Isolation and Characterization of a Sex-Specific Lectin in a Marine Red Alga, Aglaothamnion oosumiense Itono

In red algae, spermatial binding to female trichogynes is mediated by a lectin-carbohydrate complementary system. Aglaothamnion oosumiense is a microscopic filamentous red alga. The gamete recognition and binding occur at the surface of the hairlike trichogyne on the female carpogonium. Male spermatia are nonmotile. Previous studies suggested the presence of a lectin responsible for gamete recognition on the surface of female trychogynes. A novel N-acetyl-D-galactosamine-specific protein was isolated from female plants of A. oosumiense by affinity chromatography and named AOL1. The lectin was monomeric and did not agglutinate horse blood or human erythrocytes. The N-terminal amino acid sequence of the protein was analyzed, and degenerate primers were designed. A full-length cDNA encoding the lectin was obtained using rapid amplification of cDNA ends-PCR (RACE-PCR). The cDNA was 1,095 bp in length and coded for a protein of 259 amino acids with a deduced molecular mass of 21.4 kDa, which agreed well with the protein data. PCR analysis using genomic DNA showed that both male and female plants have this gene. However, Northern blotting and two-dimensional electrophoresis showed that this protein was expressed 12 to 15 times more in female plants. The lectin inhibited spermatial binding to the trichogynes when preincubated with spermatia, suggesting its involvement in gamete binding.

Molecular Characterization of a Nicotiana tabacum NtRDR6 Gene

RNA-dependent RNA polymerases (RDRs) play key roles in RNA silencing, developmental regulation, antiviral defense and the maintenance of genome integrity. In this work, we isolated an RDR gene (NtRDR6) from Nicotiana tabacum using reverse-transcription PCR (RT-PCR) and rapid amplification of cDNA ends PCR (RACE-PCR) technology. The full-length cDNA of NtRDR6 encodes a protein of 1,197 amino acids, which contains all of the five highly conserved regions in plant RDRs and the DxDGD signature motif. An amino acid sequence alignment revealed that NtRDR6 shares a high level of similarity with other known RDRs. The genomic sequence of NtRDR6 contains two introns and a group of putative cis-acting elements in the 5′-flanking region. Expression analysis performed using 6-week-old seedlings revealed that the transcript level of NtRDR6 is up-regulated by diverse signalling molecules, including jasmonate (JA), abscisic acid (ABA), gibberellin A (GA) and salicylic acid (SA). The expression of NtRDR6 is also elevated markedly by Cucumber mosaic virus (CMV) and Potato virus X (PVX) but not by Potato virus Y (PVY) and Tobacco mosaic virus (TMV). Notably, abiotic stresses have alternative effects on the transcription of NtRDR6, with heat (37°C) significantly increasing the expression of NtRDR6, whereas low temperature (15°C) does not affect expression. These results indicated that NtRDR6 may be involved in several signal transduction pathways to defend against plant virus attack and in the response to abiotic stresses.

Identification and Characterization of Two General Odorant Binding Protein Genes from the Oriental Fruit Moth, Grapholita molesta (Busck)

Two novel general odorant-binding protein (GOBP) cDNAs (GmolGOBP1 and GmolGOBP2) were cloned and characterized from female antennal tissue of the oriental fruit moth, Grapholita molesta. We focused our investigation on this olfactory protein family by using reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends-PCR (RACE-PCR). The full-length open reading frames of GmolGOBP1 and GmolGOBP2 were 492 and 483 nucleotides long, which encode 164 and 161 amino acid residue peptides, respectively. Protein signature analyses revealed that they each contained six conserved cysteines with an N-terminal signal sequence of 20 amino acids. The alignment of the two deduced protein sequences with other Lepidoptera GOBPs showed high sequence similarity (70-80%) with other full-length sequences from GenBank. Sequence similarity between the two GOBPs was only 48%, suggesting that the two proteins belong to different classes of lepidopteran GOBPs. RT-PCR analysis revealed that the two GOBP genes were expressed only in antennae of both sexes. Real-time PCR analysis further indicated that the transcript level of GmolGOBP1 was higher in males than in females, whereas the transcript level of GmolGOBP2 was higher in females than in males. Temporally, the two GOBP genes were expressed during the complete photoperiod (15L:9D). The highest transcript levels of GmolGOBP1 in both sexes and GmolGOBP2 in females were detected at the end of photophase and during scotophase. The expression of GmolGOBP2 in males remained at similar levels during the complete photoperiod. Based on these results, the possible physiological functions of GmolGOBPs are discussed.

Expression profiles of three types of GnRH during sex-change in the protandrous cinnamon clownfish, Amphiprion melanopus: Effects of exogenous GnRHs

Gonadotropin-releasing hormones (GnRHs) play pivotal roles in the control of reproduction and gonadal maturation in teleost fish. Fish have multiple GnRH genes that encode structurally distinct peptides. We identified salmon GnRH (sGnRH), seabream GnRH (sbGnRH), and chicken GnRH-II (cGnRH-II) by cDNA cloning in cinnamon clownfish (Amphiprion melanopus) using reverse transcription-PCR (RT-PCR) and rapid amplification of cDNA ends-PCR (RACE-PCR). Gene identity was confirmed by sequence alignment and subsequent phylogenetic analyses. We also investigated GnRH mRNA expression in the gonads by quantitative real time-PCR (Q-PCR), and measured plasma estradiol-17β (E(2)) levels in immature fish following treatment with the three molecular forms of GnRHs. The expression levels of sGnRH, sbGnRH, and cGnRH-II mRNA were higher in mature testes and ovaries, as compared to the levels in gonads at earlier stages of maturity. The levels of the three prepro-GnRH mRNA species and the plasma E(2) levels increased after injection of the three GnRH variants. These findings support the hypothesis that GnRH peptides play important roles in the regulation of the hypothalamic-pituitary-gonadal axis and are probably involved in paracrine control of gonadal development and sex change in cinnamon clownfish.

Agammaglobulinemia associated with BCR− B cells and enhanced expression of CD19

AbstractExpression of a BCR is critical for B-cell development and survival. We have identified 4 patients with agammaglobulinemia and markedly reduced but detectable B cells in the peripheral circulation. These B cells have an unusual phenotype characterized by increased expression of CD19 but no BCR. The cells are positive for CD20, CD22, and CD38, but not for Annexin 5 or activation markers, including CD69, CD83, or CD86. EBV lines derived from these B cells lack functionally rearranged immunoglobulin heavy-chain transcripts, as shown by PCR–rapid amplification of cDNA ends (PCR-RACE). Analysis of BM from 2 of the patients showed a severe reduction in the number of pro-B cells as well as pre-B cells. Functionally rearranged heavy-chain transcripts were identified, indicating that machinery to rearrange immunoglobulin genes was intact. Flow cytometry of B-lineage cells suggested accelerated acquisition of maturation markers in early B-cell precursors and increased phosphorylation of signal transduction molecules. Further, expression of TdT, a molecule that is normally down-regulated by a functional pre-BCR complex, was decreased. We hypothesize that the accelerated maturation, increased expression of CD19, and lack of a BCR were due to the constitutive activation of the BCR signal transduction pathway in these patients.

Specific regulation of low-abundance transcript variants encoding human Acyl-CoA binding protein (ACBP) isoforms

Despite intensive efforts on annotation of eukaryotic transcriptoms, little is known about the regulation of low-abundance transcripts. To address this question, we analysed the regulation of novel low-abundance transcript variants of human acyl-CoA binding protein (ACBP), an important multifunctional housekeeping protein, which we have identified by screening of human expressed sequence tags in combination with ab initio gene prediction. By using RT-, real-time RT- and rapid amplification of cDNA ends-PCR in five human tissues, we find these transcripts, which are generated by a consequent use of alternative promoters and alternate first or first two exons, to be authentic ones. They show a tissue-specific distribution and intrinsic responsiveness to glucose and insulin. Promoter analyses of the corresponding transcripts revealed a differential regulation mediated by sterol regulatory element-binding protein-2, hepatocyte nuclear factor-4α and nuclear factor κB (NF-κB), central transcription factors of fat and glucose metabolism and inflammation. Subcellular localization studies of deduced isoforms in liver HepG2 cells showed that they are distributed in different compartments. By demonstrating that ACBP is a target of NF-κB, our findings link fatty acid metabolism with inflammation. Furthermore, our findings show that low-abundance transcripts are regulated in a similar mode than their high-abundance counterparts.

A Crassostrea gigas Toll-like receptor and comparative analysis of TLR pathway in invertebrates

Toll-like receptor (TLR) signaling pathway was an important and evolutionarily conserved innate immune pathway. Evolutionary lineage of this pathway in the Lophotrochozoans is still less understood. In this study, we cloned a novel TLR, a key component of TLR pathway, from Crassostrea gigas, and named it CgToll-1. The 4343 base pairs full-length cDNA was assembled with the 3′ and 5′ RACE (rapid amplification of cDNA ends) PCR results, and containing a 3540 bp open reading frame, which encoding a putative TLR protein of 1179 amino acid residues. Real-time reverse transcription polymerase chain reaction analysis revealed that the highest CgToll-1 expression level was in hemolymph, and the expression pattern in hemolymph dramatically increased in the presence of bacteria Vibrio anguillarum. Furthermore, TLR pathway core genes of mollusks were searched and compared with model invertebrates. Phylogenetic trees of two downstream genes ( IκB, Rel) showed that mollusks genes were closer to Drosophila melanogaster than Strongylocentrotus purpuratus, while three upstream genes ( MyD88, IRAK, TRAF6) showed the opposite propensity. We have also detected that these two downstream genes were significantly more conservative than the three upstream genes based on amino acid sequence alignment. We found no significant difference between the codon usage biases of TLR pathway genes. This study suggests that CgToll-1 was a constitutive and inducible protein and thus could play an important role in the immune responses against bacterium infection. Besides, comparative analysis of TLR pathway showed that gene loss and divergence might exist during evolution in invertebrate.

197 MOLECULAR CLONING AND EXPRESSION OF THE CASHMERE GOAT IZUMO GENE

During the fertilization process, complex events are involved in the fusion between the reacted spermatozoa and the mature oocyte. Fusion implies that many proteins are present on the cell membrane of the gametes. Recently, a new protein, Izumo, has been shown to play a role in the sperm–egg fusion. Izumo was identified through the generation of a monoclonal antibody that inhibits this fusion. This protein belongs to the immunoglobulin superfamily type I transmembrane protein. Izumo can be detected only after acrosome reaction at the spermatozoal surface. The cashmere goat Izumo gene was identified and cloned by 3′ and 5′ rapid amplification of cDNA ends-PCR. The expression of cashmere goat Izumo was examined by RT-PCR and Northern blotting. The full-length cDNA of cashmere goat Izumo contains 1536 bp and an open reading frame of 1035 bp, encoding a polypeptide of 344 amino acids with a calculated molecular mass of 38.76 kDa and a theoretical isoelectric point of 8.18. This predicted amino acid sequence showed 89.82% amino acid identity with the bovine Izumo. The deduced amino acid sequence contained 3 conserved domains of the signal peptide, immunoglobulin-like domain, and transmembrane region. Reverse transcription-PCR and Northern blotting analysis showed that cashmere goat Izumo transcripts were highly expressed in the testis, caput, corpus, cauda epididymis. Cashmere goat Izumo may play a role in the biological process of fertilization. TPC2 Is a Novel NAADP-sensitive Ca2+ Release Channel, Operating as a Dual Sensor of Luminal pH and Ca2+

Nicotinic acid adenine dinucleotide phosphate (NAADP) is a molecule capable of initiating the release of intracellular Ca2+ required for many essential cellular processes. Recent evidence links two-pore channels (TPCs) with NAADP-induced release of Ca2+ from lysosome-like acidic organelles; however, there has been no direct demonstration that TPCs can act as NAADP-sensitive Ca2+ release channels. Controversial evidence also proposes ryanodine receptors as the primary target of NAADP. We show that TPC2, the major lysosomal targeted isoform, is a cation channel with selectivity for Ca2+ that will enable it to act as a Ca2+ release channel in the cellular environment. NAADP opens TPC2 channels in a concentration-dependent manner, binding to high affinity activation and low affinity inhibition sites. At the core of this process is the luminal environment of the channel. The sensitivity of TPC2 to NAADP is steeply dependent on the luminal [Ca2+] allowing extremely low levels of NAADP to open the channel. In parallel, luminal pH controls NAADP affinity for TPC2 by switching from reversible activation of TPC2 at low pH to irreversible activation at neutral pH. Further evidence earmarking TPCs as the likely pathway for NAADP-induced intracellular Ca2+ release is obtained from the use of Ned-19, the selective blocker of cellular NAADP-induced Ca2+ release. Ned-19 antagonizes NAADP-activation of TPC2 in a non-competitive manner at 1 μm but potentiates NAADP activation at nanomolar concentrations. This single-channel study provides a long awaited molecular basis for the peculiar mechanistic features of NAADP signaling and a framework for understanding how NAADP can mediate key physiological events.

Genetic diversity of levamisole receptor subunits in parasitic nematode species and abbreviated transcripts associated with resistance

The molecular mechanisms of levamisole (LEV) activity and expression of resistance remain largely unknown in parasitic nematodes. In contrast, genetic screens for mutants that survive exposure to LEV in the free-living nematode Caenorhabditis elegans have led to the identification of five genes (unc-38, unc-63, unc-29, lev-1 and lev-8) that encode a LEV-sensitive acetylcholine receptor (L-AChR). Loss of these genes leads to LEV resistance. In this study, orthologues of these genes were identified in three species of trichostrongylid nematodes that have a major impact on small ruminants: Haemonchus contortus, Teladorsagia circumcincta and Trichostrongylus colubriformis. Polymorphism associated with LEV resistance have been investigated by comparing transcripts of these subunits in LEV susceptible and LEV-resistant isolates of the three strongylid species. Partial sequences were identified by PCR experiments and full-length cDNA sequences corresponding to AChR subunits in the three trichostrongylid species were obtained using 3'-rapid amplification of cDNA ends-PCR and 5' rapid amplification of cDNA ends anchored with the spliced leader sequence, SL1. Expression of L-AChR subunits was investigated in LEV-resistant and LEV-susceptible isolates of H. contortus, T. circumcincta and T. colubriformis using reverse transcription PCR. We have identified a total of 20 full-length cDNA sequences corresponding to L-AChR subunits in three parasitic trichostrongylid species of which 14 correspond to novel sequences. Genes orthologous to unc-29, unc-63, unc-38 and lev-1 were found in each trichostrongylid species, whereas no gene corresponding to lev-8 has yet been identified. We have found 11 distinct paralogous sequences corresponding to the C. elegans unc-29 gene clustered in four groups revealing an unexpected diversity of unc-29-like genes. Complete coding sequences of the L-AChR subunits in two LEV-resistant and three susceptible isolates of H. contortus, T. circumcincta and T. colubriformis were essentially unchanged, but abbreviated transcripts of the unc-63 subunit were specifically expressed in resistant isolates of all three species. The candidate gene strategy developed in this study revealed an unexpectedly high diversity of L-AChR subunits specific to the trichostrongylid parasites that are a principal target for the drug LEV. Abbreviated variants, predicted to produce nonfunctional unc-63, were associated with LEV resistance. This study contributes significantly to a better understanding of LEV receptor constitution in parasitic nematodes and highlights the putative role of aberrant mRNA encoding L-AChR subunits in LEV resistance.

Peptide mass-assisted antibody cloning strategy for accurate characterization of potential therapeutic monoclonal antibodies against neurodegenerative diseases

The development of therapeutic recombinant antibodies involves accurate characterization of immunoglobulin variable light (VL) and heavy (VH) chains. However, it has been reported that the use of subgroup or isotype-specific primers for the amplification of monoclonal antibody (mAb) variable domains introduces heterogeneities within the variable domains, or amplifies aberrant productive Ig domains. To address these issues, we have combined the rapid amplification of cDNA ends PCR (RACE-PCR) for the full-length VL and VH amplification, with peptide mass fingerprinting of the corresponding Ig chain. Using this strategy, we amplified full-length cDNA chains of SAF34 and SAF32, two potential therapeutic mAbs against neurodegenerative diseases directed to the prion protein (PrP). We report an unambiguous correlation between hybridoma cDNA sequences and protein fingerprints of the variable domains of each mAb, indicating the discrimination between mutated, pseudo-genes and functional Ig genes. As a proof of principle for this dual strategy of full-length PCR amplification of variable domains and their characterization by MALDI-TOF, we show that the corresponding scFvs recognize the native PrP and retain full capacity to bind to human PrP, as does the parental mAb. This finding addresses the need for reliable light and heavy chain characterization, a key factor for humanization of mouse antibodies and for its use in passive immunotherapy applications.

Gene expression and characterization of a stress-induced tyrosine decarboxylase from Arabidopsis thaliana

Full-length tyrosine decarboxylase cDNA ( TyrDC) from Arabidopsis thaliana was identified by rapid amplification of cDNA ends-PCR and isolated by RT-PCR. The TyrDC mRNA was substantially induced by drought stress and wounding, and was considerably decreased by salt stress. By using TyrDC protein fusions with green fluorescent protein, an intracellular localization to the cytoplasm was shown. Recombinant (His) 6-TyrDC was expressed in Escherichia coli and enzymatically characterized: it exclusively catalyzed the conversion of l-tyrosine to tyramine, exhibited an optimum temperature of 50 °C, and an optimum pH at approximately 8.5–9. Recombinant TyrDC protein formed tetramers, as shown by blue native gel electrophoresis. Cloning, expression and functional analysis of a general odorant‐binding protein 2 gene of the rice striped stem borer, Chilo suppressalis (Walker) (Lepidoptera: Pyralidae)

A full-length cDNA encoding a general odorant binding protein 2 (GOBP2) was cloned from the antennae of the rice striped stem borer, Chilo suppressalis (Lepidoptera: Pyralidae), by the combination of reverse transcription PCR (RT-PCR) and rapid amplification of cDNA ends PCR (RACE-PCR). The cDNA contains a 489 bp open reading frame, which encodes a 162 amino acid protein, termed as Ch. suppressalis GOBP2 (CsupGOBP2). CsupGOBP2 is similar in the number of amino acids and protein sequence to GOBP2s in other species of Lepidoptera. RT-PCR results showed that CsupGOBP2 mRNA was highly expressed in the adult antennae of both females and males, as was CsupGOBP2 protein as revealed by Western blot analysis. CsupGOBP2 expressed in Escherichia coli was purified by affinity chromatography, refolding and gel filtration from the inclusion body. Fluorescence emission spectra and competitive binding assays by using N-phenyl-1-naphthylamine as first binding ligand and odorants as potential competitors revealed that the CsupGOBP2 protein has significant affinity to cis-11-hexadecenal (Z11-16:Ald), the main component of Ch. suppressalis pheromone and to laurinaldehyd and benzaldehyde, two general plant volatile aldehydes.

Isolation and characterization of the c‐type lysozyme gene from the common cutworm spodoptera litura

We have isolated and characterized a new insect chicken type (c‐type) lysozyme gene from the common cutworm, Spodoptera litura. The full‐length cDNA of Spodoptera lysozyme is cloned by rapid amplification of cDNA ends PCR (RACE‐PCR). The isolated cDNA consists of 1039 bp including the coding region for a 142‐amino acid residue polypeptide, which included a signal peptide of 21‐amino acid residue and a mature protein of 121‐amino acid residue. The predicted molecular weight of mature lysozyme and its theoretical isoelectric point from amino acid composition is 13964.8 Da and 9.05, respectively. The deduced amino acid sequence of Spodoptera lysozyme gene shows the highest similarity (96.7%) to Spodoptera exigua lysozyme among other lepidopteran species. Amino acid sequence comparison with other the c‐type lysozymes, Spodoptera lysozyme has the completely conserved Glu32 and Asp50 of the active site and eight Cys residues are completely conserved in the same position as that of other lepidopteran lysozymes.

Molecular cloning and characterization of a salinity stress-induced gene encoding DEAD-box helicase from the halophyte Apocynum venetum

The genes encoding DEAD-box helicases play a key role in various abiotic stresses, including temperature, light, oxygen, and salt stress. A salt-responsive gene, designated AvDH1, was isolated from the halophyte dogbane (Apocynum venetum) by using suppression subtractive hybridization and RACE (rapid amplification of cDNA ends) PCR. The deduced amino acid sequence has nine conserved helicase motifs of the DEAD-box protein family. The AvDH1 gene is present as a single copy in the dogbane genome. This gene is expressed in response to NaCl and not polyethlene glycol (PEG) nor abscisic acid, and its expression increases with time. The transcription of AvDH1 is also induced by low temperature (4 degrees C), but its accumulation first increases then decreases with time. The purified recombinant protein contains ATP-dependent DNA helicase activity, ATP-independent RNA helicase activity, and DNA- or RNA-dependent ATPase activity. The ATPase activity of AvDH1 is stimulated more by single-stranded DNA than by double-stranded DNA or RNA. These results suggested that AvDH1 belonging to the DEAD-box helicase family is induced by salinity, functions as a typical helicase to unwind DNA and RNA, and may play an important role in salinity tolerance.

Essential Role of Chitinase in the Development of the Filarial Nematode Acanthocheilonema viteae

Chitinases of pathogens have been proposed as potential targets of vaccines or specific inhibitors. We studied the genomic organization, transcript levels, developmental expression, and biological function of chitinases in the rodent filarial nematode Acanthocheilonema viteae, a model organism for human-pathogenic filarial worms. Characterization of nine genomic clones from an A. viteae phage library and Southern blot experiments revealed the existence of three different chitinase genes, two of which could theoretically yield functional transcripts. The deduced proteins of these genes had the common modular organization of family 18 chitinases. Northern blot experiments and rapid amplification of cDNA ends-PCR with adult worms and larval stages showed that only one gene is expressed, with high variation in transcript levels, as determined by real-time PCR. Chitinase transcript levels were lowest in the late male stage 4 larva (L4) and peaked in the stage 3 larva (L3), which was corroborated by Western blotting. RNA interference (RNAi) experiments showed that treatment of L3 and adult female worms with double-stranded RNA of chitinase inhibited molting of L3 worms and hatching of microfilariae. RNAi also led to the death of 50% of female worms, revealing the essential role of chitinase in the life cycle of filarial nematodes.

Molecular cloning and expression study of pi-class glutathione S-transferase (pi-GST) and selenium-dependent glutathione peroxidase (Se-GPx) transcripts in the freshwater bivalve Dreissena polymorpha

Glutathione S-transferases (GST) and glutathione peroxidases (GPx) are essential components of cellular detoxification systems. We identified GST and GPx transcripts in the freshwater bivalve Dreissena polymorpha, their full-length coding sequences were obtained by reverse-transcription PCR using degenerated primers followed by 5′ and 3′ RACE-PCR (rapid amplification of cDNA ends-PCR). The cDNA identified encoded proteins of 205 and 243 amino acids corresponding respectively to a pi-class GST and a selenium-dependent GPx. The comparison of the deduced amino acid sequences with GST and GPx from other species showed that the residues essential to the enzymatic function of these two proteins are highly conserved. We studied their expression pattern in the digestive gland, the gills and the excretory system of D. polymorpha. The results showed that pi-GST mRNA expression is higher in the digestive gland than in the gills or the excretory system. Se-GPx transcripts are expressed at high, medium and very low levels in the digestive gland, the excretory system and the gills, respectively.

SigM-Responsive Genes of Bacillus subtilis and Their Promoters

Promoters of nine Bacillus subtilis genes (bcrC, yacK, ydaH, yfnI, yjbD, ypbG, ypuA, yraA, and ysxA), all responsive to artificially induced increases in the stress-responsive extracytoplasmic function sigma factor, SigM, were mapped by rapid amplification of cDNA ends-PCR. The resulting promoter consensus suggests that overlapping control by SigX or SigW is common.

Molecular Characterization of Two Pheromone Binding Proteins and Quantitative Analysis of their Expression in the Beet Armyworm, Spodoptera exigua Hübner

Pheromone binding proteins (PBP) play an important role in insect pheromone communication. However, the PBP for the beet armyworm, Spodoptera exigua Hübner (Lepidoptera: Noctuidae), an important agricultural pest worldwide, remains unaddressed. We report the cloning of two PBP genes, SexigPBP1 and SexigPBP2, from the antennal cDNA of S. exigua by reverse transcriptase-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends-PCR (RACE-PCR). The deduced PBP amino acid sequences are characteristic of the odorant binding protein (OBP) family, although the two PBPs are only 44% identical. From an analysis of the genomic DNA, two introns and a similar intron/extron structural pattern were identified in each of the two PBP genes. RT-PCR analysis revealed that the two PBP genes are only expressed in antennae. Real-time PCR further indicated that the expression of SexigPBP1 is much higher than that of SexigPBP2, regardless of sex. However, the female expression levels for SexigPBP1 and SexigPBP2 are about 39% and 73%, respectively, relative to male levels. Finally, phylogenetic analysis suggested that PBPs from the Noctuidae are divided into three distinct groups based on the primary sequences.

5′ end cDNA amplification using classic RACE

The 5' ends of transcripts provide important information about transcription initiation sites and the approximate locations of local cis-acting enhancer elements; it is therefore important to establish the 5' ends with some precision. RACE (rapid amplification of cDNA ends) PCR is useful for quickly obtaining full length cDNAs for mRNAs for which only part of the sequence is known and to identify alternative 5' or 3' ends of fully sequenced genes. The method consists of using PCR to amplify, from complex mixtures of cellular mRNA, the regions between the known parts of the sequence and non-specific tags appended to the ends of the cDNA. Whereas the poly(A) tail serves to provide such a tag at the 3' end of the mRNA, an artificial one needs to be generated at the 5' end, and various approaches have been described to address this step. The classical scheme for 5' RACE described here is simple, suffices in many instances in which RACE is needed and can be performed in 1-3 days.
