Mechanisms and Applications of Pseudomonas monteilii SX001: A Promising Agent for Improving Cucumber Tolerance to Salt Stress

To investigate the effects of Pseudomonas monteilii SX001 on various parameters of cucumber plants under salt stress, the salt-sensitive cucumber variety “Jinyou No. 4” was used as the test material, and coconut bran was used to simulate salt stress by applying NaCl solution. The results indicated that salt stress significantly reduced the morphological structure, relative growth rate, root morphology, and photosynthetic parameters of the cucumber plants. Leaf starch, soluble sugar, and sucrose contents significantly increased, whereas their levels in roots decreased. Cell membrane damage leads to the accumulation of reactive oxygen species and malondialdehyde, with notable increases in the activities of major antioxidant enzymes such as SOD, CAT, and POD. Nitrogen metabolism was disrupted, as evidenced by a significant decrease in nitrate nitrogen content and an increase in ammonium nitrogen content, as well as a significant reduction in the activity of NR enzymes involved in nitrogen metabolism. The enzyme activity in the cucumber rhizosphere soil decreased. However, Pseudomonas monteilii SX001 significantly enhanced the growth of cucumber seedlings under salt stress, improved photosynthetic efficiency, and facilitated sugar transformation and transport via glucose metabolism. Additionally, Pseudomonas monteilii SX001 reduced the reactive oxygen content and increased antioxidant enzyme activity. It also increased the activity of substrate enzymes and decreased the diversity of rhizosphere soil microorganisms but also increased the abundance of Asticcacaulis, Acinetobacter, Brevundimonas, Pseudomonas, and Enterobacter. These findings demonstrate that Pseudomonas monteilii SX001 is a promising bioinoculant for alleviating salt stress in cucumber production and improving soil health.

Impact of alternate partial root-zone irrigation on the rhizosphere microbiota of alfalfa plants inoculated with rhizobia.

Water is an important constraint on alfalfa (Medicago sativa) production in arid and semiarid areas, and alternate irrigation in root areas has water-saving potential for alfalfa production. To investigate the impact of alternate partial root-zone irrigation (APRI) on the rhizosphere soil microorganisms of alfalfa, this study subjected alfalfa plants to different irrigation methods and irrigation levels. The growth status and rhizosphere soil microbial community diversity of alfalfa plants under alternate root-zone watering treatment were analyzed through laboratory experiments and high-throughput sequencing. The results showed that at soil moisture levels of 80% field moisture capacity (FMC) and 60% FMC, APRI had no significant impact on the biomass or nodule number of alfalfa. However, 40% FMC significantly reduced the individual plant dry weight, chlorophyll content, and nodule number of the alfalfa plants. APRI increased the relative abundance of Actinomycetes in the alfalfa rhizosphere soil. Moreover, at 60% FMC, the MBC and MBN of rhizosphere, relative abundance of Actinobacteria and unclassified K fungi and Chao 1 index of bacteria significantly increased under APRI treatment. While relative abundance of Ascomycetes and Proteobacteria in the alfalfa rhizosphere significantly reduced under 60% FMC + APRI treatment. In summary, under the same irrigation conditions, APRI did not significantly affect the growth of alfalfa in the short term. And 60%FMC + APRI treatment did significantly affect the groups, structure and diversity of the rhizosphere soil microbial communities.

Effects of nitrogen application on ammonium assimilation and microenvironment in the rhizosphere of drip-irrigated sunflower under plastic mulch.

This study investigated the effect of nitrogen application on the rhizosphere soil microenvironment of sunflower and clarified the relationship between ammonium assimilation and the microenvironment. In a field experiment high (HN, 190 kg/hm2), medium (MN, 120 kg/hm2) and low nitrogen (CK, 50 kg/hm2) treatments were made to replicate plots of sunflowers using drip irrigation. Metagenomic sequencing was used to analyze the community structure and functional genes involved in the ammonium assimilation pathway in rhizosphere soil. The findings indicated that glnA and gltB played a crucial role in the ammonium assimilation pathway in sunflower rhizosphere soil, with Actinobacteria and Proteobacteria being the primary contributors. Compared with CK treatment, the relative abundance of Actinobacteria increased by 15.57% under MN treatment, while the relative abundance decreased at flowering and maturation stages. Conversely, the relative abundance of Proteobacteria was 28.57 and 61.26% higher in the MN treatment during anthesis and maturation period, respectively, compared with the CK. Furthermore, during the bud stage and anthesis, the abundance of Actinobacteria, Proteobacteria, and their dominant species were influenced mainly by rhizosphere soil EC, ammonium nitrogen (-N), and nitrate nitrogen (-N), whereas, at maturity, soil pH and -N played a more significant role in shaping the community of ammonium-assimilating microorganisms. The MN treatment increased the root length density, surface area density, and root volume density of sunflower at the bud, flowering, and maturity stages compared to the CK. Moreover, root exudates such as oxalate and malate were positively correlated with the dominant species of Actinobacteria and Proteobacteria during anthesis and the maturation period. Under drip irrigation, applying 120 kg/hm2 of nitrogen to sunflowers effectively promoted the community structure of ammonium-assimilating microorganisms in rhizosphere soil and had a positive influence on the rhizosphere soil microenvironment and sunflower root growth.

Changes in Rhizosphere Soil Microorganisms and Metabolites during the Cultivation of Fritillaria cirrhosa.

Fritillaria cirrhosa is an important cash crop, and its industrial development is being hampered by continuous cropping obstacles, but the composition and changes of rhizosphere soil microorganisms and metabolites in the cultivation process of Fritillaria cirrhosa have not been revealed. We used metagenomics sequencing to analyze the changes of the microbiome in rhizosphere soil during a three-year cultivation process, and combined it with LC-MS/MS to detect the changes of metabolites. Results indicate that during the cultivation of Fritillaria cirrhosa, the composition and structure of the rhizosphere soil microbial community changed significantly, especially regarding the relative abundance of some beneficial bacteria. The abundance of Bradyrhizobium decreased from 7.04% in the first year to about 5% in the second and third years; the relative abundance of Pseudomonas also decreased from 6.20% in the first year to 2.22% in the third year; and the relative abundance of Lysobacter decreased significantly from more than 4% in the first two years of cultivation to 1.01% in the third year of cultivation. However, the relative abundance of some harmful fungi has significantly increased, such as Botrytis, which increased significantly from less than 3% in the first two years to 7.93% in the third year, and Talaromyces fungi, which were almost non-existent in the first two years of cultivation, significantly increased to 3.43% in the third year of cultivation. The composition and structure of Fritillaria cirrhosa rhizosphere metabolites also changed significantly, the most important of which were carbohydrates represented by sucrose (48.00-9.36-10.07%) and some amino acid compounds related to continuous cropping obstacles. Co-occurrence analysis showed that there was a significant correlation between differential microorganisms and differential metabolites, but Procrustes analysis showed that the relationship between bacteria and metabolites was closer than that between fungi and metabolites. In general, in the process of Fritillaria cirrhosa cultivation, the beneficial bacteria in the rhizosphere decreased, the harmful bacteria increased, and the relative abundance of carbohydrate and amino acid compounds related to continuous cropping obstacles changed significantly. There is a significant correlation between microorganisms and metabolites, and the shaping of the Fritillaria cirrhosa rhizosphere's microecology by bacteria is more relevant.

Metagenomic analysis of soil microbial communities associated with Poa alpigena Lindm in Haixin Mountain, Qinghai Lake.

To investigate the impact of Poa alpigena Lindm on rhizosphere and bulk soil microorganisms in Haixin Mountain, Qinghai Lake, this study employed metagenomics technology to analyze the microbial communities of the samples. Results showed that 65 phyla, 139 classes, 278 orders, 596 families, 2376 genera, and 5545 species of soil microorganisms were identified from rhizosphere and bulk soil samples. Additionally, a microbial gene library specific to Poa alpigena Lindm was established for Qinghai Lake. Through α-diversity analysis, the richness and diversity of bulk microorganisms both significantly had a higher value than that in rhizosphere soil. The indicator microorganisms of rhizosphere and bulk soil at class level were Actinobacteria and Alphaproteobacteria, respectively. KEGG pathway analysis indicated that Carotenoid biosynthesis, Starch and sucrose metabolism, Bacterial chemotaxis, MAPK signaling pathway, Terpenoid backbone biosynthesis, and vancomycin resistance were the key differential metabolic pathways of rhizosphere soil microorganisms; in contrast, in bulk soil, the key differential metabolic were Benzoate degradation, Glycolysis gluconeogenesis, Aminobenzoate degradation, ABC transporters, Glyoxylate and dicarboxylate metabolism, oxidative phosphorylation, Degradation of aromatic compounds, Methane metabolism, Pyruvate metabolism and Microbial metabolism diverse environments. Our results indicated that Poa alpigena Lindm rhizosphere soil possessed selectivity for microorganisms in Qinghai Lake Haixin Mountain, and the rhizosphere soil also provided a suitable survival environment for microorganisms.

The effect of white grub (Maladera Verticalis) larvae feeding on rhizosphere microbial characterization of aerobic rice (Oryza sativa L.) in Puer City, Yunnan Province, China

BackgroundRhizosphere microorganisms are vital in plants’ growth and development and these beneficial microbes are recruited to the root-zone soil when experiencing various environmental stresses. However, the effect of white grub (Maladera verticalis) larvae feeding on the structure and function of rhizosphere microbial communities of aerobic rice (Oryza sativa L.) is unclear.ResultsIn this study, we compared physicochemical properties, enzyme activities, and microbial communities using 18 samples under healthy and M. verticalis larvae-feeding aerobic rice rhizosphere soils at the Yunnan of China. 16 S rRNA and ITS amplicons were sequenced using Illumina high throughput sequencing. M. verticalis larvae feeding on aerobic rice can influence rhizosphere soil physicochemical properties and enzyme activities, which also change rhizosphere microbial communities. The healthy and M. verticalis larvae-feeding aerobic rice rhizosphere soil microorganisms had distinct genus signatures, such as possible_genus_04 and Knoellia genera in healthy aerobic rice rhizosphere soils and norank_f__SC − I−84 and norank_f__Roseiflexaceae genera in M. verticalis larvae-feeding aerobic rice rhizosphere soils. The pathway of the metabolism of terpenoids and polyketides and carbohydrate metabolism in rhizosphere bacteria were significantly decreased after M. verticalis larvae feeding. Fungal parasite–wood saprotroph and fungal parasites were significantly decreased after M. verticalis larvae feeding, and plant pathogen–wood saprotroph and animal pathogen–undefined saprotroph were increased after larvae feeding. Additionally, the relative abundance of Bradyrhizobium and Talaromyces genera gradually increased with the elevation of the larvae density. Bacterial and fungal communities significantly correlated with soil physicochemical properties and enzyme activities, respectively.ConclusionsBased on the results we provide new insight for understanding the adaptation of aerobic rice to M. verticalis larvae feeding via regulating the rhizosphere environment, which would allow us to facilitate translation to more effective measures.

Effects of γ-polyglutamic acid supplementation on alfalfa growth and rhizosphere soil microorganisms in sandy soil

This study aimed at exploring the effects of γ-polyglutamic acid on the growth of desert alfalfa and the soil microorganisms in the rhizosphere. The study examined the effects of varying concentrations of γ-polyglutamic acid (0%-CK, 2%-G1, 4%-G2, 6%-G3) on sandy soil, the research investigated its impact on the growth characteristics of alfalfa, nutrient content in the rhizosphere soil, and the composition of bacterial communities. The results indicated that there were no significant differences in soil organic matter, total nitrogen, total phosphorus, total potassium, and available phosphorus content among the G1, G2, and G3 treatments. Compared to CK, the soil nutrient content in the G2 treatment increased by 14.81–186.67%, showing the highest enhancement. In terms of alfalfa growth, the G2 treatment demonstrated the best performance, significantly increasing plant height, chlorophyll content, above-ground biomass, and underground biomass by 54.91–154.84%. Compared to the CK treatment, the number of OTUs (operational taxonomic units) in the G1, G2, and G3 treatments increased by 14.54%, 8.27%, and 6.84%, respectively. The application of γ-polyglutamic acid altered the composition and structure of the bacterial community, with Actinobacteriota, Proteobacteria, Chloroflexi, Acidobacteriota, and Gemmatimonadota accounting for 84.14–87.89% of the total bacterial community. The G2 treatment significantly enhanced the diversity and evenness of soil bacteria in the rhizosphere. Redundancy analysis revealed that organic matter, total nitrogen, total potassium, moisture content, and pH were the primary factors influencing the structure of bacterial phyla. At the genus level, moisture content emerged as the most influential factor on the bacterial community. Notably, moisture content exhibited a strong positive correlation with Acidobacteriota, which in turn was positively associated with indicators of alfalfa growth. In summary, the application of γ-polyglutamic acid at a 4% ratio has the potential for improving sandy soil quality, promoting plant growth, and regulating the rhizosphere microbial community.

Biochar's role in improving pakchoi quality and microbial community structure in rhizosphere soil.

Biochar amendments enhance crop productivity and improve agricultural quality. To date, studies on the correlation between different amounts of biochar in pakchoi (Brassica campestris L.) quality and rhizosphere soil microorganisms are limited, especially in weakly alkaline soils. The experiment was set up to explore the effect of different concentrations of biochar on vegetable quality and the correlation between the index of quality and soil bacterial community structure changes. The soil was treated in the following ways via pot culture: the blank control (CK) without biochar added and with biochar at different concentrations of 1% (T1), 3% (T2), 5% (T3), and 7% (T4). Here, we investigatedthe synergistic effect of biochar on the growth and quality of pakchoi, soil enzymatic activities, and soil nutrients. Microbial communities from pakchoi rhizosphere soil were analyzed by Illumina MiSeq. The results revealed that adding 3% biochar significantly increased plant height, root length, and dry weight of pakchoi and increased the contents of soluble sugars, soluble proteins, Vitamin C (VC), cellulose, and reduced nitrate content in pakchoi leaves. Meanwhile, soil enzyme activities and available nutrient content in rhizosphere soil increased. This study demonstrated that the the microbial community structure of bacteria in pakchoi rhizosphere soil was changed by applying more than 3% biochar. Among the relatively abundant dominant phyla, Gemmatimonadetes, Anaerolineae, Deltaproteobacteria and Verrucomicrobiae were reduced, and Alphaproteobacteria, Gammaproteobacteria, Bacteroidia, and Acidimicrobiia relative abundance increased. Furthermore, adding 3% biochar reduced the relative abundance of Gemmatimonas and increased the relative abundances of Ilumatobacter, Luteolibacter, Lysobacter, Arthrobacter, and Mesorhizobium. The nitrate content was positively correlated with the abundance of Gemmatimonadetes, and the nitrate content was significantly negatively correlated with the relative abundance of Ilumatobacter. Carbohydrate transport and metabolism in the rhizosphere soil of pakchoi decreased, and lipid transport and metabolism increased after biochar application. Overall, our results indicated that applying biochar improved soil physicochemical states and plant nutrient absorption, and affected the abundance of dominant bacterial groups (e.g., Gemmatimonadetes and Ilumatobacter), these were the main factors to increase pakchoi growth and promote quality of pakchoi. Therefore, considering the growth, quality of pakchoi, and soil environment, the effect of using 3% biochar is better.

Analysis on metabolic functions of rhizosphere microbial communities of Pinus massoniana provenances with different carbon storage by Biolog Eco microplates.

Rhizosphere microorganisms are influenced by vegetation. Meanwhile, they respond to vegetation through their own changes, developing an interactive feedback system between microorganisms and vegetation. However, it is still unclear whether the functional diversity of rhizosphere soil microorganisms varies with different carbon storage levels and what factors affect the functional diversity of rhizosphere soil microorganisms. In this study, the Biolog-Eco microplate technique was used to analyze the metabolic diversity of carbon source of rhizosphere soil microorganisms from 6 Pinus massoniana provenances with three levels of high, medium and low carbon storage. The results showed that the average well color development(AWCD) value of rhizosphere microorganisms was significantly positive correlated with carbon storage level of Pinus massoniana (p < 0.05). The AWCD value, Simpson and Shannon diversity of high carbon sequestrance provenances were 1.40 (144h incubation) 0.96 and 3.24, respectively, which were significantly higher (p < 0.05) than those of other P. massoniana provenances. The rhizosphere microbial AWCD, Shannon and Simpson diversity of the 6 provenances showed the same variation trend (SM>AY>QJ>SX>HF>SW). Similarly, microbial biomass carbon (MBC) content was positively correlated with carbon storage level, and there were significant differences among high, medium and low carbon storage provenances. The PCA results showed that the differences in the carbon source metabolism of rhizosphere microorganisms were mainly reflected in the utilization of amino acids, carboxylic acids and carbohydrates. Pearson correlation analysis showed that soil organic carbon (SOC), total nitrogen (TN) and pH were significantly correlated with rhizosphere AWCD (p < 0.05). Soil properties are important factors affecting rhizosphere microbial carbon source metabolism. The study confirmed that the microorganisms of high carbon storage provenances had relatively high carbon metabolic activity. Among them, the carbon metabolic activity of rhizosphere microorganisms of SM provenance was the highest, which was the preferred provenances in effective ecological service function.

Variations in rhizosphere soil dominant and pathogenic flora improve boron-efficient Beta vulgaris L. yield under boron deficit

Boron (B)-efficient sugarbeet can potentially adapt better to the B deficient soil environments, however the characteristics of root yield or quality and rhizosphere soil microbial dynamics associated with B-efficient beet roots are extremely understudied. This study aimed to clarify the root yield characteristics and rhizosphere microbial response of B-efficient sugarbeet variety under B deficiency. Pertinently, the findings of this study will contribute to the reduction of B resource consumption and cleaner production in sugarbeet planting management. The study utilized two sugarbeet varieties with different B utilization propensities and subjected them to normal and B deficient field conditions. Results demonstrated a reduction in the yield quality of both varieties under B deficiency, but the B-efficient variety exhibited a smaller reduction. Boron deficiency reduced the species richness and diversity of the rhizospheric microbial community, decreasing the relative abundance of Actinobacteriota, Acidobacteria, while increasing that of Proteobacteria, Chloroflexi and the pathogenic fungus Fusarium. The proportion of dominant bacterial was severely reduced in B-inefficient varieties (0.86%) compared to B-efficient varieties (0.14%). The efficient variety displayed higher species richness of rhizospheric soil fungi and a higher abundance and proportion of dominant flora, i.e., Bacillus and Actinobacteria, albeit lower abundance of the pathogenic fungi Mortierellomycota and Rhizoctonia. These advantages allowed B-efficient sugarbeet variety to maintain higher yield levels under B deficit. Overall, under B-deficient conditions, B-efficient variety of sugarbeet was less limited than B-inefficient one, and its rhizosphere microbial communities were more conducive to sugarbeet yield in terms of composition and function. Succinctly, this study revealed the advantages of rhizosphere soil microorganisms associated with B-efficient beet under B-deficient environments and provided the theoretical basis for reducing boron fertilizer application to protect the ecosystem and realize green and clean agricultural production, particularly of sugar beet.

Response of rhizosphere soil physicochemical properties and microbial community structure to continuous cultivation of tobacco

PurposeThe health of rhizosphere soil microorganisms is an important indicator to evaluate soil quality. Therefore, understanding the response of rhizosphere soil microorganisms to tobacco crop succession is crucial for promoting the sustainable development of agriculture.MethodsThe microbial diversity and community structure of rhizosphere soil in continuous cropping and non-cropped tobacco for 7 years were analyzed by the Illumina platform.Result(1) Continuous cropping tobacco cause rhizosphere soil acidification and reduction in alkaline nitrogen (AN) and soil organic matter (SOM). (2) Continuous cropping tobacco reduces the diversity of rhizosphere soil microbial communities, increasing harmful functional microorganisms and declining beneficial ones. (3) The abundance of bacteria that perform nitrification and saprophytic fungi in the rhizosphere soil of continuous cropping areas decreases, inhibiting carbon and nitrogen cycling processes. (4) The composition and diversity of the soil rhizosphere microbial community are affected by the imbalance in the physicochemical property of the rhizosphere.ConclusionContinuous cropping tobacco cause rhizosphere soil acidification and nutrient imbalance, and the carbon and nitrogen cycles involved in microorganisms were damaged. Furthermore, the decreased diversity of rhizosphere soil microorganisms and the increased abundance of pathogenic fungi contribute to the continuous cropping obstacles of tobacco.

Rhizosphere microbial community changes due to weed-weed competition

Many species of weeds are present in agricultural areas, but weeds with greater competitive ability normally become dominant in the field. Rhizosphere soil microbiota can influence weed-weed interactions. However, the role of rhizosphere soil microorganisms in weed-weed interactions remains largely unexplored. In this study, we investigated the ecological relationships and microbial taxa present in the rhizosphere of weeds in monoculture and coexistence systems. The weed species Ageratum conyzoides, Ipomoea ramosissima, and Bidens pilosa were grown in monoculture and coexistence pairs under greenhouse conditions for 80 days. The ecological relationships between weeds were analyzed by calculating the Relative Interaction Index (RII) based on the total dry mass of the plants. The rhizosphere microbiome was analyzed after extracting the metagenomic DNA from rhizosphere microbial populations, followed by PCR amplification and sequencing of the 16S rDNA gene and ITS region, using the Illumina MiSeq platform. Competitive interactions were observed for all combinations of weed species. Ageratum conyzoides showed the greatest decrease in dry matter production due to competition. Weed-weed competition changed rhizosphere microbial community composition and bacterial diversity. The abundance of different bacterial genera in rhizosphere soil varied according to the treatments. When comparing the competition between A. conyzoides and B. pilosa and their respective monocultures, the abundances of Opitutus, Diplorickettisiales uncultured and Bdellovibrio increased in B. pilosa monoculture. When comparing the competition between A. conyzoides and I. ramosissima and their respective monocultures, the abundance of Pseudonocardia increased while the abundance of Fimbriiglobus decreased in A. conyzoides monoculture. Already the abundances of Actinospica, Chitinophaga, Gemmatirosa, 1921-2 and Hymenobacter decreased I. ramosissima monoculture. When comparing the competition between B. pilosa and I. ramosissima, and their respective monocultures, the abundances of Flexibacter and Candidatus Xiphinematobacter decreased I. ramosissima monoculture. The abundances of Clostridium and Rhodobacter increased in competition while the abundance of Pajaroellobacter increased in B. pilosa monoculture. The fungal genera that had their abundances significantly altered were Scytalidium, whose abundance decreased in competition of A. conyzoides and B. pilosa when compared to the respective monocultures, while the abundance of Robillarda increased in B. pilosa monoculture. Already the abundance of Myriodontium increased in B. pilosa monoculture when compared with I. ramosissima monoculture and competition between both plants. Weeds can recruit specific taxa depending on the competition situation in which they are involved. Such changes in the rhizobiome are possibly related to the lower competitive ability and dry matter production of A. conyzoides. B. pilosa have a greater capacity to determine the structure of the rhizobiome when in competition with A. conyzoides. I. ramosissima seem to be less affected by rhizobiome taxon-specific changes.

Effects of coastal saline-alkali soil on rhizosphere microbial community and crop yield of cotton at different growth stages.

Soil salinization is a global constraint that significantly hampers agricultural production, with cotton being an important cash crop that is not immune to its detrimental effects. The rhizosphere microbiome plays a critical role in plant health and growth, which assists plants in resisting adverse abiotic stresses including soil salinization. This study explores the impact of soil salinization on cotton, including its effects on growth, yield, soil physical and chemical properties, as well as soil bacterial community structures. The results of β-diversity analysis showed that there were significant differences in bacterial communities in saline-alkali soil at different growth stages of cotton. Besides, the more severity of soil salinization, the more abundance of Proteobacteria, Bacteroidota enriched in rhizosphere bacterial composition where the abundance of Acidobacteriota exhibited the opposite trend. And the co-occurrence network analysis showed that soil salinization affected the complexity of soil bacterial co-occurrence network. These findings provide valuable insights into the mechanisms by which soil salinization affects soil microorganisms in cotton rhizosphere soil and offer guidance for improving soil salinization using beneficial microorganisms.

Stability and carbon uptake of the soil microbial community is determined by differences between rhizosphere and bulk soil

The interactions between plants and soil microorganisms are fundamental for ecosystem functioning. However, it remains unclear if seasonality of plant growth impacts plant-microbial interactions, such as by inducing shifts in the microbial community composition, their biomass, or changes in the microbial uptake of plant-derived carbon. Here, we investigated the stability of the microbial community and their net assimilation of plant-derived carbon over an entire growing season. Using a C3–C4 vegetation change experiment, and taking advantage of a natural 13C label, we measured the plant-derived carbon in lipid biomarkers of soil microorganisms in rhizosphere and bulk soil in two soils with contrasting textures. We found that temporal stability was higher in bacterial than in fungal biomass, whereas the spatial stability of the fungal biomass was higher than that of bacterial biomass. Moreover, symbiotic AM fungi tended to be more stable in the uptake of plant-derived carbon than bacteria and saprophytic fungi. While soil texture did influence microbial community composition as expected, it had no effect on the microbial plant carbon assimilation and the differences between rhizosphere and bulk soil. In addition, the putative differences in carbon utilization between microbial groups, with the exception of AM fungi, were generally smaller than expected, reflecting opportunistic utilization of energy sources. Our results suggest that microbial uptake of plant carbon is primarily limited by plant carbon allocation rather than by environmental factors such as soil texture and seasonality. This indicates that the ongoing carbon assimilation during the growing season is supported by a functional redundancy within the microbial community, which, in turn, helps sustain ecosystem functioning.

Rare and abundant bacterial communities in poplar rhizosphere soils respond differently to genetic effects

Interactions between plants and soil microbes are important to plant hybrid breeding under global change. However, the relationship between host plants and rhizosphere soil microorganisms has not been fully elucidated. Understanding the rhizosphere microbial structure of parents and progenies would provide a deeper insight into how genetic effects modulate the relationship between plants and soil. In this study, two family groups of poplar trees (A: parents and their two progenies; B: parents and their one progeny) with different genetic backgrounds (including seven genotypes) were selected from a common garden, and their rhizobacterial communities were analyzed to explore parent-progeny relationships. Our results showed significant differences in phylogenetic diversity, the number of 16S genes and the structure of rhizosphere bacterial communities (Adonis: R2 = 0.166, P < 0.01) between different family groups. Rhizosphere bacterial community structure was significantly dominated by genetic effects. Compared with abundant taxa, genetic effects were more powerful drivers of rare taxa. In addition, bacterial communities of hybrid progenies were all significantly more similar to their parents compared to the other group of parents, especially among rare taxa. The two poplar family groups exhibited differences between their rhizosphere bacterial co-occurrence networks. Group B had a relatively complex network with 2380 edges and 468 nodes, while group A had 1829 edges and 304 nodes. Soil organic carbon and carbon to nitrogen ratio (C/N) also influenced the rhizosphere bacterial community assembly. This was especially true for soil C/N, which explained 23 % of the β-nearest taxon index (βNTI) variation in rare taxa. Our results reveal the relationship of rhizosphere microorganisms between parents and progenies. This can help facilitate an understanding of the combination of plant breeding with microbes resource utilization and provide a theoretical basis for scientific advancement to support the development of forestry industry.

Correlation between rhizosphere environment and content of medicinal components of Arnebia euchroma

This study aimed to explore the correlation between rhizosphere soil microorganisms of wild Arnebia euchroma and the content of medicinal components to provide guidance for the selection of the ecological planting base. The total DNA of rhizosphere soil microorganisms of wild A. euchroma was extracted, and the microbial community structure of rhizosphere soil microorganisms was analyzed by IlluminaMiseq high-throughput sequencing technology. The content of total hydroxynaphthoquinone pigment and β,β'-dimethylacrylalkannin in medicinal materials was determined by high-performance liquid chromatography(HPLC). The physicochemical pro-perties of rhizosphere soil of wild A. euchroma in main producing areas were determined, and the correlation of soil microbial abundance with index component content and soil physicochemical properties was analyzed by SPSS software. The results showed that the species composition of rhizosphere fungi and bacteria in A. euchroma from different habitats was similar at the phylum and genus levels, but their relative abundance, richness index(Chao1), and community diversity(Simpson) index were different. Correlation analysis showed that the content of available phosphorus in soil was positively correlated with the content of total hydroxynaphthoquinone pigment and β,β'-dimethylacrylalkannin, and the abundance of five fungal genera such as Solicoccozyma and six bacterial genera such as Pseudo-nocardia and Bradyrhizobium was positively correlated with the content of medicinal components in medicinal materials. The abundance of Bradyrhizobium was significantly positively correlated with the content of β,β'-dimethylacrylalkanin. The abundance of fungi such as Archaeorhizomyces was significantly positively correlated with the content of available phosphorus in rhizosphere soil, and Bradyrhizobium was significantly negatively correlated with soil pH. Therefore, the abundance of fungi and bacteria in the rhizosphere of A. euchroma has a certain correlation with the medicinal components and the physicochemical properties of the rhizosphere soil, which can provide a scientific basis for the selection of ecological planting bases in the later stage.

Intervention of rhamnolipid improves the rhizosphere microenvironment of cotton in desert saline lands

Rhamnolipids have been extensively studied for the remediation of soils contaminated with petroleum hydrocarbons. However, the literature on the effects of rhamnolipids on the soil microenvironment is scarce. In this study, we adopted a drip irrigation technique to apply a rhamnolipid solution to soil planted with cotton in a saline desert area. The results showed that the addition of rhamnolipids increased soil organic matter by 5.0–31.6 % and reduced soil electrical conductivity by 0.3–42.4 %. Additionally, it improved soil nutrient conditions, reshaped the composition and function of the microbial community in the rhizosphere soil, and ultimately promoted cotton growth by 3.3 – 9.0 %. Simultaneously, the addition of rhamnolipids enriched the diversity of the bacterial community. Although it reduced the diversity of fungal communities, it optimized the community structure of fungi. It is worth noting that higher concentrations of rhamnolipids may have toxic effects on soil fungi, the mechanism of which is unknown. These findings shed new light on understanding the effects of rhamnolipids on rhizosphere soil microorganisms and provide a new direction for sustainable agricultural production.

Selenium nanomaterials alleviate Brassica chinensis L cadmium stress: Reducing accumulation, regulating microorganisms and activating glutathione metabolism

Agricultural heavy metal contamination can cause significant crop damage, highlighting the urgent need to mitigate its negative effects. Under Cd2+ stress, selenium nanomaterials (Se NMs, 2 mg kg−1) can significantly improve Brassica chinensis L. root growth and vigor, enhance photosynthesis (31.4%), and increase biomass. Se NMs treatment also reduces Brassica chinensis L root and shoot Cd concentration by 67.2 and 72.9%, respectively. This reduction is mainly due to the gene expression of Cd2+ absorption (BcITR1 and BcHMA2) which was down-regulated 51.9 and 67.0% by Se NMs, respectively. Meanwhile, Se NMs can increase the abundance of Cd-resistant microorganisms (Gemmatimonas, RB41, Haliangium, Gaiella, and Steroidobacter) in rhizosphere soil while also reducing Cd migration from soil to plants. Additionally, Se NMs also contribute to reducing ROS accumulation by improving the oxidation-reduction process between GSH and GSSG through enhancing γ-ECS (15.6%), GPx (50.2%) and GR (97.3%) activity. Remarkably, crop Se content can reach 50.8 μg/100 g, which fully meets the standards of Se-rich vegetables. These findings demonstrate the potential of Se NMs in relieving heavy metal stress, while simultaneously increasing crop Se content, making it a promising technology for sustainable agricultural production.

Geographical, climatic, and soil factors control the altitudinal pattern of rhizosphere microbial diversity and its driving effect on root zone soil multifunctionality in mountain ecosystems

Shifts in rhizosphere soil microorganisms of dominant plants' response to climate change profoundly impact mountain soil ecosystem multifunctionality; relatively little is known about the relationship between them and how they depend on long-term environmental drivers. Here, we conducted analyses of rhizosphere microbial altitudinal pattern, community assembly, and co-occurrence network of 6 dominant plants in six typical vegetation zones ranging from 1350 to 2900 m (a.s.l.) in Helan Mountains by absolute quantitative sequencing technology, and finally related the microbiomes to root zone soil multifunctionality (‘soil multifunctionality’ hereafter), the environmental dependence of the relationship was explored. It was found that the altitudinal pattern of rhizosphere soil bacterial and fungal diversities differed significantly. Higher co-occurrence and more potential interactions of Stipa breviflora and Carex coninux were found at the lowest and highest altitudes. Bacterial α diversity, the identity of some dominant bacterial and fungal taxa, had significant positive or negative effects on soil multifunctionality. The effect sizes of positive effects of microbial diversity on soil multifunctionality were greater than those of negative effects. These results indicated that the balance of positive and negative effects of microbes determines the impact of microbial diversity on soil multifunctionality. As the number of microbes at the phylum level increases, there will be a net gain in soil multifunctionality. Our study reveals that geographical and climatic factors can directly or modulate the effects of soil properties on rhizosphere microbial diversity, thereby affecting the driving effect of microbial diversity on soil multifunctionality, and points to the rhizosphere bacterial diversity rather than the fungi being strongly associated with soil multifunctionality. This work has important ecological implications for predicting how multiple environment-plant-soil-microorganisms interactions in mountain ecosystems will respond to future climate change.

Enzymatic fermentation of rapeseed cake significantly improved the soil environment of tea rhizosphere

BackgroundRapeseed cake is an important agricultural waste. After enzymatic fermentation, rapeseed cake not only has specific microbial diversity but also contains a lot of fatty acids, organic acids, amino acids and their derivatives, which has potential value as a high-quality organic fertilizer. However, the effects of fermented rapeseed cake on tea rhizosphere microorganisms and soil metabolites have not been reported. In this study, we aimed to elucidate the effect of enzymatic rapeseed cake fertilizer on the soil of tea tree, and to reveal the correlation between rhizosphere soil microorganisms and nutrients/metabolites.ResultsThe results showed that: (1) The application of enzymatic rapeseed cake increased the contents of soil organic matter (OM), total nitrogen (TN), total phosphorus (TP), available nitrogen (AN), and available phosphorus (AP); increased the activities of soil urease (S-UE), soil catalase (S-CAT), soil acid phosphatase (S-ACP) and soil sucrase (S-SC); (2) The application of enzymatic rapeseed cake increased the relative abundance of beneficial rhizosphere microorganisms such as Chaetomium, Inocybe, Pseudoxanthomonas, Pseudomonas, Sphingomonas, and Stenotrophomonas; (3) The application of enzymatic rapeseed cake increased the contents of sugar, organic acid, and fatty acid in soil, and the key metabolic pathways were concentrated in sugar and fatty acid metabolisms; (4) The application of enzymatic rapeseed cake promoted the metabolism of sugar, organic acid, and fatty acid in soil by key rhizosphere microorganisms; enzymes and microorganisms jointly regulated the metabolic pathways of sugar and fatty acids in soil.ConclusionsEnzymatic rapeseed cake fertilizer improved the nutrient status and microbial structure of tea rhizosphere soil, which was beneficial for enhancing soil productivity in tea plantations. These findings provide new insights into the use of enzymatic rapeseed cake as an efficient organic fertilizer and expand its potential for application in tea plantations.
