Desiccation and thermo-tolerance of Fucus guiryi (Phaeophyceae) from its southernmost populations (Canary Islands, Eastern Atlantic)

Abstract Populations inhabiting the periphery of a species’ distribution suffer the immediate pressure of the environmental changes. Studying their physiology under stressful conditions is essential in the context of climate change. We characterized two morphotypes with measurements of length, thickness and width of the thallus of the macroalga Fucus guiryi in two localities (north and south) in Tenerife (Canary Islands), the southern limit of distribution of the species. We hypothesized that the individuals from the north locality, especially the large morphotype, would be more vulnerable to desiccation and high temperatures than those from the south. Tolerance to desiccation was tested through two long desiccation experiments and by comparing the vegetative and reproductive blades. Tolerance to high temperatures was tested by keeping the individuals in seawater (26–32 °C) for 24 h. The recovery of physiological functions after the stressful conditions was estimated through maximal photochemical efficiency of PSII. Results indicate that both of the two morphometrically differentiated morphotypes were tolerant to prolonged desiccation; however, the small one was more affected, particularly in the northern locality. The reproductive blades were hardly affected, indicating greater tolerance to desiccation than the vegetative blades. Fucus guiryi showed tolerance to high temperatures (32 °C) suggesting plasticity to future warming scenarios.

Use of Acupoint Stimulation to Regulate Perioperative Stress-Induced Immunosuppression

The stress response can be triggered during the perioperative period by tension, fear, anesthesia, surgical trauma, and various postoperative adverse stimuli, leading to significant changes in the endocrine, metabolic, and immune systems of patients. In particular, immunosuppression induced by the stress response has adverse effects on the postoperative recovery of patients. As acupuncture stimulation-related technologies have been rapidly developed and applied in recent years, a number of basic and clinical studies have confirmed that acupoint stimulation can regulate the immune system via local, neurological, and endocrine pathways. Moreover, acupoint stimulation treatment has also been shown to reduce the adverse effects of immunosuppression by affecting the release of various cytokines, such as interleukin (IL) 1, IL-2, IL-4, IL-6, IL-10, IFN-γ, and tumor necrosis factor (TNF) β, immunoglobulins, complement proteins, and T cell markers such as CD3, CD4, and CD4/CD8 via the regulation of macrophages, neutrophils, NK cells, and endogenous opioids. In addition, acupuncture stimulation treatment during the perioperative period can also significantly decrease the amount of anesthetic required for anesthesia, effectively reduce nausea and vomiting, relieve post-operative pain, and accelerate the recovery of physiological functions. Therefore, acupuncture stimulation treatment has shown important potential for clinical applications.

Electrospun Heparin-Loaded Core-Shell Nanofiber Sutures for Achilles Tendon Regeneration In Vivo.

Achilles tendon reconstruction surgery is the primary clinical method for repairing acute Achilles tendon ruptures. However, the efficacy of the postoperative healing process and the recovery of physiological function are inadequate. This study examines the healing mechanism of ruptured rat Achilles tendons seamed with heparin-loaded core-shell fiber sutures fabricated via near-field electrospinning. High-heparin-concentration sutures (PPH3.0) perform better than the low-heparin-concentration sutures and commercial sutures (CSs). The PPH3.0 suture recruits fewer inflammatory cells and shows good histocompatibility in peritoneal implantation experiments. Staining of the Achilles tendon rupture repair zone demonstrates that a high heparin concentration in sutures reduces immune-inflammatory responses. Immunohistochemical analysis reveals that the transforming growth factor-β staining scores of the PPH3.0 sutures are not significantly different from those of the corresponding control group but are significantly different from those of the CSs and non-heparin-loaded-suture groups. According to vascular endothelial growth factor (VEGF) analysis, the concentration of VEGF in the group treated with the PPH3.0 suture increases by 37.5% compared with that in its control group. No significant difference in tension strength is observed between the PPH3.0 group and healthy Achilles tendons. These findings illustrate that this novel method effectively treats Achilles tendon rupture and promotes healing and regeneration.

Anesthesia Management and Recovery after Laparoscopic Colorectal Surgery

Open abdominal procedures are associated with high levels of postoperative surgical stress that can impede recovery of physiological functions. Trauma resulting from aggressive manipulation of organs, and especially the intestine, can increase postoperative complications and result in delay in normal recovery. In an attempt to minimize complications and overall duration of hospital stay, fast track strategies, called “early recovery after surgery” (ERAS) protocols, have been adopted. Laparoscopic surgery has recently been identified as a major component of rapid recovery. One of the key differences between the open and the laparoscopic approach is the use of pneumoperitoneum for organ exposure during laparoscopy. Prolonged increase in abdominal pressure in addition to patient positioning can have a profound effect on hemodynamic parameters such as central venous pressure (CVP), cardiac output and intracranial pressure. Modifications in fluid administration, the choice of anesthetic agent and the depth of neuromuscular blockade are indicated to accommodate to the conditions imposed by pneumoperitoneum. Postoperatively, laparoscopic surgery is usually associated with less aggressive analgesia management. Based on ERAS protocols, patients are encouraged to initiate early oral feeding and to resume physical activity as soon as possible after abdominal surgery. While laparoscopic abdominal surgery is associated with reduced needs for pain medication, there appear to be no significant differences in early enteral feeding and mobilization between laparoscopic procedures and open surgery. When all factors are taken into consideration, however, laparoscopic colorectal surgery is associated with significant reduction in the duration of the total hospital stay, and is therefore recommended whenever feasible.

Monoterpene responses to interacting effects of drought stress and infection by the fungus Heterobasidion parviporum in two clones of Norway spruce (Picea abies)

To study simultaneous biotic and abiotic stress effects on monoterpene mediated defence response, four-year-old seedlings of two clones of Picea abies (c386: “relatively resistant” c386; c171: “relatively susceptible” clone 171) were inoculated with an isolate of rot fungus Heterobasidion parviporum and simultaneously subjected to a drought stress recovery cycle. Fungal growth into the wood was generally higher in well-watered (W) than in drought-stressed (D) plants. As predawn water potential decreased, gas exchange and maximal photochemical efficiency of PSII also declined. Water potential and chlorophyll fluorescence showed differences between clones in D plants, c386 being more tolerant than c171, while stomatal conductance and net photosynthesis differed between clones in W plants; c171 showing higher values than c386. A recovery of physiological functions was observed after re-watering. The clones showed different constitutive monoterpene profiles. The infection generally caused some extensive compositional changes in the relative contents of several monoterpenes and their enantiomers; also, sterile infection (wounded only samples) affected the relative proportions of monoterpenes, although to a lesser extent than reactions elicited by the fungus. The trend in monoterpene response to disease treatments was the same in D and W plants of both clones. In particular, higher proportion of δ-3-carene was found constitutively in c386 in comparison with c171; besides, infection with H. parviporum increased the relative contents of δ-3-carene in all the samples, even if proportions of this monoterpene in wounded plus inoculated tissues of c386 were significantly higher than cortical tissues from non-inoculated branches only in W seedlings 44 days after treatment.Although drought stress decreased the total absolute contents of monoterpenes, total monoterpene concentrations significantly increased in response to infection by H. parviporum.

Optical Imaging of the Motor Cortex Following Antidromic Activation of the Corticospinal Tract after Spinal Cord Injury.

Spinal cord injury (SCI) disrupts neuronal networks of ascending and descending tracts at the site of injury, leading to a loss of motor function. Restoration and new circuit formation are important components of the recovery process, which involves collateral sprouting of injured and uninjured fibers. The present study was conducted to determine cortical responses to antidromic stimulation of the corticospinal tracts, to compare changes in the reorganization of neural pathways within normal and spinal cord-injured rats, and to elucidate differences in spatiotemporal activity patterns of the natural progression and reorganization of neural pathways in normal and SCI animals using optical imaging. Optical signals were recorded from the motor cortex in response to electrical stimulation of the ventral horn of the L1 spinal cord. Motor evoked potentials (MEPs) were evaluated to demonstrate endogenous recovery of physiological functions after SCI. A significantly shorter N1 peak latency and broader activation in the MEP optical recordings were observed at 4 weeks after SCI, compared to 1 week after SCI. Spatiotemporal patterns in the cerebral cortex differed depending on functional recovery. In the present study, optical imaging was found to be useful in revealing functional changes and may reflect conditions of reorganization and/or changes in surviving neurons after SCI.

Wearing Compression Garment after Endurance Exercise Promotes Recovery of Exercise Performance.

The present study examined the effects of wearing a lower-body compression garment (CG) after endurance exercise on recovery of physiological function. 18 males were divided into 2 experiments, the downhill running (n=10, DHR) experiments and the level running (n=8, LR) experiments. Subjects performed 30 min of DHR (gradient: - 10%) or LR (gradient: 0%) at 70% of ˙VO2max with either wearing a CG (CG trial) or normal garment (CON trial) for 24 h after running. Changes in jump performance (counter movement jump; CMJ, rebound jump; RJ, drop jump; DJ), subjective feelings, circumferences of leg, and blood variables (creatine kinase, myoglobin, interleukin-6, high-sensitivity C-reactive protein) were evaluated before exercise, immediately after exercise, 1, 3 and 24 h following exercise. Running economy was evaluated at 24 h following exercise. CMJ height and RJ index were significantly higher in the CG trial than in the CON trial 24 h after running (P<0.05). Although changes in muscle soreness and blood variables were significantly greater in the DHR experiment than in the LR experiment, there was no significant difference between the trials in either experiment. Wearing a CG following endurance exercise facilitated recovery of jump performance under situations with severe exercise-induced muscle damage.

Perioperative Care with Fast‐Track Management in Patients Undergoing Pancreaticoduodenectomy

It has been considered that allowing patients to return to daily life earlier after surgery helps recovery of physiological function and reduces postoperative complications and hospital stay. We investigated the usefulness of fast-track management in perioperative care of patients undergoing pancreaticoduodenectomy (PD). Patients (n = 90) who received conventional perioperative management from 2005 to 2009 were included as the 'conventional group' (historical control group), and patients who received perioperative care with fast-track management (n = 100) from 2010 to March 2013 were included as the 'fast-track group'. To evaluate the efficacy of perioperative care with fast-track management, the incidence of postoperative complications and the length of hospital stay were compared between the two groups (comparative study). For statistical analysis, univariate analysis was performed using the χ (2) test or Fisher's exact test. There was no significant difference between the two groups in sex, mean age, presence/absence of diabetes mellitus, preoperative drainage for jaundice, previous disease, operative procedure, mean duration of operation, or blood loss (p < 0.01). The incidence of surgical site infection in the conventional group and fast-track group was 28.9 and 14.0 %, respectively, with a significant difference between the two groups (p = 0.019). In addition, the incidence of pancreatic fistula (grade B, C) significantly differed between the two groups (27.8 % in the conventional group, 9.0 % in the fast-track group; p = 0.001). The mean postoperative hospital stay was 36.3 days in the conventional group and 21.9 days in the fast-track group (p < 0.001). Perioperative care with fast-track management may reduce postoperative complications and decrease the length of hospital stay in patients undergoing PD.

Platelet-Rich Plasma: A Promising Product for Treatment of Peripheral Nerve Regeneration After Nerve Injury

ABSTRACTNerve regeneration includes regrowth of injured axons as well as myelination, restoration of synaptic connections and recovery of physiological functions. Platelet-rich plasma (PRP) is prepared from the patient's own blood and contains growth factors that influence wound healing and used in various surgical fields including oral and maxillofacial surgery. When platelets are activated either ex vivo or in vivo, growth factors and proteins were released from platelets' alpha granules. Recent studies proved that PRP could promote regeneration of injured peripheral nerve. This review focuses on current trials using PRP to promote nerve regeneration and repairment, and proposes potential clinical application of PRP for nerve injury in the future.

Effect of pulsed electromagnetic fields (PEMFs) on condrogenic phenotype maintenance of MSCs in presence of pro-inflammatory cytochines: preliminary results

The aim of the study was to evaluate in vitro the effects of PEMFs on maintenance over time of chondrocyte phenotype of conditioned Mesenchimal Stem Cells (MSCs), in presence of pro-inflammatory cytokines (IL-s1). MSCs, taken from bone marrow, were pellet-cultured in medium conditioning towards the chondrogenic lineage. Two targets were pursued: the first was to standardize the method to obtain chondrocyte pellets in terms of type/amount of withdrawal, time/degree of differentiation and amount of extracellular matrix production; the second was to extend over time chondrocyte differentiation, checking the phenotype maintenance, after adding pro-inflammatory IL-s1 cytokine in culture medium with/without the application of PEMFs (device provided by IGEA-Carpi). The pellets obtained were coltured for different times (21, 28, 34 days), verifying the presence of type-II collagen (as index of chondrocyte differentiation) both by means of TEM analysis and immunoreaction. The best differentiation was obtained after 28 days of culture; in such pellets the studies were performed in triplicate for 15 days, identifying four experimental conditions: 1) without IL-s1 and PEMFs; 2) with IL-s1, without PEMFs; 3) without IL-s1 and with PEMFs; 4) with IL-s1 and PEMFs. The parameters of applied PEMFs were 1.7mT and 75Hz, and the time of application was 4 hours/day. Medium was changed every 3-4 days and stored for the evaluations of PGE2 (indicative of inflammation) and proteoglycans (indicative of chondrogenic differentiation). At the end of the experiment, each pellet was fixed with paraformaldehyde 4% and embedded in paraffin; sections (5 µm thick) were obtained and stained with Toluidin Blue in order to evaluate metachromasia. The results show that: 1) only the pellets treated with IL-s1 without PEMFs did not show metachromasia, indicanting a chondrocyte de-differentiation towards fibroblastic phenotype; 2) only in pellets treated with IL-s1 and with PEMF application, after about 12 days of treatment the amount of PGE2 in medium decreases (31%), indicating the anti-inflammatory effect of PEMFs, while the proteoglycan production slightly increases (2%). In conclusion, pulsed electromagnetic fields could be proposed in preventing chondrocyte de-differentiation due to inflammation induced by IL-s1; this with the final aim to integrate regenerative medicine techniques to apply in the healing of joint cartilage lesions with bio-physic energy devices, useful to obtain a stable-in-time recovery of physiologic function of articular surfaces that suffered a severe injury.

Effects of Sleep Deprivation on Measures of the Febrile Reaction and the Recovery of Somatovisceral Functions and Sleep in Endotoxemia

Electroencephalographic methods were used to study the effects of total sleep deprivation on thermoregulatory measures of the fever response in pigeons (Columba livia): brain temperature, peripheral vasomotor reactions, thoracic muscle contractile activity, and the recovery of somatic functions and the time characteristics of waking and sleep in lipopolysaccharide (LPS)-induced endotoxemia. Sleep deprivation during the period in which the quantity of slow-wave sleep increased on administration of LPS induced decreases in the latent period of fever onset and in the duration of fever, along with more significant increases in brain temperature and the level of muscle contractile activity as compared with the effects of LPS alone. The period after sleep deprivation was characterized by more prolonged recovery of muscle contractile activity and the time characteristics of sleep and waking states, along with more prolonged compensatory "rebound" of slow-wave sleep as compared with the effects of sleep deprivation alone. Thus, sleep deprivation in endotoxemia led to decreases in the latent period of fever onset, exacerbation of fever, and increases in the latent period of recovery of physiological functions.

Study on the influence of the heating transfusion on early physiology function among the prematurities

Objective To investigate the influence of heating transfusion on early physiologic function among prematurities. Methods 80 prematurities who accepted transfusion routinely were devided into control group and experimental group randomly. There were 40 cases in each group. Solutions heated up to 35 ～ 37 ℃ was used in experimental group, routine method (room temperature) was used in control group in contrast. Got record of changes of the temperature, heart rate, respiration, oxygen saturation and time took in temperature changing. Compared complication and hospital day with each other group. Results The differences of vital sign, time took in temperature changing, incidence of complications of two groups are statistically significant. Effects of two different nursing interventions on postoperative pain of patients with thoracic tumor

Objective To explore the effects of two diffeerent nursing interventions on postoperative pain of patients with thoracic tumor. Methods Postoperative patients with thoracic tumor (126 cases) were ran-domly divided into the experimental group(64 cases) and the control group (62 cases). The experimental group combined stages of assessment methods with the three-step analgesic ladder for cancer pain which the WHO recommended. Timely and reasonable use of analgesic agents ensured control of pain effectively. The control group was given disposal only when patients complained pain. Sleep, effective cough of sputum, related indica-tors of vital signs and the time of first ambulation were observed in the two groups. Results The inci-dence of patients' abnormal respiratory rate, accelerated heart rate and decreased oxygen saturation levels in fingertip in the experimental group were significantly lower than those of the control group(P<0.05). Within 3 days after surgery, the proportion of the experimental group patients whose cough and expectoration re-flex score was good, without insomnia and with first ambulation was significantly higher than that of the control group (P<0.05). Conclusions Adoption of stages of assessment and initiative and effective pain control for patients with thoracic tumor of postoperative pain can significantly improve the quality of care, pro-mote the recovery of physiological function, and reduce the incidence of postoperative complications. Ephrin A2 May Play a Role in Axon Guidance during Hair Cell Regeneration

Hair cell regeneration in the avian cochlea is accompanied by frequency specific reinnervation and recovery of physiologic function. The molecular cues that guide ganglion cells to tonotopically appropriate new hair cells have not been identified. We investigated the potential of ephrin A2 in this process. Ephrin A2 expression was characterized in acoustic ganglion cells of normal and gentamicin-treated early post hatch chicks. Ephrin A2 expression was determined by Western analysis of total protein isolated from acoustic ganglia in normal animals. Protein localization was characterized by fluorescence immunohistochemistry in sections of acoustic ganglia of normal and gentamicin treated animals. Patterns of ephrin A2 expression in acoustic ganglia were determined and quantified during hair cell regeneration. Ephrin A2 expression was found in acoustic ganglia by Western analysis. Localization of this protein by immunofluorescence revealed its presence in acoustic ganglion cells in normal chicks. After gentamicin treatment, ephrin A2 expression was lost in a subset of acoustic ganglion cells. The spatial and temporal pattern of ephrin A2 loss coincides with the pattern of hair cell loss and regeneration. The changes in ephrin A2 immunoreactivity in acoustic ganglion cells during cochlear damage and regeneration suggests that ephrin A2 may be involved in the guidance of ganglion cells to tonotopically appropriate hair cell targets during regeneration. Protein Kinase C Mediates Repair of Mitochondrial and Transport Functions after Toxicant-Induced Injury in Renal Cells

Previously, we have shown that renal proximal tubular cells (RPTCs) recover physiological functions after injury induced by the oxidant tert-butylhydroperoxide (TBHP), but not by the nephrotoxic cysteine conjugate dichlorovinyl-l-cysteine (DCVC). This study examined the role of protein kinase C (PKC) in the repair of RPTC functions after sublethal injury produced by these toxicants. Total PKC activity decreased 65 and 86% after TBHP and DCVC exposures, respectively, and recovered in TBHP-injured but not in DCVC-injured RPTCs. Mitochondrial function, active Na+ transport, and Na+-dependent glucose uptake decreased after toxicant exposure and recovered in TBHP- but not in DCVC-injured RPTCs. PKC inhibition decreased the repair of RPTC functions after TBHP injury. PKC activation promoted recovery of mitochondrial function and active Na+ transport in TBHP- and DCVC-injured RPTCs but had no effect on recovery of Na+-dependent glucose uptake. We conclude that in RPTCs, 1) total PKC activity decreases after TBHP and DCVC injury and recovers after TBHP but not after DCVC exposure, 2) recovery of PKC activity precedes the return of physiological functions after oxidant injury, 3) PKC inhibition decreases recovery of physiological functions, and 4) PKC activation promotes recovery of mitochondrial function and active Na+ transport but not Na+-dependent glucose uptake. These results suggest that the repair of renal functions is mediated through PKC-dependent mechanisms and that cysteine conjugates may inhibit renal repair, in part, through inhibition of PKC signaling.

The problem of recovery of physiological functions in humans in deep accidental hypothermia (to the problem of limits of physiological adaptation)

At present, the recovery of physiological functions in a cooled man after different kinds of accidents and sea catastrophes (accidental hypothermia) is a topical task of emergency medicine and, at the same time, an important problem of thermobiology, thermoregulation, and bioenergetics. This work outlines the disturbances of the physiological functions of the body at different stages of the development of accidental hypothermia, the main modern methods of recovery of these functions, and some unresolved problems of the physiology of resuscitating hypothermia victims. New achievements in the study of the mechanisms of cold-induced disorders of physiological functions are presented in this work, and the correction of the physiological methods for restoring the life activity of a cooled man is proposed.

Effect of Corrective Make-Up Training on Patients with Facial Nerve Paralysis

Two women who suffered from facial nerve paralysis following surgical resection of a posterior fossa tumor were instructed by an expert beautician in the application of make-up to correct their facial imbalances. The results of a personality assessment administered before and after instruction showed positive changes including reductions in depression and feelings of inferiority. Postoperative restoration of normal facial appearance and recovery of physiologic function are related and important concerns to patients. It is noteworthy that daily use of simple low-cost make-up techniques can significantly reduce the "psychological load" of patients with facial paralysis during the recovery period.
