Abstract Study question Can leukocyte telomere length be a marker of ovarian decline? Summary answer The significant correlation found between leukocyte and cumulus cell telomere lengths supports the use of leukocyte telomere length (TL) as a marker of ovarian aging. What is known already The telomere-based theory of reproductive aging proposes that age-related ovarian dysfunction is a consequence of progressive telomere shortening from fetal oogenesis to the adult ovary, compromising genome stability and proper chromosome segregation. Telomere length in cumulus cells (CCTL) is known to be a biomarker of oocyte and embryo quality, but its assessment is not practical. Hence, we previously proposed leukocyte telomere length (LTL) as an alternative to directly measuring telomere length in follicular cells, but so far it has only been assessed in samples of moderate size and homogeneous populations without consistent results. Study design, size, duration This prospective, non-interventional cohort study involved 300 women aged 18 to 46 years undergoing controlled ovarian stimulation over a 24-month period. The cohort included 169 egg donors and 131 IVF patients. Oocyte retrieval was performed 36 hours after maturation induction, with cumulus cells collected during oocyte stripping and blood samples taken via peripheral venous access. Participants/materials, setting, methods Genomic DNA was isolated from leukocytes and cumulus cells of all subjects for TL measurements. Relative TL was determined using a SYBR-Green quantitative real-time PCR protocol. A Taqman assay for the multicopy gene Alu was used to normalize the measurements. A Pearson’s correlation test was used to analyze the correlation between CCTL and LTL, as well as between TL and age, antral follicle count, Anti-Müllerian hormone, and number of mature oocytes retrieved. Main results and the role of chance Mean age of the subjects was 30.70±7.61 years. Anti-Müllerian hormone (AMH) mean level was 2.60±1.87 ng/ml, antral follicle count (AFC) was 17.8±7.4 and the number of mature oocytes was 14.3±9.3. A significant correlation was observed between CCTL and LTL (R = 0.6, p-value=2.2E-16). Notably, the relationship between leukocyte and cumulus cell telomere lengths was stronger in the 97 participants over 35 years (R = 0.67, p-value=1.55E-13), especially those between 39 to 46 years (R = 0.70, p-value=1.19E-12). Telomere length was inversely correlated with age in both leukocytes and cumulus cells (R=-0.23, p-value=6.98E-05; R=-0.4, p-value=2.38E-05), meaning the older the subject, the shorter the telomeres in both cells. Furthermore, longer telomeres in leukocytes and cumulus cells were significantly correlated with higher AFC (R = 0.20, p-value=0.0006; R = 0.25, p-value=1.83E-05), and more mature oocytes retrieved (R = 0.16, p-value=0.0043; R = 0.27, p-value=2.83E-06). However, we did not find any association between AMH levels and LTL or CCTL (R = 0.07, p-value=0.2666; R = 0.07, p-value=0.2747). Limitations, reasons for caution The method used to measure the average telomere length from pooled leukocyte samples may not reflect individual cell variability. Additionally, it allows for relative but not absolute telomere length comparisons. Wider implications of the findings This study is the first to demonstrate in a large and heterogeneous population that LTL could be a viable marker for ovarian aging by reflecting follicular cell telomere length. This finding may provide a valuable tool for assessing and monitoring the reproductive health of IVF patients. Trial registration number Not applicable

Cellular senescence, whereby cells cease to proliferate, is known to contribute to the aging process and age-related pathologies. It is elicited either by cell-intrinsic mechanisms such as progressive telomere shortening or due to the extrinsic stress-related factors, which via p53-p21 and p16-pRB tumor suppressor pathways signal cells to cease proliferation. A proper identification and characterization of senescent cells is necessary to understand the process of aging, age-related pathologies, and the development of therapeutics to treat age-related dysfunctions. The landmark discovery of Senescence-Associated-Beta-Galactosidase (SA-β-Gal) marker, and a simple colorimetric method to detect SA-β-Gal greatly facilitated identification of the senescent cells in human and rodent cells pertaining to age-related diseases (Dimri et al., 1995). Despite the availability of additional senescence biomarkers, the SA-β-Gal marker and histochemical detection method remain the most widely used tool to identify senescent cells in vitro and in vivo. Here, we revisit the original colorimetric method to detect senescent cells that was first published in 1995 (Dimri et al., 1995).

Influence of EE2 exposure, age and sex on telomere length in European long-snouted seahorse (Hippocampus guttulatus)

After a Telomere Lengthening in juvenile stage, a progressive telomere shortening occurs with age despite higher telomerase level. Telomere Length (TL) may also reflect past physiological state such as a chronic chemical stress. Several studies have revealed a correlation between TL, ageing and/or sex in vertebrates, including teleosts; however, the patterns of telomere dynamics with telomerase mRNA expression, sex, lifespan or chemical stress in teleosts are unclear. The first aim of this study is to verify if telomere length is age and sex-dependent. The second aim is to consider if TL is a useful indicator of stress response in European long-snouted seahorse, Hippocampus guttulatus, an ectothermic and non-model system. We showed that after telomere lengthening during the juvenile stage, a telomeric attrition became significant in sexually mature individuals (p = 0.042). TL decreased in older seahorses despite the presence of somatic telomerase mRNA expression at all life stages studied. There was no difference in TL between males and females, but telomerase mRNA expression was consistently higher in females than males. Exposure to EE2 had no effect on TL in young seahorses, but was correlated with a significant increase in telomerase mRNA expression and various physiological disruptions. Here, a growth retardation of −10 % for body length (p = 0.016) and approximately −45 % for mass (p = 0.001) compared to healthy juvenile seahorses was observed. Our data suggest that telomere dynamics alone should not be used as a marker of EE2 exposure in juvenile seahorses.

Telomere maintenance in African trypanosomes

Telomere maintenance is essential for genome integrity and chromosome stability in eukaryotic cells harboring linear chromosomes, as telomere forms a specialized structure to mask the natural chromosome ends from DNA damage repair machineries and to prevent nucleolytic degradation of the telomeric DNA. In Trypanosoma brucei and several other microbial pathogens, virulence genes involved in antigenic variation, a key pathogenesis mechanism essential for host immune evasion and long-term infections, are located at subtelomeres, and expression and switching of these major surface antigens are regulated by telomere proteins and the telomere structure. Therefore, understanding telomere maintenance mechanisms and how these pathogens achieve a balance between stability and plasticity at telomere/subtelomere will help develop better means to eradicate human diseases caused by these pathogens. Telomere replication faces several challenges, and the “end replication problem” is a key obstacle that can cause progressive telomere shortening in proliferating cells. To overcome this challenge, most eukaryotes use telomerase to extend the G-rich telomere strand. In addition, a number of telomere proteins use sophisticated mechanisms to coordinate the telomerase-mediated de novo telomere G-strand synthesis and the telomere C-strand fill-in, which has been extensively studied in mammalian cells. However, we recently discovered that trypanosomes lack many telomere proteins identified in its mammalian host that are critical for telomere end processing. Rather, T. brucei uses a unique DNA polymerase, PolIE that belongs to the DNA polymerase A family (E. coli DNA PolI family), to coordinate the telomere G- and C-strand syntheses. In this review, I will first briefly summarize current understanding of telomere end processing in mammals. Subsequently, I will describe PolIE-mediated coordination of telomere G- and C-strand synthesis in T. brucei and implication of this recent discovery.

Dyskeratosis Congenita: A Case Report

Dyskeratosis Congenita (DC) is a rare inherited bone marrow failure disorder characterized by lacy reticulated skin pigmentation, nail dystrophy and mucosal leukoplakia. In DC bone marrow failure is the main cause of premature death with an additional predisposition to malignancy. DC results from progressive shortening of telomeres resulting in DNA replication. X-linked recessive is the main mode of inheritance. Most of the patients respond to Androgen therapy. This is a case report of a 5-year-old boy presented with features of bone marrow failure and had abnormality of skin, oral mucosa and nail. His complete blood count with peripheral blood film showed pancytopenia and no blast cell. Hypocellular marrow was present in his Bone marrow examination. Chromosomal analysis- fluorescence in situ hybridization (FISH) revealed very short telomere of chromosome. Microarray genotyping showed DKC1 gene abnormality. Intrinsic and environmental basis of aging: A narrative review

Longevity has been a topic of interest since the beginnings of humanity, yet its aetiology and precise mechanisms remain to be elucidated. Aging is currently viewed as a physiological phenomenon characterized by the gradual degeneration of organic physiology and morphology due to the passage of time where both external and internal stimuli intervene. The influence of intrinsic factors, such as progressive telomere shortening, genome instability due to mutation buildup, the direct or indirect actions of age-related genes, and marked changes in epigenetic, metabolic, and mitochondrial patterns constitute a big part of its underlying endogenous mechanisms. On the other hand, several psychosocial and demographic factors, such as diet, physical activity, smoking, and drinking habits, may have an even more significant impact on shaping the aging process. Consequentially, implementing dietary and exercise patterns has been proposed as the most viable alternative strategy for attenuating the most typical degenerative aging changes, thus increasing the likelihood of prolonging lifespan and achieving successful aging.

Deletion of MEC1 suppresses the replicative senescence of the cdc13-2 mutant in Saccharomyces cerevisiae.

In Saccharomyces cerevisiae, telomerase recruitment to telomeres depends on a direct interaction between Cdc13, a protein that binds single-stranded telomeric DNA, and the Est1 subunit of telomerase. The cdc13-2 allele disrupts telomerase association with telomeres, resulting in progressive telomere shortening and replicative senescence. The Mec1/ATR kinase is both a positive and a negative regulator of telomerase activity and is required for the cell cycle arrest in telomerase-deficient senescent cells. In this study, we find that the deletion of MEC1 suppresses the replicative senescence of cdc13-2. This suppression is dependent on telomerase, indicating that Mec1 antagonizes telomerase-mediated telomere extension in cdc13-2 cells to promote senescence.

A subset of gut leukocytes has telomerase-dependent “hyper-long” telomeres and require telomerase for function in zebrafish

BackgroundTelomerase, the enzyme capable of elongating telomeres, is usually restricted in human somatic cells, which contributes to progressive telomere shortening with cell-division and ageing. T and B-cells cells are somatic cells that can break this rule and can modulate telomerase expression in a homeostatic manner. Whereas it seems intuitive that an immune cell type that depends on regular proliferation outbursts for function may have evolved to modulate telomerase expression it is less obvious why others may also do so, as has been suggested for macrophages and neutrophils in some chronic inflammation disease settings. The gut has been highlighted as a key modulator of systemic ageing and is a key tissue where inflammation must be carefully controlled to prevent dysfunction. How telomerase may play a role in innate immune subtypes in the context of natural ageing in the gut, however, remains to be determined.ResultsUsing the zebrafish model, we show that subsets of gut immune cells have telomerase-dependent”hyper-long” telomeres, which we identified as being predominantly macrophages and dendritics (mpeg1.1+ and cd45+mhcII+). Notably, mpeg1.1+ macrophages have much longer telomeres in the gut than in their haematopoietic tissue of origin, suggesting that there is modulation of telomerase in these cells, in the gut. Moreover, we show that a subset of gut mpeg1.1+ cells express telomerase (tert) in young WT zebrafish, but that the relative proportion of these cells decreases with ageing. Importantly, this is accompanied by telomere shortening and DNA damage responses with ageing and a telomerase-dependent decrease in expression of autophagy and immune activation markers. Finally, these telomerase-dependent molecular alterations are accompanied by impaired phagocytosis of E. coli and increased gut permeability in vivo.ConclusionsOur data show that limiting levels of telomerase lead to alterations in gut immunity, impacting on the ability to clear pathogens in vivo. These are accompanied by increased gut permeability, which, together, are likely contributors to local and systemic tissue degeneration and increased susceptibility to infection with ageing.Graphical

Mitochondrial dysfunction caused by targeted deletion of Mfn1 does not result in telomere shortening in oocytes.

Telomere shortening during oocyte growth and development is related to reproductive ageing and infertility. The main mechanism involved in the maintenance of telomeres is based on telomerase activity, a specialized enzyme complex, which is capable of adding TTAGGG repeats at the ends of the chromosomes. Mitochondrial dysfunction may cause progressive shortening of telomeres by promoting the generation of reactive oxygen species. Mitofusin-1 is a protein required for mitochondrial fusion. Mice with the mitofusin-1 (Mfn1) deletion in the oocyte are characterized by accelerated follicular depletion and infertility, associated with defective oocyte maturation and follicular development. We hypothesized whether mitochondrial dysfunction in oocytes with targeted deletion of Mfn1 causes telomere shortening. We analyzed telomere length in oocyte and somatic cells in 3-, 6- and 9-month-old Mfn1-/- and wild-type mice. Immunofluorescence in oocyte mice of TRF1 and H2A.X was assessed to evaluate the interplay between the end-protection functions and the response to DNA damage occurring inside the telomeric repeats. Mitochondrial dysfunction due to the deletion of Mfn1 does not seem to affect telomere length in mouse oocytes.

Telomerase gene therapy: a remission toward cancer.

Telomerase is an enzyme which is culpable for the aliment and stability of telomeres. It also maintains the genomic integrity and chromosomal stability. The progressive shortening of telomeres may cause chromosomal instability and alternation in the telomerase. It may cause telomere attrition which can lead to oncogenic incidence in human. Cancer is a disease which is induced by genetic alternations in genes. The genetic mutation within the hTERT is a common type of scenario which is generally found above 90% of cancer. In cancer, the length of telomere and the activity of telomerase are very important for cancer cells to proliferate and also for the survival of tumors. Cancer cells regulate through several pathways to increase telomerase activity. There have been several advancements developed to inhibit the telomerase activity in cancer cell but the repercussion of those have demonstrated many adverse effects. Research on AAVs-mediated telomerase gene therapy has demonstrated prominent outcomes in animal trials. Thus, it has the potential to bring significant shine in the telomerase cancer therapeutics. Here, in this review article, we have analyzed studies related to telomerase gene therapeutics to cure cancer. We also have summarized the telomerase function and mechanism of action to cause cancer. Moreover, other current development in the clinical advances of telomerase inhibition in cancer is described.

An inducible CRISPR/Cas9 screen identifies DTX2 as a transcriptional regulator of human telomerase

SummaryMost tumor cells reactivate telomerase to ensure unlimited proliferation, whereas the expression of human telomerase reverse transcriptase (hTERT) is tightly regulated and rate-limiting for telomerase activity maintenance. Several general transcription factors (TFs) have been found in regulating hTERT transcription; however, a systematic study is lacking. Here we performed an inducible CRISPR/Cas9 KO screen using an hTERT core promoter-driven reporter. We identified numerous positive regulators including an E3 ligase DTX2. In telomerase-positive cancer cells, DTX2 depletion downregulated hTERT transcription and telomerase activity, contributing to progressive telomere shortening, growth arrest, and increased apoptosis. Utilizing BioID, we characterized multiple TFs as DTX2 proximal proteins, among which NFIC functioned corporately with DTX2 in promoting hTERT transcription. Further analysis demonstrated that DTX2 mediated K63-linked ubiquitination of NFIC, which facilitated NFIC binding to the hTERT promoter and enhanced hTERT expression. These findings highlight a new hTERT regulatory pathway that may be exploited for potential cancer therapeutics.

Telomere Shortening and Its Association with Cell Dysfunction in Lung Diseases.

Telomeres are localized at the end of chromosomes to provide genome stability; however, the telomere length tends to be shortened with each cell division inducing a progressive telomere shortening (TS). In addition to age, other factors, such as exposure to pollutants, diet, stress, and disruptions in the shelterin protein complex or genes associated with telomerase induce TS. This phenomenon favors cellular senescence and genotoxic stress, which increases the risk of the development and progression of lung diseases such as idiopathic pulmonary fibrosis, chronic obstructive pulmonary disease, SARS-CoV-2 infection, and lung cancer. In an infectious environment, immune cells that exhibit TS are associated with severe lymphopenia and death, whereas in a noninfectious context, naïve T cells that exhibit TS are related to cancer progression and enhanced inflammatory processes. In this review, we discuss how TS modifies the function of the immune system cells, making them inefficient in maintaining homeostasis in the lung. Finally, we discuss the advances in drug and gene therapy for lung diseases where TS could be used as a target for future treatments.

Roles for the 8-Oxoguanine DNA Repair System in Protecting Telomeres From Oxidative Stress.

Telomeres are protective nucleoprotein structures that cap linear chromosome ends and safeguard genome stability. Progressive telomere shortening at each somatic cell division eventually leads to critically short and dysfunctional telomeres, which can contribute to either cellular senescence and aging, or tumorigenesis. Human reproductive cells, some stem cells, and most cancer cells, express the enzyme telomerase to restore telomeric DNA. Numerous studies have shown that oxidative stress caused by excess reactive oxygen species is associated with accelerated telomere shortening and dysfunction. Telomeric repeat sequences are remarkably susceptible to oxidative damage and are preferred sites for the production of the mutagenic base lesion 8-oxoguanine, which can alter telomere length homeostasis and integrity. Therefore, knowledge of the repair pathways involved in the processing of 8-oxoguanine at telomeres is important for advancing understanding of the pathogenesis of degenerative diseases and cancer associated with telomere instability. The highly conserved guanine oxidation (GO) system involves three specialized enzymes that initiate distinct pathways to specifically mitigate the adverse effects of 8-oxoguanine. Here we introduce the GO system and review the studies focused on investigating how telomeric 8-oxoguanine processing affects telomere integrity and overall genome stability. We also discuss newly developed technologies that target oxidative damage selectively to telomeres to investigate roles for the GO system in telomere stability.

Telomeres and Age-Related Diseases.

Telomeres are at the non-coding ends of linear chromosomes. Through a complex 3-dimensional structure, they protect the coding DNA and ensure appropriate separation of chromosomes. Aging is characterized by a progressive shortening of telomeres, which compromises their structure and function. Because of their protective function for genomic DNA, telomeres appear to play an important role in the development and progression of many age-related diseases, such as cardiovascular disease (CVD), malignancies, dementia, and osteoporosis. Despite substantial evidence that links telomere length with these conditions, the nature of these observations remains insufficiently understood. Therefore, future studies should address the question of causality. Furthermore, analytical methods should be further improved with the aim to provide informative and comparable results. This review summarize the actual knowledge of telomere biology and the possible implications of telomere dysfunction for the development and progression of age-related diseases. Furthermore, we provide an overview of analytical techniques for the measurement of telomere length and telomerase activity.

MITOCHONDRIAL DYSFUNCTION CAUSED BY TARGETED DELETION OF CLPP Results IN TELOMERE SHORTENING IN OOCYTES AND SOMATIC CELLS

Telomeres are highly preserved (TTAGGG)n repeats located at the ends of chromosomes, which serve as protective caps that maintain genomic stability over sequential cycles of cell division. Progressive shortening of telomeres over time due to inefficient DNA repair and chronic damage from oxidative stress has been suggested as a primary driver underlying somatic and reproductive aging. Another mechanism that has been linked to accelerated aging is mitochondrial dysfunction. However, the link between telomere shortening and mitochondrial dysfunction have not yet been characterized.

Increased tissue stiffness triggers contractile dysfunction and telomere shortening in dystrophic cardiomyocytes.

SummaryDuchenne muscular dystrophy (DMD) is a rare X-linked recessive disease that is associated with severe progressive muscle degeneration culminating in death due to cardiorespiratory failure. We previously observed an unexpected proliferation-independent telomere shortening in cardiomyocytes of a DMD mouse model. Here, we provide mechanistic insights using human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). Using traction force microscopy, we show that DMD hiPSC-CMs exhibit deficits in force generation on fibrotic-like bioengineered hydrogels, aberrant calcium handling, and increased reactive oxygen species levels. Furthermore, we observed a progressive post-mitotic telomere shortening in DMD hiPSC-CMs coincident with downregulation of shelterin complex, telomere capping proteins, and activation of the p53 DNA damage response. This telomere shortening is blocked by blebbistatin, which inhibits contraction in DMD cardiomyocytes. Our studies underscore the role of fibrotic stiffening in the etiology of DMD cardiomyopathy. In addition, our data indicate that telomere shortening is progressive, contraction dependent, and mechanosensitive, and suggest points of therapeutic intervention.

Abstract PO-020: Telomerase reverse transcriptase inhibitor NU-1 enhances radiation and chemotherapy sensitivity by disrupting DNA repair and accelerating cellular senescence

Abstract Upon reactivation in cancer, telomerase adds TTAGGG DNA repeats to the ends of chromosomes to prevent telomere erosion and senescence despite deregulated cell proliferation. The telomerase catalytic core consists of telomerase reverse transcriptase (TERT) and the template RNA, telomerase RNA (TR). While TR is broadly expressed, TERT is silenced in somatic cells, leading to progressive telomere shortening with each cell division leading to replicative senescence or apoptosis. That most cancers can maintain telomere integrity despite continued cell division has made TERT an attractive target for cancer therapy. We recently reported NU-1, a novel covalent telomerase inhibitor derived from the natural product chrolactomycin, that irreversibly blocks the telomerase activity of TERT. In examining mechanisms of action, our RNA-seq analysis found that NU-1 affects not only telomere maintenance but also DNA damage response (DDR) pathways. TERT has previously been implicated in multiple cancer-related pathways, including cell survival and repair of DNA double strand breaks (DSBs). To further explore the effects of TERT inhibition on the DDR, we used the human ER+ breast cancer cell line MCF7 to examine radiation response in the presence or absence of TERT inhibitors, including NU-1, chrolactomycin, BIBR1532 and MST-312. TERT inhibition was sufficient to delay both the resolution of γH2AX foci, a marker for DDR signaling, and DSB repair based on neutral comet assay. Surviving cells displayed increased cellular senescence after TERT inhibition. Importantly, only a minor fraction of the persistent γH2AX foci induced by TERT inhibitors colocalized with telomeres, based on binding of PNA probes. Examining mechanisms of DSB repair, NU-1 or BIBR1532 appeared to primarily inhibit the non-homologous end joining (NHEJ) DSB repair pathway versus less effect on homologous recombination (HR) repair, using Traffic Light Reporter assays. Similarly, NU-1 enhanced loss of cell proliferation after treatment with irinotecan, doxorubicin, etoposide, or paclitaxel in multiple TERT-expressing cancer cell lines. This effect was absent in the TERT negative Saos2 cell line. These results establish NU-1 as a chemical probe to explore TERT catalytic activity as a determinant of DNA damage tolerance and suggest telomerase inhibitors as tumor-specific radio/chemo-sensitizers. Imaging assay to probe the role of telomere length shortening on telomere-gene interactions in single cells

Telomeres are repetitive non-coding nucleotide sequences (TTAGGGn) capping the ends of chromosomes. Progressive telomere shortening with increasing age has been associated with shifts in gene expression through models such as the telomere position effect (TPE), which suggests reduced interference of the telomere with transcriptional activity of increasingly more distant genes. A modification of the TPE model, referred to as Telomere Position Effects over Long Distance (TPE-OLD), explains why some genes 1–10 MB from a telomere are still affected by TPE, but genes closer to the telomere are not. Here, we describe an imaging approach to systematically examine the occurrence of TPE-OLD at the single cell level. Compared to existing methods, the pipeline allows rapid analysis of hundreds to thousands of cells, which is necessary to establish TPE-OLD as an acceptable mechanism of gene expression regulation. We examined two human genes, ISG15 and TERT, for which TPE-OLD has been described before. For both genes, we found less interaction with the telomere on the same chromosome in old cells compared to young cells; and experimentally elongated telomeres in old cells rescued the level of telomere interaction for both genes. However, the dependency of the interactions on the age progression from young to old cells varied. One model for the differences between ISG15 and TERT may relate to the markedly distinct interstitial telomeric sequence arrangement in the two genes. Overall, this provides a strong rationale for the role of telomere length shortening in the regulation of gene expression.

Shorter telomere lengths in patients with severe COVID-19 disease

The incidence of severe manifestations of COVID-19 increases with age with older patients showing the highest mortality, suggesting that molecular pathways underlying aging contribute to the severity of COVID-19. One mechanism of aging is the progressive shortening of telomeres, which are protective structures at chromosome ends. Critically short telomeres impair the regenerative capacity of tissues and trigger loss of tissue homeostasis and disease. The SARS-CoV-2 virus infects many different cell types, forcing cell turn-over and regeneration to maintain tissue homeostasis. We hypothesize that presence of short telomeres in older patients limits the tissue response to SARS-CoV-2 infection. We measure telomere length in peripheral blood lymphocytes COVID-19 patients with ages between 29 and 85 years-old. We find that shorter telomeres are associated to increased severity of the disease. Individuals within the lower percentiles of telomere length and higher percentiles of short telomeres have higher risk of developing severe COVID-19 pathologies.
