(2001). The changes had the following consequences for the results of the qRT-PCR analyses: (i) transcript levels of dsGFP treatments are closer to untreated controls (UT); (ii) CsIAP1 transcripts were reduced on average by 33% instead of the initially reported 40%; (iii) variation of both dsGFP and dsCsIAP1 transcript levels increased slightly; (iv) the combination of increased variation and lowered reduction in transcript levels resulted in a loss of the originally reported statistical significance (Student's t-test, P=0.07). Figure 4 and its legend (Page 109) were updated: Figure 4. Relative expression of CsIAP1 transcripts after dsRNA injection (changes italicized): Graph depicts CsIAP1 mean transcript levels at 5 dpi for dsGFP and dsCsIAP1-injected midges relative to untreated controls. QRT-PCR results were analyzed using EF1b as reference gene and untreated midges as calibrator condition. On average, mRNA levels of CsIAP1 were unaffected by dsGFP injection, and reduced by 33% after dsCsIAP1 treatment compared to untreated controls (UT). However, expression levels between dsGFP and dsCsIAP1-treated midges were not statistically significantly different (Student's t-test, P = 0.07). Data are presented as mean ± one S.E.M., from four biological replicates (Figure S2). The last paragraph of the Experimental Procedures (Page 112) should read (changes italicized): Fold change for each treatment (dsGFP or dsCsIAP1) was assessed using a modified ΔΔCt method (Pfaffl, 2001), which takes potentially different primer efficiencies (Figure S10) into account. All qRT-PCRs were performed with 3 technical and four biological replicates. Untreated controls were used as the calibrator in each biological replicate. Expression data between dsGFP and dsCsIAP1 treatments were compared statistically with Student's t-test. Statistical analyses were performed using GraphPad Prism software version 5.01 (GraphPad Software Inc., La Jolla, CA, USA). Sentences 5 and 6 of paragraph 5 (Discussion - Page 110) should read (changes italicized): The 33% transcript reduction of CsIAP1 induced by a comparatively high dose of dsRNA (Miller et al., 2012) suggests that the initial RNAi trigger is not amplified in C. sonorensis, and is likely not taken up by all tissues. Similar results have been obtained from closely related Culicidae (Blandin et al., 2002, Zhu et al., 2003).
