Development and validation of a pressurised liquid extraction liquid chromatography–electrospray–tandem mass spectrometry method for β-lactams and sulfonamides in animal feed

This study presents the development and validation of a sensitive and fast (30 min extraction time and 10 min chromatographic run) method for the detection of penicillins, cephalosporins and sulfonamides in animal feed using pressurised liquid extraction and solid phase extraction as extraction and pre-concentration procedures, followed by liquid chromatography–quadrupole-linear ion-trap mass spectrometry. The developed method was validated showing limits of detection ranging from 0.12 (ampicillin) to 3.94 ng/g (amoxicillin), instrumental and analytical linearity coefficients above 0.99 in both standard and matrix-based solutions as well as relative recoveries ranging from 71% (cefoperazone) to 115% (cefazolin). Repeatability of the method was in the range of 1–9% (RSD %), whereas reproducibility ranged from 3% to 13% (RSD %). The developed and validated method was finally applied to the analysis of real feed samples. The results showed 10 out of 18 analytes to be present in at least one sample and all 14 samples to contain at least one analyte. Penicillin V, oxacillin, ceftiofur, cefoperazone, cefalexin, cefazolin, sulfamethoxypyridazine and sulfapyridine were not detected in any of the samples analysed. Considering the ban of antibacterials as growth promoters added in animal feed, this method is capable of detecting the low concentrations that could result from failure to comply with the regulations or on-site contamination.

Phenolic Compounds in Different Barley Varieties: Identification by Tandem Mass Spectrometry (QStar) and NMR; Quantification by Liquid Chromatography Triple Quadrupole-Linear Ion Trap Mass Spectrometry (Q-Trap)

Barley (Hordeum vulgare) is an important cereal that has many applications; as a human food, in malt products and as livestock feed. The content of soluble health-promoting fibers, beta-glucans, varies substantially among barley varieties. In the present study, the content of secondary metabolites with potential positive health effects in different high- and low-beta-glucan barley varieties was studied. Five different flavanols were isolated and identified: (2R,3S)-catechin-7-O-beta-D-glucopyranoside (1), prodelphinidin B3 (2), procyanidin B3 (3), (+)-catechin (4) and procyanidin B1 (5). Procyanidin B1 has never been reported in barley grains before. The compounds were identified using 1H NMR and quadrupolar time-of-flight mass spectrometry. A quantitative analytical method was developed for prodelphinidin B3, procyanidin B3 and (+)-catechin in liquid chromatography triple quadrupole-linear ion trap mass spectrometry and these compounds were quantified in all varieties, together with four phenolic acids: ferulic acid, vanillic acid, p-coumaric acid and p-hydroxybenzoic acid. Catechin was the compound that was present at the highest concentration in all varieties. The variation, between cultivars, in catechin concentration varied four fold. A Principal Component Analysis indicated no correlation between concentrations of beta-glucan and secondary metabolites. Concentrations of catechin and prodelphinidin B3 were strongly correlated, whereas the concentration of procyanidin B3 was not correlated with that of catechin or prodelphinidin B3. Either two different enzymes could be responsible for the dimerization of prodelphinidin B3 and procyanidin B3, or the affinity of the enzyme could be different whether the dimerization is between two catechin units or between units of gallocatechin and catechin.

Fast liquid chromatography–quadrupole-linear ion trap mass spectrometry for the analysis of pharmaceuticals and hormones in water resources

This paper presents the development of a fast multi-residue method for the determination of 49 pharmaceuticals and 6 metabolites from different therapeutic classes in water resources by means of Ultra-performance™ liquid chromatography (UPLC) coupled to tandem mass spectrometry. The use of the UPLC technology enabled all the 55 compounds to be separated chromatographically in less than 9 min (6.3 min positive mode and 2.7 min negative mode) and with a total analysis time of 18 min when considering column conditioning. Improved resolution, sensitivity and a reduction of matrix effects were obtained under these conditions. Unequivocal identification and quantification of the target compounds was also performed by using the dual acquisition modes of the hybrid triple quadrupole-linear ion trap (QqLIT) system. Triple quadrupole mode by means of selected reaction monitoring (SRM) was used for quantification, whilst a second SRM transition together with information-dependent analysis (IDA) experiments was used for confirmation. Additionally, one general, single solid-phase extraction (SPE) method was developed by using Oasis HLB cartridges. Quality parameters of the method in wastewaters were established obtaining a fast, robust, reproducible and cost-effective method for all the target pharmaceuticals. Finally, the optimized SPE-UPLC/QqLIT method was used for the analysis of the target compounds in wastewaters from Spain. Thirty-one out of fifty-five compounds were identified in the samples collected.

Simultaneous eicosanoid profiling and identification by liquid chromatography and hybrid triple quadrupole-linear ion trap mass spectrometry for metabolomic studies in human plasma / Multiparametrische Bestimmung und Identifizierung von Eikosanoiden mit Flüssigchromatographie in Kombination mit Hybrid-Quadrupol/lineare Ionenfallen Massenspektrometrie für Metabolom-Studien in Humanplasma

Abstract Eicosanoids play a key role in many physiological and pathological processes and might therefore serve as interesting diagnostic targets. Methods for the analysis of arachidonic acid metabolites in cells and body fluids require high sensitivity and specificity because of the very low concentrations, similar chemical structures and short half-lives of these metabolites. We established a mass spectrometric method for the simultaneous identification and quantification of arachidonic acid metabolites in human plasma samples using solid phase extraction followed by liquid chromatography (LC) and hybrid triple quadrupole-linear ion trap (QqLIT) mass spectrometry. Quantitative analysis was performed using the 4000 QTrap tandem mass spectrometer in multiple reaction monitoring (MRM) mode. As part of an independent data acquisition experiment MRMs were used as survey scans, which dependently triggered enhanced product ion (EPI) scans. Compound identification was carried out by library search using a library based on EPI spectra of standard components (prostaglandins, thromboxanes, leukotrienes and isoprostanes). The newly developed compound library enables the verification of known and structural elucidation of unknown eicosanoid metabolites in human plasma. In conclusion, our mass spectrometric method allows the simultaneous identification and quantification of arachidonic acid metabolites in one single LC-MS/MS run.

Development and validation of a pressurized liquid extraction liquid chromatography–tandem mass spectrometry method for perfluorinated compounds determination in fish

This paper describes the development and validation of an analytical methodology to determine eight perfluorinated compounds (PFCs) in edible fish using pressurized liquid extraction (PLE) with water and solid-phase extraction (SPE) with an ion-exchanger as extraction and pre-concentration procedures, followed by liquid chromatography–quadrupole-linear ion trap mass spectrometry (LC–QqLIT–MS). The rapidity and effectiveness of the proposed extraction procedure were compared with those most commonly used to isolate PFCs from fish (ion-pairing and alkaline digestion). The average recoveries of the different fish samples, spiked with the eight PFCs at three levels (the LOQ, 10 and 100 μg kg −1 of each PFC), were always higher than 85% with relative standard deviation (RSD) lower than 17%. A good linearity was established for the eight PFCs in the range from 0.003–0.05 to 100 μg kg −1, with r > 0.9994. The limits of quantification (LOQs) were between 0.003 and 0.05 μg kg −1, which are well below those previously reported for this type of samples. Compared with previous methods, sample preparation time and/or LOQs are reduced. The method demonstrated its successful application for the analysis of different parts of several fish species. Most of the samples tested positive, mainly for perfluoropentanoic acid (PFPA), perfluorobutane sulfonate (PFBS) and perfluorooctanoic acid (PFOA) but other of the eight studied PFCs were also present.

Rapid Detection and Characterization of in Vitro and Urinary N-Acetyl-l-cysteine Conjugates Using Quadrupole-Linear Ion Trap Mass Spectrometry and Polarity Switching

The present study describes a novel methodology for the rapid detection and structural characterization of unknown N-acetyl-L-cysteine (NAC) conjugates using polarity switching of triple quadrupole mass spectrometry. This method utilizes a negative neutral loss (NL) scan of 129 Da or multiple reaction monitoring (MRM) from predicted m/z values to product ions derived from the NL of 129 Da as a survey scan to trigger the acquisition of enhanced product ion (EPI) spectra in the positive ion mode. Thus, selective detection of NAC conjugates and acquisition of fragment-rich MS/MS spectra were accomplished in a single LC/MS run. The utility of this methodology was evaluated through analysis of NAC conjugates of acetaminophen in human urine after an oral dose. The MRM-EPI approach, which showed better sensitivity than the NL-EPI approach in analyzing urine samples, revealed three NAC-acetaminophen conjugates in the human urine, including two minor NAC conjugates that were derived from hydroxyl acetaminophen and methoxy acetaminophen. In addition, the methodology was applied to screening for reactive metabolites of clozapine and diclofenac using NAC as a trapping agent. Results showed reactive metabolite profiles comparable to those obtained from glutathione (GSH) trapping experiments, while MS/MS spectra of NAC conjugates provided more valuable structural information than those of GSH adducts. The study demonstrates that NAC trapping followed by NL-EPI analysis is a useful approach for high-throughput screening of reactive metabolites and that the MRM-EPI method is well-suited for analysis of low levels of NAC conjugates in urine.

Pharmaceutical metabolite profiling using quadrupole/ion mobility spectrometry/time‐of‐flight mass spectrometry

The use of hybrid quadrupole ion mobility spectrometry time-of-flight mass spectrometry (Q/IMS/TOFMS) in the metabolite profiling of leflunomide (LEF) and acetaminophen (APAP) is presented. The IMS drift times (T(d)) of the drugs and their metabolites were determined in the IMS/TOFMS experiments and correlated with their exact monoisotopic masses and other in silico generated structural properties, such as connolly molecular area (CMA), connolly solvent-excluded volume (CSEV), principal moments of inertia along the X, Y and Z Cartesian coordinates (MI-X, MI-Y and MI-Z), inverse mobility and collision cross-section (CCS). The correlation of T(d) with these parameters is presented and discussed. IMS/TOF tandem mass spectrometry experiments (MS(2) and MS(3)) were successfully performed on the N-acetyl-p-benzoquinoneimine glutathione (NAPQI-GSH) adduct derived from the in vitro microsomal metabolism of APAP. As comparison, similar experiments were also performed using hybrid triple quadrupole linear ion trap mass spectrometry (QTRAPMS) and quadrupole time-of-flight mass spectrometry (QTOFMS). The abilities to resolve the product ions of the metabolite within the drift tube and fragment the ion mobility resolved product ions in the transfer travelling wave-enabled stacked ring ion guide (TWIG) demonstrated the potential applicability of the Q/IMS/TOFMS technique in pharmaceutical metabolite profiling.

ω‐(Bis(trifluoromethyl)amino)alkane‐1‐sulfonates: synthesis and mass spectrometric study of the biotransformation products

In search of fluorinated functional groups which could undergo defluorination, and therefore be included in novel non-polluting fluorinated surfactants, omega-(bis(trifluoromethyl)amino)alkane-1-sulfonates (BTFMA-AS) with a homologue distribution from seven to thirteen methylene groups were synthesized and investigated for aerobic biodegradation applying both a standardized test and a fixed-bed bioreactor (FBBR). These compounds were prepared as part of a screening study for potentially mineralizable fluorinated endgroups.Application of hybrid triple quadrupole-linear ion trap mass spectrometry (QqQ(LIT)-MS) coupled to high-performance liquid chromatography (HPLC) allowed the tracking of primary degradation as well as the detection and structural elucidation of biotransformation intermediates. An understanding of the fragmentation pathway of the test compounds allowed selective precursor ion scans to reveal the presence of stable fluorinated metabolites. Structures were confirmed by enhanced product ion scans and MS(3) scans in the linear ion trap mode.The primary biodegradation rate and the extent of biodegradation were found to be chain-length dependent, with higher homologues being completely primarily degraded within 10 days. For the first time, two simultaneous metabolic pathways for substituted linear alkane-1-sulfonates were discovered: Desulfonation, oxidation to a carboxylic acid and subsequent chain-length shortening by beta-oxidation dominated the metabolism. This pathway resulted in the formation of 3-(bis(trifluoromethyl)amino)propionic acid and bis(trifluoromethyl)aminoacetic acid, which showed recalcitrance in this experiment. Oxidation of the alkyl chain to the respective carbonyl derivative represents the minor pathway. Only the long-chain homologues of these oxidized species were partially degraded; the short-chain homologues were not attacked.

Highly sensitive simultaneous determination of sulfonamide antibiotics and one metabolite in environmental waters by liquid chromatography–quadrupole linear ion trap–mass spectrometry

The present work describes the development of a highly sensitive analytical method based on liquid chromatography–quadrupole linear ion trap–mass spectrometry (LC–QqLIT–MS) for the determination of nine sulfonamide antibiotics and one N 4-acetylated metabolite in environmental waters (wastewater, surface water and groundwater) and bottled mineral water. Special emphasis was devoted to the elimination of matrix components during solid-phase extraction (SPE) by the evaluation of three different extraction/purification strategies: single cartridges (Oasis HLB and Oasis MCX) and tandem (TD) extraction (combination of both). The method developed proved to be suitable for sulfonamide determination in all kinds of waters tested. The method was shown to be linear in a wide concentration range, with correlation coefficients higher than 0.999 for all compounds except for sulfadimethoxine ( R 2 0.997). The overall instrumental repeatability was satisfactory, with the exception of the metabolite (RSD 34%). Method limits of detection achieved for sulfonamides were in the range 0.01–1.13 ng L −1 and for the metabolite 0.08–461 ng L −1. Recovery rates were estimated at 500 ng L −1 spike level in the four water matrices selected. The highest recovery achieved in all matrices was that corresponding to the Oasis HLB cartridge. In environmental waters, recovery values obtained were higher than 61% for the surface water and, in general, higher than 90% for groundwater and wastewater. Bottled mineral water exhibited recovery rates higher than 92%, with the exception of sulfamethoxypiridazine (82%) and sulfapyridine (86%) In order to demonstrate the applicability of the developed method, several water samples were analyzed. Results evidenced the requirement for consideration of N 4-acetylated metabolites of sulfonamides in environmental residue analysis to avoid the underestimation of removal rates of such pharmaceutical compounds during wastewater treatments.

Trace level determination of β-blockers in waste waters by highly selective molecularly imprinted polymers extraction followed by liquid chromatography–quadrupole-linear ion trap mass spectrometry

This paper describes the development of an analytical methodology to determine eight β-blockers in waste waters using molecularly imprinted polymers (MIPs) as extraction and pre-concentration material, followed by liquid chromatography–quadrupole-linear ion trap mass spectrometry (LC–QqLIT MS). The advantages offered by MIPs, in terms of selectivity and specificity, were compared with the most commonly polymeric materials used (the lipophilic–hydrophilic balanced Oasis ® HLB cartridges). Even though recoveries achieved with both sorbents were similar, ranging from 50 to 110% for sewage treatment plant (STP) effluent and 40–110 for STP influent, respectively, MIPs provided lower method detection limits than Oasis ® HLB, due to their specificity for target analytes and closely related analogues. Method detection limits (MDL) achieved using MIPs ranged from 0.2 to 6.4 ng/L for STP effluent and from 0.4 to 6.5 ng/L for STP influent. To highlight the advantages of MIPs against conventional polymeric cartridges, a detailed matrix effects study as well as cross reactivity tests were performed. For the latter purpose, the extraction efficiency of some pharmaceuticals and pesticides belonging to different therapeutic classes was assessed. LC–QqLIT MS, used for quantification and confirmation, proved to be a powerful analytical tool, as instrumental detection limits (IDL) achieved ranged from 0.2 to 2.7 pg injected (in multiple reaction monitoring mode (MRM)). In addition the inclusion of high sensitive MS/MS scans for each compound when working in Information Dependent Acquisition mode (IDA) provided extra confirmation for unequivocal identification of target compounds in complex environmental matrices.

Comprehensive profiling of N-acylhomoserine lactones produced by Yersinia pseudotuberculosis using liquid chromatography coupled to hybrid quadrupole–linear ion trap mass spectrometry

A method for the comprehensive profiling of the N-acylhomoserine lactone (AHL) family of bacterial quorum-sensing molecules is presented using liquid chromatography (LC) coupled to hybrid quadrupole-linear ion trap (QqQLIT) mass spectrometry. Information-dependent acquisition (IDA), using triggered combinations of triple-quadrupole and linear ion trap modes in the same LC-MS/MS run, was used to simultaneously screen, quantify and identify multiple AHLs in a single sample. This MS method uses common AHL fragment ions attributed to the homoserine moiety and the 3-oxo-, 3-hydroxy- or unsubstituted acyl side chains, to identify unknown AHLs in cell-free culture supernatants in an unbiased manner. This LC-MS technique was applied to determine the relative molar ratios of AHLs produced by Yersinia pseudotuberculosis and the consequences of inactivating by mutation either or both of the AHL synthase genes (ypsI and ytbI) on AHL profile and concentration. The Y. pseudotuberculosis wild type but not the ypsI ytbI double mutant produced at least 24 different AHLs with acyl chains ranging from C4 to C15 with or without 3-oxo or 3-hydroxy substituents. YtbI, in contrast to YpsI, could direct the synthesis of all of the AHLs identified. The most abundant and hence most biologically relevant Y. pseudotuberculosis AHLs were found to be the 3-oxo-substituted C6, C7 and C8 AHLs and the unsubstituted C6 and C8 compounds. The LC-QqQLIT methodology is broadly applicable to quorum-sensing signal molecule analysis and can provide comprehensive AHL profiles and concentrations from a single sample and simultaneously collect confirmatory spectra for each AHL identified.

Identification of phospholipid structures in human blood by direct-injection quadrupole-linear ion-trap mass spectrometry

Direct-injection electrospray ionization mass spectrometry in combination with information-dependent data acquisition (IDA), using a triple-quadrupole/linear ion trap combination, allows high-throughput qualitative analysis of complex phospholipid species from child whole blood. In the IDA experiments, scans to detect specific head groups (precursor ion or neutral loss scans) were used as survey scans to detect phospholipid classes. An enhanced resolution scan was then used to confirm the mass assignments, and the enhanced product ion scan was implemented as a dependent scan to determine the composition of each phospholipid class. These survey and dependent scans were performed sequentially and repeated for the entire duration of analysis, thus providing the maximum information from a single injection. In this way, 50 different phospholipids belonging to the phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol, phosphatidylcholine and sphingomyelin classes were identified in child whole blood.
