Design and synthesis of a TICT-based red-emissive fluorescent probe for the rapid and selective detection of HSA in human biofluids and live cell imaging.

Here, we report the design and synthesis of a D⋯π⋯A-based fluorescent probe, (E)-4-(4-(dibutylamine)-2-hydroxystyryl)-1-methylquinolin-1-ium (DHMQ), which is nonfluorescent in ∼100% PBS buffer medium due to a twisted intra molecular charge transfer (TICT) phenomenon and it becomes highly fluorescent (∼149 fold) in the presence of human serum albumin (HSA), owing to the restriction of its intramolecular free rotation inside the hydrophobic binding cavity of HSA. The site-selective fluorescence displacement assay and molecular docking studies clearly reveal that DHMQ selectively binds at subdomain IB of HSA. The 3σ/slope method was adopted to determine the limit of detection (LOD) value, which was as low as 2.39 nM in ∼100% PBS medium, indicating its high sensitivity towards HSA. The low dissociation constant value [Kd = (1.066 ± 0.017) μM] suggests a strong complexation between the DHMQ and HSA. Importantly, it has been demonstrated that DHMQ is capable of detecting HSA in real human serum and urine samples and was found to be suitable for live cell imaging of HSA.

A highly specific red-emitting fluorescent probe for monitoring of Cu2+ in living cells and zebrafish.

A novel seminaphthorhodafluor-based fluorescent probe was designed and prepared. It exhibited an outstanding off-on fluorescence response (about 45-fold enhancement) to Cu2+ because the structure changed from a spirocyclic form to a ring-open state. It showed satisfactory detection performances for Cu2+ with fast response speed, high selectivity and anti-interference, and a low detection limit (22.9nM) due to its long emission wavelength and off-on fluorescence response mode. This probe was also satisfactorily applied for the monitoring of Cu2+ in living cells and zebrafish.

Selective and Sensitive Detection of Fe3+ Ions Using a Red-Emissive Fluorescent Probe Based on Triphenylamine and Perylene-Linked Conjugated Microporous Polymer.

The Expansion of modern industry underscores the urgent need to address heavy metal pollution, which is a threat to human-health and environment. Efforts are underwent to develop precise technologies for detecting heavy metal ions (M+-ion). One promising approach involves the use of Conjugated Microporous Polymers (CMPs) modified with Triphenylamine (TPA) anderylene (Peryl), known as TPA-Peryl-CMP, which emits strong refluorescence. Various analytical techniques, such as Brunauer-Emmett-Teller analysis, Fourier transform infrared (FTIR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, and thermogravimetric analysis (TGA), are utilized to characterize the synthesized TPA-Peryl-CMP and understand its functional properties. In addition to its remarkable fluorescence behavior, TPA-Peryl-CMP shows promise as a sensor for Fe3+ ions using a turn-off strategy. Due to its exceptional stability and robust π-electron system, this platform demonstrates remarkable sensitivity and selectivity, significantly improving detection capabilities for specific analytes. Detailed procedures related to the mechanism for detecting Fe3+ ions are outlined for sensing Fe3+ ions, revealing a notably strong linear correlation within the concentration range of 0-3µM, with a correlation coefficient of 0.9936 and the Limit of detection (LOD) 20nM. It is anticipated that development of such a kind of TPA-Peryl-CMP will observe broader applications in detecting various analytes related to environmental and biological systems.

A water-soluble red-emitting fluorescence probe for detecting hazardous hydrazine in environmental waters and biosystems

Hydrazine (N2H4), a crucial chemical raw material, enhances people's lives and fosters human progress. Hydrazine usage or leakage has caused environmental contamination, affecting water, soil, and living beings. Hydrazine simultaneously presents a possible risk to human health due to its carcinogenic properties. Thus, quick and precise detection of hydrazine is crucial in environmental studies and biological contexts. We prepared a red-emitting fluorescence turn-on probe (XT-HZ) to detect hydrazine specifically. The probe has a low detecting limit for hydrazine (63 nM) with excitation wavelength at 570 nm and emission wavelength at 625 nm. Besides, the probe XT-HZ had excellent water solubility, high selectivity, and good sensitivity for detecting hydrazine. Finally, probe XT-HZ was applied in the imaging of N2H4 in living cells, zebrafish and environmental water samples.

Real-time and specific monitoring of nitric oxide and evaluating of the oxidative stress in living cells and zebrafish under the pollutant exposure using a carbon dot-based composite fluorescent probe

Nitric oxide (NO) functions as an essential signalling molecule in various physiological and pathological pathways. In vitro and vivo redox processes mediated by reactive oxygen species (ROS) and nitric oxide (NO) directly influence the intracellular state. In this study, a red-emitting fluorescent nanoprobe, N,S-CDs@Zn-ICA, was synthesized to monitor NO fluctuations in living cells and zebrafish under the exposure to various pollutants. Red-emissive carbon dots (N,S-CDs) were synthesized by a hydrothermal method using o-phenylenediamine and urea as carbon / nitrogen sources, and H2SO4 as sulfur source. Glutathione (GSH) was introduced to link N,S-CDs with metal organic complexes (Zn-ICA) through an amidation reaction to fabricate a carbon dot-based composite fluorescent probe, which greatly improved the selectivity, stability, and response time of the N,S-CDs. The composite probe has high selectivity and sensitivity with limit of detection (LOD) of 96.0 nM. Furthermore, the proposed probe was successfully used to monitor the dynamic changes in NO levels and evaluate oxidative stress in MCF-7 cells and zebrafish under the exposure to various pollutants, including seven heavy metal ions (such as Pb2+, Cd2+, and Hg2+) and nine organic pollutants at different concentrations and exposure times. This work provides a novel strategy for constructing highly selective and red-emitting fluorescent probe for real-time and dynamic monitoring of NO and further evaluating oxidative stress induced by pollutants in vitro and in vivo via fluorescence imaging.

A Red-Emission Fluorescent Probe with Large Stokes Shift for Detection of Viscosity in Living Cells and Tumor-Bearing Mice.

Abnormal viscosity is closely related to the occurrence of many diseases, such as cancer. Therefore, real-time detection of changes in viscosity in living cells is of great importance. Fluorescent molecular rotors play a critical role in detecting changes in cellular viscosity. Developing red emission viscosity probes with large Stokes shifts and high sensitivity and specificity remains an urgent and important topic. Herein, a novel viscosity-sensitive fluorescent probe (TCF-VIS1) with a large stokes shift and red emission was prepared based on the 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) skeleton. Due to intramolecular rotation, the probe itself does not fluorescence at low viscosity. With the increase in viscosity, the rotation of TCF-VIS1 is limited, and its fluorescence is obviously enhanced. The probe has the advantages of simple preparation, large Stokes shift, good sensitivity and selectivity, and low cytotoxicity, which make it successfully used for viscosity detection in living cells. Moreover, TCF-VIS1 showed its potential for cancer diagnosis at the cell level and in tumor-bearing mice by detecting viscosity. Therefore, the probe is expected to enrich strategies for the detection of viscosity in biological systems and offer a potential tool for cancer diagnosis.

A Red-Emission Fluorescent Probe for Intracellular Biothiols and Hydrogen Sulfide Imaging in Living Cells.

This research centers on the development and synthesis of a longwave fluorescence probe, labeled as 60T, designed for the simultaneous detection of hydrogen sulfide, cysteine/homocysteine, and glutathione. The probe showcases a swift response, good linearity range, and heightened sensitivity, boasting that the detection limits of the probe for Cys, Hcy, GSH and H2S were 0.140, 0.202, 0.259 and 0.396 μM, respectively. Notably, its efficacy in monitoring thiol status changes in live MCF-7 cells is underscored by a substantial decrease in fluorescence intensity upon exposure to the thiol trapping reagent, N-ethyl maleimide (NEM). With an impressive red emission signal at 630 nm and a substantial Stokes shift of 80 nm, this probe exhibits remarkable sensitivity and selectivity for biothiols and H2S, indicating promising applications in the diagnosis and surgical navigation of relevant cancers.

Development of a hypoxia-activated red-emission fluorescent probe for in vivo tumor microenvironment imaging and anti-tumor therapy.

Hypoxia, a significant feature of the tumor microenvironment, is closely associated with tumor growth, metastasis, and drug resistance. In the field of tumor microenvironment analysis, accurately imaging and quantifying hypoxia - a critical factor associated with tumor progression, metastasis, and resistance to therapy - remains a significant challenge. Herein, a hypoxia-activated red-emission fluorescent probe, ODP, for in vivo imaging of hypoxia in the tumor microenvironment is presented. Among various imaging methods, optical imaging is particularly convenient due to its rapid response and high sensitivity. The ODP probe specifically targets nitroreductase (AzoR), an enzyme highly expressed in hypoxic cells, playing a vital role by catalyzing the cleavage of azo bonds. The optical properties of ODP exhibited excellent performance in terms of fluorescence enhancement, fluorescence lifetime (0.81 ns), and detection limit (0.86 µM) in response to SDT. Cell imaging experiments showed that ODP could effectively detect and image intracellular hypoxia and the imaging capability of ODP was studied under various conditions including cell migration, antioxidant treatment, and different incubation times. Through comprehensive in vitro and in vivo experiments, including cellular imaging and mouse tumor models, this work demonstrates the efficacy of ODP in accurately detecting and imaging hypoxia. Moreover, ODP's potential in inducing apoptosis in cancer cells offers a promising avenue for integrating diagnostic and therapeutic strategies in cancer treatment. This innovative approach not only contributes to the understanding and assessment of tumor hypoxia but also opens new possibilities for targeted cancer therapy.

Rational design of a red-emission fluorescent probe for imaging mitochondrial viscosity and fatty liver tissues

Abnormal mitochondrial viscosity is closely associated with various diseases. Thus, it is imperative to design and synthesized fluorescent probes for monitoring changes in mitochondrial viscosity. In this work, two series of fluorescence probes with triphenylamine unit and 2-hydroxyphenylbenzothiazole unit respectively were constructed. The impact of varying chain lengths on the photophysical properties of these probes was investigated. The results indicated that probe TPAC12 with dodecane group exhibited the largest fluorescence enhancement. The results of cell imaging experiments demonstrated that all these probes exhibited specific targeting towards mitochondria and were not influenced by changes in mitochondrial membrane potential. Additionally, probe TPAC12 facilitated the monitoring of viscosity alterations within mitochondria. Importantly, probe TPAC12 has been effectively utilized for the identification of both fatty liver and normal liver tissues. These results suggested that probe TPAC12 holds great potential as a valuable tool for detecting mitochondrial viscosity in biological systems.

A reaction-based bright red-emitting fluorescence probe for hypochlorite detection and its biological application

Monitoring the fluctuations of endogenous ClO−in vivo is very important to understand the physiological roles of ClO−and the mechanisms of ClO− relevant diseases. Herein, a new fluorescent probe, RhB-FL exhibited excellent sensitivity and anti-interference for ClO− detection, and the detection limit was as low as 4.8 × 10−7 M. Furthermore, RhB-FL was effectively applied to detect ClO− in cells.

Selectively lighting up glyoxal in living cells using an o-phenylenediamine fused hemicyanine.

Glyoxal (GL) is a reactive α-dicarbonyl compound generated from glycated proteins in the Maillard reaction. It has attracted particular attention over the past few years because of its possible clinical significance in chronic and age-related diseases. In this work, a reaction-based red emission fluorescent probe GL1 has been synthesized successfully by grafting an alkyl group onto an amino group to regulate its selectivity for GL. Under physiological conditions, the fluorescence intensity of GL1 at 640 nm obviously increased with the increase of GL concentration, and it exhibited high selectivity for GL over other reactive carbonyl compounds, as well as a lower detection limit (0.021 μM) and a larger Stokes shift (112 nm). At the same time, GL1 can selectively accumulate in mitochondria and can be used to detect exogenous and endogenous GL in living cells with low cytotoxicity.

Monoamine oxidase A probe synthesis and fluorescence imaging in cells and zebrafish based on AIE mechanism

Monoamine oxidase A (MAO-A) is a crucial mammalian flavoenzyme that plays a significant role in various neurodegenerative diseases such as depression, Parkinson’s disease, schizophrenia, and epilepsy, as well as cancer, cardiac disorders, and liver disease. Thus, a real-time and targeted MAO-A detection approach is desperately needed to study the onset and management of disorders linked to MAO-A. In this study, we have successfully synthesized a red-emitting fluorescent probe AIE-M, which is based on the specificity of MAOs for the structural catalysis of propylamine group substrates. This probe utilizes quinoline-malononitrile (QM) with AIE properties as the fluorescent master. The probe containing QM exhibits excellent aggregation-induced luminescence (AIE), biocompatibility, and high photostability, enabling specific recognition of MAO-A. Moreover, it can be employed for the detection and imaging of endogenous MAO-A in neuroblastoma cells and zebrafish, thereby offering a novel research instrument for diagnosing associated ailments.

A Red-Emitting Fluorescence Probe for Rapid Detecting Exogenous and Endogenous Peroxynitrite in Living Cells with High Sensitivity and Selectivity

Peroxynitrite (ONOO−) has been revealed to play crucial roles in many physiological and pathological processes, and many diseases were proven to be associated with its misregulated production. The development of fluorescent probes meets the need for tracking ONOO− and gives a better understanding of its diverse mechanisms. In this work, a red-emitting fluorescent probe BP-ONOO was synthesized via functionalization of the rhodol-like fluorophore with a reactive site of hydrazide. The probe BP-ONOO exhibited high sensitivity, excellent selectivity, and short response time (less than 4 s) towards ONOO− under neutral or weak alkaline conditions. These attractive properties favor its application in real-time imaging of ONOO− in living cells, and the probe has been successfully applied for imaging the concentration levels of ONOO− in RAW 264.7 macrophage cells under drug stimulation.

Red-emitting fluorescent probe for hydrogen sulfide detection and its applications in food freshness determination and in vivo bioimaging.

We report the development of a red-emitting fluorescence probe (XDS) for hydrogen sulfide (H2S) detection in biosystems, real-world food samples, and application of this probe for monitoring of H2S production during food spoilage. The XDS probe is developed by coupling of coumarin derivative to rhodanic-CN through a H2S responsive CC bond. Remarkable fluorescence quenching of XDS is observed as a result of the response to H2S. Semi-quantitative detection of H2S in three real-world water and two beer samples and monitoring of H2S production during food spoilage in real-time by "naked-eye" and smartphone colorimetric analysis are then achieved using XDS as the probe. Moreover, XDS is low toxicity, allowing it being used for visualizing endogenous and exogenous H2S in vivo in a mouse model. It is expected that the successful development of XDS could provide an effective tool for investigating the roles of H2S in biomedical system and for future food safety evaluation.

A red-emitting ultrasensitive fluorescent probe for specific detection and biological visualization of cysteine in vitro and in vivo

Developing efficient strategies for specific detection of cysteine (Cys) is of great importance for identifying complicated biological roles in physiological and pathological processes. Herein, an ultrasensitive red-emission fluorescent probe (termed 1) is constructed for specific detection and biological visualization of Cys. The linked-anthocyanin fluorophore modified with a twisted N, N-diethylamino moiety shows improved red-shifted emission (642 nm) and absolute quantum yield (0.224 in dimethyl sulfoxide), as well as minimal fluorescence background signal and good water solubility. Meanwhile, utilizing acryloyl chloride as recognition group endows the probe 1 with excellent sensitivity and selectivity towards Cys (limit of detection: 2.93 nM). More importantly, the in vitro and in vivo results confirm that the probe 1 has the capacity of fluorescence imaging of Cys and good biological safety, which holds great promise for bioanalysis and biosensing of Cys.

A simple indanone-based red emission fluorescent probe for the rapid detection of cysteine in vitro and in vivo

Cysteine is a vital biothiols that plays an important role in numerous physiological and pathological processes. The development of simple molecule tools for detection and analysis Cys in subcellar environment is significant for further exploring their pathophysiological. In this work, a simple but activated fluorescent probe AMIA was constructed with a donor-π-accepter (D- π -A) structure, which using an indanone as the electron-withdrawing unit acting as the fluorophore, dimethylamino group attached to the position 4 of the benzene ring as the electron-donating, two double bonds as the linker group, and the acryloyl ester group as the trigger and response unit. This probe AMIA was exhibited highly selective and sensitive response to Cys over other amino acids and ions under physiological conditions. It was found that AMIA showed a red turn-on fluorescence response at 630 nm towards Cys with a large stroke shift of 170 nm and a very low detection limit of 26.3 nM. HRMS, 1H NMR and TD-DFT calculation further confirmed that the response mechanism is the Cys triggered the addition-cyclization reaction between AMIA’ acryloyl group and Cys’ sulfhydryl and amino unit, leading to the release of a red fluorescent dye AMIA-OH, which can be identified by naked eyes. Furthermore, AMIA was successfully applied for simultaneous determination of Cys in living cells and zebrafish with lower cytotoxicity and good cell permeability. We hope that this novel indanone-based probe AMIA will provide a new reference for visualized Cys in other complex biological system.

Hemicyanine-based sensor for mitochondrial viscosity imaging in BV2 cells

Mitochondrial viscosity is a critical factor affecting numerous physiological processes, including phagocytosis. Abnormal viscosity in mitochondria is related to some pathological activities and diseases. Evaluating and detecting the changes in mitochondrial viscosity in vivo is crucial. Thus, a mitochondria-targeted red-emitting fluorescent probe (VP) was prepared, and can be used to detect viscosity with high selectivity and sensitivity. The synthesis of probe VP was as simple as two steps and the cost was low. In addition, the fluorescence intensity (log I615) exhibited an excellent relationship with viscosity (log η) in the range of 0.5 – 2.5 (R2 = 0.9985) in water/glycerol mixture. It is noteworthy that the probe VP displayed the highest signal-to-noise ratio (about 50-fold) for viscosity in water and glycerol system. The probe VP can visualize the mitochondrial viscosity change in living cells. More importantly, phagocytic test for BV2 cells further demonstrated that phagocytosis decreased with increased viscosity. Furthermore, VP was successfully used for monitoring the mitophagy process induced by starvation, and mitochondrial viscosity exhibited enhancement during mitophagy. The probe was a potential tool for studying viscosity and phagocytosis.

A highly selective and sensitive red-emitting fluorescent probe for bioimaging of hypobromous acid in living cells

Hypobromous acid (HOBr) is an indispensable reactive oxygen species which is necessary for the functioning of many cellular functions. However, abnormal HOBr can cause serious harm to the environment and organisms. Nowadays, fluorescent probe techniques are widely used in the detection of biomolecules in living bodies, but few fluorescent probes have been investigated for the detection of HOBr in living cells. Based on the above considerations, we synthesized a fluorescent probe RD-HOBr for detecting HOBr. Probe RD-HOBr has the characteristics of fast response (<4 s), high selectivity and naked eye detectability. Notably, because the detection limit of the probe RD-HOBr for detecting HOBr is only 1.37 nM, it has the advantage of high sensitivity to HOBr. In addition, the probe RD-HOBr can not only detect HOBr in actual water samples, but also has the advantage of low toxicity and can detect HOBr in cells sensitively. Therefore, the construction of probe RD-HOBr not only provides a new tool for the sensitive detection of HOBr, but is also expected to provide a new method for the detection of HOBr in biological organisms.

GFP-based red-emissive fluorescent probes for dual imaging of β-amyloid plaques and mitochondrial viscosity

Alzheimer’s disease (AD), with incurable neurodegenerative damage, has attracted growing interest in exploration of better AD biomarkers in its early diagnosis. Among various biomarkers, amyloid-β (Aβ) aggregates and mitochondrial viscosity are closely related to AD and their dual imaging might provide a potential and feasible strategy. In this work, five GFP-based red-emissive fluorescent probes were rationally designed and synthesized for selective detection of β-amyloid plaques and viscosity, among which C25e exhibited superior properties and could successfully image β-amyloid plaques and mitochondrial viscosity with different fluorescence wavelength signals “turn-on” at around 624 and 640 nm, respectively. Moreover, the staining of brain sections from a transgenic AD mouse showed that probe C25e showed higher selectivity and signal-to-noise ratio towards Aβ plaques than commercially-available Thio-S. In addition, the probe C25e was, for the first time, employed for monitoring amyloid-β induced mitochondrial viscosity changes. Therefore, this GFP-based red-emissive fluorescent probe C25e could serve as a dual-functional tool for imaging β-amyloid plaques and mitochondrial viscosity, which might provide a unique strategy for the early diagnosis of Alzheimer’s disease.
