Discovering oxidized polar lipids in microalgae lipidome using liquid chromatography mass spectrometry approaches

Microalgae are rich in polar lipids, such as glycolipids, phospholipids and betaine lipids esterified with omega-3 polyunsaturated fatty acids (PUFA), which are prone to oxidation. Under stress conditions, the redox balance in microalgae is altered, leading to an abnormal production of reactive oxygen species (ROS) that can affect surrounding lipids. While few oxidized polar lipids have been described to play important roles in mammals and possess interesting bioactive properties, their occurrence in microalgae is poorly described, hindering their exploitation as novel bioactive compounds. To enhance our understanding of these lipids, we conducted a targeted analysis of oxidized polar lipids in lipid extracts obtained from five microalgae species: Chlorella vulgaris, Chlorococcum amblystomatis, Scendesmus obliquus, Nanochloropsis oceanica and Phaeodactylum ticornutum, using reverse-phase liquid chromatography mass spectrometry (RP-LC-MS). A detailed analysis of the LC-MS data identified 150 oxidized polar lipid species across different classes of phospholipids, glycolipids and betaine lipids in the five microalgae species. These modified oxidized lipids featured oxygenated species with 1–3 additional oxygen atoms in the fatty acyl chains. The predominant oxidized fatty acids esterified to these lipids were omega-3 eicosapentaenoic acid (EPA, 20:5 n-3) and α-linonelic acid (ALA, 18:3 n-3). Notably, most oxidized lipid species were identified in diacylglyceryl-N,N,N-trimethylhomoserine (DGTS) in C. amblystomatis, S. obliquus and N. oceanica, monogalactosyldiacylglycerol (MGDG) in P. tricornutum, and phosphatidylethanolamine (PE) in C. vulgaris. Furthermore, distinct fragmentation patterns across lipid classes allowed the unequivocal identification of oxidized polar lipids. These findings reveal a diverse array of oxidized polar lipids in microalgae, predominantly enriched with oxidized omega-3 PUFA, highlighting microalgae as a natural source of oxidized polar lipids that may serve as a natural reservoir of bioactive omega-3 oxylipins.

Untargeted plasma metabolomics and risk of colorectal cancer—an analysis nested within a large-scale prospective cohort

BackgroundColorectal cancer (CRC) is a leading cause of cancer-related death worldwide, but if discovered at an early stage, the survival rate is high. The aim of this study was to identify novel markers predictive of future CRC risk using untargeted metabolomics.MethodsThis study included prospectively collected plasma samples from 902 CRC cases and 902 matched cancer-free control participants from the population-based Northern Sweden Health and Disease Study (NSHDS), which were obtained up to 26 years prior to CRC diagnosis. Using reverse-phase liquid chromatography–mass spectrometry (LC–MS), data comprising 5015 metabolic features were obtained. Conditional logistic regression was applied to identify potentially important metabolic features associated with CRC risk. In addition, we investigated if previously reported metabolite biomarkers of CRC risk could be validated in this study population.ResultsIn the univariable analysis, seven metabolic features were associated with CRC risk (using a false discovery rate cutoff of 0.25). Two of these could be annotated, one as pyroglutamic acid (odds ratio per one standard deviation increase = 0.79, 95% confidence interval, 0.70–0.89) and another as hydroxytigecycline (odds ratio per one standard deviation increase = 0.77, 95% confidence interval, 0.67–0.89). Associations with CRC risk were also found for six previously reported metabolic biomarkers of prevalent and/or incident CRC: sebacic acid (inverse association) and L-tryptophan, 3-hydroxybutyric acid, 9,12,13-TriHOME, valine, and 13-OxoODE (positive associations).ConclusionsThese findings suggest that although the circulating metabolome may provide new etiological insights into the underlying causes of CRC development, its potential application for the identification of individuals at higher risk of developing CRC is limited.

#5668 PERITONEAL DIALYSIS ADDITIVE REDUCES TRANSPERITONEAL LOSS OF BLOOD MICROPARTICLE AND EXTRACELLULAR MATRIX PROTEINS IN A CONTROLLED CROSSOVER TRIAL

Abstract Background and Aims About 11% of patients requiring dialysis worldwide are treated with peritoneal dialysis (PD). With each PD exchange high amounts of protein are removed from the peritoneal cavity stemming from the systemic circulation (reflecting toxin removal, loss of serum proteins, inflammatory processes) and local inflammatory processes. Thereby, the clinically measured peritoneal protein loss (PPL) is the result of transperitoneal serum loss, lymphatic drainage, transmesothelial reabsorption, local production, and cellular components. Although high PPL has been found to be predictive of mortality and cardiovascular morbidity, the pathophysiological concept is still poorly understood. Method Peritoneal dialysis effluent (PDE) samples were obtained from peritoneal equilibrium tests during a prospective, multicenter, double-blinded, controlled, randomized, dual-period, 2-treatment, crossover, phase II, proof-of-concept study in Austria. PDE samples were submitted to proteomic analysis by tandem mass tag (TMT) labeling and 2D reversed phase liquid chromatography mass spectrometry. To disentangle transmembrane serum protein loss from local protein production we developed a novel and unique analytical approach. Briefly, partitioning around medoids and proximity in the Euclidean space for between visit (non-treatment visits only) single-protein kinetics was utilized to achieve this separation. A multivariate mixed-effects model was calculated to identify treatment (intraperitoneal alanyl-glutamine administration) effects. Results We identified 2,624 different proteins within the PDE of 12 patients across two time-points. A large cluster of proteins with stable abundances in collinearity with well-established clinical characteristics of transperitoneal transport (e.g., dialysate-to-plasma-albumin) in accordance with the three-pore-model was identified. After clustering and separation, the proteins resembling similar time-course kinetics to clinical transperitoneal membrane transport characteristics (one of five clusters) were all identified to be able to theoretically traverse through the small peritoneal pore (< 4-6 nm). Of these 549 proteins lost from the systemic circulation via transperitoneal small pore diffusion, the appearance of 51 in the PD effluent was significantly reduced by intraperitoneal administration of alanyl-glutamine. The affected proteins are mainly involved in extracellular matrix organization, part of extracellular exosome signaling and blood microparticles, e.g. high density lipoprotein (HDL) formation (Apolipoprotein A1). Conclusion PPL is an important but poorly understood undesired effect of PD associated with cardiovascular morbidity and mortality. Our novel analytical approach enables us to disentangle the complexity of transperitoneal serum protein loss and local protein production on single-protein level across the peritoneal proteome. Furthermore, we were able to delineate the potential beneficial effects of intraperitoneal alanyl-glutamine treatment on PPL and peritoneal health which might even suggest potential beneficial effects on cardiovascular morbidity (e.g., HDL formation).

Metabolites, elemental profile and chemical activities of Pinus strobus high temperature-derived pyroligneous acid

BackgroundPyroligneous acid (PA) is an aqueous smoky fraction produced during pyrolysis of biomass. The chemical composition of PA from different plant biomass has been studied, but reports on PA metabolites and elemental profiles are rare. In this study, we examined the metabolites, elemental profiles and the associated chemical activities of PA derived from white pine (Pinus strobus) at 1100 °C compared to similar work done elsewhere using different biomass at lower temperatures.ResultsPA from P. strobus biomass exhibited a lower electrical conductivity (2.05 mS/cm), salinity (1.03 g/L) and total dissolved solids (1.42 g/L) but higher oBrix content (9.35 ± 0.06) compared to PA from other feedstock. The P. strobus PA showed a higher antioxidant activity characterized by enhanced radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl free-radical (78.52%) and accumulation of higher total phenolic (95.81 ± 1.45 gallic acid equivalents (GAE)/mL) and flavonoid content (49.46 µg quercetin/mL). Metabolite profiling by direct injection mass spectrometry with a reverse-phase liquid chromatography–mass spectrometry (DI/LC–MS/MS) identified a total of 156 metabolites. Four (4) main groups including organic acids (90.87%), hexose (8.60%), carnitine (0.3%) and phospholipids (0.24%) were found in the PA. Mineral element analysis revealed that the P. strobus PA contained high concentrations of nitrogen (N), potassium (K), calcium (Ca) and zinc (Zn), while the content of sodium (Na) and trace/heavy metals were present at levels below the reported limit.ConclusionThis study indicates that P. strobus PA is a valuable product that can be used in agriculture to improve plant growth and productivity under normal and environmentally stressful conditions.Graphical

Purification and Characterization of Novel Antihypertensive and Antioxidative Peptides From Whey Protein Fermentate: In Vitro, In Silico, and Molecular Interactions Studies

Objective The goal of this research was to purify and characterize the novel angiotensin-converting enzyme (ACE)–inhibitory and antioxidant peptides from fermented whey protein concentrate produced by Lactobacillus paracasei and Saccharomyces cerevisiae in a co-fermentation system. Method Whey protein fermented with lactic acid bacteria and yeast culture was analyzed for antioxidative, ACE inhibition, as well as anti-inflammatory activity followed by SDS-PAGE, isoelectric focusing, and 2-dimensional (2D) analysis. Anti-inflammatory activity of whey protein fermentate was also studied on the RAW 264.7 cell line. The bioactive peptides were separated from the whey protein fermentate using reverse-phase high-performance liquid chromatography (RP-HPLC) and reverse-phase liquid chromatography mass spectrometry (RPLC/MS), and thus identification and characterization of purified bioactive peptide was performed. Results Whey protein fermentate samples’ bioactivity was analyzed at specific time intervals at 12, 24, 36, and 48 hours at 37 °C for M11 and at 25 °C for WBS2A. The development settings (incubation time [12, 24, 36, and 48 hours) and inoculation rates [1.5%, 2.0%, and 2.5%]) were optimized for peptide synthesis via the o-phthaldialdehyde (OPA) method (proteolytic activity). Maximum proteolytic activity was observed at 37 °C for M11 (6.50 mg/mL) and at 25 °C for WBS2A (8.59 mg/mL) for 48 hours of incubation. Protein profiling was carried out using SDS-PAGE and 2D gel electrophoresis, in which Sodium dodecyl-sulfate (SDS) exhibited protein bands in the 10- to 55-kDa range, while 2D showed protein bands varying from 10 to 70 kDa. Every spot from 2D was digested by trypsin and identified by RPLC/MS. Protein fractionations (3- and 10-kDa permeates) were carried out employing RP-HPLC. Whey protein fermentate has anti-inflammatory action in RAW 264.7 macrophages that have been exposed to lipopolysaccharide. A molecular docking system was also used to investigate the interactions of peptides (AFLDSRTR, ILGAFIQIITFR) with human myeloperoxidase enzyme. Conclusions The antihypertensive and antioxidative peptides discovered from whey protein fermentate may be helpful in the design of pharmacologically active healthy ingredients in the upcoming years.

Method for absolute quantification of short chain fatty acids via reverse phase chromatography mass spectrometry.

Short chain fatty acids (SCFAs; including acetate, propionate, and butyrate) are an important class of biological molecules that play a major role in modulating host-microbiome interactions. Despite significant research into SCFA-mediated biological mechanisms, absolute quantification of these molecules in their native form by liquid chromatography mass spectrometry is challenging due to their relatively poor chromatographic properties. Herein, we introduce SQUAD, an isotope-based strategy for absolute quantification of SCFAs in complex biological samples. SQUAD uses aniline derivatization in conjunction with isotope dilution and analysis by reverse-phase liquid chromatography mass spectrometry. We show that SQUAD enables absolute quantification of biologically relevant SCFAs in complex biological samples with a lower limit of detection of 40 nM and a lower limit of quantification ranging from 160 nM to 310 nM. We observed an intra- and inter-day precision under 3% (relative standard deviation) and errors in intra- and inter-day accuracy under 10%. To demonstrate this quantification strategy, we analyzed SCFAs in the caecal contents of germ free versus conventionally raised specific pathogen free (SPF) mice. We showed that acetate was the most abundant SCFA in both types of mice and was present at 200-fold higher concentration in the SPF mice. We also illustrated the use of our quantification strategy in in vitro microbial cultures from five different species of bacteria grown in Mueller Hinton media. This study illustrates the diverse SCFA production rates across microbial taxa with acetate production serving as one of the key differentiating factors across the species. In summary, we introduce an isotope dilution strategy for absolute quantification of aniline-dativized SCFAs and illustrate the utility of this approach for microbiome research.

Sex-specific differences in plasma lipid profiles are associated with Gulf War Illness

BackgroundNearly 250,000 veterans from the 1990–1991 Gulf War have Gulf War Illness (GWI), a condition with heterogeneous pathobiology that remains difficult to diagnose. As such, availability of blood biomarkers that reflect the underlying biology of GWI would help clinicians provide appropriate care to ill veterans. In this study, we measured blood lipids to examine the influence of sex on the association between blood lipids and GWI diagnosis.MethodsPlasma lipid extracts from GWI (n = 100) and control (n = 45) participants were subjected to reversed-phase nano-flow liquid chromatography-mass spectrometry analysis.ResultsAn influence of sex and GWI case status on plasma neutral lipid and phospholipid species was observed. Among male participants, triglycerides, diglycerides, and phosphatidylcholines were increased while cholesterol esters were decreased in GWI cases compared to controls. In female participants, ceramides were increased in GWI cases compared to controls. Among male participants, unsaturated triglycerides, phosphatidylcholine and diglycerides were increased while unsaturated cholesterol esters were lower in GWI cases compared to controls. The ratio of arachidonic acid- to docosahexaenoic acid-containing triglyceride species was increased in female and male GWI cases as compared to their sex-matched controls.ConclusionDifferential modulation of neutral lipids and ratios of arachidonic acid to docosahexaenoic acid in male veterans with GWI suggest metabolic dysfunction and inflammation. Increases in ceramides among female veterans with GWI also suggest activation of inflammatory pathways. Future research should characterize how these lipids and their associated pathways relate to GWI pathology to identify biomarkers of the disorder.

Studies on potential biomarkers related to drug efficacy and toxicity of an anti-tumor drug candidate by pharmaco-metabonomics

Based on high-performance liquid chromatography-mass spectrometry (HPLC-MS) and reversed-phase high-performance liquid chromatography-mass spectrometry (RPLC-MS), the changes of drug metabolism and endogenous metabolites in normal rats and tumor model rats were studied by drug metabonomics. The changes of vital signs were used to evaluate the toxicity and pharmacological effects of the new anti-cancer candidate ST on rats, and the differences of endogenous metabolites caused by ST were analyzed by multivariate statistical method. The changes of endogenous metabolites related to ST toxicity and pharmacological effects were analyzed. Studies have shown that the correlation analysis of metabonomics based on metabonomics and changes of animal signs is helpful to screen endogenous metabolites related to drug toxicity and pharmacological effects.

Effect of Diet on the Vitamin B Profile of Bovine Milk-Based Protein Ingredients.

The influence of diet on the water-soluble vitamin composition of skim milk powder and whey protein ingredients produced from the milk of cows fed pasture or concentrate-based diets was examined. Fifty-one Holstein-Friesian cows were randomly assigned into three diets (n = 17) consisting of outdoor grazing of perennial ryegrass (GRS), perennial ryegrass/white clover (CLV), or indoor feeding of total mixed ration (TMR) for an entire lactation. Raw mid-lactation milk from each group was processed into skim milk powder and further processed to yield micellar casein whey and acid whey. Sweet whey was also produced by renneting of pasteurised whole milk from each system. The water-soluble vitamin profile of each sample was analysed using a combination of direct injection mass spectrometry and reverse-phase liquid chromatography–mass spectrometry. Vitamin B3 and B3-amide concentrations were significantly higher (p < 0.05) in TMR-derived samples than in those from CLV and GRS, respectively. Vitamin B1, B2, and B7 concentrations were significantly higher in GRS and CLV-derived samples than those from TMR. Significant differences in vitamins B1, B2, and B3-amide were also observed between protein ingredient types. This study indicates that bovine feeding systems have a significant effect on B vitamin composition across a range of protein ingredient types.

Metabolomic identification of placental alterations in fetal growth restriction

Introduction Fetal growth restriction (FGR), viz., birth weight <10th percentile is a common pregnancy complication which increases the risk of adverse fetal and newborn outcomes. The placenta is the key organ for fetal growth as it controls oxygen and nutrient availability. This study aims to elucidate the mechanisms of and identify putative placental biomarkers for FGR using high-resolution metabolomics. Methods Placenta samples from 19 FGR cases and 30 controls were analyzed using proton magnetic resonance (1H NMR) spectroscopy and direct flow injection mass spectrometry with reverse-phase liquid-chromatography mass spectrometry (DI-LC-MS/MS). Significant concentration differences (p-value <.05) in 179 of the 220 metabolites were measured. Results Of the 179 metabolites, 176 (98.3%) had reduced placental levels in FGR cases. The best performing metabolite model: 3-hydroxybutyrate, glycine and PCaaC42:0 achieved an AUC (95% CI) = 0.912 (0.814–1.000) with a sensitivity of 86.7% and specificity of 84.2% for FGR detection. Metabolite set enrichment analysis (MSEA) revealed significant (p < .05) perturbation of multiple placental metabolite pathways including urea metabolism, ammonia recycling, porphyrin metabolism, bile acid biosynthesis, galactose metabolism and perturbed protein biosynthesis. Conclusion The placental metabolic pathway analysis revealed abnormalities that are consistent with fetal hepatic dysfunction in FGR. Near global reduction of metabolite concentrations was found in the placenta from FGR cases and metabolites demonstrated excellent diagnostic accuracy for FGR detection.

A metabolomic study of Asian and American Ginseng based on RRLC-QTOF/MS methods

In order to understand the significant differences in activities and effects between Asian ginseng (ASG) and American ginseng (AMG), it is important to study the correlation between chemical structures and biological activities in the two types of herbs. However, more attention has been paid to the analysis of ginsenosides in previous reports distinguishing between ASG and AMG. There are some other bioactive compounds besides ginsenosides, however, few studies have focused on a systematic comparison of these types of compounds. Two metabolomic methods were developed in this study by qualitative data acquisition using normal phase liquid chromatography mass spectrometry (NPLC-MS) and reverse phase liquid chromatography mass spectrometry (RPLC-MS) respectively, in combination with principal component analysis (PCA). Results show that both NPLC-MS and RPLC-MS-based metabolomic methods are feasible in composition profiling, biomarker screening as well as in discrimination of ASG and AMG. 17 ginsenosides were identified as analytical markers in RPLC-MS-based metabolomic method. In comparison, using NPLC-MS-based method, 5 ginsonosides, two amino acids as well as 1 oligosaccharide were identified as analytical markers. Therefore, RPLC-MS-based metabolomic method exhibits better profiling in ginsenosides, while NPLC-MS-based metabolomic method offers the advantage that multiple active ingredients can be determined simultaneously. The two methods are both helpful in identification of biomarker as well as in discrimination of American ginsengs from Asian ginsengs.

Discovery and characterization of CHO host cell protease-induced fragmentation of a recombinant monoclonal antibody during production process development

Monoclonal antibody (mAb) fragmentation is a widespread issue of protein stability that needs to be carefully monitored for critical mAb quality control during the production process development. This study describes here the discovery and characterization of CHO host cell protease-induced fragmentation of a therapeutic mAb-X in the formulation samples from an early production process. The fragmentation was observed in the sodium dodecyl sulfate capillary electrophoresis (CE-SDS) analysis of mAb-X formulation samples incubated at elevated temperature. Size exclusion liquid chromatography (SEC-HPLC) was used to analyze and collect these cleaved fragments derived from mAb-X. Reversed phase liquid chromatography mass spectrometry (RP-LC-MS) and tandem mass (MS/MS) analysis demonstrated that the fragment was generated mainly due to the hinge region cleavage of mAb-X. The fragmentation rate was characterized in the mAb-X formulation samples at pH from 4.0 to 6.0 using CE-SDS and SDS-PAGE analysis. The percentage of the main fragment increased dramatically from 2.8% to 31.2% as pH decreased from 6.0 to 4.0 at 40 °C for 28 days, which indicated the fragmentation was highly pH-dependent. The SDS-PAGE analysis further verified the pH-dependent property of the framentation of mAb-X. Moreover, the fragmentation was characterized in the presence and absence of pepstatin A, an inhibitor of acidic proteases. Significant inhibition of mAb-X fragmentation was observed with the addition of pepstatin A to mAb-X formulation samples. These results suggested residual acidic host cell protease(s) in the formulation samples from an early production process caused the fragmentation of mAb-X. To prove evidence, we developed an optimized protein A chromatography to enhance the residual host cell protease(s) removal capability of mAb-X purification process and consequently eliminate the above described cleaved fragment of mAb-X, which further supported the hypothesis that the fragmentation of mAb-X was catalyzed by the residual host cell protease(s) in the formulation samples from the early production process. This case study reiterated that residual host cell protease is a critical quality attribute (CQA) that should be carefully controlled and evaluated to guarantee successful manufacture processes for mAb products.

Reversed-phase liquid chromatography separation and mass spectrometry fragmentation analysis of mitiglinide and three isomers

The separation of mitiglinide and its three isomer impurities was achieved by reversed-phase ultra-performance liquid chromatography-high resolution mass spectrometry. An ACQUITY UPLC HSS T3 column (100 mm×2.1 mm, 1.8 μm) was used as the stationary phase and water-acetonitrile-n-pentanol (75:25:1, v/v/v; formic acid was added to adjust pH to 1.8) was used as the mobile phase with a flow rate of 0.4 mL/min. According to the exact mass and high resolution mass spectrometry fragmentation (Q Exactive), significant differences were observed in the fragment ion abundance in the secondary mass spectra of mitiglinide and its three isomer impurities. Two of these isomer impurities were newly discovered. The possible fragmentation mechanisms of mitiglinide and its three isomer impurities were also deduced. The limit of detection of the developed method was 1 μg/L. The linearity of the developed method was good from the limit of quantitation (2 μg/L) to 10000 μg/L with a correlation coefficient of 0.9999. The relative standard deviation of the peak area was 2.0%. On the basis of these results, the sources of the mitiglinide isomer impurities were discussed. Isomer impurity 1 was degraded at high temperature, while isomer impurities 2 and 3 were determined to be synthetic impurities. In addition, samples of mitiglinide calcium raw materials were analyzed.

Nonaqueous capillary electrophoresis mass spectrometry method for determining highly hydrophobic peptides.

A nonaqueous capillary electrophoresis mass spectrometry (NACE-MS) method was developed to separate and determine highly hydrophobic temporin peptides. The nonaqueous background electrolyte solution was a mixture of 20% acetonitrile, 78% methanol and 2% formic acid, with 20 mM ammonium formate. The separation of six peptides was completed within 12 min. The CE system was connected to a triple quadrupole mass spectrometer operating in MRM mode using a chemical modifier solution of 2 mM ammonium formate in ethanol with the flow through microvial interface. The mass spectrometer offered a second dimension of separation for peptides having identical migration times but different structures. The new method represents the first system capable of reliably determining hydrophobic peptides without using reversed phase liquid chromatography mass spectrometry.

Optimization of lipid extraction and analytical protocols for UHPLC-ESI-HRMS-based lipidomic analysis of adherent mammalian cancer cells.

Lipidomics, which reveals comprehensive characterization of molecular lipids, is a rapidly growing technology used in biomedical research. Lipid extraction is a critical step in lipidomic analysis. However, the effectiveness of different lipid extract solvent systems from cellular samples still remains unclear. In the current study, the protocol of reverse-phase liquid chromatography mass spectrometry (LC/MS)-based lipidomics was optimized for extraction and detection of lipids from human pancreatic cancer cell line PANC-1. Four different extraction methods were compared, including methanol/methyl-tert-butyl ether (MTBE)/H2O, methanol/chloroform, methanol/MTBE/chloroform, and hexane/isopropanol. Data were acquired using high-resolution mass spectrometry in positive and negative ion modes respectively. The number of total detected and identified lipids was assessed with the aid of automated lipid identification software LipidSearch. Results demonstrated that methanol/MTBE/H2O provided a better extraction efficiency for different lipid classes, which was chosen as the optimized extraction solvent system. This validated method enables highly sensitive and reproducible analysis for a variety of cellular lipids, which was further applied to an untargeted lipidomic study on human pancreatic cancer PANC-1 cell lines. Moreover, this optimized extraction solvent system can be further applied to other cancer cell lines with similar chemical and physical properties. MiR-375 Regulates Invasion-Related Proteins Vimentin and L-Plastin

Invasion is a hallmark of advanced head and neck squamous cell carcinoma (HNSCC). We previously determined that low relative miR-375 expression was associated with poor patient prognosis. HNSCC cells with increased miR-375 expression have lower invasive properties and impaired invadopodium activity. Using stable isotope labeling with amino acids in cell culture and reverse-phase liquid chromatography mass spectrometry, we assessed the impact of miR-375 expression on protein levels in UM-SCC-1 cells. Increased miR-375 expression was associated with down-regulation of proteins involved in cellular assembly and organization, death and survival, and movement. Two invasion-associated proteins, vimentin and L-plastin, were strongly down-regulated by miR-375. Luciferase reporter assays demonstrated that high miR-375 expression reduced vimentin promoter activity, suggesting that vimentin is an indirect target of miR-375. Runt-related transcription factor 1 (RUNX1) is a potential miR-375 direct target, and its knockdown reduced vimentin and L-plastin expression. Data in The Cancer Genome Atlas HNSCC database showed a significant inverse correlation between miR-375 expression and RUNX1, vimentin, and L-plastin RNA expression. These clinical correlations validate our invitro model findings and support a mechanism in which miR-375 suppresses RUNX1 levels, resulting in reduced vimentin and L-plastin expression. Furthermore, knockdown of RUNX1, L-plastin, and vimentin resulted in significant reductions in cell invasion invitro, indicating the functional significance of miR-375 regulation of specific proteins involved in HNSCC invasion.

Identification and characterization of monoclonal antibody fragments cleaved at the complementarity determining region using orthogonal analytical methods.

Sodium dodecyl sulfate capillary electrophoresis (CE-SDS) has been widely used to separate and quantify monoclonal antibody (mAb) fragments and impurities. Two fragments with different masses smaller than monomer were observed in the CE-SDS analysis of a recombinant IgG1 mAb treated at 40°C for 28days. Reversed phase liquid chromatography mass spectrometry (RP-LC-MS) and trypsin digestion peptide mapping followed by LC-MS/MS analysis were employed to conclusively demonstrate that these antibody fragments were the degradation products derived from a Ser-Ser peptide bond cleavage at the heavy chain complementarity determining region (CDR). The total percentage of the cleaved antibody heavy chain was increased from 1.1% in the control sample to 4.6% in the forced degradation sample stored at 40°C for 28days by using CE-SDS analysis. Native size exclusion liquid chromatography (SEC-HPLC) was used to analyze the fragment profile of this treated mAb but did not reveal fragments smaller than the monomer. However, a small fragment was detected using denaturing size exclusion liquid chromatography (dSEC-HPLC) and was confirmed to be the N-terminal fragment of the heavy chain cleaved at the CDR region using RP-LC-MS analysis. These results suggested that this N-terminal fragment may be closely associated with the rest of the cleaved antibody molecule through non-covalent bond interactions. Moreover, when dSEC-HPLC and RP-LC-MS were used to analyze the aggregate and monomer samples individually collected from SEC-HPLC, both samples contained the same amount of the cleaved N-terminal fragment. These results suggested that the CDR region cleaved fragments likely not be involved in aggregates formation of this antibody. This case study reiterated that a combination of orthogonal analytical methods plays a crucial role in comprehensively analyzing and characterizing antibody fragmentation.

Analysis of sorghum wax and carnauba wax by reversed phase liquid chromatography mass spectrometry

Sorghum is a genus of plant in the grass family, which is used for both grain and forage production throughout the world. In the United States, sorghum grain is predominantly used as livestock feed, and in ethanol production. In recent years however, sorghum grain has been investigated for other industrial applications, including gluten free food sources for the US food market, and waxes. The United States is the world's largest producer of grain sorghum, which is grown in the arid regions of the southern Great Plains, Arizona and California.Carnauba wax is used in a variety of products; including cosmetics, industrial polishes, food products, and paper products. The United States has no domestic source of carnauba wax, and imports 100% of its carnauba wax supply. Sorghum wax has demonstrated similar physical properties to carnauba wax, and could potentially be a viable substitute for carnauba wax.In this paper we present the first successful reversed phase HPLC method, via a C30 column, for the analysis and characterization of waxes, without the need for specialized columns or sample derivation. Sorghum wax is composed of a heterogeneous mixture of compounds, dominated by C28 and C30 saturated and unsaturated species, while carnauba is more homogeneous in nature, and composed primarily of C56-C60 saturated wax esters.

Novel post-translational oligomerization of peptidyl dehydrodopa model compound, 1,2-dehydro-N-acetyldopa methyl ester

Post-translational modification of peptidyl tyrosine to peptidyl dopa is widely observed in different marine organisms. While peptidyl dopas are oxidatively converted to dehydrodopa derivatives, nothing is known about the further fate of dehydrodopyl compounds. To fill this void, we studied the oxidation chemistry of a peptidyl dehydrodopa mimic, 1,2-dehydro-N-acetyldopa methyl ester with mushroom tyrosinase. We employed both routine biochemical studies and reversed phase liquid chromatography mass spectrometry to investigate the course of the reaction. Tyrosinase catalyzed the oxidation of 1,2-dehydro-N-acetyldopa methyl ester readily generating its typical o-quinone as the transient two-electron oxidation product. This quinone was extremely unstable and rapidly reacted with the parent compound forming benzodioxan type oligomeric products. Reaction mixture containing chemically made o-benzoquinone and 1,2-dehydro-N-acetyldopa methyl ester generated a mixed adduct of benzoquinone and 1,2-dehydro-N-acetyldopa methyl ester. Based on this finding, we propose that peptidyl dehydrodopa also exhibits a similar transformation accounting partially for the adhesive and cementing properties of dopyl proteins in nature.

Sensitivity improvement in hydrophilic interaction chromatography negative mode electrospray ionization mass spectrometry using 2-(2-methoxyethoxy)ethanol as a post-column modifier for non-targeted metabolomics

The application of ammonia acetate buffered liquid chromatography (LC) eluents is known to concomitantly lead to ion suppression when electrospray ionization mass spectrometry (ESI–MS) detection is used. In negative ESI mode, post column infusion of 2-(2-methoxyethoxy)ethanol (2-MEE) was shown in the literature to help to compensate this adverse effect occurring in reversed phase liquid chromatography mass spectrometry (RP-LC–MS) analyses. Here a setup of direct infusion and hydrophilic interaction chromatography (HILIC) post-column infusion experiments was established in order to investigate systematically the beneficial effects of 2-MEE. We demonstrate that, 2-MEE can help to improve ESI–MS sensitivity in HILIC too and reveal analyte structure specific behaviors. Our study indicates that 2-MEE especially improves ESI response for small and polar molecules. The ESI response of stable isotope labeled amino acids spiked into biological matrices increases up to 50-fold (i.e. D5-l-glutamic acid) when post column infusion of 2-MEE is applied. A non-targeted analysis of a pooled urine sample via HILIC–ESI–QTOF–MS supports this hypothesis. In direct infusion, the combined application of an ammonia acetate buffered solution together with 2-MEE results in an improved ESI response compared to a non-buffered solution. We observed up to 60-fold increased ESI response of l-lysine. We propose this effect is putatively caused by the formation of smaller ESI droplets and stripping of positive charge from ESI droplets due to evaporation of acetic acid anions. In summary, post-column infusion of 2-MEE especially enhances ESI response of small and polar molecules. Therefore it can be regarded as a valuable add-on in targeted or non-targeted metabolomic HILIC–MS studies since this method sets a focus on this molecule category.
