Use of Mucilage from Opuntia ficus-indica in the Manufacture of Probiotic Cream Cheese

Cream cheese is a type of fresh cheese with a thin consistency with great potential for adding probiotics. However, artificial thickeners have been used in its production, decreasing consumer satisfaction. This study suggests natural mucilage, specifically from the Cactaceae Opuntia ficus-indica, as a replacement for artificial thickeners due to its thick gelatinous properties. This study evaluated different cream cheese formulations by adding varying concentrations of Opuntia ficus-indica mucilage and the probiotic Lactobacillus acidophilus (L. acidophilus). Four formulations were created: formulation C (control, without mucilage), F1 (containing 1 mL/kg mucilage), F2 (2 mL/kg), and F3 (3 mL/kg mucilage). The physicochemical characteristics (pH, 4.90–5.57; 0.15–0.20% acidity; 1.78–2.42% protein; 29.98–30.88% fat; 38.27–41.63% moisture; and 1.25–1.63% ash) and microbiological analysis met the quality standards required by Brazilian legislation, and the cream cheese showed probiotic potential, with L. acidophilus counts above 108 CFU/mL within four weeks of storage. Regarding sensory evaluation, the texture received one of the highest scores (7.89), followed by aroma (7.11). Therefore, the Cactaceae mucilage has proven to be a viable alternative to replace artificial thickeners in cream cheese, making it an excellent option for probiotic supplementation.

Biosynthesis of Selenium and Copper Oxide Nanoparticles using Probiotic Bacteria against Food Spoilage Microorganisms

The study’s aim is to synthesize selenium (SeNPs) and copper oxide (CuONPs) nanoparticles using probiotic Lactobacillus acidophilus as a reducing agent. The biosynthesized probiotic Lactobacillus acidophilus selenium and copper oxide nanoparticles were known by color change. The characterization of SeNPs and CuoNPs was completed by Ultraviolet-visible (UV-VIS) spectroscopy, Field Emission Scanning electron microscope (FESEM), Atomic force microscope (AFM), X-Ray diffraction analysis (XRD) and Fourier transform infrared spectroscopy (FTIR). These tests are utilized for detecting stability, morphology, size, crystalline nature and functional groups on nanoparticles prepared surface. Outcomes revealed appearance of the brick-red and green color, representing a specific color of selenium and copper oxide nanoparticles. It was also disclosed that UV-VIS spectroscopy indicated band absorbance at 236 and 264 nanometer of intense surface Plasmon resonance, manifesting the formation and stability of prepared SeNPs and CuONPs. The FESEM image displayed mulit-shapes between spherical and vaulted for selenium and copper oxide nanosized. XRD at 2 theta revealed crystalline selenium and copper oxide nanoparticles where the average size is 75.52-153.22 nm. FTIR revealed the presence of functional groups of the supernatant that act as stabilizing and reducing agents. The antibacterial activity of synthesized nanoparticles was studied against five different bacteria causing food spoilage. As a result of this study, selenium nanoparticle and copper nanoparticles were biosynthized successfully in less time and easy consuming way by green synthesis of the supernatant probiotic Lactobacillus acidophilus.

Probiotics and magnesium orotate for the treatment of major depressive disorder: a randomised double blind controlled trial

Following on from our pilot studies, this study aimed to test the efficacy of a combination of probiotics (Lactobacillus acidophilus, Bifidobacterium bifidum, Streptococcus thermophilus), magnesium orotate and coenzyme 10 for the treatment of major depressive disorder (MDD) through a double-blind placebo controlled clinical trial. The participants were 120 adults diagnosed with MDD randomised to daily oral administration, over 8 weeks, of either the intervention or placebo, with a 16-week follow-up period. Intent-to-treat analysis found a significantly lower frequency of the presence of a major depressive episode in the intervention group compared with placebo at the end of the 8-week treatment phase, with no difference between the two conditions at 8-week follow-up. Both the categorical and continuous measure of depressive symptoms showed a significant difference between the two conditions at 4 weeks, but not 8 and 16 weeks. The secondary end-point was demonstrated with an overall reduction in self-rated symptoms of anxiety and stress in the active treatment group compared with placebo. These findings suggest that the combination of probiotics, magnesium orotate and coenzyme 10 may be an effective treatment of MDD over an 8-week period.

Antimicrobial activities of Alchornea cordifolia leaf extract on Candida albican and some selected Probiotics

The quest for novel and effective antimicrobial agents has become increasingly urgent because of the increasing threat of drug-resistant microbial pathogens. Alchornea cordifolia, a plant species widely used in traditional medicine, has been reported to possess antimicrobial properties. However, its antifungal activity and interactions with beneficial probiotic microorganisms remain poorly understood. This study aimed to investigate the antifungal efficacy of Alchornea cordifolia extracts and their bioactive components against Candida albicans and explore their interactions with probiotic microorganisms, Lactobacillus acidophilus and Sacchromyces bourdii, which are often used for control. By elucidating the antimicrobial potential and probiotic interactions of Alchornea cordifolia, this study aims to contribute to the development of innovative and sustainable solutions for the prevention and treatment of Candida albicans infections while also promoting a balanced microbiota. The crude extract was subjected to phytochemical screening. The bioactive components were isolated and purified via column chromatography and thin layer chromatography, whereas FTIR was used to analyse the functional groups present in the extract and isolated components. The extract and isolated components were further subjected to antimicrobial, minimum inhibitory concentration (MIC) and minimum fungicidal/bactericidal concentration (MFC/MBC) assays against C. albicans, S. bourdii and L. acidophilus via the agar diffusion and broth macrodilution technique. Phytochemical analysis revealed the presence of various bioactive components, including tannins, flavonoids, phenols, saponins, alkaloids, anthocyanins, cardiac glycosides, cyanogenic glycosides, oxalates, carotene and sterols. Four spots with Rf values between 0.73 and 0.75, 0.83 and 0.73, 0.87 and 0.98 and 0.32 and 0.23 assigned as components A, B, C and D, respectively, were isolated after elution with a gradient of solvents. The FTIR results of the crude extract revealed functional groups consistent with those of secondary metabolites, such as hydroxyl groups (O-H, peaks at 3812–3500 cm-1), amines (N–H stretching peaks at 3431 cm-1), alkenes (C-H stretching CH3 CH2, CH peaks at 3175–2800 cm-1), and C=C peaks at 1623 cm-1. These functional groups were identified during preliminary tests. The crude extract and components A, B, and C exhibited similar activities against Candida albicans, with inhibition zones ranging from 21 mm to 32 mm. Component C had the highest activity, with an inhibition zone of 32 mm. Component D showed moderate activity, with an inhibition zone of 26 mm. Antimicrobial activity against Sacromyces boulardii and Lactobacillus acidophilus varied among the components, with Component C showing the highest activity against Sacromyces boulardii. There was some selectivity for the pathogen rated as the crude extract ˃C ˃B = A ˃D. The crude extract and components A, B, and C had MICs of 100 µg/ml against Candida albicans. Component C had an MIC of 1000 µg/ml against Sacromyces boulardii. Component D had an MIC of 10 µg/ml against Candida albicans. The methanol crude extract and its components have shown promising antimicrobial activity against Candida albicans, making it a potential natural solution for candidiasis management. Formulation optimization of a probiotic low-calorie chocolate milk and investigating its qualitative properties during storage

Due to concern regarding the consumption of high amount of sugar in diet and role of diet in combating overweigh and related disease, the aim of present study was to optimize a reduced calorie probiotic chocolate milk formula with suitable physicochemical properties. The formula comprising inulin, stevia (Stevia rebaudiana Bertoni), chia (Salvia hispanica L.) seed gum (CSG), and whey protein concentrate (WPC) which optimized using Box-Behnken design (BBD) and then enriched with an encapsulated probiotic strain Lactobacillus acidophilus (DSM1643). The independent variables included inulin (2–8%), CSG (0.1–0.5 %), stevia (50–100 % replacement of sugar), and WPC (1–3%). The dependent variables were selected as viscosity, average particle size, sedimentation percentage, and general acceptance. Optimization done toward achieving the highest viscosity and general acceptance and the lowest sedimentation percentage and average particle size. The optimal conditions were found to be 7.99 % inulin, 70 % stevia, 0.34 % CSG, and 1 % WPC. Under these conditions, the viscosity, sedimentation percentage, average particle size, and general acceptance of the product were equal to 40.69 mPa s, 2.2 %, 434.221 nm, and 5.1, respectively. Next, the chocolate milk was enriched with at 109 CFU/g probiotic bacteria and evaluated. The probiotic strain was resistant to simulated gastrointestinal conditions and under this condition the free bacterial cells count declined by 8 logCFU/g while the encapsulated cells decreased approximate 3 logCFU/g. The bacteria count did not undergo a significant change until the 5th day of storage. The results showed that the inulin, stevia, CSG, and WPC at optimal concentrations and encapsulated probiotic bacteria could be simultaneously applied to produce a product with good properties. This formula could be considered as a new product with health improving properties, low calorie which is suitable for people suffering from diabetes and obesity.

Cassava (Manihot esculenta) Brazilian cultivars have different chemical compositions, present prebiotic potential, and beneficial effects on the colonic microbiota of celiac individuals

The purpose of this study was to investigate the potential prebiotic properties of cassava cultivars from Northeast [Doce mel and Ourinho (OUR)] and South [Baiana, and IPR-Upira (UPI)] of Brazil in in vitro fermentation systems. The cultivars were evaluated for their chemical composition, and, then, two cultivars were selected (OUR and UPI) and subjected to in vitro gastrointestinal digestion to assess the effects on probiotics Lacticaseibacillus casei, Lactobacillus acidophilus, and Bifidobacterium animalis growth, metabolic activity, and prebiotic activity scores. Finally, the impact of cassava cultivars on the fecal microbiota of celiac individuals was evaluated using the 16S rRNA gene. Cassava cultivars have variable amounts of fiber, resistant starch, fructooligosaccharides (FOS), organic acids, phenolic compounds, and sugars, with OUR and UPI cultivars standing out. OUR and UPI cultivars contributed to the increase in the proliferation rates of L. casei (0.04–0.19), L. acidophilus (0.34–0.27), and B. animalis (0.10–0.03), resulting in more significant effects than FOS, an established prebiotic compound. Also, the positive scores of prebiotic activities with probiotic strains indicate OUR and UPI’s ability to stimulate beneficial bacteria while limiting enteric competitors selectively. In addition, OUR and UPI promoted increased relative abundance of Bifidobacteriaceae, Enterococcaceae, and Lactobacillaceae in the fecal microbiota of celiac individuals while decreased Lachnospirales, Bacteroidales, and Oscillospirales. The results show that cassava cultivars caused beneficial changes in the composition and metabolic activity of the human intestinal microbiota of celiacs. OUR and UPI cultivars from the Northeast and South of Brazil could be considered potential prebiotic ingredients for use in the formulation of functional foods and dietary supplements.

Antagonistic and antibacterial activity of <i>Lactobacillus rhamnosus</i> HN001 and <i>Lactobacillus acidophilus</i> La-14 included in the oral probiotic

BACKGROUND: A microbial imbalance in the vaginal biotope leads to the development of vaginal infections and is associated with urinary tract infections. The ineffectiveness of the therapy and the frequent recurrence of these infections are important reasons for searching for alternative treatment strategies. A progressive solution to this issue has been the use of oral probiotics with a targeted effect on the intestinal and vaginal microflora. AIM: The aim of this study was to evaluate the antagonistic and antibacterial activity of the probiotic strains Lactobacillus rhamnosus HN001 and Lactobacillus acidophilus La-14, which are the components of the oral probiotic, against opportunistic microorganisms of intestinal origin, while assessing the sensitivity of these strains to antimicrobial drugs. MATERIALS AND METHODS: The probiotic strains Lactobacillus rhamnosus HN001 and Lactobacillus acidophilus La-14 included in the oral probiotic Duogynal® were cultivated and the grown colonies were identified using the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry method (Bruker Daltonics GmbH Co., Germany). To study the antagonism of the probiotic strains of lactobacilli and opportunistic pathogenic microorganisms, the following 11 clinical isolates were collected: Escherichia coli (two isolates), Klebsiella pneumoniae, Klebsiella oxytoca, Enterobacter asburiae, Proteus mirabilis, Citrobacter freundii, Staphylococcus aureus, Enterococcus faecalis, Streptococcus agalactiae, and Candida albicans. To assess the sensitivity of the probiotic strains to antimicrobial drugs, the Gram Positive AST panel (Autobio Diagnostics Co., Ltd, China) was used. RESULTS: When the microorganisms were co-cultivated with the probiotic lactobacilli (Lactobacillus rhamnosus HN001 and Lactobacillus acidophilus La-14), we noted a decrease in pH and the death of opportunistic microorganisms such as Escherichia coli, Klebsiella pneumoniae, Klebsiella oxytoca, Enterobacter asburiae, Proteus mirabilis, Citrobacter freundii, Staphylococcus aureus, Enterococcus faecalis, and Streptococcus agalactiae, as well as yeast-like fungi Candida albicans. When studying antibiotic resistance, it was found that the both strains of lactobacilli were resistant to ampicillin, vancomycin, daptomycin, clindamycin, linezolid, moxifloxacin, nitrofurantoin, oxacillin, oritavancin, penicillin, rifampin, teicoplanin, tigecycline, trimethoprim/sulfamethoxazole, cefoxitin, ceftarolin, and ciprofloxacin. CONCLUSIONS: The oral probiotic, which contains a combination of two lactobacilli (Lactobacillus rhamnosus HN001 and Lactobacillus acidophilus La-14), has pronounced antagonistic activity against opportunistic microorganisms. These strains are resistant to most antibacterial drugs used in widespread clinical practice, which may be the basis for recommending the use of this probiotic drug together with prescribed antibiotics for prevention of vaginal or intestinal dysbiosis.

Effect of live and inactivated probiotic strains of <i>Lactobacillus acidophilus</i> and <i>Bifidobacterium animalis</i> subsp. <i>lactis</i> on myocardial infarction size in rats with systemic inflammatory response syndrome

Within the concept of a heart-gut axis, new works are emerging to support the efficacy of probiotic strains to increase myocardial resistance to ischemia-reperfusion injury (IRI) in comorbidity. The question remains open whether the presence of live probiotic bacteria is a necessary condition for the realization of their cardioprotective effect. The aim of this work was to determine the manifestation of cardio-protective effect of living and pasteurized probiotic strains Lactobacillus acidophilus (LA-5) and Bifidobacterium animalis subsp. lactis (BB-12) in rats with systemic inflammatory response syndrome (SIRS). Myocardial resistance to IRI was assessed using an in vivo model of left coronary artery occlusion-reperfusion. Experiments were performed on male Wistar rats with improved conventional status with visceral obesity, chemically induced colitis and antibiotic-induced dysbiosis, which together provided the formation of (SIRS) against the background of oral administration of live and inactivated probiotic bacteria. Myocardial resistance to ischemia-reperfusion injury was assessed using the technique of left coronary artery occlusion in vivo. The infarct size in the group with simulated SIRS was significantly higher than in the control group 43% (39; 44) and 31% (28; 35), (p 0.05). In the SIRS group with the introduction of inactivated probiotic bacteria, the infarct size 45% (37; 48) did not differ from the SIRS group and was significantly higher than in the control (p 0.05). At the same time, the size of the infarction in the group with the introduction of live probiotics did not differ from that in the control group and amounted to 32% (28; 37). There are specific features of the action of live and inactivated probiotic microorganisms with preservation of cardioprotective effect when using live lacto- and bifidobacteria in animals with SIRS.

Proliferation of probiotics and antioxidant effects of functional oligosaccharides added in fermented dairy product

To obtain functional fermented dairy products with strong probiotic proliferation and high antioxidant ability, the proliferation ability of oligosaccharides on probiotics and their DPPH radical scavenging ability were studied. Key functional oligosaccharides (galactooligosaccharide and galactomannan oligosaccharide) and probiotics (Bifidobacterium bifidum and Lactobacillus acidophilus) were selected. The orthogonal experimental design of L9(34) was employed to study the optimal process, which was adding 5 g/L galactomannan oligosaccharide and 5 g/L galactooligosaccharide in reconstituted milk, inoculating 1% Streptococcus thermophilus, 1% Lactobacillus bulgaricus and 1% probiotics (B. bifidum and L. acidophilus = 2:1) and fermenting for 6 h. This work is important for preparing functional fermented dairy products with a high content of probiotics and strong antioxidant capacity.

Improving the physicochemical and sensory properties and volatile profile of goat ricotta cream with Limosilactobacillus mucosae CNPC007 supplementation

This study assessed the effects of the autochthonous strain Limosilactobacillus mucosae CNPC007 on the techno-functional characteristics, physicochemical properties, volatile profile, and sensory aspects of goat ricotta cream during refrigerated storage. Three cheese formulations were tested: CRC (without probiotic), RCLM (with L. mucosae CNPC007), and RCLA (with commercial probiotic Lactobacillus acidophilus La-5). L. mucosae CNPC007 influenced color, adhesiveness, lactose content, acidity, and proteolysis depth index in goat ricotta cream. RCLA and RCLM maintained a probiotic viable cell count >6 log CFU g−1. RCLM had the highest proteolysis depth index and free amino acid release. Seventeen distinct volatile compounds were identified in the formulations. Probiotic supplementation did not affect global acceptance or purchase intention. RCLA and RCLM were preferred over CRC in the ranking test and considered ideal for goat aroma in the JAR test. L. mucosae CNPC007 supplementation has the potential to improve the nutritional, functional, aromatic, and sensory aspects of goat ricotta cream.

Effect of Different Treatments on Chemical Components of Fermented Probiotic Millet Dried Mix

Aims: The purpose of this study was to develop fermented probiotic millet dried mix by incorporating a probiotic Lactobacillus acidophilus and to ascertain how different processes used in the product's development affected its chemical components. Study Design: Fermented probiotic millet dried mix was developed by fermentation of germinated finger millet flour and Reconstituted Skim Milk (RSM) with L. acidophilus. The impact of various treatments on moisture, crude protein, crude fiber, total ash, alcoholic acidity and phytate content were investigated. Place and Duration of Study: Department of Dairy Microbiology, Dairy Science College, Hebbal, Bengaluru, Karnataka, India between June 2023 and April 2024. Methodology: Moisture, crude protein, crude fiber, total ash, alcoholic acidity and phytate content were examined after various treatments such as soaking, germination, autoclaving, fermentation and drying of millet dried mix. Results: At 10% L. acidophilus inoculum level and a 1:1.5 millet to RSM ratio, a maximum viability of 9.52 log10 cfu/g was reported after 30 h of fermentation. After freeze drying of fermented probiotic millet mix with 2% Skim Milk Powder (SMP) as a cryoprotective agent, viability was retained to 9.50 log10 cfu/g. The optimized fermented probiotic millet dried mix had 4.76% moisture, 11.94% crude protein, 6.50% crude fiber, 4.74% total ash, 1.90% alcoholic acidity and 1.73 mol/kg of phytate. Conclusion: The resultant fermented probiotic millet dried mix was rich in nutrients and the probiotic L. acidophilus helps to improve gut health. Thus, the product had combined therapeutic and nutritional benefits of probiotics with millet.

The Probiotic Strain Lactobacillus acidophilus CL1285 Reduces Fat Deposition and Oxidative Stress and Increases Lifespan in Caenorhabditis elegans.

Caenorhabditis elegans was recently shown to be a powerful model for studying and identifying probiotics with specific functions. Lactobacillus acidophilus CL1285, Lacticaseibacillus casei LBC80R, and Lacticaseibacillus rhamnosus CLR2, which are three bacteria that were marketed by Bio-K+, were evaluated using the nematode C. elegans to study fat accumulation, lifespan, and resistance to oxidative stress. Although the general effects of probiotics in terms of protection against oxidative stress were highlighted, the CL1285 strain had an interesting and specific feature, namely its ability to prevent fat accumulation in nematodes; this effect was verified by both the Oil Red and Nile Red methods. This observed phenotype requires daf-16 and is affected by glucose levels. In addition, in a daf-16- and glucose-dependent manner, CL1285 extended the lifespan of C. elegans; this effect was unique to CL1285 and not found in the other L. acidophilus subtypes in this study. Our findings indicate that L. acidophilus CL1285 impacts fat/glucose metabolism in C. elegans and provides a basis to further study this probiotic, which could have potential health benefits in humans and/or in mammals.

Lactobacillus acidophilus KBL409 protects against kidney injury via improving mitochondrial function with chronic kidney disease.

Recent advances have led to greater recognition of the role of mitochondrial dysfunction in the pathogenesis of chronic kidney disease (CKD). There has been evidence that CKD is also associated with dysbiosis. Here, we aimed to evaluate whether probiotic supplements can have protective effects against kidney injury via improving mitochondrial function. An animal model of CKD was induced by feeding C57BL/6 mice a diet containing 0.2% adenine. KBL409, a strain of Lactobacillus acidophilus, was administered via oral gavage at a dose of 1 × 109CFU daily. To clarify the underlying mechanisms by which probiotics exert protective effects on mitochondria in CKD, primary mouse tubular epithelial cells stimulated with TGF-β and p-cresyl sulfate were administered with butyrate. In CKD mice, PGC-1α and AMPK, key mitochondrial energy metabolism regulators, were down-regulated. In addition, mitochondrial dynamics shifted toward fission, the number of fragmented cristae increased, and mitochondrial mass decreased. These alterations were restored by KBL409 administration. KBL409 supplementation also improved defects in fatty acid oxidation and glycolysis and restored the suppressed enzyme levels involved in TCA cycle. Accordingly, there was a concomitant improvement in mitochondrial respiration and ATP production assessed by mitochondrial function assay. These favorable effects of KBL409 on mitochondria ultimately decreased kidney fibrosis in CKD mice. In vitro analyses with butyrate recapitulated the findings of animal study. This study demonstrates that administration of the probiotic Lactobacillus acidophilus KBL409 protects against kidney injury via improving mitochondrial function.

Fermenting Acerola (Malpighia emarginata D.C.) and Guava (Psidium guayaba L.) Fruit Processing Co-Products with Probiotic Lactobacilli to Produce Novel Potentially Synbiotic Circular Ingredients.

This study evaluated the effects of acerola and guava fruit processing co-products fermented with probiotic Lactobacillus acidophilus LA-05 and Lacticaseibacillus paracasei L-10 on the abundance of different intestinal bacterial groups and microbial metabolic activity during 48 h of in vitro fecal fermentation. Digested fermented fruit co-products increased the relative abundance of beneficial bacterial groups while overall decreasing or maintaining the relative abundance of non-beneficial bacterial groups, suggesting selective stimulatory effects on beneficial bacterial intestinal populations. The fermented co-products stimulated microbial metabolic activity due to decreased pH, sugar consumption, short-chain fatty acid production, phenolic compound and metabolic profile alteration, and high antioxidant capacity during fecal fermentation. Acerola and guava co-products have high nutritional value and bioactive compounds whose fermentation with probiotics improves their potential functionalities. The results show that fermented fruit co-products could induce beneficial changes in the relative abundance of several bacterial groups as well as in the metabolic activity of the human intestinal microbiota. These results highlight their potential as novel and circular candidates for use as synbiotic ingredients.

Introduction: Radiation enteritis is damage to the small and large intestines caused by radiation therapy for regional cancer treatment. The incidence is around 90% in patients undergoing radiation in the pelvic and abdominal regions. This study aimed to report the effect of probiotics on radiation enteritis in endometrial cancer patients. Case presentation: A 57-year-old morbidly obese woman diagnosed with endometrial cancer underwent chemoradiation therapy. On the 21st out of 25 radiation sessions and the 2nd chemotherapy session, the patient visited the nutrition clinic at the Radiation Oncology Therapy Center with a complaint of diarrhea for the past 5 days. The diarrhea occurred approximately 10 times a day, with each bowel movement being approximately 50 ml and yellow in color. The patient was then treated with anti-diarrheal therapy, probiotics (Lactobacillus acidophilus, Bifidobacterium longum, and Streptococcus thermophilus) 2 times a day, zinc 20 mg once a day, and prescribed a low-fat and lactose-free diet. After monitoring on the 21st day, the frequency of liquid bowel movements decreased to 2 times a day, and the appetite improved. Conclusion: Patients with radiation enteritis who are given probiotics may experience a reduction in symptoms of acute diarrhea. Direct administration of probiotics increases beneficial bacteria in the digestive tract.

Cell-Free Culture Supernatant of Lactobacillus acidophilus AG01 and Bifidobacterium animalis subsp. lactis AG02 Reduces the Pathogenicity of NetB-Positive Clostridium perfringens in a Chicken Intestinal Epithelial Cell Line.

The worldwide reduction in the use of antibiotics in animal feed is fueling the need for alternatives for the prevention and control of poultry intestinal diseases such as necrotic enteritis (NE), which is caused by Clostridium perfringens. This is the first report on the use of an intestinal epithelial chicken cell line (CHIC-8E11) to study the pathogenic traits of C. perfringens and to investigate the mode of action of cell-free supernatants (CFS) from probiotic Lactobacillus acidophilus AG01 and Bifidobacterium animalis subsp. lactis AG02 in reducing the pathogenicity of C. perfringens. The cell adhesion, permeability and cytotoxicity were assessed under challenge with four C. perfringens strains isolated from broiler NE episodes of differing geographical origin (CP1-UK; CP10-Sweden; 25037-CP01 and CP22-USA). All the C. perfringens strains could adhere to the CHIC-8E11 cells, with varying affinity (0.05-0.48% adhesion across the strains). The CFS from one out of two strains (CP22) increased the cell permeability (+4.5-fold vs. the control, p < 0.01), as measured by the fluorescein isothiocyanate-dextran (FD4) content, with NetB toxin implicated in this effect. The CFS from all the strains was cytotoxic against the CHIC-8E11 cells in a dose- and strain-dependent manner (cytotoxicity 23-62% across the strains when dosed at 50 µL/mL, as assessed by the MTT cell viability assay). Pre-treatment of the cells with CFS from B. animalis subsp. lactis AG02 but not L. acidophilus AG01 reduced the cell adhesion of three out of four C. perfringens strains (by 77-85% vs. the control, p < 0.001) and reduced the negative effect of two NetB-positive strains on the cell permeability. The CFS of both probiotics alleviated the cytotoxicity of all the C. perfringens strains, which was dependent on the dose. The results confirm the suitability of the CHIC-8E11 cell line for the study of host-pathogen cell interactions in the context of NE caused by C. perfringens and reveal a beneficial mode of action of B. animalis subsp. lactis AG02 in reducing C. perfringens cell adhesion and, together with L. acidophilus AG01, in reducing C. perfringens cytotoxicity.

Properties of Potato Resistant Starch and Its Effect on the Growth of Probiotics

This study uses potato starch as raw material to prepare potato resistant starch (PRS3) using a pressure heating dual enzyme method and purifies it. The physicochemical indicators such as swelling power, solubility, freeze-thaw stability, transparency, oil absorption, milk absorption, and water holding capacity of purified PRS3 are measured. The growth and proliferation of probiotics (Lactobacillus acidophilus, Lactobacillus bulgaricus, Bifidobacterium, Streptococcus thermophilus, and Lactobacillus paracasei) under different carbon source concentrations of PRS3 medium, soluble starch medium, and glucose medium, as well as their effects on the growth process are studied. The authors also discusses the effects of glucose medium and PRS3 medium on salt and acid tolerance of probiotics. The results show that the freeze-thaw stability, solubility, and milk absorption capacity of PRS3 are stronger than those of original potato starch, but its oil absorption, water holding capacity, swelling capacity, and transparency are weaker than those of original potato starch. When the carbon source concentration is 2.0%, the proliferative effect of PRS3 on five bacteria is in descending order: Lactobacillus acidophilus, Lactobacillus bulgaricus, Bifidobacterium, Streptococcus thermophilus, and Lactobacillus paracasei. The ability of PRS3 medium to promote the proliferation of probiotics is stronger than that of glucose medium and soluble starch medium. It has a protective effect on probiotics in low pH environments and can improve their resistance to bile salts.
