The outgrowth of neurite from cell body of nerve cells is the first and critical step during neuronal differentiation. This process is tightly regulated by a plethora of signaling proteins. Recently, MicroRNAs have emerged as key posttranscriptional regulators of gene expression during neuronal development. Herein, we reported that a brain enriched microRNA family, micoRNA-200 family, including miR-200a/b/c, miR-141 and miR-429, are implicated in controlling neurite outgrowth in PC12 and rat superior cervical ganglia (SCG) neurons. Luciferase reporter assay and western blot analysis showed that each member of miR-200 family directly targets to 3'-UTR of Phosphatase and tensin homolog (PTEN) individually to inhibit its protein expression. Among them, the miR-200c is the major suppressor of PTEN expression. Either overexpression of miR-200c or knockdown of PTEN promotes NGF-induced neurite outgrowth in PC12 cells. In contrast, NGF-induced neuritogenesis of PC12 is blocked by knockdown of miR-200c or overexpression of PTEN, respectively. More interestingly, we also observed similar effects of miR-200c or PTEN during SCG neuronal differentiation in vitro. Taken together, our results demonstrated that miR-200 family plays an important role in neurite outgrowth by suppressing PTEN expression in PC12 cells or SCG neurons.
