The use of the concept of privileged structures significantly accelerates the search for new leads and their optimization. 6-(methylsulfonyl)-8-(4-methyl-4H-1,2,4-triazol-3-yl)-2-(5-nitro-2-furoyl)-2,6-diazaspiro[3.4]octane 1 has been identified as a lead, with MICs of 0.0124-0.0441 μg/mL against MTb multiresistant strains. Several series of structural analogues have been synthesized, including variations in the periphery and simplifications of their scaffolds. All synthesized compounds were tested against the MTb H37Rv strain and ESKAPE panel of pathogens using serial broth dilutions. However, an attempt to optimize structure of 1 did not lead to the development of more active compounds which can work against MTb, but to substances with high activity against S. aureus. Induced-fit docking and MM-GBSA calculations determined a change in the likely biotarget from deazaflavin-dependent nitroreductase to azoreductases. The privileged nature of the scaffold was demonstrated by the detection of a different type of activity.

Opuntia monacantha (Willd.) Haw. of the Grumari Sandbank, RJ, Brazil: Physical, chemical, biological and nutritional composition characterizations

Opuntia monacantha is known in Brazil and is commonly marketed in some regions, as in the ecological fairs of Porto Alegre. This study aims to investigate the physical and mineral characteristics as well as antioxidant activity, antibacterial activity, and chemical profile of the main metabolites of the fruit extracts. The fruits are a source of fiber (pulp and peel: 7.01 mg, 9.01 mg/100 g), respectively, selenium pulp and peel (0.04 mg/100 g and 0.05 mg/100 g), copper pulp (1.05 mg/100 g) and manganese peel (67,83 mg/100 g). The pulp showed phenolic content related to an antioxidant potential attributed to quercetin and rutin constitution in the extracts. The antibacterial activity was observed against S. aureus ATCC 33591 (methicillin-resistant, MRSA) and ATCC 29213 (methicillin-sensitive, MSSA) at 512 µg.mL−1 in the apolar extracts of the fruits constituted by saturated and unsaturated fatty acids, as well as phytosterols. The fruits presented a rich nutritional composition correlated with antioxidant and antibacterial action, being potential for health improvement and it can be explored by the food industry as well as in nature consumption by the population.

Antibacterial Activity, Toxicity and Drug-Likeness Profiles of Woodfordia fruticosa-Derived Metabolites Using Computational-Aided Drug Design Platform

This study presents a comprehensive investigation into the phytoconstituents reported from Woodfordia fruticosa (L.) Kurz leaf and flower extracts using gas chromatography-mass spectrometry (GC-MS) analysis, along with some existing phytochemicals, to explore their potential antibacterial properties through molecular docking studies. Followed by bio-assay-guided leave and flower extraction with two solvent systems, i.e., methanol (polar) and petroleum ether (non-polar), was used and further subjected to GC-MS to identify and quantify various secondary metabolites. Based on spectral intensity and volume area, a total of 28 compounds (P1 to P28) have been selected from GC-MS analyses, and an additional 14 compounds (P29 to P42) from previous reports were selected for molecular docking studies against DNA gyrase subunit B (GryB) of Escherichia coli (PDB ID: 7P2M) and Staphylococcus aureus (PDB ID: 5D7R) with novobiocin as the standard. Further, docking score or binding affinity (kcal/mol.) of each ligand were investigated, where the 4,5-dihydro-4,4-undeca methylene-2-phenyl-1,3-oxazin-6-one (P20) with a docking score of -8.4 kcal/mol., from the GC-MS-derived group and the chrysophanol-8-O-β-d-glucopyranoside (P37) with a docking score of -9.7 kcal/mol., from existing phytochemical groups were reported as potential antibacterial. The predicted toxicity and drug-ability profiles also suggested that GC-MS-derived candidates displayed comparatively higher non-toxic profiles but lower drug-likeness profiles than existing groups. This integrative approach explores the phytochemical profiles of W. fruticosa responsible for antibacterial activity of the crude extracts and providing insights into in selection of lead antibacterial agent through cost-effective computer-aided drug design platform to accelerate antibacterial drug discovery with higher chance of experimental success.

TC-14, a cathelicidin-derived antimicrobial peptide with broad-spectrum antibacterial activity and high safety profile

Cathelicidins, a major class of antimicrobial peptides (AMPs), hold considerable potential for antimicrobial drug development. In the present study, we identified a novel cathelicidin AMP (TC-33) derived from the Chinese tree shrew. Despite TC-33 demonstrating weak antimicrobial activity, the novel peptide TC-14, developed based on its active region, exhibited a 432-fold increase in antimicrobial activity over the parent peptide. Structural analysis revealed that TC-14 adopted an amphipathic α-helical conformation. The bactericidal mechanism of TC-14 involved targeting and disrupting the bacterial membrane, leading to rapid membrane permeabilization and rupture. Furthermore, TC-14 exhibited a high-safety profile, as evidenced by the absence of cytotoxic and hemolytic activities, as well as high biocompatibility and safety invivo. Of note, its potent antimicrobial activity provided significant protection in a murine model of skininfection. Overall, this study presents TC-14 as a promising drug candidate for antimicrobial drug development.

Functional Polyorganosilsesquioxane Nanocomposite Hydrogels With Encapsulated Silver Nanoparticles – Promising Compounds Against Gastrointestinal Pathogen Bacteria

AbstractNowadays the most common gastrointestinal related gram‐negative H. pylori and E. coli pathogen bacteria that are considered to be the main causes for the development of various gastric diseases and bloodstream infections are rapidly developing resistance against the standard antibiotics, commonly used in the clinical practices. One of the alternative high potential approaches for solving problems of bacterial resistance includes applications of silver‐based materials and silver nanoparticles (AgNPs). In the present study two types of amine‐ and triazol‐ functional polyorganosilsesquioxane nanocomposite sorbent hydrogels with narrowly dispersed AgNPs were developed. In vitro antibacterial activity and cytotoxicity tests showed concentration‐ and time‐dependent activity of the prepared nanocomposites against gram‐negative E. coli and H. pylori bacteria and a reduced cytotoxicity towards Caco‐2 human colon epithelial cells. Molecular structures and chemical compositions of the synthesized composites elucidated with transmission and scanning electron microscopy and UV‐Vis spectroscopy revealed spherical and narrowly dispersed along the polymer matrices AgNPs with average sizes of 3–5 nm. It was estimated that the chemical composition and particularly the zeta‐potentials values that were found to be dependent on the molecular structures of the synthesized nanocomposites had a high influence on the AgNPs antibacterial activity and cytoxicity profiles.

Exploring the Antibacterial Activity and Cellular Fates of Enterobactin-Drug Conjugates That Target Gram-Negative Bacterial Pathogens.

Siderophores are secondary metabolites utilized by bacteria to acquire iron (Fe), an essential transition metal nutrient. Fe levels in the host environment are tightly regulated and can be further restricted to starve invading bacterial pathogens in a host-defense process known as nutritional immunity. To survive and colonize the Fe-limited host environment, bacteria produce siderophores and express cognate siderophore transport machinery. These active transport pathways present an opportunity for selective and efficient drug delivery into bacterial cells, motivating decades of research on synthetic siderophore-antibiotic conjugates (SACs) as a Trojan-horse strategy for the development of targeted antibiotics.Enterobactin (Ent) is a triscatecholate siderophore produced and utilized by many Gram-negative bacteria, including all Escherichia coli and Salmonella species. Within these species, pathogenic strains cause a variety of human diseases including urinary tract infections, gastroenteritis, and sepsis. Infections caused by these Gram-negative pathogens can be difficult to treat because of the impermeability of the outer membrane (OM). This impermeability can be overcome by utilizing siderophores as drug delivery vectors for targeting Gram-negative pathogens. Ent is a promising delivery vector because it undergoes active transport across the OM mediated by the Ent uptake machinery after scavenging Fe(III) from the extracellular environment. Despite the well-elucidated chemistry and biology of Ent, its use for SAC development was hampered by the lack of an appropriate functional group for cargo attachment. Our laboratory addressed this need by designing and synthesizing monofunctionalized Ent scaffolds. Over the past decade, we have used these scaffolds to explore Ent-based SACs with a variety of drug warheads, including β-lactam and fluoroquinolone antibiotics, and Pt(IV) prodrugs. Investigations of the antibacterial activities of these conjugates and their cellular fates have informed our design principles and revealed approaches to achieving enhanced antibacterial potency and pathogen-targeted activity. Collectively, our studies of Ent-drug conjugates have provided discoveries, understanding, and invaluable insights for future design and evaluation of SACs.In this Account, we present the story of our work on Ent-drug conjugates that began about ten years ago with the development of monofunctionalized Ent scaffolds and the design and synthesis of various conjugates based on these scaffolds. We describe the antibacterial activity profiles and uptake pathways of Ent-drug conjugates harboring traditional antibiotics and repurposed platinum anticancer agents as well as studies that address cellular targets and fates. Finally, we discuss other applications of monofunctionalized Ent scaffolds, including a siderophore-based immunization strategy. We intend for this Account to inspire further investigations into the fundamental understanding and translational applications of siderophores and siderophore-drug conjugates.

Evaluation of Yield, Chemical Profile, and Antimicrobial Activity of Teucrium polium L. Essential Oil Used in Iranian Folk Medicine.

Teucrium polium L. is used to treat many diseases like abdominal pains, indigestion, colds, and reproductive system diseases in Iranian folk medicine. This study was designed to investigate the yield, chemical profile of essential oil, and antibacterial and antifungal activity of this species. The flowering aerial parts of T. polium were collected from the Margh region of Kashan, Iran. The essential oil of the plant was extracted and separated using the Clevenger apparatus and analyzed using gas chromatography-mass spectrometry (GC-MS). The antimicrobial activity of the essential oil against a variety of standard microbial strains was investigated with the Agar well-diffusion method and determination of the lowest growth inhibitory and lethal concentration (MIC and MBC). The results showed that the yield of T. polium essential oil (TPEO) was 0.5778%. Based on the results of GC-MS, 76 compounds (99.3%) were identified in the TPEO, of which α-pinene (9.67%), β-caryophyllene (8.07%), β-pinene (5.04%), nerolidol (4.94%), and oleic acid (4.57%) were the dominant components. The results of antimicrobial tests showed that the TPEO on Staphylococcus aureus created the largest zone of inhibition (~ 14.29 mm). The dominant inhibitory activity of this essential oil was against Gram-negative bacteria Escherichia coli (~ 9.00 mm), which was almost similar to rifampin (~ 11 mm). It seems that the predominance of terpenoid and acidic compounds of the essential oil is one of the possible factors of the anti-bacterial activity of this essential oil. Therefore, the TPEO may be a promising and potential strategy to inhibit some bacterial strains.

Medicinal plants in burns treatment – a systematic review

Introduction and aim. The World Health Organization (WHO) reports that around 11 million people experience burn injuries each year.1,2 Burn injuries occur when tissue damage is caused by various factors such as UV radiation, heat, chemicals, or electric current.3,4 While burns primarily affect the skin, they can also extend to deeper tissues like bones or muscles. When the skin is burned, its vital functions, including protection against the external environment, pathogens, and evaporation, are compromised.1 The appropriate treatment method must be chosen based on the burn stage, the patient's condition, and the cause. The aim of this paper is to provide a comprehensive review based on literature, concerning superficial burns and sunburns treatment, with a focus on nature-derived topical treatment methods.
 Material and methods. PubMed database was searched for the following terms: “burns”, “burn treatment”, “aloe vera”, “sunburn”, “centella asiatica”, “climate change”, “wound healing” for the articles published between 2005-2023 and written in the English language.
 Conclusion. Herbal-derived compounds, with their diverse mechanisms of action, antibacterial activity, and safety profiles present a competitive alternative to conventional treatment of burns and sunburns, however, there are still not enough clinical trials to assess the effectiveness and safety profiles of nature-derived compounds.

Self-Aggregation, Antimicrobial Activity and Cytotoxicity of Ester-Bonded Gemini Quaternary Ammonium Salts: The Role of the Spacer.

Five ester-bonded gemini quaternary ammonium surfactants C12-En-C12 (n = 2, 4, 6), with a flexible spacer group, and C12-Bm-C12 (m = 1, 2), with rigid benzene spacers, were synthesized via a two-step reaction and analyzed. Furthermore, the effects of the spacer structure, spacer length and polymerization degree on the self-aggregation, antimicrobial activity and cytotoxicity of C12-En-C12 and C12-Bm-C12 and their corresponding monomer N-dodecyl-N,N,N-trimethyl ammonium chloride DTAC were investigated. The results showed that C12-En-C12 and C12-Bm-C12 had markedly lower critical micellar concentration (CMC) values and lower surface tension than DTAC. Moreover, the CMC values of C12-En-C12 and C12-Bm-C12 decreased with increasing spacer length. In the case of equivalent chain length, the rigidity and steric hindrance of phenylene and 1,4-benzenediyl resulted in larger CMC values for C12-Bm-C12 than for C12-En-C12. The antibacterial ability of C12-En-C12 and C12-Bm-C12 was assessed using Escherichia coli (E. coli) and Staphylococcus albus (S. aureus) based on minimum inhibitory concentrations (MICs). Furthermore, C12-En-C12 and C12-Bm-C12 exhibited higher antimicrobial activity than DTAC and had stronger function toward S. aureus than E. coli. The antimicrobial activity was enhanced by increasing the spacer chain length and decreased with the increased rigidity of the spacers. The cytotoxic effects of C12-En-C12 and C12-Bm-C12 in cultured Hela cells were evaluated by the standard CCK8 method based on half-maximal inhibitory concentration (IC50). The cytotoxicity of C12-En-C12 and C12-Bm-C12 was significantly lower than alkanediyl-α,ω-bis(dimethyldodecylammonium) bromide surfactants and DTAC. The spacer structure and the spacer length could induce significant cytotoxic effects on Hela cells. These findings indicate that the five ester-bonded GQASs have stronger antibacterial activity and lower toxicity profile, and thus can be used in the pharmaceutical industry.

Cholinium-Based Ionic Liquids as Promising Antimicrobial Agents in Pharmaceutical Applications: Surface Activity, Antibacterial Activity and Ecotoxicological Profile.

Cholinium-based ionic liquids are compounds increasingly studied in pharmaceutics and biomedicine to enhance bioavailability in drug delivery systems and as bioactive ingredients in pharmaceutical formulations. However, their potential as antimicrobial agents has scarcely been investigated. Herein, we explored the antimicrobial activity of a series of surface-active cholinium-based ionic liquids (Chol-ILs). For this purpose, Chol-ILs with alkyl chains of 10-16 carbon atoms were synthesized and their self-assembly in aqueous medium was investigated. Subsequently, their antimicrobial activity against a panel of clinically relevant bacteria and their ability to eradicate MRSA and P. aeruginosa PAO1 biofilms was evaluated. Finally, we analyzed the ecotoxicological profile of Chol-ILs in terms of susceptibility to aerobic biodegradation and acute aquatic toxicity against D. magna and V. fisheri. Our results reveal that cholinium-based ILs with alkyl chain lengths ≥12 C show a broad spectrum of antibacterial activity. Their antimicrobial efficacy depends on their hydrophobicity, with the C14-C16 homologs being the most effective compounds. These ILs exhibit antimicrobial activity similar to that of imidazolium ILs and quaternary ammonium antiseptics. Moreover, the longer alkyl chain Chol-ILs are able to eradicate established biofilms at concentrations as low as 16-32 µg/mL. The biodegradation rate of cholinium-based ILs decreases with alkyl chain elongation. Our results reinforce the suitability of Chol-ILs as promising multifunctional compounds for application in pharmaceutical and biomedical formulation.

Antibacterial activity and GC-MS profile of secondary metabolites of Bacillus subtilis subsp. subtilis HSFI-9 associated with Holothuria scabra

The emergence of resistant bacteria has led to the importance of new antibacterial discovery. Before this research, the marine biota Holothuria scabra found in the Kodek gulf area, West Nusa Tenggara, Indonesia, was explored to isolate Bacillus sp. The present research was conducted to explore Bacillus sp. to produce secondary antibacterial metabolites, by culturing isolate using media containing starch, yeast, and peptone. Ethyl acetate was used for secondary metabolite extraction. Antibacterial activity screening was carried out by two-fold microdilution test to obtain Minimum Inhibitory Concentration (MIC) value. In this study, it was found that the ethyl acetate extract of the strain HSFI-9 can inhibit the growth of Staphylococcus aureus, Extended Spectrum ?-Lactamase (ES?L)-Escherichia coli, and Mycobacterium smegmatis with sequential minimum inhibitory concentrations (MIC) values ??of 3.125; 25; and 50 µg/mL. Based on phylogenetic tree analysis, the strain HSFI-9 show 99.71 similarity with Bacillus subtilis subsp. subtilis. The ethyl acetate extract of Bacillus subtilis subsp. subtilis HSFI-9 metabolomics profile was analyzed using Gas Chromatography-Mass Spectroscopy (GC-MS). It showed five volatile secondary metabolites, with the first dominant compound being (2S, 3S)-(-)-3-propyloxiranemethanol (MW 116, C6H12O2) with an area of ??34.78% (database similarity index of 73.3%). This study concluded that B. subtilis subsp. subtilis HSFI-9 could be further developed as an antibacterial producer.

Antibiotic resistance is rising across the world. For a very long time, bitter ginger (Zingiber zerumbet) has been used as one of the most popular herbal remedies to treat a wide range of common diseases. Ginger has been shown to have antioxidant and antibacterial activity. It has various bioactive chemicals that might be utilized as an alternative treatment option for many infectious diseases. The present study aimed to examine the biochemical profile of ginger, antioxidant, and antibacterial activity against selective endodontic microbes. Antioxidant was measured using DPPH and antibacterial activity was performed using disk diffusion tests. Streptococcus mutants, Enterococcus faecalis, Staphylococcus spp., and Lactobacillus spp. were tested for antibacterial activity. Before evaluating the dried extracts, all solvents were eliminated using rotary evaporation. The obtained IC50 value revealed that ethanol extract had the greatest antioxidant activity. Concerning each bacterium, the plant extracts demonstrated considerable antibacterial activity (p = .001). Ethanol extracts showed the strongest antibacterial activity against the studied microorganisms. This study highlights that the Zingiber zerumbet (Z. zerumbet) is a strong antibacterial herb against multidrug-resistant (MDR) gram-positive bacteria. It may also be employed as a possible natural antioxidant source.

Phytochemical and Biological Screening of Catholic Vegetable (Jatropha Tanjorensis) Aqueous Extract in Bali, Taraba State, Nigeria

Jatropha tanjorensis has been used as a medicinal herb in treatment and management of disease because they are relatively safer, more affordable and sometimes offers better therapeutic value than synthetic drugs. The study investigated the phytochemical and biological screening of catholic vegetable (Jatropha tanjorensis) aqueous extract with the objectives of evaluating some secondary metabolites in the plant using standard qualitative methods and ascertaining the antimicrobial effect of Jatropha tanjorensis aqueous leave extract on bacteria of medical importance such as Pseudomonas aeruginosa, Klebsiella pneumonia, Staphylococcus aureus, Salmonella typhi and Escherichia coli using the agar well diffusion method. The results revealed that the extract of the leaf of J. tanjorensis contain some important bioactive components (phytochemicals) in a high amount such as saponins followed by flavonoids, alkaloid and tannins in moderate amount whereas phenol, phlobatannins and glycosides are in low amount. It also shows that the extract was active against E. coli, P. aeruginosa, S. aureus, K. pneumonia and S. typhi measuring clear zones of inhibition 30.0mm, 22.0mm, 19.0mm, 18.0mm and 17.0mm respectively. Thus, the exhibition of strong antibacterial activity and phytochemical profiles obtained from the analyses of J. tanjorensis leaf indicated that it might be recommended for human consumption and adequate amount of consumption could contribute greatly towards meeting human nutritional needs for normal body growth and adequate protection and curative measures to common diseases related to the test organisms.

Antimicrobial and Antibiofilm Properties of Latvian Honey against Causative Agents of Wound Infections.

Honey is widely used in traditional medicine and modern wound healing biomaterial research as a broad-spectrum antimicrobial, anti-inflammatory and antioxidant agent. The study's objectives were to evaluate the antibacterial activity and polyphenolic profiles of 40 monofloral honey samples collected from beekeepers in the territory of Latvia. The antimicrobial and antifungal activity of Latvian honey samples were compared with commercial Manuka honey and the honey analogue sugar solutions-carbohydrate mixture and tested against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, clinical isolates Extended-Spectrum Beta-Lactamases produced Escherichia coli, Methicillin-resistant Staphylococcus aureus and Candida albicans. Antimicrobial activity was evaluated with the well-diffusion method (80% honey solution w/v) and microdilution method. The honey samples with the highest antimicrobial potential were tested to prevent biofilm development and activity against a preformed biofilm. The principal component analysis of the antimicrobial properties of honey samples vs. polyphenolic profile was performed. Eleven honey samples exhibited antibacterial activity to all investigated bacteria. The antibacterial effect of the samples was most significant on the Gram-positive bacteria compared to the studied Gram-negative bacteria. Latvian honey presents promising potential for use in wound healing biomaterials, opening the possibility of achieving long-term antibacterial effects.

SYNTHESIS AND STRUCTURE ELUCIDATION OF NEW METHYL 1H-BENZIMIDAZOLE-5-CARBOXYLATE DERIVATIVES

Objective: In this study, in connection with previous works in our department, some methyl 1H-benzimidazole-5-carboxylate derivatives were synthesized for the first time. Compounds were modified by substituting the second position of the benzimidazole ring with 4-methylpiperidinyl groups for increasing antibacterial activity. 
 Material and Method: The targeted methyl 1H-benzimidazole-5-carboxylates were synthesized by the reaction of o-phenylendiamine derivatives with urea. Oxygen at the 2nd position was converted to the chlorine in the presence of POCl3. Finally, the resulting products were obtained by the nucleophylic reaction with 4-methylpiperidine. Structures of synthesized compounds were elucidated with 1H- 13C-NMR and LC-MS techniques.
 Result and Discussion: Methyl 1H-benzimidazole-5-carboxylate derivatives that bearing 4-methyl piperidinyl groups at 2nd position were synthesized for getting better in vitro antibacterial activity profiles. In vitro antibacterial activity of the synthesized compounds in this study are under investigation.

Synthesis, spectroscopic characterization, antibacterial activity and antiproliferative profile of a new silver(I) complex of 5-fluorocytosine

In this paper, we report the synthesis, structural characterization, and explore the potential antibacterial and antiproliferative activities of a new silver(I) complex of the nucleotide analogue 5-fluorocytosine, Ag-5FC. Elemental and thermogravimetric (TGA) analyses led to the minimal formula AgC4H4FN3O·H2O. Infrared (FT-IR) and solid-state 13C nuclear magnetic resonance (NMR) spectroscopic analyses indicate that the ligand coordinates to the metal ion by its nitrogen and oxygen atoms. High resolution mass spectrometric (HRMS) data confirmed the proposed composition of the complex as well as the presence of recombination species in solution. Molecular modeling reinforces the proposed N,O-coordination of 5FC to silver as indicated by IR data with a possible structure of the complex in a dimeric form, of coordination formula [Ag(C4H4FN3O)(OH2)]2. The complex showed a modest antimicrobial activity (MIC = 4.3 mmol⋅L−1) against Gram-negative (Pseudomonas aeruginosa and Escherichia coli) and Gram-positive (Staphylococcus aureus and Bacillus cereus) strains. Despite the low solubility, the Ag-5FC complex totally inhibited the proliferation of colon (HT-29), multidrug resistant ovarian (NCI/ADR-RES), prostate (PC-3), and renal (786–0) tumor cell lines in a concentration range of 6.2 to 9.3 µmol L−1. Further studies are necessary to confirm efficiency and safety for continuing the pre-clinical evaluation of the Ag-5FC complex.
