Binding of adenosine analogs with type A1 purine receptors in the guinea pig ileum

Several N6-adenosine analogs were synthesized and their A1-purine receptor binding affinities were determined. Our results demonstrate that N6-allyl- and N6-aminopropanol-adenosine increase affinities to A1-purine receptor in guinea pig ileum.

A Synthetic Glycopeptide of Substance P Analogue (SP6–11) with Enhanced NK-1 Receptor Specificity

A new glycopeptide analogue of substance P (6–11) (SP6–11), namely, N1.6 (β-D-glucopyranosyl) [Glu6, Pro9]SP6–11, has been synthesized and found to be water soluble. The in vitro biological activity of this glycopeptide was determined for spasmogenic activity in the guinea pig ileum and for potentiation of electrically evoked contractions in the rat vas deferens. Thus, activities on NK-1, NK-2, and NK-3 receptor types have been differentiated by two assays and, in the case of NK-1 and NK-3, receptors in guinea pig ileum (GPI) were assayed using specific pharmacological procedures. The ED50 values for the analogue and reference peptides substance P (SP), neurokinin A(NKA), and neurokinin B (NKB) were determined and potencies relative to SP were calculated. The analogue is three times more potent than the potent NK-1 agonist SP on NK-1 receptors. Moreover, this glycopeptide proved to be as selective for the NK-1 receptor as the specific agonist SPOMe (the methyl ester of substance P).

Pharmacological differentiation of presynaptic M 1 muscarinic receptors modulating acetylcholine release from postsynaptic muscarinic receptors in guinea-pig ileum

1. 1. Effects of three muscarinic antagonists on electrically evoked ACh release and contractile response were investigated in longitudinal muscle strips of guinea-pig ileum suspended in an organ-bath and superfused with Krebs solution. ACh release was determined by a specific radioimmunoassay. 2. 2. Telenzepine, a selective M 1 muscarinic antagonist, increased the ACh release at a concentration of 100-fold less than that inhibiting the contractile response (10 vs 1000 nM). 3. 3. AF-DX 116, a cardioselective M 2 muscarinic antagonist, inhibited the contractile response at 10 μM, but did not affect the ACh release at this concentration. 4. 4. (−) N-Methylscopolamine (NMS) did not affect the ACh release, but inhibited the contractile response at all concentrations tested (1–1000 nM), indicating (−)NMS can be used as an ileal specific postsynaptic muscarinic antagonist. 5. 5. These data demonstrate that presynaptic muscarinic receptors modulating ACh release are distinct from postsynaptic ones involved in the contractile response and can be classified as M 1 subtype.

Antagonism at GABA B receptors by saclofen and related sulphonic analogues of baclofen and GABA

Saclofen (the direct sulphonic analogue of baclofen) is a competitive antagonist of baclofen at GABA B receptors in guinea pig ileum and rat cortical slices (estimated pA 2 = 5.3), at least twice as potent as 2-hydroxy-saclofen (pA 2 = 5). A series of related sulphonic analogues also antagonised baclofen in the guinea pig ileum, including 2-hydroxy-saclofen amide (pA 2 = 3.3), 3-amino-2-hydroxy-2-phenyl-propylsulphonic acid (pA 2 = 3.5), 3-amino-2-(benzo-(b)-furan-2-yl)-propylsulphonic acid (pA 2 = 4.3), and 3-amino-2-(4-chlorophenyl)-prop-l-enesulphonic acid (pA 2 = 2.5–3), but none were more active than saclofen which is the most potent specific GABA B antagonist yet found.

Involvement of M1 cholinergic receptors and enteric nerves in the spasmogenic activity of bacterial N-formyl oligopeptides on guinea-pig ileum.

Bacterial N-formyl-methionyl oligopeptides are spasmogenic for guinea-pig ileum in vitro but the mechanism of this effect is not understood. To investigate this phenomenon further, we have determined pA2 values (the negative logarithm of the concentration of an antagonist reducing a double-dose agonist response to a single-dose response) for a number of potential antagonists of N-formyl-met-leu-phe (F-met-leu-phe) using histamine, acetylcholine, 5HT and substance P as control agonists. Atropine, pirenzepine and tetrodotoxin were potent inhibitors of F-met-leu-phe induced contraction (pA2's 8.4, 8.0 and 7.9, respectively) suggesting involvement of neural and cholinergic pathways in the response. Sulphasalazine, known to block the F-met-leu-phe receptor on neutrophil leucocytes, was also a potent inhibitor. Tachyphylaxis induced by either 5HT, or substance P, did not diminish the response to F-met-leu-phe, suggesting that these potential mediators were not involved. These studies indicate that bacterially synthesized formyl-methionyl oligopeptides bind to cells bearing receptors in guinea-pig ileum and produce muscle contraction via enteric cholinergic (M1) neural pathways.

The affinity of some acetylenic analogues of 4-DAMP methobromide for muscarinic receptors in guinea-pig ileum and atria.

1. The replacement of 4-hydroxy-N-methyl piperidine (HO NMe) in 4-diphenylacetoxy-N-methyl piperidine (4-DAMP) metho-bromide by 4-hydroxy-but-2-ynylamines (HOCH2C=CCH2NR2) reduces the affinity for muscarine-sensitive acetylcholine receptors in guinea-pig ileum and atria. It does not abolish selectivity. The tertiary amines are more active and more selective than the corresponding quaternary salts. 2. Analogous derivatives of 4-hydroxy-but-2-ynylamines which lack the ester group (i.e. substituted 4-hydroxymethyl-propynyl amines) are less active and less selective. The quaternary compounds are more active than the tertiary bases. 3. The diphenylcarbamyl ester of 4-hydroxy-N-methylpiperidine methobromide has less than one-thousandth of the activity of the diphenylacetyl ester (4-DAMP methobromide) and is not selective. 4. Although 4-diphenylacetoxy-butynyl dimethylamine is only about one-hundredth as active as 4-DAMP methobromide it appears to have comparable selectivity. It is an interesting compound because it is a tertiary amine and should cross membranes.

Bradykinin analogues: differential agonist and antagonist activities suggesting multiple receptors.

Bradykinin analogues with specific antagonist activity in several bioassays were evaluated for effects on [3H]-bradykinin receptor binding sites and inositol phosphate production in neuroblastoma N1E-115 cells. The analogues varied in their affinities for bradykinin receptors in guinea-pig ileum and N1E-115 cell membranes, in their effects on uterine and ileal contractions and in their agonist or antagonist activity on phosphoinositide turnover in N1E-115 cells. These tissue specific effects suggest the presence of multiple bradykinin receptor subtypes.

Desethylaprophen: A Metabolite of Aprophen with Antimuscarinic Activities

The metabolic fate of aprophen* hydrochloride (2-diethyl-aminoethyl 2,2-diphenylpropionate) was studied in rats after intravenous administration. Both 14C-labeled and unlabeled aprophen were used in these studies. Blood samples were collected and analyzed to determine the identities of the metabolites formed. Utilizing high-performance liquid chromatography, desethylaprophen was identified as a major metabolite in ether-extracted samples from rats, and could be detected in blood samples 1 min after intravenous administration. It was most likely formed by N-de-ethylation of aprophen by a cytochrome P-450-dependent monooxygenase. Synthetic desethylaprophen was found to possess cholinolytic activity (i.e., it functioned as a muscarinic antagonist by blocking the contraction of acetylcholine-stimulated guinea pig ileum, the release of α-amylase from pancreatic acinar cells stimulated by carbachol, and also by inhibiting the binding of [3H]N-methyl scopolamine to the muscarinic receptors of guinea pig ileum). It was interesting that although the biological effects of desethylaprophen were 100-fold lower than those of aprophen, it was equally able to compete for the binding sites of muscarinic receptors of the guinea pig ileum.

Inactivation of dynorphin-(1-8) in isolated preparations by three peptidases.

Inactivation of dynorphin-(1-8) in three in vitro isolated preparations, guinea-pig ileum, mouse vas deferens and rabbit vas deferens, was estimated by employing the relatively specific inhibitors of enkephalin-hydrolyzing enzymes. All three enzyme inhibitors, amastatin, captopril and phosphoramidon, significantly enhanced the inhibitory potency of dynorphin-(1-8) in the three isolated preparations. The magnitude of the enhancement of the dynorphin potency by captopril was significantly higher than that by either amastatin or phosphoramidon in guinea-pig ileum; that by amastatin was significantly higher than that by either captopril or phosphoramidon in rabbit vas deferens; and that by amastatin was similar to that by captopril, but significantly higher than that by phosphoramidon in mouse vas deferens. The Ke values of three antagonists, naloxone, Mr 2266 and ICI 154129, against dynorphin-(1-8) in the presence of the three peptidase inhibitors indicated that dynorphin-(1-8) acted on kappa receptors in guinea-pig ileum and on both kappa and delta receptors in mouse vas deferens. Since amastatin, captopril and phosphoramidon produced the naloxone-reversible inhibition of contractions of guinea-pig ileum in the presence of dynorphin-(1-8), all three dynorphin-inactivating enzymes were indicated to be located very close to kappa receptors.

Muscarinic receptors on nerves and muscles in opossum esophagus muscularis mucosa

The muscarinic receptors of muscularis mucosa have some recognition properties that suggest they resemble receptors of the M1 subtype. The nerves of these tissues also contain muscarinic receptors which inhibit tonic contractions caused by release of a substance-P-like material by field stimulation. These receptors also appear to be M1 in type as they are maximally activated by McNeil A343 as well as by carbachol (pD2, 5.5 and 7.5, respectively). They are also inhibited by pirenzepine, as well as by atropine (negative logarithms of the required dose for 50% inhibition or potentiation, 6.6-6.7 compared with 8.2-8.3). Hexahydrosiladifenidol, an antagonist selective or M2 receptors of guinea pig ileum, had a low (approximately 7.1) pA2 value for antagonism of both agonists in smooth muscle in this tissue. However, it was closer to atropine in potency with respect to potentiating tonic responses to field stimulation or to inhibiting phasic responses to field stimulation than it was to antagonizing smooth muscle contractions. Thus, atropine was about 40 times more potent than pirenzepine and 2-5 times more potent than hexahydrosilafenidol. There were some quantitative differences in the effectiveness of these three antagonists in blocking the phasic (acetylcholine-mediated) response to field stimulation. Atropine was 70-100 times more potent than pirenzepine and 8-25 times more potent than hexahydrosiladifenidol. This greater potency difference for inhibition of phasic contractions compared with potentiation of tonic contractions was discussed. This tissue appears to be one of the first smooth muscles in which both nerves and muscles contain muscarinic receptors with some recognition properties resembling those of the M1 subtype.

ChemInform Abstract: Differential Blockade of Muscarinic Receptor Subtypes by Polymethylene Tetraamines. Novel Class of Selective Antagonists of Cardiac M‐2 Muscarinic Receptors.

Several N,N'-bis[6-[(2-methoxybenzyl)amino]hexyl]-1, omega-alkanediamine tetrahydrochlorides were synthesized and evaluated for their blocking activity on muscarinic receptors in guinea pig atria and rat ileum and bladder. The results were compared with those obtained for the classical nonselective muscarinic antagonist atropine. It was discovered that optimum activity is associated with an eight-carbon chain (compound 4) in guinea pig atria whereas, in both rat ileum and bladder, the 12-carbon analogue 7 had the highest activity. In addition, polymethylene tetraamines 1-6 displayed high selectivity toward guinea pig atria muscarinic receptors. The discriminatory power of 1-6 was not shared by 7. All the tetraamines were shown to be competitive antagonists of muscarinic receptors. N,N'-Bis[6-[(2-methoxybenzyl)amino]hexyl]-1,8-octanediamine was the most potent and selective toward muscarinic receptors in atria, with a pA2 value of 8.13 and a selectivity ratio (atria vs. ileum or bladder) of ca. 270. At a concentration of 10 microM, tetraamine 4 did not affect histamine and 5-hydroxytryptamine receptors of guinea pig ileum or alpha-adrenoreceptors of guinea pig atria whereas it inhibited postsynaptic alpha-adrenoreceptors of rat vas deferens with a -log K value of 5.23 and nicotinic receptors of frog rectus abdominis with an IC50 value of 0.23 microM. It is concluded that 4 is a novel, powerful, and selective tool in the characterization of muscarinic receptor subtypes.

Nature of the N6 region of the adenosine receptor in guinea-pig ileum and rat vas deferens.

This study explored the nature of the purine domain N6 regions of the presynaptic adenosine receptors of guinea-pig ileum and of rat vas deferens. The experimental design tested a model of these receptors which is complementary to the structure of the N6 substituent of the classical A1 adenosine receptor agonist N6-1-phenyl-2R-propyladenosine, (R-PIA). Assays of activity employed ileal segments or the midportions of vasa deferentia under continuous electrical stimulation at 0.2 Hz. Structure activity correlations compared the EC-50s for twitch inhibition. As shown previously, R-PIA was 60-80 times more potent than its S diastereomer, the resultant of the positive contribution of propyl C-3 to activity as well as the negative influence of steric hindrance exerted by propyl C-3 of the S diastereomer. Other pairs of diastereomers having a chiral center adjacent to N6 showed that the stereoselectivity of the PIAs was generalizable. Biological activity appears to reside wholly in the N6 alkyl moiety; the phenyl or aryl groups of similar size actually diminished potency. The receptor subregions interacting with propyl C-1 and C-3 of R-PIA are each large enough to accommodate two - but not three - methylene residues, each methylene contributing additively to activity. Hydrophobicity is a prominent attribute of the propyl C-1 and C-3 subregions. The potencies of these analogs as inhibitors of presynaptic transmission in ileum or vas deferens are covariant with inhibition of [3H]N6-cyclohexyl-adenosine binding to rat cerebral cortical membranes. Singular exceptions to this generalization may represent organ- or species-dependent differences in receptor fine structure.

5-Aminovaleric acid interactions with GABAA and GABAB receptors in guinea-pig ileum.

The potential interaction of 5-aminovaleric acid (5-AVA) on GABAA and GABAB receptors was investigated on the guinea-pig isolated ileum myenteric plexus preparation. In the unstimulated preparation 5-AVA (0.1-3 mM) produced a transient contraction which was abolished by previous exposure to picrotoxin (0.1 mM), atropine (3 microM) or tetrodotoxin (0.3 microM). Cross desensitization was observed between the contractile effects of 5-AVA and GABA, while a previous exposure to (+/-)-baclofen did not affect 5-AVA induced contractions. 5-AVA antagonized the relaxant effect of (+/-)-baclofen in unstimulated preparations. 5-AVA (1 mM) had no effect on amplitude of twitches in supramaximally stimulated preparations while GABA produced a concentration related inhibition. In the presence of 5-AVA (1 mM) the concentration response curve to GABA was shifted to the right without a reduction of the maximal effect attainable. These observations indicate that 5-AVA interacts with both GABAA and GABAB receptors in guinea-pig ileum. The concentrations required to observe a GABAA effect are of the same order as those which are effective in producing a blockade of GABAB mediated responses.

Muscarinic-receptor stimulation enhances polyphosphoinositide breakdown in guinea-pig ileum smooth muscle.

Muscarinic-receptor stimulation by 0.1 mM-carbachol in longitudinal muscle of the guinea-pig ileum increases the incorporation of [3H]inositol into inositol-containing phospholipid. This effect was blocked by 16 microM-atropine. After 60 min incubation, carbachol increased the accumulation of total inositol phosphates 20-fold in the presence of 10 mM-Li+. Less than 20% of the total inositol phosphate corresponded to inositol 1-phosphate by ion-exchange chromatography, whereas of the remainder about two-thirds corresponded to inositol bisphosphate and one third to inositol trisphosphate. It is concluded that stimulation of muscarinic receptors in guinea-pig ileum enhances breakdown of polyphosphoinositides, suggesting that this may be a primary event associated with Ca2+ mobilization in the guinea-pig ileum.

A photoaffinity reagent to label the opiate receptors of guinea pig ileum and mouse vas deferens.

An enkephalin derivative, [D-Ala2,Leu5]enkephalin N-[(2-nitro-4-azidophenyl)amino]ethylamide, has been synthesized as a photoaffinity label for the opiate receptor. This compound retains the full biological activity of [D-Ala2,Leu5]enkephalin in guinea pig ileum and mouse vas deferens tests with IC50 values of 4.4 and 2.6 nM, respectively, and inhibits the binding of [3H]naloxone to rat brain membrane preparation with an IC50 value of 2.5 nM. Photolysis of a muscle strip of the guinea pig ileum or of the mouse vas deferens in the presence of the peptide derivative caused irreversible inhibition of electrically stimulated contractions with high efficiencies (80 and 66%, respectively), while the inhibitory effect in the dark was fully reversed by washing. This irreversible inhibition during photolysis was completely prevented by the presence of [D-Ala2,Leu5]enkephalin. These results demonstrate that [D-Ala2,Leu5]-enkephalin N-[(2-nitro-4-azidophenyl)amino]ethylamide is a prominent candidate as a photoaffinity label for the opiate receptor.

Autoradiographic localisation of muscarinic receptors in guinea-pig intestine: Distribution of high and low affinity agonist binding sites

Muscarinic receptors mediate a variety of intestinal functions including smooth muscle contraction, ganglionic transmission and water and electrolyte secretion. In this study, we have used [ 3H]quinuclidinyl benzilate ([ 3H]QNB) in an in vitro autoradiographic method to map the distribution of muscarinic receptors in guinea-pig ileum, colon and caecum. In addition, the relative distribution of low and high affinity agonist binding sites was assessed by the addition of the muscarinic agonist, carbachol, to selectively inhibit the binding of [ 3H]QNB to the high affinity sites. Although quantitative differences existed, the overall distribution of muscarinic receptors was similar in the 3 regions of intestine examined. Autoradiograph grains were found distributed over the myenteric and sub-mucous plexuses, the longitudinal and circular muscle layers and in the case of the colon, the muscularis mucosa. The inclusion of carbachol demonstrated that a greater proportion of high affinity sites were associated with the musculature than with the enteric plexuses. These findings are discussed in relation to the role of muscarinic mechanisms in intestinal motility and secretion.

Muscarinic receptors in guinea pig ileum. A study by agonist--3H-labelled antagonist competition.

A series of muscarinic agonists (n = 18) derived from dioxolane, furan, piperidine, and thiacyclohexane ring structures which have a degree of conformational restraint have been examined in guinea pig ileal smooth muscle. Binding to muscarinic receptors was determined by competition with 3H-labelled (-)-quinuclidinyl benzilate. Agonist binding was characterized by shallow displacement curves and Hill coefficients (nH) of less than 1. Binding (K1) was resolved into high (K1) and low (K2) affinity components. Correlations between drug-receptor binding and pharmacological activity were examined. A good correlation was found between K1 and K2 (pK1 = 1.33 X pK2 - 0.1), suggesting that the structural requirements for binding at low and high affinity sites are similar. Good correlations also exist between pK1 and pD2 (pK1 = 0.63 X pD2 + 1.27, furans and dioxolanes; pK1 = 0.71 X pD2 + 1.30, piperidines and thiacyclohexanes) confirming that a pharmacologically potent agonist has a high binding affinity. Although K1 represents only an "average" binding affinity its correlation with pD2 is better than those between pK1 or pK2 and pD2. The significance of these several correlations is discussed and some general limitations to the correlation of binding and response data at pharmacological receptors are noted.

The effects of pH on the affinity of pirenzepine for muscarinic receptors in the guinea-pig ileum and rat fundus strip.

1 Dose-ratios obtained with pirenzepine on the guinea-pig ileum at 30 degrees C are indistinguishable from those obtained at 37 degrees C. 2. In 0.1 M NaCl at 37 degrees C the pKa of pirenzepine for the loss of its last ionizable proton is 8.2. The ionization of pirenzepine is therefore markedly affected by changes in pH in the physiological range. 3 In experiments with pirenzepine on guinea-pig ileum and rat fundus made over a range of pH, the dose-ratio increases with the proportion of the protonated form present. As expected, the slope of the graph of dose-ratio against proportion protonated depends on the concentration of antagonist. The changes in pH produce only small effects on dose-ratios obtained with pirenzepine monomethiodide. These effects of pH can account for some of the differences between estimates of the affinity of pirenzepine. 4 The logarithm of the affinity constant of the protonated form of pirenzepine for the receptors in guinea-pig ileum is estimated to be 6.93, compared with 6.94 for the receptors in rat fundus. However, for the non-protonated form the values appear to be below 5 for the ileum compared with about 6.4 for the rat fundus.
