α-Tocopherol Long-Chain Metabolite α-T-13'-COOH Exhibits Biphasic Effects on Cell Viability, Induces ROS-Dependent DNA Damage, and Modulates Redox Status in Murine RAW264.7 Macrophages.

The α-tocopherol long-chain metabolite α-tocopherol-13'-hydroxy-chromanol (α-T-13'-COOH) is a proposed regulatory intermediate of endogenous vitamin E metabolism. Effects of α-T-13'-COOH on cell viability and adaptive stress response are not well understood. The present study aims to investigate the concentration-dependent effects of α-T-13'-COOH on cellular redox homeostasis, genotoxicity, and cytotoxicity in murine RAW264.7 macrophages as a model system. Murine RAW264.7 macrophages are exposed to various dosages of α-T-13'-COOH to determine its regulatory effects on reactive oxygen species (ROS) production, DNA damage, expression of stress-related markers, and the activity of ROS scavenging enzymes including superoxide dismutases, catalase, and glutathione-S-transferases. The impact on cell viability is assessed by analyzing cell proliferation, cell cycle arrest, and cell apoptosis. α-T-13'-COOH influences ROS production and induces DNA damage in a dose-dependent manner. The metabolite modulates the activity of ROS-scavenging enzymes, with significant changes observed in the activities of antioxidant enzymes. A biphasic response affecting cell viability is noted: sub-micromolar doses of α-T-13'-COOH promote cell proliferation and enhance DNA synthesis, whereas supraphysiological doses lead to DNA damage and cytotoxicity. It hypothesizes an adaptive stress response, characterized by upregulation of ROS detoxification mechanisms, enhanced cell cycle arrest, and increased apoptosis, indicating a correlation with oxidative stress and subsequent cellular damage.

AtZAT10/STZ1 improves drought tolerance and increases fiber yield in cotton.

Drought poses a significant challenge to global crop productivity, necessitating innovative approaches to bolster plant resilience. Leveraging transgenic technology to bolster drought tolerance in crops emerges as a promising strategy for addressing the demands of a rapidly growing global populace. AtZAT10/STZ1, a C2H2-type zinc finger protein transcription factor has shown to significantly improve Arabidopsis' tolerance to various abiotic stresses. In this study, we reports that AtSTZ1 confers notable drought resistance in upland cotton (Gossypium hirsutum), amplifying cotton fiber yield under varying conditions, including irrigated and water-limited environments, in field trials. Notably, AtSTZ1-overexpressing transgenic cotton showcases enhanced drought resilience across critical growth stages, including seed germination, seedling establishment, and reproductive phases. Morphological analysis reveals an expanded root system characterized by an elongated taproot system, increased lateral roots, augmented root biomass, and enlarged cell dimensions from transgenic cotton plants. Additionally, higher contents of proline, chlorophyll, soluble sugars, and enhanced ROS-scavenging enzyme activities are observed in leaves of transgenic plants subjected to drought, underscoring improved physiological adaptations. Furthermore, transgenic lines exhibit heightened photosynthetic rate, increased water use efficiency, and larger stomatal and epidermal cell sizes, coupled with a decline in leaf stomatal conductance and density, as well as diminished transpiration rates compared to the wild type counterparts. Transcriptome profiling unveils 106 differentially expressed genes in transgenic cotton leaves post-drought treatment, including protein kinases, transcription factors, aquaporins, and heat shock proteins, indicative of an orchestrated stress response. Collectively, these findings underscore the capacity of AtSTZ1 to augment the expression of abiotic stress-related genes in cotton following drought conditions, thus presenting a compelling candidate for genetic manipulation aimed at enhancing crop resilience.

A comprehensive assessment of photosynthetic acclimation to shade in C4 grass (Cynodon dactylon (L.) Pers.)

BackgroundLight deficit in shaded environment critically impacts the growth and development of turf plants. Despite this fact, past research has predominantly concentrated on shade avoidance rather than shade tolerance. To address this, our study examined the photosynthetic adjustments of Bermudagrass when exposed to varying intensities of shade to gain an integrative understanding of the shade response of C4 turfgrass.ResultsWe observed alterations in photosynthetic pigment-proteins, electron transport and its associated carbon and nitrogen assimilation, along with ROS-scavenging enzyme activity in shaded conditions. Mild shade enriched Chl b and LHC transcripts, while severe shade promoted Chl a, carotenoids and photosynthetic electron transfer beyond QA− (ET0/RC, φE0, Ψ0). The study also highlighted differential effects of shade on leaf and root components. For example, Soluble sugar content varied between leaves and roots as shade diminished SPS, SUT1 but upregulated BAM. Furthermore, we observed that shading decreased the transcriptional level of genes involving in nitrogen assimilation (e.g. NR) and SOD, POD, CAT enzyme activities in leaves, even though it increased in roots.ConclusionsAs shade intensity increased, considerable changes were noted in light energy conversion and photosynthetic metabolism processes along the electron transport chain axis. Our study thus provides valuable theoretical groundwork for understanding how C4 grass acclimates to shade tolerance.

Genome-wide identification and role of HSFs in antioxidant response of hot water treated zucchini fruit during cold storage

Zucchini squashes are cold-sensitive and vulnerable to chilling injury (CI) resulting from reactive oxygen species (ROS) and hot water (HW) immersing effectively reduce CI symptoms during cold storage. However, mechanism involved in reduced ROS due to HW treatment has not been characterized well. In this study, tender green zucchini fruit were treated with HW for 15 min at 45 ± 1 °C and stored for 15 d at 4 ± 1 °C and above 90 % relative humidity. Results showed substantial reduction in CI index, electrolyte leakage, malonaldehyde (MDA) contents and ROS accumulation along with increased activity of ROS-scavenging enzymes due to HW treatment. To gain insight into the molecular mechanism involved in antioxidant defense system, transcriptomic analysis revealed that heat shock factors (HSF) accumulated due to HW treatment regulated the ROS pathway during cold stress. CpHSFA4a was one of the highly expressed transcription factors (TF) due to HW treatment that regulated the transcription of ROS enzymes related genes. CpHSFA4a bind actively with heat shock element (HSE) in promoter regions of CpSOD, CpCAT, CpAPX1, CpAPX2, and CpAPX3, activated and increased the expression of these genes. In conclusion, HW treatment alleviated the CI by maintaining ROS homeostasis through CpHSFA4a mediated ROS pathway in zucchini squashes during cold storage.

Impact of Probiotic Lactiplantibacillus plantarum ATCC 14917 on atherosclerotic plaque and its mechanism.

Atherosclerosis is viewed as not just as a problem of lipid build-up in blood vessels, but also as a chronic inflammatory disease involving both innate and acquired immunity. In atherosclerosis, the inflammation of the arterial walls is the key characteristic that significantly contributes to both the instability of plaque and the occlusion of arteries by blood clots. These events ultimately lead to stroke and acute coronary syndrome. Probiotics are living microorganisms that, when consumed in the right quantities, offer advantages for one's health. The primary objective of this study was to investigate the influence of Lactiplantibacillus plantarum ATCC 14917 (ATCC 14917) on the development of atherosclerotic plaques and its underlying mechanism in Apo lipoprotein E-knockout (Apoe-/-mice). In this study, Apoe-/-mice at approximately 8weeks of age were randomly assigned to three groups: a Normal group that received a normal chow diet, a high fat diet group that received a gavage of PBS, and a Lactiplantibacillus plantarum ATCC 14917 group that received a high fat diet and a gavage of 0.2ml ATCC 14917 (2 × 109CFU/mL) per day for a duration of 12weeks. Our strain effectively reduced the size of plaques in Apoe-/-mice by regulating the expression of inflammatory markers, immune cell markers, chemokines/chemokine receptors, and tight junction proteins (TJPs). Specifically, it decreased the levels of inflammatory markers (ICAM-1, CD-60 MCP-1, F4/80, ICAM-1, and VCAM-1) in the thoracic aorta, (Ccr7, cd11c, cd4, cd80, IL-1β, TNF-α) in the colon, and increased the activity of ROS-scavenging enzymes (SOD-1 and SOD-2). It also influenced the expression of TJPs (occludin, ZO-1, claudin-3, and MUC-3). In addition, the treatment of ATCC 14917 significantly reduced the level of lipopolysaccharide in the mesenteric adipose tissue. The findings of our study demonstrated that our strain effectively decreased the size of atherosclerotic plaques by modulating inflammation, oxidative stress, intestinal integrity, and intestinal immunity.

Inhibition of enzymatic browning in freeze-thawed apricot fruit by combined chlorogenic acid and osmotic dehydration treatments

In this study, the inhibitors of apricot browning were analyzed by investigating the effects of osmotic dehydration (OD) pretreatment combined with chlorogenic acid (CA) on cell disruption, loss of phenolic substrates, and changes in key enzymes during freeze-thawing. Both OD and OD-CA mitigated the deterioration caused by ice crystal damage during freeze-thawing. OD and OD-CA significantly reduces the BI of apricots by 43.27 and 59.55 compared to CK. The PPO and POD activity in OD-CA apricots are only 61.11% and 42.13% of those in OD. OD-CA could also protect SOD and CAT activity, with their activity in the OD-CA being 1.41 and 1.14 times that of OD group. This indicated that OD-CA treatment protected against the activities of ROS-scavenging enzymes. Compared with OD, OD-CA could better protect the bioactive substance of apricot. The contents of ascorbic acid, total phenols, and total flavonoids in OD-CA were 1.32 times, 1.11 times, and 1.05 times higher than those in OD. OD-CA also effectively protected chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, catechins, and rutin. In summary, OD-CA primarily reduced the browning by protecting the integrity of cell structures, minimizing the potential for exposure to air, enzymes, and phenolic substrates, and inhibiting browning enzymes activity.

Ashwagandha Diminishes Hippocampal Apoptosis Induced by Microwave Radiation by Acetylcholinesterase Dependent Neuro-Inflammatory Pathway in Male Coturnix coturnix Japonica.

Microwave radiation (MWR) has been linked to neurodegeneration by inducing oxidative stress in the hippocampus of brain responsible for learning and memory. Ashwagandha (ASW), a medicinal plant is known to prevent neurodegeneration and promote neuronal health. This study investigated the effects of MWR and ASW on oxidative stress and cholinergic imbalance in the hippocampus of adult male Japanese quail. One control group received no treatment, the second group quails were exposed to MWR at 2h/day for 30days, third was administered with ASW root extract orally 100mg/day/kg body weight and the fourth was exposed to MWR and also treated with ASW. The results showed that MWR increased serum corticosterone levels, disrupted cholinergic balance and induced neuro-inflammation. This neuro-inflammation further led to oxidative stress, as evidenced by decreased activity of antioxidant enzymes SOD, CAT and GSH. MWR also caused a significant decline in the nissil substances in the hippocampus region of brain indicating neurodegeneration through oxidative stress mediated hippocampal apoptosis. ASW, on the other hand, was able to effectively enhance the cholinergic balance and subsequently lower inflammation in hippocampus neurons. This suggests that ASW can protect against the neurodegenerative effects of MWR. ASW also reduced excessive ROS production by increasing the activity of ROS-scavenging enzymes. Additionally, ASW prevented neurodegeneration through decreased expression of caspase-3 and caspase-7 in hippocampus, thus promoting neuronal health. In conclusion, this study showed that MWR induces apoptosis and oxidative stress in the brain, while ASW reduces excessive ROS production, prevents neurodegeneration and promotes neuronal health.

MiR396b/GRF6 module contributes to salt tolerance in rice.

Salinity, as one of the most challenging environmental factors restraining crop growth and yield, poses a severe threat to global food security. To address the rising food demand, it is urgent to develop crop varieties with enhanced yield and greater salt tolerance by delving into genes associated with salt tolerance and high-yield traits. MiR396b/GRF6 module has previously been demonstrated to increase rice yield by shaping the inflorescence architecture. In this study, we revealed that miR396b/GRF6 module can significantly improve salt tolerance of rice. In comparison with the wild type, the survival rate of MIM396 and OE-GRF6 transgenic lines increased by 48.0% and 74.4%, respectively. Concurrent with the increased salt tolerance, the transgenic plants exhibited reduced H2O2 accumulation and elevated activities of ROS-scavenging enzymes (CAT, SOD and POD). Furthermore, we identified ZNF9, a negative regulator of rice salt tolerance, as directly binding to the promoter of miR396b to modulate the expression of miR396b/GRF6. Combined transcriptome and ChIP-seq analysis showed that MYB3R serves as the downstream target of miR396b/GRF6 in response to salt tolerance, and overexpression of MYB3R significantly enhanced salt tolerance. In conclusion, this study elucidated the potential mechanism underlying the response of the miR396b/GRF6 network to salt stress in rice. These findings offer a valuable genetic resource for the molecular breeding of high-yield rice varieties endowed with stronger salt tolerance.

CsHSFA1d Promotes Drought Stress Tolerance by Increasing the Content of Raffinose Family Oligosaccharides and Scavenging Accumulated Reactive Oxygen Species in Cucumber.

Drought is the most severe form of stress experienced by plants worldwide. Cucumber is a vegetable crop that requires a large amount of water throughout the growth period. In our previous study, we identified that overexpression of CsHSFA1d could improve cold tolerance and the content of endogenous jasmonic acid in cucumber seedlings. To explore the functional diversities of CsHSFA1d, we treat the transgenic plants under drought conditions. In this study, we found that the heat shock transcription factor HSFA1d (CsHSFA1d) could improve drought stress tolerance in cucumber. CsHSFA1d overexpression increased the expression levels of galactinol synthase (CsGolS3) and raffinose synthase (CsRS) genes, encoding the key enzymes for raffinose family oligosaccharide (RFO) biosynthesis. Furthermore, the lines overexpressing CsHSFA1d showed higher enzymatic activity of GolS and raffinose synthase to increase the content of RFO. Moreover, the CsHSFA1d-overexpression lines showed lower reactive oxygen species (ROS) accumulation and higher ROS-scavenging enzyme activity after drought treatment. The expressions of antioxidant genes CsPOD2, CsAPX1 and CsSOD1 were also upregulated in CsHSFA1d-overexpression lines. The expression levels of stress-responsive genes such as CsRD29A, CsLEA3 and CsP5CS1 were increased in CsHSFA1d-overexpression lines after drought treatment. We conclude that CsHSFA1d directly targets and regulates the expression of CsGolS3 and CsRS to promote the enzymatic activity and accumulation of RFO to increase the tolerance to drought stress. CsHSFA1d also improves ROS-scavenging enzyme activity and gene expression indirectly to reduce drought-induced ROS overaccumulation. This study therefore offers a new gene target to improve drought stress tolerance in cucumber and revealed the underlying mechanism by which CsHSFA1d functions in the drought stress by increasing the content of RFOs and scavenging the excessive accumulation of ROS.

Feedback regulation of the isoprenoid pathway by SsdTPS overexpression has the potential to enhance plant tolerance to drought stress.

In order to maintain the dynamic physiological balance, plants are compelled to adjust their energy metabolism and signal transduction to cope with the abiotic stresses caused by complex and changeable environments. The diterpenoid natural compound and secondary metabolites, sclareol, derived from Salvia sclarea, has gained significant attention owing to its economic value as a spice material and diverse physiological activities. Here, we focused on the roles and regulatory mechanisms of the sclareol diterpene synthase gene SsdTPS in the resistance of S. sclarea to abiotic stresses. Our results suggested that abiotic stresses could induce the response and upregulation of SsdTPS expression and isoprenoid pathway in S. sclarea. Ectopic expression of SsdTPS conferred drought tolerance in transgenic Arabidopsis, compared with wild-type. Overexpression of SsdTPS enhanced the transcription of ABA signal transduction synthetic regulators and induced the positive feedback upregulating key regulatory genes in the MEP pathway, thereby promoting the increase of ABA content and improving drought tolerance in transgenic plants. In addition, SsdTPS-overexpressed transgenic Arabidopsis improved the responses of stomatal regulatory genes and ROS scavenging enzyme activities and gene expression to drought stress. This promoted the stomatal closure and ROS reduction, thus enhancing water retention capacity and reducing oxidative stress damage. These findings unveil the potentially positive role of SsdTPS in orchestrating multiple regulatory mechanisms and maintaining homeostasis for improved abiotic stress resistance in S. sclarea, providing a novel insight into strategies for promoting drought resistance and cultivating highly tolerant plants.

Optimizing light regimes for neutral lipid accumulation in Dunaliella salina MCC 43: a study on physiological status and carbon allocation.

Dunaliella salina is a favourable source of high lipid feedstock for biofuel and medicinal chemicals. Low biomass output from microalgae is a significant barrier to industrial-scale commercialisation. The current study aimed to determine how photosynthetic efficiency, carbon fixation, macromolecular synthesis, accumulation of neutral lipids, and antioxidative defence (ROS scavenging enzyme activities) of D. salina cells were affected by different light intensities (LI) (50, 100, 200, and 400µmolm-2s-1). The cells when exposed to strong light (400µmolm-2s-1) led to reduction in chlorophyll a but the carotenoid content increased by 19% in comparison to the control (LI 100). The amount of carbohydrate changed significantly under high light and in spite of stress inflicted on the cells by high irradiation, a considerable increase in activity of carbonic anhydrase and fixation rate of CO2 were recorded, thus, preserving the biomass content. The high light exposed biomass when subjected to nitrogen-deficient medium led to increase in lipid content (59.92% of the dry cell weight). However, neutral lipid made up 78.26% of the total lipid while other lipids like phospholipid and glycolipid content decreased, showing that the lipid was redistributed in these cells under nitrogen deprivation, making the organism more appropriate for biodiesel/jet fuel use. Although D. salina cells had a relatively longer generation time (3.5 d) than other microalgal cells, an economic analysis concluded that the amount of carotenoid they produced and the quality of their lipids made them more suited for commercialization.

Exogenous melatonin orchestrates multiple defense responses against Botrytis cinerea in tomato leaves

Infection of tomato leaves and fruits by necrotrophic fungal pathogen Botrytis cinerea causes considerable economic loss. Melatonin has multiple physiological functions and is considered a potential biocontrol agent for its positive effects on plant innate immunity. In tomato, melatonin reduces Botrytis-caused post-harvest fruit decay by activating jasmonic acid (JA) signaling. In this study, we analyzed the effects of melatonin on resistance to Botrytis and associated defense mechanisms in tomato leaves. Treatment of detached tomato leaves with melatonin at a concentration as low as 100 µM induced resistance to Botrytis based on reduced lesion size and fungal growth. Melatonin-induced resistance to gray mold was associated with activation of a complex set of defense mechanisms in tomato leaves. First, melatonin suppressed Botrytis-induced cell death and accumulation of reactive oxygen species (ROS), which was correlated with increased activities of ROS-scavenging enzymes and enhanced expression of their corresponding genes. Second, melatonin treatment induced callose synthase genes and promoted rapid and strong callose deposition in tomato leaves. Melatonin also induced other anti-microbial genes encoding chitinases, β-1,3-glucanases, phenylalanine ammonialyases and polyphenol oxidases. Third, melatonin increased Botrytis-induced expression of genes encoding SlWRKY33, SlMYC2 and SlERF transcription factors, which activate defense-related genes expression through JA and ethylene (ET) signaling pathways. By contrast, melatonin suppressed salicylic acid (SA) signaling based on reduced expression of the SlPR1 marker gene, which is known to antagonize defense against Botrytis. These results collectively indicate that exogenous melatonin orchestrates multiple signaling pathways to activate a complex set of defense mechanisms against Botrytis in tomato leaves.

Melatonin alleviated cadmium accumulation and toxicity by modulating phytohormonal balance and antioxidant metabolism in rice

Cadmium (Cd) contamination is an eminent dilemma that jeopardizes global food safety and security, especially through its phytotoxicity in rice; one of the most edible crops. Melatonin (MET) has emerged as a protective phytohormone in stress conditions, but the defensive role and underlying mechanisms of MET against Cd toxicity in rice still remain unclear. To fulfill this knowledge gap, the present study is to uncover the key mechanisms for MET-mediated Cd-stress tolerance in rice. Cd toxicity significantly reduced growth by hindering the process of photosynthesis, cellular redox homeostasis, phytohormonal imbalance, and ultrastructural damages. Contrarily, MET supplementation considerably improved growth attributes, photosynthetic efficiency, and cellular ultrastructure as measured by gas exchange elements, chlorophyll content, reduced Cd accumulation, and ultrastructural analysis via transmission electron microscopy (TEM). MET treatment significantly reduced Cd accumulation (39.25%/31.58%), MDA (25.87%/19.45%), H2O2 (17.93%/9.56%), and O2 (29.11%/27.14%) levels in shoot/root tissues, respectively, when compared with Cd treatment. More importantly, MET manifested association with stress responsive phytohormones (ABA and IAA) and boosted the defense mechanisms of plant by enhancing the activities of ROS-scavenging antioxidant enzymes (SOD; superoxide dismutase, POD; peroxidase, CAT; catalase, APX; ascorbate peroxidase) and as well as regulating the key stress-responsive genes (OsSOD1, OsPOD1, OsCAT2, OsAPX1), thereby reinstate cellular membrane integrity and confer tolerance to ultrastructural damages under Cd-induced phytotoxicity. Overall, our findings emphasized the potential of MET as a long-term and cost-effective approach to Cd remediation in paddy soils, which can pave the way for a healthier and more environmentally conscious agricultural sector.

MsWRKY33 increases alfalfa (Medicago sativa L.) salt stress tolerance through altering the ROS scavenger via activating MsERF5 transcription.

Alfalfa (Medicago sativa L.) is considered to be the most important forage crop on a global scale. Nevertheless, soil salinity significantly decreases productivity, seriously threatening food security worldwide. One viable strategy is to explore salt stress-responsive factors and elucidate their underlying molecular mechanism, and utilize them in further alfalfa breeding. In the present study, we designated MsWRKY33 as a representative salt stress-responsive factor preferentially expressed in alfalfa roots and leaves. Subsequently, it was demonstrated that MsWRKY33 was localized in the cell nucleus, and functioned as a transcriptional activator of the W-box element. Transgenic alfalfa overexpressing MsWRKY33 displayed enhanced salt stress tolerance and antioxidant activities with no significant difference in other agronomic traits. Transcriptome profiling of MsWRKY33 transgenic alfalfa under control and salt treatment unveiled significantly altered expression of reactive oxygen species (ROS) scavenger genes in transgenic alfalfa. Subsequent examination revealed that MsWRKY33 binded to the promoter of MsERF5, activating its expression and consequently fine-tuning the ROS-scavenging enzyme activity. Furthermore, MsWRKY33 interacted with the functional fragment of MsCaMBP25, which participates in Ca2+ signaling transduction. Collectively, this research offers new insight into the molecular mechanism of alfalfa salt stress tolerance and highlights the potential utility of MsWRKY33 in alfalfa breeding.

Disruption of serotonin biosynthesis increases resistance to striped stem borer without changing innate defense response in rice.

Striped stem borer (SSB) is one of the most damaging pests in rice production worldwide. Previously, we preliminarily demonstrated that indica rice Jiazhe LM, an OsT5H (encoding tryptamine-5-hydroxylase) knockout mutant deficient in serotonin, had increased resistance to SSB as compared with its wildtype parent Jiazhe B. However, the full scenario of SSB resistance and the underlying mechanism remain unknown. In this study, we first demonstrated that the OsT5H knockout could generally increase rice resistance to SSB and then proved that the OsT5H knockout does not disrupt the innate defense response of rice plants to SSB infestation, that is, OsT5H knockout mutations neither had significant effect on the transcriptional response of defense genes upon SSB infestation, nor the profile of defense related metabolites and plant hormones, such as lignin, salicylic acid, jasmonic acid, and abscisic acid, nor the activity of reactive oxygen species(ROS) scavenging enzymes and the ROS contents. We then demonstrated that supplementation of serotonin promoted SSB growth and performance in artificial diet feeding experiments. We observed that SSB larvae feeding on Jiazhe B had serotonin 1.72- to 2.30-fold that of those feeding on Jiazhe LM at the whole body level, and more than 3.31 and 1.84 times in the hemolymph and head, respectively. Further studies showed that the expression of genes involved in serotonin biosynthesis and transport was ~88.1% greater in SSB larvae feeding on Jiahze LM than those feeding on Jiazhe B. These observations indicated that SSB increases serotonin synthesis when feeding on serotonin deficient rice but is unable to fully compensate the dietary serotonin deficiency. Put together, the present study strongly suggests that it is the deficiency of serotonin, not the secondary effect of OsT5H knockout on innate defense response confers the SSB resistance in rice, which implies that reducing serotonin level, particularly through inhibition of its inductive synthesis upon SSB damage, could be an efficient strategy for breeding SSB resistant varieties.

OsLPR5 Encoding Ferroxidase Positively Regulates the Tolerance to Salt Stress in Rice.

Salinity is a major abiotic stress that harms rice growth and productivity. Low phosphate roots (LPRs) play a central role in Pi deficiency-mediated inhibition of primary root growth and have ferroxidase activity. However, the function of LPRs in salt stress response and tolerance in plants remains largely unknown. Here, we reported that the OsLPR5 was induced by NaCl stress and positively regulates the tolerance to salt stress in rice. Under NaCl stress, overexpression of OsLPR5 led to increased ferroxidase activity, more green leaves, higher levels of chlorophyll and lower MDA contents compared with the WT. In addition, OsLPR5 could promote the accumulation of cell osmotic adjustment substances and promote ROS-scavenging enzyme activities. Conversely, the mutant lpr5 had a lower ferroxidase activity and suffered severe damage under salt stress. Moreover, knock out of OsLPR5 caused excessive Na+ levels and Na+/K+ ratios. Taken together, our results exemplify a new molecular link between ferroxidase and salt stress tolerance in rice.

Nordihydroguaiaretic acid (NDGA) maintains the postharvest quality of fresh daylily flower buds (Hemerocallis citrina) by enhancing antioxidant metabolism and regulating phytohormone equilibrium

Daylily (Hemerocallis citrina) bud is a functional vegetable rich in various nutrients and many bioactive compounds. However, fresh daylily buds are very susceptible to senescence and quality deterioration occurs rapidly after harvest. The effects of nordihydroguaiaretic acid (NDGA) on postharvest quality, reactive oxygen species (ROS) metabolism and phytohormone alterations in daylily buds were investigated. The results showed that NDGA treatments delayed the loss of overall quality, and alleviated yellowing and disaggregation. The NDGA treatment inhibited the production of hydrogen peroxide and malondialdehyde, enhanced activities of ROS-scavenging enzymes and delayed the decrease in total antioxidant capacity. The biosynthesis of abscisic acid was decreased, while the contents of indole-3-acetic acid and brassinosteroids were increased by NDGA treatment, and therefore equilibrium involving several phytohormones was altered by NDGA. Transcriptomic profiling of genes associated with the signal transduction pathways of the main phytohormones was performed to reveal alterations in the regulation of postharvest senescence of daylily buds in response to NDGA. Collectively, NDGA treatment is a potential approach for the preservation of fresh daylily flower buds.

Exogenous Melatonin Alters Stomatal Regulation in Tomato Seedlings Subjected to Combined Heat and Drought Stress through Mechanisms Distinct from ABA Signaling

The understanding of stomatal regulation in climate stress is essential for ensuring resilient crops. The investigation of the stomatal regulation in combined heat and drought stress aimed to link effects of exogenous melatonin on stomatal conductance (gs) and its mechanistic interactions with ABA or ROS signaling. Melatonin-treated and non-treated tomato seedlings were subjected to moderate and severe levels of heat (38°C for one or three days) and drought stress (soil relative water content of 50% or 20%) applied individually and in combination. We measured gs, stomatal anatomy, ABA metabolites and enzymatic ROS scavengers. The stomata in combined stress responded predominantly to heat at soil relative water content (SRWC) = 50% and to drought stress at SRWC = 20%. Drought stress increased ABA levels at severe stress, whereas heat stress caused an accumulation of the conjugated form, ABA glucose ester, at both moderate and severe stress. The melatonin treatment affected gs and the activity of ROS scavenging enzymes but had no effect on ABA levels. The ABA metabolism and conjugation of ABA might play a role in stomatal opening toward high temperatures. We provide evidence that melatonin increases gs in combined heat and drought stress, but the effect is not mediated through ABA signaling.

Exogenously applied melatonin enhanced the tolerance of Brassica napus against cobalt toxicity by modulating antioxidant defense, osmotic adjustment, and expression of stress response genes

The excessive accumulation of cobalt (Co) in plant tissues severely impairs plant growth that ultimately reduces the yield. However, melatonin (MT) has been known to mediate the abiotic stress tolerance in plants. The present study aimed at investigating the protective mechanisms of exogenously applied MT (0, 50 and 100 μM) under Co (0, 100, 200 and 300 μM) stress by focusing on morpho-physiological, biochemical and cellular characterizations of Brassica napus plants. Cobalt (300 μM) alone treatment drastically inhibited the stomatal conductance, plant height (45%), leaf area (30%), free amino acid (139%), relative electrolyte leakage (109%), and total soluble sugars (71%), compared with the control. However, the exogenous supply of MT notably minimized the oxidative damage, lipid peroxidation and maintained the membrane integrity under Co-toxicity by restricting the overproduction of ROS (H2O2 and O2•), and MDA in leaves and roots. Melatonin significantly enhanced the activities of ROS-scavenging antioxidant enzymes, secondary metabolism-related phenylalanine ammonia lyase (PAL), polyphenol oxidase (PPO), stress-responsive genes (heat shock protein as HSP-90, methyl transferase as MT) and regulated the Co-transporters, especially in roots. These findings indicated that an exogenous supply of MT improve the plant morphology, photosynthetic apparatus, osmotic adjustments, and antioxidant defense systems by enhancing the Co-detoxification in B. napus plants.

Hot water treatment improves peach fruit cold resistance through PpHSFA4c-mediated HSF-HSP and ROS pathways

Peach fruit is cold-sensitive and susceptible to chilling injury (CI), and hot water (HW) treatment can reduce their CI. However, the mechanism of HW treatment reduced CI has not been characterized. In this study, HW treatment suppressed the increases in CI index, electrolyte leakage, and malonaldehyde (MDA) content in peach fruit during cold storage. It also decreased reactive oxygen species (ROS) accumulation and increased the activity of ROS-scavenging enzymes. Transcriptome analysis indicated that HW treatment might contribute to the crosstalk between the heat shock factor-heat shock protein (HSF-HSP) and ROS pathways during cold storage. PpHSFA4c was a key transcription factor and could up-regulate defense genes to alleviate the CI of peach fruit during storage. Furthermore, PpHSFA4c could activate the expressions of PpHSP18.5, PpHSP70, PpHSP83, PpAPX1, and PpAPX3 by interacting with their promoters. These studies indicated that HW treatment alleviated the CI, increased antioxidant activity, and maintained ROS homeostasis of peach fruit through the PpHSFA4c-mediated HSF-HSP and ROS pathways. This study provides novel insights into the regulatory mechanisms of HW treatment alleviating CI in postharvest peach fruit, and expands the theoretical basis for commercial application of HW treatment technology to maintain quality and reduce CI in cold-sensitive fruit.
