Nutritional enhancement in black seed (Nigella sativa L.) using bacteria‐based biofertilizers

AbstractNigella sativa L. is an aromatic spice, utilized as an original and peculiar flavoring ingredient in a variety of culinary applications and pharmaceuticals. Black seed (Nigella sativa L.) belongs to the family Ranunculaceae. It is an undercultivated crop in Pakistan. The present study was planned keeping in mind sustainable development goals SDG 3 (good health and well‐being) and SDG 15 (life on land). The effects of several rhizospheric bacterial strains and synthetic fertilizers on the development of N. sativa and nutrition were studied using a completely randomized experimental design. For this purpose, plant growth‐promoting effects of different strains (Azospirillum brasilense, Azospirillum lipoferum, and Pantoea agglomerans) and synthetic fertilizers (nitrogen and phosphorus) were assembled to check their effects individually and in combination form. Azospirillum lipoferum and Pantoea agglomerans inoculation significantly enhanced the morphological characteristics of N. sativa, whether applied individually or in combination, with positive effects on seedlings, plant height, number of branches, number of leaves, number of flowers, stamens numbers, follicles number, number of tentacles and seed production. N. sativa plants that were simultaneously inoculated with Azospirillum lipoferum and Pantoea agglomerans showed the highest potential for antioxidant activity, particularly in petroleum ether extracts. In the methanolic extract, a higher amount of radical scavenging was observed as compared to positive and negative control. There was also increase in fat, moisture and carbohydrate contents of the combination inoculated plant. So, from the present study, in Pakistan, the technique is recommended to enhance the yield and nutritional value of N. sativa.

Sustainable agriculture through rhizosphere bacteria for plant growth promotion, nutrient acquisition, and stress alleviation

Rhizosphere bacteria are attracting a lot of interest in sustainable agriculture because of their capacity to stimulate plant growth, improve nutrient uptake, and reduce stress in plants. A review of recent discoveries in the field of using rhizosphere bacteria for these advantageous agricultural techniques is given in this article. Rhizosphere bacteria facilitate plant growth by solubilizing inaccessible elements like phosphorus, potassium, zinc, and manganese by producing a variety of enzymes and organic acids. Furthermore, these bacteria support plant nutrition by fixing atmospheric nitrogen, a necessary component for plant growth and development. Moreover, some rhizosphere bacterial strains can inhibit plant diseases, which lessen the need for artificial chemical pesticides. Rhizosphere bacteria have improved plant stress tolerance by producing stress-related substances, including phytohormones and osmoprotectants. These bacteria also give rise to systemic resistance in plants, successfully allowing them to survive various biotic and abiotic challenges. Concerning climate change and environmental variations, this rhizosphere bacteria-mediated stress reduction component holds a tremendous potential for sustainable agriculture. But for rhizosphere bacteria-based agricultural techniques to be successfully utilized, a thorough grasp of their variety, ecological relationships, and mode of action are necessary. Optimizing application techniques, formulating bioinoculants, and considering environmental aspects are crucial for consistent and dependable outcomes through rhizosphere bacteria.

Inoculation with plant growth-promoting rhizobacteria improves seagrass Thalassia hemprichii photosynthesis performance and shifts rhizosphere microbiome

Plant growth-promoting rhizobacteria (PGPR) inoculation is a crucial strategy for maintaining the sustainability of agriculture and presents a promising solution for seagrass ecological restoration in the face of disturbances. However, the possible roles and functions of PGPRs in the seagrass rhizosphere remain unclear. Here, we isolated rhizosphere bacterial strains from both reef and coastal regions and screened two PGPR isolates regarding their in vivo functional traits. Subsequently, we conducted microcosm experiments to elucidate how PGPR inoculation affected seagrass photosynthesis and shape within each rhizosphere microbiome. Both screened PGPR strains, Raoultella terrigena NXT28 and Bacillus aryabhattai XT37, excelled at expressing a specific subset of plant-beneficial functions and increased the photosynthetic rates of the seagrass host. PGPR inoculation not only decreased the abundance of sulfur-cycling bacteria, it also improved the abundance of putative iron-cycling bacteria in the seagrass rhizosphere. Strain XT37 successfully colonized the seagrass rhizosphere and displayed a leading role in microbial network structure. As a nitrogen-fixing bacteria, NXT28 showed potential to change the microbial nitrogen cycle with denitrification in the rhizosphere and alter dissimilatory and assimilatory nitrate reduction in bulk sediment. These findings have implications for the development of eco-friendly strategies aimed at exploiting microbial communities to confer sulfide tolerance in coastal seagrass ecosystem.

Biocontrol Potential of Some Rhizospheric Soil Bacterial Strains against Fusarium culmorum and Subsequent Effect on Growth of Two Tunisian Wheat Cultivars

PGPR (Plant Growth Promoting Rhizobacteria) are used as biofertilizers and biological control agents against fungi. The objective of this work was to evaluate the antagonistic activities of some bacterial strains isolated from soil against four phytopathogenic fungal strains (Fusarium graminearum, F. culmorum, Phytophthora sp. and Verticillium dahlia). Two strains having an antagonist effect on fungi and displaying the maximum of plant growth promoting (PGP) traits were selected for further study and identified as Bacillus subtilis and B. amyloliquefaciens respectively. In planta assays demonstrated that the two Bacillus strains are able to enhance plant growth of two wheat cultivars in absence of nitrogen and protect them against F. culmorum. Pot experiments performed in a greenhouse showed that wheat plants inoculation with two bacterial strains reduce F. culmorum disease severity correlated with the accumulation of phenolic compounds and chlorophyll content. These could partly explain the effectiveness of these bacteria in protecting Tunisian durum wheat cultivars against F. culmorum. Application B. amyloliquefaciens, showed better protection than B. subtilis although the last one enhanced more the plant growth of two wheat cultivars in absence of fungus. Hence, combination of two bacterial strains could be a strategic approach to enhance plant growth and control plant diseases.

Degradation of 8:2 fluorotelomer carboxylic acid (8:2 FTCA) by plants and their co-existing microorganisms

8:2 fluorotelomer carboxylic acid (8:2 FTCA), an important precursor of perfluorocarboxylic acids (PFCAs), is widely detected in environment and biotas. Hydroponic exposures were conducted to investigate the accumulation and metabolism of 8:2 FTCA in wheat (Triticum aestivum L.) and pumpkin (Cucurbita maxima L.). Endophytic and rhizospheric microorganisms co-existing with the plants were isolated to investigate their contributions to degrade 8:2 FTCA. Wheat and pumpkin roots could take up 8:2 FTCA efficiently with the root concentration factor (RCF) as 5.78 and 8.93, respectively. 8:2 FTCA could be biotransformed to 8:2 fluorotelomer unsaturated carboxylic acid (8:2 FTUCA), 7:3 fluorotelomer carboxylic acid (7:3 FTCA), and seven PFCAs with 2–8 carbon chain length in plant roots and shoots. Cytochromes P450 (CYP450) and glutathione-S-transferase (GST) activities in plants were significantly increased, while flavin-dependent monooxygenases (FMOs) activities were not changed, suggesting that CYP 450 and GST were involved in the transformation of 8:2 FTCA in plant tissues. Twelve 8:2 FTCA-degrading endophytic (8 strains) and rhizospheric (4 strains) bacterial strains were isolated from root interior, shoot interior and rhizosphere of plants, respectively. These bacteria were identified as Klebsiella sp. based on the morphology and 16S rDNA sequence, and they could biodegrade 8:2 FTCA to intermediates and stable PFCAs.

Comparative study of the rhizosphere microbiome of Coffea arabica grown in different countries reveals a small set of prevalent and keystone taxa

Coffee (Coffea arabica) is a globally important crop and its cultivation demands the use of agrochemicals and pesticides. Recently, plant-associated microorganisms have emerged as an unexploited resource to enhance crop performance and sustainability. In particular, the rhizosphere microbiome has been identified as a hotspot for plant health, by providing tolerance to biotic and abiotic stresses as well as regulating phenological traits. However, its composition varies widely across crops and environmental conditions. Hence, the knowledge of the “common core” taxa, which are a subset of the microbiome that is persistently associated, across different environmental gradients and conditions, to a host species, represent valuable targets for the development of microbiome-based management approaches. This study aimed to characterize and determine the rhizosphere-inhabiting microbial groups (common core and most prevalent taxa), associated with the Coffea arabica plants in different geographical locations. To assess the structure of the coffee rhizosphere microbiome, we performed the 16 S rRNA amplicon analysis of samples collected across six locations spanning four countries and grown under different climate conditions, soil type, agroecosystem management, plant vegetative stage and age. Our findings allowed us to delineate and identify a robust set of six bacterial taxa, namely Sphingobium, Rhizobium group, Acidibacter, Sphingomonas, Burkholderia group and Amycolatopsis, which are likely to play an important role for the growth and health of coffee plants. Using network analysis, we observed that some of the six “common core” taxa were highly interconnected with the resident community, being also keystone taxa of the coffee rhizosphere microbiome. Furthermore, we performed a high-throughput targeted isolation resulting into 162 rhizosphere bacterial strains, some of them related to the most prevalent taxa. These isolates represent a working bacterial culture collection to be used in the future for the development of microbiome-based solutions for a more sustainable coffee farming.

The impact of newly synthesized sulfonamides on soil microbial population and respiration in rhizospheric soil of wheat (Triticum aestivum L.).

Antibiotics released into agricultural fields through the manure of grazing animals could exert harmful impacts on soil microbes and plants. Antibiotics exert high impacts on environment than other pharmaceuticals due to their higher biological activity. However, little is known about their impacts on plants, despite indications that antibiotics exert negative effects on soil microorganisms, which ultimately harm the plants. It has been demonstrated that beneficial microorganisms promote plant growth and development under various stresses. This study evaluated the toxicity of four newly derived sulfonamides (SAs), i.e., 2-(phenylsulfonyl) hydrazine carbothioamide (TSBS-1), N, 2-bis phenyl hydrazine carbothioamide (TSBS-2), aminocarbonyl benzene sulfonamide (UBS-1), and N, N’-carbonyl dibenzene sulfonamide (UBS-2) on bacterial growth and soil microbial respiration. Each SA was tested at four different concentrations (i.e., 2.25, 2.5, 3, 4 mg/ml) against five rhizospheric bacterial strains, including AC (Actinobacteria sp.), RS-3a (Bacillus sp.), RS-7a (Bacillus subtilis), RS-4a (Enterobacter sp.), and RS-5a (Enterobacter sp.). Antimicrobial activity was checked by disc diffusion method, which showed that inhibition zone increased with increasing concentration of SAs. The UBS-1 resulted in the highest inhibition zone (11.47 ± 0.90 mm) against RS-4a with the highest concentration (4 mg/ml). Except TSBS-1, all sulfonamide derivatives reduced CO2 respiration rates in soil. Soil respiration values significantly increased till 6th day; however, exposure of sulfonamide derivatives suppressed microbial respiration after 6th day. On the 20th day, poor respiration activity was noted at 0.23, 0.2, and 0.4 (CO2 mg/g dry soil) for TSBS-1, UBS-1, and UBS-2, respectively. Our results demonstrate that sulfonamides, even in small concentrations, significantly affect soil microbial population and respiration. Soil microbial respiration changes mediated by sulfonamides were dependent on length of exposure and concentration. It is recommended that antibiotics should be carefully watched and their impact on plant growth should be tested in the future studies.

Uncovering the phytochemicals of root exudates and extracts of lead (Pb) tolerant Chrysopogon zizanioides (L.) Roberty in response to lead contamination and their effect on the chemotactic behavior of rhizospheric bacteria.

The chemical composition of root exudates and root extracts from Chrysopogon zizanioides (L.) Roberty cv KS-1 was determined in the presence of lead [Pb(II)]. Hitherto, no information is available in the literature concerning the phytochemical components of root exudates of C. zizanioides. Significantly higher concentrations of total carbohydrates (26.75 and 42.62% in root exudates and root extract, respectively), reducing sugars (21.46 and 56.11% in root exudates and root extract, respectively), total proteins (9.22 and 23.70% in root exudates and root extract, respectively), total phenolic acids (14.69 and 8.33% in root exudates and root extract, respectively), total flavonoids (14.30 and 12.28% in root exudates and root extract, respectively), and total alkaloids (12.48 and 7.96% in root exudates and root extract, respectively) were observed in samples from plants growing under Pb(II) stress in comparison to the respective controls. GC-MS profiling showed the presence of a diverse group of compounds in root exudates and extracts, including terpenes, alkaloids, flavonoids, carotenoids, plant hormones, carboxylic/organic acids, and fatty acids. Among the detected compounds, many have an important role in plant development, regulating rhizosphere microbiota and allelopathy. Furthermore, the results indicated that C. zizanioides exudates possess a chemotactic response for rhizospheric bacterial strains Bacillus licheniformis, Bacillus subtilis, and Acinetobacter junii Pb1.

Plant growth-promoting bacteria improve Triticum aestivum L. growth and photosynthetic activity in sulfonamide-contaminated soil

Antibiotics are disseminated by grazing animals or their feces throughout agricultural areas. These antibiotics, as environmental pollutants, have a negative impact on plant growth and physiology. Sulfonamides are frequently used to protect livestock production, but they also have a considerable negative impact on plant growth and rhizospheric bacteria. Plants are sensitive to their surroundings, and they employ their internal biological systems to deal with the threats to their growth and development. Soil microbes, which are the most common inhabitants, have amazing metabolic capacities that enable them to overcome abiotic constraints. The effects of sulfonamide stress on wheat cultivar Chakwal-50 were studied in terms of morphological and biochemical processes, as well as stress mitigation by microorganisms isolated from wheat rhizospheric soil. This research looked at the effects of four new derived sulfonamides (SAs): 2-phenylsulfonyl hydrazine carbothioamide (TSBS-1), N,2-bis phenylhydrazine carbothioamide (TSBS-2), aminocarbonyl benzene sulfonamide (UBS-1), and N, N'-carboxyl dibenzene sulfonamide (UBS-2). Five rhizospheric bacterial strains were used: AC (Actinobacteria sp.), RS-3a (Bacillus sp.), RS-7a (Bacillus subtilis), RS-4a (Enterobacter sp.), and RS-5a (Enterobacter sp.). T. aestivum seeds were sterilized before being cultivated in soil polluted with various sulfonamide derivatives and inoculated with various bacterial strains under natural environmental conditions. To analyze the effect of sulfonamides on several aspects of plant growth, data from each antibiotic and bacterial strain was used to determine shoot fresh weight, root fresh weight, root and shoot length, number of leaves, and photosynthetic activity on the 15th day of germination. According to the results, Inoculation of Bacillus spp. (RS-3a) produced a maximum shoot fresh weight (FW) and shoot length by 8.14 g and 15.19 cm, respectively. Actinobacteria sp. (AC) had the highest chlorophyll value of 37 μg/g FW, while Bacillus spp. (RS-3a) increased carotenoid values with the average value of 550 μg/g FW by reducing sulfonamide stress. Following this preliminary screening, some bacterial strains were found to be promising plant growth promoting rhizobacteria for improving plant growth in a way that was both safe and effective.

Rhizospheric bacteria as potential biocontrol agents against Fusarium wilt and crown and root rot diseases in tomato

The discovery of novel biocontrol agents requires the continuous scrutiny of native microorganisms to ensure that they will be useful on a regional scale. The goal of the present work was to discover novel antagonistic bacteria against Fusarium oxysporum ff. spp. lycopersici race 3 (Fol R3) and radicis-lycopersici (Forl) causing Fusarium wilt disease and Fusarium crown and root rot of tomatoes, respectively. High-throughput liquid antagonism screening of 1,875 rhizospheric bacterial strains followed by dual confrontation assays in 96-well plates was used to select bacteria exhibiting > 50% fungal growth inhibition. In a second dual confrontation assay in 10-cm Petri dishes, bacteria showing > 20% Fol R3 or Forl growth inhibition were further screened using a blood hemolysis test. After discarding β-hemolytic bacteria, a seedling antagonistic assay was performed to select five potential antagonists. A phylogenetic analysis of 16S rRNA identified one strain as Acinetobacter calcoaceticus (AcDB3) and four strains as members of the genus Bacillus (B. amyloliquefaciens BaMA26, Bacillus siamensis BsiDA2, B. subtilis BsTA16 and B. thuringiensis BtMB9). Greenhouse assays demonstrated that BsTA16 and AcDB3 were the most promising antagonists against Fol R3 and Forl, respectively. Pathogen biocontrol and growth promotion mechanisms used by these bacteria include the production of siderophores, biofilm, proteases, endoglucanases and indole acetic acid, and phosphate solubilization. These five bacteria exerted differential responses on pathogen control depending on the tomato hybrid, and on the growth stage of tomatoes. We report for the first time the use of an Acinetobacter calcoaceticus isolate (AcDB3) to control Forl in tomato under greenhouse conditions.

Antimicrobial activity of bacteria associated with the rhizosphere and phyllosphere of Avena fatua and Brachiaria reptans.

Environmental pollution especially heavy metal-contaminated soils adversely affects the microbial communities associated with the rhizosphere and phyllosphere of plants growing in these areas. In the current study, we identified and characterized the rhizospheric and phyllospheric bacterial strains from Avena fatua and Brachiaria reptans with the potential for antimicrobial activity and heavy metal resistance. A total of 18 bacterial strains from the rhizosphere and phyllosphere of A. fatua and 19 bacterial strains from the rhizosphere and phyllosphere of B. reptans were identified based on 16S rRNA sequence analysis. Bacterial genera, including Bacillus, Staphylococcus, Pseudomonas, and Enterobacter were dominant in the rhizosphere and phyllosphere of A. fatua and Bacillus, Marinobacter, Pseudomonas, Enterobacter, and Kocuria, were the dominating bacterial genera from the rhizosphere and phyllosphere of B. reptans. Most of the bacterial strains were resistant to heavy metals (Cd, Pb, and Cr) and showed antimicrobial activity against different pathogenic bacterial strains. The whole-genome sequence analysis of Pseudomonas putida BR-PH17, a strain isolated from the phyllosphere of B. reptans, was performed by using the Illumina sequencing approach. The BR-PH17 genome contained a chromosome with a size of 5774330 bp and a plasmid DNA with 80360 bp. In this genome, about 5368 predicted protein-coding sequences with 5539 total genes, 22 rRNAs, and 75 tRNA genes were identified. Functional analysis of chromosomal and plasmid DNA revealed a variety of enzymes and proteins involved in antibiotic resistance and biodegradation of complex organic pollutants. These results indicated that bacterial strains identified in this study could be utilized for bioremediation of heavy metal-contaminated soils and as a novel source of antimicrobial drugs.

Stigmasterol root exudation arising from Pseudomonas inoculation of the duckweed rhizosphere enhances nitrogen removal from polluted waters

Rhizospheric microorganisms such as denitrifying bacteria are able to affect ‘rhizobioaugmention’ in aquatic plants and can help boost wastewater purification by benefiting plant growth, but little is known about their effects on the production of plant root exudates, and how such exudates may affect microorganismal nitrogen removal. Here, we assess the effects of the rhizospheric Pseudomonas inoculant strain RWX31 on the root exudate profile of the duckweed Spirodela polyrrhiza, using gas chromatography/mass spectrometry. Compared to untreated plants, inoculation with RWX31 specifically induced the exudation of two sterols, stigmasterol and β-sitosterol. An authentic standard assay revealed that stigmasterol significantly promoted nitrogen removal and biofilm formation by the denitrifying bacterial strain RWX31, whereas β-sitosterol had no effect. Assays for denitrifying enzyme activity were conducted to show that stigmasterol stimulated nitrogen removal by targeting nitrite reductase in bacteria. Enhanced N removal from water by stigmasterol, and a synergistic stimulatory effect with RWX31, was observed in open duckweed cultivation systems. We suggest that this is linked to a modulation of community composition of nirS- and nirK-type denitrifying bacteria in the rhizosphere, with a higher abundance of Bosea, Rhizobium, and Brucella, and a lower abundance of Rubrivivax. Our findings provide important new insights into the interaction of duckweed with the rhizospheric bacterial strain RWX31 and their involvement in the aquatic N cycle and offer a new path toward more effective bio-formulations for the purification of N-polluted waters.

Glyphosate-remediation potential of selected plant species in artificial wetlands

Glyphosate is a systemic herbicide with an important environmental impact due to its adverse effect on non-target organisms. Its removal can be accomplished by wild (native) plants which, together with their associated microbiota, have acquired tolerance against glyphosate or are able to degrade it. Little is known, however, about these plants' potential for glyphosate remediation and the role that plant-microbe interactions play in this process. The objective of the present study was to evaluate plant species from locations with a history of glyphosate exposure – drainage ditches of banana plantations (Musa sp.), or riparian areas - to determine which, in combination with their microbiota, promote the removal of glyphosate. Five plant species were selected to evaluate their response to inundated growing conditions and exposure to 20 mg glyphosate acid equivalent/L for 47 days. Parameters affecting plant growth and glyphosate removal were recorded. Rhizosphere bacterial strains were identified and their tolerance to different glyphosate concentrations determined. Also, the activity of 19 rhizospheric enzymes associated with the phosphorus, carbon and nitrogen cycles were analyzed. Glyphosate-treated Panicum maximum exhibited the highest removal efficiency (87%), followed by Typha domingensis and Heliconia latispatha. All plants were associated with glyphosate-tolerant bacteria, and in the cases of Xanthosoma sagittifolium and Fimbristylis dichotoma, strains were recovered that even succeeded in growing in a medium containing 40 g glyphosate ae/L. Processes involved in carbon, nitrogen and phosphorus metabolism in the rhizosphere were affected by glyphosate exposure, and the importance of plant-microbe-substrate interactions in the elimination of glyphosate was evinced. Although all plants contributed to the removal of glyphosate, their use in phytoremediation projects will depend on the location and nature of the environmental problem to be resolved.

Rhizobacterial consortium mediated aroma and yield enhancement in basmati and non-basmati rice (Oryza sativa L.)

Basmati and non-basmati rice varieties are commercially important. Aromatic rice varieties are low yielding and recently depletion in aroma is observed due to the shift towards modern agriculture. Therefore, it is necessary to restore the aroma and increase the yield through sustainable agriculture. The use of microbial bioinoculants is one of the promising ways to achieve these targets. With these objectives, rhizospheric bacterial strains Enterobacter hormaechei (AM122) and Lysinibacillus xylanilyticus (DB25) having the property of synthesizing 2-acetyl-1-pyrroline (2AP) were isolated from the rhizosphere of two aromatic rice varieties, Ambemohar-157 and Dehradun Basmati respectively and their effect on plant growth, aroma and yield enhancement under mono-inoculation and consortium conditions was analyzed. The bacterial inoculum in consortium resulted in significant improvement in vegetative growth, yield and 2AP content over mono inoculation and control. The study highlights the potential of E. hormaechei and L. xylanilyticus in plant growth, yield and aroma enhancement in basmati and non-basmati rice varieties. These strains can be taken up further for developing a commercial bioformulation.

Study on the optimal antagonistic effect of a bacterial complex against Monilinia fructicola in peach.

Peach brown rot caused by Monilinia fructicola is one of the most economically destructive diseases of peach (Prunus persica L.) in some orchards of China. Biocontrol is a significant strategy that exhibits strong levels of control and ecologically sound concepts in disease management. The purpose of this study was to investigate the combined suppressive effects of three endophytic bacterial strains (xj-14, xj-15, and xj-16) and two soil rhizosphere bacterial strains (xj-A and xj-C) that were shown to have strong inhibitory activity toward M. fructicola in our previous study. The optimal strains and the optimized combination of strains were determined. The combination of strains xj-15 and xj-C inhibited M. fructicola more intensively for a longer period of time. Following the application of 1 × 109 CFU/mL bacterial complex to the fruits, leaves, and shoots of peach trees infected with M. fructicola, the rate of inhibition reached 73.80%, 83.33%, and 90.43%, respectively. A pot experiment using lettuce (Lactuca sativa) showed that inoculation with the bacterial complex significantly increased the growth of seedlings. In this study, some compound bacteria were more effective than those in previous study in suppressing disease and promoting growth, which have the potential to be further applied in the field.

Phenol Removal Capacity of the Common Duckweed (Lemna minor L.) and Six Phenol-Resistant Bacterial Strains From Its Rhizosphere: In Vitro Evaluation at High Phenol Concentrations.

The main topic of this study is the bioremediation potential of the common duckweed, Lemna minor L., and selected rhizospheric bacterial strains in removing phenol from aqueous environments at extremely high initial phenol concentrations. To that end, fluorescence microscopy, MIC tests, biofilm formation, the phenol removal test (4-AAP method), the Salkowski essay, and studies of multiplication rates of sterile and inoculated duckweed in MS medium with phenol (200, 500, 750, and 1000 mg L−1) were conducted. Out of seven bacterial strains, six were identified as epiphytes or endophytes that efficiently removed phenol. The phenol removal experiment showed that the bacteria/duckweed system was more efficient during the first 24 h compared to the sterile duckweed control group. At the end of this experiment, almost 90% of the initial phenol concentration was removed by both groups, respectively. The bacteria stimulated the duckweed multiplication even at a high bacterial population density (>105 CFU mL−1) over a prolonged period of time (14 days). All bacterial strains were sensitive to all the applied antibiotics and formed biofilms in vitro. The dual bacteria/duckweed system, especially the one containing strain 43-Hafnia paralvei C32-106/3, Accession No. MF526939, had a number of characteristics that are advantageous in bioremediation, such as high phenol removal efficiency, biofilm formation, safety (antibiotic sensitivity), and stimulation of duckweed multiplication.

Nitrogen Substrate Utilization in Three Rhizosphere Bacterial Strains Investigated Using Proteomics.

Nitrogen metabolism in the rhizosphere microbiome plays an important role in mediating plant nutrition, particularly under low inputs of mineral fertilizers. However, there is relatively little mechanistic information about which genes and metabolic pathways are induced by rhizosphere bacterial strains to utilize diverse nitrogen substrates. Here we investigate nitrogen substrate utilization in three taxonomically diverse bacterial strains previously isolated from Arabidopsis roots. The three strains represent taxa that are consistently detected as core members of the plant microbiome: Pseudomonas, Streptomyces, and Rhizobium. We use phenotype microarrays to determine the nitrogen substrate preferences of these strains, and compare the experimental results vs. computational simulations of genome-scale metabolic network models obtained with EnsembleFBA. Results show that all three strains exhibit generalistic nitrogen substrate preferences, with substrate utilization being well predicted by EnsembleFBA. Using label-free quantitative proteomics, we document hundreds of proteins in each strain that exhibit differential abundance values following cultivation on five different nitrogen sources: ammonium, glutamate, lysine, serine, and urea. The proteomic response to these nitrogen sources was strongly strain-dependent, with lysine nutrition eliciting widespread protein-level changes in Pseudomonas sp. Root9, whereas Rhizobium sp. Root491 showed relatively stable proteome composition across different nitrogen sources. Our results give new protein-level information about the specific transporters and enzymes induced by diverse rhizosphere bacterial strains to utilize organic nitrogen substrates.

Study of the Modulating Interactions of Multitrait Rhizobacteria Using Zea Mays L. as the Host Plant

Background: Rhizosphere is a soil region closest to roots of the plants inhabiting different types of microorganisms including rhizobacteria. Chemical fertilizers which are conventionally used for increasing crop production are dangerous in terms of minimizing the nutritional value of crops and may also be hazardous for biological agents. Therefore, the use of Plant Growth Promoting Rhizobacteria (PGPR) are favorable for improved crop production over chemical fertilizers.
 Objectives: The current study highlights the growth promoting traits of bacterial isolates through isolation of rhizospheric bacterial strains from different plants.
 Methodology: In this study, ten rhizospheric bacterial isolates were used, which were morpho-physiologically characterized and then tested for plant growth-promoting traits i.e., HCN production, ammonification and auxin production. Most of the bacterial strains gave positive results for these plant growth-promoting traits. To study the beneficial effects of these bacteria on plants, plant-microbial interaction assay was conducted using Zea mays.
 Results: Results revealed that these bacteria enhanced the growth as compared to control plants. Bacterial isolates Streptomyces lydicus (Cn6), Staphylococcus aureus (Cn7) and Bacillus pumilus (PP3) showed strong ammonia producing effects. The isolates Bacillus subtilis (Cn2), PP2 and PP5 exhibited strong potential of HCN production whereas only Streptomyces lydicus (Cn6) and Bacillus pumilus (PP3) were observed to be auxin producers. A maximum increase in fresh weight of the plants was observed in treatment with PP2 showing 94.36% increase over controls. Cn1 showed an increase (26.12%) in shoot length while Cn5 revealed a prominent increase (64.95%) in root length compared to the control plant. The isolates Cn5 and Cn4 showed improvement in the total chlorophyll content of the treated plants with a percentage increase of 100% and 99.82%, respectively compared to the control.
 Conclusion: In conclusion, these PGPR may be further used in agriculture research for growth improvement.

Phytostabilization of Polluted Military Soil Supported by Bioaugmentation with PGP-Trace Element Tolerant Bacteria Isolated from Helianthus petiolaris

Lead (Pb) and cadmium (Cd) are major environmental pollutants, and the accumulation of these elements in soils and plants is of great concern in agricultural production due to their toxic effects on crop growth. Also, these elements can enter into the food chain and severely affect human and animal health. Bioaugmentation with plant growth-promoting bacteria (PGPB) can contribute to an environmentally friendly and effective remediation approach by improving plant survival and promoting element phytostabilization or extraction under such harsh conditions. We isolated and characterised Pb and Cd-tolerant root-associated bacteria from Helianthus petiolaris growing on a Pb/Cd polluted soil in order to compose inoculants that can promote plant growth and also ameliorate the phytostabilization or phytoextraction efficiency. One hundred and five trace element-tolerant rhizospheric and endophytic bacterial strains belonging to eight different genera were isolated from the aromatic plant species Helianthus petiolaris. Most of the strains showed multiple PGP-capabilities, ability to immobilise trace elements on their cell wall, and promotion of seed germination. Bacillus paramycoides ST9, Bacillus wiedmannii ST29, Bacillus proteolyticus ST89, Brevibacterium frigoritolerans ST30, Cellulosimicrobium cellulans ST54 and Methylobacterium sp. ST85 were selected to perform bioaugmentation assays in greenhouse microcosms. After 2 months, seedlings of sunflower (H. annuus) grown on polluted soil and inoculated with B. proteolyticus ST89 produced 40% more biomass compared to the non-inoculated control plants and accumulated 20 % less Pb and 40% less Cd in the aboveground plant parts. In contrast, B. paramycoides ST9 increased the bioaccumulation factor (BAF) of Pb three times and of Cd six times without inhibiting plant growth. Our results indicate that, depending on the strain, bioaugmentation with specific beneficial bacteria can improve plant growth and either reduce trace element mobility or enhance plant trace element uptake.

Plant Growth Promoting Attributes and Antimicrobial Potential of Rhizospheric Bacterial Strain Asp49 Isolated From Juna Amrapar, Near Little Rann of Kutch, Gujarat, India

Rhizospheric bacteria are known to possess plant growth promoting properties. In this study, the bacterium was isolated from rhizospheric soil of plant Salvadora persica L. collected from Juna Amrapar, Little Rann of Kutch, Gujarat (India). The isolate was tested for the properties including nitrogen fixation, phosphate solubilization, indole acetic acid production, siderophore production and ammonia production. In addition, the salt profile of bacteria indicated its tolerance up to 12% NaCl (w/v) on starch agar. The antibiotic susceptibility test revealed that the bacterial strain Asp49 was resistant to Novobiocin (5 μg), Erythromycin (15 μg), Amikacin (30 μg), Teicoplanin (10 μg), Methicillin (5 μg) and sensitivity to Tetracycline (30 μg), Chloramphenicol (30 μg). On the whole, based on in-vitro studies, the ability of selected strain as plant growth promoter in the saline desert conditions of Kutch district is confirmed.
