Deciphering the reduction of antibiotic resistance genes (ARGs) during medium-chain fatty acids production from waste activated sludge: Driven by inhibition of ARGs transmission and shift of microbial community

Medium-chain fatty acids (MCFAs) production from waste activated sludge (WAS) by chain extension (CE) is a promising technology. However, the effects and mechanisms of CE process on the fate of antibiotic resistance genes (ARGs) remain unclear. In this study, the results showed that the removal efficiency of ARGs was 81.15 % in CE process, suggesting its efficacy in reducing environmental risks. Further, the observed decrease in mobile genetic elements (MGEs) indicated that CE process restricted the horizontal gene transfer (HGT). Complementing this, the increase in soluble organic matters and extracellular 16 S rDNA confirmed that MCFAs production caused bacterial damage. Decreased intracellular ARGs and increased extracellular ARGs further revealed that MCFAs production impaired ARGs hosts, thereby limiting the vertical gene transfer (VGT) of ARGs. Shift of microbial community combined with co-occurrence network analysis demonstrated that functional bacteria without host potential for ARGs were enriched, but potential ARGs and MGEs hosts decreased, showing the role of functional bacterial phylogeny and selection pressure of MCFAs in reducing ARGs. Finally, partial least squares path model was used to systematic verify the mechanism of ARGs removal in CE process, which was attributed to the inhibition of ARGs transmission (HGT and VGT) and shift of microbial community.

Meta-analysis addressing the potential of antibiotic resistance gene elimination through aerobic composting

The significant increase in antibiotic resistance genes (ARGs) in organic solid wastes (OSWs) has emerged as a major threat to the food chain. Aerobic composting is a widely used technology for OSW management, with the potential to influence the fate of AGRs. However, the variability of the ARG elimination effects reported in different studies has highlighted the uncertainty regarding the effects of composting on ARGs. To identify the potential of composting in reducing ARG and the factors (e.g., composting technologies and physiochemical properties) influence ARG changes, a meta-analysis was conducted with a database including 4,232 observations. The abundances of ARGs and mobile genetic elements (MGEs) can be substantially reduced by 74.3% and 78.8%, respectively, via aerobic composting. During composting, the ARG levels in chicken and swine manure tended to be reduced more significantly (81.7% and 78.0%) compared to those in cattle manure (52.3%) and sewage sludge (32.6%). The reduction rate of sulfonamide resistant genes was only 35.3%, which was much lower than those of other types. MGEs and composting duration (CD) were identified as the most important factors driving ARG changes during composting. These findings provide a comprehensive insight into the effects of composting on ARG reduction, which may help prevent the transmission in food systems.

Effect of composting and storage on the microbiome and resistome of cattle manure from a commercial dairy farm in Poland.

Manure from food-producing animals, rich in antibiotic-resistant bacteria and antibiotic resistance genes (ARGs), poses significant environmental and healthcare risks. Despite global efforts, most manure is not adequately processed before use on fields, escalating the spread of antimicrobial resistance. This study examined how different cattle manure treatments, including composting and storage, affect its microbiome and resistome. The changes occurring in the microbiome and resistome of the treated manure samples were compared with those of raw samples by high-throughput qPCR for ARGs tracking and sequencing of the V3-V4 variable region of the 16S rRNA gene to indicate bacterial community composition. We identified 203 ARGs and mobile genetic elements (MGEs) in raw manure. Post-treatment reduced these to 76 in composted and 51 in stored samples. Notably, beta-lactam, cross-resistance to macrolides, lincosamides and streptogramin B (MLSB), and vancomycin resistance genes decreased, while genes linked to MGEs, integrons, and sulfonamide resistance increased after composting. Overall, total resistance gene abundance significantly dropped with both treatments. During composting, the relative abundance of genes was lower midway than at the end. Moreover, higher biodiversity was observed in samples after composting than storage. Our current research shows that both composting and storage effectively reduce ARGs in cattle manure. However, it is challenging to determine which method is superior, as different groups of resistance genes react differently to each treatment, even though a notable overall reduction in ARGs is observed.

Removal of antibiotics, bacterial toxicity, and occurrence of antibiotic resistance genes in secondary hospital effluents treated with granular activated carbon and the impact of preceding chlorination

This comprehensive investigation highlighted the complex adsorption behaviors of antibiotics onto granular activated carbon (GAC), the effectiveness of this adsorption in reducing bacterial toxicity, and the reduction of antibiotic resistance genes (ARGs) and antibiotic resistant bacteria (ARB) in hospital wastewater (HWW) effluents. Six GACs were characterized for their physicochemical properties, and their ability to adsorb six antibiotics in the background matrix of actual HWW was evaluated. Coconut shell-derived GAC (Co-U), which had the highest hydrophobicity and lowest content of oxygen-containing acidic functional groups, demonstrated the highest adsorption capacities for the tested antibiotics. Bacterial toxicity tests revealed that GACs could eliminate the bacterial toxicity from antibiotic intermediates present in chlorinated HWW. By contrast, the bacterial toxicity could not be removed by GACs in non-chlorinated HWW due to the greater presence of intermediate components identified by LC-MS/MS. The intraparticle diffusion coefficient of antibiotics adsorbed onto Co-U could be calculated by adsorption kinetics derived from the linear driving force model and the homogenous intraparticle diffusion model associated with the linear adsorption isotherms (0–150 μg/L). Meropenem and sulfamethoxazole exhibited the highest adsorption capacities in a single-solute solution compared to penicillin G, ampicillin, cetazidime, and ciprofloxacin. However, the greater adsorption capacities of meropenem and sulfamethoxazole disappeared in mixed-solute solutions, indicating the lowest adsorption competition. GAC can eliminate most ARGs while also promoting the growth of some ARB. Chlorination (free chlorine residues at 0.5 mg Cl2/L) did not significantly affect the overall composition of ARGs and ARB in HWW. However, the accumulation of ARGs and ARB on GAC in fixed bed columns was lower in chlorinated HWW than in non-chlorinated HWW due to an increase in the adsorption of intermediates.

Aeration-driven piezoelectric activation of peroxymonosulfate achieves effective mitigation of antibiotic resistance dissemination

The antibiotic resistance dissemination in water has become a globally concerned issue, and the wastewater discharge, especially medical wastewater, is considered as one of the most important sources for antibiotic resistance genes (ARGs). However, the effectiveness of current disinfection techniques in the ARGs reduction still remains controversial. In this study, a novel aeration-driven piezoelectric peroxymonosulfate (PMS) activation system using oxygen-vacancy engineered BaTiO3 (BTO) was developed to effectively eliminate antibiotic resistant bacteria (ARB) and ARGs from water. The ARB can be completely inactivated and ∼3.0 logs of ARGs can be removed by the PMS/BTO/aeration system within 1 h, and the spent BTO nanoparticles can be facilely reused after simple rinsing. The aeration can not only provide the driving force for the piezocatalytic process but also more dissolved oxygen in water that played an important role in the generation of free radicals. The radical quenching experiments and electron spin-resonance (ESR) confirmed that all the free radicals, including singlet oxygen (1O2), hydroxyl radical (OH•), sulfate radical (SO4•−) and superoxide radical (•O2−), contributed to the ARGs reduction and 1O2 radicals were identified as the dominant active species. This work provides a high-efficiency and energy saving approach for the mitigation of ARGs from water as the universal use of aeration in water treatment processes and the good reusability of BTO nanoparticles.

To what extent do water reuse treatments reduce antibiotic resistance indicators? A comparison of two full-scale systems

Water reuse is an essential strategy for reducing water demand from conventional sources, alleviating water stress, and promoting sustainability, but understanding the effectiveness of associated treatment processes as barriers to the spread of antibiotic resistance is an important consideration to protecting human health. We comprehensively evaluated the reduction of antibiotic resistance genes (ARGs) and antibiotic-resistant bacteria (ARB) in two field-operational water reuse systems with distinct treatment trains, one producing water for indirect potable reuse (ozone/biologically-active carbon/granular activated carbon) and the other for non-potable reuse (denitrification-filtration/chlorination) using metagenomic sequencing and culture. Relative abundances of total ARGs/clinically-relevant ARGs and cultured ARB were reduced by several logs during primary and secondary stages of wastewater treatment, but to a lesser extent during the tertiary water reuse treatments. In particular, ozonation tended to enrich multi-drug ARGs. The effect of chlorination was facility-dependent, increasing the relative abundance of ARGs when following biologically-active carbon filters, but generally providing a benefit in reduced bacterial numbers and ecological and human health resistome risk scores. Relative abundances of total ARGs and resistome risk scores were lowest in aquifer samples, although resistant Escherichia coli and Klebsiella pneumoniae were occasionally detected in the monitoring well 3-days downgradient from injection, but not 6-months downgradient. Resistant E. coli and Pseudomonas aeruginosa were occasionally detected in the nonpotable reuse distribution system, along with increased levels of multidrug, sulfonamide, phenicol, and aminoglycoside ARGs. This study illuminates specific vulnerabilities of water reuse systems to persistence, selection, and growth of ARGs and ARB and emphasizes the role of multiple treatment barriers, including aquifers and distribution systems.

Gamma-ray irradiation as an effective method for mitigating antibiotic resistant bacteria and antibiotic resistance genes in aquatic environments

The prevalence of antibiotic resistance genes (ARGs) in municipal wastewater treatment plants (MWTPs) has emerged as a significant environmental concern. Despite advanced treatment processes, high levels of ARGs persist in the secondary effluent from MWTPs, posing ongoing environmental risks. This study explores the potential of gamma-ray irradiation as a novel approach for sterilizing antibiotic-resistant bacteria (ARB) and reducing ARGs in MWTP secondary effluent. Our findings reveal that gamma-ray irradiation at an absorbed dose of 1.6 kGy effectively deactivates all culturable bacteria, with no subsequent revival observed after exposure to 6.4 kGy and a 96-h incubation in darkness at room temperature. The removal efficiencies for a range of ARGs, including tetO, tetA, blaTEM-1, sulI, sulII, and tetW, were up to 90.5% with a 25.6 kGy absorbed dose. No resurgence of ARGs was detected after irradiation. Additionally, this study demonstrates a considerable reduction in the abundances of extracellular ARGs, with the transformation efficiencies of extracellular tetracycline and sulfadiazine resistance genes decreasing by 56.3–81.8% after 25.6 kGy irradiation. These results highlight the effectiveness of gamma-ray irradiation as an advanced and promising method for ARB sterilization and ARG reduction in the secondary effluent of MWTPs, offering a potential pathway to mitigate environmental risks associated with antibiotic resistance.

Simultaneous volatile fatty acids promotion and antibiotic resistance genes reduction in fluoranthene-induced sludge alkaline fermentation: Regulation of microbial consortia and cell functions

The impact and mechanism of fluoranthene (Flr), a typical polycyclic aromatic hydrocarbon highly detected in sludge, on alkaline fermentation for volatile fatty acids (VFAs) recovery and antibiotic resistance genes (ARGs) transfer were studied. The results demonstrated that VFAs production increased from 2189 to 4272 mg COD/L with a simultaneous reduction of ARGs with Flr. The hydrolytic enzymes and genes related to glucose and amino acid metabolism were provoked. Also, Flr benefited for the enrichment of hydrolytic-acidifying consortia (i.e., Parabacteroides and Alkalibaculum) while reduced VFAs consumers (i.e., Rubrivivax) and ARGs potential hosts (i.e., Rubrivivax and Pseudomonas). Metagenomic analysis indicated that the genes related to cell wall synthesis, biofilm formation and substrate transporters to maintain high VFAs-producer activities were upregulated. Moreover, cell functions of efflux pump and Type IV secretion system were suppressed to inhibit ARGs proliferation. This study provided intrinsic mechanisms of Flr-induced VFAs promotion and ARGs reduction during alkaline fermentation.

The fate of antibiotic resistance genes during anaerobic digestion of sewage sludge with ultrasonic pretreatment.

This study investigated the effect of ultrasonic (US) pretreatment at three different contact times (30, 45, and 60 min) with a power of 240 W and frequency of 40 kHz on the fate of antibiotic resistance genes (ARGs), mobile genetic elements (MGEs), and enteric pathogens during anaerobic digestion (AD) of sludge. By using real time-qPCR, three MGEs (int1, int2, and tnpA) and seven ARGs (sul1, sul2, tetW, tetA, tetO, ermF, and aac(6')-lb) were quantified that have serious human health impacts and represent the most widely used antibiotics (tetracycline, sulfonamide, macrolide, and aminoglycoside). Results indicated that US pretreatment under different contact times improved the removal of ARGs and MGEs. Compared to 30 and 45 min of US pretreatment, 60 min of US pretreatment resulted in a higher reduction of ARGs with total ARG reduction of 41.70 ± 1.13%. Furthermore, the relative abundance of ARGs and MGEs after US pretreatment was reduced more effectively in anaerobic reactors than in a control AD without US pretreatment. The total ARGs and MGEs removal efficiency of control AD was 44.07 ± 0.72% and 63.69 ± 1.43%, and if US pretreatment at different times were applied, the total ARGs and MGEs removal efficiency of the whole pretreatment AD process improved to 59.71 ± 2.76-68.54 ± 1.58% and 69.82 ± 2.15-76.84 ± 0.22%. The highest removal of total ARGs (68.54 ± 1.58%) and MGEs (76.84 ± 0.22%) was achieved after AD with US pretreatment at 45 min. However, US pretreatment and AD with US pretreatment were not effective in inactivation of enteric pathogens (total coliforms and E. coli), suggesting that posttreatment is needed prior to land application of sludge to reduce the level of enteric pathogens. There was no detection of the studied ARGs and MGEs in the enteric pathogens after US pretreatment in subsequent AD. According to this study, long contact times of US pretreatment can mitigate ARGs and MGEs in AD processes, offering valuable insight into improving environmental safety and sustainable waste management. Additionally, the study highlights the need to investigate posttreatment techniques for reducing enteric pathogens in AD effluent, a crucial consideration for agricultural use and environmental protection.

Ozone micron bubble pretreatment for antibiotic resistance genes reduction in hospital wastewater treatment

Ozone micron bubble (OMB) treatment offers a promising approach to effectively eliminate Antibiotic Resistance Genes (ARGs) from infectious medical wastewater and mitigate the threat of drug resistance transmission. This study evaluated the effectiveness of OMB treatment for reducing ARGs from infectious medical wastewater in laboratory and on-site pilot treatment setups. In part, the presence of antibiotic residues in a hospital wastewater treatment plant (WWTP) and the impact of hospital wastewater on the distribution of ARGs in a wastewater collection system were also investigated. The results of wastewater collection system survey revealed a high prevalence of ARGs in the system, particularly mcr-1, largely originating from medical wastewater discharges. Furthermore, analysis of antibiotic residues in the hospital wastewater treatment system showed significant accumulation, particularly of quinolone antibiotics, in the biomass of the biological oxidation tank, suggesting a potential risk of ARG proliferation within the system. Comparison of wastewater samples from domestic and hospital WWTPs revealed a relatively higher abundance of ARGs in the latter, with differences ranging from 2.2 to sixfold between corresponding locations in the treatment plants. Notably, the biological oxidation unit of both WWTPs exhibited a greater proportion of ARGs among all sampled points, indicating the potential proliferation of ARGs within the biomass of the treatment units. ARG degradation experiments showed that OMB treatment resulted in a significantly lower CT value (9.3 mg O3 L−1 min) compared to ozone coarse bubble treatment (102 mg O3 L−1 min) under identical test conditions. Moreover, the use of OMB on site significantly reduced the accumulation of ARGs in hospital wastewater, underscoring its potential as an effective solution for mitigating ARG spread.

Distribution and dynamics of antibiotic resistance genes in sludge under discharge plasma oxidation

Antibiotic resistance genes (ARGs), emerging environmental contaminants, have been largely accumulated in sludge during wastewater treatment. Discharge plasma oxidation, an advanced oxidation technology, has been demonstrated as an effective strategy in sludge pretreatment. However, the effect of plasma oxidation on the distribution and dynamics of ARGs in sludge is unknown. Herein, the variations of four typical ARGs (tetC, tetW, blaTEM-1, aac(3)-Ⅱ) and one integron (intI-1) in sludge during discharge plasma treatment were investigated. The results showed that under low discharge voltage or short treatment time, weak plasma oxidation enriched the contents of ARGs and intI-1 in sludge (with 1.14 log unit maximum increment) via proliferating biomass or promoting horizontal gene transfer. Further increasing discharge voltage or prolonging treatment time, strong plasma oxidation eliminated target genes due to the destroyed bacteria or inhibited horizontal transfer of ARGs. Especially, the homogeneous condition greatly promoted the reduction of ARGs in liquid phase (up to 1.52 log unit reduction). Furthermore, the correlation analysis between intI-1 and ARGs implied that tetC and tetW abundances might be significantly affected by intI-1 through horizontal transfer. At the same time, the microbial communities in sludge were investigated to help understand the effect of plasma oxidation on ARGs variation. Notably, the most predominant phylum Proteobacteria and genus Acinetobacter resisted the oxidative infringement from plasma to maintain high relative abundances (52.05 % in solid phase and 84.10 % in liquid phase for Proteobacteria, 13.82 % in solid phase and 78.00 % in liquid phase for Acinetobacter at 20 kV). Meanwhile, the co-occurrence analysis between genes and dominant genera further showed that Romboutsia sp. might be a potential host bacteria of ARGs and intI-1 in solid phase, and Acidovorax sp., Parachlamydia sp., and Armatimonadetes_gp5 sp. might be the potential hosts of tetW, tetC, and intI-1 in liquid phase.

A novel strategy for eliminating antibiotic resistance genes during fertilization of dewatered sludge by earthworms: Vermicomposting practice using Chinese herbal residues derived from Lianhua Qingwen as a bulking material

Vermicomposting is a sustainable sludge recycling technology that utilizes an eco-friendly composting using earthworms and microorganisms. However, a high abundance of antibiotic resistance genes (ARGs) remains in dewatered sludge that is not satisfactorily eliminated by vermicomposting. Chinese herbs have played a major role in curing many diseases in East Asia, leading to a large amount of Chinese herbal residues (CHRs) are difficult to dispose of. The present study investigated the feasibility of CHRs on the ARGs reduction in dewatered sludge during vermicomposting. The CHRs derived from Lianhua Qingwen were added separately to sludge with weight ratios of 0%, 10%, 30%, and 50%; sludge was then vermicomposted for 30 days. The results showed that co-vermicomposting of sludge and CHRs is a feasible strategy. The CHR treatments significantly (P < 0.05) decreased antibiotic concentration and bacterial population by 23.64%–49.68% and 42.58%–93.07%, respectively, compared to counterpart. Compared to the control, the CHR addition lowered the absolute abundances of macrolide, tetracycline, and sulfonamide ARGs by 42.69%–85.15%, 22.03%–75.24%, and 23.59%–90.66%, respectively. In addition, sludge containing 30% CHRs showed significant (P < 0.05) elimination of intⅠ-1 and tnpA-4 genes with abundance reductions of 71.40% and 52.33%, respectively, relative to the control. This study suggests that the CHRs can effectively reduce ARGs content in sludge by decreasing the bacterial population and horizontal gene transfer capacity during vermicomposting.

Simulated-sunlight enhances membrane aerated biofilm reactor performance in sulfamethoxazole removal and antibiotic resistance genes reduction

Membrane aerated biofilm reactors (MABRs) can be used to treat domestic wastewater containing sulfamethoxazole (SMX) because of their favorable performance in the treatment of refractory pollutants. However, biologics are generally subjected to antibiotics stress, which induces the production of antibiotic resistance genes (ARGs). In this study, a simulated-sunlight assisted MABR (L-MABR) was used to promote SMX removal and reduce ARGs production. The SMX removal efficiency of the l-MABR system was 9.62 % superior to that of the MABR system (83.13 %). In contrast from MABR, in the l-MABR, only 28.75 % of SMX was removed through microbial activity because functional bacteria were inactivated through radiation by simulated sunlight. In addition, photolysis (64.61 %) dominated SMX removal, and the best performing indirect photolysis process was the excited state of effluent organic matters (3EfOMs*). Through photolysis, ultraviolet (UV) and reactive oxygen species (ROS) enriched the SMX removal route, resulting in the SMX removal pathway in the l-MABR no longer being limited by enzyme catalysis. More importantly, because of the inactivation of functional bacteria, whether in the effluent or biofilm, the copy number of ARGs in the l-MABR was 1–3 orders of magnitude lower than that in the MABR. Our study demonstrates the feasibility of utilizing simulated-sunlight to enhance the antibiotic removal efficiency while reducing ARG production, thus providing a novel idea for the removal of antibiotics from wastewater.

Metagenomics insights into the performance and mechanism of soil infiltration systems on removing antibiotic resistance genes in rural sewage

The removal of antibiotic resistance genes (ARGs) in sewage is of great concern, but advanced sewage treatment technologies are not suitable for rural areas, so the multi-layer soil infiltration system (MSL) has been developed for rural sewage treatment. However, little is known about the performance and function of MSL in the treatment of ARGs in rural sewage. Here, we optimized the matrix composition and structure of MSL and explored the efficacy and mechanism of MSL systems for ARG removal under different hydraulic conditions. The ARGs removal rate of MSL ranged from 41.51% to 99.67%, in which MSL with the middle hydraulic load, high pollution load, and continuous inflowing conditions showed the best removal performance. In addition, this system can operate stably and resist the temperature fluctuation, which showed an equivalent removal rate of ARGs in warm and cold seasons, amounting to 69.0%. The structural equation model revealed that microorganisms in sewage could significantly affect ARG removal (path coefficient = 0.91), probably owing to their interspecies competition. As for the internal system, the reduction of ARGs was mainly driven by microorganisms in the system matrix (path coefficient = 0.685), especially soil-mixture-block (SMB) microorganisms. The physicochemical factors of the matrix indirectly reduce ARGs by affecting the microorganisms that adhere to the matrices. Note that the pairwise alignment of nucleotide analysis demonstrated that the system matrix, especially biochar in the SMB, adsorbed ARGs and their hosts from the sewage, and in turn eliminated them by inhibiting the spread and colonization of hosts, thereby reducing the abundance of ARGs. Collectively, this study provides a deeper insight into the removal of ARGs from rural sewage by MSL, which can help improve sewage treatment technologies.

The factors controlling antibiotic resistance genes in different treatment processes of mainstream full-scale wastewater treatment plants

The alteration of antibiotic resistance genes (ARGs) in wastewater has been less studied in wastewater treatment plants (WWTPs), making it difficult to assess ARGs' spreading risk comprehensively. Therefore, this study investigated the distribution and reduction of ARGs in the main process (Anaerobic-Anoxic-Oxic with Membrane Bio-Reactor (A2/O + MBR), Oxidation Ditch with sedimentation (OD), and Cyclic Activated Sludge System (CASS) with sedimentation) and disinfection process (Ultra-violet and Chlorination) of full-scale WWTPs. The wastewater was sampled before and after the different main process and disinfection process; then, the diversity and abundance of ARGs and mobile genetic genes (MGEs, helping the horizontal transfer of ARGs) in wastewater of different treatment stages were determined by a real-time high-throughput quantitative PCR (HT-qPCR) system. It was found that similar influents would result in similar ARGs in wastewater samples, independent of the treatment processes used. The main process could effectively reduce the abundance of ARGs and MGEs by 1.80–2.12 and 1.46–2.18 logarithm units, respectively. The main factors affecting ARGs were mainly wastewater quality index, especially COD, and MGEs like transposase and insertion sequences which were significantly associated with 66 and 48 subtypes of ARGs, respectively. Moreover, disinfection was more effective than the main process in inactivating antibiotic resistance bacteria (ARB), and the removal rate of ARB by disinfection reached 43.53 %–100 %. However, there are still risks of ARB regeneration (up to 4.22 log units) in the effluent of WWTPs. In the future, nutrient removal and disinfection process improvement is necessary to benefit ARG and ARB removal.

Transmission and regulation insights into antibiotic resistance genes in straw-sludge composting system amended with calcium peroxide

This study developed a Fenton-like system by adding calcium peroxide (CaO2) to a composting system containing straw and sludge. The objective was to examine the influence of antibiotic resistance genes (ARGs) and the structure of the bacterial community. The findings indicated that the inclusion of CaO2 facilitated the reduction of ARGs. ARGs abundance in the test group (T) with CaO2 was 19.02% lower than that in the control check group (CK) without CaO2, and the abundance of ARGs in both groups after composting was lower than the initial abundance. Additionally, the structure of bacterial community in both groups underwent significant changes. Redundancy analysis (RDA) revealed that the CaO2-induced Fenton-like reaction predominantly affected temperature, pH, and the bacterial community by means of reactive oxygen species (ROS). In conclusion, the addition of CaO2 enhanced the removal of ARGs from sewage-sludge and improved compost quality in the composting.

Attenuation effects of ZVI/PDS pretreatment on propagation of antibiotic resistance genes in bioreactors: Driven by antibiotic residues and sulfate assimilation

Sulfate radical-based advanced oxidation processes (AOPs) combined biological system was a promising technology for treating antibiotic wastewater. However, how pretreatment influence antibiotic resistance genes (ARGs) propagation remains largely elusive, especially the produced by-products (antibiotic residues and sulfate) are often ignored. Herein, we investigated the effects of zero valent iron/persulfate pretreatment on ARGs in bioreactors treating sulfadiazine wastewater. Results showed absolute and relative abundance of ARGs reduced by 59.8%− 81.9% and 9.1%− 52.9% after pretreatments. The effect of 90-min pretreatment was better than that of the 30-min. The ARGs reduction was due to decreased antibiotic residues and stimulated sulfate assimilation. Reduced antibiotic residues was a major factor in ARGs attenuation, which could suppress oxidative stress, inhibit mobile genetic elements emergence and resistant strains proliferation. The presence of sulfate in influent supplemented microbial sulfur sources and facilitated the in-situ synthesis of antioxidant cysteine through sulfate assimilation, which drove ARGs attenuation by alleviating oxidative stress. This is the first detailed analysis about the regulatory mechanism of how sulfate radical-based AOPs mediate in ARGs attenuation, which is expected to provide theoretical basis for solving concerns about by-products and developing practical methods to hinder ARGs propagation.

Evaluating the impact of the humic acid amendment on antibiotic resistance genes reduction and product quality during swine manure composting

The overuse of antibiotics in livestock farms and their subsequent accumulation in manures lead to increase antibiotic resistance genes (ARGs) in the compost products. Yet, the function of humic acid (HA) in shaping antibiotics and ARGs abundances, microbial structure, and product quality during composting remains unclear. Here, the efficiency of added HA in reducing antibiotics and ARGs abundances during swine manure (SM) composting was studied using five rates of HA: 0%, 1%, 2%, 4%, and 6%. The findings indicated that the abundance of ARGs types such as LSM, bacitracin, macrolide, sulfonamide, tetracycline, and vancomycin and ARGs subtypes including MacB, MexD, OleB, SrmB, Sul1, VanHB, VanSB, pbp2b, tetPB, and tetX decreased significantly under 6% HA amount compared with CK compost (P < 0.05). Besides, the added 6% HA shaped the abundance of the bacterial community (notably Acinetobacter, Pseudoxanthomonas, and Luteimonas), as well as the level of carbohydrate metabolism and amino acid metabolism (6.9% and 5.4%, respectively), which can be taken as an indicator for microbial activity, thereby accelerating antibiotics decrease. RDA analysis revealed that TOC, moisture content, and pH considerably influenced microbial activity and ARGs reduction (P < 0.05). The estimation of compost quality indicated that 6% HA significantly improved the total content of humic substance and HA to 120.7 ± 1.02 g/kg and 62.3 ± 0.92 g/kg, and reduced the content of fulvic acid to 12.8 ± 1.22 g/kg, thus promoting antibiotics and ARGs reduction. Overall, this study offers a promising option for improving compost quality and safety and mitigating ARGs circulation in the natural environment.
