Diurnal decline in photosynthesis and stomatal conductance in several tropical species

Photosynthesis (A) and stomatal conductance (gs) change diurnally due to internal signals, but the effects of diurnal rhythms on dynamic photosynthetic behavior are understudied. We examined diurnal changes in A and gs in ten tropical species: across species, there was a tendency for A and gs to decline diurnally when these were repeatedly measured under either steady-state or fluctuating irradiance conditions. We then examined in more detail the irradiance-induced kinetics of gas exchange in a C3 and C4 crop species each, namely fig (Ficus carica) and sugarcane (Saccharum officinarum). During the day, fig showed significantly slower photosynthetic induction and lower gs, as well as a slower gs increase, in the afternoon than in the morning and noon. Sugarcane showed a reduction in steady-state A reached under high irradiance and slower gs increase as well as lower gs reached under high irradiance, but no changes in the rate of photosynthetic induction, in the afternoon, compared to morning and noon. These reductions in the afternoon were not reverted by a dark treatment in the middle of the day, suggesting that the decrease was not proportional to diurnal time-integrated carbon fixation. Repeated exposure to light- and shadeflecks (1000 and 50 μmol m-2 s-1, lasting 20 min each) revealed fundamental differences in stomatal regulation between species: in fig, stomata opened and closed slowly, and their opening became progressively slower under a series of lightflecks, whereas sugarcane showed much faster stomatal opening than closure that was unchanged during the course of the day. Our results highlight that steady-state rates and irradiance-induced kinetics of photosynthesis and stomatal movement change diurnally in most species studied, and that they do so differently in fig and sugarcane.

Rate of photosynthetic induction in fluctuating light varies widely among genotypes of wheat.

Crop photosynthesis and yield are limited by slow photosynthetic induction in sunflecks. We quantified variation in induction kinetics across diverse genotypes of wheat for the first time. Following a preliminary study that hinted at wide variation in induction kinetics across 58 genotypes, we grew 10 genotypes with contrasting responses in a controlled environment and quantified induction kinetics of carboxylation capacity (Vcmax) from dynamic A versus ci curves after a shift from low to high light (from 50 µmol m–2 s–1 to 1500 µmol m–2 s–1), in five flag leaves per genotype. Within-genotype median time for 95% induction (t95) of Vcmax varied 1.8-fold, from 5.2 min to 9.5 min. Our simulations suggest that non-instantaneous induction reduces daily net carbon gain by up to 15%, and that breeding to speed up Vcmax induction in the slowest of our 10 genotypes to match that in the fastest genotype could increase daily net carbon gain by up to 3.4%, particularly for leaves in mid-canopy positions (cumulative leaf area index ≤1.5 m2 m–2), those that experience predominantly short-duration sunflecks, and those with high photosynthetic capacities.

Characterization of photosynthetic gas exchange in leaves under simulated adaxial and abaxial surfaces alternant irradiation.

Previous investigations on photosynthesis have been performed on leaves irradiated from the adaxial surface. However, leaves usually sway because of wind. This action results in the alternating exposure of both the adaxial and abaxial surfaces to bright sunlight. To simulate adaxial and abaxial surfaces alternant irradiation (ad-ab-alt irradiation), the adaxial or abaxial surface of leaves were exposed to light regimes that fluctuated between 100 and 1,000 μmol m−2 s−1. Compared with constant adaxial irradiation, simulated ad-ab-alt irradiation suppressed net photosynthetic rate (Pn) and transpiration (E) but not water use efficiency. These suppressions were aggravated by an increase in alternant frequency of the light intensity. When leaves were transferred from constant light to simulated ad-ab-alt irradiation, the maximum Pn and E during the high light period decreased, but the rate of photosynthetic induction during this period remained constant. The sensitivity of photosynthetic gas exchange to simulated ad-ab-alt irradiation was lower on abaxial surface than adaxial surface. Under simulated ad-ab-alt irradiation, higher Pn and E were measured on abaxial surface compared with adaxial surface. Therefore, bifacial leaves can fix more carbon than leaves with two “sun-leaf-like” surfaces under ad-ab-alt irradiation. Photosynthetic research should be conducted under dynamic conditions that better mimic nature.

Bryophyte photosynthesis in sunflecks: greater relative induction rate than in tracheophytes

Photosynthetic kinetics in changing light intensity is considered pivotal for the survival of understorey tracheophytes; however, it is virtually unknown for bryophytes because bryophytes and tracheophytes have contrasting physio-ecological strategies. Anatomy suggests that relative rate of photosynthetic induction is faster in bryophyte gametophytes due to the absence of stomata, whose slow response may limit photosynthesis of tracheophyte leaves in changing light intensity. We measured steady-state and dynamic CO2 exchange in 10 bryophyte species. We performed an in-depth survey of the moss Hypnum cupressiforme sampled from sun and shade sites. Our key results are: (1) Bryophyte photosynthesis after dark acclimatisation induced much faster than in tracheophytes, reaching 50% of maximum gross photosynthesis (Agross) in about 90 s. Such rapid induction is comparable only to tracheophytes whose stomata do not limit CO2 uptake. Times to reach 90% of Agross were also substantially shorter in bryophytes (ca 220 s) than those reported for most tracheophytes (500–2000 s). (2) Shade-grown mosses did not reveal lower photosynthetic capacity than sun ones. (3) Shade-grown H. cupressiforme induced photosynthesis slightly faster than that from forest gaps. We conclude that bryophytes are efficient utilizers of temporal light heterogeneity, increasing carbon gain during short high-light events such as sunflecks in forest understorey.

The effect of temperature on photosynthetic induction under fluctuating light in Chrysanthemum morifolium

The photosynthetic response was investigated on Chrysanthemummorifolium under dynamic light conditions in the 20–35 °C temperature range to evaluate the effect of climatic variables on photosynthetic induction. The plant material was grown under uniform, controlled conditions and its gas exchange was analyzed. The gas exchange measurements were used to investigate the rate of induction, momentary induction state, and the opening of stomata. At the varying temperature ranges and under dynamic light conditions, C. morifolium reached a quasi-steady-state induction equilibrium (ISeq(PAR,T)) within 14–45 min. For the same level of photosynthetically active radiation (PAR), the equilibrated level of steady-state induction increased as the temperature increased. It was highest approximately at 30 °C. The induction state was equilibrated at a lower level as the temperature increased to 35 °C. The interaction effect of PAR and temperature on induction state was not significant. The rate of photosynthetic induction and the time required at which the induction reached its 90 % value (t90) was influenced by PAR significantly. The light history of a leaf had a significant effect on t90, indicating that the time to reach a steady-state induction is different depending on the light environment and the period at which the leaf was exposed to light. The velocity of the photosynthetic induction was not affected by the temperature. It was associated with stomatal conductance of the leaf prior to the onset of light (gSini).

Stomatal versus biochemical limitations to dynamic photosynthetic performance in four tropical rainforest shrub species.

Photosynthetic performance under dynamic light regimes was assessed in four different species of tropical shrubs from the family Rubiaceae via field gas exchange measurements conducted on Barro Colorado Island, Panamá. Rates of photosynthetic induction and induction loss were assessed throughout the day in both the wet and dry seasons in order to determine the relative roles of stomata and biochemistry in limiting photosynthetic performance under transient light conditions. A high degree of coordination was observed between stomatal conductance and biochemical capacity for CO2 assimilation during induction. Rates of biochemical and overall photosynthetic induction sharply decreased when initial stomatal conductance fell below a narrow range of critical values. Time of day or season did not affect rates of biochemical deactivation upon shading, but did influence stomatal closure, which often exerted a significant influence over induction loss in the darkness. In measurements of total assimilation due to a 60-s light pulse, both biochemical activity and stomatal conductance were linearly related to total CO2 uptake. Only during the mornings of the wet season was stomatal conductance consistently high enough to be non-limiting to dynamic photosynthetic performance. At all other times, stomatal behavior exercised significant influence over induction times, photosynthetic induction loss, and total CO2 uptake from 60-s light pulses.
