Cortico-thalamocortical interactions for learning, memory and decision-making.

The thalamus and cortex are interconnected both functionally and anatomically and share a common developmental trajectory. Interactions between the mediodorsal thalamus (MD) and different parts of the prefrontal cortex are essential in cognitive processes, such as learning and adaptive decision-making. Cortico-thalamocortical interactions involving other dorsal thalamic nuclei, including the anterior thalamus and pulvinar, also influence these cognitive processes. Our work, and that of others, indicates a crucial influence of these interdependent cortico-thalamocortical neural networks that contributes actively to the processing of information within the cortex. Each of these thalamic nuclei also receives potent subcortical inputs that are likely to provide additional influences on their regulation of cortical activity. Here, we highlight our current neuroscientific research aimed at establishing when cortico-MD thalamocortical neural network communication is vital within the context of a rapid learning and memory discrimination task. We are collecting evidence of MD-prefrontal cortex neural network communication in awake, behaving male rhesus macaques. Given the prevailing evidence, further studies are needed to identify both broad and specific mechanisms that govern how the MD, anterior thalamus and pulvinar cortico-thalamocortical interactions support learning, memory and decision-making. Current evidence shows that the MD (and the anterior thalamus) are crucial for frontotemporal communication, and the pulvinar is crucial for frontoparietal communication. Such work is crucial to advance our understanding of the neuroanatomical and physiological bases of these brain functions in humans. In turn, this might offer avenues to develop effective treatment strategies to improve the cognitive deficits often observed in many debilitating neurological disorders and diseases and in neurodegeneration.

Regulation of cortical activity and arousal by the matrix cells of the ventromedial thalamic nucleus

The “non-specific” ventromedial thalamic nucleus (VM) has long been considered a candidate for mediating cortical arousal due to its diffuse, superficial projections, but direct evidence was lacking. Here, we show in mice that the activity of VM calbindin1-positive matrix cells is high in wake and REM sleep and low in NREM sleep, and increases before cortical activity at the sleep-to-wake transition. Optogenetic stimulation of VM cells rapidly awoke all mice from NREM sleep and consistently caused EEG activation during slow wave anesthesia, while arousal did not occur from REM sleep. Conversely, chemogenetic inhibition of VM decreased wake duration. Optogenetic activation of the “specific” ventral posteromedial nucleus (VPM) did not cause arousal from either NREM or REM sleep. Thus, matrix cells in VM produce arousal and broad cortical activation during NREM sleep and slow wave anesthesia in a way that accounts for the effects classically attributed to “non-specific” thalamic nuclei.

Cholinergic Behavior State-Dependent Mechanisms of Neocortical Gain Control: a Neurocomputational Study.

The embodied mammalian brain evolved to adapt to an only partially known and knowable world. The adaptive labeling of the world is critically dependent on the neocortex which in turn is modulated by a range of subcortical systems such as the thalamus, ventral striatum, and the amygdala. A particular case in point is the learning paradigm of classical conditioning where acquired representations of states of the world such as sounds and visual features are associated with predefined discrete behavioral responses such as eye blinks and freezing. Learning progresses in a very specific order, where the animal first identifies the features of the task that are predictive of a motivational state and then forms the association of the current sensory state with a particular action and shapes this action to the specific contingency. This adaptive feature selection has both attentional and memory components, i.e., a behaviorally relevant state must be detected while its representation must be stabilized to allow its interfacing to output systems. Here, we present a computational model of the neocortical systems that underlie this feature detection process and its state-dependent modulation mediated by the amygdala and its downstream target the nucleus basalis of Meynert. In particular, we analyze the role of different populations of inhibitory interneurons in the regulation of cortical activity and their state-dependent gating of sensory signals. In our model, we show that the neuromodulator acetylcholine (ACh), which is in turn under control of the amygdala, plays a distinct role in the dynamics of each population and their associated gating function serving the detection of novel sensory features not captured in the state of the network, facilitating the adjustment of cortical sensory representations and regulating the switching between modes of attention and learning.

Basal forebrain administration of the somatostatin-analog octreotide does not affect cortical EEG in urethane anaesthetized rats

Basal forebrain (BF) plays an important role in the regulation of cortical activation. Somatostatin (SOM) is present both in local neurons as well as in fibers in the BF. In previous studies, SOM axons were found to innervate corticopetal cholinergic cells and SOM was found to presynaptically modulate GABA and glutamate release onto cholinergic neurons in the BF. However, no systematic analysis is available about the EEG effects of SOM or its analog, octreotide (OCTR) injected directly into the BF. In the present experiments, EEG changes were examined following an OCTR injection (0.5 microliter, 500 nmol) into the BF areas containing several choline acetyl transferase-immunoreactive neurons of urethane-anaesthetized rats. Fronto-occipital EEG was recorded on both sides and relative EEG power was calculated in the delta (0-3 Hz), theta (3-9 Hz), alpha (9-16 Hz) and beta (16-48 Hz) frequency bands. OCTR injected to the BF failed to induce significant EEG changes and did not affect tail pinch-evoked cortical activation. Lack of effect may be attributed to the urethane anaesthesia as well as to the possible complex interactions between SOM and BF cholinergic and GABAergic neurons.

Cholinergic modulation of slow cortical rhythm in urethane-anesthetized rats

Slow cortical rhythm (SCR) is characterized by rhythmic cycling of active (UP) and silent (DOWN) states in cortical cells. In urethane anesthesia, SCR appears as alternation of almost isoelectrical EEG periods and low-frequency, high-amplitude large shifts with superimposed high-frequency activity in the local field potentials (LFPs). Dense cholinergic projection reaches the cortex from the basal forebrain (BF), and acetylcholine (ACh) has been demonstrated to play a crucial role in the regulation of cortical activity. In the present experiments, cholinergic drugs were administered topically to the cortical surface of urethane-anesthetized rats to examine the direct involvement of ACh and the BF cholinergic system in the SCR. SCR was recorded by a 16-pole vertical electrode array from the hindlimb area of the somatosensory cortex. Multiple unit activity (MUA) was recorded from layer V to VI in close proximity of the recording array. Neither a low dose (10 mM solution) of the muscarinic antagonist atropine or the nicotinic agonist nicotine (1 mM solution) had any effect on SCR. In contrast, the higher dose (100 mM solution) of atropine, the cholinergic agonist carbachol (32 mM solution), and the cholinesterase inhibitor physostigmine (13 mM solution) all decreased the number of UP states, delta power (0-3 Hz) and MUA. These results suggest that cholinergic system may influence SCR through muscarinic mechanisms during urethane anesthesia. Cholinergic activation obstructs the mechanisms responsible for local or global synchronization seen during SCR as this rhythm was disrupted or aborted. Muscarinic antagonism can evoke similar changes when high dose of atropine is applied.

Neuromodulation for intractable OAB

To review the current status of sacral neuromodulation (SNM) in the treatment of intractable overactive bladder symptoms, the PubMed English-language literature was used to address efficacy, technical issues, alternative techniques, the mechanism of action, adverse events, the cost-effectiveness, and current considerations on the use of SNM as second-line treatment in OAB. Several studies attest to the short- and long-term efficacy of SNM in reducing urgency incontinence and the remaining OAB symptoms. Few, medium-sized sample, randomized controlled trials exist. Little is known about long-term patient satisfaction from SNM and prognostic factors have not been identified. Technical advances have significantly improved patient selection and outcomes, as well as complication rates, but revision rates still remain high. The mechanism of action remains largely unknown, but appears to be associated with reorganization of spinal reflexes and regulation of cortical activity. Reports on cost-effectiveness are conflicting. Less invasive forms of SNM may offer a viable alternative to central SNM. In conclusion, current evidence supports the short- and long-term efficacy of SNM in treating intractable OAB, but further research is needed to improve patient selection, identify prognostic factors, clarify the mechanism of action and reduce complications and revision rates.

Immunolocalization of the voltage‐gated potassium channel Kv2.2 in GABAergic neurons in the basal forebrain of rats and mice

The Kv2 voltage-gated potassium channels, Kv2.1 and Kv2.2, are important regulators of neuronal excitability in mammalian brain. It has been shown that Kv2.1 channels are expressed in virtually all neurons in the brain. However, the cellular localization of Kv2.2 has not been fully elucidated. In this article we report that Kv2.2 is highly expressed in a subset of neurons in the magnocellular preoptic nucleus (MCPO) and the horizontal limb of the diagonal band of Broca (HDB) of the basal forebrain complex, which are areas highly implicated in the regulation of cortical activity and the sleep/wake cycle. It has been shown that MCPO and HDB contain distinct populations of neurons that differ in their neurochemicals, cholinergic, glutamatergic, and gamma-aminobutyric acid (GABA)ergic neurons. Using specific immunolabeling and knockin mice in which green fluorescent protein (GFP) is expressed in GABAergic neurons, we found that Kv2.2 is abundantly expressed in a large subpopulation of the GABAergic neurons in the MCPO and HDB. These data offer Kv2.2 as a molecular target to study the role of the specific subpopulation of basal forebrain GABAergic neurons.

Effect of basal forebrain neuropeptide Y administration on sleep and spontaneous behavior in freely moving rats

Neuropeptide Y (NPY) is present both in local neurons as well as in fibers in the basal forebrain (BF), an area that plays an important role in the regulation of cortical activation. In our previous experiments in anaesthetized rats, significant EEG changes were found after NPY injections to BF. EEG delta power increased while power in theta, alpha, and beta range decreased. The aim of the present experiments was to determine whether NPY infusion to BF can modulate sleep and behavior in freely moving rats. In this study, microinjections were made into the BF. Saline was injected to the control side, while either saline or one of two doses of NPY (0.5 μl, 300–500 pmol) to the treated side. EEG as well as behavioral changes were recorded. Behavioral elements after the NPY injections changed in a characteristic fashion in time and three consecutive phases were defined. In phase I (half hour 2), activated behavioral items (moving, rearing, grooming) appeared frequently. In phase II (half hours 3 and 4) activity decreased, while motionless state increased. Reappearance of activity was seen in phase III (half hours 5 and 6). NPY injections caused sleep–wake changes. The three phases described for behavioral changes were also reflected in the sleep data. During phase I, lower NPY dose increased wakefulness and decreased deep sleep. Reduced behavioral activity seen in phase II was partially reflected in the sleep. In this phase, wakefulness tended to increase in the third half hour, while decreased in the 4th half hour. Deep sleep and total slow wave sleep non-significantly decreased in the third and increased in the 4th half hour. In most cases, wakefulness was elevated again during Phase III, while sleep decreased. Length of single sleep–wake epochs did not change after NPY injections. Our results suggest a role for NPY in the integration of sleep and behavioral stages via the BF.

EEG effect of basal forebrain neuropeptide Y administration in urethane anaesthetized rats

Neuropeptide Y (NPY) is present both in local neurons as well as in fibers in the basal forebrain (BF), an area that plays an important role in the regulation of cortical activation. In previous studies, NPY axons were found to innervate corticopetal cholinergic cells in this area. In addition, identified NPY positive neurons have been shown to be silent during cortical activation, but active during slow EEG waves. However, no in vivo studies have shown the effect of local NPY release in the BF on the EEG. In the present experiments, the EEG was examined following NPY injection (0.5 μl, 300–500 pmol) into the BF of urethane-anaesthetized rats. Fronto-parietal EEG was recorded on both sides and relative EEG power was calculated in the delta (0–3 Hz), theta (3–9 Hz), alpha (9–16 Hz) and beta (16–48 Hz) frequency bands. We found a significant increase in relative delta power and a decrease in the power of all higher frequency bands (theta, alpha, beta) after NPY injection. These results suggest that NPY can inhibit cortical activation via the BF.

Regional cerebral cortical activation in monoamine oxidase A-deficient mice: differential effects of chronic versus acute elevations in serotonin and norepinephrine

Mice deficient in monoamine oxidase A have previously been shown to demonstrate a chronic elevation of serotonin and norepinephrine in the brain. Using the autoradiographic [ 14C]iodo-antipyrine method, we examined cerebral cortical blood flow in conscious, restrained four- to five-month-old knock-out and wild-type animals following the intraperitoneal administration of either saline or d-fenfluramine. Knock-out animals administered saline, compared to their wild-type counterparts, demonstrated a significantly higher regional cortical blood flow in somatosensory and barrel field neocortex, an area which previous histological studies have shown to be characterized by abnormal serotonergic projection fibers and absent barrel formation. Regional cortical blood flow was significantly lower in knock-out than in wild-type mice in the entorhinal and midline motor cortex, with non-significant decreases noted in the olfactory, piriform and retrosplenial cortices and the amygdala. We compared the above findings to those obtained in response to d-fenfluramine which, in conjunction with its metabolite d-norfenfluramine, results in acute elevations of brain levels of serotonin and norepinephrine. Administration of d-fenfluramine (21.2 mg/kg) resulted in changes in regional cortical perfusion in most brain regions of both knock-out and wild-type mice that were opposite to the genotypic differences seen in perfusion in response to saline. Fenfluramine significantly increased regional cortical blood flow in the allocortex (olfactory, piriform, entorhinal) and the amygdala, and significantly decreased regional cortical blood flow in the somatosensory, barrel field, midline motor and retrosplenial cortices. Changes in regional perfusion in response to fenfluramine were topographically equivalent in knock-out and wild-type mice, although in knock-out mice such changes were of greater magnitude. Our study suggests that the effects on regional cortical blood flow of a lifelong absence of monoamine oxidase A, and the consequent chronic increase in serotonin and norepinephrine, differ from those attributable to acute increases in these neurotransmitters following fenfluramine administration. Such a differential response may reflect neurodevelopmental abnormalities and/or effects of a chronic physiological adaptation on the regulation of cortical activation.

Tonic and phasic influence of basal forebrain unit activity on the cortical EEG

Changes in arousal levels are normally accompanied by modification of gross electrical activity (EEG) in the cortex, with low amplitude fast waves characterizing high levels and large slow waves low levels of arousal. These changes in cortical EEG patterns depend mainly on two factors: on the input from the thalamus and on the state of various membrane channels in the cortical pyramidal cells, which are both regulated by ascending modulatory systems. Several lines of evidence indicate that of the activating systems the cholinergic is the most effective in activating the cortex. Its blockade with atropine induces large slow waves in the EEG, while inhibition of other systems has no such profound effect. The effect of atropine can be mimicked by lesioning the basal forebrain. Neurons in this area show very close tonic and phasic correlation with the cortical EEG, further supporting the suggestion that projections of these neurons have a special role in the regulation of cortical activity. However, there is a discrepancy between the effects of excitotoxic and selective cholinotoxic lesions of the basal forebrain. The immunohistochemical diversity of the corticopetal basal forebrain projection and the electrophysiological heterogeneity of the neurons also indicate that, in addition to cholinergic cells, other types of neurons do also participate in the regulation of cortical activity from this area. To understand the intimate details the activity of identified basal forebrain neurons must be recorded and correlated with cortical events.

Dynamics of the EEG parameters in younger schoolchildren during change in the CNS functional state (relaxation) under the action of rhythmical auditory stimulation

The article is devoted to the problem of neurophysiological mechanisms of relaxation. Formation of the relaxation state under the influence of auditory rhythmic stimulation (RS) was studied using spectral and coherent EEG characteristics and electrodermal resistance. Notable shifts of the coherence (Coh) function of biopotentials during transition to the state of relaxation induced by the RS were established, namely, an increase in the distant Coh of the frontal area. A pronounced RS influence on the regulation of the CNS functional state was shown. On the basis of the results of the study and their comparison with the published data, a conclusion is drawn about the crucial role of the frontal areas in the regulation of cortical activation during relaxation and about the physiological justification of the application of the rhythmical auditory stimulation for the formation of the relaxation state its informational value.

Discharge Properties of Juxtacellularly Labeled and Immunohistochemically Identified Cholinergic Basal Forebrain Neurons Recorded in Association with the Electroencephalogram in Anesthetized Rats

Multiple lines of evidence indicate that cholinergic basal forebrain neurons play an important role in the regulation of cortical activity and state. However, the discharge properties of cholinergic cells in relation to the electroencephalogram (EEG) are not yet known. In the present study, cells were recorded in the basal forebrain in association with cortical EEG activity in urethane-anesthetized rats, and their discharge was examined during EEG irregular slow activity and during stimulation-induced cortical activation, characterized by rhythmic slow (theta) and high-frequency (gamma) activities. Recorded cells were labeled with Neurobiotin (Nb), using the juxtacellular technique and identified as cholinergic by immunohistochemical staining for choline acetyltransferase (ChAT). Nb-positive/ChAT-positive neurons were distinctive and significantly different from Nb-positive/ChAT-negative neurons, which were heterogeneous in their discharge properties. All Nb(+)/ChAT(+) cells increased their discharge rate with stimulation, and most shifted from an irregular tonic discharge during EEG slow irregular activity to a rhythmic burst discharge during rhythmic slow activity. The stimulation-induced rhythmic discharge was cross-correlated with the EEG rhythmic slow activity. In some units the rhythmic discharge matched the rhythmic slow activity of the retrosplenial cortex; in others, it matched that of the prefrontal cortex, which occurred at a slower frequency, suggesting that subsets of cholinergic neurons may influence their cortical target areas rhythmically at particular frequencies. Cholinergic basal forebrain neurons thus may evoke and enhance cortical activation via both an increase in rate and a change in pattern to rhythmic bursting that would stimulate rhythmic slow (theta-like) activity in cortical fields during active waking and paradoxical sleep states.

Responses of cortical EEG-related basal forebrain neurons to brainstem and sensory stimulation in urethane-anaesthetized rats.

The basal forebrain can be considered to be a rostral extension of the ascending reticular activating system. A large number of neurons in the basal forebrain have been shown to display higher firing rates when low-voltage fast activity is present in the cortical EEG as opposed to states characterized by large slow waves in both unanaesthetized and anaesthetized animals. However, a smaller number of cells with increased discharge rate during slow waves was also observed in most of these studies. While it is likely that these two types of neurons have opposite roles in the regulation of cortical activation, it is not known how they respond to inputs from the brainstem or the periphery. In the present study, extracellular recordings were made in the basal forebrain of urethane-anaesthetized rats. A total of 52 neurons were studied in which the firing rate was significantly higher during fast cortical EEG waves (F-cells), and 14 neurons in which activity was significantly greater during slow waves (S-cells). The two cell types responded differently to stimulation of the pedunculopontine tegmental nucleus (PPT) and dorsal raphe nucleus (DRN) with short (0.5-1 s) trains of pulses and to noxious sensory stimuli (tail pinch). These stimulations excited most F-cells (80-96%) and inhibited the majority of S-cells (55-67%). In the few F-cells that were inhibited by stimulation, the response varied with the background firing rate of the cell: the higher the firing rate at the time of stimulation, the higher the probability of observing an inhibitory response. In contrast, single electrical pulses delivered to the PPT and DRN excited the majority (72%) of both F- and S-cells. Previous in vitro studies have shown that the application of acetylcholine or serotonin has predominantly inhibitory effects on basal forebrain cholinergic neurons. The predominantly excitatory effect of noxious, PPT and DRN stimulation on F-cells therefore suggests that glutamatergic or other excitatory afferents play a more dominant role in regulating basal forebrain neurons. We have previously shown that F-cells are more prevalent than S-cells. In combination, these findings suggest that basal forebrain neurons, and F-cells in particular, are important in mediating the ascending excitatory drive from the brainstem to the cerebral cortex.

Conative Regulation of Cortical Activity by the Reticular Formation, Hypothalamus, and Thalamus

Evolution of neural regions suggests the requirement for a common format for information units exchanged among regions. Short-term memory experiments suggest a format of six or seven items. A similar number of configurations of primary cerebral interactions is proposed, associated with arousal, sleep, approach, withdrawal, perseveration, alert scanning, and commanded by the reticular formation. A comparable number of basic states (feeding, mating, grooming, shelter-seeking, fighting, etc.) is proposed as operating within these configurations, under regulation by the hypothalamus. Conative variables from this region are transformed into patterns of regulation for local cortical populations by the thalamus. Elaboration of these configurations and states by higher structures leads to new forms of cortical activity only loosely coupled to brain stem systems.
