Echo Time Dependence of BOLD fMRI Studies of the Piriform Cortex

In functional magnetic resonance imaging (fMRI) studies, brain areas that are commonly associated with the processing of olfactory stimuli, i.e., piriform cortex and orbitofrontal cortex, are often obscured by susceptibility-induced signal loss. The authors hypothesized that using a short echo time (TE) should not only reduce susceptibility artifacts but also increase the overall signal-to-noise ratio and allow to retrieve a blood oxygenation level-dependent (BOLD) signal in regions normally affected by these artifacts. Two sequences with TEs of 60 and 32 ms were compared using a 1.5-T MRI scanner: in a standard motor paradigm, activations of the contralateral motor cortex were measured. In an olfactory stimulation paradigm, activations in piriform cortex were compared. Reducing TE from 60 to 32 ms reduced the observed signal intensity changes in the motor paradigm by 51%. Concomitant to this, geometric distortions and signal dropout artifacts were decreased at orbitofrontal and temporomesial brain areas in both paradigms. Contrary to the authors' expectations, the signal intensity changes in the piriform cortex were also reduced by 48% in the olfactory paradigm. Moreover, piriform cortex activation was detected in less subjects at TE = 32 ms than at TE = 60 ms. Changes in cortical activation were significant in the right, but not in the left piriform cortex. Although a shorter TE reduces signal dropouts due to susceptibility artifacts, this shorter TE is not sufficient to recover the BOLD signal from regions affected by susceptibility artifacts such as the piriform cortex. Thus, reducing the TE to the T2* of the investigated region is not an effective approach to improve the results of olfactory fMRI studies.

Metabotropic glutamate receptors mGluR4 and mGluR8 regulate transmission in the lateral olfactory tract–piriform cortex synapse

The piriform cortex (PC) is the primary terminal zone of projections from the olfactory bulb, termed the lateral olfactory tract (LOT). The PC plays a critical role in processing of olfactory stimuli and is also a highly seizure prone area thought to be involved in some forms of temporal lobe epilepsy. Pharmacological and immunohistochemical studies provide evidence for the localization of various metabotropic glutamate receptors (GluRs) in the PC. We employed whole-cell patch clamp recordings from PC pyramidal cells to determine the roles of group III mGluRs in modulating synaptic transmission at the LOT–PC synapse. The group III mGluR agonist, l-AP4, induced a concentration-dependent inhibition of synaptic transmission at the LOT–PC synapse at concentrations that activate mGluR4 and mGluR8, but not mGluR7 or other mGluR subtypes (EC 50 = 473 nM). In addition, the selective mGluR8 agonist, DCPG (300 nM), also suppressed synaptic transmission at the LOT synapse. Furthermore, the inhibitory actions of l-AP4 and Z-cyclopentyl-AP4, a selective mGluR4 agonist, were potentiated by the mGluR4 positive allosteric modulator, PHCCC (30 μM). The high potency of l-AP4, combined with the observed effects of DCPG and PHCCC, suggests that both mGluR4 and mGluR8 play a role in the l-AP4-induced inhibition of synaptic transmission at the LOT–PC synapse.

Activation of olfactory and trigeminal cortical areas following stimulation of the nasal mucosa with low concentrations of S(-)-nicotine vapor--an fMRI study on chemosensory perception.

Applied to the nasal mucosa in low concentrations, nicotine vapor evokes odorous sensations (mediated by the olfactory system) whereas at higher concentrations nicotine vapor additionally produces burning and stinging sensations in the nose (mediated by the trigeminal system). The objective of this study was to determine whether intranasal stimulation with suprathreshold concentrations of S(-)-nicotine vapor causes brain activation in olfactory cortical areas or if trigeminal cortical areas are also activated. Individual olfactory detection thresholds for S(-)-nicotine were determined in 19 healthy occasional smokers using a computer-controlled air-dilution olfactometer. Functional magnetic resonance images were acquired using a 1.5T MR scanner with applications of nicotine in concentrations at or just above the individual's olfactory detection threshold. Subjects reliably perceived the stimuli as being odorous. Accordingly, activation of brain areas known to be involved in processing of olfactory stimuli was identified. Although most of the subjects never or only rarely observed a burning or painful sensation in the nose, brain areas associated with the processing of painful stimuli were activated in all subjects. This indicates that the olfactory and trigeminal systems are activated during perception of nicotine and it is not possible to completely separate olfactory from trigeminal effects by lowering the concentration of the applied nicotine. In conclusion, even at low concentrations that do not consistently lead to painful sensations, intranasally applied nicotine activates both the olfactory and the trigeminal system.

Inhibiting dopamine reuptake blocks the induction of long-term potentiation and depression in the lateral entorhinal cortex of awake rats

Synaptic plasticity in olfactory inputs to the lateral entorhinal cortex may result in lasting changes in the processing of olfactory stimuli. Changes in dopaminergic tone can have strong effects on basal evoked synaptic responses in the superficial layers of the entorhinal cortex, and the current study investigated whether dopamine may modulate the induction of long-term potentiation (LTP) and depression (LTD) in piriform cortex inputs to layer II of the lateral entorhinal cortex in awake rats. Groups of animals were pretreated with either saline or the selective dopamine reuptake inhibitor GBR12909 prior to low or high frequency stimulation to induce LTD or LTP. In saline-treated groups, synaptic responses were potentiated to 122.4+/-6.4% of baseline levels following LTP induction, and were reduced to 84.5+/-4.9% following induction of LTD. Changes in synaptic responses were maintained for up to 60min and returned to baseline levels within 24h. In contrast, induction of both LTP and LTD was blocked in rats pretreated with GBR12909. Dopaminergic suppression of synaptic plasticity in the entorhinal cortex may serve to restrain activity-dependent plasticity during reward-relevant behavioral states or during processing of novel stimuli.

From taste to touch: sensory signaling in model organisms

The ability to perceive our environment has fascinated scientists and philosophers for thousands of years. According to some interpretations of Aristotle’s writings, the sense organs are able to perceive the environment by being transformed so that they are more like the object they are detecting. An eye can see a specific color by taking on that color; an animal can perceive warmth by becoming warm, and so forth. In recent years, our understanding of the inner workings of the Aristotelian senses of sight, smell, taste, touch, and hearing has gone through a revolution, and many of the key insights have emerged through analyses of model organisms. The reviews in this special issue of Pflugers Archiv highlight the progress achieved through work in genetically tractable metazoan animals ranging from worms to flies and mice. Although the various sensory modalities operate through a diversity of activation mechanisms and timescales, a recurring theme is that members of the transient receptor potential (TRP) superfamily of cation channels function broadly in sensory physiology and throughout animal phylogeny. The most ancient among the five classical senses are touch, taste, and smell. These three sensory modalities are present in the worm, Caenorhabditis elegans, and all metazoan organisms analyzed. In fact, the first mutations to affect touch were identified in the worm, and a review by Alexander Bounoutas and Martin Chalfie describes the current state of knowledge of the cells and proteins that function in the response to gentle touch. The fruitfly, Drosophila melanogaster, has also been invaluable for dissecting the genetics of gentle and noxious touch, and Maurice Kernan reviews the functions and development of the various types of sense organs in flies and the proteins essential for mechanotransduction. Among the players identified is no mechanoreceptor potential C (NOMPC), which is required for light touch and is the first TRP channel shown to function in mechanotransduction. The other of the primordial sensory modalities, taste and smell are important not only for identifying nutrient-rich sources but for avoiding noxious chemicals and predators. In many animals, the pheromone response contributes to mate selection, mating behavior, aggressiveness, and other behaviors. In worms, chemosensation impacts on a developmental program as the animals arrest at a nonreproductive, nonfeeding stage in environments with a paucity of food or food-derived odors. The perception of chemical cues in the environment can also impact lifespan, at least in invertebrates. Four reviews in the current special issue focus on the responses to volatile (olfactory) and nonvolatile (gustatory) chemical stimuli. These include a contribution by Piala Sengupta, outlining the cells, circuitry, and molecules involved in chemically induced behaviors in worms. Michelle Ebbs and Hubert Amrien provide an overview of the anatomical sites and receptors required for the taste and pheromone responses in Drosophila. In addition, they review fly taste transduction and the behaviors controlled by nonvolatile chemicals. As outlined in a separate review by Dean Smith, Drosophila has also provided important insights into the detection and processing of olfactory stimuli. While most pheromones are nonvolatile, at least one volatile chemical serves as a pheromone in flies, and this latter review describes the requirement for this pheromone for mating and for aggressive behavior. Functional neuroanatomy of the rhinophore of Archidoris pseudoargus

For sea slugs, chemosensory information represents an important sensory modality, because optical and acoustical information are limited. In the present study, we focussed on the neuroanatomy of the rhinophores and processing of olfactory stimuli in the rhinophore ganglion of Archidoris pseudoargus, belonging to the order of Nudibranchia in the subclass of Opisthobranchia. Histological techniques, fluorescent markers, and immunohistochemistry were used to analyse neuroanatomical features of the rhinophore. A large ganglion and a prominent central lymphatic channel are surrounded by longitudinal muscles. Many serotonin-immunoreactive (IR) processes were found around the centre and between the ganglion and the highly folded lobes of the rhinophore, but serotonin-IR cell bodies were absent inside the rhinophore. In contrast to the conditions recently found in Aplysia punctata, we found no evidence for the presence of olfactory glomeruli within the rhinophore. Using calcium-imaging techniques with Fura II as a calcium indicator, we found differential calcium responses in various regions within the ganglion to stimulation of the rhinophore with different amino acids. The lack of glomeruli in the rhinophores induces functional questions about processing of chemical information in the rhinophore.

Modulation of olfactory bulb tyrosine hydroxylase and catecholamine transporter mRNA by estrogen

Since estrogen exerts wide ranging effects within the central nervous system, it is important to investigate the sites and actions of this gonadal steroid hormone at extra-hypothalamic locations. In the present report, the effects of estrogen upon catecholaminergic function within the olfactory bulb were examined. To assess the role of estrogen at this site, ovariectomized mice received either no further hormonal treatment or were treated with estrogen, the anti-estrogen, tamoxifen, or a combination of estrogen and tamoxifen as administered in a 21-day release pellet. At 14 days post-hormonal treatment, the olfactory bulbs were assayed for mRNA levels of tyrosine hydroxylase, dopamine transporter and norepinephrine transporter using competitive-PCR. Tyrosine hydroxylase mRNA levels in either estrogen or estrogen+tamoxifen treated females were significantly decreased compared with non-hormonally treated controls. In addition, tyrosine hydroxylase mRNA levels of tamoxifen-treated mice were significantly greater than that of estrogen-treated mice. Dopamine transporter mRNA levels of tamoxifen-treated females were significantly greater than that of non-hormonally treated controls and estrogen treated mice. The combination of estrogen+tamoxifen significantly increased dopamine transporter mRNA levels compared to that of estrogen treated mice. No overall statistically significant differences in norepinephrine transporter mRNA levels were obtained among the four treatment groups. The data demonstrate that estrogen can exert significant modulatory effects upon olfactory bulb catecholaminergic function. Therefore, events which alter estrogen levels (menstrual/estrogen cycle, pregnancy/lactation, menopause, tamoxifen treatment) can modulate olfactory bulb catecholaminergic functions which may be involved with the detection and processing of olfactory stimuli.

Morphology of interneurons in the procerebrum of the snailHelix aspersa

Terrestrial snails have a highly developed sense of olfaction. Because the procerebrum has a large number of cells and is located at the entry site of the olfactory nerve into the brain, the structure is thought to have a significant role in the processing of olfactory stimuli. The morphology of the procerebral neurons in the snail Helix aspersa was investigated through intracellular injections of biocytin. No formal categorization of neuronal types was possible, but some cells were seen to have neurites entirely intrinsic to the procerebrum, whereas others had both intrinsic and extrinsic arborizations, and still others had only extrinsic arborizations. These interneurons were previously thought to have arborizations restricted to the procerebral lobe. We demonstrated the extent of the neurite projections outside of the procerebral lobe by making focal injections of biocytin or Neurobiotin into various regions of the cerebral ganglion. This technique revealed subsets of cells that send neurites not only in the ipsilateral ganglion but also through the cerebral commissure into the contralateral cerebral ganglion. Our results demonstrate not only that the procerebral cell population is heterogeneous but also that the procerebrum interacts more directly with the rest of the central nervous system than was formerly believed.

Simultaneous monitoring of intracerebral release and behavior: endogenous vasopressin improves social recognition.

We previously reported that direct osmotic stimulation of the supraoptic nucleus (SON) of rats via a microdialysis probe produces a robust 'rebound' release of endogenous vasopressin (AVP) into the extracellular fluid of this hypothalamic nucleus. We now demonstrate in a combined microdialysis and push-pull perfusion study that this intranuclear release is accompanied by increased AVP release in the septum. Simultaneous monitoring of intranuclear release and behavioral performance in the same animal indicated a significant correlation between the amount of endogenously released AVP and improved social memory based on the olfactory discriminative capacities of adult male rats. This memory improvement was partially blocked by local administration of a AVP V1 receptor antagonist either into the SON or septum. These findings indicate that direct osmotic stimulation of the supraoptic nucleus, which increases intracerebral vasopressin release, improves the acquisition and/or processing of olfactory stimuli. Thus, the endogenous neuropeptide fulfills one of the major criteria for being causally involved as a neurotransmitter/neuromodulator in behavioral performance.

A neural network for processing olfactory-like stimuli

Several critical issues associated with the processing of olfactory stimuli in animals (but focusing on insects) are discussed with a view to designing a neural network which can process olfactory stimuli. This leads to the construction of a neural network that can learn and identify the quality (direction cosines) of an input vector or extract information from a sequence of correlated input vectors, where the latter corresponds to sampling a time varying olfactory stimulus (or other generically similar pattern recognition problems). The network is constructed around a discrete time content-addressable memory (CAM) module which basically satisfies the Hopfield equations with the addition of a unit time delay feedback. This modification improves the convergence properties of the network and is used to control a switch which activates the learning or template formation process when the input is "unknown". The network dynamics are embedded within a sniff cycle which includes a larger time delay (i.e. an integer ts greater than 1) that is also used to control the template formation switch. In addition, this time delay is used to modify the input into the CAM module so that the more dominant of two mingling odors or an odor increasing against a background of odors is more readily identified. The performance of the network is evaluated using Monte Carlo simulations and numerical results are presented.

Effects of fluprazine hydrochloride on conspecific odor preferences in rats

The effect of Fluprazine Hydrochloride (DU 27716) on preference for conspecific male, estrous female and food odors was examined in male rats utilizing a two-compartment choice apparatus. Treatment with 8.0 mg/kg Fluprazine enhanced the preference of males for male odors but had no effect on preference for either estrous female or food odors. The drug-induced enhancement of male odor preference is consistant with the suggestion that Fluprazine interferes in some way with the processing of olfactory stimuli which normally precede offensive attack. The failure of the drug to alter the preference of males for estrous female odors suggests that the increased sniffing of estrous females noted during social testing may be secondary to other sources of conspecific stimulation or may reflect a highly transitory effect on olfactory processes. These results suggest that the suppressive effects of Fluprazine on intermale aggression and copulation are mediated by somewhat distinct mechanisms.
