Tissue Engineering Material KLD-12 Polypeptide /TGF-β1 the Protective Effect and Mechanism of Nanofiber Gel on Early Intervertebral Disc Degeneration

Intervertebral disc degeneration (IDD) is a common clinical symptom of multifactorial disease. The treatment and expenditure of IDD cause huge economic and psychological harm to patients, and there is no root treatment in the clinic. However, the appearance of tissue engineering materials provides a new idea for the treatment of early IDD. KLD-12 polypeptide material is a new kind of polypeptide scaffold material, which can be used to repair early IDD and TGF-β1Transforming growth factor-1 plays an important role in the proliferation of Intervertebral disc cells and inhibition of inflammatory response. In order to further understand the tissue engineering material kld-12 polypeptide / TGF-β1 the biomechanical properties of nanofiber gel, and to clarify tissue engineering material KLD-12 polypeptide TGF-β1nanofiber gel provides an experimental basis for the protection and mechanism of early IDD. In this paper, tissue engineering material KLD-12 polypeptide /TGF-β1 is mainly studied as the protective effect and mechanism of nanofiber gel on early IDD. In this paper, through the study of the tissue structure of the intervertebral disc, the composition of the nucleus pulposus, annulus fibrosus and cartilage endplate was studied. The objective was to study the relationship between transforming growth factor TGF-β1 and IDD and to understand its important role in the proliferation of intervertebral disc cells and inhibition of inflammatory response. In this paper, we studied the molecular basis of IDD, the main reason is the imbalance of extracellular matrix synthesis and degradation of Intervertebral disc cells, to understand the structural characteristics of cartilage endplate and the composition of Intervertebral disc fibroblasts. In this study, we studied the cell proliferation activity, the ratio of surviving dead cells, the content of glucosaminoglycans, the content of polyproteoglycan and type II collagen in the gel, and studied the protective effect and mechanism of tissue engineering material KLD-12 polypeptide /TGF-β1 nanofiber gel on early IDD. The results showed that kld-12 polypeptide / TGF-β1 was more effective in the proliferation activity of annulus fibrosus cells of nanofiber gel is higher than that of KLD-12 polypeptide/annulus fibroin nanofiber gel. On 2d, the difference in cell proliferation activity was not obvious, KLD-12 polypeptide / TGF- β 1 the fibrous annulus cell proliferation activity of nanofiber gel was 0.796, and the proliferation activity of KLD-12 polypeptide/annulus fibroin nanofiber gel was 0.786. On the 14d, KLD-12 polypeptide / TGF- β 1 the fibrous annulus cell proliferation activity of nanofiber gel was 1.204, and the proliferation activity of KLD-12 polypeptide/annulus fibroin nanofiber gel was 1.034.

The Effect of Timing of rhBMP-2 Injection on Intervertebral Disc Degeneration in a Rat Tail Model

In vitro studies have demonstrated that Bone Morphogenetic Protein-2 (BMP-2) stimulates the proliferation of intervertebral disc cells and promotes the secretion of extracellular matrix components. We have previously demonstrated that the rat tail intervertebral disc degenerates in a time dependent manner after injury. Few studies have demonstrated the ability of BMP-2 to improve disc degeneration in vivo, or investigated the extent to which the disc can be degraded and still be partly or completely recovered.

Effects of Chinese herbal medicine Yiqi Huayu Bushen Recipe on expressions of aggrecan and type Ⅹ collagen mRNAs in cells from degenerated human intervertebral discs

To investigate the effects of serum containing Yiqi Huayu Bushen Recipe, a compound Chinese herbal medicine, on expressions of aggrecan and type X collagen mRNAs of the degenerated intervertebral disc cells in patients with cervical spondylotic myelopathy. Six inpatients diagnosed as cervical spondylotic myelopathy and undergoing cervical decompression and fusion surgery to remove the degenerated disc tissue were enrolled in this study. Primary intervertebral disc cells were isolated using small tissue mass method. Serum containing Yiqi Huayu Bushen recipe were obtained after SD rats were fed with Yiqi Huayu Bushen recipe for 3 days. MTT method was used to detect proliferation of the first-passage cells isolated from degenerated intervertebral discs and growth curve was drawn. In the following tests, the first-passage cells were cultured with the sera containing drugs and sodium chloride (NaCl), respectively. Proliferation of intervertebral disc cells was detected by MTT method and expressions of aggrecan and type X collagen mRNAs were detected by real-time polymerase chain reaction. The growth curve of the first-passage cells of the degenerated intervertebral discs showed that the time of cell proliferation peak was the sixth day. Compared with the same concentrations of NaCl solution, serum containing Yiqi Huayu Bushen Recipe at 5%, 10% and 20% promoted cell proliferation and expression of aggrecan mRNA and decreased expression of type X collagen mRNA (P<0.01). 10% serum containing Yiqi Huayu Bushen recipe was more effective than 5% and 20%. Yiqi Huayu Bushen Recipe inhibits degeneration of the intervertebral disc cells by regulating collagen and proteoglycan expressions.

PDGF, bFGF and IGF-I stimulate the proliferation of intervertebral disc cells in vitro via the activation of the ERK and Akt signaling pathways

Intervertebral disc (IVD) degeneration is frequently characterized by increased cell proliferation, probably as a tissue regenerative response. Although many growth factors and their receptors have been shown to be expressed normally in the disc, and generally to be over-expressed during degeneration, not all of them have been thoroughly studied concerning their effects on IVD cell proliferation. In the present report, three potent mitogens, platelet-derived growth factor (PDGF), basic fibroblast growth factor (bFGF) and insulin-like growth factor-I (IGF-I) are examined regarding their capacity to induce proliferation in vitro of bovine coccygeal nucleus pulposus (NP) and annulus fibrosus (AF) cells, as well as to activate major intracellular signal transduction pathways. PDGF, bFGF and IGF-I were found to induce DNA synthesis in quiescent IVD cells in a dose-dependent manner. Maximum stimulation was induced by PDGF, while stimulation by all three factors simultaneously exceeded only slightly that caused by PDGF alone. All three growth factors were shown to phosphorylate immediately extracellular-signal regulated kinases (ERKs), while the stimulation by bFGF especially resulted in sustained ERK phosphorylation. Furthermore, all three growth factors induced phosphorylation of Akt in both Thr308 and Ser473 residues immediately after stimulation, although bFGF-induced phosphorylation was much weaker than that provoked by PDGF and IGF-I. In addition, the MEK inhibitor PD98059 and the PI 3-K inhibitor wortmannin were shown to block growth factor-induced ERK- and Akt-phosphorylation, respectively, in IVD cells. Inhibition of the MEK/ERK or the PI 3-K/Akt pathways provoked a significant decline of the proliferative effects of PDGF, bFGF or IGF-I on IVD cell cultures, while the simultaneous inhibition of both signaling pathways abolished completely the mitogenicity of these growth factors. The above effects of the three growth factors were reproduced similarly in both NP and AF cell cultures. Overall, the above results indicate that PDGF, bFGF and IGF-I stimulate the proliferation of IVD cells via the ERK and Akt signaling pathways.

Platelet-Rich Plasma (PRP) Stimulates the Extracellular Matrix Metabolism of Porcine Nucleus Pulposus and Anulus Fibrosus Cells Cultured in Alginate Beads

In vitro assessment of the effects of platelet-rich plasma on the extracellular matrix metabolism of porcine intervertebral disc cells. To determine whether platelet-rich plasma is effective in stimulating cell proliferation and extracellular matrix metabolism by porcine disc cells cultured in alginate beads. Platelet-rich plasma is used to accelerate wound healing and tissue regeneration. Activated platelets release multiple growth factors that regulate cell proliferation, differentiation, and morphogenesis. Individual growth factors present in platelet-rich plasma have been demonstrated to affect the metabolism of intervertebral disc cells. Platelet-poor and platelet-rich plasma was isolated from fresh porcine blood using a commercially available platelet concentration system. After preculture for 7 days and serum starvation for 24 hours, the beads containing nucleus pulposus and anulus fibrosus cells were then cultured for another 72 hours in serum-free medium, 10% fetal bovine serum, 10% platelet-poor plasma, or 10% platelet-rich plasma. The synthesis of proteoglycans and collagen, the accumulation of proteoglycans, and the DNA content were biochemically assessed. Platelet-rich plasma had a mild stimulatory effect on cell proliferation of intervertebral disc cells. Platelet-rich plasma treatment significantly upregulated proteoglycan and collagen synthesis and proteoglycan accumulation when compared with platelet-poor plasma. Platelet-rich plasma was effective in stimulating cell proliferation and extracellular matrix metabolism. The response to platelet-rich plasma was greater in the case of anulus fibrosus cells than of nucleuspulposus cells. The local administration of platelet-rich plasma might stimulate intervertebral disc repair. In addition, given the risks of using animal serum for tissue engineering, autologous blood may gain favor as a source of growth factors and serum supplements needed for stimulating cells to engineer intervertebral disc tissues.
