Polyurethanes are among the most ubiquitous materials in the biomedical and pharmaceutical areas. Thus, in this study, polycaprolactone-triol-based polyurethane conjugated ibuprofen (coded as PU-ibu) was synthesized by a two-step condensation method and characterized according to composition, morphological, thermal, and mechanical. The synthesized PU film had a smoother surface and superior mechanical properties compared to PU-ibu film.Moreover, PU was capable of swelling in PBS while PU-ibu film exhibited a surprising “de-swelling” behavior. Additionally, PU-ibu film not only had adequate wettability, excellent hemocompatibility, and sustained delivery capability but also greatly enhanced the proliferation of fibroblast cells. Therefore, the PU-ibu film provides a potential novel system for sustained drug delivery.
