Evaluation of radioprotective efficacy of Drymaria cordata extract on whole-body radiation-induced haematological damage in mice

Introduction: This study aimed to evaluate the radioprotective potential of Drymaria cordata (DC) extract on mice’s haematological parameters following exposure to X-rays radiation.Materials and Methods: Sixty female mice weighing 38-45g, 10-12 weeks old, were used for this study. The mice were divided into six different groups containing ten mice, sub-divided into irradiated and un-irradiated groups. The animals received 250mg/kg extract of DC by oral gavage for thirteen days in addition to feeding and water ad libitum. Mice were irradiated at the Radiotherapy and Oncology Department of Grey’s Hospital using a linear accelerator. Blood samples were collected at different time intervals for the haematology test with post-irradiation monitoring for 30 days. Results: Exposure of mice to 4Gy and 8Gy of X-ray radiation produced significant changes in the mice’s erythrocytes, haematocrit, leukocytes and platelets, in a dose and time-dependent manner compared with the control (CNT) group. The present study revealed a progressive decrease in all the haematological parameters until 30 days among the irradiated groups. However, animals treated with DC extract before irradiation and animals who received extract only exhibited a significant time-dependent increase in the studied haematological parameters compared to the animals in the CNT group. Furthermore, the pre-treatment of mice with the DC delayed the onset of mortality, thereby increasing mice's survival rate compared with the irradiated control. Conclusion: Our findings showed that DC is a potent natural radioprotective agent through its ability to reduce radiation-induced damage in mice’s haematopoietic system and increase the survival rate.

Amelioration of Radiation-Induced Damages in Mice by Curcuminoids: The Role of Bioavailability

Purpose The present study investigated the role of free curcuminoids bioavailability on the relative radioprotective efficacy of natural unformulated curcuminoids. Materials and Methods A food-grade bioavailable formulation of curcuminoids as curcumagalactomannosides (CGM) and unformulated curcuminoids (UC) were employed for the study. Swiss albino mice were randomized into Normal control, Radiation control, Radiation + UC, and Radiation + CGM groups and irradiated with γ-radiation of 6, 8, 10 and 12 Gy. Survival rate, hematological and biochemical parameters, bone marrow cellularity, chromosomal aberrations and histopathology of intestine were followed as a measure of the relative efficacy. Results and Discussion: Oral administration with both UC and CGM at 100 mg/kg. b.wt. produced significant radioprotective effect over the untreated control group of animals. However, CGM treatment was found to provide better clastogenic and genotoxic potential as compared to UC. Further, the histopathology analysis of intestine confirmed the better protective effect of CGM over UC-treated animals. Conclusion The present study demonstrated the positive role of the bioavailability of curcuminoids in the amelioration of radiation-induced damages in mice since CGM treatment exerted better survival rate and radioprotective effect as compared with UC, despite the relatively low concentrations of curcuminoids in CGM (39% w/w).

IKKβ Inhibitor IMD-0354 Attenuates Radiation Damage in Whole-body X-Irradiated Mice.

Nuclear factor-kappa B (NF-κB) transcription factor plays a critical role in regulating radiation-induced inflammatory and immune responses. Intracellular reactive oxygen species generation induces the activation of NF-κB via the inhibitor of κB (IκB) kinase (IKK) complex signaling. Previous studies have reported that the inhibition of IKK-driven NF-κB activation offers a therapeutic strategy for managing inflammatory disorders and various cancers, but it has additionally been reported that treatment targeting NF-κB also shows a radioprotective effect. IMD-0354 is an IKKβ inhibitor that blocks IκBα phosphorylation in the NF-κB pathway. This compound is known to exert anti-inflammatory and antitumor effects, but its radioprotective effects are unclear. Therefore, in the present study, we examined whether or not IMD-0354 has a mitigative effect on radiation-induced damages in mice. IMD-0354 was dissolved in soybean oil and subcutaneously administered to C57BL/6J Jcl mice for 3 consecutive days after 7 Gy of whole-body X-irradiation. The survival rate on day 30 and the NF-κB p65 and IκBα in bone marrow and spleen cells based on flow cytometry were assessed. IMD-0354 administration significantly suppressed the lethality induced by whole-body X-irradiation, and the survival rate increased by 83%. The NF-κB p65 and IκBα in bone marrow and spleen cells were significantly lower in IMD-0354-treated mice than in irradiated mice, suggesting that the IKKβ inhibitor IMD-0354 exerts a radiomitigative effect by suppressing the NF-κB.

The Radioprotective Effect of Resveratrol Against Genotoxicity Induced by γ-Irradiation in Mice Blood Lymphocytes.

In this study, we evaluated whether the protective potential of resveratrol (RSV; 3,5,4′-trihydroxy-trans-stilbene) against γ-radiation caused damages in peripheral blood lymphocyte of mice. Resveratrol as a polyphenolic compound scavenges free radicals. Various doses of RSV were administered intraperitoneally 2 hours to adult male mice before a single dose of whole-body γ-irradiation (2 Gy). To assess the protective ability of RSV, the alkaline comet assay in blood lymphocyte of mice was performed and the total comet score was evaluated. The results of the alkaline comet assay showed that RSV significantly inhibited radiation-induced DNA damage. We observed that RSV protects blood lymphocyte against radiation-induced damage in mice.

Protective effects of Sasa quelpaertensis nakai extract on radiation-induced oxidative stress in immune cells

Sasa quelpaertensis Nakai is a type of edible bamboo grass distributed on Jeju Island, Korea. S. quelpaertensis has been used as afolk medicine for treatment of a variety of ailments. It has been reported to present biological effects, including anti-inflammatory and antioxidant effects. In this study, we demonstrate that S. quelpaertensis Nakai extract (SNE) rescues immunocytes from gamma radiation-induced apoptosis and oxidative DNA damage. We examined the cytotoxicity, cell proliferation, DNA damage, apoptosis, and generation of reactive oxygen species (ROS) in mice given SNE for 45 days in immune cells. To determine the splenocytes protection capability of SNE, gamma-ray was irradiated to the whole body of C57BL/6 mice. Our results suggest that SNE stimulated the proliferation of splenocytes without cytotoxic effects. In addition, SNE not only decreased DNA damage but also reduced apoptosis of splenocytes, and attenuated the production of ROS generation in hydrogen peroxide-induced splenocytes. Therefore, SNE can protect against gamma radiation-induced damage in mice.

Amelioration of radiation-induced damages in mice by carotenoid meso-zeaxanthin

Purpose: The present study was undertaken to evaluate the radioprotective effect of meso-zeaxanthin, a xanthophyll carotenoid with profound antioxidant activity.Materials and methods: Swiss albino mice were treated with different doses of meso-zeaxanthin (50 and 250 mg/kg body weight, orally) five days before irradiation and sacrificed at different time points. The protective effects of meso-zeaxanthin on mortality, haematological parameters, bone marrow cellularity and gastrointestinal system of irradiated mice were studied. The anti-genotoxic action of meso-zeaxanthin was studied by measuring micronuclei formation, chromosomal aberrations and DNA damage (comet assay).Results: Meso-zeaxanthin administration significantly increased the lifespan of irradiated mice and reduced myelosuppression as evident from increases in white blood cell counts, bone marrow cellularity and the number of maturing monocytes when compared to the myelosuppression in radiation control animals. Meso-zeaxanthin significantly elevated the radiation-induced reduction in the activities of superoxide dismutase, catalase, glutathione peroxidase and glutathione in both liver and intestinal mucosa. The carotenoid-treated animals showed a profound reduction in genotoxic activity which was apparent in decreases in micronuclei formation and chromosomal aberrations. Irradiation also induced damage to cellular DNA as was obvious from increases in comet parameters like tail DNA%, tail moment, tail length and Olive tail moment in the radiation control group. These parameters were decreased by meso-zeaxanthin treatment.Conclusion: Results indicated a radioprotective potential of meso-zeaxanthin.

Protective effect of anthocyanin from Lonicera Caerulea var. Edulis on radiation-induced damage in mice.

The radioprotective effect of anthocyanin extracted from Lonicera caerulea var. edulis (ALC), was studied in ICR mice. Different doses of ALC were intragastrically administered to mice once a day, prior to radiation. After two weeks, the mice received a one-time 5 Gy whole body 60Coγ radiation. The spleen index, thymus index, activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), malondialdehyde (MDA) content, and glutathione (GSH) content in liver tissue were measured. Compared with the radiation control group, the levels of MDA in all ALC treated groups decreased significantly (p < 0.05). Moreover, the GSH content, activities of SOD and GSH-Px in liver tissue were enhanced significantly (p < 0.05) in all ALC groups. These results demonstrate that ALC may be a potential radioprotector, and a further study of the molecular mechanism is needed for further application.

Protective effect of an extract of Guipi Pill (归脾丸) against radiation-induced damage in mice

To study the protective effect of an extract of Guipi Pill () against radiation-induced damage. A total of 100 Kunming mice were randomly divided into normal group, model group, positive drug group (treated with radioprotective agent "523", 5 mg/kg at 24 h before irradiation) and two treatment groups, with 20 mice in each group. The extract of water extraction-alcohol precipitation (WAP) from Guipi Pill were administered orally to the mice in the two treatment groups at the dose of 500 and 1,000 mg/kg, respectively, for 6 days prior to whole body radiation (8 Gy). Fifty mice with 10 in each group were used to observe the survival rate 30 days after radiation. The other 50 mice with 10 in each group were sacrificed on day 10 after radiation (6 Gy) in order to take blood, liver and unilateral femur. Pretreatment prior to irradiation with WAP resulted in a significantly higher 30-day survival rate of mice after exposure to a potentially lethal dose of 8-Gy radiation. WAP could significantly increase the total white blood cell count and DNA content of bone marrow, and it also increased the activity of various antioxidant enzymes, such as superoxide dismutase, catalase, total antioxidant capacity and glutathione peroxidase in liver tissue of mice, which were reduced by radiation treatment. Maleic dialdehyde level and bone marrow micronucleus rate were significantly reduced by WAP, which were increased after 6-Gy radiation. WAP of Guipi Pill could increase the 30-day survival rate and the antioxidant capacity as well as protect bone marrow in mice. WAP of Guipi Pill is an effective radioprotective agent.

Protective Effect of Polysaccharides Isolated from &lt;i&gt;Tremella fuciformis&lt;/i&gt; against Radiation-induced Damage in Mice

WTF-B, a type of water-soluble homogeneous polysaccharide, was isolated and purified from Tremella Fuciformis. To investigate the radioprotective effect of WTF-B, we employed a 30-day survival assay. Mice were treated with WTF-B once per day for three consecutive days before 8-Gy gamma irradiation. The treatment groups receiving 54 and 72 mg/kg body weight (b.w.) of WTF-B showed 50% survival post-irradiation. The hematological parameters of the peripheral blood indicated that WTF-B, when administered at doses of 72 mg/kg b.w., significantly restored hemoglobin, white blood cell counts and red blood cell counts by the 14th day and 18th day. In addition, spleen colony forming units (CFU-S), the number of nucleated cells in bone marrow (BMNC) and spleen index were used to investigate the radioprotective effect of WTF-B on the hematopoietic system. The treatment groups receiving WTF-B at 18, 54 and 72 mg/kg b.w. doses presented significantly higher BMNC compared to radiation-only group. The group administered 72 mg/kg b.w. WTF-B presented a significant change in CFU-S compared to the radiation-only group. We also completed micronucleus and chromosome aberration assays to explore genotoxicity. The results of those assays indicated that the number of micronuclei induced by 2-Gy irradiation in a group treated with 72 mg/kg b.w. WTF-B decreased from 30.30‰ to 11.32‰. The chromosomal aberration produced by 3-Gy irradiation in the group receiving 72 mg/kg b.w. WTF-B decreased from 56.01% to 28.13%. The results of the present study indicate a potential use for WTF-B as a radioprotector.

Protective Role of Tea Polyphenols in Combination against Radiation‐induced Haematopoietic and Biochemical Alterations in Mice

The purpose of this study was to investigate the radioprotective effects of tea polyphenols (TPs) in various combinations against radiation-induced damage in mice. Mice were divided into different groups: non-irradiated control, irradiated control, amifostine (43.6 mg/kg, i.v. 30 min before irradiation; positive control) and various combinations of tea polyphenols in different doses. The radioprotective effect on the haematopoietic system, serum cytokines and endogenous antioxidant enzymes were studied. TP50, containing approximately 50% of (-)-epigallochatechin-3-gallate in addition to other catechins, showed the greatest radioprotective effect against radiation-induced changes in haematological parameters (red blood cell count, white blood cell count and haemoglobin), and maintained the spleen and thymus indices unchanged (spleen or thymus weight/body weight × 1000). Tea polyphenols also significantly decreased radiation-induced lipid peroxidation (malondialdehyde levels), elevated endogenous antioxidant enzymes (superoxide dismutase) and reduced the serum cytokines which were elevated in radiation-induced toxicity. This evidence shows the potential of tea polyphenols, particularly in the combination found in TP50, as radioprotectors in mice, especially regarding recovery of the haematopoietic system, antioxidant potential activity and reduction of inflammatory cytokines.

Protective Effect of an Extract of Emblica officinalis Against Radiation-Induced Damage in Mice

The radioprotective effect of Emblica officinalis extract (EOE) was studied in mice. Swiss albino mice were exposed to gamma rays (5 Gy) in the absence (control) or presence (experimental) of EOE, orally 100 mg/kg body weight, once daily for 7 consecutive days. A specimen of small intestine (jejunum) was removed from the mice and studied at different autopsy intervals from 12 hours to 30 days. In control animals, crypt cell population, mitotic figures, and villus length were markedly reduced on day 1; these later started to increase progressively but did not attain the normal level even at the last autopsy interval. The animals receiving EOE prior to irradiation had a higher number of crypt cells and mitotic figures when compared with non-drug-treated control at all the autopsy intervals. Irradiation of animals resulted in a dose-dependent elevation in lipid peroxidation and a reduction in glutathione as well as catalase concentration in the intestine at 1 hour post-irradiation. In contrast, EOE treatment before irradiation caused a significant depletion in lipid peroxidation and elevation in glutathione and catalase levels.
