Implications and Current Limitations of Oogenesis from Female Germline or Oogonial Stem Cells in Adult Mammalian Ovaries.

A now large body of evidence supports the existence of mitotically active germ cells in postnatal ovaries of diverse mammalian species, including humans. This opens the possibility that adult stem cells naturally committed to a germline fate could be leveraged for the production of female gametes outside of the body. The functional properties of these cells, referred to as female germline or oogonial stem cells (OSCs), in ovaries of women have recently been tested in various ways, including a very recent investigation of the differentiation capacity of human OSCs at a single cell level. The exciting insights gained from these experiments, coupled with other data derived from intraovarian transplantation and genetic tracing analyses in animal models that have established the capacity of OSCs to generate healthy eggs, embryos and offspring, should drive constructive discussions in this relatively new field to further exploring the value of these cells to the study, and potential management, of human female fertility. Here, we provide a brief history of the discovery and characterization of OSCs in mammals, as well as of the in-vivo significance of postnatal oogenesis to adult ovarian function. We then highlight several key observations made recently on the biology of OSCs, and integrate this information into a broader discussion of the potential value and limitations of these adult stem cells to achieving a greater understanding of human female gametogenesis in vivo and in vitro.

Endocrine disruption: From a whole-lake experiment to a calibrated ecosystem model

The unique Canadian whole-ecosystem study on the impact of 17α-ethinylestradiol (EE2) on a freshwater food web (Karen et al., 2014) provides evidence of the value of whole-ecosystem experiments for understanding indirect effects of endocrine disrupting compounds (EDCs) and other aquatic stressors. To further explain the indirect effects of EE2 observed in the experimental lake, an ecosystem model based on AQUATOX equations was successfully developed and calibrated. Discussions with the scientists who gathered the experimental data were necessary to ensure the consistency of the parameters used in the model and the realism of the biomass dynamics observed in such ecosystems. The prediction results helped further explain how the other fishes were impacted by the fathead minnow collapse. This study also suggests that a mix of reduced gamete production, increased gamete mortality and fish mortality is a potential mechanism for the collapse of the fathead minnow population.

A rigorous measure of genome-wide genetic shuffling that takes into account crossover positions and Mendel’s second law

Comparative studies in evolutionary genetics rely critically on evaluation of the total amount of genetic shuffling that occurs during gamete production. Such studies have been hampered by the absence of a direct measure of this quantity. Existing measures consider crossing-over by simply counting the average number of crossovers per meiosis. This is qualitatively inadequate, because the positions of crossovers along a chromosome are also critical: a crossover toward the middle of a chromosome causes more shuffling than a crossover toward the tip. Moreover, traditional measures fail to consider shuffling from independent assortment of homologous chromosomes (Mendel's second law). Here, we present a rigorous measure of genome-wide shuffling that does not suffer from these limitations. We define the parameter [Formula: see text] as the probability that the alleles at two randomly chosen loci are shuffled during gamete production. This measure can be decomposed into separate contributions from crossover number and position and from independent assortment. Intrinsic implications of this metric include the fact that [Formula: see text] is larger when crossovers are more evenly spaced, which suggests a selective advantage of crossover interference. Utilization of [Formula: see text] is enabled by powerful emergent methods for determining crossover positions either cytologically or by DNA sequencing. Application of our analysis to such data from human male and female reveals that (i) [Formula: see text] in humans is close to its maximum possible value of 1/2 and that (ii) this high level of shuffling is due almost entirely to independent assortment, the contribution of which is ∼30 times greater than that of crossovers.

Patterns of structural change in gonads of the divine dwarfgoby Eviota epiphanes as they sexually transition.

This study documents changes in gonadal structure for the serial hermaphrodite (or bidirectional sex changer) divine dwarfgoby Eviota epiphanes (family Gobiidae) as individuals transition in both directions. To evaluate transitional gonad morphology, individuals actively producing the same gamete type (oocytes or sperm) were set up into pairs and euthanised over a period of 14 days to get a time series of morphological changes during gonad transformation. Results from this study show that rapid changes in the gonad take place at a structural level as individuals change their reproductive function and gamete production. Changing from oocyte production (o-phase) to sperm production (s-phase) starts with the breakdown of vitellogenic oocytes (i.e., atresia) followed by the appearance and proliferation of spermatogenic tissue which, in most cases, was not previously visible. Changing from sperm production to oocyte production included the cessation of sperm production, a reduction in size and number of seminiferous lobules and the maturation of previtellogenic oocytes already present in the gonads. Experimental fish changed from oocyte production to sperm production more readily than from sperm production to oocyte production. The hypothesis that shifts in sexual function among serially hermaphroditic fish species have a similar cost in either direction is not supported in E. epiphanes.

Triploid production via heat-induced diploidisation of megaspores in Populus pseudo-simonii

Triploid breeding in Populus enhances the commercial properties of cultivated trees. Sexual polyploidization through the production of diploid gametes produces elite triploids with heterosis and ploidy vigour. Populus pseudo-simonii is a species indigenous to China. It is highly adaptable and resistant drought, cold, and saline–alkaline stresses. To improve the properties in growth and pulpwood traits of P. pseudo-simonii, we used heat treatments to induce chromosome doubling in the megaspores as a first step in the production of triploid saplings. The heat treatments were applied during development stages of the female flower buds. We produced 117 triploids via the diploidisation of megaspores. The highest proportion of triploids among the offspring (51.43%) was obtained by applying a heat treatment of 40 °C over 4 h to flower buds in which one-third of the catkins were outside the bract scales. The proportion of pachytene was positively correlated with the average rate of triploid production in each bud developmental stage. Thus, among the phases of meiosis, the pachytene stage may be most susceptible to heat treatments aiming to diploidise the megaspores in P. pseudo-simonii. The megasporocytes in these treated flower buds were mostly in the first meiotic division stages, suggesting that the first meiotic division restitution (FDR) type 2n megaspores may have been induced by our experimental procedures. These triploid hybrids derived from FDR type 2n megaspores will probably contribute significantly to programmes aimed at improving commercial properties of Populus congeners in the section Tacamahaca.

Colchicine did not affect the viability of induced 2n pollen in <i>Populus tomentosa</i>

Colchicine is widely used as a mutagen to induce production of diploid gametes in plants. However, whether colchicine affects induced pollen viability remains unclear. To clarify whether colchicine affected the viability of induced pollen, we induced production of diploid pollen by colchicine, followed by pollen germination and crossing induced pollen with normal gametes to produce triploid in Carrière. The results showed that the predominant meiotic stages and the number of colchicine injections had significant effects on the occurrence rates of induced 2n pollen. When the colchicine injection was given at diakinesis, a significant decrease in the pollen production per bud was observed ( < 0.001). The morphology of the colchicine-induced 2n pollen was similar to that of the natural 2n pollen in its ectexine structure. The pollen germination experiments revealed that there was also no significant difference in germination rates between the induced diploid pollen and natural 2n pollen grains, and 68 triploids were created by crossing colchicine-induced pollen. Our findings revealed that colchicine injection could induce to produce 2n pollen and will not lead to dysfunction of induced diploid pollen.in vitroPopulus tomentosapP. tomentosa

Germline stem cell homeostasis.

In many species, germline stem cells (GSCs) function to sustain gametogenesis throughout the life of organismal life span. As the source of gametes, the only cell type that can pass the genetic information to the next generation, GSCs play a fundamental role in maximizing the quantity of gametes that animals produce, while ensuring their highest quality. GSCs are maintained by the signals from their niches, and germ cells that exited the niche undergo differentiation to generate functional gametes. GSC population is sustained by a multitude of mechanisms such as asymmetric stem cell divisions and dedifferentiation of partially differentiated germ cells. In this review, we summarize the mechanisms that maintain GSC homeostasis to ensure life-long production of functional gametes.

From the Ivory Tower to Reality! Conclusions of the New Edition.

While many of the traditional scientific disciplines have developed over centuries, animal conservation is a relative newcomer. It relies on multiple specialties with different levels of expertise that, eventually, generate vast amounts of data. More specifically, conservation physiology is an emerging area that can be defined as 'an integrative scientific discipline applying physiological concepts, tools, and knowledge to characterizing biological diversity and its ecological implications; understanding and predicting how organisms, populations, and ecosystems respond to environmental change and stressors; and solving conservation problems across the broad range of taxa, including microbes, plants, and animals' (Cooke et al. 2013). Reproductive biology is more focused, given that it mainly deals with the physiology underlying the production of gametes, embryos, and offspring, and the many associated processes that control these events. However, it is integrated into the different components of conservation physiology. In bringing together the various contributors for this book, the editors' purpose was to provide readers with a new perspective about the complexity behind reproduction and the role it plays in species conservation. Chapters highlight the diversity of reproductive mechanisms across taxa, and provide insight into how they may have evolved, and likely will continue to evolve in a changing environment. To conservation physiologists, the hope is that this information will be applied to sustain populations in both natural habitats and managed facilities. Ultimately, a major goal is to forecast and mitigate negative impacts of environmental change or anthropogenic pressures on animal fitness, which will only follow once we have acquired a solid understanding of reproductive processes.

Loss-of-function of sox3 causes follicle development retardation and reduces fecundity in zebrafish

Folliculogenesis is essential for production of female gametes in vertebrates. However, the molecular mechanisms underlying follicle development, particularly apoptosis regulation in ovary, remain elusive. Here, we generated sox3 knockout zebrafish lines using CRISPR/Cas9. sox3 knockout led to follicle development retardation and a reduced fecundity in females. Comparative analysis of transcriptome between sox3−/− and wild-type ovaries revealed that Sox3 was involved in pathways of ovarian steroidogenesis and apoptosis. Knockout of sox3 promoted follicle apoptosis and obvious apoptosis signals were detected in somatic cells of stages III and IV follicles of sox3−/− ovaries. Moreover, Sox3 can bind to and activate the promoter of cyp19a1a. Up-regulation of Cyp19a1a expression promoted 17β-estradiol synthesis, which inhibited apoptosis in follicle development. Thus, Sox3 functions as a regulator of Cyp19a1a expression, via 17β-E2 linking apoptosis suppression, which is implicated in improving female fecundity.

DNA Damage and Repair in Human Reproductive Cells.

The fundamental underlying paradigm of sexual reproduction is the production of male and female gametes of sufficient genetic difference and quality that, following syngamy, they result in embryos with genomic potential to allow for future adaptive change and the ability to respond to selective pressure. The fusion of dissimilar gametes resulting in the formation of a normal and viable embryo is known as anisogamy, and is concomitant with precise structural, physiological, and molecular control of gamete function for species survival. However, along the reproductive life cycle of all organisms, both male and female gametes can be exposed to an array of “stressors” that may adversely affect the composition and biological integrity of their proteins, lipids and nucleic acids, that may consequently compromise their capacity to produce normal embryos. The aim of this review is to highlight gamete genome organization, differences in the chronology of gamete production between the male and female, the inherent DNA protective mechanisms in these reproductive cells, the aetiology of DNA damage in germ cells, and the remarkable DNA repair mechanisms, pre- and post-syngamy, that function to maintain genome integrity.

The evolution and patterning of male gametophyte development.

The reproductive adaptations of land plants have played a key role in their terrestrial colonization and radiation. This encompasses mechanisms used for the production, dispersal and union of gametes to support sexual reproduction. The production of small motile male gametes and larger immotile female gametes (oogamy) in specialized multicellular gametangia evolved in the charophyte algae, the closest extant relatives of land plants. Reliance on water and motile male gametes for sexual reproduction was retained by bryophytes and basal vascular plants, but was overcome in seed plants by the dispersal of pollen and the guided delivery of non-motile sperm to the female gametes. Here we discuss the evolutionary history of male gametogenesis in streptophytes (green plants) and the underlying developmental biology, including recent advances in bryophyte and angiosperm models. We conclude with a perspective on research trends that promise to deliver a deeper understanding of the evolutionary and developmental mechanisms of male gametogenesis in plants.

Molecular and physiological characterization of a natural interspecific coffee hybrid

Natural hybridization is a common phenomenon in plants and considered to be a driving force of biodiversity and evolution. In coffee spontaneous hybridization between two different species is earlier documented. Here we investigated the morphological, molecular and physiological characteristics of a natural tetraploid interspecific coffee hybrid involving allopolyploid Coffea arabica and diploid C. excelsa species. The formation of the tetraploid hybrid was postulated on account of the production of unreduced female gametes in C. excelsa. In our findings, most morphological characters of the interspecific hybrid were found to be intermediate between the putative parental species, but the young leaf colour was found to be distinctively transgressive. Sequence-related amplified polymorphism marker analysis unequivocally supported the involvement of C. excelsa and C. arabica in hybrid formation. The gas exchange parameters, chlorophyll fluorescence parameters were compared between the hybrid and parents which revealed that the interspecific hybrid had the significantly higher substomatal CO2 concentration and non-photochemical quenching compared to parents. This tetraploid interspecific hybrid encompassing the genome of two divergent species could be of ascertainable importance in arabica coffee breeding program.

QTL mapping of natural variation reveals that the developmental regulator bruno reduces tolerance to P-element transposition in the Drosophila female germline.

Transposable elements (TEs) are obligate genetic parasites that propagate in host genomes by replicating in germline nuclei, thereby ensuring transmission to offspring. This selfish replication not only produces deleterious mutations—in extreme cases, TE mobilization induces genotoxic stress that prohibits the production of viable gametes. Host genomes could reduce these fitness effects in two ways: resistance and tolerance. Resistance to TE propagation is enacted by germline-specific small-RNA-mediated silencing pathways, such as the Piwi-interacting RNA (piRNA) pathway, and is studied extensively. However, it remains entirely unknown whether host genomes may also evolve tolerance by desensitizing gametogenesis to the harmful effects of TEs. In part, the absence of research on tolerance reflects a lack of opportunity, as small-RNA-mediated silencing evolves rapidly after a new TE invades, thereby masking existing variation in tolerance. We have exploited the recent historical invasion of the Drosophila melanogaster genome by P-element DNA transposons in order to study tolerance of TE activity. In the absence of piRNA-mediated silencing, the genotoxic stress imposed by P-elements disrupts oogenesis and, in extreme cases, leads to atrophied ovaries that completely lack germline cells. By performing quantitative trait locus (QTL) mapping on a panel of recombinant inbred lines (RILs) that lack piRNA-mediated silencing of P-elements, we uncovered multiple QTL that are associated with differences in tolerance of oogenesis to P-element transposition. We localized the most significant QTL to a small 230-kb euchromatic region, with the logarithm of the odds (LOD) peak occurring in the bruno locus, which codes for a critical and well-studied developmental regulator of oogenesis. Genetic, cytological, and expression analyses suggest that bruno dosage modulates germline stem cell (GSC) loss in the presence of P-element activity. Our observations reveal segregating variation in TE tolerance for the first time, and implicate gametogenic regulators as a source of tolerant variants in natural populations.

Both male and female gamete generating cells produce processed pseudogenes in the human genome

The human genome contains an unusually large number of processed pseudogenes. The fact that processed pseudogenes are roughly 33% more abundant in our X chromosome than in our autosomes suggests that this overabundance is the result of the fact that human oogenesis is much longer than that of non-mammalian species. Here, we analyze the origins of the processed pseudogenes found on the human Y chromosome to determine whether human spermatogenesis also contribute to this overabundance.Our results show that human processed pseudogenes not only retrotranspose to the Y chromosome, but are also produced by genes on the Y chromosome. Furthermore, the fact that X chromosomes are three times more abundant than Y chromosomes likely explains why the euchromatic density of processed pseudogenes is three times higher in the X chromosome than in the Y chromosome.The large number of processed pseudogenes found in our genome is therefore due to the low substrate specificity of the L1 reverse transcriptase responsible for the reverse transcription of germline mRNA molecules into processed pseudogenes, as well as the life-long production of both male and female gametes.

Cloning and characterization of the homoeologous genes for the Rec8-like meiotic cohesin in polyploid wheat

BackgroundMeiosis is a specialized cell division critical for gamete production in the sexual reproduction of eukaryotes. It ensures genome integrity and generates genetic variability as well. The Rec8-like cohesin is a cohesion protein essential for orderly chromosome segregation in meiotic cell division. The Rec8-like genes and cohesins have been cloned and characterized in diploid models, but not in polyploids. The present study aimed to clone the homoeologous genes (homoeoalleles) for Rec8-like cohesin in polyploid wheat, an important food crop for humans, and to characterize their structure and function under a polyploid condition.ResultsWe cloned two Rec8-like homoeoalleles from tetraploid wheat (TtRec8-A1 and TtRec8-B1) and one from hexaploid wheat (TaRec8-D1), and performed expression and functional analyses of the homoeoalleles. Also, we identified other two Rec8 homoeoalleles in hexaploid wheat (TaRec8-A1 and TaRec8-B1) and the one in Aegilops tauschii (AetRec8-D1) by referencing the DNA sequences of the Rec8 homoeoalleles cloned in this study. The coding DNA sequences (CDS) of these six Rec8 homoeoalleles are all 1,827 bp in length, encoding 608 amino acids. They differed from each other primarily in introns although single nucleotide polymorphisms were detected in CDS. Substantial difference was observed between the homoeoalleles from the subgenome B (TtRec8-B1 and TaRec8-B1) and those from the subgenomes A and D (TtRec8-A1, TaRec8-A1, and TaRec8-D1). TtRec8-A1 expressed dominantly over TtRec8-B1, but comparably to TaRec8-D1, in polyploid wheat. In addition, we developed the antibody against wheat Rec8 and used the antibody to detect Rec8 cohesin in the Western blotting and subcellular localization analyses.ConclusionsThe Rec8 homoeoalleles from the subgenomes A and D are transcriptionally more active than the one from the subgenome B in polyploid wheat. The structural variation and differential expression of the Rec8 homoeoalleles indicate a unique cross-genome coordination of the homoeologous genes in polyploid wheat, and imply the distinction of the wheat subgenome B from the subgenomes A and D in the origin and evolution.

Neuroanatomical Framework of the Metabolic Control of Reproduction.

A minimum amount of energy is required for basic physiological processes, such as protein biosynthesis, thermoregulation, locomotion, cardiovascular function, and digestion. However, for reproductive function and survival of the species, extra energy stores are necessary. Production of sex hormones and gametes, pubertal development, pregnancy, lactation, and parental care all require energy reserves. Thus the physiological systems that control energy homeostasis and reproductive function coevolved in mammals to support both individual health and species subsistence. In this review, we aim to gather scientific knowledge produced by laboratories around the world on the role of the brain in integrating metabolism and reproduction. We describe essential neuronal networks, highlighting key nodes and potential downstream targets. Novel animal models and genetic tools have produced substantial advances, but critical gaps remain. In times of soaring worldwide obesity and metabolic dysfunction, understanding the mechanisms by which metabolic stress alters reproductive physiology has become crucial for human health.

Sperm quality in wild-caught and farmed males of the European eel (Anguilla anguilla)

Sperm density and performance of wild and farmed male European eels were evaluated to investigate the effect of maintenance in captivity on ejaculate quality. Hormonal stimulation (with human chorionic gonadotropin) lasted for 17 weeks. Different origins of the male European eels partially influenced the quality of the ejaculates. Indeed, wild animals (i.e., collected at a natural site) had greater sperm longevity during the hormonal stimulation, whereas there was no significant effect of the origin on sperm density, spermatocrit, the percentage of motile sperm, and plasma testosterone concentrations. The different origins of the males also affected fatty acid sperm content, with greater unsaturated fatty acid and omega-6 concentrations in wild eels and lesser concentrations of saturated fatty acids and omega-3 fatty acids in farmed eels. Regarding sperm quality of European eels, this is the first study that takes into account the effect of different origins of stimulated males (wild-caught compared with farmed) on sperm quality, and these findings may help to improve the production of high-quality gametes in this endangered species.

New insights into the reproductive cycle of two Great Scallop populations in Brittany (France) using a DEB modelling approach

The present study aimed to improve understanding of the environmental conditions influencing the reproductive cycle of the great scallop Pecten maximus in two locations in Brittany (France). We also evaluated potential consequences of future climate change for reproductive success in each site.We simulated reproductive traits (spawning occurrences and synchronicity among individuals) of P. maximus, using an existing Dynamic Energy Budget (DEB) model. To validate and test the model, we used biological and environmental datasets available for the Bay of Brest (West Brittany, France) between 1998 and 2003. We also applied the scallop DEB model in the Bay of Saint-Brieuc (North Brittany, France) for the same period (1998–2003) to compare the reproductive cycle in different environmental conditions. In order to accurately model the P. maximus reproductive cycle we improved the scallop DEB model in two ways: through (1) energy acquisition, by incorporating microphytobenthos as a new food source; and (2) the reproductive process, by adding a new state variable dedicated to the gamete production. Finally, we explored the effects of two contrasting IPCC climate scenarios (RCP2.6 and RCP8.5) on the reproductive cycle of P. maximus in these two areas at the 2100 horizon.In the Bay of Brest, the simulated reproductive cycle was in agreement with field observations. The model reproduced three main spawning events every year (between May and September) and asynchronicity in the timing of spawning between individuals. In the Bay of Saint-Brieuc, only two summer spawning events (in July and August) were simulated, with a higher synchronicity between individuals. Environmental conditions (temperature and food sources) were sufficient to explain this well-known geographic difference in the reproductive strategy of P. maximus. Regarding the forecasting approach, the model showed that, under a warm scenario (RCP8.5), autumnal spawning would be enhanced at the 2100 horizon with an increase of seawater temperature in the Bay of Brest, whatever the food source conditions. In the Bay of Saint-Brieuc, warmer temperatures may impact reproductive phenology through an earlier onset of spawning by 20 to 44 days depending on the year.

Tip60 complex promotes expression of a differentiation factor to regulate germline differentiation in female Drosophila.

Germline stem cells (GSCs) self-renew and differentiate to sustain a continuous production of gametes. In the female Drosophila germ line, two differentiation factors, bag of marbles (bam) and benign gonial cell neoplasm (bgcn), work in concert in the stem cell daughter to promote the generation of eggs. In GSCs, bam transcription is repressed by signaling from the niche and is activated in stem cell daughters. In contrast, bgcn is transcribed in both the GSCs and stem cell daughters, but little is known about how bgcn is transcriptionally modulated. Here we find that the conserved protein Nipped-A acts through the Tat interactive protein 60-kDa (Tip60) histone acetyl transferase complex in the germ line to promote GSC daughter differentiation. We find that Nipped-A is required for efficient exit from the gap phase 2 (G2) of cell cycle of the GSC daughter and for expression of a differentiation factor, bgcn. Loss of Nipped-A results in accumulation of GSC daughters. Forced expression of bgcn in Nipped-A germline-depleted ovaries rescues this differentiation defect. Together, our results indicate that Tip60 complex coordinates cell cycle progression and expression of bgcn to help drive GSC daughters toward a differentiation program.
