Co‐release of inhibitory neurotransmitters in the RVLM

Sympathetic nerve activity (SNA) and blood pressure (BP) are largely determined by the level of excitability of presympathetic neurons (C1 neurons) in the rostral ventrolateral medulla (RVLM). GABAergic inhibition in the RVLM is the primary mechanism for depressor and sympathoinhibitory responses; however, the role of glycinergic inhibition in the control of RVLM excitability is unknown. There is evidence that both, GABA and glycine efficiently inhibit RVLM neurons, but functional evidence for their co‐release or the mechanism of co‐inhibition is lacking. In our study we used both male and female glycine transporter 2 Cre (GlyT2‐Cre) mice. Glycinergic neurons sending projections to the RVLM were identified with viral labeling. Targeted expression of channelrhodopsin 2 (ChR2) in GlyT2‐expressing neurons and whole‐cell recordings from presympathetic RVLM neurons were used to study the mechanism of glycine release. We found that under steady‐state conditions spontaneous inhibitory postsynaptic current (sIPSC) are primarily mediated by GABA. Photostimulation of glycinergic fibers expressing GlyT2 in the RVLM resulted in evoked IPSCs with both glycinergic and GABAergic components. The time course of synaptic signaling were identical for the two transmitters. Bath application of GABAA receptors antagonist bicuculline (10 μM) or glycine receptor antagonist strychnine (1 μM) revealed that ~40% of the evoked inhibitory currents are GABAergic. High frequency photostimulation (20 Hz) of co‐releasing inhibitory fibers produced a short‐term synaptic depression. Blockade of GlyT2 prevented the glycinergic component of the light evoked IPSCs. In this study we showed that GABA and glycine are co‐released in the RVLM. Our findings suggest that the two neurotransmitters may be functionally interchangeable or complement each other, and thus provide diverse mechanisms for maintaining inhibitory homeostasis.

Rostral ventrolateral medulla neuron activity is suppressed by Klotho and stimulated by FGF23 in newborn Wistar rats.

Hypertension often occurs in patients with chronic kidney disease (CKD). Considering the decrease in serum Klotho and increase in serum FGF23 levels in such patients, decreased Klotho and increased FGF23 levels were thought to be associated with hypertension. Presympathetic neurons at the rostral ventrolateral medulla (RVLM) contribute to sympathetic activity and regulation of blood pressure. Therefore, we hypothesized that Klotho would reduce the activities of RVLM neurons and FGF23 would stimulate them. Accordingly, this study examined the effects of Klotho and FGF23 on bulbospinal neurons in the RVLM. We used a brainstem-spinal cord preparation to record from RVLM presympathetic neurons and to evaluate the effects of Klotho and FGF23 on firing rate and membrane potentials of these neurons. Our results showed that Klotho-induced RVLM neuron hyperpolarization, while ouabain, a Na+/K+-ATPase inhibitor, suppressed the effects of Klotho on such neurons. Moreover, FGF23 induced RVLM neuron depolarization, while SU5402, an FGF23 receptor (FGFR1) antagonist, induced RVLM neuron hyperpolarization. Histological examinations revealed that Klotho, Na+/K+-ATPase, FGF23, and FGFR1 were present in RVLM neurons and that Klotho was localized in the same neurons as FGFR1. These results suggest that Klotho and FG23 regulate the activity of RVLM neurons. Klotho may reduce the activity of RVLM neurons via stimulating Na+/K+-ATPase on those neurons while FGF23 may activate those neurons via FGFR1.

Glycinergic neurotransmission in the rostral ventrolateral medulla controls the time course of baroreflex-mediated sympathoinhibition.

To maintain appropriate blood flow to various tissues of the body under a variety of physiological states, autonomic nervous system reflexes regulate regional sympathetic nerve activity and arterial blood pressure. Our data obtained in anaesthetized rats revealed that glycine released in the rostral ventrolateral medulla (RVLM) plays a critical role in maintaining arterial baroreflex sympathoinhibition. Manipulation of brainstem nuclei with known inputs to the RVLM (nucleus tractus solitarius and caudal VLM) unmasked tonic glycinergic inhibition in the RVLM. Whole-cell, patch clamp recordings demonstrate that both GABA and glycine inhibit RVLM neurons. Potentiation of neurotransmitter release from the active synaptic inputs in the RVLM produced saturation of GABAergic inhibition and emergence of glycinergic inhibition. Our data suggest that GABA controls threshold excitability, wherreas glycine increases the strength of inhibition under conditions of increased synaptic activity within the RVLM. The arterial baroreflex is a rapid negative-feedback system that compensates changes in blood pressure by adjusting the output of presympathetic neurons in the rostral ventrolateral medulla (RVLM). GABAergic projections from the caudal VLM (CVLM) provide a primary inhibitory input to presympathetic RVLM neurons. Although glycine-dependent regulation of RVLM neurons has been proposed, its role in determining RVLM excitability is ill-defined. The present study aimed to determine the physiological role of glycinergic neurotransmission in baroreflex function, identify the mechanisms for glycine release, and evaluate co-inhibition of RVLM neurons by GABA and glycine. Microinjection of the glycine receptor antagonist strychnine (4mm, 100nL) into the RVLM decreased the duration of baroreflex-mediated inhibition of renal sympathetic nerve activity (control= 12± 1min; RVLM-strychnine= 5.1± 1min), suggesting that RVLM glycine plays a critical role in regulating the time course of sympathoinhibition. Blockade of output from the nucleus tractus solitarius and/or disinhibition of the CVLM unmasked tonic glycinergic inhibition of the RVLM. To evaluate cellular mechanisms, RVLM neurons were retrogradely labelled (prior injection of pseudorabies virus PRV-152) and whole-cell, patch clamp recordings were obtained in brainstem slices. Under steady-state conditions GABAergic inhibition of RVLM neurons predominated and glycine contributed less than 25% of the overall inhibition. By contrast, stimulation of synaptic inputs in the RVLM decreased GABAergic inhibition to 53%; and increased glycinergic inhibition to 47%. Thus, under conditions of increased synaptic activity in the RVLM, glycinergic inhibition is recruited to strengthen sympathoinhibition.

NOVEL ROLE OF GLYCINE IN CONTROL OF SYMPATHETIC OUTFLOW

The arterial baroreflex is one of the essential negative‐feedback mechanisms that compensates rapid changes in blood pressure (BP) through inhibition of presympathetic neurons in the rostral ventrolateral medulla (RVLM). There is general consensus that GABAergic projections originating from the caudal VLM (CVLM) provide the primary inhibitory input that controls the excitability of presympathetic RVLM neurons and sympathetic output. However, glycine, another inhibitory neurotransmitter, is also present in the RVLM. In anesthetized rats, we found that RVLM glycine played a critical role in arterial baroreflex function by controlling the time course of baroreflex mediated inhibition of renal sympathetic nerve activity. Blockade of glycine receptors in the RVLM, following an increase in BP, increased recovery of renal sympathetic nerve activity to baseline levels (5.1 ± 1.0 vs 11.7 ± 1.3 min, N=6, P<0.01) in comparison to control. In addition, tonic glycinergic inhibition of the RVLM was unmasked following blockade of output from the nucleus tractus solitaries and/or during disinhibition of the CVLM. Our whole‐cell, patch‐clamp recordings were obtained from brainstem slices containing presympathetic neurons in the RVLM labeled with PRV‐152. We confirmed that in steady state conditions GABAergic inhibition of RVLM neurons predominated and glycine contributed less than 25% of overall inhibition. In contrast, activation of the network produced saturation of GABAergic inhibition and glycinergic inhibition became prominent. Medullary neuronal loss and alpha-synuclein burden in Multiple System Atrophy

Spinally projecting neurons in the rostral ventrolateral medulla (RVLM) are thought to play a critical role in the generation of vasomotor sympathetic tone and represent a major site of convergence for multiple descending and reflex pathways that co-ordinate sympathetic nerve activity. Elucidating the organization of the circuits that drive these neurons is a key research objective. Here we present brain-wide connectomic maps of neurons that provide monosynaptic drive to putative RVLM sympathetic premotor neurons, generated using a two-step restricted trans-synaptic viral tracing strategy. We made focal microinjections of a genetically restricted reporter-expressing rabies vector, SADΔG(EnvA)-RFP, into the RVLM, and restricted its entry to neurons that project to the T2 spinal cord by first transducing them with a cassette required for rabies entry and trans-synapsis, YTB. By precisely targeting rabies injections to the core of the RVLM bulbospinal population we were able to restrict rabies ‘seeding’ to small numbers (as low as one single neuron) of RVLM presympathetic neurons immediately caudal to the facial nucleus, and then map the locations of neurons that provide monosynaptic input to the seed population. We observed reproducible patterns of inputs arising from the dorsal, contralateral, and midline medulla, and local RVLM interneurons including catecholaminergic non-bulbospinal neurons and neurons likely to reside within the ventral respiratory column. Distant inputs were identified in the pons, cerebellum and midbrain, and included previously suspected sites of monosynaptic drive such as the paraventricular nucleus of the hypothalamus.

Leptin Inhibits Presympathetic Neurons in the Rostral Ventrolateral Medulla

Leptin is greatly involved in the regulation of food intake, and dysfunction of leptin or its receptors is associated with obesity. Numerous evidence indicate that obesity is linked to increased prevalence of hypertension. The sympathetic nervous system (SNS) is greatly involved in the regulation of arterial blood pressure and neurons in the rostral ventrolateral medulla (RVLM) are critical components of both the SNS and cardiovascular regulation. In this study we used whole-cell patch-clamp recordings from presympathetic kidney-related RVLM neurons identified with retrograde viral labeling and tested the hypothesis that leptin reduces neuronal excitability of RVLM neurons. Application of leptin (500 nM) caused a rapid membrane hyperpolarization (3.7 ± 1.4 mV change) in half of the recorded presympathetic RVLM neurons without affecting the rest of the recorded cells. The hyperpolarization was accompanied with an increase of input resistance from 287 ± 95 MΩ to 336 ± 97 MΩ. In addition, leptin decreased the frequency of spontaneous excitatory postsynaptic currents (sEPSC) by 35.6 ± 0.1% without changing sEPSC amplitude. In contrast, leptin did not alter the inhibitory neurotransmission. In summary, our data demonstrate that leptin reduces the excitability of a subset of presympathetic RVLM neurons via pre- and postsynaptic mechanisms and suggest a potential control mechanism of SNS activity. Electrophysiological Properties of Rostral Ventrolateral Medulla Presympathetic Neurons Modulated by the Respiratory Network in Rats

The respiratory pattern generator modulates the sympathetic outflow, the strength of which is enhanced by challenges produced by hypoxia. This coupling is due to the respiratory-modulated presympathetic neurons in the rostral ventrolateral medulla (RVLM), but the underlining electrophysiological mechanisms remain unclear. For a better understanding of the neural substrates responsible for generation of this respiratory-sympathetic coupling, we combined immunofluorescence, single cell qRT-pCR, and electrophysiological recordings of the RVLM presympathetic neurons in in situ preparations from normal rats and rats submitted to a metabolic challenge produced by chronic intermittent hypoxia (CIH). Our results show that the spinally projected cathecholaminergic C1 and non-C1 respiratory-modulated RVLM presympathetic neurons constitute a heterogeneous neuronal population regarding the intrinsic electrophysiological properties, respiratory synaptic inputs, and expression of ionic currents, albeit all neurons presented persistent sodium current-dependent intrinsic pacemaker properties after synaptic blockade. A specific subpopulation of non-C1 respiratory-modulated RVLM presympathetic neurons presented enhanced excitatory synaptic inputs from the respiratory network after CIH. This phenomenon may contribute to the increased sympathetic activity observed in CIH rats. We conclude that the different respiratory-modulated RVLM presympathetic neurons contribute to the central generation of respiratory-sympathetic coupling as part of a complex neuronal network, which in response to the challenges produced by CIH contribute to respiratory-related increase in the sympathetic activity.

Patterning of somatosympathetic reflexes reveals nonuniform organization of presympathetic drive from C1 and non-C1 RVLM neurons

To determine the organization of presympathetic vasomotor drive by phenotypic populations of rostral ventrolateral medulla (RVLM) neurons, we examined the somatosympathetic reflex (SSR) evoked in four sympathetic nerves together with selective lesions of RVLM presympathetic neurons. Urethane-anesthetized (1.3 g/kg ip), paralyzed, vagotomized and artificially ventilated Sprague-Dawley rats (n = 41) were used. First, we determined the afferent inputs activated by sciatic nerve (SN) stimulation at graded stimulus intensities (50 sweeps at 0.5-1 Hz, 1-80 V). Second, we recorded sympathetic nerve responses (cervical, renal, splanchnic, and lumbar) to intensities of SN stimulation that activated A-fiber afferents (low) or both A- and C-fiber afferents (high). Third, with low-intensity SN stimulation, we examined the cervical SSR following RVLM microinjection of somatostatin, and we determined the splanchnic SSR in rats in which presympathetic C1 neurons were lesioned following intraspinal injections of anti-dopamine-β-hydroxylase-saporin (anti-DβH-SAP). Low-intensity SN stimulation activated A-fiber afferents and evoked biphasic responses in the renal, splanchnic, and lumbar nerves and a single peak in the cervical nerve. Depletion of presympathetic C1 neurons (59 ± 4% tyrosine hydroxylase immunoreactivity profiles lesioned) eliminated peak 2 of the splanchnic SSR and attenuated peak 1, suggesting that only RVLM neurons with fast axonal conduction were spared. RVLM injections of somatostatin abolished the single early peak of cervical SSR confirming that RVLM neurons with fast axonal conduction were inhibited by somatostatin. It is concluded that unmyelinated RVLM presympathetic neurons, presumed to be all C1, innervate splanchnic, renal, and lumbar but not cervical sympathetic outflows, whereas myelinated C1 and non-C1 RVLM neurons innervate all sympathetic outflows examined. These findings suggest that multiple levels of neural control of vasomotor tone exist; myelinated populations may set baseline tone, while unmyelinated neurons may be recruited to provide actions at specific vascular beds in response to distinct stressors.

Low dose of moxonidine within the rostral ventrolateral medulla improves the baroreflex sensitivity control of sympathetic activity in hypertensive rat

To determine the effects of the centrally antihypertensive drug moxonidine injected into the rostral ventrolateral medulla (RVLM) on baroreflex function in spontaneously hypertensive rats (SHR). Baroreflex sensitivity control of renal sympathetic nerve activity (RSNA) and barosensitivity of the RVLM presympathetic neurons were determined following application of different doses of moxonidine within the RVLM. Three doses (0.05, 0.5, and 5 nmol in 50 nL) of moxonidine injected bilaterally into the RVLM dose-dependently reduced the baseline blood pressure (BP) and RSNA in SHR. At the highest dose (5 nmol) of moxonidine injection, the maximum gain (1.24%+/-0.04%/mmHg) of baroreflex control of RSNA was significantly decreased. However, the lower doses (0.05 and 0.5 nmol) of moxonidine injection into the RVLM significantly enhanced the baroreflex gain (2.34%+/-0.08% and 2.01%+/-0.07%/mmHg). The moxonidine-induced enhancement in baroreflex function was completely prevented by the imidazoline receptor antagonist efaroxan but not by the alpha(2)-adrenoceptor antagonist yohimbine. A total of 48 presympathetic neurons were recorded extracellularly in the RVLM of SHR. Iontophoresis of applied moxonidine (30-60 nA) dose-dependently decreased the discharge of RVLM presympathetic neurons but also significantly increased the barosensitivity of RVLM presympathetic neurons. These data demonstrate that a low dose of moxonidine within the RVLM has a beneficial effect on improving the baroreflex function in SHR via an imidazoline receptor-dependent mechanism.

Tonic glutamatergic input in the rostral ventrolateral medulla is increased in rats with chronic heart failure.

Chronic heart failure (CHF) is characterized by increased sympathetic tone. The glutamatergic input in the rostral ventrolateral medulla (RVLM), which is a key region involved in sympathetic outflow, seems not to be involved in the generation of sympathetic tone in the normal state. The aim of this study was to determine the role of the RVLM glutamate receptors in the generation of sympathetic tone in CHF. CHF was produced by coronary artery ligation. Bilateral microinjection of the glutamate receptor antagonist kynurenic acid, the N-methyl-d-aspartate (NMDA) receptor antagonist D-2-amino-5-phosphonopentanoate, or the non-NMDA receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione into the RVLM dose-dependently reduced resting blood pressure and renal sympathetic nerve activity in CHF but not in sham rats. Picoinjection of kynurenic acid (100 pmol in 5 nL) significantly decreased the basal discharge by 47% in 25 RVLM presympathetic neurons in CHF rats. In contrast, kynurenic acid had no effect on the discharge in all 22 of the RVLM presympathetic neurons tested in sham rats. These data suggest that upregulated glutamate receptors, including NMDA and non-NMDA, in the RVLM are involved in tonic control of elevated sympathetic tone in CHF.

Somatostatin 2A Receptor-Expressing Presympathetic Neurons in the Rostral Ventrolateral Medulla Maintain Blood Pressure

Bulbospinal neurons in the rostral ventrolateral medulla (RVLM) are critical for the maintenance of sympathetic vasomotor tone and normal cardiovascular reflex function. So far, selectively eliminating/inhibiting distinct subpopulations of RVLM neurons has not significantly altered arterial pressure. Here we show that RVLM presympathetic neurons that express somatostatin 2A receptors are essential for maintaining and potentially generating sympathetic vasomotor tone. Combined immunocytochemistry and in situ hybridization were used to map the expression of somatostatin receptors 1, 2A, 2B, 3, and 4 (sst1 through 4, respectively) in the rat RVLM. sst1 and sst2B were absent; sst3 and sst4 were sparse. However, sst2A was found postsynaptically and detected in 35+/-5% of bulbospinal RVLM neurons a population that included 54+/-4% of catecholaminergic and 30+/-3% of enkephalinergic neurons. Bilateral microinjection into the RVLM of either somatostatin or the receptor-selective agonist lanreotide evoked dramatic, dose-dependent sympathoinhibition, hypotension, and bradycardia that were blocked by the sst2 receptor antagonist BIM-23627 in anesthetized rats. Bilateral RVLM microinjection of somatostatin also attenuated chemoreceptor and somatosympathetic reflex function. Somatostatin only eliminated the first sympathoexcitatory peak evoked by somatosympathetic reflex activation, whereas muscimol abolished both excitatory peaks providing functional evidence that the activity of only a subpopulation of RVLM presympathetic neurons is inhibited by somatostatin. We suggest that the subpopulation of bulbospinal RVLM neurons that expresses the sst2A receptor sets sympathetic vasomotor output. These neurons are essential for maintaining resting blood pressure under anesthesia and contribute to adaptive reflexes mediated through the RVLM.

Role of ventrolateral medulla in generating the 10-Hz rhythm in sympathetic nerve discharge

We recorded changes in right inferior cardiac and either left inferior cardiac or left vertebral sympathetic nerve discharge (SND) produced by unilateral microinjections of GABA-A and excitatory amino acid (EAA) receptor antagonists into the ventrolateral medulla (VLM) of urethane-anesthetized, baroreceptor-denervated cats. Unilateral microinjections of GABA-A receptor antagonists, SR-95531 or bicuculline, into single tracks in VLM anywhere between 1 and 5 mm rostral to the obex eliminated or markedly reduced 10-Hz power in SND on both sides of the body. Low-frequency components (<6 Hz) of SND were unaffected. Complete blockade of the 10-Hz rhythm occurred with a dose of SR-95531 as low as 6.25 pmol in a 50-nl volume. Unilateral microinjections of the nonselective EAA receptor antagonist, kynurenate (KYN; 7.5 nmol), into the caudal or rostral VLM significantly reduced, but did not eliminate, 10-Hz SND ipsilateral to the injection sites, while 10-Hz SND contralateral to the injection sites was not significantly changed. These observations suggest that 1) GABAergic transmission in VLM is critical for generation of the 10-Hz rhythm, 2) the caudal and rostral portions of VLM act together to generate the 10-Hz rhythm, and 3) 10-Hz rhythm generation depends, at least in part, on tonic or phasic excitatory drive to GABAergic interneurons in caudal VLM and presympathetic neurons in rostral VLM. The data also suggest that pathways interconnecting the two halves of the brain stem play an important role in promoting 10-Hz rhythm generation.

Direct Evidence for Imidazoline (I1) Receptor Modulation of Ethanol Action on Norepinephrine‐Containing Neurons in the Rostral Ventrolateral Medulla in Conscious Spontaneously Hypertensive Rats

Enhancement of the rostral ventrolateral medulla (RVLM) presympathetic (norepinephrine, NE) neuronal activity represents a neurochemical mechanism for the pressor effect of ethanol. In this study, we tested the hypothesis that ethanol action on RVLM presympathetic neurons is selectively influenced by the signaling of the local imidazoline (I1) receptor. To support a neuroanatomical and an I1-signaling selectivity of ethanol, and to circumvent the confounding effects of anesthesia, the dose-related neurochemical and blood pressure effects of ethanol were investigated in the presence of selective pharmacological interventions that cause reduction in the activity of RVLM or nucleus tractus solitarius (NTS) NE neurons via local activation of the I1 or the alpha2-adrenergic receptor in conscious spontaneously hypertensive rats. Local activation of the I1 receptor by rilmenidine (40 nmol) or by the I1/alpha2 receptor mixed agonist clonidine (1 nmol), and local activation of the alpha2-adrenergic receptor (alpha2AR) by the pure alpha2AR agonist alpha-methylnorepinephrine (alpha-MNE, 10 nmol) caused reductions in RVLM NE, and blood pressure. Intra-RVLM ethanol (1, 5, or 10 microg), microinjected at the nadir of the neurochemical and hypotensive responses, elicited dose-dependent increments in RVLM NE and blood pressure in the presence of local I1--but not alpha2-receptor activation. Only intra-NTS alpha-MNE, but not rilmenidine or clonidine, elicited reductions in local NE and blood pressure; ethanol failed to elicit any neurochemical or blood pressure responses in the presence of local activation of the alpha2AR within the NTS. The findings support the neuroanatomical selectivity of ethanol, and support the hypothesis that the neurochemical (RVLM NE), and the subsequent cardiovascular, effects of ethanol are selectively modulated by I1 receptor signaling in the RVLM.

Pancreatic vasoconstrictor responses are regulated by neurons in the rostral ventrolateral medulla

The pancreas receives sympathetic input which arises from several premotor cell groups in the CNS including the rostral ventrolateral medulla (RVLM). In this study, we examined the influence of electrical stimulation of the RVLM on pancreatic blood flow measured by laser Doppler flowmetry and gastric blood flow measured by ultrasonic Doppler flowmetry in halothane-anesthetized rats. The laser Doppler flow measurement technique was validated by demonstration that pancreatic conductance was reduced by systemic administration of the vasoconstrictor phenylephrine and increased by the vasodilator sodium nitroprusside. Sympathetic vasomotor withdrawal induced by either administration of phenylbiguanide (2 and 10 μg/kg, i.v.) or electrical stimulation of the central end of the cervical vagal trunk (5 Hz, 2 ms, 50–150 μA) produced depressor responses and increases in pancreatic and gastric vascular conductance. Electrical stimulation of the RVLM (50 Hz, 0.5 ms, 25–75 μA) produced pressor and tachycardic responses accompanied by decreases in pancreatic and gastric vascular conductance. All responses to RVLM stimulation were abolished by blockade of ganglionic neurotransmission (hexamethonium bromide, 20 mg/kg, i.v.). These data suggest that RVLM presympathetic vasomotor neurons are a primary source of tonic sympathetic vasomotor drive to the pancreatic and gastric vasculature.

Baroreceptor reflex stimulation does not induce cytomegalovirus promoter-driven transgene expression in the ventrolateral medulla in vivo

Adenoviruses are being employed to induce transgene expression in the central nervous system in vivo. In these studies, the cytomegalovirus (CMV) promoter is commonly employed to drive expression of the transgene because of its strong, constitutive activity in a wide range of cell types. However, using this promoter, expression in neurons is variable, with strongest expression being observed in non-neuronal cells. Indeed, even in vitro, CMV driven expression in neurons is variable. In cultured sympathetic ganglion cells it has been demonstrated that CMV-driven expression requires activation of cAMP-response element-binding protein (CREB) and that this can be induced by depolarization. In this study we tested whether depolarization might induce CMV-driven transgene expression, delivered by microinjection of an adenovirus, in the rostral ventrolateral medulla (RVLM) of rats. Prior to stimulation, transgene expression occurs in non-neuronal cells in the RVLM. Some neuronal expression was observed in neighbouring regions, in the nucleus ambiguus and in facial motor neurons. Within the RVLM, depolarization, induced by intraperitoneal administration of the ganglion blocking drug, pentolinium, did not lead to induction of transgene expression. This stimulus is known to induce expression of the immediate early gene c- fos. We conclude that either this experimental paradigm was not sufficient for activation of the CREB pathway or that possibly the virus does not gain access to the neurons of the RVLM. The adoption of specific promoters or viruses with higher neuronal transduction efficiency appears to be essential for the genetic modification of RVLM presympathetic neurons in vivo.

Fractal noises and motions in time series of presympathetic and sympathetic neural activities

We used Allan factor analysis to classify time series of the discharges of single presympathetic neurons in the cat medullary lateral tegmental field (LTF) and rostral ventrolateral medulla (RVLM) and of the postganglionic vertebral sympathetic nerve. These time series fell into two classes of fractal signals characterized by statistically self-similar behavior reflecting long-range correlations among data points. Classification of a time series as either fractional Gaussian noise (fGn) or fractional Brownian motion (fBm) was based on the scaling exponent, alpha, of the power law in the Allan factor curve. fGn is defined as 0 < alpha < 1 and fBm as 1 < alpha < 3. All but one of the spike trains of 12 classifiable LTF neurons with sympathetic nerve-related activity were fGn. In contrast, the spike trains of 8 of 9 RVLM presympathetic neurons that could be classified were fBm. The time series of simultaneously recorded vertebral sympathetic nerve discharge and heart period fell into the fBm class. Because a fBm signal is the cumulative sum of the elements comprising the corresponding fGn signal, these results demonstrate smoothing of fractal time series in a feedforward direction from medullary presympathetic neurons to postganglionic sympathetic neurons. Whether this involves integration by RVLM neurons of their LTF inputs or independent fractal processes acting at different levels of the network controlling sympathetic nerve discharge remains to be determined. Cholecytokinin activates baro‐activated neurons in rat caudal ventrolateral medulla

Cholecystokinin (CCK) is a gastointestinal hormone that produces intestinal vasodilation, which appears to be mediated by reduced sympathetic nerve activity to the gut. Intravenous injection of CCK inhibits splanchnic nerve activity (SNA) and reduces the firing of a subset of pre-sympathetic neurons in the rostral ventrolateral medulla (RVLM) in chloralose-anesthetized rats. We determined whether the inhibition of RVLM neurons could be produced by the activation of GABAergic neurons in the caudal ventrolateral medulla (CVLM). In chloralose-anesthetized, artificially ventilated, paralyzed rats, we measured the effects of CCK (10 μg/kg, iv) on arterial pressure (AP), SNA, heart rate, and the firing of baro-activated, pulse-modulated CVLM neurons. CCK decreased SNA (49±10 %), heart rate (42±7 bpm), and AP (24±10 mmHg). The firing of 3 of 5 baro-activated CVLM neurons was increased by CCK (4 to 11 spikes/sec), but 2 of 5 did not change or were inhibited during the accompanying decrease in AP. In contrast, 8 of 10 neighboring baro-inhibited neurons were inhibited by CCK (3 to 0.4 spikes/sec). Four of these cells were silenced briefly. These data suggest a subset of baro-activated CVLM neurons are activated by CCK and may produce the inhibition observed in some pre-sympathetic RVLM neurons. Furthermore, these data suggest that baro-activated CVLM neurons may be subdivided into CCK-sensitive and CCK-insensitive neurons. Supported by NIH: RO1 HL075174.

The time series of simultaneously recorded vertebral sympathetic nerve discharge and the arterial pulse also were fBm-based signals. Because a fBm signal is the cumulative sum of the elements comprising the corresponding fGn signal, these results show smoothing of fractal time series in a feedforward direction from medullary presympathetic neurons to postganglionic sympathetic neurons. This may involve integration by RVLM neurons of their LTF inputs or independent fractal processes acting at different levels of the network controlling sympathetic nerve discharge. Whether feedforward smoothing of fractal signals is a feature in other neural systems is open to investigation.
