Breaking boundaries in microbiology: customizable nanoparticles transforming microbial detection.

The detection and identification of microorganisms are crucial in microbiology laboratories. Traditionally, detecting and identifying microbes require extended periods of incubation, significant manual effort, skilled personnel, and advanced laboratory facilities. Recent progress in nanotechnology has provided novel opportunities for detecting and identifying bacteria, viruses, and microbial metabolites using customized nanoparticles. These improvements are thought to have the ability to surpass the constraints of existing procedures and make a substantial contribution to the development of rapid microbiological diagnosis. This review article examines the customizability of nanoparticles for detecting bacteria, viruses, and microbial metabolites and discusses recent cutting-edge studies demonstrating the use of nanotechnology in biomedical research.

Exploring current and emerging therapies for porphyrias.

Porphyrias are rare, mostly inherited disorders resulting from altered activity of specific enzymes in the haem synthesis pathway that lead to accumulation of pathway intermediates. Photocutaneous symptoms occur when excess amounts of photoreactive porphyrins circulate in the blood to the skin, whereas increases in potentially neurotoxic porphyrin precursors are associated with neurovisceral symptoms. Current therapies are suboptimal and their mechanisms are not well established. As described here, emerging therapies address underlying disease mechanisms by introducing a gene, RNA or other specific molecule with the potential to cure or slow progression of the disease. Recent progress in nanotechnology and nanoscience, particularly regarding particle design and formulation, is expanding disease targets. More secure and efficient drug delivery systems have extended our toolbox for transferring specific molecules, especially into hepatocytes, and led to proof-of-concept studies in animal models. Repurposing existing drugs as molecular chaperones or haem synthesis inhibitors is also promising. This review summarizes key examples of these emerging therapeutic approaches and their application for hepatic and erythropoietic porphyrias.

Geometric Control of Cell Behavior by Biomolecule Nanodistribution.

Many dynamic interactions within the cell microenvironment modulate cell behavior and cell fate. However, the pathways and mechanisms behind cell-cell or cell-extracellular matrix interactions remain understudied, as they occur at a nanoscale level. Recent progress in nanotechnology allows for mimicking of the microenvironment at nanoscale in vitro; electron-beam lithography (EBL) is currently the most promising technique. Although this nanopatterning technique can generate nanostructures of good quality and resolution, it has resulted, thus far, in the production of only simple shapes (e.g., rectangles) over a relatively small area (100 × 100 μm), leaving its potential in biological applications unfulfilled. Here, we used EBL for cell-interaction studies by coating cell-culture-relevant material with electron-conductive indium tin oxide, which formed nanopatterns of complex nanohexagonal structures over a large area (500 × 500 μm). We confirmed the potential of EBL for use in cell-interaction studies by analyzing specific cell responses toward differentially distributed nanohexagons spaced at 1000, 500, and 250 nm. We found that our optimized technique of EBL with HaloTags enabled the investigation of broad changes to a cell-culture-relevant surface and can provide an understanding of cellular signaling mechanisms at a single-molecule level.

Nanotechnology in reproduction, breeding and conservation of fish biodiversity: Current status and future potential

AbstractThe reproductive capability of an organism is the main factor to determine the stability of the population. Till now, numerous reproductive strategies have been used, to successfully breed the fish in captivity but the desired performance is yet to be achieved. Nanotechnology has transformed the reproductive capacity of higher animals to a greater extent and has a huge potential in the aquaculture sector too. The use of nanotechnology in fish reproduction and breeding is limited to the delivery of drugs, hormones, vaccines, or genes and still, an immense scope is left in the area that needs to be explored. Here, the recent progress of nanotechnology in fish reproduction, breeding and conservation of germplasm is reviewed. Further, the key successes in the development of cryopreservation, semen imaging, sperm sorting, germplasm preservation, prototypes of smart nanobiosensors and drug delivery systems in animals including fish are highlighted. The potential future opportunities for nanotechnology to enhance reproductive output and conservative management of fish have been presented. In general, the current review summarises some of the applications of nanotechnology that are performing well in higher animals and could be a game‐changer in manipulating the reproductive potential or conserving the fish germplasm.

Convective Heat Transport in Bidirectional Water Driven Hybrid Nanofluid Using Blade Shaped Cadmium Telluride and Graphite Nanoparticles under Electromagnetohydrodynamics Process

The recent progress in nanotechnology provides the concept of hybrid-class nano-fluids having advanced thermal features comparing to regular nanofluids. The idea of a hybrid nanofluid has motivated many researchers because of its convincible performance in thermal systems. The novel theme of the present effort is to scrutinize the consequences of convective heat transfer in bidirectional water driven hybridclass nanofluid involving blade shaped cadmium telluride CdTe and graphite C nanoparticles with electromagnetohydrodynamics (EMHD) process. The transport equations representing the aforementioned topic are firstly nondimensionalized by using scaling-group transformation and then tackled by the Keller-box method, numerically. The significant results for pertinent parameters have been simulated and presented graphically as well as in tabular forms. Coefficients of drag forces are diminished with the more loadings of cadmium telluride and graphite nanoparticles and opposite results are noticed in the case of the Nusselt number. Heat transport has been improved significantly with more loadings of nanoparticles from 1 wt% to 10 wt%. A comparison benchmark for a limited version of the investigation is made with the previously published data.

Adsorptive removal of Ag/Au quantum dots onto covalent organic frameworks@magnetic zeolite@arabic gum hydrogel and their catalytic microwave-Fenton oxidative degradation of Rifampicin antibiotic

Recent progress in nanotechnology via incorporation of small particle size as quantum dots (QDs) (1–10 nm) in many industrial activities and commercial products has led to significant undesired environmental impacts. Therefore, QDs removal from wastewater represents an interesting research topic with a lot of challenges for scientists and engineers nowadays. In this work, the coagulative removal of metal quantum dots as silver and gold from industrial water samples is explored. A novel biosorbent was assembled via binding of covalent organic frameworks (COFs) with magnetic zeolite and Arabic gum hydrogel (COFs@MagZ@AGH) as a promising removal material for Ag-QDs and Au-QDs. This was fully characterized by EDX, SEM, TEM, FT-IR, XPS, XRD and surface area and applied in coagulative removal of Au-QDs and Ag-QDs in presence of several experimental factors as pH, presence of other electrolytes, stirring time, initial QDs concentration, coagulant dosage, and temperature in order to optimize the removal processes. At optimum conditions, COFs@MagZ@AGH was able to recover 99.19% and 87.57% of Ag-QDs and Au-QDs QDs, respectively via chemical adsorption mechanism with perfect fitting to pseudo-second order model. Reuse of the recovered Ag/Au-QDs@COFs@MagZ@AGH as efficient catalysts in catalytic degradation of Rifampicin antibiotic (Rf) from water was additionally investigated and optimized via microwave-Fenton catalysts with excellent oxidative degradation efficiency (100%). Reusability and applicability of the biosorbent (COFs@MagZ@AGH) and catalysts (Ag/Au-QDs@COFs@MagZ@AGH) in real industrial water samples were also explored and successfully accomplished.

Quantification of Nanoscale Dose Enhancement in Gold Nanoparticle-Aided External Photon Beam Radiotherapy.

Simple SummaryGold nanoparticles (AuNPs) have become common in radiation oncology research through the past decades. Their radiosensitization effect could offer a novel approach to cancer diagnosis and radiotherapy as well. The aim of this study is the assessment of dose enhancement attributed to the incubation of AuNPs in an irradiated target. The present work is focused on investigating the impact of AuNPs properties in dose increase under different irradiation conditions using 6 MV photon beams with and without a flattening filter via Monte Carlo simulations. Results from the simulated scenarios depict a sufficient dose raise especially at close distances to AuNPs, depending on the presence of a flattening filter in the path of the photon beam, AuNPs size, the type of modeled distribution and their concentration. Therefore, the obtained enhanced dose deposition due to AuNPs presence in an irradiated region could lead to more sufficient tumor cell destruction than irradiation alone.The recent progress in Nanotechnology has introduced Gold Nanoparticles (AuNPs) as promising radiosensitizing agents in radiation oncology. This work aims to estimate dose enhancement due to the presence of AuNPs inside an irradiated water region through Monte Carlo calculations. The GATE platform was used to simulate 6 MV photon histories generated from a TrueBeam® linear accelerator with and without a Flattening Filter (FF) and model AuNPs clusters. The AuNPs size, concentration and distribution pattern were examined. To investigate different clinical irradiation conditions, the effect of field size, presence of FF and placement of AuNPs in water were evaluated. The range of Dose Enhancement Factors (DEF = DoseAu/DoseWater) calculated in this study is 0.99 ± 0.01–1.26 ± 0.02 depending on photon beam quality, distance from AuNPs surface, AuNPs size and concentration and pattern of distribution. The highest DEF is reported for irradiation using un-flattened photon beams and at close distances from AuNPs. The obtained findings suggest that dose deposition could be increased in regions that represent whole cells or subcellular targets (mitochondria, cell nucleus, etc.). Nevertheless, further and consistent research is needed in order to make a step toward AuNP-aided radiotherapy in clinical practice.

Thermal applications of copper oxide, silver, and titanium dioxide nanoparticles via fractional derivative approach

The thermal investigation is encountered by utilizing the copper oxide (CuO), silver (Ag), and titanium dioxide (TiO2) nanoparticles. The thermal selection of these three types of nanoparticles is associated with impressive characteristics and stability. The comparative results for the hybrid nanofluid and modified hybrid models are pronounced. The semi-analytical simulations are performed via the Laplace technique for nanoparticles temperature and velocity inspection. The significances of the fractional parameter and volume fraction are observed for the velocity profile.

Antibacterial and antibiofilm efficacy of Ag NPs, Ni NPs and Al2O3 NPs singly and in combination against multidrug-resistant Klebsiella pneumoniae isolates

BackgroundAlthough traditional antibiotic therapy provided an effective approach to combat pathogenic bacteria, the long-term and widespread use of antibiotic results in the evolution of multidrug-resistant bacteria. Recent progress in nanotechnology offers an alternative opportunity to discover and develop novel antibacterial agents. MethodsA total of 51 K. pneumoniae strains were collected from several specimens of hospitalized patients and identified by two parallel methods (biochemical tests and Vitek-2 system). The antibiotic sensitivity of isolates was evaluated by disk diffusion antibiogram and Vitek-2 system. The biofilms formation ability of antibiotic-resistant strains was examined by microtiter plate and tube methods based on crystal violet staining. The molecular technique was used to determine key genes responsible for biofilms formation of clinical isolates. The antibacterial and antibiofilm activities of Ag NPs, Ni NPs, Al2O3 NPs singly (NPs) and in combination (cNPs) were investigated against selected strains using standard methods. Moreover, the cytotoxicity of NPs was evaluated on mouse neural crest-derived (Neuro-2A) cell line. ResultsThe results of bacterial studies revealed that more than 80 % of the isolates were resistant to commonly used antibiotics and about 95 % of them were able to form biofilms. Moreover, the presence of fimA and mrkA genes were determined in all biofilm-producing strains. The results of antibacterial and antibiofilm activities of NPs and cNPs demonstrated the lower MIC and MBEC values for Al2O3 NPs singly as well as for Ag/Ni cNPs and Ag/Al2O3 cNPs in combination, respectively. Overall, the inhibitory effects of cNPs were superior to NPs against all strains. Furthermore, the results of the checkerboard assays showed that Ag NPs act synergistically with two other NPs against multidrug-resistant Klebsiella pneumoniae (MDR-K. pneumoniae) isolates. The in vitro cytotoxicity assay revealed no significant toxicity of NPs against Neuro-2A cells. ConclusionIn the present study, the combination of Ag NPs, Ni NPs, and Al2O3 NPs were used against MDR-K. pneumoniae strains and antibacterial and antibiofilm activities were observed for Ag/Ni cNPs and Ag/Al2O3 cNPs.

Ultrasmall FeS2 Nanoparticles-Decorated Carbon Spheres with Laser-Mediated Ferrous Ion Release for Antibacterial Therapy.

Recent progress in nanotechnology and the ancient use of sulfur in treating dermatological disorders have promoted the development of nano-sulfides for antimicrobial applications. However, the variable valences and abundant forms of nano-sulfides have complicated investigations on their antibacterial activity. Here, carbon nanospheres (CNSs) with decoration of ultrasmall FeS2 nanoparticles (CNSs@FeS2 ) is synthesized, and their antibacterial ability and mechanism are explored. The CNSs@FeS2 released Fe2+ and sulfur ions simultaneously through dissolution and disproportionation. In vitro study indicated that the released Fe2+ killed bacteria by increasing the oxidative state of bacterial surfaces and intracellular molecules. Importantly, the released sulfur exhibited a protective effect on Fe2+ , ensuring the stable existence of Fe2+ to continuously combat bacteria. Moreover, the carbon shells of CNSs@FeS2 not only prevented the aggregation of FeS2 but also accelerated the release of Fe2+ through photothermal effects to achieve synergistic hyperthermia/Fe2+ therapy. In vivo experiments indicated that treatment with CNSs@FeS2 resulted in a marked reduction in bacterial number and improvement in survival in an acute peritonitis mouse model, and antibacterial wound experiments demonstrated high efficacy of CNSs@FeS2 -enabled synergistic hyperthermia/Fe2+ therapy. Thus, this study clarifies the antibacterial mechanism of FeS2 and offers a synergetic therapeutic platform with laser-mediated Fe2+ release for antibacterial applications.

Nanotechnology for the Treatment of Spinal Cord Injury.

Spinal cord injury (SCI) affects the central nervous system (CNS) and there is currently no treatment with the potential for rehabilitation. Although several clinical treatments have been developed, they are still at an early stage and have not shown success in repairing the broken fiber, which prevents cellular regeneration and integral restoration of motor and sensory functions. Considering the importance of nanotechnology and tissue engineering for neural tissue injuries, this review focuses on the latest advances in nanotechnology for SCI treatment and tissue repair. The PubMed database was used for the bibliographic survey. Initial research using the following keywords "tissue engineering and spinal cord injury" revealed 970 articles published in the last 10 years. The articles were further analyzed, excluding those not related to SCI or with results that did not pertain to the field of interest, including the reviews. It was observed that a total of 811 original articles used the quoted keywords. When the word "treatment" was added, 662 articles were found and among them, 529 were original ones. Finally, when the keywords "Nanotechnology and spinal cord injury" were used, 102 articles were found, 65 being original articles. A search concerning the biomaterials used for SCI found 700 articles with 589 original articles. A total of 107 articles were included in the discussion of this review and some are used for the theoretical framework. Recent progress in nanotechnology and tissue engineering has shown promise for repairing CNS damage. A variety of in vivo animal testing for SCI has been used with or without cells and some of these in vivo studies have shown successful results. However, there is no translation to humans using nanotechnology for SCI treatment, although there is one ongoing trial that employs a tissue engineering approach, among other technologies. The first human surgical scaffold implantation will elucidate the possibility of this use for further clinical trials. This review concludes that even though tissue engineering and nanotechnology are being investigated as a possibility for SCI treatment, tests with humans are still in the theoretical stage. Impact statement Thousands of people are affected by spinal cord injury (SCI) per year in the world. This type of lesion is one of the most severe conditions that can affect humans and usually causes permanent loss of strength, sensitivity, and motor function below the injury site. This article reviews studies on the PubMed database, assessing the publications on SCI in the study field of tissue engineering, focusing on the use of nanotechnology for the treatment of SCI. The review makes an evaluation of the biomaterials used for the treatment of this condition and the techniques applied for the production of nanostructured biomaterials.

Recent advances in bionanomaterials for liver cancer diagnosis and treatment.

According to the World Health Organization, liver cancer is the fourth leading cause of cancer associated with death worldwide. It demands effective treatment and diagnostic strategies to hinder its recurrence, complexities, aggressive metastasis and late diagnosis. With recent progress in nanotechnology, several nanoparticle-based diagnostic and therapeutic modalities have entered into clinical trials. With further developments in nanoparticle mediated liver cancer diagnosis and treatment, the approach holds promise for improved clinical liver cancer management. In this review, we discuss the key advances in nanoparticles that have potential for liver cancer diagnosis and treatment. We also discuss the potential of nanoparticles to overcome the limitations of existing therapeutic modalities.

Advances in Natural or Synthetic Nanoparticles for Metastatic Melanoma Therapy and Diagnosis.

Simple SummaryMalignant melanoma is the most aggressive skin cancer; its incidence is constantly growing in the white population. In the advanced stage of the disease, after metastatic dissemination, patients have a poor prognosis. Nanomedicine represents a new frontier in cancer treatment; in this field, synthetic and natural nanoparticles (NPs) may represent an important therapeutic and diagnostic opportunity. This review provides an overview of current knowledge in this area: several kinds of NPs, like PLGA, chitosan, liposome and gold-NPs, are used to increase the specificity of drug delivery, allowing a dose reduction and, consequently, a lower toxic effect. Particular attention is given to exosomes (EXOs), an example of natural NPs, important both in conveying molecules with a therapeutic function and in the diagnostic field.Advanced melanoma is still a major challenge in oncology. In the early stages, melanoma can be treated successfully with surgery and the survival rate is high, nevertheless the survival rate drops drastically after metastasis dissemination. The identification of parameters predictive of the prognosis to support clinical decisions and of new efficacious therapies are important to ensure patients the best possible prognosis. Recent progress in nanotechnology allowed the development of nanoparticles able to protect drugs from degradation and to deliver the drug to the tumor. Modification of the nanoparticle surface by specific molecules improves retention and accumulation in the target tissue. In this review, we describe the potential role of nanoparticles in advanced melanoma treatment and discuss the current efforts of designing polymeric nanoparticles for controlled drug release at the site upon injection. In addition, we highlight the advances as well as the challenges of exosome-based nanocarriers as drug vehicles. We place special focus on the advantages of these natural nanocarriers in delivering various cargoes in advanced melanoma treatment. We also describe the current advances in knowledge of melanoma-related exosomes, including their biogenesis, molecular contents and biological functions, focusing our attention on their utilization for early diagnosis and prognosis in melanoma disease.

A review on numerical modeling for magnetic nanoparticle hyperthermia: Progress and challenges.

Recent progress in nanotechnology has advanced the development of magnetic nanoparticle (MNP) hyperthermia as a potential therapeutic platform for treating diseases. Due to the challenges in reliably predicting the spatiotemporal distribution of temperature in the living tissue during the therapy of MNP hyperthermia, critical for ensuring the safety as well as efficacy of the therapy, the development of effective and reliable numerical models is warranted. This article provides a comprehensive review on the various mathematical methods for determining specific loss power (SLP), a parameter used to quantify the heat generation capability of MNPs, as well as bio-heat models for predicting heat transfer phenomena and temperature distribution in living tissue upon the application of MNP hyperthermia. This article also discusses potential applications of the bio-heat models of MNP hyperthermia for therapeutic purposes, particularly for cancer treatment, along with their limitations that could be overcome.

Applications and strategies in nanodiagnosis and nanotherapy in lung cancer.

Lung cancer is the second most common cancer and the leading cause of death in both men and women in the world. Lung cancer is heterogeneous in nature and diagnosis is often at an advanced stage as it develops silently in the lung and is frequently associated with high mortality rates. Despite the advances made in understanding the biology of lung cancer, progress in early diagnosis, cancer therapy modalities and considering the mechanisms of drug resistance, the prognosis and outcome still remains low for many patients. Nanotechnology is one of the fastest growing areas of research that can solve many biological problems such as cancer. A growing number of therapies based on using nanoparticles (NPs) have successfully entered the clinic to treat pain, cancer, and infectious diseases. Recent progress in nanotechnology has been encouraging and directed to developing novel nanoparticles that can be one step ahead of the cancer reducing the possibility of multi-drug resistance. Nanomedicine using NPs is continuingly impacting cancer diagnosis and treatment. Chemotherapy is often associated with limited targeting to the tumor, side effects and low solubility that leads to insufficient drug reaching the tumor. Overcoming these drawbacks of chemotherapy by equipping NPs with theranostic capability which is leading to the development of novel strategies. This review provides a synopsis of current progress in theranostic applications for lung cancer diagnosis and therapy using NPs including liposome, polymeric NPs, quantum dots, gold NPs, dendrimers, carbon nanotubes and magnetic NPs.

The Use of Nanomaterials in Tissue Engineering for Cartilage Regeneration; Current Approaches and Future Perspectives.

The repair and regeneration of articular cartilage represent important challenges for orthopedic investigators and surgeons worldwide due to its avascular, aneural structure, cellular arrangement, and dense extracellular structure. Although abundant efforts have been paid to provide tissue-engineered grafts, the use of therapeutically cell-based options for repairing cartilage remains unsolved in the clinic. Merging a clinical perspective with recent progress in nanotechnology can be helpful for developing efficient cartilage replacements. Nanomaterials, < 100 nm structural elements, can control different properties of materials by collecting them at nanometric sizes. The integration of nanomaterials holds promise in developing scaffolds that better simulate the extracellular matrix (ECM) environment of cartilage to enhance the interaction of scaffold with the cells and improve the functionality of the engineered-tissue construct. This technology not only can be used for the healing of focal defects but can also be used for extensive osteoarthritic degenerative alterations in the joint. In this review paper, we will emphasize the recent investigations of articular cartilage repair/regeneration via biomaterials. Also, the application of novel technologies and materials is discussed.

Overcoming multidrug resistance in cancer: Recent progress in nanotechnology and new horizons.

Multidrug resistance (MDR), defined as the ability of cancer cells to gain resistance to both conventional and novel chemotherapy agents, is an important barrier in treating malignancies. Initially, it was discovered that cellular pumps dependent on ATP were the cause of resistance to chemotherapy, and further studies have found that other mechanisms such as increased metabolism of drugs, decreased drug entry, and defective apoptotic pathways are involved in this process. MDR has been the focus of numerous initiatives and countless studies have been undertaken to better understand MDR and formulate strategies to overcome its effects. The current review highlights various nano-drug delivery systems including polymeric/solid lipid/mesoporous silica/metal nanoparticles, dendrimers, liposomes, micelles, and nanostructured lipid carriers to overcome the mechanism of MDR. Nanoparticles are novel gateways to enhance the therapeutic efficacy of anticancer agents at the target site of action due to their tumor-targeting abilities, which can limit the unwanted systemic effects of chemotherapy agents and also reduce drug resistance. Additionally, other innovative strategies including RNA interference as a biological process used to inhibit or silence specific gene expression, natural products as MDR modulators with little systemic toxic effects, which interfere with the functions of proteins involved in drug efflux, and physical approaches such as combination of conventional drug administration with thermal/ultrasound/photodynamic strategies are also highlighted.

Analysis for time-dependent flow of Carreau nanofluid over an accelerating surface with gyrotactic microorganisms: Model for extrusion systems

Recent progress in nanotechnology and nanoscience has attended the researchers’ interest due to its inspiring industrial and technological applications. This contribution investigates the bioconvection flow of Carreau fluid over a periodically moving stretched surface with important feature of magnetic field and thermal radiation. Bioconvection in non-Newtonian nanofluid is utilized with sustaining the joint features of buoyancy and magnetic forces. In addition, thermal radiation consequences are summarized in the energy equation while further computations have been operated under “one parametric approach.” The attended problem is contested with a famous convergent technique having excellent accuracy. A comprehensive graphical approach is constituted to describe the thermo-physical features of interesting physical parameters. The reported results may use in enhancement of extrusion system, bio-fuels, and biotechnology.

On Strength of Brittle Nanomaterials: Confinement Effect on Weibull Distributions

Recent progress in nanotechnology enables us to utilize the elastic strain engineering, the emerging technology capable of controlling the physio-chemical properties of materials via externally-imposed elastic strains, for hard materials. Because the range of accessible properties by imposing elastic strains are set by materials' elasticity limits, it is of great importance to suppress the occurrence of any inelastic deformations and failure, and thereby the fundamental knowledge on fracture behavior at nanoscale is highly required. The conventional Weibull theory, which has been widely used for last a few decades to explain the failure statistics of brittle bulk materials, has a limitation to be directly applied to the samples of nanometer dimensions because the baseline assumption on statistical equivalence becomes intractable for small samples. In this study, we suggest an integrated equation presenting the sample size effect on fracture strength for brittle nanomaterials by further considering the confinement of the flaw size distribution. This new approach is applicable to any homogeneous brittle nanomaterials whose failure is governed by linear elastic fracture mechanics, and shows good agreement with experimental data collected from literatures. We expect that this theoretical study offers new guideline to employ brittle nanomaterials in designing and fabricating the advanced strain engineering system.

Simple Non-Destructive Method of Ultrathin Film Material Properties and Generated Internal Stress Determination Using Microcantilevers Immersed in Air

Recent progress in nanotechnology has enabled to design the advanced functional micro-/nanostructures utilizing the unique properties of ultrathin films. To ensure these structures can reach the expected functionality, it is necessary to know the density, generated internal stress and the material properties of prepared films. Since these films have thicknesses of several tens of nm, their material properties, including density, significantly deviate from the known bulk values. As such, determination of ultrathin film material properties requires usage of highly sophisticated devices that are often expensive, difficult to operate, and time consuming. Here, we demonstrate the extraordinary capability of a microcantilever commonly used in a conventional atomic force microscope to simultaneously measure multiple material properties and internal stress of ultrathin films. This procedure is based on detecting changes in the static deflection, flexural and torsional resonant frequencies, and the corresponding quality factors of the microcantilever vibrating in air before and after film deposition. In contrast to a microcantilever in vacuum, where the quality factor depends on the combination of multiple different mechanical energy losses, in air the quality factor is dominated just by known air damping, which can be precisely controlled by changing the air pressure. Easily accessible expressions required to calculate the ultrathin film density, the Poisson’s ratio, and the Young’s and shear moduli from measured changes in the microcantilever resonant frequencies, and quality factors are derived. We also show that the impact of uncertainties on determined material properties is only minor. The validity and potential of the present procedure in material testing is demonstrated by (i) extracting the Young’s modulus of atomic-layer-deposited TiO2 films coated on a SU-8 microcantilever from observed changes in frequency response and without requirement of knowing the film density, and (ii) comparing the shear modulus and density of Si3N4 films coated on the silicon microcantilever obtained numerically and by present method.
