Alfalfa Mosaic Virus and White Clover Mosaic Virus Combined Infection Leads to Chloroplast Destruction and Alterations in Photosynthetic Characteristics of Nicotiana benthamiana.

Alfalfa mosaic virus (AMV) is one of the most widely distributed viruses; it often exhibits combined infection with white clover mosaic virus (WCMV). Even so, little is known about the effects of co-infection with AMV and WCMV on plants. To determine whether there is a synergistic effect of AMV and WCMV co-infection, virus co-infection was studied by electron microscopy, the double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), and real-time fluorescence quantitative PCR (RT-qPCR) of AMV and WCMV co-infection in Nicotiana benthamiana. Meanwhile, measurements were carried out on the photosynthetic pigments, photosynthetic gas exchange parameters, and chlorophyll fluorescence parameters. The results showed that the most severe disease development was induced by AMV and WCMV co-infection, and the disease grade was scale 7. N. benthamiana leaves induced mottled yellow-green alternating patterns, leaf wrinkling, and chlorosis, and chloroplasts were observed to be on the verge of disintegration. The relative accumulation of AMV CP and WCMV CP was significantly increased by 15.44-fold and 10.04-fold upon co-infection compared to that with AMV and WCMV single infection at 21 dpi. In addition, chlorophyll a, chlorophyll b, total chlorophyll, the net photosynthetic rate, the water use efficiency, the apparent electron transport rate, the PSII maximum photochemical efficiency, the actual photochemical quantum yield, and photochemical quenching were significantly reduced in leaves co-infected with AMV and WCMV compared to AMV- or WCMV-infected leaves and CK. On the contrary, the carotenoid content, transpiration rate, stomatal conductance, intercellular CO2 concentration, minimal fluorescence value, and non-photochemical quenching were significantly increased. These findings suggest that there was a synergistic effect between AMV and WCMV, and AMV and WCMV co-infection severely impacted the normal function of photosynthesis in N. benthamiana.

Phytoplankton Diversity, Spatial Patterns, and Photosynthetic Characteristics Under Environmental Gradients and Anthropogenic Influence in the Pearl River Estuary.

The Pearl River Estuary (PRE) is one of the world's most urbanized subtropical coastal systems. It presents a typical environmental gradient suitable for studying estuarine phytoplankton communities' dynamics and photosynthetic physiology. In September 2018, the maximum photochemical quantum yield (Fv/Fm) of phytoplankton in different salinity habitats of PRE (oceanic, estuarine, and freshwater zones) was studied, revealing a complex correlation with the environment. Fv/Fm of phytoplankton ranged from 0.16 to 0.45, with taxa in the upper Lingdingyang found to be more stressed. Community composition and structure were analyzed using 18S rRNA, accompanied by a pigment analysis utilized as a supplementary method. Nonmetric multidimensional scaling analysis indicated differences in the phytoplankton spatial distribution along the estuarine gradients. Specificity-occupancy plots identified different specialist taxa for each salinity habitat. Dinophyta and Haptophyta were the predominant taxa in oceanic areas, while Chlorophyta and Cryptophyta dominated freshwater. Bacillariophyta prevailed across all salinity gradients. Canonical correlation analysis and Mantel tests revealed that temperature, salinity, and elevated nutrient levels (i.e., NO3--N, PO43--P, and SiO32--Si) associated with anthropogenic activities significantly influenced the heterogeneity of community structure. The spatial distribution of phytoplankton, along with in situ photosynthetic characteristics, serves as a foundational basis to access estuarine primary productivity, as well as community function and ecosystem health.

Elucidating and Optimizing the Photochemical Mechanism of Coumarin-Caged Tertiary Amines.

Photoactivatable or "caged" pharmacological agents combine the high spatiotemporal specificity of light application with the molecular specificity of drugs. A key factor in all optopharmacology experiments is the mechanism of uncaging, which dictates the photochemical quantum yield and determines the byproducts produced by the light-driven chemical reaction. In previous work, we demonstrated that coumarin-based photolabile groups could be used to cage tertiary amine drugs as quaternary ammonium salts. Although stable, water-soluble, and useful for experiments in brain tissue, these first-generation compounds exhibit relatively low uncaging quantum yield (Φu < 1%) and release the toxic byproduct formaldehyde upon photolysis. Here, we elucidate the photochemical mechanisms of coumarin-caged tertiary amines and then optimize the major pathway using chemical modification. We discovered that the combination of 3,3-dicarboxyazetidine and bromine substituents shift the mechanism of release to heterolysis, eliminating the formaldehyde byproduct and giving photolabile tertiary amine drugs with Φu > 20%─a 35-fold increase in uncaging efficiency. This new "ABC" cage allows synthesis of improved photoactivatable derivatives of escitalopram and nicotine along with a novel caged agonist of the oxytocin receptor.

Perspective on Theoretical and Experimental Advances in Atmospheric Photochemistry.

Research that explores the chemistry of Earth's atmosphere is central to the current understanding of global challenges such as climate change, stratospheric ozone depletion, and poor air quality in urban areas. This research is a synergistic combination of three established domains: earth observation, for example, using satellites, and in situ field measurements; computer modeling of the atmosphere and its chemistry; and laboratory measurements of the properties and reactivity of gas-phase molecules and aerosol particles. The complexity of the interconnected chemical and photochemical reactions which determine the composition of the atmosphere challenges the capacity of laboratory studies to provide the spectroscopic, photochemical, and kinetic data required for computer models. Here, we consider whether predictions from computational chemistry using modern electronic structure theory and nonadiabatic dynamics simulations are becoming sufficiently accurate to supplement quantitative laboratory data for wavelength-dependent absorption cross-sections, photochemical quantum yields, and reaction rate coefficients. Drawing on presentations and discussions from the CECAM workshop on Theoretical and Experimental Advances in Atmospheric Photochemistry held in March 2024, we describe key concepts in the theory of photochemistry, survey the state-of-the-art in computational photochemistry methods, and compare their capabilities with modern experimental laboratory techniques. From such considerations, we offer a perspective on the scope of computational (photo)chemistry methods based on rigorous electronic structure theory to become a fourth core domain of research in atmospheric chemistry.

Effects of nitrogen addition and Bothriochloa ischaemum and Lespedeza davurica mixture on plant chlorophyll fluorescence and community production in semi-arid grassland.

Grass-legume mixture can effectively improve productivity and stimulate overyielding in artificial grasslands, but may be N-limited in semi-arid regions. This study investigated the effects of N addition on chlorophyll fluorescence and production in the grass-legume mixtures community. An N addition experiment was conducted in the Bothriochloa ischaemum and Lespedeza davurica mixture community, with seven mixture ratios (B0L10, B2L8, B4L6, B5L5, B6L4, B8L2, and B10L0) according to the sowing abundance of B.ischaemum and L.davurica and four N addition levels, N0, N25, N50, and N75 (0,25,50,75kgNhm-2 a-1), respectively. We analyzed the response of chlorophyll fluorescence parameters of the two species, the rapid light-response curves of chlorophyll fluorescence, as well as aboveground biomass (AGB) and overyielding. Our results showed that the two species showed different photosynthetic strategies, with L.davurica having significantly higher initial fluorescence (Fo), effective photochemical quantum yield of PSII (ΦPSII), and coefficient of photochemical fluorescence quenching (qP) than B. ischaemum, consisting with results of rapid light-response curves. N addition and mixture ratio both had significant effects on chlorophyll fluorescence and AGB (p<0.001). The ΦPSII and qP of L.davurica were significantly lowest in B5L5 and B6L4 under N addition, and the effect of N varied with mixture ratio. The photosynthetic efficiency of B. ischaemum was higher in mixture than in monoculture (B10L0), and ΦPSII was significantly higher in N50 than in N25 and N50 at mixture communities except at B5L5. The community AGB was significantly higher in mixture communities than in two monocultures and highest at B6L4. In the same mixture ratio, the AGB was highest under the N50. The overyielding effects were significantly highest under the N75 and B6L4 treatments, mainly attributed to L.davurica. The partial least squares path models demonstrated that adding N increased soil nutrient content, and complementary utilization by B.ischaemum and L.davurica increased the photosynthetic efficiency. However, as the different photosynthetic strategies of these two species, the effect on AGB was offset, and the mixture ratio's effects were larger than N. Our results proposed the B6L4 and N50 treatments were the optimal combination, with the highest AGB and overyielding, moderate grass-legume ratio, optimal community structure, and forage values.

Enhanced quantum efficiency of dicyano BODIPYs over difluoro BODIPYs: A DFT approach

The modified BODIPY (pyrromethene) dyes with cyano (-CN), substituting usual fluorine (-F), at the 4-position may give enhanced photochemical stability and quantum yield of fluorescence (QYF) when compared to the corresponding fluoro derivative. We have investigated and discussed comparatively the structural parameters of the ground (S0) and excited (S1) state geometries of a few widely used BODIPY dyes, both 4-cyano and 4-fluoro pyrromethene (PM), through DFT and TD-DFT studies and established the reasons for improved QYF of the cyano derivatives. The electrophilicity index indicates the stability of the excited geometry of the cyanated BODIPY and the calculated transition dipole moments reveal a longer lifetime of the excited state (S1) for the cyano BODIPY. The higher singlet and triplet gap in cyanated BODIPY accounts for higher QYF due to restricted transition from singlet to triplet state. This suggests that a population inversion, crucial for laser action, is easier to build up for cyano derivative than for the commonly used fluoro analogue while other gain/loss parameters of the PM dye lasers remain similar. Hence, 4-cyano derivatives of the PM dyes may be useful for more efficient lasing applications. The non-linear optical properties of the PM dyes are also investigated.

Diverse nitrogen enrichments enhance photosynthetic resistance of Sargassum horneri to ultraviolet radiation

In recent years, golden tides caused by floating Sargassum have induced severe ecological disasters globally. Eutrophication is a significant factor contributing to the massive spread of Sargassum golden tides. Furthermore, the thalli of Sargassum that float on the ocean surface are subjected to more ultraviolet radiation (UVR). The coupled impact of eutrophication and UVR on the photosynthetic physiology of golden tide species remains unclear. In this study, the thalli of Sargassum horneri, known to cause golden tide, were cultured and acclimated to three distinct nitrogen (N) conditions (natural seawater, NSW; NH4+-N enrichment; and NO3–N enrichment) for 6 days. Subsequently, the thalli were exposed to two different radiation treatments (photosynthetically active radiation (150 W m-2), PAR, 400–700 nm; PAR (150 W m-2) + UVR (28 W m-2), 280–700 nm) for 120 min, to investigate the photosynthetic effects of UVR and N on this alga. The findings demonstrated that exposure to UVR impeded the photosynthetic capacity of S. horneri, as evidenced by a decrease in the maximum photochemical quantum yield (Fv/Fm), photosynthetic efficiency (α) and chlorophyll content. Under diverse N-enrichment conditions, the alga tended to adopt various strategies to mitigate the adverse effects of UVR. NH4+-enrichment dissipated excess UVR energy through a greater increase in non-photochemical quenching (NPQ). While NO3–enrichment protected alga by enhancing N assimilation (higher nitrate reductase activity (NRA) and soluble protein content), and maintained a stable energy captured per unit reaction center for electron transfer (ET0/RC) and a higher net photosynthetic rate. Although different N enrichments could not completely offset the damage caused by UV radiation, they secured the photoprotective ability of S. horneri in several ways.

Chlorophyll fluorescence characteristics and H2O2 contents of Chinese tallow tree are dependent on population origin, nutrients and salinity.

Plants from invasive populations often have higher growth rates than conspecifics from native populations due to better environmental adaptability. However, the roles of improved chlorophyll fluorescence or antioxidant defenses in helping them to grow better under adverse situations are insufficient, even though this is a key physiological question for elucidating mechanisms of plant invasion. Here, we conducted experiments with eight native (China) and eight introduced (USA) populations of Chinese tallow tree (Triadica sebifera). We tested how salinity, nutrients (overall amount or N:P in two separate experiments) and their interaction affected T. sebifera aboveground biomass, leaf area, chlorophyll fluorescence and antioxidant defenses. Plants from introduced populations were larger than those from native populations, but salinity and nutrient shortage (low nutrients or high N:P) reduced this advantage, possibly reflecting differences in chlorophyll fluorescence based on their higher PSII maximum photochemical efficiency (F v/F m) and PSI maximum photo-oxidizable P700 in higher nutrient conditions. Native population plants had lower F v/F m with saline. Except in high nutrients/N:P with salinity, introduced population plants had lower electron transfer rate and photochemical quantum yield. There were no differences in antioxidant defenses between introduced and native populations except accumulation of hydrogen peroxide (H2O2), which was lower for introduced populations. Low nutrients and higher N:P or salinity increased total antioxidant capacity and H2O2. Our results indicate that nutrients and salinity induce differences in H2O2 contents and chlorophyll fluorescence characteristics between introduced and native populations of an invasive plant, illuminating adaptive mechanisms using photosynthetic physiological descriptors in order to predict invasions.

Impact of exogenous melatonin foliar application on physiology and fruit quality of wine grapes (Vitis vinifera) under salt stress.

Soil salinisation is an important abiotic stress faced in grape cultivating, leading to weakened plant vigour and reduced fruit quality. Melatonin as a novel hormone has shown positive exogenous application value. Therefore, this study used wine grape (Vitis vinifera ) 'Pinot Noir' as a test material to investigate the changes of foliar spraying with different concentrations of melatonin on the physiology and fruit quality of wine grapes in a field under simulated salt stress (200mmolL-1 NaCl). The results showed that foliar spraying of melatonin significantly increased the intercellular CO2 concentration, maximum photochemical quantum yield of PSII, relative chlorophyll and ascorbic acid content of the leaves, as well as the single spike weight, 100-grain weight, transverse and longitudinal diameters, malic acid, α-amino nitrogen and ammonia content of fruits, and decreased the initial fluorescence value of leaves, ascorbate peroxidase activity, glutathione content, fruit transverse to longitudinal ratio and tartaric acid content of plants under salt stress. Results of the comprehensive evaluation of the affiliation function indicated that 100μmolL-1 melatonin treatment had the best effect on reducing salt stress in grapes. In summary, melatonin application could enhance the salt tolerance of grapes by improving the photosynthetic capacity of grape plants under salt stress and promoting fruit development and quality formation, and these results provide new insights into the involvement of melatonin in the improvement of salt tolerance in crop, as well as some theoretical basis for the development and industrialisation of stress-resistant cultivation techniques for wine grapes.

Effects of foliar iron oxide nanoparticles (Fe3O4) application on photosynthetic parameters, distribution of mineral elements, magnetic behaviour, and photosynthetic genes in tomato (Solanum lycopersicum var. cerasiforme) plants

This study aims to examine the effect of foliar magnetic iron oxide (Fe3O4) nanoparticles (IONP) application on the physiology, photosynthetic parameters, magnetic character, and mineral element distribution of cherry tomatoes (Solanum lycopersicum var. cerasiforme). The IONP suspension (500 mg L−1) was sprayed once (S1), twice (S2), thrice (S3), and four times (S4) a week on seedlings. Upon 21 days of the treatments, photosynthetic parameters (chlorophyll, carotenoids, photosynthetic yield, electron transport rate) were elucidated. Inductively-coupled plasma-optical emission spectrometer (ICP-OES) and vibrating sample magnetometer (VSM) were used to determine the mineral elements and abundance of magnetic power in the seedlings. In addition, the RT-qPCR method was performed to quantify the expressions of photosystem-related (PsaC, PsbP6, and PsbQ) and ferritin-coding (Fer-1 and Fer-2) genes. Results revealed that the physiological and photosynthetic indices were improved upon S1 treatment. The optimal dosage of IONP spraying enhances chlorophyll, carotenoid, electron transport rate (ETR), and effective photochemical quantum yield of photosystem II (Y(II)) but substantially diminishes non-photochemical quenching (NPQ). However, frequent IONP applications (S2, S3, and S4) caused growth retardation and suppressed the photosynthetic parameters, suggesting a toxic effect of IONP in recurrent treatments. Fer-1 and Fer-2 expressions were strikingly increased by IONP applications, suggesting an attempt to neutralize the excess amount of Fe ions by ferritin. Nevertheless, frequent IONP treatment fluctuated the mineral distribution and caused growth inhibition. Although low-repeat foliar applications of IONP (S1 in this study) may help improve plant growth, consecutive applications (S2, S3, and S4) should be avoided.

Comprehensive evaluation and screening identification indexes of heat‐resistance indices in cotton (Gossypium hirsutum L.)

AbstractCotton (Gossypium hirsutum L.) cultivars exhibit varying responses to heat stress. To investigate the heat resistance of various cotton and establish an index system for evaluating their heat resistance, 21 cotton cultivars were selected and subjected to two temperature regimes (CK, average temperature 28°C, 32/24°C; HT, average temperature 38°C, 42/34°C). The results showed that under high temperatures, different changes occurred in individual indexes of cotton, reflecting the differences in heat resistance in cotton cultivars. A total of 21 cotton cultivars could be classified into four types: heat‐tolerant, moderate heat‐tolerant, moderate heat‐sensitive and heat‐sensitive cultivars by multivariate statistical analysis. Moreover, the indexes of net photosynthetic rate (Pn), leaf superoxide dismutase activity (LSOD), the maximum photochemical quantum yield (Fv/Fm), the actual photochemical quantum yield (ΦPSII), leaf malondialdehyde content (LMDA), leaf catalase activity (LCAT), dry matter weight of shoot (SDW) and root malondialdehyde content (RMDA) were determined to be useful for evaluating the cotton heat tolerance by stepwise regression analysis. The pot experiment showed that the reduction of boll number, boll weight and seed cotton yield was more remarkable under HT in the heat‐sensitive cultivar CCRI‐92 than in the heat‐resistant cultivar CCRI‐69, which further verified the screening results. In conclusion, the heat‐sensitive cultivars CCRI‐92 and heat‐resistant cultivar CCRI‐69 which are identified by seedling experiment could serve as ideal experimental materials for studying heat resistance in cotton. The physiological indices such as Pn, LSOD, Fv/Fm, ΦPSII, LMDA, LCAT, SDW and RMDA be employed for assessing the heat tolerance in cotton.

Soil water availability modulates the response of grapevine leaf gas exchange and PSII traits to a simulated heat wave

Background and aimsA better understanding of plant carbon assimilation, water status and photosystem performance responses to combined heat and drought stress would help to optimize grapevine management under such limiting conditions.MethodsGas exchange and chlorophyll fluorescence parameters were measured in potted grapevines, cv Sauvignon Blanc, before, during and after simulated six-day heat (Tmax = 40 °C) wave using heated well-watered (HW), heated drought-stressed (HD), non-heated well-watered (CW) and non-heated dry (CD) vines.ResultsPhotosynthesis and stomatal conductance in HW vines increased during the morning and dropped in the afternoon with respect to CW vines. Daily plant transpiration in HW almost doubled that of CW vines. When grapevines were already exposed to drought, the effects of the heat wave were negligible, with HD plants showing similar leaf photosynthesis and transpiration to their CD counterparts. Heat, but not drought stress, decreased the maximum (Fv/Fm) and effective photochemical quantum yield of PSII (φPSII), and also affected the use of absorbed energy. HW plants dissipated more radiative energy as heat, a protective mechanism of the photosystem, while HD vines increased the energy dissipated by non-regulated non-photochemical pathways, which might lead to photoinhibition damages. The different behavior could be due to the enhanced transpiration rate and consequent decrease in leaf temperature in HW as compared to HD vines. After the heat wave, only HW vines recovered the afternoon values of photosynthesis, stomatal conductance and φPSII to similar levels as those in CW vines.ConclusionDrought had a more significant effect than heat stress on photosynthesis, stomatal conductance and transpiration. The combined heat and drought stress, however, increased the proportion of energy lost by the leaves through harmful non-regulated dissipative pathways. With adequate soil water availability, grapevines withstood the heat wave period through an increase in leaf transpiration, which decreased leaf temperature and protected the PSII from heat damage.

Improved chlorophyll fluorescence, photosynthetic rate, and plant growth of Brassica napus L. after co‐application of biochar and phosphorus fertilizer in acidic soil

AbstractAimsBiochar has been used as an amendment to improve soil fertility and increase crop yield. A pot trial and incubation experiment were conducted to evaluate the effects of combined biochar and phosphate fertilizer applications on changes in soil physiochemical properties, photosynthetic parameters, and plant growth of Brassica napus L. in acidic soil.MethodsBiochar (B) was applied at rates (B0: 0%, B1: 2%, and B2: 4% w/w) to the soil, while phosphorus (P) as KH2PO4 was added at P0: 0 mg kg–1 soil, P1: 100 mg kg–1 soil, and P2: 150 mg kg–1 soil. Non‐amended soil served as the control (Ck).ResultsThe study found that the maximum photochemical quantum yield of photosystem II (Fv/Fm) and non‐photochemical quenching were affected by biochar and P fertilizer treatments. The Fv/Fm was significantly lower in the Ck while the highest in B2P1. Compared to the Ck treatment, the electron transport rate and photosynthetic rate were significantly increased under the application of biochar with P fertilizer. Additionally, malondialdehyde content declined by 68.9% when biochar was added with P fertilizer treatments. Furthermore, the plant P content and plant growth parameters were significantly increased. Biochar with and without P fertilizer treatments increased soil pH significantly, compared to the application of P fertilizer alone. Water‐soluble P decreased with incubation time, while Al‐P decreased significantly by 69.9% and 76.4% under B1 and B2 treatments, respectively, compared to CK.ConclusionThe findings revealed a positive effect of the combined application of biochar with phosphate fertilizer on the growth of oilseed rape in acidic soil.

Effects of Exogenous Melatonin on the Growth and Physiological Characteristics of Ginkgo biloba L. under Salinity Stress Conditions

Ginkgo (Ginkgo biloba L.) is a cherished relic among plants, commonly planted as a street tree. However, it faces cultivation challenges due to escalating soil salinization and widespread snowmelt application. Therefore, this study used 4-year-old Ginkgo seedlings to investigate how exogenous melatonin at varying concentrations affects seedling growth and physiology under salinity stress. The results revealed that appropriate melatonin concentrations (0.02, 0.1 mmol·L−1) significantly mitigated leaf yellowing under different NaCl stress levels. Furthermore, they increased ground diameter, current-year branch growth, relative water concentration, free proline, and soluble sugars in leaves. Melatonin also reduced electrolyte exudation rates, flavonoids, and malonic dialdehyde concentration, while enhancing peroxidase and superoxide dismutase activities. This led to reduced chlorophyll content, photosynthetic rate, stomatal conductance, and transpiration rate, stabilizing intercellular CO2 concentration, preserving photosynthetic structures, and enhancing photosynthetic rates. Additionally, the decline in the photosynthetic electron transport rate, the effective photochemical quantum yield of PSII, and the potential efficiency of primary conversion of light energy of PSII was alleviated. Minimal fluorescence and the non-photochemical quenching coefficient also improved. However, high melatonin concentration (0.5 mmol·L−1) exacerbated salinity stress. After analyzing composite scores, the 0.02 mmol·L−1 melatonin treatment was most effective in alleviating NaCl stress, while the 0.5 mmol·L−1 treatment intensified physiological stress under 200 mmol·L−1 NaCl stress. Principal component analysis and correlation analysis identified seven physiological indicators (photosynthetic rate, transpiration rate, photosynthetic electron transport rate, minimal fluorescence, superoxide dismutase, free proline, and chlorophyll a) and three growth indicators (ground diameter, branch length, and current-year branch thickness) as key markers for rapid salinity stress assessment in Ginkgo. These findings are crucial for addressing challenges associated with snowmelt’s impact on roadside Ginkgo trees, expanding planting areas, and breeding exceptional salt-tolerant Ginkgo varieties.

Enhanced photochemical efficiency of PSII in Prosopis juliflora suggests contribution to invasion advantage over native C3 xero-halophytes under salt stress.

Chlorophyll a fluorescence parameters related to PSII photochemistry, photoprotection and photoinhibition were investigated in four C3 plant species growing in their natural habitat: Prosopis juliflora ; Abutilon indicum ; Salvadora persica ; and Phragmites karka . This study compared the light reaction responses of P. juliflora , an invasive species, with three native co-existing species, which adapt to varying water deficit and high salt stress. Chlorophyll a fluorescence quenching analyses revealed that P. juliflora had the highest photochemical quantum efficiency and yield, regulated by higher fraction of open reaction centres and reduced photoprotective energy dissipation without compromising the integrity of photosynthetic apparatus due to photoinhibition. Moreover, the elevated values of parameters obtained through polyphasic chlorophyll a fluorescence induction kinetics, which characterise the photochemistry of PSII and electron transport, highlighted the superior performance index of energy conservation in the transition from excitation to the reduction of intersystem electron carriers for P. juliflora compared to other species. Enhanced pigment contents and their stoichiometry in P. juliflora apparently contributed to upregulating fluxes and yields of energy absorbance, trapping and transport. This enhanced photochemistry, along with reduced non-photochemical processes, could explain the proclivity for invasion advantage in P. juliflora across diverse stress conditions.

Physiological and Photosynthetic Characteristics of Pumpkin Seedlings Cultivated Under Low-Light Stress

Low-light stress is known to have a significant impact on pumpkin yield in greenhouse conditions, it is crucial to understand the characteristics of low-light tolerance and identify resistant germplasm resources. In this study, four pumpkins (Cucurbita moschata) varieties were used, including three inbred lines (10–32, NY-1, and JB-1) and one hybrid line (NY-1×JB-1). Various low-light treatments (T0, no shade; T40, 40% shading; T80, 80% shading) were set, to explore their effects on morphology, photosynthetic characteristics, osmotic regulation substance content, and antioxidant enzyme activity in pumpkin seedling leaves. In the results, seedlings of inbred lines 10–32 and JB-1, as well as hybrid NY-1×JB-1, exhibited low-light resistance with shading rate increases. The maximum photochemical quantum yield of photosystem II (Fv/Fm) of all varieties decreased under low-light stress. Catalase (CAT) activity initially increased and then decreased, while superoxide dismutase (SOD) activity showed the opposite trend. Peroxidase (POD) activity gradually increased in 10–32, JB-1, and NY-1×JB-1, with JB-1 showing a significant increase at T80, and NY-1 displaying an initial decrease followed by an increase. Noteablely, the soluble proline (Pro) content decreased in 10–32, NY-1, and NY-1×JB-1. Significant differences in malondialdehyde (MDA) content were observed among the seedlings of four varieties. The MDA contents increased in 10–32 and JB-1 under T0 and T40, while showed no significant change in NY-1. The MDA content at T80 significantly decreased in NY-1, and first decreased and then significantly increased in NY-1×B-1. The soluble sugar content in 10–32 remained unchanged between T0 and T40, but significantly increased at T80. While in JB-1 and NY-1, it initially increased then decreased with increasing shading rate. The trend in NY-1×JB-1 was opposite to that of JB-1 and NY-1, with initially decreased followed by a significant increase. In conclusion, morphological characteristics associated with low-light tolerance can serve as reference indices for breeding low-light tolerance pumpkins. These parameters can be used as reference indices to assess pumpkin resistance to low-light stress.

Effect of growth medium composition on the viability and genetic stability of Chrysanthemum × morifolium Ramat. explants under in vitro cold storage conditions

Cultivation of plants at low positive temperatures is promising for the in vitro preservation of valuable Chrysanthemum × morifolium Ramat. varieties. The study aims to examine the effect of chlorocholine chloride concentrations in a specialized medium on the viability and the physiological and morpho-anatomical parameters of chrysanthemum plants (Excel, Sheer Purple, and William Seward varieties), as well as their genetic stability after 12 months of cold storage under in vitro gene bank conditions. In vitro microshoot tips (without leaves) were placed on a 1/4 Murashige and Skoog medium supplemented with 60.0 g/L sucrose and 0.2–0.8 g/L chlorocholine chloride. The explants were stored in cooling chambers at a temperature of 4 °C. After 12 months of preservation, a reduction was observed in growth kinetics with increasing concentration of chlorocholine chloride in the medium. The viability of explants decreased from 96 to 91% at higher concentrations of chlorocholine chloride. The effective photochemical quantum yield of photosystem II decreased more than twice under cold storage conditions as compared to the control. Several histological features were identified in the leaves of in vitro explants cold-stored in the gene bank: decreased in leaf thickness and mesophyll density in leaf blades, degradation of palisade tissue, and a high degree of transpiration regulation. The SSR and ISSR analyses were used to show that the microshoots of the examined chrysanthemum varieties were identical to the original plants following in vitro cold storage. The conducted studies revealed that the presence of 0.6–0.8 g/L chlorocholine chloride in the medium reduces the growth kinetics of explants while maintaining their high viability and genetic stability.

The GmCYP2-GmHAL3 module regulates salt tolerance in soybean seedlings

Soybean (Glycine max L. Merr) production is severely affected by soil salinization as an important crop. The discovery of salt stress-responsive genes is essential for soybean breeding. To our knowledge, cyclophilins (CYPs) play important roles in salt stresses in crops such as rice and cotton, except in soybean. This study cloned GmCYP2 from soybean salt-tolerant cultivar Suxie No. 1 and obtained ten stable transgenic soybean lines overexpressing GmCYP2 by Agrobacterium rhizogenes-mediated transformation. Two of the higher-expressing transgenic lines (OE1 and OE2) were used to compare salt tolerance with the wild type (WT). Under the salt stress, the chlorophyll content, the maximum efficiency of PSII photochemistry (Fv/Fm) and actual photochemical quantum yield [Y(II)] of OE1 and OE2 lines were significantly higher than those of the WT. Meanwhile, the accumulation of malondialdehyde (MDA) in the OE1 and OE2 lines was significantly lower than that in the WT. In addition, the Na+/K+ ratio was also significantly lower than that of the WT in leaves and roots. The results also confirmed that the salt tolerance of GmCYP2-silencing soybean seedlings obtained by virus-induced gene silencing (VIGS) was reduced compared to the control. Subcellular localization showed that GmCYP2 was expressed in the plasma membrane and nucleus of Nicotiana benthamiana. RNA-seq results demonstrated that GmCYP2 is involved in key regulators of the ABA and Ca2+ signaling pathway. Yeast two-hybrid, bimolecular fluorescence complementation (BiFC) and split-luciferase complementation assay demonstrated that GmCYP2 protein interacts with salt-associated halotolerance 3 (HAL3) protein. These results suggest that the GmCYP2-GmHAL3 module enhances salt tolerance of soybean seedlings by maintaining the Na+/K+ ratio and good photosynthetic state and it may be involved in the ABA and Ca2+ signaling pathway.

Effect of Electron-donating Group on NO Photolysis of {RuNO}6 Ruthenium Nitrosyl Complexes with N2 O2 Lgands Bearing π-Extended Rings.

In this study, we introduced the electron-donating group (-OH) to the aromatic rings of Ru(salophen)(NO)Cl (0) (salophenH2 =N,N'-(1,2-phenylene)bis(salicylideneimine)) to investigate the influence of the substitution on NO photolysis and NO-releasing dynamics. Three derivative complexes, Ru((o-OH)2 -salophen)(NO)Cl (1), Ru((m-OH)2 -salophen)(NO)Cl (2), and Ru((p-OH)2 -salophen)(NO)Cl (3) were developed and their NO photolysis was monitored by using UV/Vis, EPR, NMR, and IR spectroscopies under white room light. Spectroscopic results indicated that the complexes were diamagnetic Ru(II)-NO+ species which were converted to low-spin Ru(III) species (d5 , S=1/2) and released NO radicals by photons. The conversion was also confirmed by determining the single-crystal structure of the photoproduct of 1. The photochemical quantum yields (ΦNO s) of the photolysis were determined to be 0>1, 2, 3 at both the visible and UV excitations. Femtosecond (fs) time-resolved mid-IR spectroscopy was employed for studying NO-releasing dynamics. The geminate rebinding (GR) rates of the photoreleased NO to the photolyzed complexes were estimated to be 0≃1, 2, 3. DFT and TDDFT computations found that the introduction of the hydroxyl groups elevated the ligand π-bonding orbitals (π (salophen)), resulting in decrease of the HOMO-LUMO gaps in 1-3. The theoretical calculations suggested that the Ru-NNO bond dissociations of the complexes were mostly initiated by the ligand-to-ligand charge transfer (LLCT) of π(salophen)→π*(Ru-NO) with both the visible and UV excitations and the decreasing ΦNO s could be explained by the changes of the electronic structures in which the photoactivable bands of 1-3 have relatively less contribution of transitions related with Ru-NO bond than those of 0.

Nitrogen enrichment ameliorates the stimulatory effects of reduced salinity on photosynthesis and growth of phytoplankton assemblages in the northern South China sea

Anthropogenic activities and climate change are exacerbating the occurrence of extreme rainfall that normally brings large amounts of nutrient-rich freshwater from the land to the sea, resulting in acute salinity decrease and nutrient increase. To evaluate the effects of such changes in salinity and nutrients, we tracked the changes in photosynthetic efficiency and growth of phytoplankton assemblages from the northern South China Sea at 5 salinity levels and at an intermediate salinity level with 3 or 4 nitrogen concentrations. The results showed that the reduction of salinity reduced the maximum photochemical quantum yield (FV/FM) of photosystem II of phytoplankton within a short-term cultivation (i.e. 24-72 h), followed by a stimulatory effect. The reducing effect of reduced salinity lasted longer in the nearshore area than in the offshore area, so the stimulatory effect occurred later in the former area. Nitrogen enrichment mitigated the negative effect of reduced salinity in short-term cultivation and showed a positive effect on FV/FM in long-term cultivation. Moreover, both reduced salinity and enriched nitrogen stimulated phytoplankton growth after an acclimation period. Our results suggest that the reduced salinity stresses phytoplankton in the short term, which is mitigated by nitrogen enrichment, but benefits them in the long term. This sheds light on how phytoplankton thrive and even flourish in coastal or estuarine environments where salinity and nutrients typically covary strongly after extreme rainfall.
