in the F group. In conclusion, under harsh conditions in the Tibetan alpine meadow, barn feeding increased ruminal VFAs concentrations (especially propionate and butyrate), which stimulated gene expression related to cell proliferation in rumen epithelium, appearing to be superior to natural grazing and semi-grazing in gaining body weight of the local Gangba sheep.

Application of 3-nitrooxypropanol and canola oil to mitigate enteric methane emissions of beef cattle results in distinctly different effects on the rumen microbial community

BackgroundThe major greenhouse gas from ruminants is enteric methane (CH4) which in 2010, was estimated at 2.1 Gt of CO2 equivalent, accounting for 4.3% of global anthropogenic greenhouse gas emissions. There are extensive efforts being made around the world to develop CH4 mitigating inhibitors that specifically target rumen methanogens with the ultimate goal of reducing the environmental footprint of ruminant livestock production. This study examined the individual and combined effects of supplementing a high-forage diet (90% barley silage) fed to beef cattle with the investigational CH4 inhibitor 3-nitrooxypropanol (3-NOP) and canola oil (OIL) on the rumen microbial community in relation to enteric CH4 emissions and ruminal fermentation.Results3-NOP and OIL individually reduced enteric CH4 yield (g/kg dry matter intake) by 28.2% and 24.0%, respectively, and the effects were additive when used in combination (51.3% reduction). 3-NOP increased H2 emissions 37-fold, while co-administering 3-NOP and OIL increased H2 in the rumen 20-fold relative to the control diet. The inclusion of 3-NOP or OIL significantly reduced the diversity of the rumen microbiome. 3-NOP resulted in targeted changes in the microbiome decreasing the relative abundance of Methanobrevibacter and increasing the relative abundance of Bacteroidetes. The inclusion of OIL resulted in substantial changes to the microbial community that were associated with changes in ruminal volatile fatty acid concentration and gas production. OIL significantly reduced the abundance of protozoa and fiber-degrading microbes in the rumen but it did not selectively alter the abundance of rumen methanogens.ConclusionsOur data provide a mechanistic understanding of CH4 inhibition by 3-NOP and OIL when offered alone and in combination to cattle fed a high forage diet. 3-NOP specifically targeted rumen methanogens and partly inhibited the hydrogenotrophic methanogenesis pathway, which increased H2 emissions and propionate molar proportion in rumen fluid. In contrast, OIL caused substantial changes in the rumen microbial community by indiscriminately altering the abundance of a range of rumen microbes, reducing the abundance of fibrolytic bacteria and protozoa, resulting in altered rumen fermentation. Importantly, our data suggest that co-administering CH4 inhibitors with distinct mechanisms of action can both enhance CH4 inhibition and provide alternative sinks to prevent excessive accumulation of ruminal H2.

Trace mineral source influences digestion, ruminal fermentation, and ruminal copper, zinc, and manganese distribution in steers fed a diet suitable for lactating dairy cows

High solubility of certain trace minerals (TM) in the rumen can alter nutrient digestibility and fermentation. The objectives of the present studies were to determine the effects of TM source on 1) nutrient digestibility and ruminal fermentation, 2) concentrations of soluble Cu, Zn, and Mn in the rumen following a pulse dose of TM, and 3) Cu, Zn, and Mn binding strength on ruminal digesta using dialysis against a chelating agent in steers fed a diet formulated to meet the requirements of a high producing dairy cow. Twelve Angus steers fitted with ruminal cannulae were adapted to a diet balanced with nutrient concentrations similar to a diet for a high producing lactating dairy cow for 21 d. Steers were then randomly assigned to dietary treatments consisting of 10 mg Cu, 40 mg Mn, and 60 mg Zn/kg DM from either sulfate (STM), hydroxychloride (HTM) or complexed trace minerals (CTM). The experimental design did not include a negative control (no supplemental Cu, Mn, or Zn) because the basal diet did not meet the National Research Council requirement for Cu and Zn. Copper, Mn, and Zn are also generally supplemented to lactating dairy cow diets at concentrations approximating those supplied in the present study. Following a 14-d adaptation period, total fecal output was collected for 5-d. Following the fecal collection period, rumen fluid was collected for Volatile fatty acid (VFA) parameters. On the following day, the same diet was provided for 14 d, without supplemental Cu, Zn, and Mn. This period served as a wash-out period. A pulse dose of 100, 400, and 600 mg of Cu, Zn, Mn, respectively, from either STM, HTM, or CTM, was administered via ruminal cannulae to the steers on day 15. Over a 24-h period ruminal samples were obtained every 2-h. Following centrifugation, the supernatant was analyzed for Cu, Mn, and Zn. Ruminal solid digesta samples from times 0, 12, and 24 h after bolus dosing were exposed to dialysis against Tris-EDTA. Digestibility of NDF and ADF were lesser in STM vs. HTM and vs. CTM supplemented steers. Steers receiving HTM and CTM had greater total VFA concentrations than STM, and molar proportions of individual VFA were not affected by treatment. Ruminal soluble Cu and Zn concentrations were greater post dosing in STM and CTM supplemented steers at 2, 4, and 6 h for Cu and 4, 6, 8, 10 and 12 h for Zn when compared to HTM supplemented steers. The release of Cu and Zn from ruminal solid digesta following dialysis against Tris-EDTA at 12 and 24 h postdosing was greater for steers receiving HTM compared to those receiving STM or CTM. Results indicate trace mineral source impacts: 1) how tightly bound Cu and Zn are to ruminal solid digesta; 2) fiber digestion; 3) and ruminal total VFA concentrations.
