A Trio of Active Zone Proteins Comprised of RIM-BPs, RIMs, and Munc13s Governs Neurotransmitter Release

At the presynaptic active zone, action-potential-triggered neurotransmitter release requires that fusion-competent synaptic vesicles are placed next to Ca2+ channels. The active zone resident proteins RIM, RBP, and Munc13 are essential contributors for vesicle priming and Ca2+-channel recruitment. Although the individual contributions of these scaffolds have been extensively studied, their respective functions in neurotransmission are still incompletely understood. Here, we analyze the functional interactions of RIMs, RBPs, and Munc13s at the genetic, molecular, functional, and ultrastructural levels in a mammalian synapse. We find that RBP, together with Munc13, promotes vesicle priming at the expense of RBP's role in recruiting presynaptic Ca2+ channels, suggesting that the support of RBP for vesicle priming and Ca2+-secretion coupling is mutually exclusive. Our results demonstrate that the functional interaction of RIM, RBP, and Munc13 is more profound than previously envisioned, acting as a functional trio that govern basic and short-term plasticity properties of neurotransmission.

Developmental Synaptic Changes at the Transient Olivocochlear-Inner Hair Cell Synapse.

In the mature mammalian cochlea, inner hair cells (IHCs) are mainly innervated by afferent fibers that convey sound information to the CNS. During postnatal development, however, medial olivocochlear (MOC) efferent fibers transiently innervate the IHCs. The MOC-IHC synapse, functional from postnatal day 0 (P0) to hearing onset (P12), undergoes dramatic changes in the sensitivity to acetylcholine (ACh) and in the expression of key postsynaptic proteins. To evaluate whether there are associated changes in the properties of ACh release during this period, we used a cochlear preparation from mice of either sex at P4, P6-P7, and P9-P11 and monitored transmitter release from MOC terminals in voltage-clamped IHCs in the whole-cell configuration. The quantum content increased 5.6× from P4 to P9-P11 due to increases in the size and replenishment rate of the readily releasable pool of synaptic vesicles without changes in their probability of release or quantum size. This strengthening in transmission was accompanied by changes in short-term plasticity properties, which switched from facilitation at P4 to depression at P9-P11. We have previously shown that at P9-P11, ACh release is supported by P/Q- and N-type voltage-gated calcium channels (VGCCs) and negatively regulated by BK potassium channels activated by Ca2+ influx through L-type VGCCs. We now show that at P4 and P6-P7, release is mediated by P/Q-, R- and L-type VGCCs. Interestingly, L-type VGCCs have a dual role: they both support release and fuel BK channels, suggesting that at immature stages presynaptic proteins involved in release are less compartmentalized.SIGNIFICANCE STATEMENT During postnatal development before the onset of hearing, cochlear inner hair cells (IHCs) present spontaneous Ca2+ action potentials that release glutamate at the first auditory synapse in the absence of sound stimulation. The IHC Ca2+ action potential frequency pattern, which is crucial for the correct establishment and function of the auditory system, is regulated by the efferent medial olivocochlear (MOC) system that transiently innervates IHCs during this period. We show here that developmental changes in synaptic strength and synaptic plasticity properties at the MOC-IHC synapse upon MOC fiber activation at different frequencies might be crucial for tightly shaping the pattern of afferent activity during this critical period.

Frequency-Dependent Gating of Hippocampal-Neocortical Interactions.

How and where hippocampal-neocortical interactions required for memory formation take place is a major issue of current research. Using a combined in vivo functional magnetic resonance imaging/electrophysiology approach, we have investigated whether specific frequencies of CA3 neuronal activation, inducing different forms of short-term plasticity at CA1 synapses, contribute to differential activity propagation in brain-wide networks connected to the hippocampus. We report that localized activation of CA3 neurons in dorsal hippocampus produced activity propagation within the hippocampal formation, including the subiculum and entorhinal cortex, which increased monotonically with frequency to a maximum at 20-40 Hz. However, robust extrahippocampal propagation was seen specifically at theta-beta frequencies (10-20 Hz), reaching a network of midline neocortical and mesolimbic structures. Activation in those regions correlated with a frequency-dependent facilitation of spiking activity recorded in CA1. These results provide a mechanistic link between the dynamic properties of short-term plasticity in the efferent synapses of CA3 neurons in CA1 and activity propagation in brain-wide networks, and identify polysynaptic information channels segregated in the frequency domain.

Cortical Balance of Excitation and Inhibition Is Regulated by the Rate of Synaptic Activity

Cortical activity is determined by the balance between excitation and inhibition. To examine how shifts in brain activity affect this balance, we recorded spontaneous excitatory and inhibitory synaptic inputs into layer 4 neurons from rat somatosensory cortex while altering the depth of anesthesia. The rate of excitatory and inhibitory events was reduced by ∼50% when anesthesia was deepened. However, whereas both the amplitude and width of inhibitory synaptic events profoundly increased under deep anesthesia, those of excitatory events were unaffected. These effects were found using three different types of anesthetics, suggesting that they are caused by the network state and not by local specific action of the anesthetics. To test our hypothesis that the size of inhibitory events increased because of the decreased rate of synaptic activity under deep anesthesia, we blocked cortical excitation and replayed the slow and fast patterns of inhibitory inputs using intracortical electrical stimulation. Evoked inhibition was larger under low-frequency stimulation, and, importantly, this change occurred regardless of the depth of anesthesia. Hence, shifts in the balance between excitation and inhibition across distinct states of cortical activity can be explained by the rate of inhibitory inputs combined with their short-term plasticity properties, regardless of the actual global brain activity.

A cortical attractor network with Martinotti cells driven by facilitating synapses.

The population of pyramidal cells significantly outnumbers the inhibitory interneurons in the neocortex, while at the same time the diversity of interneuron types is much more pronounced. One acknowledged key role of inhibition is to control the rate and patterning of pyramidal cell firing via negative feedback, but most likely the diversity of inhibitory pathways is matched by a corresponding diversity of functional roles. An important distinguishing feature of cortical interneurons is the variability of the short-term plasticity properties of synapses received from pyramidal cells. The Martinotti cell type has recently come under scrutiny due to the distinctly facilitating nature of the synapses they receive from pyramidal cells. This distinguishes these neurons from basket cells and other inhibitory interneurons typically targeted by depressing synapses. A key aspect of the work reported here has been to pinpoint the role of this variability. We first set out to reproduce quantitatively based on in vitro data the di-synaptic inhibitory microcircuit connecting two pyramidal cells via one or a few Martinotti cells. In a second step, we embedded this microcircuit in a previously developed attractor memory network model of neocortical layers 2/3. This model network demonstrated that basket cells with their characteristic depressing synapses are the first to discharge when the network enters an attractor state and that Martinotti cells respond with a delay, thereby shifting the excitation-inhibition balance and acting to terminate the attractor state. A parameter sensitivity analysis suggested that Martinotti cells might, in fact, play a dominant role in setting the attractor dwell time and thus cortical speed of processing, with cellular adaptation and synaptic depression having a less prominent role than previously thought.

Interactions between short-term and long-term plasticity: shooting for a moving target

AbstractFar from being static transmission units, synapses are highly dynamical elements that change over multiple time scales depending on the history of the neural activity of both the pre- and postsynaptic neuron. Moreover, synaptic changes on different time scales interact: long-term plasticity (LTP) can modify the properties of short-term plasticity (STP) in the same synapse. Most existing theories of synaptic plasticity focus on only one of these time scales (either STP or LTP or late-LTP) and the theoretical principles underlying their interactions are thus largely unknown. Here we develop a normative model of synaptic plasticity that combines both STP and LTP and predicts specific patterns for their interactions. Recently, it has been proposed that STP arranges for the local postsynaptic membrane potential at a synapse to behave as an optimal estimator of the presynaptic membrane potential based on the incoming spikes. Here we generalize this approach by considering an optimal estimator of a non-linear function of the membrane potential and the long-term synaptic efficacy -- which itself may be subject to change on a slower time scale. We find that an increase in the long-term synaptic efficacy necessitates changes in the dynamics of STP. More precisely, for a realistic non-linear function to be estimated, our model predicts that after the induction of LTP, causing long-term synaptic efficacy to increase, a depressing synapse should become even more depressing. That is, in a protocol using trains of presynaptic stimuli, as the initial EPSP becomes stronger due to LTP, subsequent EPSPs should become weakened and this weakening should be more pronounced with LTP. This form of redistribution of synaptic efficacies agrees well with electrophysiological data on synapses connecting layer 5 pyramidal neurons.

Monitoring synaptic transmission in primary neuronal cultures using local extracellular stimulation

Various techniques have been applied for the functional analysis of synaptic transmission in cultured neurons. Here, we describe a method of studying synaptic transmission in neurons cultured at high-density from different brain regions such as the cortex, striatum and spinal cord. We use postsynaptic whole-cell recordings to monitor synaptic currents triggered by presynaptic action potentials that are induced by brief stimulations with a nearby extracellular bipolar electrode. Pharmacologically isolated excitatory or inhibitory postsynaptic currents can be reliably induced, with amplitudes, synaptic charge transfers, and short-term plasticity properties that are reproducible from culture to culture. We show that the size and kinetics of pharmacologically isolated inhibitory postsynaptic currents triggered by single action potentials or stimulus trains depend on the Ca 2+ concentration, temperature and stimulation frequency. This method can be applied to study synaptic transmission in wildtype neurons infected with lentiviruses encoding various components of presynaptic release machinery, or in neurons from genetically modified mice, for example neurons carrying floxed genes in which gene expression can be acutely ablated by expression of Cre recombinase. The preparation described in this paper should be useful for analysis of synaptic transmission in inter-neuronal synapses formed by different types of neurons.

Temperature-Dependent Shift of Balance among the Components of Short-Term Plasticity in Hippocampal Synapses

Studies of short-term plasticity (STP) in the hippocampus, performed mostly at room temperature, have shown that small central synapses rapidly depress in response to high-frequency stimulation. This decrease in synaptic strength with synapse use places constraints on the use of STP as a dynamic filter for processing of natural high-frequency input. Here we report that, because of a strong but differential temperature dependence of STP components, the properties of STP in excitatory hippocampal synapses change dramatically with temperature. By separating the contributions of various STP processes during spike trains at different temperatures, we found a shift from dominating depression at 23 degrees C to prevailing facilitation and augmentation at 33-38 degrees C. This shift of balance among STP components resulted from a large increase in amplitudes of facilitation and augmentation (Q10 approximately 2.6 and approximately 5.1, respectively) and little change in the amplitude of depression (Q10 approximately 1.1) with temperature. These changes were accompanied by the accelerated decay of all three processes (Q10 = 3.2, 6.6, and 2.1, respectively). The balance of STP components achieved at higher temperatures greatly improved the maintenance of synaptic strength during prolonged synaptic use and had a strong effect on the processing of natural spike trains: a variable mixture of facilitated and depressed responses at 23 degrees C changed into a significantly more reproducible and depression-free filtering pattern at 33-38 degrees C. This filtering pattern was highly conserved among cells, slices, and animals, and under various physiological conditions, arguing for its physiological significance. Therefore, the fine balance among STP components, achieved only at near body temperatures, is required for the robust function of STP as a dynamic filter during natural stimulation.

Long- and short-term plasticity at mossy fiber synapses on mossy cells in the rat dentate gyrus

Mossy cells give rise to the commissural and associational pathway of the dentate gyrus, and receive their major excitatory inputs from the mossy fibers of granule cells. Through these feed-back excitatory connections, mossy cells have been suggested to play important roles in both normal signal processing in learning and memory, as well as in seizure propagation. However, the nature of the activity-dependent modifications of the mossy fiber inputs to mossy hilar cells is not well understood. We studied the long- and short-term plasticity properties of the mossy fiber-mossy cell synapse, using the minimal stimulation technique in slices in whole cell recorded mossy cells retrogradely prelabeled with the fluorescent dye DiO from the contralateral dentate gyrus. Following tetanic stimulation, mossy fiber synapses showed significant NMDA receptor-independent long-term potentiation (LTP), associated with increased excitatory postsynaptic currents (EPSC) amplitude and decreased failure rates. Coefficient of variance and failure rate analyses suggested a presynaptic locus of LTP induction. Mossy fiber synapses on mossy cells also showed activity-dependent short-term modification properties, including both frequency-dependent facilitation (stimuli at higher frequencies evoked larger EPSCs with lower failure rates) and burst facilitation (each EPSC in a burst had a larger amplitude and higher probability of occurrence than the preceding EPSCs within the burst). The data show that mossy fiber-mossy cell synapses exhibit both long- and short-term plasticity phenomena that are generally similar to the mossy fiber synapses on CA3 pyramidal cells.
