Reduction of carbonyl compounds by using polymethylhydro-siloxane: reactivity and selectivity

Reduction of aldehydes and ketones with PMHS [Me 3SiO-(SiMe(H)O) n-SiMe 3] proceeded smoothly in the presence of Bu 4NF at -70° or 0° within 60 min in THF. High stereo- and chemoselectivities as well as functional group tolerance of this system are also presented.

The site-selective incorporation of a NAD + cofactor mimic into a folded helix–loop–helix polypeptide motif

LA-42, a polypeptide with 42 amino acid residues, has been designed to fold into a hairpin helix–loop–helix motif that can dimerise in solution to form a four-helix bundle. On the surface of the folded motif a reactive site has been introduced that contains a histidine and a lysine residue in a helical sequence. The reaction between the His-Lys site and a p-nitrophenyl ester has previously been shown to lead to the site-selective formation of an amide at the side chain of the flanking lysine residue. An N-methylnicotinoyl group has now been incorporated into LA-42, in a reaction between the peptide and the N-methylnicotinic acid ester in aqueous solution at pH 5.9, to form a template for the engineering of selective catalysts for the reduction of carbonyl compounds. The formation of an amide bond between LA-42 and the cofactor mimic was established by electro-spray mass spectrometry and NMR spectroscopy and the reduction of the N-methylnicotinoyl residue by sodium dithionite in aqueous solution at pH 6.5–7 was demonstrated by UV and NMR spectroscopy. Key problems with NAD+/NADH models in aqueous solution include poor solubility of the ox and/or red forms of the catalysts and short lifetimes of the red form due to hydrolysis of the enamine. Both the red and the ox forms of the peptide-linked nicotinoyl cofactor are soluble in aqueous solution, which is a necessary condition for the development of turnover systems, and the lifetime of the reduced form of the polypeptide catalyst has been increased by more than a factor of three over that of the 1-methylnicotinamide.

Beta–type zeolites as selective and regenerable catalysts in the Meerwein–Ponndorf–Verley reduction of carbonyl compounds

Zeolite Beta, in the Al–form as well as in the Al–free, Ti–containing form, appears to be a selective and regenerable catalyst in the Meerwein–Ponndorf–Verley and Oppenauer (MPVO) reactions. In the liquid–phase MPV reduction of 4–tert–butylcyclohexanone with secondary alcohols, both catalysts display a high stereoselectivity to cis–4–tert–butylcyclohexanol, the isomer of industrial relevance. This stereoselectivity can be explained by considering the two transition states inside the pores of zeolite Beta. By using (S)–2–butanol as the reductant enantioselective reduction of phenylacetone was observed. 4–methylcyclohexanone was studied as the substrate in the gas–phase MPV reduction. Catalyst deactivation is much more pronounced with the acidic Al–Beta catalyst than with the non–acidic Ti–Beta.

Ab Initio Computational Examination of Carbonyl Reductions by Borane: The Importance of Lewis Acid−Base Interactions

Calculations of the energies and geometries of various complexes of diborane and borane with acetaldehyde, acetone, acetyl chloride, formaldehyde, methyl acetate, THF, dimethyl sulfide (DMS), and ammonia have been performed. Adducts and transition states involving methoxyborane, dimethoxyborane, and trimethyl borate have also been examined. The 6-31+G(d,p) basis set was used to optimize geometries with the Møller−Plesset (second order, MP2) perturbation method. These calculations characterize the probable intermediates and transition states involved in borane, diborane, THF·BH3, and DMS·BH3 reductions of carbonyl compounds as well as THF- and DMS-catalyzed diborane reductions. Four-centered transition states were located for borane reductions of acetaldehyde, acetone, acetyl chloride, formaldehyde, and methyl acetate, the accessibility of which correlates to carbonyl π-orbital energies. Diborane adducts of acetone, acetaldehyde, DMS, THF, and ammonia were also located. The ability to form such adducts depends strongly on Lewis basicity, and subsequent adduct disruption to give free borane and Lewis base−borane adducts is critical in diborane reductions. THF and DMS are predicted to disrupt completely diborane under typical conditions to give THF·BH3 and DMS·BH3. Borane transfer from one Lewis base to another can occur by SN1- or SN2-like pathways of comparable energy. Methoxyborane has a low-energy disproportionation pathway to give borane and dimethoxyborane. Dimethoxyborane is a poor reducing agent that probably does not disproportionate during reductions.

Highly regio- and stereoselective reductions of carbonyl compounds in aqueous glycosidic media

Highly regioselective reductions of α,β-unsaturated ketones to the corresponding allylic alcohols were performed in essentially quantitative yields in aqueous media containing either glycosidic surfactants or amphiphilic carbohydrates. Reductions of cyclohexanones and cyclohexenones lead under the same conditions, stereoselectively to reduced compounds bearing an equatorial alcohol function. Hydrophobic interactions between amphiphilic carbohydrates and lipophilic substrates were modelized and should account for the observed stereodifferentiation.
