A Facile Method for the Synthesis of Novel Quinazolinone Compounds

Abstract A simple and easy method is provided for the synthesis of the previously unknown 2-substituted-methylene-4(3H)-1,2-dihydroquinazolinones 3 in good yields by the reaction of ketene dithioacetals 1 with anthranilamide (2).

SYNTHESIS AND ANTIMICROBIAL EVALUATION OF SOME QUINAZOLINONE DERIVATIVES

Several quinazolinone compounds were prepared having thiadiazole moiety joined to the 4(3H)-quinazolinone ring at 2-position through thiomethyl bridge. The antimicrobial activities of the prepared compounds have been investigated.

Synthesis of 5‐fluoro‐2‐methyl‐3‐(2‐trifluoromethyl‐1,3,4‐thiadiazol‐5‐yl)‐4(3H)‐quinazolinone and related compounds with potential antiviral and anticancer activity

AbstractThe synthesis of ten new substituted 1,3,4‐thiadiazolyl‐4(3H)‐quinazolinones 8–11, 13, 17, and 20–23 is reported. Compounds 8–11 were prepared by condensation of 5‐fluoro‐2‐methyl‐3,1‐benzoxazin‐4‐one (3) and 5‐substituted 2‐amino‐1,3,4‐thiadiazoles 4–7. Compound 13 was obtained by condensation of 5‐fluoro‐2‐methyl‐3,1‐benzoxazin‐4‐one (3) with DL‐α‐amino‐ϵ‐caprolactam (12). Compound 17 was synthesized by condensation of 6‐bromo‐2‐methyl‐3,1‐benzoxazin‐4‐one (16) and 2‐amino‐5‐t‐butyl‐1,3,4‐thiadiazole (5). Compounds 20–23 were obtained by condensation of 5‐chloro‐6,8‐dibromo‐2‐methyl‐3,1‐benzoxazin‐4‐one (19) and 5‐substituted 2‐amino‐1,3,4‐thiadiazoles 4–7, respectively. The substituted 3,1‐benzoxazin‐4‐ones 3, 16, and 19 were obtained in good yield by refluxing the appropriate anthranilic acid, 1,15, and 18 with acetic anhydride (2).

A New Tablet Disintegrating Agent: Cross-Linked Polyvinylpyrrolidone

Cross-linked polyvinylpyrrolidone was studied for its disintegration property in comparison to starch USP and alginic acid. Certain physical parameters of the disintegrants (maximum moisture sorption, hydration capacity, bulk density, and specific surface area) were determined for the purpose of differentiating their relative efficiency. A linear relationship was found to exist when the maximum moisture sorption was plotted versus the specific surface area for each disintegrant. It was postulated that capillary activity of the cross-linked polyvinylpyrrolidone for water appears responsible for its tablet disintegration property. Cross-linked polyvinylpyrrolidone demonstrated superiority over starch USP and alginic acid in most of the experimental tablet formulations made by either dry or wet granulation. A quinazolinone compound was formulated into tablets employing each disintegrant to provide identical disintegration times, and these tablets were submitted to dissolution rate analysis. The dissolution results showed some differences for those made by direct compression but no variation for wet granulated tablets.

Dissolution of Poorly Water-Soluble Drugs I: Some Physical Parameters Related to Method of Micronization and Tablet Manufacture of a Quinazolinone Compound

An investigation was conducted to analyze the dissolution behavior of a poorly water-soluble drug. Two specific methods of micronization were employed which provided drug forms of different physical characteristics, specific surface area, and particle-size range. The drug, a quinazolinone compound, was micronized by employing spray drying and air attrition. The micronsized drugs, pelletized pure drugs, and tablets prepared from same by direct-compaction were subjected to dissolution-rate studies at pH 1.2 and 3. In vitro dissolution data obtained have demonstrated a significant variation for the pure spray-dried and air-attritioned drugs at pH 1.2, although physical specifications had suggested the contrary of that reported. The pure drug forms, when pelletized, resulted in elimination of the difference observed for dissolution rates with the pure drug powders. Dilution of the pure drugs with tablet excipients and subsequent direct compaction into tablets resulted in an improvement of dissolution behavior compared to that obtained with the pure drug forms.

The direct effects of metolazone diuresis on the isolated bloodless perfused canine kidney.

AbstractRenal bloodless perfusions under normothermic, normobaric conditions with oxygen or helium gas equilibrations were conducted in 14 canine kidneys. A new diuretic, metolazone (SR‐720—22), a quinazolinone compound, was evaulated for possible direct renal effects. Metolazone exerts direct renal effects with or without oxygen characterized by (1) fall in renal resistance; (2) rise in urine flow rate and osmolar clearance; (3) no change in GFR, in inulin extraction, or in the proximal tubular reabsorption of sodium. The agent at low doses is thus a potent and direct active diuretic. Lack of alteration in renal acidification processes as well as activity under anoxic conditions may indicate future usefulness in renal insufficiency states.
